The objective of this experiment was to evaluate the effect of supplementing rumen-protected Lys based on a Lys-deficient diet on liver metabolism in growing Holstein heifers. The experiment was conducted for 3 months with 36 Holstein heifers (initial body weight: 200 ± 9.0 kg; 7-month-old). Heifers were randomly assigned to 2 diets based on corn, soybean meal, alfalfa hay, and wheat bran: control, Lys-deficient diet (LD; 0.66% Lys in diet), and Lys-adequate diet (LA; 1.00% Lys in diet). The results showed no difference in growth performance between the 2 groups (P > 0.05). However, there was a clear trend of increasing feed conversion rate with Lys supplementation (0.05 < P < 0.01). The serum urea nitrogen concentration was significantly decreased, and the aspartate aminotransferase-to-alanine aminotransferase ratio was significantly decreased by Lys supplementation (P < 0.05). Moreover, growing heifers fed a Lys-adequate diet had lower levels of urine nitrogen excretion and higher levels of the biological value of nitrogen (P < 0.05). Metabolomic analysis revealed that 5 types of phosphatidylcholine and 3 types of ceramide were significantly increased and enriched in sphingolipid metabolism and glycerophospholipid metabolism (P < 0.05). His, Leu, and Asp levels were significantly decreased in the liver following Lys supplementation (P < 0.05). In conclusion, Lys supplementation may promote the synthesis of body tissue proteins, as evidenced by significantly decreased amino acids in the liver and urine N excretion, it also improves hepatic lipid metabolism by providing lipoprotein precursors.  相似文献

This study was conducted to investigate the effects of spray-dried porcine plasma protein (SDPP) or spray-dried chicken plasma protein (SDCP) supplementation in diets without the inclusion of antibiotics and zinc oxide (ZnO) on growth performance, fecal score, and fecal microbiota in early-weaned piglets. A total of 192 healthy weaning piglets (Duroc × Landrace × Yorkshire, 21 d old) were blocked by BW (6.53 ± 0.60 kg) and randomly assigned to 4 dietary treatments: negative control (NC, basal diet), positive control (PC), basal diet + ZnO at 2 g/kg and antibiotics at 0.8 g/kg), SDPP (containing 5% SDPP), and SDCP (containing 5% SDCP). The experiment lasted 14 d. The SDPP group had higher (P < 0.05) final BW, average daily gain and average daily feed intake than the NC and SDCP groups. The percentage of piglets with fecal scores at 2 or ≥2 was higher (P < 0.05) in the NC and SDCP groups than in the PC group. A decreased (P < 0.05) bacterial alpha diversity and Bacteroidetes abundance, but increased (P < 0.05) Firmicutes abundance were observed in the PC and SDPP groups when compared to the NC group. The relative abundance of Lactobacillus was higher (P < 0.05) in the SDPP than in the SDCP group, and that of Streptococcus was higher (P < 0.01) in the PC and SDPP groups than in the NC group. The PC group also had higher (P < 0.01) Faecalibacterium abundance than the NC and SDCP groups. Additionally, the SDCP group had higher (P < 0.05) serum urea nitrogen than those fed other diets, and lower (P < 0.10) short-chain fatty acids to branched-chain fatty acids ratio than the PC and SDPP groups. Overall, SDPP was a promising animal protein for piglets in increasing feed intake, modifying gut microbiota profile, reducing gut protein fermentation and alleviating diarrhea frequency, thus promoting growth performance, under the conditions with limited in-feed utilization of antibiotics and ZnO.  相似文献

The purpose of this study was to investigate the potential benefits of yeast cell wall (YCW) on the gastrointestinal development of weaned calves. Twenty healthy Holstein male calves (BW = 92 ± 8.29 kg and 60 ± 5 d of age) were randomly allocated into 2 groups: CON with no YCW, and YCW (accounted for 0.16% of the basal diet). The dietary concentrate-to-roughage ratio was 40:60. All the calves were fed regularly twice a day at 09:00 and 16:00 and had free access to water. The experiment lasted for 60 d. The results showed that calves fed YCW showed higher (P < 0.05) length, width, and surface area of papillae in the ventral sac of the rumen as compared to CON. For the dorsal sac of the rumen, the muscularis thickness was thicker (P < 0.05) in the YCW group when compared with CON group. The villus height of YCW calves was higher (P < 0.05) than that of CON in the ileum. Calves supplemented with YCW also showed a higher (P < 0.05) villus height-to-crypt depth ratio in the ileum. The YCW calves exhibited a greater (P < 0.05) thickness of the wall in the duodenum and jejunum. Calves supplemented with YCW improved (P < 0.05) the claudin 1 mRNA expression in the ileum and occludin mRNA expression in the jejunum and ileum. The YCW increased (P < 0.05) the contents of secretory immunoglobulin A in the jejunum and ileum of calves. In conclusion, dietary supplementation with YCW could improve the gastrointestinal development of weaned calves.  相似文献

The uptake and metabolism of methionine (Met) are critical for epigenetic regulation, redox homeostasis, and embryo development. Our previous study showed that appropriate supplementation of dietary Met promoted the birth weight and placental angiogenesis of high-prolific sows. To further explore the metabolic effect of Met on pregnant sows, we have evaluated the influence of dietary Met level on Met metabolism, and the relationship between metabolites of Met and reproductive performance, antioxidant ability, and placental angiogenesis throughout the gestation of high-prolific sows. Sixty sows (the 3rd parity, Large White) were randomly divided into 5 groups that were fed diets with standardized ileal digestible (SID) methionine-to-lysine (Met:Lys) ratios of 0.27 (control), 0.32, 0.37, 0.42, and 0.47 from the mating day (gestational d 0, G0d) until the farrowing day. HPLC-MS/MS analysis was used to simultaneously evaluate the metabolites related to Met, e.g. S-adenosylmethionine (SAM), S-adenosylhomocysteine (SAH), homocysteine (Hcy), cysteine (Cys), and glutathione (GSH). The concentration of SAM and SAH in plasma had significant fluctuations, especially in late pregnancy. Increasing dietary Met supplementation significantly improved the plasma SAM and methylation potential (SAM-to-SAH ratio) at d 114 of pregnancy (G114d). Moreover, a positive association of the plasma SAM concentration at G114d was observed with the litter weight of born alive (P < 0.05; R2 = 0.58). Furthermore, Hcy concentration in plasma was at the lowest level for 0.37 ratio group at G114d. However, it significantly increased during late pregnancy. Moreover, there were negative correlations between plasma Hcy concentration at G114d (P < 0.05) and the placental vascular density in the fold and stroma (P < 0.05). Compared with the control group, the expression of vascular endothelial growth factor-A (VEGF-A) in the placenta tissue of 0.37 ratio group increased significantly (P < 0.05). Collectively, these findings indicate that dietary Met:Lys ratio (0.37 to 0.57) in the pregnant diet dose not influence the antioxidant ability of the high-prolific sows; however, the improvement of fetal development and placental angiogenesis of high-prolific sows by supplementation of Met are closely associated to the key Met-related metabolite of SAM and Hcy, respectively.  相似文献

This study was performed to evaluate the effect of folic acid (FA) on performance, egg quality and yolk 5-methyltetrahydrofolate (5-MTHF) content. A total of 384 Hy-line W36 strain hens from 52 to 58 weeks of age were randomly assigned to 4 groups, and each group received one of following dietary treatments: 0, 5, 10 and 15 mg FA/kg diet. A completely randomized design was used. Egg production percentage, egg mass and egg weight were increased significantly (P < 0.05) and feed conversion ratio (FCR) was reduced significantly (P < 0.05) by increasing FA content in diets. No significant differences were detected among treatments on egg quality except for shell thickness. The dietary supplementations of laying hens diets with FA significantly increased yolk 5-MTHF content (P = 0.02). Overall, these data demonstrate that dietary supplementation with FA raised 5-MTHF content of yolk and productivity of egg production in laying hens.  相似文献

Lycium barbarum polysaccharide (LBP) is a traditional Chinese herbal medicine possessing many properties such as anti-oxidant and immunomodulation. This study was conducted to evaluate the clinical efficacy of LBP in broilers. A total of 240 newly hatched broilers were randomly allotted into 4 treatments with 6 replicates. Diets in 3 treatment groups were supplemented with 2, 4, and 8 g/kg LBP, respectively, based on the control diet. Lymphocytes isolated from broiler blood were used to detect immunoregulation function of LBP in vitro. Results in vivo showed that average daily feed intake (ADFI) and feed conversion ratio (FCR) in the 4 g/kg LBP group were decreased significantly (P < 0.05). Dietary 4 g/kg LBP significantly increased immune organ indexes of broilers at 21 d (P < 0.05). Moreover, levels of serum total protein, globulin, albumin, and lysozyme of broilers were higher in the 8 g/kg LBP group (P < 0.05). CD4+/CD8+ ratio in blood T cells was higher in the 4 g/kg LBP group (P < 0.05). Results in vitro suggested that 100 and 1,600 μg/mL LBP supplementation significantly promoted blood B and T lymphocyte proliferation of broilers, respectively (P < 0.05). TNF-α mRNA abundance was observably decreased in the lower LBP concentration groups (P < 0.05). In conclusion, LBP may possess the clinical efficacy for growth promotion and immunomodulation and could be used as an alternative for nutritive additive in broilers.  相似文献

An experiment was designed to determine the influence of fibre and betaine on the development of the intestine, liver and pancreas of broilers from hatch to 14 d of age. A total of 250-day-old Cobb 500 male broilers were allocated to 16 cages with 15 broilers each. Treatments were arranged in a 2 × 4 factorial design, consisting of 2 feed formulations (low and high fibre diets) and 4 levels of betaine (0, 1, 3 or 5 kg/t). At hatch, 10 birds in total were euthanised, and samples of the liver, pancreas, yolk sac and intestine were collected for reference of the analysed parameters before the start of the trial. On d 4, 9 and 14, 5 birds per cage (10 birds per treatment) were selected, euthanised and treated as the same as the birds at hatch. Villus height and width and crypt depth were determined on the duodenum samples, and absorptive area was calculated. The number of enterocytes in mitosis at the villus was determined by a positive reaction to antibody for Ki67 protein, and fused villus was evaluated visually. The relative weight of the yolk sac reduced (P < 0.05) as birds aged while the intestine and liver reached a maximum (P < 0.05) at around d 4 and the pancreas at d 9. Birds fed the high fibre diet had greater feed intake, lower relative weight of the pancreas and higher villus (P < 0.05) than birds fed the low fibre diet. Villus width increased (P < 0.05) at 4 d of age, and this was associated with fused villus. Betaine inclusion reduced (P < 0.05) villus width, increased (P < 0.05) villus size and absorptive area, and reduced (P < 0.05) the number of enterocytes with positive reaction for the antibody Ki-67. Betaine inclusion reduced the width and increased the absorptive area and the villus height of the duodenum of birds up to 14 d of age. The higher fibre diet increased feed intake and villus height, yet reduced pancreas relative weight, while not affecting body weight gain. This response was possibly due to a dilution effect of the fibre, reducing nutrient absorption and consequently stimulating villus growth to improve absorption rates.  相似文献

In ovo feeding of vitamin C (VC) has positive effects on the growth performance, immune and antioxidant function in poultry, which indicates that increasing VC content in eggs may be of benefit. This study was to investigate the effects of dietary VC supplementation on VC synthesis and transportation and egg deposition. In Exp. 1, in order to select a suitable animal model, VC content was detected in different eggs from different layer species. Vitamin C content was lower in ISA Brown breeder eggs and Hy-Line Brown layer eggs (P < 0.05) then in Arbor Acres breeder eggs. In Exp. 2, a total of 24 Hy-Line Brown layers (42-week-old) were randomly divided into 3 treatments with 8 replicates and fed a basal diet with VC at 0, 200 and 400 mg/kg. Sodium-dependent VC transporter 1 and 2 (SVCT1 and SVCT2) expressions were higher in ileum than in duodenum and jejunum (P < 0.05). SVCT1 expression was higher but SVCT2 expression was lower in the magnum than in the ovary (P < 0.05). L-Gulonolactone oxidase (GLO) and SVCT1 expressions were higher but SVCT2 was lower in the kidney than in the liver (P < 0.05). Dietary VC supplementation at 400 mg/kg increased SVCT1 expression in duodenum, ovary and magnum, but decreased GLO and SVCT1 expression in liver (P < 0.05). Dietary VC supplementation at 200 and 400 mg/kg increased SVCT2 expression in duodenum, but decreased GLO and SVCT1 expression in kidney and SVCT2 expression in liver (P < 0.05). Dietary VC supplementation promoted VC absorption in duodenum and jejunum, but reduced endogenous VC synthesis in liver and kidney. Although dietary VC supplementation enhanced VC transportation in ovary and magnum, it did not increase VC deposition in produced eggs.  相似文献

The objective of this experiment was to investigate the potential benefits of active dry yeast (ADY) on the growth performance, rumen fermentation, nutrient digestibility, and serum parameters of weaned beef calves. Thirty Simmental crossbred male calves (body weight = 86.47 ± 4.41 kg and 70 ± 4 d of age) were randomly divided into 2 groups: control (CON) (fed basal ration) and ADY (fed basal ration and 5 g/d ADY per calf). The dietary concentrate-to-roughage ratio was 35:65. All the calves were regularly provided rations 3 times a day at 07:00, 13:00, and 19:00 and had free access to water. The experiment lasted for 60 d. The average daily gain of ADY group was higher (P = 0.007) than that of the CON group, and the ratio of feed intake to average daily gain in the ADY group was reduced (P = 0.022) as compared to the CON group. The concentration of ruminal ammonia-N was higher (P = 0.023) in the CON group than that in the ADY group, but an opposite trend of microbial protein was found between the 2 groups. Also, the ruminal concentrations of propionate and butyrate were higher (P < 0.05) in the ADY group than those in the CON group. Calves fed ADY exhibited higher (P < 0.05) crude protein and neutral detergent fiber digestibility. Supplementation of ADY increased (P < 0.05) the contents of glucose, glutathione peroxidase, superoxide dismutase, immunoglobulin A, immunoglobulin M, and interleukin 10 in the serum of calves, but an opposite trend was observed in malondialdehyde, interleukin 1 beta, and tumor necrosis factor alpha contents between the 2 groups. In conclusion, dietary supplementation with ADY could improve the growth performance, rumen fermentation, nutrient digestibility, antioxidant ability, and immune response of weaned beef calves.  相似文献

Animal protein sources such as fishmeal and plasma powder are excellent and indispensable sources of energy, amino acids, and minerals in animal production. Amino acid imbalance, especially methionine-to-sulfur amino acid (Met:SAA) ratio, caused by an imbalance of animal protein meal leads to growth restriction. This study was conducted to evaluate the effects of imbalanced Met:SAA ratio supplementation of different animal protein source diets on growth performance, plasma amino acid profiles, antioxidant capacity and intestinal morphology in a piglet model. Twenty-four weaned piglets (castrated males; BW = 10.46 ± 0.34 kg), assigned randomly into 3 groups (8 piglets/group), were fed for 28 d. Three experimental diets of equal energy and crude protein levels were as follows: 1) a corn-soybean basal diet with a Met:SAA ratio at 0.51 (BD); 2) a plasma powder diet with a low Met:SAA ratio at 0.41 (L-MR); 3) a fishmeal diet with a high Met:SAA ratio at 0.61 (H-MR). Results revealed that compared to BD, L-MR significantly decreased (P < 0.05) the activities of plasma total antioxidant capacity and glutathione peroxidase, plasma amino acid profiles, and significantly reduced (P < 0.05) villus height and crypt depth in the duodenum and jejunum. Additionally, L-MR significantly reduced (P < 0.05) the mRNA expression level of solute carrier family 7 member 9 (SlC7A9) in the ileum, and significantly increased (P < 0.05) mRNA expression levels of zonula occludens-1 (ZO-1) in the duodenum, and Claudin-1, ZO-1, sodium-coupled neutral amino acid transporters 2 (SNAT2) and SlC7A7 in the jejunum. H-MR significantly increased (P < 0.05) plasma SAA levels, and significantly reduced (P < 0.05) average daily feed intake, villus height, and villus height-to-crypt depth (VH:CD) ratio in the ileum compared to BD. In conclusion, L-MR may result in oxidative stress and villous atrophy but proves beneficial in improving intestinal barrier function and the activity of amino acid transporters for compensatory growth. H-MR may impair intestinal growth and development for weaned piglets. The research provides a guidance on the adequate Met:SAA ratio (0.51) supplementation in diet structure for weaned piglets.  相似文献

Current study investigated the effect of feeding systems (conventional vs organic rearing) on carcass characteristics, meat quality and fatty acid composition of Musculus longissimus dorsi of Korean native black barrows (KNP). Thirty pigs were reared under a conventional feeding system at indoor area of 1 m2 per head, while another thirty pigs were fed an indoor area with organic saw dusts of 1 m2 and an outdoor area with free ranges of 1 m2 for each pig for organic system. Diet for the organic rearing was also provided according to the guideline for organic pork products. Warner–Bratzler shear force was lower and water holding capacity was higher for pork produced under the organic guidelines. However, pork produced by an organic system did not affect sensory traits compared to pork produced by a conventional one. In addition, longissimus muscle from organically reared pigs had significantly (P < 0.05) higher myoglobin content and consequently higher CIE a?-values compared with those for the conventionally-reared pigs (P < 0.05). The organic pork resulted in significantly (P < 0.05) higher polyunsaturated fatty acid (PUFA) and unsaturated fatty acid contents, as well as a higher n-3 PUFA than the conventional one (P < 0.05).  相似文献

Certain hormones play important roles in modulating mammalian reproductive behaviour. Daidzein is a well-known isoflavonic phytoestrogen that possesses oestrogenic activity. This study was conducted to probe the effects of daidzein supplementation in gestation diets on the reproductive performance in sows. A total of 120 multiparous sows (Landrace × Yorkshire) were randomly assigned to 2 groups (n = 60) and fed either a base diet (control) or one containing 200 mg/kg daidzein during gestation. We discovered that daidzein supplementation significantly increased the total number of piglets born per litter and number of piglets born alive per litter (P < 0.05), decreased the farrowing time (P < 0.05) and increased the serum oestrogen and progesterone concentrations (P < 0.05) at 35 d of gestation. Moreover, serum immunoglobulin G (IgG) concentration and superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities were higher in the daidzein-treated group than in the control group at 35 d of gestation (P < 0.05). Daidzein increased the serum SOD activity and total anti-oxidative capacity (T-AOC) at 85 d of gestation (P < 0.05). Interestingly, daidzein elevated the expression levels of the sodium-coupled neutral amino acid transporter 1 (SLC38A1) and insulin-like growth factor 1 (IGF-1) genes in the placenta (P < 0.05). These results suggest that daidzein ingestion could improve sow reproductive performance by changing serum hormones, elevating anti-oxidative capacity and up-regulating critical functional genes in the placenta.  相似文献

This experiment was conducted to study the effects of dietary supplementation with acidifiers on the growth performance, meat quality, and intestinal health of broiler chickens. A total of 648 male Arbor Acres broiler chickens at 1 d old were randomly divided into 6 groups, and each group consisted of 6 replicates with 18 broilers per replicate. The dietary treatments were as follows: negative control (NC, the basal diet), NC + antibiotic (enramycin, 8 mg/kg, positive control [PC]), NC + phosphoric acid (PA, 0.1, 0.2, and 0.3 g/kg), and NC + lactic acid (LA, 0.3 g/kg). The feeding trial lasted for 42 d. The results showed that the feed-to-gain ratio of the NC + acidifier groups was lower than that of the NC and PC groups from 1 to 42 d (P < 0.05). Compared with the values in the NC group, the pH of breast muscle was significantly higher in the NC + PA (0.2 g/kg) and LA (0.3 g/kg) groups (P < 0.05), and the cooking loss was lower in the breast muscle of the NC + PA (0.1 g/kg) and LA (0.3 g/kg) groups (P < 0.05). In addition, the shear force of the breast muscle and thigh muscle and the pH value in the crop, gizzard and duodenum of the antibiotic and acidifier groups were significantly decreased (P < 0.05). Moreover, the trypsin, chymotrypsin, and lipase activities of the duodenum in the NC + PA (0.2 and 0.3 g/kg) groups, as well as the villus height-to-crypt depth (VH:CD) ratio of the duodenum in the NC + PA (0.1 g/kg) group was significantly greater (P < 0.05) compared with those in the NC group. Meanwhile, the number of total aerobic bacteria, Escherichia coli and Salmonella in the cecum of the NC + PA (0.1 g/kg) and LA (0.3 g/kg) groups were decreased (P < 0.05). Collectively, diet supplementation with acidifiers could improve the growth performance, meat quality, and intestinal health of broilers, in which the effects of PA (0.1 g/kg and 0.2 g/kg) are better than the other supplementations.  相似文献

This study was conducted to evaluate the effect of pyridoxine on the development of hair follicles in Rex rabbits and the underlying molecular mechanism. Two hundred 3-month-old Rex rabbits were randomly divided into 5 groups and fed diets supplemented with 0, 5, 10, 20, or 40 mg/kg pyridoxine. The hair follicle density on the dorsal skin and the gene and protein expression levels of components of the phosphoinositide 3-kinase (PI3K)/protein kinase B (PKB or Akt), Wnt, Notch and bone morphogenetic protein (BMP) signalling pathways were measured. In addition, free hair follicles were isolated from Rex rabbits and cultured with pyridoxine in vitro to measure hair shaft growth. Furthermore, dermal papilla cells (DPC) were isolated from the skin of Rex rabbits and cultured with pyridoxine in vitro to measure the gene and protein expression levels of components of the PI3K/Akt, Wnt, Notch and BMP signalling pathways. The results showed that the addition of dietary pyridoxine significantly increased the total follicle density, secondary follicle density, and secondary-to-primary ratio (S/P, P < 0.05), that the growth ratio of hair stems was promoted by pyridoxine in basic culture medium, and that the growth length of tentacle hair follicles cultured in the pyridoxine group was longer than that in the control group (P < 0.05). In addition, pyridoxine changed the DPC cycle progression and promoted cell proliferation, and appropriate concentrations of pyridoxine (10 and 20 μmol/L) significantly inhibited cell apoptosis (P < 0.05). Pyridoxine significantly affected the gene expression of components of the PI3K/Akt, Wnt and Notch signalling pathways in the skin and DPC of Rex rabbits (P < 0.05), increased the levels of phosphorylated catenin beta 1 (CTNNB1) and Akt, and decreased the level of phosphorylated glycogen synthase kinase 3 beta (GSK-3β) (P < 0.05). Therefore, the molecular mechanism by which pyridoxine promotes hair follicle density in Rex rabbits probably occurs through activation of the PI3K/Akt, Wnt and Notch signalling pathways, prolonging hair follicle growth and delaying the onset of telogen.  相似文献

Seventeen crossbred lambs were assigned randomly to low-protein (LP; 8% crude protein [CP]; n = 9) and high-protein (HP; 13% CP; n = 8) diets for 9 weeks. The final body weight, average daily feed intake (ADFI), and average daily gain (ADG) of the HP lambs were significantly higher (P < 0.05) than the LP lambs; however, gain to feed ratio (G:F) for the LP lambs was significantly higher (P < 0.05) than the HP lambs. Hot carcass weight (HCW), adjusted fat thickness, and drip loss of longissimus dorsi (LD) muscle were significantly higher (P < 0.05) for the HP than LP lambs. In contrast, instrumental color values L*, a*, b*, C*, and hue angle (H) of meat from the LP lambs scored significantly higher (P < 0.05) than the HP lambs. The LD muscle from HP lambs had significantly greater CLA of cis-9 trans-11 isomer (P < 0.05) than the LP lambs. The gene expression of metabolism and meat quality-related genes of LP was significantly higher than HP (P < 0.05). These results suggest that a higher dietary CP level promotes growth performance for finishing lambs, whereas lower dietary CP level is beneficial for meat quality, especially when evaluating color characteristics in the final product.  相似文献

The aim of present study was to evaluate whether diets supplemented with dihydroartemisinin (DHA) could alleviate intestinal inflammatory injury in weaned piglets with intrauterine growth retardation (IUGR). Twelve normal birth weight (NBW) piglets and 12 piglets with IUGR were fed a basal diet (NBW-CON and IUCR-CON groups), and another 12 piglets with IUGR were fed the basal diet supplemented with DHA at 80 mg/kg (IUGR-DHA group) from 21 to 49 d of age. At 49 d of age, 8 piglets with similar body weight in each group were sacrificed. The jejunal and ileal samples were collected for further analysis. The results showed that IUGR impaired intestinal morphology, increased intestinal inflammatory response, raised enterocyte apoptosis and reduced enterocyte proliferation and activated transmembrane toll-like receptor 4 (TLR4)/nucleotide-binding and oligomerization domain (NOD)/nuclear factor-κB (NF-κB) signaling pathway. Dihydroartemisinin inclusion ameliorated intestinal morphology, indicated by increased villus height, villus height-to-crypt depth ratio, villus surface area and decreased villus width of piglets with IUGR (P < 0.05). Compared with NBW piglets, IUGR piglets supplemented with DHA exhibited higher apoptosis index and caspase-3 expression, and lower proliferation index and proliferating cell nuclear antigen expression in the intestine (P < 0.05). Dihydroartemisinin supplementation attenuated the intestinal inflammation of piglets with IUGR, indicated by increased concentrations of intestinal inflammatory cytokines and lipopolysaccharides (P < 0.05). In addition, DHA supplementation down-regulated the related mRNA expressions of TLR4/NOD/NF-κB signaling pathway and upregulated mRNA expressions of negative regulators of TLR4 and NOD signaling pathway in the intestine of piglets with IUGR (P < 0.05). Piglets in the IUGR-DHA group showed lower protein expressions of TLR4, phosphorylated NF-κB (pNF-κB) inhibitor α, nuclear pNF-κB, and higher protein expression of cytoplasmic pNF-κB in the intestine than those in the IUGR-CON group (P < 0.05). In conclusion, DHA supplementation could improve intestinal morphology, regulate enterocyte proliferation and apoptosis, and alleviate intestinal inflammation through TLR4/NOD/NF-κB signaling pathway in weaned piglets with IUGR.  相似文献

Different dietary nitrogen (N) patterns may have different effects on gut microbiota. To investigate the effects of different crude protein (CP) levels or essential amino acids (EAA) supplementation patterns on the structure and functions of colonic microbiota, 42 barrows (25 ± 0.39 kg) were randomly assigned to 7 dietary treatments including: diet 1, a high CP diet with balanced 10 EAA; diet 2, a medium CP diet with approximately 2% decreased CP level from diet 1 and balanced 10 EAA; diets 3, 4, 5, 6 and 7, low CP diets with 4% decreased CP level from diet 1. Specifically, diet 3 was only balanced for Lys, Met, Thr and Trp; diets 4, 5 and 6 were further supplemented with Ile, Val and Ile + Val on the basis of diet 3, respectively; and diet 7 was balanced for 10 EAA. Results over a 110-d trial showed that reducing the CP level by 2% or 4% dramatically decreased N intake and excretion (P < 0.05) in the presence of balanced 10 EAA, which was not observed when altering the EAA supplementation patterns in low CP diet (−4%). With balanced 10 EAA, 2% reduction in dietary CP significantly reduced Firmicutes-to-Bacteroidetes (F:B) ratio and significantly elevated the abundance of Prevotellaceae NK3B31 (P < 0.05); whereas 4% reduction evidently increased the abundances of Proteobacteria, Succinivibrio and Lachnospiraceae XPB1014 (P < 0.05). Among the 5 low CP diets (−4%), supplementation with Ile, or Val + Ile, or balanced 10 EAA increased F:B ratio and the abundance of Proteobacteria. In addition, the predicted functions revealed that different CP levels and EAA balanced patterns dramatically altered the mRNA expression profiles of N-metabolizing genes, the “N and energy metabolism” pathways or the metabolism of some small substances, such as amino acids (AA) and vitamins. Our findings suggested that reducing the dietary CP levels by 2% to 4% with balancing 10 EAA, or only further supplementation with Ile or Val + Ile to a low protein diet (−4%) reduced the N contents entering the hindgut to various degrees, altered the abundances of N-metabolizing bacteria, and improved the abilities of N utilization.  相似文献

This study was conducted to evaluate the effects of different stocking densities on growth performance, blood parameters, intestinal morphology and intestinal immunity of growing pigs. A total of 288 male pigs (44.35 ± 0.50 kg) were randomly assigned to groups with stocking densities of 2.46, 1.23 and 0.82 m2/pig for a month. The results showed that there was no significant difference on growth performance among groups. Pigs in the stocking density of 0.82 m2/pig had the lowest backfat thickness and spleen weight index among groups (P < 0.05). With increasing stocking density, the serum concentrations of blood urea nitrogen (BUN), transglutaminase (TGG), alkaline phosphatase (AKP) and immunoglobulin A (IgA) were increased, and albumin (ALB), albumin-to-globulin ratio (ALB:GLO), insulin-like growth factor-1 (IGF-1) and immunoglobulin (IgM) were decreased (P < 0.05), and cortisol tended to increase and glucose tended to decrease (0.05 < P < 0.1). Compared with the stocking density of 2.46 m2/pig, the ileal villus height and jejunal villus width decreased in stocking densities of 1.23 and 0.82 m2/pig (P < 0.05). The duodenal villus height and ileal villus width in the stocking density of 0.82 m2/pig were the lowest among 3 groups (P < 0.05). The content of immunoglobulin A in duodenum, jejunum and ileum mucosa increased along with increasing density (P < 0.05). The contents of interleukin (IL)-2 in the spleen or liver and IL-10 in the spleen were higher in the stocking density of 0.82 m2/pig than in other 2 groups. These results showed that stocking density could affect the metabolism, intestinal morphology, and immunity of growing pigs and 1.23 m2/pig may be the suitable stocking density for the growing pigs in the present study.  相似文献

This study was conducted to evaluate the effects of dietary live yeast (LY) supplementation during late gestation and lactation on reproductive performance, colostrum and milk composition, blood biochemical and immunological parameters of sows. A total of 40 multiparous sows were randomly fed either the control (CON) diet or the CON diet supplemented with LY at 1 g/kg from d 90 of gestation to weaning. Results showed that the number of stillborn piglets and low BW piglets were significantly decreased in the LY-supplemented sows compared with sows in the CON group (P < 0.05). Moreover, the concentrations of protein, lactose and solids-not-fat were increased in the colostrum of LY-supplemented sows (P < 0.05). Interestingly, the plasma activities of aspartate aminotransferase (AST), alkaline phosphatase (ALP), gamma-glutamyl transpeptidase (γ-GGT) at d 1 of lactation and alanine aminotransferase (ALT) at weaning day were decreased by feeding LY diet (P < 0.05). Meanwhile, sows fed LY diet had higher plasma concentration of immunoglobulin G compared with sows fed CON diet at d 1 of lactation (P < 0.05). In conclusion, LY supplementation in maternal diets decreased the number of stillborn piglets and low BW piglets, improved colostrum quality and health status of sows.  相似文献

This study compared the growth performance, nutrient utilization, and slaughter traits of Dorper crossbred male lambs fed as per the established nutrition recommendations for sheep, with an aim to verify the efficacy of different feeding standards. A total of 576 lambs (4 months of age, 28.3 ± 0.86 kg BW) were randomly allotted to 3 treatments with 12 replicates per treatment (16 lambs per replicate). The lambs were fed diets formulated according to the following 3 nutritional systems: the nutrient requirements of Dorper crossbred lambs established by Chinese Academy of Agricultural Sciences (CAAS), NRC (National Research Council), (2007), and AFRC (Agricultural and Food Research Council) (1993). The experiment lasted for 81 d. Feed intake was recorded every 3 days, and lambs were weighed every 20 days. Digestibility trials were conducted with 6 lambs each group from d 42 to 53 and d 70 to 81. At the end of the experiment, 10 lambs randomly chosen from each group were sacrificed to determine the carcass traits and meat quality. The results indicated that the lambs in the NRC group had the highest dry matter intake (DMI), followed by those in the AFRC and CAAS groups (P < 0.05). The average daily gain, carcass weight, and dressing percentage were higher for lambs in the CAAS group than those in the NRC group (P < 0.05). The lambs in the CAAS group had the lowest feed conversion ratio, followed by those in the AFRC and NRC groups (P < 0.05). The apparent digestibility of DM was higher for the lambs in the CAAS group than those in the NRC group (P < 0.05). Water losing rate, as well as the lightness (L*), redness (a*), and yellowness (b*) values of the longissimus thoracis were not different among groups (P > 0.05). In conclusion, Dorper crossbred lambs fed diets formulated according to the CAAS recommendations exhibited superior growth performance than those fed diets formulated according to the American or British feeding standards.  相似文献
