Summary

In dairy cows, overfeeding during the dry period leads to overcondition at calving and to depression of appetite after calving. As a consequence, at calving overconditioned high‐producing dairy cows inevitably go into a more severe negative energy balance (NEB) postpartum than cows that have a normal appetite. During the period of NEB, the energy requirements of the cow are satisfied by lipolysis and proteolysis. Lipolysis results in an increased concentration of non esterified fatty acids (NEFA) in the blood. In the liver, these NEFA are predominantly esterified to triacylglycerols (TAG) that are secreted in very low density lipoproteins (VLDL). In early lactation in cows with a severe NEB, the capacity of the liver to maintain the export of the TAG in the form of VLDL in balance with the hepatic TAG production is not always adequate. As a result, the excess amount of TAG accumulates in the liver, leading to fatty infiltration of the liver (hepatic lipidosis or fatty liver). The NEB and/or fatty liver postpartum are frequently associated with postparturient problems. In general, a severe NEB induces changes in biochemical, endocrinological, and metabolic pathways that are responsible for production, maintenance of health, and reproduction of the postparturient dairy cow. These changes include a decrease in blood glucose and insulin concentrations, and an increase in blood NEFA concentrations. High NEFA concentrations caused by intensive lipolysis are accompanied by impairment of the immune system, making the cows more vulnerable to infections. Metabolic diseases such as ketosis, milk fever, and displaced abomasum are related to overcondition at calving. The changes in biochemical, endocrinological, and metabolic pathways are associated with delay of the first visible signs of oestrus, an increase in the interval from calving to first ovulation, a decrease in conception rate, and a prolonged calving interval. It is possible that the increased blood NEFA concentration directly impairs ovarian function.  相似文献

The previous experiments have shown that some phenothiazines have antioxidant and anti-inflammatory properties in vitro . In this study the inhibition of the production of reactive oxygen species (ROS) by neutrophils was studied in two groups of horses, which received a dose of 0.1 mg/kg of either acepromazine or promethazine intravenously. Blood samples were collected before (T0) and 0.5, 1, 3 and 5 h after drug administration. The chemiluminescence (CML) response of neutrophils was measured ex vivo in the presence of luminol for a period of 10 min and the maximum CML value (peak value) recorded. There was a significant inhibition of the ROS production in the acepromazine treated group (49% inhibition) at 5 h after administration and in the promethazine group (24% inhibition) at 3 h after administration ( P  < 0.05 vs. T0). These findings are of therapeutic relevance in the use of phenothiazines in equine patients with inflammatory diseases where neutrophil activation and ROS production are implicated.  相似文献

The aim of this study was to evaluate the effect of postpartum mastitis between first calving and subsequent conception on production and reproduction performance as well as culling of Holstein cows. A data set of 9,183 first lactation cows was used. Results showed that the first cumulative 100?days' milk production and the milk yield standardized to 305?days were affected by the interval from calving to first mastitis (P??0.05). Dairy cows are highly susceptible to infectious diseases, like mastitis, during the period around calving. Although factors contributing to increased susceptibility to infection have not been fully elucidated, impaired neutrophil recruitment to the site of infection and changes in the concentrations of some micronutrients related with the function of the immune defence has been implicated. Most of the current information is based on studies outside the Nordic countries where the conditions for dairy cows are different. Therefore, the aim of the study was to evaluate changes in blood concentrations of the vitamins A and E, the minerals calcium (Ca), phosphorous (P), and magnesium (Mg), the electrolytes potassium (K) and sodium (Na) and the trace elements selenium (Se), copper (Cu) and zinc (Zn), as well as changes in total and differential white blood cell counts (WBC) and expression of the adhesion molecules CD62L and CD18 on blood neutrophils in Swedish dairy cows during the period around calving. Blood samples were taken from 10 cows one month before expected calving, at calving and one month after calving. The results were mainly in line with reports from other countries. The concentrations of vitamins A and E, and of Zn, Ca and P decreased significantly at calving, while Se, Cu, and Na increased. Leukocytosis was detected at calving, mainly explained by neutrophilia, but also by monocytosis. The numbers of lymphocytes tended to decrease at the same time. The mean fluorescent intensity (MFI) of CD62L and CD18 molecules on blood neutrophils remained constant over time. The proportion of CD62L+ neutrophils decreased significantly at calving. The animals were fed according to, or above, their requirements. Therefore, changes in blood levels of vitamins, minerals and trace elements were mainly in response to colostrum formation, changes in dry matter intake, and ruminal metabolism around calving. Decreased levels of vitamins A and E, and of Zn at calving might have negative implications for the functions of the immune defence. The lower proportion of CD62L+ neutrophils at calving may result in less migration of blood neutrophils into the tissues, and might contribute to the increased susceptibility to infections at this time.  相似文献

Gastrointestinal nematodes rarely cause signs of clinical disease in adult cattle. However, they have been shown to exert a negative impact on production in lactating animals, as seen by improved production following elimination of the worms using anthelmintics. A double blind, randomized clinical trial was performed in 28 dairy herds in Canada. The objective of the study was to evaluate the effect of treatment with eprinomectin pour-on solution (IVOMEC EPRINEX) at calving on production, in cattle that have had some exposure to pasture. Cows were randomly allocated to treatment or placebo in blocks of 10, based on calving date, and treated with eprinomectin or placebo on the day of calving.Information on milk production was obtained from all animals, as well as recorded cases of selected diseases. Milk production results from the Canadian dairy herd management system database were analysed using a mixed model with herd as a random effect and test within-cow as a repeated measurement. Test day milk yields from the first six tests after treatment were included in the model, representing a period of between 180 and 200 days in milk (dim). Treated cows produced an additional 0.94 kg of milk per day when compared to the controls over this period. The production effect was independent of calving season, age of the animal and geographical location. No effect of treatment was seen on milk composition, somatic cell count (scc) or on the selected health parameters that were recorded for all included animals.Monthly fecal egg counts (FEC) were performed for eight randomly selected animals in each herd. The observed FEC were low in this study, with a range from 0 to 419 trichostrongyle type eggs per 5g (ep5g) of feces in animals not yet treated with the anthelmintic. The average count was 9.8 and the median was 1.0. FECs dropped immediately after calving and stayed lower for at least 100 days in treated animals when compared to controls.In conclusion, gastrointestinal nematodes appear to have an effect on milk production in Canadian dairy cows that have had some degree of pasture exposure. Eliminating the present subclinical parasite burdens produced a consistent increase in milk production that can yield economic benefits for the dairy producer.  相似文献

The optimum policy of insemination and replacement of dairy cows was determined by the dynamic programming technique. The model used in the previous study was extended to allow variation in time of conception. From 2 to 7 months after calving three alternatives were considered for an open cow namely (a) inseminating the cow, with a calculated probability of success, (b) leaving her open, and (c) replacing her immediately. When it was profitable to leave a cow open, the optimum time for replacement during the lactation period was determined.The minimum production level for insemination to be the optimum choice depended on the stage of lactation and the parity of the cow. In the optimum situation the average calving interval was 371 days, while 13% of the cows had an interval of 14 months or longer.The optimum policy was greatly affected by changes in the replacement heifer price. Changes in the probability of conception and persistency of milk production had significant but smaller effects. In herds with a smaller decline in production after the peak, insemination should be continued for longer than in herds with a larger decline.The relation between production and calving interval that resulted from the optimum policy was determined. When a measure of milk production was used that was not affected by gestation, the average correlation was 0.09. The correlation increased to 0.35 when the total 305-day production was used.  相似文献

The objective of the present study was to quantify the effect of strain of Holstein–Friesian and pasture-based feeding system on calving performance, blood parameters and overall survival within a seasonal calving milk production system. The three strains of Holstein–Friesian compared were high production North American (HP), selected entirely for milk production, high durability North American (HD), selected based on milk production, fertility and muscularity traits, and New Zealand (NZ) selected from a seasonal calving pasture-based system. The three feed systems compared were a high grass allowance feed system typical of spring calving herds in Ireland (MP; control); an increased stocking rate system (HS) and an increased concentrate supplementation system (HC). Data comprised up to 585 lactations on 240 cows. Data were analysed using linear mixed models, generalised estimating equations and survival analysis. Strain of Holstein–Friesian had no significant effect on calving difficulty or rate of stillbirths while the effect of feed system on calving difficulty and rate of stillbirths approached significance. Strain of Holstein–Friesian had no significant effect on plasma metabolite or hormone concentrations around parturition, with the exception of urea concentration. At the beginning of the breeding season the NZ strain had lower plasma concentrations of NEFA (0.20 mmol/l) and higher IGF-I (86.8 ng/ml) than the HD (0.24 mmol/l and 66.9 ng/ml, respectively) and HP (0.26 mmol/l and 64.0 ng/ml, respectively) strains. The higher IGF-I concentration of the NZ strain was maintained during the breeding season. The NZ and HD strains survived longer than the HP strain with median survival days post-first calving of 1372, 1367 and 1033 respectively, while feed system had no significant effect on overall survival rate over the study period.  相似文献

The development of the carrier state in swine after infection with Salmonella serovar Typhimurium (S. Typhimurium) has not been elucidated yet. Possibly, phagocytes like macrophages play a crucial role. It was the aim of the present study to characterize the interaction of a S. Typhimurium strain and its hilA and ssrA mutants with porcine peripheral blood monocytes (PBM). Exposure of porcine PBM to S. Typhimurium induced the production of reactive oxygen species (ROS), requiring bacterial protein synthesis. The numbers of intracellular bacteria sharply decreased over a period of 3h. Monocytes obtained from different pigs differed markedly in their ROS production and in their ability to kill the bacteria. Interestingly, high ROS production did not coincide with increased intracellular killing. Using diphenylene iodonium inhibition of nicotinamide adenine dinucleotide phosphate (NADPH) oxidase activity, it was shown that bacterial killing was ROS-dependent only within 1h post inoculation, but was ROS-independent from 1h post inoculation onwards. This might be explained by the finding that metabolically active Salmonella bacteria were capable of suppressing the respiratory burst activity in a SPI-1- and SPI-2-independent manner without causing measurable cell damage. Opsonization with complement did not alter the ROS production. Nevertheless, it increased intracellular survival of the bacteria. In conclusion, survival of S. Typhimurium inside porcine PBM is promoted by suppression of respiratory burst activity and complement binding.  相似文献

Neutrophils, eosinophils and macrophages interact with invading parasites and naive hosts. The initial reaction of leukocytes is the generation of reactive oxygen species (ROS). The cytotoxic effects of extracts derived from intact Cysticercus cellulosae and from the scolex or membrane fractions on neutrophils were examined. DNA fragmentation of neutrophils was observed when cells were incubated with an extract from the intact metacestode; however, the addition of antioxidant enzymes to the incubation medium had a protective effect. The scolex and membrane extracts did not affect DNA fragmentation of neutrophils. Hydrogen peroxide production of neutrophils incubated with metacestode fractions from C. cellulosae increased by 190% (total extract), 120% (scolex) or 44% (membrane). An increase in antioxidant catalase activity (28%) concomitant with the increased production of ROS was observed in neutrophils incubated with metacestode fractions, which could be an attempt at self-protection. ROS production by neutrophils in the presence of the intact cysticerci extract did not alter phagocytosis. In contrast, the scolex and membrane fractions increased the phagocytic capacity of neutrophils by 44 and 28%, respectively. The results showed that the extract from intact C. cellulosae was toxic for neutrophils via ROS production, leading to DNA fragmentation and inhibition of phagocytic capacity, but neutrophils are able to protect themselves against oxidative stress by via catalase activity.  相似文献

Genetic and environmental correlations were estimated both between the ability to show oestrus and milk production, and among different fertility traits (heat-intensity score, number of days between consecutive inseminations, number of inseminations per service period, interval between calving and first or last insemination, and interval between first and last insemination). Milk production was measured as the average of the energy-corrected milk yield on second and third monthly test days. The number of records were approximately 450000, 350000, 180000 and 75000 in the heifer period, first, second and third lactations, respectively. A linear, trivariate model that included the effects of herd-year, year, month, age and cow's sire was applied. The results indicated that further selection for increased milk production is not expected to deteriorate heat intensity. The number of days between calving and first insemination, the number of inseminations and the heat intensity were complementary, and can be recommended for a selection index for fertility.  相似文献

The periparturient or transition period of 4 weeks before and 4 weeks after calving is characterised by a greatly increased risk of disease. Hypocalcaemia around calving is a risk factor for many of these diseases and is an indirect risk factor for increased culling. The incidence of clinical hypocalcaemia (milk fever) in the field generally ranges from 0-10%, but may exceed 25% of cows calving. In research trials conducted on milk fever the incidence has approached 80% of cows calving. Homeostasis of calcium (Ca) is regulated by calcitonin, parathyroid hormone and 1,25(OH)(2) vitamin D(3). Age increases the risk of milk fever by approximately 9% per lactation. Control of milk fever has revolved around stimulation of homeostatic mechanisms through feeding a pre-calving diet low in Ca. More recently, the role of the dietary cation anion difference (DCAD) in the prevention of Ca disorders has been examined, both by field research and meta-analysis. The most appropriate form of the DCAD equation has been contentious, but recent meta-analyses have shown that the equation (Na(+)+K(+))-(Cl(-)+S(2-)) is most effective for predicting milk fever risk. Decreased risk of milk fever is linear with DCAD, whereas the effect of DCAD on urinary pH is curvilinear. A pivotal role of providing dietary magnesium (Mg) before calving has been confirmed by meta-analysis, and a quadratic effect of Ca on milk fever risk was found with a peak occurring with dietary levels of 1.1-1.3% of dry matter. Risks of milk fever increase with increased dietary phosphorus (P) fed pre-calving and with increasing days of exposure to a pre-calving diet. Meta-analysis has revealed that the important roles of dietary Ca, Mg and P, as well as the duration of exposure to the pre-calving diet in milk fever control strategies are independent of DCAD. Studies on the effect of exposure to well designed pre-calving diets have shown that substantial improvements in production, reproduction and animal health can be made but further examination of the influence of the period of exposure to different diets is warranted.  相似文献

Relative date of first calving of beef heifers was studied in relation to production efficiency and subsequent reproductive performance. Crossbred heifers were managed in a drylot environment for 1 yr with feed intake measured through weaning of the first calf. Mean heifer age at entry into drylot was 572 d. Production traits were evaluated by calving group (CG), where CG1 included records of heifers calving (and calves born) in the first 21 d of the calving season for a particular year, CG2 included those calving from 22 through 42 d and CG3 included those calving after 42 d. Calving groups did not differ significantly for preweaning calf ADG, but weaning age differences resulted in heavier weaning weights for CG1 compared with CG2 and CG3. An earlier relative calving date was associated with increased cumulative feed energy intake of heifers and their calves during the 1-yr test period. In terms of production efficiency, the weaning weight advantage of earlier calving was offset only partly by the increased feed energy intake of the dam-calf unit, resulting in 6.3 Mcal (12.4%) less ME per kg calf weaning weight for CG1 vs CG3 for the 1-yr period. Within a limited calving season, earlier calving dams tended to be biologically and economically more efficient because a greater proportion of their annual production cycle was in a productive (lactating) mode, diluting maintenance costs as a fraction of all costs. Heifers in CG1 also tended to calve earlier than CG3 heifers for the second calf. Calving interval was a biased measure under the management conditions of a limited breeding season and culling of open cows.  相似文献

The aim of this study was to evaluate the behaviour of adenosine deaminase (ADA) activity, as well as its participation in the immunomodulation of pregnant cows. Thus, sixteen cows were divided into two groups (A and B): the group A was composed by cows not pregnant (= 8), while the group B was composed by pregnant cows (= 8). Serum levels of interleukin‐10 (IL‐10), IL‐6, tumour necrosis factor alpha (TNF‐α), reactive oxygen species (ROS) and C‐reactive protein (CRP), as well as ADA and glutathione S‐transferase (GST) activities, were measured on five sampling times (3, 5, 7 and 8 months of gestation, and soon after calving). Serum ADA activity was similar throughout the experiment in the cows belonging to the group A, but its activity increased during the experiment in cows from the group B, that is it was lower in the third and fifth months of pregnancy, and higher on months 7, 8 and after calving when compared to the group A. TNF‐α and IL‐6 serum levels were lower in pregnant cows compared to non‐pregnant animals; however, they significantly increased after calving. Serum levels of IL‐10 increased after 8 months of gestation, but it reduced after calving when compared to the group A, while CRP increased on month 8 of gestation and after calving compared to the group A. Pregnant cows showed lower serum ROS levels on months 3, 5 and 7 of gestation, and higher levels at the post‐partum. Serum GST activity was higher on month 5 of gestation in pregnant cows, but it was lower on months 7, 8 and in the post‐partum compared to the group A. Based on these evidence, we concluded that ADA activity and the others mediators or inflammatory modulators have important role in the maintenance of cow′s gestation due to their immunomodulatory effects.  相似文献

Alpha-1-acid glycoprotein (AGP) is an acute-phase protein with anti-inflammatory and immunomodulating properties. AGP is described as a potent inhibitor of the production of reactive oxygen species (ROS) in human neutrophils. However, published reports about the mechanism of inhibition are conflicting. The influence of bovine AGP on the production of ROS by bovine peripheral blood polymorphonuclear leucocytes (PMN) was studied using a highly sensitive method approaching its inhibitory mechanism. ROS production in PMN was induced with phorbol 12-myristate 13-acetate (PMA) or opsonized Staphylococcus aureus bacteria. ROS generation was quantified and evaluated by flow cytometry. AGP efficiently suppressed PMA, but did not opsonize bacteria-induced ROS generation in vitro. The suppressive effect was concentration-dependent and adversely proportional to PMA concentration. The selective inhibitory potential of AGP in comparison with ovalbumin (OVA) and bovine serum albumin (BSA) showed that ROS inhibition was not a mere protein effect. ROS production was suppressed only if AGP and PMA were simultaneously present with PMN. Pre-incubation of PMN with AGP did not alter the PMN response to PMA. Moreover, AGP could not suppress ROS production after pre-stimulation of PMN with PMA. Human and bovine AGP did not differ in their inhibitory potential to the PMA-induced ROS production in bovine, human and equine PMN. The results show that AGP does not modulate bovine neutrophil functions directly, but acts as a scavenger of PMA.  相似文献

The goal of this study was to compare production variables and veterinary costs between dairy herds enrolled in an integrated herd health program and herds with a conventional, non-computerized herd management. Four variables were used to assess the performance of the herds, including calving interval, milk production per lactation, as well as the product of calving interval x veterinary costs per year and the ratio of production to veterinary costs per year. A total of 22 dairy herds, serviced by the ambulatory clinic, University of Zurich, were investigated. There were 11 experimental herds that had been enrolled in an integrated herd health program, INTERHERD?, and 11 control herds. Data of the latter were derived from a computerized accounting system, OBLON DATA?. A total of 92'350 records from the years 1999 - 2005 were analyzed retrospectively. During the investigation period the calving interval did not significantly increase in experimental herds, whereas milk production steadily increased in both groups. The integrated herd health program did not result in additional costs when the dairy farms have no problems on a herd basis. From our study, differences in farms with and without herd health program are only marginal.  相似文献

In the cow the foetal endocrine signals that initiate the calving process result in prepartum luteolysis. Withdrawal of progesterone (P4) action is a prerequisite for a normal calving. The rather abrupt declining influence of P4 is followed by a cascade of physiological processes in the myometrium and cervix. This contribution will focus on some of these events. Like in many other species, the myometrium in cows is not completely inactivated during pregnancy. So-called contractures have been registered during the final weeks of gestation and their EMG-characteristics in cows show a low frequency (on average: 13.6 per day) and long duration (on average 12.1 min). They are not evenly spread over the day because they occur less frequently when the cows are disturbed for feeding or cleaning their stables. Contractures affect several foetal functions. In the cow these contractures disappear during a period of about 8-9h when maternal plasma P4 levels are rapidly declining before calving. There is experimental evidence that this temporary inhibition is associated with prepartal luteal regression. The cause of this inhibition is still unknown. Because nitrous oxide inhibits smooth muscle cells and evidence in laboratory animals indicates that expression of the inducible form of nitrous oxide (iNOS) is downregulated in myometrium, but upregulated in the cervix around the onset of parturition, we started to investigate the role of this enzyme in bovine tissues around calving. By means of a RT-PCR technique, we obtained a first indication that iNOS is hardly expressed in the myometrium during calving, while expression was clearly detected at day 4 after calving. Analysis of prepartum en periparturient biopsies from myometrium and cervix with quantitative PCR is still underway. In six pregnant cows, provided with uterine EMG-electrodes and with ultrasonic crystals implanted on the caudal cervical rim to measure cervical dilatation, calving was induced with an injection of prostaglandin (PG) F2alpha. While maternal plasma P4 levels had significantly declined within 8h after PG treatment, the myometrium escaped from temporary inhibition with the development of a parturient contractility pattern on average at 13.5h after injection. However, it was only at 28 h after PG treatment that the first sustained increase of the opening of the vaginal ostium of the cervix was measured.  相似文献

The study was undertaken to determine the trends in the reproductive performance of Holstein dairy cows in Iran during 1994 to 2008. Reproductive performance data for 528,034 lactations of 246,132 cows in 1,822 Holstein dairy herds of Iran were used. The potential effect of calving season, herd, parity, calving year, as well as herd size and 305-day milk production on reproductive performance traits was investigated using multiple regression models. The least squares means of age at first calving decreased by 3.1 (±0.06) days per year from 806.5 (±96.3) days in 1994 to 788 (±89.9) days in 2008. The least squares means of calving interval increased 1.02 (±0.03) days per year from 394.1 (±65) days in 1994 to 413.2 (±81) days in 2008. Greater 305-day milk production was associated with an average increase of 6.55 (±0.08) days in calving interval per 1,000-kg increase in milk yield. Larger herd size was associated with an average decrease of 0.22 (±0.02) days in calving interval per 50 cows per herd. The mean number of days dry was 88.6 (±51.3) days and increased by 0.82 (±0.02) days per year. In conclusion, reproductive performance in Holstein dairy herds has generally decreased, whereas herd size and milk production have increased over time. Producers may make significant improvements in herd reproduction by reviewing management strategies including the sire selection, reproductive management, inseminator training and techniques, and improved estrous detection. Moreover, it may be advisable for the fertility traits to be included in the genetic selection indices to reduce the rate of reproductive decline.  相似文献

The objectives of this study were to provide baseline data for alpha-tocopherol, selenium and polyunsaturated fatty acid concentrations in the serum and feed of New Zealand dairy cattle, and to assess the likelihood that abnormal peroxide metabolism has a role in the impaired lactational and reproductive performance noted in selenium-deficient cattle. Twenty-four Friesian heifers were randomly allocated one of four winter diets consisting of hay with or without selenium supplementation, or pasture and silage with or without selenium supplementation. A winter diet consisting exclusively of hay (alpha-tocopherol concentration 19 mg/kg of dry matter) resulted in a pre-calving serum alpha-tocopherol concentration of 1.2 mg/l compared to 4.5 mg/l for pastured heifers (p<0.01). The pre-calving alpha-tocopherol concentration for the heifers fed hay fell into the range considered deficient (<2.0 mg/l), whereas heifers fed pasture and silage remained in the range considered adequate throughout the study period. Serum fatty acid concentration, and the proportion of fatty acids that were polyunsaturated, were lowest in the hay-fed heifers before calving (1.0 mg/ml, 37.1% respectively), and remained unchanged following re-introduction to pasture after calving in late July and August. Serum fatty acid concentration did not increase following the re-introduction of the heifers to pasture because of the unexpectedly low fatty acid concentration (4.8 g/kg of dry matter) of the mature winter pasture. In October, however, the proportion of fatty acids in serum that were polyunsaturated increased (50%) as did serum cy-tocopherol concentrations (greater than 13 mg/l). Mean serum selenium concentrations in the unsupplemented heifers ranged from 139 to 204 nmol/l, being lowest in October (p<0.01). Supplementation with intraruminal selenium pellets (two pellets delivering about 3 mg of selenium/day) increased serum selenium concentration and glutathione peroxidase activity (p<0.01) whereas the type of winter diet had no effect (p>0.05). These results suggest that dairy cattle wintered on hay can become Vitamin E-depleted, whereas the feeding of pasture and silage should provide adequate Vitamin E. The pasture offered following calving during July and August provided a low dietary polyunsaturated fatty acid challenge, suggesting that abnormal peroxide metabolism is unlikely to be an important mechanism in the impaired performance of selenium-deficient adult cattle which calve at this the of year.  相似文献

This study was designed to evaluate if the immunosuppression typically observed during the immediate periparturient period (3 weeks before and after calving) in dairy cows influences the effectiveness of diagnostic tests for the detection of Johne's disease; and, if providing additional energy to the cows during this period would minimize any immunosuppressive effects. Twelve dairy cows naturally infected with Mycobacterium paratuberculosis were fitted with rumen cannulas in late gestation and assigned to treatment groups: control, n = 6; or stuffed, n = 6. Cows in the control group were allowed to consume feed ad libitum. Cows assigned to the stuffed treatment group were also fed ad libitum but received additional total mixed ration by manually stuffing their rumens with refused feed to maintain a dry matter intake of 2% body weight/day before calving and 2.5% body weight/day after calving. Parturition had a significant impact on immune function with significant reductions in M. paratuberculosis-specific antibodies detected in the serum and milk regardless of treatment group. Similarly, in vitro immunoglobulin production was decreased at calving for both treatment groups. In addition, stuffing cows modulated cell-mediated immune function by reducing antigen-specific lymphocyte proliferation and interferon-gamma production after calving. Shedding of M. paratuberculosis in the milk was apparent in 58% (7/12) of cows after parturition with no difference noted between control and stuffed animals. Parturition had no major effect on fecal shedding of cows regardless of treatment. These data suggest that parturition had a significant effect on immune function parameters including diagnostic tests for paratuberculosis. Furthermore, providing additional energy to cows with Johne's disease did not preclude immunosuppressive effects during the periparturient period.  相似文献
