Hepatic monoxygenases metabolize steroids that in turn influence reproduction. Experiments were conducted to establish the effect of dose of phenobarbital and level of dietary protein on hepatic monoxygenase. In Exp. 1, ewes were given either 0, .5, 1, 2 or 3 g phenobarbital (PB) daily for 4 to 8 d. After 48 h after last dose of PB, cytochrome P-450 was higher (P less than .01) in all ewes given PB than in controls and was higher (P less than .01) in ewes given PB for 8 d than in ewes treated for 4 d. In Exp. 2, either 0 or 1 g of PB was given orally for 8 d. Liver samples were collected 1, 4, 7 or 10 d after last treatment. Both cytochrome P-450 (P less than .01) and nicotinamide adenine dinucleotide phosphate (NADPH) cytochrome c reductase (P less than .06) were higher in ewes receiving PB than in controls. Cytochrome P-450 was twice as high in treated ewes as in control ewes on the 8th d of treatment, but concentrations returned to control levels 10 d after last treatment. Microsomal protein and cytochrome b5 were not affected by PB (P greater than .10). Ewes in Exp. 3 receiving a diet containing 14.8% crude protein for 10 d had higher levels of cytochrome P-450 (P less than .01) than ewes fed 4.7% crude protein. Protein did not affect microsomal protein or NADPH cytochrome c reductase. These data suggest that a relationship among PB, dietary protein and monoxygenases exists and provide information that will lead to a better understanding of the relationship between diet and reproduction.  相似文献

The administration of xenobiotics such as phenobarbital (PB) and chlorinated hydrocarbons to rats, mice and several other species has been shown to increase the level of hepatic mixed function oxidases. Experiments were conducted to establish the effect of dose of PB, sex of animals, and effect of interval from dose to measurement on concentration of hepatic enzymes in barrows and gilts 6 to 9 mo of age and weighing 100 to 120 kg. Animals given 1 or 2 g PB for 3 d had higher concentrations of microsomal protein cytochrome b5 and P-450 and NADPH cytochrome c reductase than control animals (P less than .01). Animals given 2 g PB had 3.5 times as much cytochrome P 450 as did controls. Barrows and gilts did not differ from each other in any variables measured (P less than .20). In a study of the time course of induction all animals given PB had higher levels of microsomal protein, cytochrome P-450 and reductase in 24 h than controls; however, cytochrome b5 was depressed on d 1 and was elevated by d 3. Concentration of cytochrome P-450 reached a maximum by d 7 (P less than .01); cytochromes b5 and c reductase reached a maximum by d 9 and 3, respectively (P less than .01). Levels of cytochrome P-450 were higher in gilts than in barrows on all days following PB treatment (P less than .04). Twenty fine-wool wether lambs were allotted randomly at weaning to be fed a conventional diet (CD) formulated for growing lambs, or the same basal mixture plus dried gamma-irradiated solids from primary sewage sludge (SS) incorporated as 7% of the dry matter. Feedlot performance and chemical elements in whole blood were monitored during 90 d, after which lambs were slaughtered and carcass data were collected. Concentrations of Ag, Ca, Cd, Co, Cr, Cu, Fe, K, Mg, Mn, Na, Ni, P, Pb and Zn were measured in livers, kidneys, bones, spleens and muscles. Sewage solids in the diet did not affect performance appreciably, although dressing percentages from lambs fed SS were 4% lower than CD-fed animals (P less than .05). Rib eye areas, fat thicknesses, quality and yield grades were similar (P greater than .05). Element concentrations in whole blood at weaning, after 56 d of the feeding trial and at slaughter did not differ (P greater than .05) between dietary groups. Serum chemistry determinations showed no biologically meaningful patterns related to diets. Lambs fed SS had higher (P less than .05) concentrations of Cu in livers (51.1 vs 34.3 +/- 3.8 micrograms/g, mean +/- SE) and Pb in kidneys (4.0 vs 2.2 +/- .3 micrograms/g) and lower (P less than .05) Mg in kidneys (.69 vs .72 +/- .01 mg/g). None of the elements in spleen and muscle tissue differed (P greater than .05) between diet groups. Microsomal and cytosolic enzymes that metabolize xenobiotics were measured in composite samples representing entire livers and in samples from three lobes, using livers of cattle, goats and sheep. Within individual species, concentrations of cytochrome P-450 and b5 and activities of NADPH cytochrome c reductase, aldrin epoxidase, aminopyrine N-demethylase, ethoxycoumarin O-deethylase, microsomal and cytosolic stilbene oxide (epoxide) hydrolase and glutathione S-transferase were not different (P greater than .05) among the various hepatic lobes. Among species, several activities differed (P less than .05), with cattle livers generally having lower values than sheep or goats.  相似文献

Physiological effects of feeding high levels of magnesium to sheep   总被引:1,自引:0,他引:1  
The effects of feeding high levels of Mg to sheep on animal health, nutrient digestibility, metabolism of macro-minerals, blood and tissue mineral concentrations and hematological and histological tissue changes were evaluated. Six lambs were allotted to each of four diets supplemented with MgO to attain .2, .6, 1.2 or 2.4% Mg. Total fecal and urinary collections were made for the initial 20 d and last 10 d of a 50 d study. Diarrhea was noted in lambs fed the two highest Mg levels. Apparent digestibility of DM, ADF and CP decreased linearly (P less than .01) with increasing Mg levels. Dietary Mg levels did not affect (P greater than .05) rectal temperature, respiration rate or pulse rate. Apparent absorption and retention of Mg (g/d) were lowest in lambs fed 2.4% Mg until 15 d on trial, after which values increased linearly (P less than .01) with dietary Mg. Phosphorus absorption and retention decreased linearly (P less than .01) with increasing Mg levels. Increasing dietary Mg resulted in a linear increase (P less than .01) in serum Mg and variable increases (P less than .05) in erythrocyte Mg. Serum Ca was decreased linearly (P less than .05) and quadratically (P less than .05) by added Mg during the d 1 to 30 and 40 to 50 periods, respectively. Serum inorganic P was elevated consistently in lambs fed 2.4% Mg. A linear response to increasing dietary Mg (P less than .05) occurred in the Mg content of liver, kidney, rib bone and femur bone. Sixteen crossbred beef heifers were used to determine the efficacy of serum clinical profiles as diagnostic tools for detection of early stages of locoweed toxicity and to test the ability of two mineral supplements for prevention or therapy of toxicosis. Dietary treatments were (DM basis) 1) 100% sorghum sudangrass hay, 2) 80% sorghum sudangrass hay:20% locoweed, 3) 80% sorghum sudangrass hay:20% locoweed plus 100 g of Silent Herder Mineral Mix (a mineral supplement reported to alleviate locoweed toxicity), and 4) 80% sorghum sudangrass hay:20% locoweed plus 100 g of clinoptilolite (a natural zeolite clay). Diets were fed at 1.5% of BW for 28 d, after which heifers had ad libitum access to sorghum sudangrass hay for 14 d. Jugular blood was sampled before feeding every 7 d, and at 2, 4, 6, and 8 h after feeding on d 28. Compared with controls, heifers fed locoweed had elevated (P less than .01) serum alkaline phosphatase activities from d 7 through 35. On d 42, alkaline phosphatase activities in heifers previously fed locoweed were lower (P less than .05) than in control heifers. Serum glutamic oxaloacetic transaminase activities were elevated (P less than .01) in heifers fed locoweed from d 7 through 42 compared with control heifers. Kochia hay that had elicited mild toxicosis in sheep and cattle when fed for 21 d was fed to young, growing steers to evaluate the efficacy of metoclopramide for preventing kochia toxicosis. Twelve steers (271 +/- 39 kg) were given ad libitum access to Kochia hay for 38 d. Six steers were dosed orally with 15 mg of metoclopramide/kg of BW three times each week in gelatin capsules, and six steers received empty gelatin capsules. Steers were housed in individual outside pens for the first 28 d, then inside in metabolism stalls for the last 10 d. Kochia intake averaged 1.2 +/- .04 and 1.1 +/- .05% of BW throughout 38 d for kochia-fed control and kochia plus metoclopramide-dosed steers, respectively. During the last 10 d, metoclopramide had no effect (P greater than .15) on digestibility of DM, OM, NDF, ADF, or CP. However, metoclopramide reduced N retention (P less than .01; 20 vs 8 g/d). Kochia hay decreased serum prolactin and insulin concentrations (P less than .01) from 12.4 to 1.5 ng/ml and from .53 to .23 ng/ml, respectively. Metoclopramide had no effect (P greater than .50) on prolactin or insulin in steers fed kochia hay. Serum growth hormone was not affected by kochia but was suppressed by metoclopramide in steers fed kochia hay (P less than .07). Serum bilirubin (total and unconjugated) was elevated (P less than .05), indicating early, mild hepatotoxicosis characteristic of kochia toxicosis. Two split-plot designed experiments were conducted to determine the effects of breed (Angus, A, or Brangus, B) and diet (fescue hay, FH; corn silage, CS; or concentrate) on composition and rate of growth, diet digestibility and plasma hormones of steers. In Exp. 1, 10 steers (five of each breed) were fed a CS-based diet followed by a FH-based diet for two consecutive 60-d periods. Both breeds had lower (P less than .01) DM intake and digestibility when fed FH than when fed CS diets. The B steers had higher (P less than .01) plasma insulin concentrations than A steers when fed the CS diet. In Exp. 2, during two consecutive years, 10 steers previously fed CS- and FH-based diets were finished with a corn silage-whole shelled corn-based diet. During yr 1, A steers had higher (P less than .01) DM intake and plasma triiodothyronine (T3) and thyroxine (T4) concentrations (P less than .05) than B steers did. Although final weights were similar (P greater than .10), A steers had heavier (P less than .05) carcass weights than B steers did. During yr 2, A steers had higher (P less than .07) DM and starch digestibilities and higher (P less than .01) plasma T4 concentrations than B steers did. The greater (P less than .01) energetic efficiency of A steers was attributed to the greater rates of fat deposition during yr 2. Regardless of type of diet fed, A steers were more efficient at depositing energy. Higher circulating T4 concentrations of A than B steers may explain some of the physiological differences between these breeds.  相似文献

The effects of feeding high levels of Mg were evaluated in a 130-d study with 24 steers. Six steers were allotted to each of four diets supplemented with MgO to attain .3, 1.4, 2.5 or 4.7% Mg, DM basis. Chromic oxide was used as digesta marker; fecal grab samples were collected once daily during nine consecutive 10-d periods. Steers fed 2.5 and 4.7% Mg refused some feed during the study, so their respective dietary Mg intakes were 2.4 and 3.7% (DM basis). Severe diarrhea and a lethargic appearance were observed in steers fed the two higher Mg levels. Fecal DM and apparent DM digestibility decreased linearly (P less than .01) with increased dietary Mg. Apparent Mg absorption (g/d) increased linearly (P les than .01) and apparent Ca and P absorption (g/d) decreased linearly (P less than .01) with addition of Mg to the diet. Increasing dietary Mg linearly elevated blood serum and erythrocyte Mg (P less than .01; P less than .05, respectively) and serum inorganic P (P less than .05) and linearly decreased serum Ca (P less than .01). Magnesium concentration in liver, kidney, skeletal muscle and rib-bone and P in skeletal muscle all increased linearly (P less than .05) with dietary Mg. Increasing dietary Mg in the steers caused a progressive degeneration of the stratified squamous epithelium of rumen papillae. A progressively more serious Mg toxicosis condition developed over time in steers fed diets containing 2.4 and 4.7% Mg.  相似文献

Microsomal arylhydrocarbon (benzo[a]pyrene) hydroxylase activity in hyperplastic nodules in the livers of rats was decreased by feeding 2-acetylaminofluorene in their diet to 10% of that without 2-acetylaminofluorene feeding. By immunoblotting analysis with the antibodies raised against P-450/B[a]P, which is a form of cytochrome P-450 catalyzing benzo[a]pyrene hydroxylation in liver microsomes of untreated rats, it was shown that P-450/B[a]P content was decreased in the microsomes prepared from the nodular tissues. Microsomal UDP-glucuronyltransferase activity toward phenolic xenobiotics such as 1-naphthol and 4-nitrophenol was increased by 4.5-5.0-fold in the nodular tissues. This increase was inhibited by the addition of antibodies raised against GT-1, a form of UDP-glucuronyltransferase glucuronizing phenolic xenobiotics. Immunoblotting analysis with the antibodies against GT-1 showed that a protein band corresponding to GT-1 was increased in the microsomes from nodular tissues. The increased UDP-glucuronyltransferase also had about 4,000 daltons "high mannose" oligosaccharide(s) like GT-1. The decreases of P-450/B[a]P and increases of GT-1 were observed mainly in nodular foci of the liver tissues. These results indicate that in liver nodules, decreased cytochrome P-450-dependent benzo[a]pyrene hydroxylase activity and increased UDP-glucuronyltransferase activity toward phenolic xenobiotics result from the decrease of the P-450 corresponding to P-450/B[a]P and the increase of the GT corresponding to GT-1.  相似文献

Twenty-four crossbred Florida native wether lambs, 28 kg initially, were assigned randomly to a basal diet (31 ppm Mn dry matter basis) supplemented with either 0, 500, 1,000, 2,000 or 4,000 ppm Mn from feed grade MnO or 2,000, 4,000 or 8,000 ppm Mn from reagent grade MnCO3. There were three sheep/treatment, with ad libitum access to feed and tap water. After 84 d, all animals were slaughtered and tissues removed for analysis. Dietary Mn at the highest levels from either source reduced (P less than .05) average daily feed intake and average daily gain. Hemoglobin and hematocrit were not affected by dietary Mn. Manganese concentration in kidney, spleen, heart, skeletal muscle, bone, serum (P less than .01) and liver (P less than .05) increased as dietary Mn increased. Kidney P was reduced (P less than .05) as dietary Mn increased; however, no other tissue minerals were affected. At comparable levels of supplementation, sheep fed feed-grade MnO exhibited numerically greater tissue Mn concentration than did those fed reagent-grade MnCO3 and regression analysis indicated higher rates of tissue uptake in MnO-supplemented sheep. Reduced feed intake and gain and apparent Mn homeostatic failure in liver were the only observed effects that may have been related to Mn toxicosis.  相似文献

This experiment had two objectives. The first was to test a hypothesis that the presence of mature cows can influence the growth rate and physiological maturation rate of growing heifers. The second objective was to define a phenomenon termed nonpuberal estrus (NPE). Nonpuberal estrus in a prepuberal heifer is a behavioral estrus that is not followed by ovulation and formation of a corpus luteum. Two years of data were collected. All heifers were crossbred and were sired by Hereford, Charolais and Tarentaise bulls in yr 1 (n = 153) and by the same three breeds plus Jersey, Shorthorn, Brahman and Longhorn in yr 2 (n = 207). All heifers were born in the spring and weaned in October when approximately 180 d old. After weaning, heifers were assigned at random (within breed of sire) to control or mature cow (MC) treatment pens. Control pens contained 25 or 26 heifers and a sterile marker bull, while MC pens contained 25 or 26 heifers and a sterile marker bull plus four nonpregnant, mature cows. The presence of the mature cows did not (P greater than .10) affect rate of gain or yearling weight. Sire breed did (P less than .01) influence weight on test and average daily gain during the test in both years and affected (P less than .01) yearling weight during yr 2 but not (P greater than .10) during yr 1. Six crossbred steers (315 kg) with cannulas in the rumen, proximal duodenum and distal ileum were used to study the influence of level and source of dietary fat on characteristics of digestion. Dietary treatments consisted of a steam-rolled barley-based finishing diet containing 1) no supplemental fat; 2) 4% yellow grease (YG); 3) 4% blended animal-vegetable fat (BVF); 4)8% YG; 5) 8% BVF or 6) 6% BVF and 2% crude lecithin. Increasing level of fat supplementation resulted in linear decreases (P less than .01) in ruminal and total tract digestion of OM and ADF and intestinal digestion of fat (P less than .05). At the 4 and 8% levels of supplementation, intestinal true digestibility of fat averaged 80.1 and 69.3%, respectively. Ruminal molar proportions of acetate decreased, and propionate molar proportion, as well as DE and ME values of the diet, increased linearly (P less than .01) with level of fat supplementation. The DE and ME values for fat were 8.17 and 9.76 at the 4% level and 7.35 and 8.72 Mcal/kg at the 8% level of supplementation, respectively. Yellow grease supplementation resulted in greater (P less than .05) ruminal fiber digestion and greater ruminal molar proportions of propionate than BVF. Intestinal fat digestion was similar (P greater than .10) for YG and BVF. Adding 25% lecithin to BVF resulted in greater ruminal fiber digestion and greater ruminal molar proportions of acetate; however, lecithin tended (P less than .10) to have a lower ME value than BVF.  相似文献

Effectiveness of vaccination in controlling ovine footrot   总被引:2,自引:0,他引:2  
Efficacy of reducing footrot by use of a commercial vaccine was evaluated over 2 yr with 223 ewes from five crossbred genotypes. Ewes were assigned randomly within genotype, age and initial footrot status to a 2 x 3 factorial array of vaccination in the 1st yr and fall vaccination, winter vaccination or control in the 2nd yr. The flock contained a single Bacteroides nodosus serotype (XV) known to be highly cross-reactive with one of the vaccine serotypes (British serotype H). Vaccination reduced footrot incidence by 61% (P less than .01) in the 1st yr and by 45% (P less than .01) in the 2nd yr. Vaccination of previously uninfected ewes did not significantly reduce the rate of new infection, but vaccination of infected ewes reduced re-infection by 92% (P less than .01). Vaccination in the 1st yr failed to produce any carryover or additive protection in the 2nd yr. Genotypes differed in both footrot incidence (P less than .05) and footrot reduction following vaccination (P less than .05), but the effect was inconsistent between years. Vaccination increased serum antibody titers in both years (P less than .01). Among vaccinates in the 1st yr, infected ewes had lower titer levels than uninfected ewes (P less than .01), but no correlation between titer level and incidence was observed in the 2nd yr.  相似文献

A 3 x 2 trial was conducted to determine the effects of adding canola oil (0, 5, or 10%) and copper sulfate (0 or 250 ppm Cu) to diets of growing-finishing swine on performance, carcass characteristics, and carcass fat fatty acid composition. The trial used 180 pigs (27 kg). Grower diets (.80% lysine for 0% canola oil diet) were given from 27 to 57 kg of BW and finisher diets (.64% lysine) from 57 to 102 kg. Diets were formulated to constant ME:lysine ratio within the grower and finisher phases. Over the entire growing-finishing period, the addition of canola oil to the diets resulted in linear improvements in rate of gain (P less than .05) and feed efficiency (P less than .01). Dietary additions of canola oil had no effect (P greater than .10) on resulting backfat thickness or longissimus muscle area but resulted in reductions (P less than .01) in loin marbling and color and carcass fat firmness, mostly noted in pigs fed the diets with 10% canola oil. Canola oil additions at 5 and 10% levels, respectively, resulted in a 23 and 37% reduction (P less than .01) in saturated fatty acids, 3 and 8% increase (P less than .01) in monounsaturated fatty acids, and 37 and 77% increase (P less than .01) in polyunsaturated fatty acids in the carcass fat compared with the diets without canola oil. Twenty-eight Hereford x Angus cows (4 yr of age) were used to determine the effects of pre- and postpartum dietary energy on performance and reproductive function in suckled beef cows. The experiment was designed as a 2 x 2 factorial with cows receiving either 70 (L) or 150% (H) of NRC recommended level of dietary energy before and(or) after parturition, resulting in four treatment combinations (L-L, L-H, H-L, H-H). Prepartum diets were fed for approximately 110 d, and postpartum diets were fed until either 10 d after the second postpartum ovulation or 150 d postpartum for those cows that failed to ovulate. Cows receiving low compared with high levels of energy before calving lost more (P less than .01) weight, body condition, subcutaneous fat, and longissimus muscle area before parturition and had calves with lighter (P less than .01) birth weights. Cows receiving low compared with high levels of energy postpartum lost more (P less than .01) weight, body condition, and longissimus muscle area after parturition. Low levels of energy before and after parturition decreased (P less than .01) milk production and calf weight at 70 d of age. Rates of cervical and uterine involution were unaffected by dietary energy treatments. Cows receiving high levels of energy prepartum had increased (P less than .01) mean concentrations and pulse frequency of LH in serum after parturition, and cows receiving high levels of energy postpartum had increased (P less than .05) pulse frequency of LH. Low levels of energy postpartum decreased (P less than .01) appearance rate of small (5.0 to 7.9 mm) and large (greater than or equal to 10 mm) follicles, and low levels of energy prepartum decreased (P less than .02) appearance rate of large follicles based on transrectal ultrasonography. Cows receiving high levels of energy prepartum had shorter (P less than .02) intervals from parturition to ovulation, and a higher (P less than .01) percentage of the cows that received high levels of energy postpartum ovulated by 150 d postpartum. In summary, prepartum level of dietary energy influenced birth weight and weight gain of calves, milk production, concentrations and pulse frequency of LH in serum, appearance rate of large follicles, and the interval to first ovulation. Postpartum level of dietary energy influenced milk production, weight gain of calves, pulse frequency of LH, appearance rate of small and large follicles, and the percentage of cows that ovulated after parturition.  相似文献

The influence of a biological response modifier (FK-565) on weanling pig performance was evaluated. One hundred twenty-five weanling pigs (weaned at 21 +/- 3 d) averaging 6.3 kg were utilized in a 35-d growth trial. Dietary treatments included a basal diet (1.25% lysine, corn-soybean meal-dried whey), the basal plus .1, 1 or 10 ppm FK-565 and the control plus an antibacterial combination containing chlortetracycline, sulfamethazine and penicillin. Performance was recorded weekly, and on d 35 all pigs were bled for whole blood and serum chemistry profiles and then were euthanatized. Heart, liver, kidneys and spleen weights were recorded. Also, gross and histological examinations were made of these organs, as well as sections of lung, ileum, bone marrow, thymus and mesenteric lymph node. By d 14, pigs fed the antibacterial diet gained faster (P less than .06) than pigs fed the control and FK-565 diets. However, no differences (P less than .10) in feed intake at d 14 or efficiency of feed utilization at either d 14 or 35 were observed. For the overall 35-d trial, ADG was greater (P less than .01) for pigs consuming the antibacterial diet than for pigs consuming control and the FK-565 diets. Twenty calves (7 mo old) were blocked by breed, sex, and weight into five groups of four calves and randomly assigned to either a 90% forage (alfalfa) or a 90% grain (50% sorghum and 50% wheat) diet fed at one (1M) or two (2M) times NEm for 140 d. Samples of ruminal epithelial tissue were collected from the anterior ventral sac, and papillae were cut free by hand and used for in vitro incubations and acyl-CoA synthetase assays. Substrates were acetate (90 mM), propionate (60 mM), butyrate (30 mM), and glucose (20 mM). Net productions of beta-hydroxybutyrate and acetoacetate from acetate were greater (P less than .01) with the 2M feeding; however, 14CO2 production from acetate was greater (P less than .05) with the grain diet. Net production of lactate (P = .09) and pyruvate (P less than .01) from propionate increased with the 2M feeding, whereas net lactate production from glucose decreased (P less than .01). Uptakes of VFA were similar with 1M and 2M feeding and were about 10-fold greater than uptakes of glucose. Production of 14CO2 from propionate was two- to fivefold greater than from acetate, butyrate, or glucose. Oxygen consumption was greater (P less than .01) with 2M feeding and unaffected by substrate. Activities of butyryl-CoA synthetase (nmol.mg of tissue-1.h-1) were greater (P less than .05) for animals consuming the forage diets. Addition of butyrate inhibited acetyl- and propionyl-CoA synthetase activity by 63 and 92%, respectively for all dietary treatments. Overall, metabolism of ruminal epithelial tissue tended to increase with the 2M feeding. Influence of dietary forage content on metabolism and activity of acyl-CoA synthetases was minimal, but the high-forage diet caused slight increases.  相似文献

Twenty-four lambs, averaging 29 kg, were used to determine the effect of supplemental dietary Fe on performance and Cu, P, Zn and Mn utilization. Treatments consisted of supplemental Fe at 0, 300, 600 or 1,200 mg/kg diet as ferrous carbonate. The basal diet contained 154 mg Fe/kg diet and consisted of 90% Coastal bermudagrass pellets, 9.45% group corn, .5% sodium chloride and .05% vitamin mix. Lambs were slaughtered after having ad libitum access to diets for 98 to 121 d. Dietary Fe did not affect lamb gain or feed intake. Supplemental Fe increased Fe concentrations in liver (P less than .01), spleen (P less than .01) and bone (P less than .10), but not in kidney and muscle. Serum Fe concentrations and percentage transferrin saturation in serum were increased (P less than .01) by supplemental Fe at 28 and 84 d, but not at the termination of the study. Plasma Cu was decreased (P less than .01) at 56 d, whereas serum ceruloplasmin activity was reduced (P less than .01) at 28 d in lambs fed 1,200 mg Fe/kg diet compared with lambs fed 600 mg Fe/kg diet. Lower levels of Fe (300 and 600) reduced (P less than .01) ceruloplasmin by 56 d and plasma Cu by 84 d compared with controls. Liver Cu also was decreased (P less than .05) by supplemental Fe. Experiments were conducted with rats and swine to determine the potential of dietary alfalfa as a treatment for zearalenone (Z) toxicosis. Ninety-six female weanling Wistar rats were fed a casein-based semipurified diet containing 0, 15 or 25% alfalfa and 0 or 250 micrograms Z/g feed. Exposure to Z for 14 d resulted in reduced growth, feed consumption and feed efficiency as well as kidney and liver enlargement and reduced activity of 3 alpha-hydroxysteroid dehydrogenase (3 alpha-HSD). Z had no effect on uterine weight. Including alfalfa in these diets reduced the inhibitory effects of Z on growth and feed consumption, minimized Z-induced liver enlargement and increased hepatic 3 alpha-HSD activity. Dietary alfalfa also reduced concentrations of residual Z and zearalenols (Zl) in liver. In a second experiment, 108 Yorkshire gilts weighing 8 to 11 kg were fed diets containing 0, 15 or 25% alfalfa and 0, 10, 20 or 40 micrograms Z/g feed for 4 wk. Z caused uterine enlargement when fed as low as 10 micrograms/g feed, although no effects were seen in growth rate, feed consumption or feed efficiency. Alfalfa decreased uterine enlargement (P less than .05), but caused a depression in feed efficiency (P less than .05). Hepatic 3 alpha-HSD activity was five times lower in swine than in rats, although activity still tended to decrease with Z and increase with alfalfa feeding. Residues of Z and Zl in pig liver indicated species differences in the metabolism of Z. These studies show that dietary alfalfa promotes Z metabolism in rats and that this feedstuff may also be useful for treating Z toxicosis in livestock.  相似文献
