An 8-year-old grey Arabian mare was presented for reproductive evaluation with a history of failing to become pregnant during two previous breeding seasons. An enlargement was identified on the right uterine horn on rectal examination which was confirmed as a fluid-filled nonechogenic mass on ultrasonography. Subsequent examination procedures and laparotomy revealed an old hematoma in the wall of the uterine horn.  相似文献

South American camelids show high embryo loss rate, during the first 60 days of pregnancy. One of the factors which may be related to this situation is that over 98% of the embryos implant in the left uterine horn (LUH) even though both ovaries contribute similarly to ovulation. There is scarce information about the uterine environment of female camelids at any physiological state that could explain the capability of the LUH to attract the embryo and maintain pregnancy. We describe, for the first time, the biochemical and protein profile of uterine fluid (UF), addressing the right and LUH environment in non‐pregnant and pregnant alpacas. Different substrates, electrolytes and metabolites were assayed in both uterine horn fluids. Small changes were observed in glucose and total protein levels, which were more noticeable during pregnancy. In addition, 10 specific proteins were found in the left horn fluid in 5‐week‐pregnant alpacas, and two protein bands were identified in non‐pregnant alpaca right horn fluid. These results would provide basic information for identification of possible markers for pregnancy diagnosis, reproductive diseases and hormone‐treated animals evaluation and hence contributing to improve the pregnancy rate.  相似文献

This short communication reports the clinical, ultrasonographic and histopathological findings in a cat with atresia of the uterine cervix and mucometra. After 6 months of continuous oestrous behaviour, a remarkable abdominal enlargement was observed in a 14-year-old queen. A presumptive diagnosis of mucometra was concluded after the ultrasound evaluation and based on clinical signs and blood analyses. Ovariohysterectomy revealed a notable symmetrical distension (4-5 cm in diameter) of both uterine horns that were filled with fluid (690 ml); microbiological analyses confirmed the aseptic nature of the uterine fluid. Ovarian follicular cysts and cystic subsurface epithelial structures, >1.5 cm in diameter, were present in both ovaries and no corpora lutea were observed. Gross and microscopic evaluation of the uterus confirmed the development of cystic endometrial hyperplasia and the absence of an internal cervical os. The endometrial hyperplasia and mucometra could have developed as a consequence of repeated oestrogenic stimulation.  相似文献

Maternal recognition of pregnancy refers to the requirement for the conceptus (embryo and its associated extra-embryonic membranes) to produce a hormone that acts on the uterus and/or corpus luteum (CL) to ensure maintenance of a functional CL for production of progesterone; the hormone required for pregnancy in most mammals. The pregnancy recognition signal in primates is chorionic gonadotrophin which acts directly on the CL via luteinizing hormone receptors to ensure maintenance of functional CL during pregnancy. In ruminants, interferon tau (IFNT) is the pregnancy recognition signal. IFNT is secreted during the peri-implantation period of pregnancy and acts on uterine epithelia to silence expression of estrogen receptor alpha and oxytocin receptor which abrogates the oxytocin-dependent release of luteolytic pulses of prostaglandin F2-alpha (PGF) by uterine epithelia; therefore, the CL continues to produce progesterone required for pregnancy. Pig conceptuses secrete interferon delta and interferon gamma during the peri-implantation period of pregnancy, but there is no evidence that they are involved in pregnancy recognition signaling. Rather, pig conceptuses secrete abundant amounts of estrogens between Days 11 to 15 of pregnancy required for maternal recognition of pregnancy. Estrogen, likely in concert with prolactin, prevents secretion of PGF into the uterine venous drainage (endocrine secretion), but maintains secretion of PGF into the uterine lumen (exocrine secretion) where it is metabolized to a form that is not luteolytic. Since PGF is sequestered within the uterine lumen and unavailable to induce luteolysis, functional CL are maintained for production of progesterone. In addition to effects of chorionic gonadotrophin, IFNT and estrogens to signal pregnancy recognition, these hormones act on uterine epithelia to enhance expression of genes critical for growth and development of the conceptus.  相似文献

OBJECTIVE: To macroscopically, histomorphologically, and morphometrically compare uterine tissues obtained from nulliparous and multiparous dogs. ANIMALS: 12 dogs constituting 2 homogenous groups (7 nulliparous Beagles and 5 multiparous Beagles, each of which had at least 7 pregnancies). PROCEDURE: Serum concentrations of progesterone and estradiol-17beta were determined. Samples of uterine tissues were fixed, embedded in paraffin, and cut into serial cross sections. Morphometric analysis was performed on systematically selected trichrome-stained sections. RESULTS: Mean absolute volume of the uterine wall did not differ between groups. Volume of blood vessels was significantly higher and relative mean value for myometrial connective tissue was significantly lower in the multiparous dogs. Arteries in the myometrium of multiparous dogs had pronounced thickening of the intima (ie, pregnancy sclerosis) and a concomitant thinning of the smooth muscle layer (tunica media). Furthermore, the elastica interna of these arteries appeared to be disintegrating and was highly and irregularly folded. Adventitia of the myometrial veins in multiparous dogs contained numerous layers of elastic fibers; however, only a few layers were observed in the adventitia of corresponding veins in nulliparous dogs. CONCLUSIONS AND CLINICAL RELEVANCE: In this study, we documented that in contrast to other species, the uterus of nonpregnant dogs does not increase in size and volume even after at least 7 pregnancies. Furthermore, sclerotic alterations of uterine arteries are proof of at least 1 pregnancy. Results of this study may be useful in the evaluation of uterine diseases.  相似文献

ABSTRACT: There is a dialogue between the developing conceptus (embryo-fetus and associated placental membranes) and maternal uterus which must be established during the peri-implantation period for pregnancy recognition signaling, implantation, regulation of gene expression by uterine epithelial and stromal cells, placentation and exchange of nutrients and gases. The uterus provide a microenvironment in which molecules secreted by uterine epithelia or transported into the uterine lumen represent histotroph required for growth and development of the conceptus and receptivity of the uterus to implantation. Pregnancy recognition signaling mechanisms sustain the functional lifespan of the corpora lutea (CL) which produce progesterone, the hormone of pregnancy essential for uterine functions that support implantation and placentation required for a successful outcome of pregnancy. It is within the peri-implantation period that most embryonic deaths occur due to deficiencies attributed to uterine functions or failure of the conceptus to develop appropriately, signal pregnancy recognition and/or undergo implantation and placentation. With proper placentation, the fetal fluids and fetal membranes each have unique functions to ensure hematotrophic and histotrophic nutrition in support of growth and development of the fetus. The endocrine status of the pregnant female and her nutritional status are critical for successful establishment and maintenance of pregnancy. This review addresses the complexity of key mechanisms that are characteristic of successful reproduction in sheep and pigs and gaps in knowledge that must be the subject of research in order to enhance fertility and reproductive health of livestock species.  相似文献

Uteroferrin,also known as type 5 tartrate resistant acid phosphatase ( ACP5 ) or TRAP,is an iron-containing glycoprotein secreted by uterine gland epithelium (GE) in response to progesterone and transported across the placental areoalae into the fetal circulation and allantoic fluid to deliver iron and to stimulate hematopoeisis in pigs.This study determined if ACP5 was expressed in the ovine uterus in response to pregnancy,progesterone,interferon tau,placental lactogen,and placental growth hormone.ACP5 protein was present in uterine GE of cycfic and early pregnant ewes,particularly between days 18 and 120 of pregnancy.ACP5 mRNA was expressed in uterine GE of cyclic and pregnant ewes in the same temporal and cell-specific manner.ACP5 was present in secretions from uterine glands,i.e.,uterine milk,and aUantoic fluid from days 40 to 80 of pregnancy,and in uterine flushings from cyclic and early pregnant ewes.Progesterone induced expression of ACP5 mRNA and intrauterine infusion of recombinant ovine interferon tau further stimulated ACP5 expression in uterine GE of ewes,but intrauterine injections of ovine placental lactogen and ovine growth hormone had no effect on ACP5 expression in uterine GE.These results indicate that ACP5 is:1 ) expressed only in GE in response to progesterone ;2 ) secreted into the uterine lumen and transported into the conceptus via plaeental areolae during pregnancy;and 3) present in secretions from uterine GE and in allantoic fluid.The roles of ACP5 in the ovine uterus may include transport of iron across the placenta and stimulation of hematopoiesis.  相似文献

A 6-year-old female Alaska Malamute dog was presented for evaluation of abdominal enlargement referred by a local veterinarian. On the history, the owner complained of chronic abdominal enlargement initiated more than 4 months ago, reduced appetite, occasional vomiting and general dullness. He also complained of greenish mucous intermittent vaginal discharge starting 10 days ago. The bitch was chronically treated with medroxiprogesterone acetate. A laparatomy was performed and fluid in the abdomen was found and aspirated during the surgery. Also a very fluid-filled distended uterus and a mass in the distal part of the left uterine horn were found. The mass was encapsulated by the omentum, but areas of necrosis and calcification were identified. Histopathological diagnosis was endometrial adenocarcinoma.  相似文献

The reproductive tracts of 22 Friesian dairy cows were examined from seven to 35 days after insemination using a real time B-mode ultrasound scanner with a 7.5 MHz transducer. The earliest detection of pregnancy was at nine days when a vesicle was imaged within the lumen of the uterine horn. The early conceptus was seen at day 13 within the vesicle and these structures were followed ultrasonically until day 35. There was a sudden enlargement of the vesicle at day 19 and a heart beat was detected in the embryo at day 22. The allantois was imaged at day 23 and the amnion by day 29. The embryonic outline was clearly defined by day 33 when the body cavities could be discerned. This ability to determine pregnancy at an early stage should prove to be a useful technique in investigating the problems associated with early embryonic death in cattle.  相似文献

Deep intra‐uterine insemination is commonly accepted as a routine procedure for artificial insemination in horses. The motives and principles of deep insemination are well described, but the equipment used may differ. In this trial, the efficiency of two different insemination pipettes for deep intra‐uterine insemination in the mare was compared with insemination into the uterine body using commercially available frozen–thawed semen of two stallions of proven fertility. These inseminations were performed using two different doses. The semi‐flexible Minitube pipette was compared with a newly designed insemination device with a more flexible telescopic insemination catheter (Ghent device). The semi‐flexible Minitube pipette performed better than the newly designed insemination device with respect to pregnancy outcome (p = 0.008). The superiority of deep horn insemination over uterine body insemination was reflected by the better pregnancy rates obtained after deep insemination using the same low doses (30.6% better pregnancy rates) (p = 0.0123).  相似文献

The Siberian polecat (Mustela eversmanni) is the preferred species to assess procedures and establish normative values for application in the related and endangered black-footed ferret (Mustela nigripes). This study was undertaken to physically, ultrasonographically, and radiographically evaluate fetal development in a spontaneously breeding captive Siberian polecat population. Ultrasonographically, fetal sac enlargement allowed presumptive pregnancy detection as early as 12 days of gestation, the fetal pole was the first definitive sign of pregnancy at about 18 days of gestation, when the fetal heart beat also appeared, and definitive pregnancy detection by ultrasound was essentially 100% accurate after 18 days. The estimation of fetal number by ultrasound was less reliable than by radiography, as it is in other litter-bearing species. Crown-rump growth, organ differentiation, and calcification patterns resembled those of domestic carnivores except that comparable developmental stages in polecats occurred at disproportionately later times, suggesting that young Siberian polecats are delivered in a less developed state. Careful palpation permitted detection of pregnancy after day 17 but with less certainty than with ultrasound. Radiographic evaluation was insensitive and of limited value for pregnancy detection until near term. Litter number and fetal detail were difficult to assess until ossification could be observed, 3-6 days before parturition.  相似文献

This study determined ultrasonographic parameters of fetuses and uterine adnexa in late pregnancy in normal, cloned, and high-risk pregnancies in relation to perinatal and neonatal outcome. Ten cows with normal pregnancies (CONTROL, mean pregnancy length 273 d), 10 sick cows with potentially compromised pregnancies (HIGH-RISK, mean pregnancy length 267 d), and 10 heifers with cloned pregnancies (CLONED, mean pregnancy length 274 d) were examined at more than 260 d of gestation. There was no difference in mean fetal heart rates among the groups. The cloned calves were heavier (57 ± 8 kg) than calves from CONTROL group (36 ± 7 kg), and calves from HIGH-RISK group (37 ± 13 kg) (P = 0.003). The diameter of the thoracic aorta was positively correlated (R = 0.62) with fetal birth weight in the CONTROL group (P = 0.01). Fetal activity was not associated with survival. The results suggest that transabdominal ultrasonographic assessment of the fetal well-being may serve as a potential tool for evaluation of the fetoplacental unit.  相似文献

The effect of exogenous iron and folate on reproductive performance in swine is equivocal. However, the effect of exogenous iron and folate on secretion of their respective uterine transport proteins has never been reported. Twenty gilts were infused (n = 5 per treatment) with either 1) saline, 2) alpha-tocopherol, 3) alpha-tocopherol plus iron citrate, or 4) alpha-tocopherol plus tetrahydrofolate on d 11 to 14 of pregnancy. Intravenous infusion of iron citrate and tetrahydrofolate increased (P < 0.05) plasma iron and folate, respectively, for 6 to 8 h after treatment. Treatments had no effect on uterine content of uteroferrin or secreted folate-binding protein in uterine flushings obtained on d 15 of pregnancy. These data suggest that uterine secretion of uteroferrin and secreted folate-binding protein are not influenced by plasma levels of iron and folate, respectively, and may provide an explanation for the equivocal effect of iron and folate treatment on reproductive performance in gilts.  相似文献

To assess the effects of a single supraphysiological postnatal administration of a progestogen on uterine glands in dogs, 10 females were randomly assigned to a medroxyprogesterone acetate 35 mg (MPA; n = 6) or placebo (n = 4) group within the first 24 h of birth. The safety of the treatment was also evaluated. A transient mild clitoris enlargement appeared in MPA-treated females. Microscopic postpubertal uterine assessment revealed the presence of uterine glands in all cases without significant differences in the area occupied by the glands per µm2 of endometrium nor in the height of the uterine epithelium.  相似文献

The effects of norepinephrine (NE) uptake inhibitors on the porcine uterine artery's contractile response to NE and their potential alteration during early pregnancy (Day 13 postcoitus; day of coitus = Day 0) in comparison with the end of the luteal phase (Days 11-14; first day of behavioral estrus = Day 0) was investigated. This pregnancy time point is characterized by a transient increase in resting uterine blood flow dependent on the presence of blastocysts, an increased endometrial vascular permeability, and the beginning of the endometrial attachment of the blastocysts. A cumulative concentration-response curve (CCRC) to NE was produced in isometrically-mounted rings. Cocaine (5 microM) was used to inhibit neuronal NE uptake and hydrocortisone (30 microM) was used to inhibit extraneuronal NE uptake. Either drug alone induced a leftward shift of the CCRC to NE without affecting the maximal response. This shift had the same amplitude at the end of the luteal phase and in early pregnancy. However, the leftward shift induced by cocaine was larger than that induced by hydrocortisone only in early pregnancy, and the leftward shift induced by exposure to both hydrocortisone and cocaine was larger in early pregnancy than at the end of the luteal phase. These results suggest that a significant sensitization of the contractile response of the porcine uterine artery to NE is induced by neuronal and extraneuronal uptake inhibitors and that this effect is altered in early pregnancy, possibly reflecting the existence of a mechanism protecting the uterine artery against excessive sympathetic stimulation.  相似文献

Between January 1993 and April 1998 in a total of 27,378 sheep an ultrasonographic early pregnancy diagnosis was done. The determined pregnancy rates (mean = 82.2%) were related to the mating systems, duration of mating season and time of the year. The benefits of scanning are described. If the pregnancy rate is lower than 97% the early pregnancy diagnosis is economical to flock management. Cases of uterine and fetal pathology (fetal mortality, maceration, hydrometra, pyometra) are presented.  相似文献

This short communication reports the impact of endometrial biopsies, uterine flushings and follicular fluid aspiration procedures at day 6 post artificial insemination (AI) on pregnancy rates. In Experiment 1, cows were timed AI (TAI) and assigned to the following treatment groups: control (n = 37), uterine flushing (n = 35) and endometrial biopsy (n = 38). On day 30 post AI, pregnancy rates were 40.5%, 33% and 28.5%, respectively (p > 0.1). Pregnancy rate on day 60 was lower (p < 0.004) in flushed cows than in the controls. In Experiment 2, oestrus was detected and cows were assigned to flushing (n = 32) or biopsy (n = 33) treatments 6 days after AI, which resulted in pregnancy rates of 31% and 36%, respectively (p > 0.1). In Experiment 3, cows were, 6 days after TAI, randomly assigned to the following treatments: control (n = 84) or aspiration of the largest follicle (n = 73). Pregnancy rates on day 30 post AI were 63.5% for the control group and 53% for the aspirated group (p > 0.1). In conclusion, uterine flushing and endometrial biopsy negatively affect pregnancy rates, but neither procedure can be considered to be incompatible with pregnancy maintenance. Follicular aspiration during pregnancy does not interact with pregnancy success. The amount and quality of samples obtained are compatible with the use of cellular and molecular analysis of uterine variables from cows that failed or succeeded on maintaining pregnancy.  相似文献

Background: A major,unresolved issue is how the uterine microenvironment determines pregnancy success in cattle.Before implantation,conceptus development depends on the uterine secretome(i.e.,histotroph).Despite its pivotal role,little is known about the dynamics of histotroph synthesis and changes in composition throughout the early diestrus and the relevance to pregnancy establishment.We hypothesize that disturbances on histotroph composition affect the establishment of pregnancy.Aim was to disturb histotroph composition at early diestrus and verify the effects on:(Exp.1) timing to restore its composition; and(Exp.2) pregnancy rate after multiple-embryo transfer.Estrous cycle of multiparous Nelore cows were synchronized and estrus was considered d 0(D0) of the experiments.Disturbance was through flushing each uterine horn with 30 m L of DMPBS and collecting the resulting uterine luminal flushing(ULF) on D1; D4; D7; D1 + D4 + D7.Control group remained not-collected.In Exp.1,ULF was collected on D7.5 from all animals and used for quantification of total protein concentration and abundance of albumin.In Exp.2,three in vitro-produced embryos were transferred to the uterine horn ipsilateral to the ovary containing the CL on D7.5 and pregnancy was checked on D25 by ultrasound.Results: In Exp.1,ULF collection on D4 or D7 increased(1.5-to 2.2-folds) the total protein concentration and albumin abundance.ULF collection on D1 did not alter(P 0.10) these endpoints.In Exp.2,ULF collected on D4 or D7 decreased pregnancy rates to approximately half of that measured in the remaining groups.Conclusions: Subtle perturbations imposed to the native intrauterine milieu,such as those caused by a single,low-volume collection of ULF,profoundly disturbs intrauterine composition and pregnancy success.At least 4 d were necessary for the uterus to recover its composition and the functional capacity to carry post-implantation gestation.  相似文献

In 1977 Bazer and Thatcher proposed that maternal recognition of pregnancy in the pig involves the secretion of PGF(2alpha) towards the uterine lumen (exocrine) rather than towards the uterine venous drainage (endocrine) as occurs in the non-pregnant pig during the mid to late stages of the estrous cycle. The retrograde transfer of PGF(2alpha) from the venous blood and uterine lymph into the uterus and the ability of the uterine vein and artery wall to accumulate PGF(2alpha) could constitute a part of putative mechanism of corpus luteum protection during early pregnancy. A luteotropic/anti-luteolytic effect of PGE(2) in the pig also has been frequently demonstrated and it seems that the most effective agent in changing PGE(2):PGF(2alpha) secretion is estradiol. The role for oxytocin during luteolysis and early pregnancy is controversial. It appears, however, that the main function of this hormone is autocrine and/or paracrine stimulation of PGF(2alpha) secretion. Pig trophoblastic interferons, unlike those of ruminants, do not themselves exert an anti-luteolytic effect in pigs. It is likely, that cytokines and angiogenic growth factors are involved in the initiation of luteolysis and/or maintenance of corpora lutea (CL).A discovery of functional LH receptors in porcine endometrium opened a new possibility for this hormone in luteolysis and perhaps in recognition of pregnancy in pigs. The endogenous LH pulses can provoke prostaglandin secretion from endometrium in pigs. On the other hand prolongation of up-regulation of LH receptors in endometrium of early pregnant gilts can additionally increase angiogenic factor production before the process of implantation is completed. Finally new integrated concepts of luteolysis and inhibition of luteolysis in pigs based on selectively reviewed information are presented.  相似文献
