Quantities and types of ceramides and their relationships to physical properties of the horn covering the claws of clinically normal cows and cows with subclinical laminitis were investigated. Total ceramide content of the horn covering the sole and wall from cows with subclinical laminitis was 872.2 +/- 146.6 microg/g and 528.6 +/- 61.3 microg/g, respectively, and was significantly (P < 0.01, 0.05) lower than that from clinically normal cows. The mean moisture content in the claws from cows with subclinical laminitis (43.5% +/- 4.3%) was significantly (P < 0.05) higher than that in the claws from clinically normal cows. The hardness of claws from cows with subclinical laminitis (35.2 +/- 3.5) was significantly (P < 0.05) less than that of claws from clinically normal cows. Significant correlations between ceramides and moisture content (P < 0.001) and between ceramide and hardness (P < 0.001) were found in clinically normal cows and cows with subclinical laminitis. Our results indicate that decreases in ceramide contents may be related to changes in physical properties of the horn covering the claw in cows with subclinical laminitis.  相似文献

Effects of dietary biotin supplementation on serum biotin levels and physical properties of sole horn of 40 Holstein cows were evaluated. The mean serum biotin level in biotin-supplemented cows after 10 mo of biotin supplementation (1163.2 +/- 76.2 pg/mL) was significantly higher (P = 0.007) than that in control cows (382.0 +/- 76.2 pg/mL). The sole horn of biotin-supplemented cows was significantly harder (P = 0.026) and had a significantly lower moisture content (P = 0.021) than that of control cows. No morphologic differences in horn tubules or intertubular horn were found between the biotin-supplemented and control cows. The total lipid content of sole horn was significantly higher (P = 0.030) in the biotin-supplemented cows than in the control cows. These results suggest that dietary biotin supplementation causes increases in serum biotin levels and changes in physical properties and fat content of sole horn.  相似文献

The field study investigated severity, localisation and incidence of claw lesions of dairy cows and their healing process during a period of three months on selected mountain pastures in the central part of Switzerland. In 60 cows, which were at least 120 days in their lactation, the healing process was compared with the biochemical profiles. In 141 cows 197 claw lesions were recorded. Diagnosed were only sole ulcers (38%) and white line lesions (62%). In the first and second half of the summer term, the number of claw lesions was equal, although more severe lesions occurred mainly during the second half (89%). The lesions were treated surgically and the affected claw was elevated on a wood block or a plastic shoe. Average time for formation of a close layer of horn was 14 days. A delayed healing process was observed in dairy cows with an milk yield over 5500 kg per lactation, as well as in the second half of the summer term. Cows with a delayed healing process had significantly higher concentrations of free fatty acids and beta-hydroxybutyrate, and higher plasma enzyme activities for AST than cows with adequate healing process. This indicates that cows with a relatively high milk production touch upon the limits of their physical capacity under harder conditions on alpine pastures, which may affect also the healing process of claw lesions.  相似文献

Immunoglobulin (Ig) G1 concentrations in milk from Holstein cows was measured to determine if transfer and concentration was influenced by production factors (lactation number, stage of lactation, daily milk production), milk composition (milk fat, protein, lactose, and total solids content) or by serum IgG1 concentration. Two hundred and ninety-nine Chinese Holstein cows were randomly selected from four herds containing a total of more than 1600 lactating animals. The concentration of IgG1 in the milk and serum was determined by ELISA.Milk IgG1 concentrations varied between 0.030 and 0.614 mg/mL and significantly correlated with lactation number, stage of lactation, daily milk production and somatic cell count. The IgG1 mass was found to highly correlate with lactation number, stage of lactation, daily milk production and milk protein content. Lactation number had the highest positive direct relationship with IgG1 concentration.  相似文献

Forty Holstein dairy cows receiving a 38% concentrate diet based on maize silage were assigned to either a control group, either a biotin group, receiving 20 mg of biotin per day from 15 days before expected calving date and for 120 days after calving. Milk production was measured daily, milk fat content, protein content, urea and somatic cell counts were determined weekly from week 2 to week 17 of lactation. The profile of milk fatty acids was determined at weeks 3 and 10. Plasma glucose and blood beta-hydroxybutyrate were determined before calving and at weeks 1, 2, 3, 5, 7 and 10 of lactation.

Biotin supplementation resulted in an increased milk production in multiparous cows during weeks 2 to 6, but the effect was no more significant between 7 and 17 weeks of lactation. Milk protein percent was decreased by 0.1% in multiparous cows. Milk fat content was not affected by biotin, and milk fat daily production tended to increase during early lactation. In milk fat, biotin supplementation tended to decrease the proportion of fatty acids with less than 16 carbons at week 3, but the daily amount was not affected. Biotin tended to decrease biohydrogenation intermediates, increased C16:1 at week 3, and tended to increase cis-9 C18:1 at weeks 3 and 10. After 7 weeks of lactation, biotin tended to increase blood beta-hydroxybutyrate in multiparous cows with values remaining in a normal range, and decreased plasma glucose in primiparous cows. These modifications of plasma parameters, milk protein content and profile of milk fatty acids could be due to a higher lipid mobilisation from adipose tissue driven by the increased milk production.  相似文献

A cross-over study was conducted to investigate the effect of intramammarily infused lipopolysaccharide (LPS) on the acute phase reaction in early (EL) and in late (LL) lactation. Nine cows received intramammary injections of 100 microg of Escherichia coli 0111:B4 LPS during EL and LL. The severity of each cows systemic and local signs and change in milk appearance were recorded and scored throughout the experiment. Systemic and local signs were found to be more serious in EL cows. Tumor necrosis factor alpha (TNF alpha) was detected in milk but not in serum. Serum amyloid A (SAA) concentrations increased both in serum and in milk. The milk TNF alpha concentrations peaked at 8 h post-challenge (PC). SAA concentrations started to increase at 8 h PC, and peak concentrations were seen at 32 and 48 h PC in milk and serum, respectively. The milk TNF alpha and SAA seemed to be correlated, being on average higher in EL. Serum SAA concentration was not correlated with milk TNF alpha or SAA, nor with the severity of local or systemic signs, but was correlated with changes in milk appearance.  相似文献

Milk fever occurring during the peripartum period has been suggested to be caused by fatty liver developed during the nonlactating stage because diseased cows have increased serum concentrations of non-esterified fatty acids (NEFA) and show hepatic lipidosis. In cows with fatty liver and related diseases such as ketosis, serum concentrations of apolipoprotein (apo) B-100 and apoA-I are decreased. The purpose of the present study was to examine whether apoB-100 and apoA-I concentrations are similarly decreased in cows with milk fever. Apolipoprotein concentrations were also measured in cows with downer syndrome, which has been suggested to be related, at least in part, to milk fever. Compared with healthy cows during early lactation, apoB-100 and apoA-I concentrations were decreased in cows with milk fever and also in downer cows. In cows with milk fever, the decreases in apoB-100 and apoA-I concentrations were associated with increased NEFA and decreased cholesterol and phospholipid concentrations. However, in downer cows, serum lipid concentration changes were not as distinct as in cows with milk fever. These results, coupled with previous findings on the decreases in apoB-100 and apoA-I concentrations of cows with fatty liver-related diseases, suggest that fatty liver is involved in the development of milk fever and partly in that of downer cow syndrome.  相似文献

The influence of different copper and zinc contents in rations on blood serum concentrations was tested in a feeding trial at the Institute of Animal Nutrition of the Federal Research Institute for Animal Health (FLI). A total of 60 German Holstein breed cows (38 first lactating and 22 second lactating cows) were tested. All animals received a diet based on maize‐ and grass silage ad libitum. The animals were divided into two groups with 30 cows each. Group A received concentrates supplemented with copper and zinc as recommended, whereas Group B was offered a concentrate with roughly double the amount of copper and zinc. Blood samples were taken several times during the lactation. The concentrations of copper and zinc in the serum were measured and correlations between trace element intake and their serum concentrations were calculated. The mean dry matter intake was significantly higher for cows in the second lactation (19.1 kg/day) than for cows in the first lactation (16.4 kg/day), whereas the DM‐intake was constant in the different feeding groups (17.3 and 17.4 kg/day). The correlation between feed intake in the first and the second lactation was r = 0.76. The copper concentration in the serum was not influenced by the different feedings (11.9 and 12.5 μmol/l), but high variations were found between the cows. The influence of the lactation number was not significant (12.1 and 12.4 μmol/l). In contrast to copper, the zinc concentration in the serum was significantly higher for Group B (14.0 μmol/l) than for Group A (12.2 μmol/l), respectively, for second lactating cows (13.8 μmol/l) in relation to 12.7 μmol/l for first lactating cows. The individual variation was also high as for copper. Neither the correlation between copper intake and copper serum concentration nor the correlation between zinc intake and zinc serum concentration was significant.  相似文献

The prevalence of digital dermatitis (DD) in first lactation cows (FLCs) presented at 10 breeding cattle auctions was determined in a single auction centre in Austria over a period of 14 months. At each auction, FLCs were selected randomly for claw examination from the monthly auction catalogue. After obtaining owner consent, the hindclaws were examined in a walk-in crush. The type of lesion (M1–M4) and the diameter of DD lesions together with claw horn lesions and the trimming status of the claws were evaluated.From a total of 1110 FLCs registered for the ten auctions, 399 (36%) were randomly chosen for examination and 199 FLCs (17.9%) could be defined. In 63 cows (15.7%), the owners refused consent. A total of 24 FLCs were found to have DD lesions on one or both hindlimbs, resulting in a DD prevalence of 12.1%. The size of lesions ranged from 0.5–3 cm in diameter. No influence of environmental factors (such as season or housing) on the prevalence of DD could be found. In 30/199 (15.1%) of FLCs examined, claw horn lesions such as double sole, white line disease and sole ulcer were observed, but both claw horn and DD lesions were seen in only three cows. Potential buyers at auctions should be aware of both conditions, as the 12.1% prevalence of DD and the 15.1% prevalence of claw horn lesions in FLCs are considerable. Critical evaluation of claw health and DD is essential when purchasing breeding cattle.  相似文献

The correlation between the serum hydroxyproline concentration and serum activity levels of TRAP and BALP was examined in 41 cows. The correlated coefficient (r) was 0.6391 for TRAP and 0.3147 for BALP, respectively. Judging from the significant correlation to the serum hydroxyproline concentration, serum TRAP activity is an index for bone metabolism in cows. Serum TRAP activity was therefore measured in 205 healthy cows (2-9 years old) in order to observe the changes in bone resorption with aging and milk production. TRAP levels differed slightly between group A (< or =4 yrs) and B (5 yrs< or =) at the same stage of lactation. The activity levels rose slightly at the height of lactation stage and during the dry stage.  相似文献

The effects of ammonia and hydrogen sulfide on the physical and biochemical properties of the claw horn of Holstein cows were evaluated. Significant (P < 0.05, 0.01) decreases in hardness and elasticity were found in claw horns soaked in ammonia (NH3) and hydrogen sulfide (H2S) solutions compared with those that were soaked in water for 12, 24, and 48 h. Water absorption rate, as a indicator of permeability barrier function, increased significantly (P < 0.05) over time during the soaking period and was found to be dependent on the concentrations of NH3 and H2S in the solutions. The contents of ceramide, the main lipid component for the permeability barrier system of the stratum corneum, were significantly decreased in claw horns soaked in NH3 and H2S solutions compared with the values before soaking. Quantities of eluted protein released from claw horns treated with NH3 and H2S solutions were approximately 20 times and 30 to 40 times greater than those released from claw horns treated with water alone. Interestingly, the quantities of cytokeratin 10, the main cytoskeletal protein of the stratum corneum, eluted from claw horns treated with NH3 and H2S solutions were markedly greater than the quantity released from horns soaked in water. Our results suggest that abnormal changes in the physical property of claw horn caused by NH3 and H2S treatment are due to disruption of the biochemical property of the claw horn induced by these chemical agents derived from slurry.  相似文献

We investigated the effect of parity number on the dry matter intake (DMI) of cows during the first week after calving. Eighty‐three cows were evaluated from 14 days before to 7 days after calving. DMI and milk yield were measured for 7 days after calving, and the calving score was measured. Blood samples were collected throughout the experiment. The average DMI during the first week after calving was reduced in the first‐lactation heifers and high‐parity number cows. A quadratic relationship between the parity number and the DMI was observed. The first‐lactation heifers had lower prepartum serum total protein (TP) concentration and milk yield, higher prepartum serum nonesterified fatty acid (NEFA) concentration and calving score than the multiparous cows. The recovery rate of serum calcium (Ca) after calving was slow in the cows in the parity 6. The DMI was positively affected by the serum Ca concentration after calving, milk yield, and prepartum serum TP concentration and was negatively affected by the calving score and prepartum serum NEFA concentration. We conclude that the DMI immediately after calving tends to be lower in first‐lactation heifers and high‐parity number cows, but factors that reduce the DMI differ according to parity number.  相似文献

As a result of research conducted in the US, recommendations for dry cow vitamin E intakes have increased seven fold there, however there has been no change to recommendations in the UK. As part of a larger study comparing the impact of existing UK and new US recommended vitamin E intakes on the health and fertility of commercial dairy cows in the UK, a study was set up to investigate the effect of route of supplementation and stage of lactation, over a 21 day period, on the response to mega-supplementation of cattle receiving supposedly adequate vitamin E. The study assessed the response of dry, peak lactation and mid lactation cows to in-feed or parenteral vitamin E supplementation (7 animals per treatment/lactation stage group) by measuring plasma and milk vitamin E concentrations, blood glutathione peroxidase (GSH-Px) activity and milk yields over a 21 day period. Plasma vitamin E concentrations were significantly influenced by a time, stage and treatment interaction (P = 0.046). Both dry and lactating animals had significantly higher plasma vitamin E concentrations at some time points in the parenteral supplemented cows compared to the in-feed supplementated animals (P ≤ 0.011 and P < 0.01, respectively). Milk vitamin E concentrations did not significantly differ between lactation stages but treatment had a significant effect on concentrations (P < 0.008) when lactation stage was removed from the model. There was no significant difference in milk yield between treatment groups. A significant relationship between plasma and milk vitamin E concentrations was only found in the parenterally supplemented cows (r = 0.435, P < 0.001). In cattle with intakes greater than the ARC recommendations, measurement of plasma vitamin E concentration may be of limited value in determining whether there has been a response to supplementation. The relationship between plasma and milk vitamin E concentrations is too poor for milk vitamin E concentrations to be used as a proxy for plasma vitamin E.  相似文献

The objective of this study was to evaluate the effects of malic acid (MA) on feed intake, milk yield and composition, blood metabolites and energy balance in early lactation Holstein dairy cows from 1 to 63 day in milk (DIM). Twenty-eight multiparous Holstein dairy cows, blocked by lactation number, previous 305-d mature equivalent milk production, and expected calving date, were arranged into four groups in a randomized block design. Treatments were: control (without MA), LMA, MMA and HMA with 70, 140 and 210 g malic acid per cow per day, respectively. The supplement of food grade MA (99.8% of MA) was hand-mixed into the top one-third of the daily ration. Cows were fed ad libitum a total mixed ration consisting of equal proportion of forage and concentrate. Milk yield increased (P = 0.04), but feed intake and milk components were not affected (P > 0.05) by MA supplementation. The energy balance, expressed as the difference between energy input and output, tended to be higher (P = 0.08) for MA supplemented cows during the 63-DIM period MA and supplemented cows showed a trend (P = 0.07) toward less loss of BW during the 63-day period, especially during the first 21-day of lactation. Concentrations of plasma glucose and serum insulin were higher for cows fed LMA, MMA, and HMA relative to control and linearly (P < 0.01) increased with increasing MA supplementation. Concentrations of non-esterified fatty acids (NEFA), beta-hydroxybutyrate (BHBA) and urine ketones were lower for MA-supplemented cows at 7, 14 and 21 DIM of lactation and linearly (P < 0.01) decreased with increasing MA supplementation. Although feed intake was not affected, milk yield increased, MA-supplemented cows experienced higher concentrations of plasma glucose and serum insulin, lower concentrations of plasma BHBA and NEFA, and lower concentrations of urine ketones, suggesting that nutrient digestibilities and energy availability may have been improved.  相似文献

The objective of this study was to investigate the influence of orally administered biotin on the healing of uncomplicated sole ulcers in dairy cattle. In a double-blind controlled study, 24 dairy cows with a mild, uncomplicated sole ulcer on a lateral hind claw were given either 40 mg biotin per day or a placebo feed over a period of 50 days. An orthopaedic shoe was fitted to the medial claw of the affected foot. The healing process was assessed clinically and by histological examination of horn samples. In the biotin-treated animals, the newly formed epidermis covering the sole ulcers was found to be of significantly better histological quality after 50 days than at the start of the study. The significant improvement in histological horn quality found in the biotin-treated animals suggests that biotin exerts a positive influence on the healing of sole ulcers, however the study period of 50 days appears to have been too short to permit macroscopic detection of the improvement in horn quality.  相似文献

OBJECTIVE: To examine the relationship between lameness and milk yield in dairy cows. DESIGN: Cohort study. ANIMALS: 531 dairy cows. PROCEDURE: Cows affected with lameness were classified into 1 of 3 groups on the basis of type of diseases or lesions observed, including interdigital phlegmon (foot rot), papillomatous digital dermatitis (foot warts), or claw lesions. Cows not affected with lameness were classified as healthy. From Dairy Herd Improvement Association records, 305-day mature equivalent milk yield data were collected at the end of lactation or when the cow left the herd. Milk yield was compared between cows affected with lameness and healthy cows. RESULTS: 167 (31%) cows were affected with lameness during lactation. Lame cows had claw lesions (60%), papillomatous digital dermatitis (31%), or interdigital phlegmon (9%). Milk yield in lame cows with interdigital phlegmon (mean, 17,122 lb) was significantly less, compared with healthy cows (19,007 lb). CONCLUSIONS AND CLINICAL RELEVANCE: In this herd, interdigital phlegmon was associated with a 10% decrease in milk production. Lame cows with claw lesions or papillomatous digital dermatitis produced less milk than healthy cows, but the difference was not significant.  相似文献

OBJECTIVE: To use threshold concentrations of acetone and beta-hydroxybutyrate in milk and serum, respectively; identify risk for ketosis and endometritis; and assess analyses of blood and milk samples as predictors of risk for ketosis in high-yielding dairy cows. ANIMALS: 90 multiparous Holstein cows. PROCEDURE: At intervals before and after parturition, blood samples were obtained for determination of glucose, nonesterified fatty acids, leptin, insulin, insulin-like growth factor-1, and beta-hydroxybutyrate concentrations. Samples of milk were obtained at similar intervals after parturition for determination of fat content and concentrations of acetone, protein, and lactose. Reproductive examination of each cow was performed weekly. RESULTS: For each cow, threshold concentrations of acetone and beta-hydroxybutyrate were calculated as 75th and 90th percentiles of maximum postpartum concentrations of acetone in milk (0.40 and 0.87 mmol/L) and beta-hydroxybutyrate in serum (2.30 and 3.51 mmol/L). Significant decrease in milk production (442 to 654 kg of energy-corrected milk/305-day period per cow) was associated with acetone or beta-hydroxybutyrate in excess of threshold values. Milk acetone concentrations > 0.40 mmol/L were associated with 3.2 times higher risk for endometritis. Low plasma glucose, high serum beta-hydroxybutyrate, and high milk acetone concentrations during week 1 after parturition were indicators of increased risk for ketosis later during lactation. CONCLUSIONS AND CLINICAL RELEVANCE: Determination of milk acetone concentration during the week after parturition may identify cows at risk for ketosis and endometritis; with appropriate interventions, development of disease and production losses may be reduced.  相似文献

The objective was to evaluate the effect of body condition score (BCS) at 30 days before calving (−30 days) induced by a differential nutritional management, parity and week of lactation (WOL) on milk yield and composition, and milk casein and fatty acid composition. Primiparous and multiparous Holstein cows with high BCS (PH, n = 13; MH, n = 9) and low BCS (PL, n = 9; ML = 8) under grazing conditions were sampled at WOL 2 and 8 (before and after peak of lactation). Milk yield was greater in multiparous than in primiparous cows and tended to decrease from WOL 2 to 8 only in ML cows. Milk protein, fat and casein yields were greater in multiparous than in primiparous cows and decreased from WOL 2 to 8. Milk casein concentration in milk protein was greater in MH cows than in ML, PH and PL cows at WOL 2. Milk κ‐casein was greater, and β‐casein was less in multiparous than in primiparous cows. As lactation progressed, proportion of casein fractions were not altered. Only κ‐casein fraction was affected by BCS at −30 days as PL showed a higher concentration than PH. The de novo (4:0–15:1) and mixed‐origin fatty acids (16:0–16:1) in milk fat increased, whereas preformed fatty acids (≥17:0) decreased from WOL 2 to 8. Saturated (SAT) fatty acids tended to be greater and monounsaturated fatty acids (MUFA) were less in multiparous than in primiparous cows. High‐BCS cows had greater concentrations of polyunsaturated (PUFA), conjugated linoleic acid (CLA) as well as n‐6 and n‐3 fatty acids in milk fat than low‐BCS cows. The results indicate that casein and fatty acid fractions in milk were affected by parity and may be modified by a differential nutritional management during the pre‐calving period (BCS at −30 days) in cows under grazing conditions.  相似文献

The aim of this study, which was part of the EU-financed project Life Ammonia, was to evaluate the effects of dietary components and milk production on nitrogen efficiency of dairy cows. The study included examining the effects of decreased crude protein (CP) concentration in a grass-clover silage based diet and results of mixing whole-crop barley silage (WCBS) with grass-clover silage in the diet, on feed intake, milk production and nitrogen efficiency. Rations were formulated and milk production data were registered individually each month for 42 cows of the Swedish Red Cattle breed during four indoor periods from 1999 to 2003. The range in nitrogen efficiency by the cows, 11 to 398 days in milk, was 18 to 40%, when fed a diet containing 135 to 184 g CP/kg DM, 44 to 56% of NDF as rumen degradable fibre (RDF) and milking 13 to 57 kg of ECM daily. The average CP concentration of the diet, containing mainly grass-clover silage and concentrate, was decreased from 168 g/kg DM (170 g in early lactation) in the control treatment period to 160 g/kg DM (163 g in early lactation) during the following treatment period. The CP concentration was 170 g/kg DM (171 g in early lactation) during the third treatment period, when the grass-clover silage was fed in a mixture with WCBS. Using the whole data set (n = 284 for primiparous, n = 440 for multiparous cows based on measurements each month) resulted in models, in which total DM intake, ECM yield, dietary CP concentration and RDF were the most important factors affecting nitrogen utilisation of primiparous and multiparous cows. Increases in both average DM intake and milk yield by multiparous cows and no changes in average intake and milk yield by primiparous cows fed the low CP diet or the normal CP diet containing WCBS, compared to cows fed the normal CP diet, resulted in similar nitrogen efficiencies among the treatments. Hence, dietary CP concentrations of 160 to 170 g/kg DM can be used for cows in early lactation in commercial herds to improve nitrogen utilisation without causing a simultaneous decrease in milk yield.  相似文献

This study was designed to evaluate serum bile acid measurements as indicatory, of liver function and/or hepatic fat infiltration in dairy cattle. Serum bile acid concentrations were measured in healthy dairy cattle at different stages of lactation after fasting or feeding. Bile acid concentrations were compared with liver fat content and sulfobromophthalein (BSP) half-life (T 1/2). Serum bile acid concentrations were higher in cows in early lactation and with higher daily milk production. Compared with prefasting values, bile acid concentrations were decreased at 8,14, and 24 hours of fasting. Blood samples from fed cows at 1 - to 2-hour intervals had wide and inconsistent variations in bile acid concentration. Because serum bile acids correlated well with BSP T 1/2, it is suggested that both measurements evaluate a similar aspect of liver function. Neither bile acids nor BSP T I correlated with differences in liver fat content among cows. Because of large variability in serum bile acid concentrations in fed cows and the lack of correlation of measured values with liver fat content, bile acid determinations do not appear useful for showing changes in hepatic function in fed cows with subclinical hepatic lipidosis nor serve as a screening test for this condition.  相似文献
