Background: In ruminants, dietary C18:3 n-3 can be lost through biohydrogenation in the rumen; and C18:3 n-3 that by-passes the rumen still can be lost through oxidation in muscle, theoretically reducing the deposition of C18:3 n-3,the substrate for synthesis of poly-unsaturated fatty acids(n-3 LCPUFA) in muscle. In vitro studies have shown that rumen hydrogenation of C18:3 n-3 is reduced by supplementation with palm oil(rich in cis-9 C18:1). In addition, in hepatocytes, studies with neonatal rats have shown that cis-9 C18:1 inhibits the oxidation of C18:3 n-3. It therefore seems likely that palm oil could reduce both rumen biohydrogenation of C18:3 n-3 and muscle oxidation of C18:3 n-3. The present experiment tested whether the addition of palm oil to a linseed oil supplement for goat kids would prevent the losses of C18:3 n-3 and thus improve the FA composition in two muscles, Longissimus dorsi and Biceps femoris. To investigate the processes involved, we studied the rumen bacterial communities and measured the mRNA expression of genes related to lipid metabolism in Longissimus dorsi. Sixty 4-month-old castrated male Albas white cashmere kids were randomly allocated among three dietary treatments. All three diets contained the same ingredients in the same proportions, but differed in their fat additives: palm oil(PMO), linseed oil(LSO) or mixed oil(MIX; 2 parts linseed oil plus 1 part palm oil on a weight basis).Results: Compared with the LSO diet, the MIX diet decreased the relative abuandance of Pseudobutyrivibrio, a bacterial species that is positively related to the proportional loss rate of dietary C18:3 n-3 and that has been reported to generate the ATP required for biohydrogenation(reflecting a decrease in the abundance of rumen bacteria that hydrogenate C18:3 n-3 in MIX kids). In muscle, the MIX diet increased concentrations of C18:3 n-3, C20:5 n-3, C22:6 n-3,and n-3 LCPUFA, and thus decreased the n-6/n-3 ratio; decreased the mRNA expression of CPT1β(a gene associated with fatty acid oxidation) and increased the mRNA expression of FADS1 and FADS2(genes associated with n-3 LCPUFA synthesis), compared with the LSO diet. Interestingly, compared to Longissimus dorsi, Biceps femoris had greater concentrations of PUFA, greater ratios of unsaturated fatty acids/saturated fatty acids(U/S), and poly-unsaturated fatty acids/saturated fatty acids(P/S), but a lesser concentration of saturated fatty acids(SFA).Conclusions: In cashmere goat kids, a combination of linseed and palm oils in the diet increases the muscle concentration of n-3 LCPUFA, apparently by decreasing the relative abundance of rumen bacteria that are positively related to the proportional loss rate of dietary C18:3 n-3, by inhibiting m RNA expression of genes related to C18:3 n-3 oxidation in muscle, and by up-regulating m RNA expression of genes related to n-3 LCPUFA synthesis in muscle,especially in Longissimus dorsi.  相似文献

Many compounds have been screened for their potential anti-cryptosporidial activity in ruminants but none of them has been totally efficient in controlling the disease. A product containing activated charcoal and wood vinegar liquid demonstrated a good efficacy in controlling clinical signs and oocyst excretion in calves experimentally infected. This product (Obionekk(?), Obione, Charentay, France) was given to goat kids in field conditions. The product was administered in a preventive way at the dose rate of 3.75 g/day/kid of Obionekk(?) from the age of 3 days to the age of 17 days in suspension in milk replacer twice or three times a day. A significant reduction of oocyst excretion was observed when the product was given 3 times a day when compared to control group. The clinical signs (diarrhoea) were significantly reduced whatever the rhythm of distribution when compared to control group. These results demonstrated the efficacy of Obionekk(?) in preventing cryptosporidiosis in goat kids in field conditions.  相似文献

Larval development, mucosal mast cell (MMC) and eosinophil responses in worm-nai;ve lambs, yearling goats and goat kids were compared using two different experimental challenge regimes involving oral administration of infective Teladorsagia circumcincta L(3). Experimental challenge regimes enabled primary and secondary immune responses in the two species to be compared. Goats carried higher worm burdens than lambs and there were significant differences in the stages of development attained by the larval challenge that established in the two species. Possible physiological reasons for these differences are discussed. There were also differences in the establishment and development of larvae in individual yearlings which may indicate the development of a weak age-related immune response. Quantitative analysis of MMC and globule leukocyte (GL) recruitment and functional activity in the form of mast cell-specific proteinase (MCP) production demonstrated differences between the species with goat tissues containing significantly higher numbers of GL and lower concentrations of MCP than the lambs. Quantitative analysis of blood and tissue eosinophil responses failed to demonstrate any significant differences in either species under the two challenge regimes.  相似文献

To be successful, initiatives to improve smallholder’s goat production should directly address the needs and objectives of the keepers while promoting rational use of local genetic resources. This paper identifies the objectives, constraints and needs of goat farmers in southern Benin and discusses their relevance to the development of improvement programmes. Between November and December 2005, structured questionnaires, focus group discussions and participant observation were used to collect information from 38 goat farmers in two selected locations. Goats were kept mainly for sale whenever cash was needed. Traits related to reproduction, to behaviour, to health and to meat production were considered equally important and were ranked very highly by goat keepers. Increased net income per flock through increased number of marketable animals is the derived breeding objective from the trait analysis. Disease outbreaks resulting in high mortality, poor housing, and feed shortages were, in descending order, the most important problems. It was concluded that the development of initiatives to improve management practices is an overriding priority. It will lead to increases in productivity in the short term and foster farmers’ participation in the development of long-term improvement strategies, which should include selection and controlled mating.  相似文献

Gastrointestinal helminthiasis represents an obstacle to goat raising, causing severe damage to herds such as growth retardation, weight loss, and even death. In this study, we aimed to determine the prevalence and risk factors associated to goat gastrointestinal helminthiasis in the Sertão region of Paraíba State, Brazil. A total of 256 goats from 54 farms were systematically sampled. Blood and fecal samples were collected from each animal for egg per gram (EPG), larval culture, and packed cell volume (PCV) analyses. We found that 79.3 % of the goats investigated were parasitized with gastrointestinal helminths. Significant correlation (p?=?0.004) was observed between the EPG and PCV of the animals studied, and it was observed that the EPG increases as the PCV decreases. In the larval culture, the most prevalent helminth was Haemonchus sp. (83.2 %). Age and sex were significant variables (p?≤?0.20) for the development of gastrointestinal helminths: 86.8 % of animals over 36 months of age and 81.7 % of females were infected. The variable type of animal exploitation was also significant, with 90.3 % (p?≤?0.20) of the animals presenting double suitability (milk and meat). The Sertão region of Paraíba State presents high prevalence of gastrointestinal helminthiasis in goats, and age and type of animal exploitation are the most relevant risk factors to the development of these parasites.  相似文献

Rearing of goats under communal farming conditions is characterised by high kid mortality and low weaning percentages. A survey was conducted to determine farmers’ perceptions on the causes of kid mortality during summer under the communal farming system in Nkonkobe Local Municipality in the Eastern Cape, South Africa. This was done by administering questionnaires to a total of 162 respondents in 14 villages around Nkonkobe Local Municipality. The study showed that majority of farmers (75 %) keep flock sizes of less than 10 goats and kids, and this indicates that goat production in Nkonkobe Local Municipality is suppressed. According to the farmers, diseases (89 %), endo-parasites (72 %) and ecto-parasites (68 %) were perceived as the major causes of kid mortality. Other causes reported include starvation (15 %), extreme weather conditions (28 %), abortion (7 %), theft (35 %), diarrhoea (43 %), accidents (10 %) and wounds (9 %). The low number of goats could be attributed to high mortalities. It was also found that all causes reported by farmers played a role in high kid mortality in Nkonkobe Local Municipality. However, the causes which require more emphasis to formulate extension support were tick-borne diseases and parasites. This study provided baseline information on possible causes of kid mortalities in Nkonkobe Local Municipality. There is, however, a need to conduct further studies to determine actual causes of high kid mortalities so as to develop preventive strategies that would minimize kid mortality for good economic returns.  相似文献

The objective of this study was to compare the effects of the type of Haemonchus contortus experimental infection (trickle infection, TI versus single infection, SI) on feed intake, nutrients digestibility, parasitological and haematological measures, and plasma leptin in Creole kids. The animals were infected over 2 periods (challenge 1 and challenge 2) of 6 weeks each, corresponding respectively to the primary and the secondary infection. Periods prior infection (1 week each) were considered as controls. The primary infection was realized with 35 Creole kids (18.40 ± 3.76 kg BW) housed in individual boxes and fed a hay-based diet. The secondary infection continued with 29 kids (21.90 ± 3.40 kg BW) from the initial 35. A total of 6 kids and 8 kids were slaughtered for measuring nematode burden at the end of the primary and the secondary infection, respectively. Measurements of nutrients digestibility were made at 0, 3 and 5 weeks post-infection for both challenges. Faecal egg count (FEC), blood eosinophilia and packed cell volume (PCV) were monitored weekly. Feed intake (dry matter intake, DMI) and nutrients digestibility were negatively affected by H. contortus infection only during the primary infection. Plasma leptin changed significantly over time (P = 0.0002) but was not affected by the infection type. Effect of infection type was observed only on crude protein digestibility during the primary infection, which was lower in the TI group (P < 0.01). The overall level of blood eosinophilia was significantly higher in the TI group (P < 0.0001) during both challenges. The overall FEC mean was significantly higher in the SI compared with the TI groups, during both challenges (P < 0.02). These results were related to the mean female length significantly higher in the SI group compared with the TI group during challenge 1 (P = 0.004), and the number of adult nematode significantly lower in the TI group compared with the SI group during the challenge 2 (P = 0.05). The results showed that the response of Creole kids to H. contortus experimental infection was in part dependent on the type of experimental infection. Our data suggest that plasma leptin would not be involved in the response of Creole kids against H. contortus infection, as no relationship between its plasma level and the transient reduction in voluntary feed intake observed in both groups during the primary infection was observed.  相似文献

The development of a bacteria-inducible expression system has several advantages compared with persistent expression of anti-bacterial proteins in milk to prevent and treat mastitis. The present study determined whether mastitis responsive promoters could regulate enhanced green fluorescent protein (EGFP) expression in goat mammary epithelial cells (GMECs) in response to challenges with Escherichia coli, Staphylococcus aureus or Streptococcus agalactiae. The level of expression of interleukin (IL)-1α was significantly increased in GMECs challenged with E. coli, S. aureus or S. agalactiae compared with untreated GMECs. IL-1β was induced by E. coli and S. aureus, while Toll-like receptor 2 (TLR2) was induced by E. coli only.GMECs were transfected with IL-1α, IL-1β and TLR2 promoter-EGFP reporter gene lentiviral expression vectors and the levels of expression of EGFP were measured by flow cytometry and Western blot analysis after bacterial challenge. EGFP expression driven by the IL-1α and IL-1β promoters was higher in GMECs challenged with E. coli, S. aureus or S. agalactiae than in untreated GMECs. There were no differences in EGFP expression driven by the TLR2 promoter between GMECs challenged with S. aureus or S. agalactiae and untreated GMECs, but EGFP expression was significantly increased in GMECs challenged with E. coli. Overall, these results indicate that the promoters of some bacteria-inducible genes can regulate EGFP expression in GMECs in response to bacterial challenges. This bacteria-inducible expression strategy could be used for production of mastitis resistant animals by regulating the expression of anti-bacterial proteins in the mammary gland.  相似文献

The aim of this study was to determine the fattening performance and the slaughter and carcass characteristics of Akkeçi (Saanen × Kilis (B1) crossbred) male kids raised in individual crates (IC) or in group pen housing (GP). In both GP and IC groups, total 13 heads of male kids, whose initial body weights were 21.43 ± 1.03 kg and 20.61 ± 0.98 kg, respectively, evaluated for their fattening performances. After 56 days of the fattening period, the final body weights and average daily weight gains of the GP and IC kids were recorded as 30.84 ± 0.82 kg and 25.84 ± 0.76 kg; 182.42 ± 14.77 g and 92.09 ± 13.76 g, respectively (P < 0.001). Adrenal gland weights of the GP and IC groups were similar as 1.66 ± 0.25 g and 1.77 ± 0.22 g, respectively. Although, the carcass conformation of kids was similar between groups, there were significant differences between GP and IC groups for some slaughter and carcass characteristics. In general, although housing kids reared in individual crates provides opportunity to individually feed and monitor a kid, the results obtained from the individual crates in the present study were not satisfactorily from the ethological and production points of view.  相似文献

Summary

The fertility of ten West African Dwarf goats in which oestrus was induced with PGF2α was comparable to that of the five West African Dwarf goats in the control group. Performance indices as determined by duration of oestrus and length of gestation were not affected. Similarly the birth weight of kids was not significantly influenced. However, treated goats were superior to controls when the interval from commencement of experiment to oestrus is considered, although there was no difference between the two treated groups of goats (5 mg vs 10 mg PGF2α).

Results showed that PGF2α could be an important tool in oestrus synchronization and thus in control of the reproductive performance of the West African Dwarf goats under humid lowland tropical environmental conditions.  相似文献

Caprine brucellosis is a bacterial zoonotic infection affecting goats especially in developing countries all over the world. In Malaysia, the risk factors associated with this infection in farms have not been studied. A case–control study was carried out in goat farms in four states of Malaysia to elucidate the risk factors associated with the infection on the farms using structured questionnaires and face-to-face interviews. Results indicate that the introduction of new animals (OR?=?5.25; 90 % CI?=?1.46, 18.88), younger age category of farms (OR?=?5.53; 90 % CI?=?1.09, 21.66), and farms with single breed of goats (OR?=?8.50; 90 % CI?=?1.27, 41.97) were significant risk factors for brucellosis. In order to control brucellosis or possibly eradicate it in goat farms, these factors need to be dealt with. Enforcing stringent importation protocols or complete ban of goat importation from brucellosis endemic countries will help reduce risk of introducing new infection into the country.  相似文献

One hundred and fifty West African Dwarf (WAD) kids were tested at the National Goats Breeding Station to determine the effects of some neonatal factors on their vigour levels within the first 24 h post-partum. The kids were also tested to establish the relationship between maternal weight, rectal temperature, times of first standing and sucking, sucking period and vigour. The distance covered (in meters) by a kid towards its mother during a 5-min test period was considered as vigour trait. Vigour levels increased significantly (p?<?0.05) with increasing age within the first 24 h post-partum. Kids born to second parity mothers had higher (p?<?0.05) vigour than those born to first parity mothers. Similarly, high birth weight kids had higher (p?<?0.05) vigour levels than low birth weight kids. Weak positive and negative correlations (p?<?0.05), respectively, were found between vigour and rectal temperature, and vigour and the time of first successfully standing. However, there was moderate positive correlation (p?<?0.01) between vigour and sucking period. The time it took for the newborn kid to stand up for the first time also correlated weakly and negatively (p?<?0.01) with the duration of sucking by the newborn within the first 5 min of accessing the teat. Vigour within the first 24 h post-partum in WAD kids increased with increasing age, parity and birth weight and might be responsible for the earlier and longer sucking time and periods, respectively, in high birth weight kids and those dropped by multiparous does.  相似文献

Background

The study aimed at examining hypoxia-inducible factor (HIF)1α expression in adenocarcinomas and adenomas in bitches in regard to tumour malignancy grade, proliferation, apoptosis and vascularisation. Therefore, paraffin sections of 15 adenomas and 64 adenocarcinomas sampled from 79 dogs aged 6 to 16 years were analysed.

Results

A significantly higher HIF-1α expression was noted in adenocarcinomas in comparison to adenomas (P < 0.0004). Moreover, HIF-1α expression in adenocarcinomas correlated positively with tumour malignancy grade (r = 0.59, P < 0.05), Ki-67 antigen expression (r = 0.43; P < 0.0005), TUNEL-positive cells (r = 0.62, P < 0001) and tumour vascularity measured by quantification of vessels characterized by the expression of von Willebrand Factor (r = 0.57, P < 0.05).

Conclusion

Results of this study indicate a similar biological role of HIF-1α in dogs and in humans, which may confirm suitability of the animal model in investigations on progression of tumours in humans.  相似文献

Summary

The disposition of sulphadimidine (SDM) and of its N4‐acetyl (N4‐SDM) and two hydroxyl metabolites, 6‐hydroxymethyl‐ (SCH2OH) and 5‐hydroxyasulphadimidine (SOH), was studied in plasma and milk of dairy cows following intramuscular or intravenous administration of sulphadimididine‐33.3% at doses of 10, 45, 50, and 100 mg/kg. The main metabolite in plasma as well as in milk was SCH2OH. The metabolite percentages, the final plasma elimination half‐lives, and the time of peak SDM concentrations in milk are presented for different dosages. The concentrations of SDM and its metabolites in milk ran parallel to those in plasma beyond4 hours p.i. The metabolite concentrations in plasma and milk were lower than those of the parent SDM. Sulphate and glucuronide metabolites could not be detected in milk.

At high doses (45 mg/kg or more) and SDM plasma concentrations exceeding 20 μg/ml, a capacity limited metabolism of SDM to SCH2OH was noticed, viz, asteady state concentration of SCH2OH and a biphasic elimination pattern for SDM and SCH,OH in plasma and milk.

The mean ultrafiltrate ratios of the milk to plasma concentrations with respect to SDM, SCH2OH, SOH, and N4‐SDM were: 0.69, 0.22, 020, and 0.63, respectively.

The total amount of SDM and its metabolites recovered from the milk after milking twice daily over the whole experimental time was less than 2% of the applied dose.

A bioassay method allowed of detecting qualitatively SDM concentrations exceeding 0.2 μg/ml in plasma or milk. Withholding times for edible tissues and milk are suggested.  相似文献

At present,China's traditional agriculture and animal husbandry has no way to rely on the "fine fodder + straw" feeding model. Only relying on grass and animal husbandry can guarantee China's consumer demand and food safety. However, due to the restraints such as policy, infrastructure, technology, market and organizational factors, the "lacking" status of China's pasture industry is not optimistic. In this paper, we analyze the pasture production, commercial grass production, grass seed production,forage product trade, the import and export volume of different types of pasture products, the current status of pastoral products trading partners and the causes behind it in depth. And concluded that the supply of forage products in China needs to be imported. Imports are growing while exports are generally declining,the pasture trade market is highly concentrated while grassland trading partners are dispersed. Five suggestions are given to provide theoretical guidance for the development of China's pasture industry: increase policy support, promote infrastructure, increase investment in scientific research, strengthen market construction, and develop industrial organizations. © 2019 China Agricultural University.
