Objective
To evaluate replicate effects and test–retest reliability of mechanical and thermal quantitative sensory testing (QST) in normal dogs and dogs with osteoarthritis (OA)-associated pain.Study design
A prospective clinical study.Animals
A total of 54 client owned dogs (OA, n = 31; controls, n = 23).Methods
Mechanical [electronic von Frey (EVF) and blunt pressure] and thermal (hot and cold) sensory thresholds were obtained in dogs with OA-associated pain and control dogs at two visits, 7 days apart, to assess test–retest reliability. Thresholds were measured at the OA-affected joint (hip or stifle), over the tibial muscle and over the midpoint of the metatarsals. Five replicates were obtained for each modality at each site bilaterally.Results
Overall, there was no significant effect of replicates on QST response. EVF thresholds were significantly lower at the second visit in OA dogs at the affected and metatarsal sites (p = 0.0017 and p = 0.0014, respectively). Similarly for control dogs, EVF thresholds were significantly lower at the second visit at the metatarsal site (p = 0.001). Significantly higher hot thermal latencies were seen in OA dogs at the affected and tibial testing sites (p = 0.014 and p = 0.012, respectively), and in control dogs at the tibial site (p = 0.004).Conclusions
In QST, a replicate does not show a strong effect. However, QST results show variability over time, particularly for EVF and hot thermal stimuli.Clinical relevance
If QST is to be used clinically to evaluate a sensitized state, the variability over time needs to be accounted for in the study design. 相似文献2.

Objective
Variants in the MC1R gene have been associated with red hair color and sensitivity to pain in humans. The study objective was to determine if a relationship exists between MC1R genotype and physiological thermal or mechanical nociceptive thresholds in Labrador Retriever dogs.Study design
Prospective experimental study.Animals
Thirty-four Labrador Retriever dogs were included in the study following public requests for volunteers. Owner consent was obtained and owners verified that their dog was apparently not experiencing pain and had not been treated for pain during the previous 14 days. The study was approved by the Institutional Animal Care and Use Committee.Methods
Nociceptive thresholds were determined from a mean of three thermal and five mechanical replications using commercially available algometers. Each dog was genotyped for the previously described MC1R variant (R306ter). Data were analyzed using one-way anova with post hoc comparisons using Tukey’s test (p < 0.05).Results
Thirteen dogs were homozygous wild-type (WT/WT), nine were heterozygous (WT/R306ter), and eight were homozygous variant (R306ter/R306ter) genotype. Four dogs could not be genotyped. A significant difference (p = 0.04) in mechanical nociceptive thresholds was identified between dogs with the WT/WT genotype (12.1 ± 2.1 N) and those with the WT/R306ter genotype (9.2 ± 2.4 N).Conclusion
A difference in mechanical, but not thermal, nociceptive threshold was observed between wild-type and heterozygous MC1R variants. Differences in nociceptive thresholds between homozygous R306ter variants and other genotypes for MC1R were not observed.Clinical relevance
Compared with the wild-type MC1R genotype, nociceptive sensitivity to mechanical force in dogs with a single variant R306ter allele may be greater. However, in contrast to the reported association between homozygous MC1R variants (associated with red hair color) and nociception in humans, we found no evidence of a similar relationship in dogs with the homozygous variant genotype. 相似文献3.

Objective
To test the efficacy of intraoperative intrafragmentary administration of bupivacaine (haematoma block) in controlling postoperative pain in dogs undergoing osteosynthesis of long-bone isolated diaphyseal fractures.Study design
Randomized, ‘blinded’, placebo-controlled, prospective study.Animals
A total of 23 client-owned dogs with isolated long-bone fractures.Methods
Dogs were allocated randomly to two groups: bupivacaine group (B) or placebo group (P). Group B dogs (n = 11) were administered an intraoperative intrafragmentary injection of 0.5% bupivacaine (1.1 mg kg–1) just before fracture fixation, whereas group P dogs (n = 12) were administered normal saline. Postoperative pain evaluations using the University of Melbourne Pain Scale (UMPS) and algometer were performed upon arrival to the recovery room and 1, 2, 4, 6, 8, 20 and 32 hours later. Algometer measurements were performed on: the incision site, a healthy region near the fracture line and the contralateral healthy limb. When the pain score exceeded 14 points in the UMPS, rescue analgesia was administered. The time-standardised area under the curve (AUCst) was used to compare UMPS scores and mechanical pain thresholds between the two groups.Results
None of the group B dogs required rescue analgesia, whereas eight of the 12 group P dogs did (p = 0.001). The pain threshold AUCst at the incision line was higher in group B [16.3 (2.9–41.6) N] than in group P [5.6 (2.5–17.4) N] (p = 0.029). The mean UMPS score AUCst was lower in group B (3.7 ± 1.8) than in group P (9.4 ± 4.6) (p = 0.016). In a small number of animals of both groups that were evaluated radiologically, adequate bone healing was noted.Conclusions and clinical relevance
An intraoperative bupivacaine haematoma block is a simple, quick and effective method that can be used to aid in postoperative pain control in dogs submitted to long-bone osteosynthesis. 相似文献4.
Objectives
To determine the prevalence of mitral valve regurgitation (MR) in asymptomatic Swedish Norfolk terriers.Animals
Seventy-nine privately owned Norfolk terriers.Materials and methods
A prospective observational study was conducted where dogs were recruited via the Swedish Norfolk terrier club. All dogs were examined using the same protocol including physical examination and Doppler echocardiography.Results
Fifteen dogs (19%) had a murmur at the time of the examination. A total of 35 dogs (44%) had MR, including 23 dogs (29%) with both MR and tricuspid valve regurgitation and 12 dogs (15%) with MR only, identified on Doppler echocardiography. In addition, 7 dogs (9%) had tricuspid valve regurgitation only. The prevalence of MR increased with increasing age (p < 0.0001).Conclusions
Mitral valve regurgitation is common in asymptomatic Norfolk terriers with and without murmurs and the prevalence increases with age. The impact of MR in this breed on survival remains to be elucidated by a longitudinal study. 相似文献5.

Objective
To investigate the expression and distribution of desmosomal and gap junction proteins of the intercalated disc in the atria of boxers with arrhythmogenic right ventricular cardiomyopathy (ARVC).Animals
Nineteen control dogs and 13 boxers with histopathologically confirmed ARVC.Methods
Right and left atrial samples were examined using immunofluorescence and Western blots. The intercalated disc proteins investigated included total and phosphorylated connexin43 (Cx43 and pCx43), connexin45, connexin40, plakoglobin, plakophilin-2, desmoplakin, and N-cadherin.Results
Histopathological changes characteristic of ARVC were present in the left or right atrium of 12 out of 13 boxers and were absent in all control dogs. When compared to the 19 control dogs, immunofluorescence analysis revealed a decrease in signal intensity for pCx43 and plakoglobin in the left (p = 0.03 and p = 0.014, respectively) and right atrium (p = 0.015 and p = 0.002, respectively) of affected boxers. Connexin43 and pCx43 Western blot band density was significantly decreased in the left (p = 0.025 and p = 0.027, respectively) and right atrium (p = 0.001 and p = 0.044, respectively) of affected boxers.Conclusion
Altered intercalated disc and gap junction proteins were identified in atrial myocardium of ARVC boxers, supporting atrial involvement as part of this disorder. Reduction in pCx43 in conjunction with histological changes could represent the substrate for atrial arrhythmias associated with ARVC. Furthermore, these findings detected in boxer dogs, lend support for the broader term, arrhythmogenic cardiomyopathy, as preferred nomenclature used to describe this disease in humans. 相似文献6.

Objective
To investigate whether pulse pressure variation (PPV) can predict fluid responsiveness in healthy dogs during clinical surgery.Study design
Prospective clinical study.Animals
Thirty-three isoflurane-anesthetized dogs with arterial hypotension during orthopedic surgery.Methods
Fluid challenge with lactated Ringer's solution (15 mL kg?1 in 15 minutes) was administered in mechanically ventilated dogs (tidal volume 10 mL kg?1) with hypotension [mean arterial pressure (MAP) < 65 mmHg]. The volume expansion was considered effective if cardiac output (CO; transesophageal Doppler) increased by ≥ 15%. Cardiopulmonary data were analyzed using two-way ANOVA, receiver operating characteristics (ROC) curves and Spearman coefficient; p < 0.05 was considered significant.Results
Effective volume expansion, mean ± standard deviation 42 ± 4% increase in CO (p < 0.0001) was observed in 76% of the dogs, resulting in a decrease in PPV (p < 0.0001) and increase in MAP (p < 0.0001), central venous pressure (CVP; p = 0.02) and ejection fraction (p < 0.0001) compared with before the fluid challenge. None of these changes occurred when volume expansion resulted in a nonsignificant CO increase of 4 ± 5%. No significant differences were observed in blood gas analysis between responsive and nonresponsive dogs. The increase in CO was correlated with the decrease in PPV (r = ?0.65; p < 0.0001) but absolute values of CO and PPV were not correlated. The PPV performance (ROC curve area: 0.89 ± 0.06, p = 0.0011) was better than that of CVP (ROC curve area: 0.54 ± 0.12) and MAP (ROC curve area: 0.59 ± 0.13) to predict fluid responsiveness. The best cut-off for PPV to distinguish responders and nonresponders was 15% (50% sensitivity and 96% specificity).Conclusions and clinical relevance
In mechanically ventilated, healthy, isoflurane-anesthetized dogs, PPV predicted fluid responsiveness to volume expansion, and MAP and CVP did not show such applicability. 相似文献7.
Objective
To compare the running-drip and hanging-drop techniques for locating the epidural space in dogs.Study design

Forty-five healthy dogs requiring epidural anaesthesia.Methods
Dogs were randomized into four groups and administered epidural anaesthesia in sternal (S) or lateral (L) recumbency. All blocks were performed by the same person using Tuohy needles with either a fluid-prefilled hub (HDo) or connected to a drip set attached to a fluid bag elevated 60 cm (RDi). The number of attempts, ‘pop’ sensation, clear drop aspiration or fluid dripping, time to locate the epidural space (TTLES) and presence of cerebrospinal fluid (CSF) were recorded. A morphine–bupivacaine combination was injected after positive identification. The success of the block was assessed by experienced observers based on perioperative usage of rescue analgesia. Data were checked for normality. Binomial variables were analysed with the chi-squared or Fisher’s exact test as appropriate. Non-parametric data were analysed using Kruskal–Wallis and Mann–Whitney tests. Normal data were studied with an anova followed by a Tukey's means comparison for groups of the same size. A p-value of < 0.05 was considered to indicate statistical significance.Results
Lateral recumbency HDo required more attempts (six of 11 dogs required more than one attempt) than SRDi (none of 11 dogs) (p = 0.0062). Drop aspiration was observed more often in SHDo (nine of 11 dogs) than in LHDo (two of 11 dogs) (p = 0.045). Mean (range) TTLES was longer in LHDo [47 (18–82) seconds] than in SHDo [20 (14–79) seconds] (p = 0.006) and SRDi [(34 (17–53) seconds] (p = 0.038). There were no differences in ‘pop’ sensation, presence of CSF, rescue analgesia or pain scores between the groups.Conclusion and clinical relevance
The running-drip method is a useful and fast alternative technique for identifying the epidural space in dogs. The hanging-drop technique in lateral recumbency was more difficult to perform than the other methods, requiring more time and attempts. 相似文献8.

Stroke volume (SV), stroke volume variation (SVV) and cardiac index were measured using the EV device in sevoflurane-anaesthetized, mechanically ventilated dogs undergoing thoracotomies for experimental creation of right ventricular failure. The dogs were considered fluid responsive if stroke volume (SVI; indexed to body weight), measured using pulmonary artery thermodilution, increased by 10% or more after volume loading (10 mL kg–1). Relationships of SVV, central venous pressure (CVP) and pulmonary artery occlusion pressure (PAOP) with SVI were analysed to estimate fluid responsiveness.Results
Better prediction of fluid responsiveness, with a significant area under the receiver operating characteristic curve, was observed for SVV (0.85 ± 0.07; p = 0.0016) in comparison with CVP (0.65 ± 0.11; p = 0.17) or PAOP (0.60 ± 0.12; p = 0.35), with a cut-off value of 13.5% (84% specificity and 73% sensitivity).Conclusions and clinical relevance

Objective
To describe a transorbital approach to the maxillary nerve block in dogs and compare it with a traditional approach.Study design

Heads from 17 euthanized dogs (10 Greyhounds, three Border Collies and four of mixed breed).Methods
A volume of 1 mL of methylene blue dye was injected by each of two techniques, a traditional percutaneous approach and a transorbital approach to the maxillary nerve block. Both techniques were used on each head, alternating the left and right sides after random assignment to the first head. The heads were dissected to reveal the maxillary nerve and the length of nerve stained was measured.Results
There was no significant difference (p = 0.67) in the proportion of nerves stained for a length >6 mm by either technique (88.2% transorbital versus 82.3% percutaneous). The mean length of nerve stained did not differ significantly between the techniques (p = 0.26).Conclusions and clinical relevance
The transorbital approach to the maxillary nerve block described here presents a viable alternative to the traditional percutaneous approach. Further study is required to confirm its efficacy and safety under clinical conditions. 相似文献10.

To compare electrical velocimetry (EV) noninvasive measures of cardiac output (CO) and stroke volume variation (SVV) in dogs undergoing cardiovascular surgery with those obtained with the conventional thermodilution technique using a pulmonary artery catheter.Study design

Seven adult Beagle dogs with a median weight of 13.6 kg.Methods
Simultaneous, coupled cardiac index (CI; CO indexed to body surface area) measurements by EV (CIEV) and the reference pulmonary artery catheter thermodilution method (CIPAC) were obtained in seven sevoflurane-anaesthetized, mechanically ventilated dogs undergoing experimental open-chest cardiovascular surgery for isolated right ventricular failure. Relationships between SVV or central venous pressure (CVP) and stroke volume (SV) were analysed to estimate fluid responsiveness. Haemodynamic data were recorded intraoperatively and before and after fluid challenge.Results
Bland–Altman analysis of 332 matched sets of CI data revealed an overall bias and precision of – 0.22 ± 0.52 L minute?1 m?2 for CIEV and CIPAC (percentage error: 30.4%). Trend analysis showed a concordance of 88% for CIEV. SVV showed a significant positive correlation (r2 = 0.442, p < 0.0001) with SV changes to a volume loading of 200 mL, but CVP did not (r2 = 0.0002, p = 0.94). Better prediction of SV responsiveness (rise of SV index of ≥ 10%) was observed for SVV (0.74 ± 0.09; p = 0.014) with a significant area under the receiver operating characteristic curve in comparison with CVP (0.53 ± 0.98; p = 0.78), with a cut-off value of 14.5% (60% specificity and 83% sensitivity).Conclusions and clinical relevance
In dogs undergoing cardiovascular surgery, EV provided accurate CO measurements compared with CIPAC, although its trending ability was poor. Further, SVV by EV, but not CVP, reliably predicted fluid responsiveness during mechanical ventilation in dogs. 相似文献11.

Objectives
To provide reference intervals for 2-dimensional linear and area-based estimates of left atrial (LA) function in healthy dogs and to evaluate the ability of estimates of LA function to differentiate dogs with subclinical myxomatous mitral valve disease (MMVD) and similarly affected dogs with congestive heart failure (CHF).Animals
Fifty-two healthy adult dogs, 88 dogs with MMVD of varying severity.Methods
Linear and area measurements from 2-dimensional echocardiographs in both right parasternal long and short axis views optimized for the left atrium were used to derive estimates of LA active emptying fraction, passive emptying fraction, expansion index, and total fractional emptying. Differences for each estimate were compared between healthy and MMVD dogs (based on ACVIM classification), and between MMVD dogs with subclinical disease and CHF that had similar LA dimensions. Diagnostic utility at identifying CHF was examined for dogs with subclinical MMVD and CHF. Relationships with bodyweight were assessed.Results
All estimates of LA function decreased with increasing ACVIM stage of mitral valve disease (p<0.05) and showed negative relationships with increasing LA size (all r2 values < 0.2), except for LA passive emptying fraction, which did not differ or correlate with LA size (p=0.4). However, no index of LA function identified CHF better than measurements of LA size. Total LA fractional emptying and expansion index showed modest negative correlations with bodyweight.Conclusions
Estimates of LA function worsen with worsening MMVD but fail to discriminate dogs with CHF from those with subclinical MMVD any better than simple estimates of LA size. 相似文献12.
Objective
To compare the effectiveness of ultrasound- and electrostimulation-guided nerve blocks of the brachial plexus and to determine whether ultrasound guidance is feasible in conscious dogs.Study design

The nerves of the brachial plexus of the right thoracic limb were blocked under ultrasound guidance (UNB) in conscious dogs and under electrostimulation guidance (ENB) in anesthetized dogs with bupivacaine (0.4 mL kg–1, 0.25%). Saline (0.4 mL kg–1) was injected in control animals. Sensory nerve blockade was evaluated by scoring cutaneous sensation in targeted nerves. Motor nerve blockade was evaluated based on weight bearing, conscious proprioception and withdrawal reflex scores. Times to execute the technique in UNB and ENB were compared using t tests (p < 0.05). Scores for sensory and motor nerve blockades in each treatment were compared with scores before treatment and with control treatment scores using nonparametric repeated-measures two-way analysis of variance. Time to onset and duration of sensory nerve block were assessed using scores for four sensory nerve functions. A successful sensory nerve block was defined by decreases in scores for these functions. Success rates of nerve blocks were compared among treatments using McNemar’s test.Results
In UNB and ENB, onset times of sensory nerve blocks were 1 hour and 1.5 hours, respectively. Onset times of motor nerve blocks were 0.5 hour in both treatments. In UNB and ENB, durations of sensory nerve block were 3 hours and 0.5 hour, respectively, and durations of motor nerve block were 7.5 hours and 6.5 hours, respectively. Success rates did not differ between the techniques.Conclusions and clinical relevance
The UNB brachial plexus block had a shorter onset time and longer duration than ENB. UNB can be performed in conscious dogs or those under mild sedation. 相似文献13.

Objective
To compare the effects of thiopentone, propofol and alfaxalone on arytenoid cartilage motion and establish the dose rates to achieve a consistent oral laryngoscopy examination.Study design

Six healthy adult Beagle dogs.Methods
Each dog was randomly administered three induction agents with a 1-week washout period between treatments. Thiopentone (7.5 mg kg?1), propofol (3 mg kg?1) or alfaxalone (1.5 mg kg?1) was administered over 1 minute for induction of anaesthesia. If the dog was deemed inadequately anaesthetised, then supplemental boluses of 1.8, 0.75 and 0.4 mg kg?1 were administered, respectively. Continual examination of the larynx, using a laryngoscope, commenced once an adequate anaesthetic depth was reached until examination end point. The number of arytenoid motions and vital breaths were counted during three time periods and compared over time and among treatments. Data were analysed using Friedman and Mann–Whitney U tests, Spearman rho and a linear mixed model with post hoc pairwise comparison with Tukey correction.Results
The median (range) induction and examination times were 2.8 (2.0–3.0), 2.7 (2.0–3.3) and 2.5 (1.7–3.3) minutes (p = 0.727); and 14.1 (8.0–41.8), 5.4 (3.3–14.8) and 8.5 (3.8–31.6) minutes (p = 0.016) for thiopentone, propofol and alfaxalone, respectively. The median dose rates required to achieve an adequate anaesthetic depth were 6.3 (6.0–6.6), 2.4 (2.4–2.4) and 1.2 (1.2–1.2) mg kg?1 minute?1, respectively. There was no significant difference for the total number of arytenoid motions (p = 0.662) or vital breaths (p = 0.789) among induction agents.Conclusion and clinical relevance
The number of arytenoid motions were similar among the induction agents. However, at the dose rates used in this study, propofol provided adequate conditions for evaluation of the larynx with a shorter examination time which may be advantageous during laryngoscopy in dogs. 相似文献14.

To compare the motor and sensory block efficacy and duration of a modified paravertebral brachial plexus block (PBPB) after administration of lidocaine alone (LI) or combined with epinephrine (LE).Study design

A total of eight healthy female Beagle dogs.Methods
Under general anesthesia, modified PBPB was performed on the left thoracic limb using neurostimulation and/or ultrasound guidance to administer lidocaine (2 mg kg–1; 0.2 mL kg–1) either alone (treatment LI, n = 10) or with epinephrine (1:100,000; treatment LE, n = 9). Sensory block was evaluated through reaction to a painful mechanical stimulus applied at five sites on the limb. Motor block effect was evaluated according to visual gait assessments and thoracic limb vertical force measurements under dynamic and static conditions. Data were analyzed using repeated-measures generalized estimating equations. All statistical tests were performed two-sided at the α = 0.05 significance threshold.Results
The duration of sensory block did not differ significantly between treatments. Visible gait impairment was more persistent in LE than in LI (118 ± 63 minutes for LI and 163 ± 23 minutes for LE; mean ± standard deviation) (p = 0.027). At nadir value, dynamic peak vertical force was lower in LE than in LI (p = 0.007). For both dynamic and static evaluations, the nadir and the return to baseline force were delayed in LE (return to normal at 180–200 minutes) when compared with LI (130–140 minutes) (p < 0.005).Conclusions and clinical relevance
The addition of epinephrine to lidocaine prolonged the duration and increased the intensity of the regional block, as verified by visual gait assessment and kinetic analysis. No significant difference was noted between treatments regarding sensory blockade. Kinetic analysis could be useful to evaluate regional anesthetic effect in dogs. 相似文献15.

PWTT was calculated as the time from the electrocardiogram R-wave peak to the rise point of the pulse oximeter wave. Estimated CO (esCO) was derived from PWTT after calibration with arterial pulse pressure (both non-invasive and invasive methods) and TDCO. Haemodynamic changes were induced by administration of phenylephrine (vasoconstriction), high isoflurane (vasodilatation and negative inotropy) and dobutamine (vasodilatation and positive inotropy). Trending between percentage change in PWTT and TDSV was assessed using concordance analysis and receiver operator characteristic (ROC) curve. The agreement between esCO and TDCO was evaluated using the Bland–Altman method.Results
The direction of percentage change between consecutive PWTT and the corresponding TDSV showed a concordance rate of 95%, with correlation coefficients of ?0.86 (p < 0.001). Area under the ROC curve for the change in PWTT to detect 15% change in TDSV was 0.91 (p < 0.001). TDCO compared with esCO calibrated with invasive and non-invasive blood pressure showed a bias (precision of agreement) of 0.58 (1.54) and 0.57 (1.59) L minute?1 with a percentage error of ±61% and ±63%, respectively.Conclusions and clinical relevance
In isoflurane-anaesthetized dogs, PWTT showed a good trending ability to detect 15% changes in SV. This technique is easy to use, inexpensive, non-invasive and could become routine anaesthetic monitoring. However, the agreement between absolute esCO and TDCO was unacceptable. 相似文献16.

To evaluate the efficacy, in terms of the amount of rescue analgesia required, and the clinical usefulness of epidural injection of morphine with bupivacaine or levobupivacaine for elective pelvic limb surgery in dogs during a 24-hour perioperative period.Study design

A group of 26 dogs weighing 31.7 ± 14.2 (mean ± standard deviation) kg and aged 54 ± 36 months.Methods
All dogs were premedicated with methadone intravenously (0.2 mg kg–1) and anaesthesia induced with diazepam (0.2 mg kg–1) and propofol intravenously to effect. After induction of anaesthesia, dogs randomly received a lumbosacral epidural injection of morphine 0.1 mg kg–1 with either levobupivacaine 0.5% (1 mg kg–1; group LevoBM) or bupivacaine 0.5% (1 mg kg–1; group BM). Cardiovascular, respiratory and temperature values were recorded during the intra- and postoperative period. A visual analogue scale, subjective pain scale, sedation scale and the short form of the Glasgow pain scale were assessed every 6 hours after epidural injection during 24 hours. The ability to stand and walk, neurological deficits and other side effects were assessed at the same time points. The amount of rescue analgesia (sufentanil intraoperatively and methadone postoperatively) was recorded.Results
No statistically significant differences were found between groups for any of the recorded data, with the exception of the incidence of spontaneous urination and postoperative rescue analgesia requirement. In group LevoBM four dogs spontaneously urinated at recovery while none of the dogs in group BM did (p = 0.03) and seven dogs of group LevoBM required postoperative rescue analgesia versus none of the dogs in the BM group (p = 0.005).Conclusions

To determine the cardiovascular and acid-base effects of 6% hydroxyethyl starch (HES) 130/0.4 and 0.9% sodium chloride (NaCl) administered to anaesthetized greyhounds with haemorrhagic shock.Study design

Twelve healthy adult greyhounds.Methods
After 60 minutes of isoflurane anaesthesia, 48 mL kg?1 of blood was removed to induce hypotension. Dogs were randomized to receive either 20 mL kg?1 of HES 130/0.4 or 80 mL kg?1 of 0.9% NaCl over 20 minutes. Haemoglobin, arterial and central venous blood gas and electrolytes, lactate, mean arterial pressure (MAP) and cardiac index were measured at: T0, 60 minutes after induction of anaesthesia, immediately prior to blood removal; T1, immediately after blood removal; T2, immediately after fluid administration; and T3, 40 minutes after fluid administration. Oxygen extraction ratio (O2ER) was calculated at each sample time.Results
O2ER increased at T1 and decreased at T2 and T3, with no difference between the two groups. Dogs administered HES 130/0.4 had higher lactate at T2 [mean (95% confidence interval) 1.3 (0.8–1.9) mmol L?1] than dogs administered 0.9% NaCl [0.8 (0.5–1.1) mmol L?1]; p = 0.045. Dogs administered HES 130/0.4 had a higher MAP at T3 [88 (74–102) mmHg] than dogs administered 0.9% NaCl [69 (60–79) mmHg]; p = 0.019. Dogs administered 0.9% NaCl were more acidaemic at T2 and T3, including higher hydrogen ion, lower bicarbonate, lower base excess and higher chloride concentrations.Conclusion
and clinical relevance The effect of 20 mL kg?1 of HES 130/0.4 on shock, as measured by O2ER, was no different than that of 80 mL kg?1 of 0.9% NaCl in dogs under general anaesthesia. Dogs were enrolled in a modified 3 + 3 phase I trial. Three dogs initially received doses of 2.5 and 5 μg/kg SC syncBNP followed by an additional three dogs dosed at 5 and 10 μg/kg. Hemodynamic monitoring was performed for 120 min after each injection. Blood and urine samples were collected at 45 and 120 min after injection of 5 μg/kg. Major adverse clinical events that would potentially halt testing were pre-defined.Results
Four healthy dogs and two dogs with stage B1 mitral valve disease were recruited. Synthetic canine B-type natriuretic peptide was well tolerated at all doses. Synthetic canine B-type natriuretic peptide at 5 μg/kg significantly increased median plasma cGMP (baseline cGMP, 131.5 pmol/mL [range, 91.9–183.6 pmol/mL]; 45 min, 153.6 pmol/mL [140.3–214.3 pmol/mL]; 120 min, 192.7 pmol/mL [139.1–240.1 pmol/mL]; p=0.041).Discussion and conclusions

All dogs were premedicated with intramuscular acepromazine. General anaesthesia was induced with propofol and maintained with isoflurane in oxygen. Intraoperatively, nociception was assessed based on changes in heart rate, respiratory rate and mean arterial pressure above baseline values. Postoperatively, pain was evaluated with a Sammarco pain score, a Glasgow pain scale and a visual analogue scale (VAS). Tarlov’s scale was used to quantify motor block. All dogs were evaluated at recovery and then 1, 2, 3, 4, 5 and 24 hours after that. Rescue analgesia was provided during surgery with fentanyl and, postoperatively, with buprenorphine. Groups were compared using one-way repeated-measure analysis of variance followed by the Holm–Sidak method for multiple comparison or nonparametric tests when appropriate.Results
The two treatment groups did not differ (p > 0.05) with respect to intraoperative physiological variables, rescue analgesia, postoperative pain scores (Sammarco q = 1.00; Glasgow q = 3.10; VAS q = 0.50) and duration of the motor block (Tarlov’s q = 2.40).Conclusions and clinical relevance
The addition of epidural magnesium to ropivacaine did not improve or prolong the analgesia provided by ropivacaine alone.
