Events acting prenatally on developing foetuses are important determinants for disorders later in life. Prenatal stress (PNS) is one of these events. The purpose of this study was to determine the consequences of a repeated social stress applied during late gestation of the pregnant gilt on the immune system and hypothalamo-pituitary-adrenal (HPA) axis activity of the piglets from birth to two months of age. Pregnant gilts were submitted to repeated social stress which was induced by housing unfamiliar gilts in pairs modified twice a week during 4 weeks between days 77 and 105 of gestation (S group, n = 18). Control gilts were housed in stable pairs during the same period (C group, n = 18). Blood cortisol, haptoglobin and IgG levels, immune cell counts, mitogen-induced whole-blood proliferation and TNF-α production were evaluated in piglets at 4 days of age (D4), before and after weaning (D26 and 28) and before and after relocation to a new building (D60 and 62). We found that PNS did not affect growth rate of the progeny. It decreased the relative weight of adrenal glands on D4 (P < 0.05) but plasma cortisol levels were similar in both groups at all ages. IgG levels in colostrum and in the serum of piglets were not affected. PNS decreased the total numbers of white blood cells, lymphocytes and granulocytes from D26 to D60 (P < 0.05), the CD4+/CD8+ T cell ratio on D4 (P < 0.05), and LPS induced-TNF-α production on D60 (P < 0.05). PNS increased the ConA-induced lymphocyte proliferation on D4 and D60 and the PWM-induced proliferation on D60 (P < 0.05). Our results demonstrate that a repeated social stress applied to pregnant sows during late gestation can induce long-lasting effects on several parameters of the immune function of the offspring. These effects are not due to modifications of the HPA axis activity and may impair the abilities of the piglets to efficiently react against infections during the suckling period and around weaning.  相似文献

Chitosan oligosaccharides (COS) are the hydrolyzed product of chitosan and have multifunctional health benefits. The objective of this study was to elucidate the effect of COS as a dietary supplement to gilts on their productivity and health and that of their litters. Gilts were randomly assigned to either a treatment (n = 30) or control group (n = 30). The treatment gilts were fed a standard dry sow ration supplemented with COS at 0.12 and 0.24 g/gilt per d during gestation and lactation, respectively, and the control group was fed the standard dry sow ration only. The body weight, reproductive performance, milk production and litter size for each gilt and body weight of corresponding litters were recorded. The serum immunoglobulins (IgA, IgG, IgM) and secretory immunoglobulin A (sIgA) concentrations of gilts and piglets and fecal sIgA concertation of gilts were measured by Enzyme-linked immunosorbent assay (ELISA). Our study showed that maternal COS supplementation 1) significantly increased gilt body weight in late pregnancy (P < 0.05), 2) significantly increased milk production of gilts at different stages (d 1, 3, 7 and 19) of lactation (P < 0.05), 3) significantly increased body weight gain of piglets at weaning (P < 0.05), 4) significantly increased the serum concentrations of IgM and sIgA in piglets, and sIgA in fecal sample of gilts (P < 0.05), and 5) tended to increase the pregnancy success rate (P > 0.05) in the treatment group compared to the control group. These results suggest that maternal COS intervention in gilts can improve gilt milk production, piglet pre-weaning growth and immunity parameters in both gilts and piglets.  相似文献

Cushing's disease caused by pituitary corticotroph adenoma in dogs is usually treated by medical treatment, and the efficacy of this treatment has been reported. However, controversy remains as to whether reduced negative feedback through the inhibition of cortisol secretion, similar to Nelson's syndrome, may appear as an adverse effect. The purpose of this study was to investigate the effect of reduced negative feedback through the inhibition of cortisol secretion by daily trilostane administration on the pituitary–adrenal axis in clinically normal dogs. Dogs were administered 5 mg/kg trilostane twice a day every day for 8 weeks (n = 8) or 16 weeks (n = 3). After the initiation of trilostane administration, plasma adrenocorticotropic hormone (ACTH) concentrations were increased remarkably. As assessed by magnetic resonance imaging (MRI) during administration, the pituitary became enlarged. After trilostane administration, the cytoplasmic areas of the pituitary corticotrophs were increased and the ratio of pituitary corticotrophs to all cells in the anterior lobe was greater in the trilostane-treated dogs than that in untreated animals. In addition, histological examinations revealed bilateral adrenal cortical hyperplasia. Using real-time PCR quantification, the expression of proopiomelanocortin (POMC) mRNA in the pituitary and ACTH receptor (ACTH-R) mRNA in the adrenal gland was greater in the dogs treated with trilostane than in untreated dogs. These results indicate that reduced negative feedback induced hyperfunction of the pituitary corticotrophs and pituitary enlargement in healthy dogs. These changes suggest that the inhibition of cortisol secretion by trilostane may increase the risk for accelerating the growth of corticotroph adenomas in dogs with Cushing's disease.  相似文献

Two experiments were conducted to determine if the secretory patterns of luteinizing hormone (LH), follicle stimulating hormone (FSH) and prolactin (PRL) and serum concentrations of progesterone change immediately preceding induced puberty in gilts. To help predict when prepubertal gilts would attain puberty, gilts were induced into puberty by relocation from confinement housing to an outdoor lot and exposure to mature boars. In Exp. 1, 17 prepubertal gilts were bled on two successive days from 0800 to 1200 h before relocation and boar exposure and until the second day of estrus or for 8 d in gilts that failed to exhibit estrus. Blood samples were collected from indwelling cannulas at 20-min intervals for 4 h. In Exp. 2, blood samples were collected from 20 prepubertal gilts at 20-min intervals from 0800 to 1200 h and from 2000 to 2400 h until the second day of estrus or for 6 d if the gilt failed to exhibit estrus. In each experiment, 11 gilts exhibited pubertal estrus 3 to 6 d after relocation and boar exposure. When the frequency of LH spikes in each gilt was normalized to the day of her preovulatory surge of LH (d 0), a decline in the frequency of LH secretory spikes was observed as gilts approached puberty. However, neither the average magnitude of LH spikes nor mean LH concentrations were different among these days. This study was conducted to evaluate the effect of estrus induction with gonadotropins on endometrial and conceptus expression of HoxA10, transforming growth factor (TGF) β1, leukemia inhibitory factor (LIF), and prostaglandin H synthase-2 (PGHS-2) during early pregnancy in pigs. Twenty-four prepubertal gilts received 750 IU of pregnant mare serum gonadotropin (PMSG) and 500 IU of human chorionic gonadotropin (hCG) 72 h later. Gilts in the control group (n = 23) were observed daily for estrus behavior. Endometrial tissue samples, conceptuses, blood serum, and uterine luminal flushings (ULFs) were collected on days 10, 11, 12, and 15 after insemination. There was no effect of estrus induction on estradiol content in ULFs, or on ovulation and fertilization rates in studied gilts. However, the content of progesterone in the blood serum was greater in naturally ovulated gilts in comparison to gonadotropin-treated animals on day 12 of pregnancy (P < 0.05). HoxA10 expression was up-regulated in the endometrium of pregnant gilts, with natural ovulation on days 12 (P < 0.05) and 15 (P < 0.001) in comparison to days 10 and 11. When compared to control gilts, administration of PMSG/hCG resulted in decreased expression of endometrial HoxA10, TGFβ, LIF, and PGHS-2 on day 12 of pregnancy (P < 0.05). Conceptus expression of studied factors was not affected by gonadotropin treatment. Overall, these results suggest improper endometrial preparation for implantation in prepubertal gilts induced to ovulate with PMSG/hCG.  相似文献

The effects of floor heating at parturition on sow and piglet behaviour in pens were examined in a Danish production herd. A total of 50 gilts and sows were split into two groups; one (n = 25) experienced no treatment; one (n = 25) was exposed to floor heating in the area of the sow around parturition. The sows and piglets were recorded on video 24 h a day for behavioural analyses.Floor heating tended to improve piglet weight at weaning (P = 0.05) but did not affect average daily gain. Piglets in pens with floor heating were more often in proximity of the sow at posture changes than piglets from pens without floor heating on day one postpartum (P = 0.01). Floor heating did not affect duration of parturition or any of the recordings of sow behaviour significantly.  相似文献

The objective of this study was to estimate the effect of piglet birth weight on future BW, growth, backfat, and longissimus muscle area of pigs in a commercial U.S. production system. Pigs (n = 5727) at a commercial farm were individually weighed and identified within 24 h of birth. Weights were collected prior to weaning (n = 4108), after finisher placement (n = 3439), and 7 (n = 1622) and 16 (n = 1586) weeks into finishing; hot carcass weight was also collected (n = 1693). Average daily gain during lactation, nursery, finishing, and overall (birth to 16 weeks into finishing) was calculated. During BW collection 16 weeks into finishing, real-time ultrasound backfat thickness and longissimus muscle area were measured. Sex × birth weight (linear and quadratic) interactions were observed for BW at weaning and finisher placement and daily gain during pre-weaning and nursery. Linear birth weight × cross foster interactions were observed for weaning weight and pre-weaning gain. Linear and quadratic effects of birth weight on BW at weaning, finisher placement, 7 and 16 weeks into finishing, and hot carcass weight and average daily gain during pre-weaning, nursery, finishing, and total were observed. For all measures of BW and average daily gain, as birth weight increased subsequent BW and average daily gain increased at a decreasing rate; however, for the sex × birth weight (linear and quadratic) interactions, heavier birth weight barrows were lighter and grew slower than gilts of comparable birth weight. Worth noting, the birth weight × sex interactions described very few pigs in the extreme portion of the birth weight distribution. For birth weight × cross foster interactions, non-cross fostered pigs were increasingly heavier and faster growing as birth weight increased compared to cross fostered pigs. Heavier birth weight pigs tended to have increased backfat depth (P = 0.07). Linear and quadratic effects of birth weight on longissimus muscle area were observed; as birth weight increased muscling increased at a decreasing rate. Regardless of interactions or period of production, increased birth weight resulted in heavier future BW, faster daily gain along with larger longissimus muscle area prior to harvest. In all instances the magnitude of the negative effect of birth weight increased as birth weight decreased.  相似文献

Clinical outbreaks due to Actinobacillus pleuropneumoniae occur recurrently, despite the wide-scale use of antimicrobials or vaccination. Therefore, new approaches for the prevention and control of these outbreaks are necessary. For the development of alternative measures, more insight into the transmission of the bacterium on farms is necessary. The aim of this cohort study was to quantify transmission of A. pleuropneumoniae amongst weaned piglets on farms. We investigated three possible transmission routes: (i) indirect transmission by infected piglets within the same compartment, (ii) transmission by infected pigs in adjacent pens and (iii) transmission by direct contact within pens. Additionally, we evaluated the effect of independent litter characteristics on the probability of infection. Two farms participated in our study. Serum and tonsil brush samples were collected from sows pre-farrowing. Serum was analysed for antibodies against Apx toxins and Omp. Subsequently, tonsil brush samples were collected from all piglets from these dams (N = 542) in three cohorts, 3 days before weaning and 6 weeks later. Tonsil samples were analysed by qPCR for the presence of the apxIVA gene of A. pleuropneumoniae. Before weaning, 25% of the piglets tested positive; 6 weeks later 47% tested positive. Regression and stochastic transmission models were used to assess the contribution of each of the three transmission routes and to estimate transmission rates. Transmission between piglets in adjacent pens did not differ significantly from that between non-adjacent pens. The transmission rate across pens was estimated to be 0.0058 day−1 (95% CI: 0.0030–0.010), whereas the transmission rate within pens was ten times higher 0.059 day−1 (95% CI: 0.048–0.072). Subsequently, the effects of parity and serological response of the dam and litter age at weaning on the probability of infection of pigs were evaluated by including these into the regression model. A higher dam ApxII antibody level was associated with a lower probability of infection of the pig after weaning; age at weaning was associated with a higher probability of infection of the pig after weaning. Finally, transmission rate estimates were used in a scenario study in which the litters within a compartment were mixed across pens at weaning instead of raising litter mates together in a pen. The results showed that the proportion of infected piglets increased to 69% if litters were mixed at weaning, indicating that farm management measures may affect spread of A. pleuropneumoniae.  相似文献

Fall-born calves grazing Neotyphodium coenophialum-infected tall fescue [E+; Lolium arundinaceum (Schreb.) Darbysh.] pastures should benefit from early weaning because of reduced exposure to fungal toxins. However, fall-born calves that grazed E+ and were weaned in mid-April had reduced post-weaning performance compared with calves managed similarly but weaned in early June. Gelbvieh × Angus calves (n = 238) were used in a 3-year study to determine the optimal time to wean fall-born calves grazing E+. Cow/calf pairs were allocated randomly to one of four weaning date treatments: 1) March 16 (177 ± 4.7 days of age; MarW), 2) April 13 (204 ± 4.7 days of age; AprW), 3) May 11 (236 ± 4.7 days of age; MayW), and 4) June 8 (264 ± 4.8 days of age; JuneW). On their assigned weaning date, calves were weighed, vaccinated, blood samples were collected, and calves were moved to 3.2-ha pastures adjacent to their dams for 14 days. After the weaning period, calves were weighed, blood samples were collected, and then calves were moved to pastures containing non-toxic forages. Birth weight and calf weights on MarW, AprW, and MayW weaning dates did not differ (P ≥ 0.21) across weaning date treatments. However, calf weaning weights on their respective weaning dates, weight on JuneW and on June 22 (14 days following the JuneW), daily gain between birth and June 22, and weight change between MarW and JuneW increased linearly (P < 0.05) across weaning dates. Linear decreases (P < 0.05) across weaning dates were noted for antibody titers to bovine virus diarrhea (BVD) and infectious bovine rhinotracheitis (IBR), serum Cu, and platelets at the time of actual weaning. Linear increases (P < 0.05) across weaning dates were noted for: i) total antioxidant potential and eosinophils at weaning; ii) changes in antibody titers to BVD and IBR, total antioxidant potential, and serum Zn during the 14-day post-weaning period; and iii) and changes in BVD and BRSV titers and serum Cu between actual weaning date and June 22. Heifer weight at breeding increased (P < 0.05) and steer weight at shipping, hot carcass weight and backfat thickness tended (P ≤ 0.10) to increase linearly across weaning dates. Final feedlot weight and gain did not differ (P ≥ 0.18) among weaning dates. Therefore, delaying weaning of fall-born calves grazing E+ pastures until early June may be beneficial for calf weight and immune function at weaning, and heifer weight at breeding, but those benefits in steers may be mitigated through the feedlot period.  相似文献

Forty piglets (average body weight = 5.32 kg) were used to investigate the effect of dietary crude protein (CP) content on immunological responses following a challenge with enterotoxigenic Escherichia coli (ETEC) K88. Pigs, housed 4 per pen, were randomly allotted to 2 diets: 1) a high, 225 g/kg CP diet (HCP) or 2) a low, 176 g/kg CP diet (LCP) supplemented with crystalline amino acids. Pigs were orally challenged with 6 mL of an ETEC K88 suspension containing 1010 cfu/mL on d 8 after weaning. Blood samples were collected from 10 pigs (1 pig/pen) on d 7 (at weaning), −24 h, 8 h, 72 h and 7 d after the challenge for determination of plasma urea N (PUN) and serum concentrations of tumor necrosis factor alpha (TNF-α) and interleukin 1 beta (IL-1β) and haptoglobin (Hp). Tumor necrosis factor alpha, IL-1β and Hp were measured as indicators of inflammatory responses. The concentrations of serum TNF-α at 8 h, 72 h and 7 d after challenge were similar to the level observed at 24 h before challenge but higher (P < 0.05) than the weaning level. Pigs fed the LCP diet had lower (P = 0.032) concentrations of IL-1β (72 vs. 116 pg/mL) at 8 h post-challenge compared with those fed the HCP diet. Likewise, pigs fed the LCP diet tended to have lower (P = 0.088) concentration of Hp (9 vs. 25 mg/dL) compared with those fed the HCP diet at 8 h post-challenge. Compared with the weaning concentration, PUN concentration at 72 h after ETEC challenge was higher (P < 0.05) in pigs fed the HCP diet. The results indicate that the LCP diet supplemented with crystalline amino acids reduced inflammatory responses, as indicated by serum IL-1β, in piglets infected with ETEC K88.  相似文献

Growth rates during rearing affect the age and body weight (BW) of replacement heifers at first calving. Diet and disease can affect growth via altered metabolic hormone concentrations, but are difficult to monitor accurately on commercial farms. This study investigated the effect of management and metabolic indices (IGF-I, insulin, glucose and urea) on the growth rate of 509 Holstein-Friesian heifers on 19 UK dairy farms. Size (BW, heart girth, height and crown-rump length) was measured at approximately 1, 6 and 15 months. The mean daily weight gain up to 6 months for all calves was 0.77 kg/day, with extreme variability both between cohorts of calves (range 0.49–1.02 kg/day) and between individual calves within farms (range 0.45–1.13 kg/day). Growth was enhanced by supplemental colostrum, by milk replacer as opposed to whole milk and by ad libitum milk feeding and was reduced by gradual weaning and dehorning after weaning. Larger group size slowed growth before weaning (>6 calves) but increased it post-weaning (>20 calves). These management differences were reflected in altered plasma IGF-I concentrations, which were positively associated with growth throughout the rearing period. Larger calves at 1 month had a greater weight gain up to 6 months. Sub-optimum growth of some heifers within each cohort was established at an early age and resulted in animals reaching the start of breeding at an inadequate size (BW range 209–498 kg at 15 months). This could be alleviated by altered management strategies and improved monitoring of growing heifers.  相似文献

To study the effect of GnRH in prepubertal gilts, seven crossbred gilts were treated with saline solution and 250 fig GnRH. In connection with saline and GnRH treatments blood was sampled every 15 min for 4 h, thereafter every 30 min for 2 h and every 60 min for 3 h, and finally every 3 h for 6 days. The ovaries were inspected by laparo-scopy just before and 6 days after GnRH treatment. The first GnRH treatment was undertaken when the gilts had a mean age of 141 days and mean body weight of 66 kg. One gilt was in prooestrus at this treatment. In the other 6 gilts the mean LH level was around 0.5 μg/l during a 4 h period after the saline injection. After the GnRH treatment a LH peak was seen with a mean duration of 4 h and a mean maximum level of 9.2 ± 2.07 μg/1. None of the gilts ovulated or showed oestrus within a week after GnRH treatment, which was confirmed by laparoscopy. The seventh gilt which was in prooestrus had high levels of oestradiol-17β (> 40 pmol/1) at GnRH treatment and no LH peak was seen during a 4 h period after treatment.Two gilts which had not shown oestrus at an age of 173 days and a mean body weight of 93 kg were treated a second time with 250 μg GnRH. The LH peak had a duration of 4 h and a mean maximum level of 5.3 ± 3.04 μg/l. Neither of these 2 gilts showed oestrus or ovulated within a week after GnRH injection. It was concluded that a single injection of GnRH results in a LH peak but is not enough to stimulate ovulation or oestrus in prepubertal gilts at a mean age either of 141 or 173 days.Key words: GnRH-treatment, prepubertal gilts, LH, oestradiol-17β  相似文献

Altered nursing or nutrition before artificial insemination (AI) can be used as a doe biostimulant to improve lactating rabbits reproduction. The timing of a shift from free to a 3-day controlled nursing to AI or the nursing method at fast-refeeding can affect the efficacy of these stimulations. In an earlier study the effects of a 3-day controlled nursing on days 8, 9 and 10 or in controlled nursing rabbits, the impact of a 24-h fast with 48–50 h re-feeding were investigated. This follow-up work tested a 3-day biostimulation with controlled nursing on days 9, 10 and 11. Another aim was to assess the same doe fast-refeeding but now in free-nursing rabbits. Pannon White does (n=480) were artificially inseminated 11 days post-partum. Control (C) does nursed freely. Rabbits simulating local farm practice (F) had controlled nursing until day 14 using a metal-plate as separation and then free nursing to weaning (day 35). In biostimulations with altered nursing, there was a shift from free to a 3-day controlled nursing (days 9–11) with a wire-mesh separation (BW), a metal-plate insertion (BM) or nest-tray removal (BN) and return to free nursing on day 12 until weaning. The C, F, BW, BM and BN does were fed ad libitum. At biostimulation with fast-refeeding (BF), the free-nursing does were subjected to a 24 h water-only fast between days 8 and 9 and a 48–50 h ad libitum re-feeding before AI. Doe reproduction and growth of the current litter were differently affected by the treatments. In the C, F, BW, BM, BN and BF does, sexual receptivity was 83, 90, 68, 80, 74 and 85% (P=0.05), the kindling rate was 79, 76, 74, 89, 68, 70% (P=0.05), the number of kits born alive was 7.9, 8.0, 8.8, 9.1, 7.9, 6.8 (P=0.005), kit weight at weaning 982, 991, 953, 986, 955, 964 g (P=0.012) and at 70 days of age 2383, 2407, 2220, 2350, 2279, 2382 g, respectively (P=0.001). Among biostimulations with altered nursing, the 3-day controlled nursing with a metal-plate separation (BM) can be advised for the practice because only this method was efficient in this (days 9–11) and previous (days 8–10) studies. There appears to be an interaction between doe nursing and feeding, since the same fast led to different production of free-nursing does compared to those in a previous work that nursed controlled.  相似文献

This study aimed to determine the effects of weaning age and postweaning feeding programme on pig performance and health. In experiment 1, 96 same gender pairs of pigs weaned at 3, 4 and 5 weeks of age were used in a 3 (weaning age)×4 (dietary programme) factorial design experiment. Pigs received different amounts of a phase 1 diet (16.2 MJ/kg digestible energy (DE) and 16.2 g/kg lysine) and phase 2 diet (15.3 MJ/kg DE and 15.0 g/kg lysine): (A) very low (VL, 1 kg phase 1 and 3 kg phase 2 diet per pig); (B) low (L, 2 kg phase 1 and 6 kg phase 2 diet per pig); (C) medium (M, 3 kg phase 1 and 9 kg phase 2 diet per pig) or (D) high (H, 4 kg phase 1 and 12 kg phase 2 diet per pig), followed by a cereal based phase 3 diet (15.0 MJ/kg DE and 13.8 g/kg lysine) to 10 weeks of age. In experiment 2, faecal samples from 60 pigs weaned at 3, 4 and 5 weeks of age were collected at 10 days postweaning and analysed for Escherichia coli and lactic acid bacteria counts. In experiment 1, there were no interaction effects of age×dietary programme on growth performance. Dietary programme did not affect growth performance from weaning to 10 weeks of age. From weaning to 10 weeks of age, increasing weaning age increased average daily gain (ADG; 363, 402, and 476 g for 3, 4 and 5 weeks respectively; s.e. 17.6; P<0.001), average daily feed intake (ADFI; 560, 620, and 680 g; s.e. 26.1; P<0.001), and improved feed conversion ratio (FCR; 1.57, 1.55, and 1.43; s.e. 0.045; P<0.05). However, weaning age had no effect (P>0.05) on pig weight at 10 weeks of age. In experiment 2, 3 week weaned pigs had higher faecal counts of E. coli (P<0.05) than 4 week weaned pigs and higher faecal counts of lactic acid bacteria (P>0.01) than 5 week weaned pigs at 10 days postweaning. In conclusion, feeding higher amounts of phases 1 and 2 diets did not affect performance at any of the weaning ages tested. Increasing weaning age increased growth performance between weaning and 10 weeks of age, but had no effect on the resulting body weight. Pigs weaned at 3 weeks of age had increased counts of selected faecal bacteria compared to those weaned later.  相似文献

Porcine endometrial development between birth (postnatal day = PND 0) and PND 56 involves differentiation of glandular epithelium (GE) from luminal epithelium (LE) and estrogen receptor-alpha (ER) expression. Juvenile ER architecture evolves after birth, as stroma and nascent GE first express ER. Mature ER architecture is evident after PND 30, when stroma, GE and LE are ER-positive. When administered during discrete periods between PND 0 and 56, effects of estradiol-17beta valerate (EV) on the neonatal porcine uterus relate to endometrial ER architecture. Transient EV exposure from birth reduces embryo survival in pregnant adult gilts. Effects of EV, administered as juvenile endometrial ER architecture develops (P1, PND 0-13), or after mature ER architecture is established (P2, PND 42-55), were evaluated in uteri from gilts treated with corn oil or EV in P1 or P2 and hysterectomized on PND 100 without additional steroids (NSt), on PND 102 after EV on PND100-101 (EV2), or on PND 117 after EV2 followed by progesterone on PND 102-116 (EP). Neonatal EV reduced uterine weight (P < 0.02), size (P < 0.01), luminal protein content (P < 0.07), and percent incorporation of 3H-leucine into nondialyzable endometrial products in vitro (P < 0.01). Group (NSt, EV2, EP) -specific treatment effects detected for endometrial ER, progesterone receptor, uteroferrin, and/or retinol binding protein mRNA levels were frequently related to period (P1,P2). Results support the idea that estrogen-sensitive postnatal organizational events, including those defined, in part, by endometrial ER architecture, are likely components of genetic and epigenetic programs governing uterine morphogenesis and ontogeny of endometrial function in the pig.  相似文献

The temporal pattern and sex effect of immune and stress hormone responses to a lipopolysaccharide (LPS) challenge were assessed using a pig model. Secretion of the pro-inflammatory cytokines tumor necrosis factor (TNF)-α, interleukin (IL)-1β, and IL-6 increased in a time-dependent manner following LPS infusion. There was also a time-dependent increase in secretion of the stress-related hormones cortisol, epinephrine (E), and norepinephrine (NE) following LPS, with peak concentrations attained within 30 min. The magnitude of the TNF-α and IL-1β responses were both positively associated (P < 0.05) with the magnitude of cortisol response following LPS, whereas serum IL-1β and IL-6 were positively correlated with the magnitude of E and NE responses following LPS. Acute-phase protein production was also time-dependently increased following LPS. The concentration of immune cells in circulation was decreased (P < 0.05) at 5.5 h post-LPS and negatively correlated with pro-inflammatory cytokine production. By 24 h post-LPS, immune cell counts increased (P < 0.05) and were positively associated with both pro-inflammatory cytokine and stress hormone production. The amplitude of pro-inflammatory cytokine response following LPS was affected (P < 0.05) by sex classification; however, the magnitude of elevated cytokine concentrations was not. The magnitude of the NE response, but not of the E and cortisol responses, to LPS was influenced by sex (P < 0.05). Similar to the pro-inflammatory cytokines, the magnitude of exposure to the stress hormones following LPS was not influenced by sex. The production of serum amyloid A (SAA) was influenced by sex, with barrows producing more SAA than gilts at 24 h post-LPS (P < 0.05). Collectively, these results demonstrate sex-specific, concomitant temporal changes in innate immune- and stress-related hormones.  相似文献

Exposing a pregnant sow to stress has been shown to affect the resulting offspring. Our objective was to determine if rough handling of pregnant sows altered the physiology of her offspring and if these alterations were different from an experimentally induced model of prenatal stress. Sow treatments consisted of i.v. injections of ACTH (1 IU/kg of BW), exposure to rough handling for 10 min (Rough), or no treatment (Control) once a week during d 42 to 77 of gestation. To determine the plasma cortisol response to treatments, blood (5 mL) was collected from 30 sows after treatment administration. To conduct the prenatal stress study, a separate group of 56 sows was used in 1 of 4 replicates. At birth, production data were collected for each litter, including birth weight, number born, anogenital distance, and pig viability. At weaning, pigs were blocked by BW and sex, and placed in a nursery pen of 6 pigs, with 2 pigs from each treatment group. To assess the effect of treatments on cortisol, corticosteroid-binding globulin (CBG), and hematological cell profiles, blood was collected every other day for 10 d after weaning. Application of treatments caused plasma cortisol concentrations to be greatest in ACTH sows compared with Control sows (P < 0.001), with Rough sows having intermediate values (P = 0.07). Treatments did not affect the number of pigs born, number of stillborn, or pig viability (P > 0.40). The ratio of cortisol to CBG did not differ between treatments (P = 0.09). Hematological variables did not differ between treatments (P > 0.19). Pigs born to ACTH sows had a smaller anogenital distance compared with controls (P < 0.03), with pigs from Rough sows being intermediate. Our data indicate that swine exposed to prenatal stress (ACTH injection) can have alterations in sexual morphology without effects on growth or the immune cell populations measured in this study.  相似文献

Recent evidence suggests that different patterns of prenatal survival causing different levels of intrauterine crowding are occurring in prolific sows bearing litters with 10–15 piglets born, resulting in marked differences in average litter birth weight. Because early intrauterine crowding of embryos is known to have negative effects on prenatal development, the objective of the study was to compare litter characteristics, as well as histomorphometrical and molecular traits of myofibers of selected offspring born from litters of high (H: >1.7 kg) and low (L: <1.3 kg) average birth weight. The selected newborn littermates were either two females representing the intermediate (HI: 1.73 kg) and low (HL: 1.41 kg) birth weight from H-litters or two females representing the intermediate (LI: 1.26 kg) and high (LH: 1.55 kg) birth weight from L-litters. These piglets were sacrificed the day of birth and the longissimus (LM) and semitendinosus (ST) muscles were collected. The average birth weight of the selected HI and LI piglets differed (P<0.05) whereas that of the HL and LH piglets was similar. From birth to weaning, H-offspring grew faster (P<0.05), which was associated with greater (P<0.05) body weight loss of H-compared with L-sows during lactation, where their feed intake did not differ. In LI offspring, brain proportion of birth weight and brain to ST ratio was greater (P<0.05) than in HI but similar between LH and HL progenies. Expressed per mm2, the dark portion of the ST of HI piglets had fewer (P<0.05) primary but similar secondary myofiber numbers compared to LI progeny. Nevertheless, due to the larger ST, total myofiber number was greater in HI and LH than LI piglets. In the LM, myofiber characteristics were similar across birth weight classes, except for the greater (P<0.10) secondary to primary myofiber ratio in HL than LH offspring. In the ST, but not in the LM, the expression of the myoblast differentiation factor 1 was greater (P<0.05) in LH than HL offspring. No birth weight class differences occurred in myosin heavy chain isoform distribution in the LM. In conclusion, female progenies falling in the mid-weight range of the L-litters, but not their heavier littermates, carried some negative phenotypic traits normally associated with intrauterine growth restriction, such as greater brain to ST ratio and impaired myofiber hyperplasia.  相似文献

A three-year longitudinal study was conducted to evaluate effects of strategic anthelmintic treatment regimes on age at first lambing (AFL), weight at first lambing (WFL) and lambing interval (LI) of 356 communally grazed ewes and 675 lambs owned by 10 smallholder farmers in the central highlands of Ethiopia. The ewes were stratified by weight and randomly allocated to three treatment groups as untreated control (TG1), twice-dosed per year (TG2) for both nematodes and trematodes in mid-January and mid-June and four-time-dosed per year (TG3) in June for nematodes, in August–September for nematodes and adult Fasciola, in November–December and January–February for immature flukes. The fixed effect of anthelmintic treatments, parity, season and year of lambing on AFL and LI was evaluated. Mean ± standard error (SE) of lambing interval was 292 ± 3 days. Both anthelmintic treatments (TG2 and TG3) shortened (P < 0.01) LI by about 23 days compared to non-treated ewes (TG1). Season and year of lambing had a significant (P < 0.001) effect on LI. Mean ± SE of AFL and WFL was 598 ± 10 days and 17.2 ± 1.37 kg, respectively. Anthelmintic treatments and parity of dam of the ewe lamb did not affect AFL (P > 0.05). Birth and lambing seasons of the ewe lamb had significant (P < 0.05) effect on AFL. On the other hand, lambing season of the ewe had significant (P < 0.05) effect on WFL. Ewe lambs born in the long rains lambed more than 50 days earlier than the ewe lambs born in short rainy and dry seasons. Lambing occurred year-round with two peaks in August–September and December–January, each 5 months after the two rainy seasons. Anthelmintic treatment at the beginning of the two rainy seasons should improve reproductive performance of Menz ewe lambs in similar agro-ecology in the central highlands of Ethiopia.  相似文献

This paper describes the parasite Henneguya corruscans n. sp. which infects the gills of Pseudoplatystoma corruscans Spix and Agassiz, 1829 found in the Paraná River, Brazil. The parasites belong to the interlamellar-epithelial type as defined by Molnár (2002) [Molnár, K., 2002. Site preference of fish myxosporeans in the gills. Dis. Aquat. Org. 48, 197–207]. The spores examined had thin, smooth walls with symmetric valves; the total length of the spores was 27.6 (25–29) μm. The spore body was ellipsoidal in frontal view and biconvex in lateral view and they measured 14.3 (13–15) μm long by 5 μm wide and 4 μm in thickness. The polar capsules were small and elongated, equally sized, with a rounded posterior extremity and tapering anteriorly, and they corresponded more or less the half the length of the spore body; they were 6.8 (6–7) μm long by 2 μm wide, and the polar filament formed 5–6 coils obliquely to the axis of the polar capsule. The tail was 13.7 (12–15) μm long and bifurcated shortly after the end of the spore body. The importance of the infection for the farming of P. corruscans is discussed.  相似文献
