Aquaflor® [50% w w?1 florfenicol (FFC)], is approved for use in freshwater‐reared warmwater finfish which include tilapia Oreochromis spp. in the United States to control mortality from Streptococcus iniae. The depletion of florfenicol amine (FFA), the marker residue of FFC, was evaluated after feeding FFC‐medicated feed to deliver a nominal 20 mg FFC kg?1 BW d?1 dose (1.33× the label use of 15 mg FFC kg?1 BW d?1) to Nile tilapia O. niloticus and hybrid tilapia O. niloticus × O. aureus held in a recirculating aquaculture system (RAS) at production‐scale holding densities. Florfenicol amine concentrations were determined in fillets taken from 10 fish before dosing and from 20 fish at nine time points after dosing (from 1 to 240 h post‐dosing). Water samples were assayed for FFC before, during and after the dosing period. Parameters monitored included daily feed consumption and biofilter function (levels of ammonia, nitrite and nitrate). Mean fillet FFA concentration decreased from 13.77 μg g?1 at 1‐h post dosing to 0.39 μg g?1 at 240‐h post dosing. Water FFC concentration decreased from a maximum of 1400 ng mL?1 at 1 day post‐dosing to 847 ng mL?1 at 240 h post‐dosing. There were no adverse effects noted on fish, feed consumption or biofilter function associated with FFC‐medicated feed administration to tilapia.  相似文献

Aeromonas hydrophila and Flavobacterium columnare, the aetiological agents of motile aeromonas septicaemia (MAS) and columnaris disease respectively, have been recently causing crippling mortalities to the sunshine bass, Morone chrysops female ×Morone saxatilis male (Percichthyidae), industry in the United States. Isolates of A. hydrophila and F. columnare obtained from fish that died during farm outbreaks were subjected to in vitro evaluation of florfenicol (FFC), copper sulphate (CuSO4) and potassium permanganate (KMnO4). Florfenicol inhibited the growth of A. hydrophila and F. columnare more than CuSO4 and KMnO4. The minimum inhibition concentration (MIC) of FFC was 0.04 ± 0 and 0.2 ± 0.1 mg L?1 for A. hydrophila and F. columnare respectively, while the 50% inhibition concentration (IC50) for A. hydrophila and F. columnare was 0.23 ± 0.01 and 0.4 ± 0.2 mg L?1 respectively. Copper sulphate was more effective against A. hydrophila than KMnO4; CuSO4 had a MIC of 83.2 ± 0 mg L?1 compared to 158.0 ± 0 mg L?1 for KMnO4. Copper sulphate was also more effective against F. columnare than KMnO4. The IC50 values of CuSO4 and KMnO4 towards F. columnare were 4.8 ± 0.3 and 8.7 ± 1.6 mg L?1 respectively, and the minimum bactericidal concentration values of CuSO4 and KMnO4 towards F. columnare were 25.0 ± 0 and > 158.0 mg L?1 respectively. In addition, F. columnare was more sensitive to CuSO4 and KMnO4 than A. hydrophila.  相似文献

Sanguinarine (SA), with antimicrobial and antiparasitic activities against fish pathogens, exhibits great potential commercial use in aquaculture. However, little information on pharmacokinetics of SA restricts further application in aquaculture. In this study, pharmacokinetics of SA in common carp (Cyprinus carpio) following a single intraperitoneal administration [10 mg kg?1 BW (body weight)] was evaluated by high‐performance liquid chromatography (HPLC). The peak concentration (Cmax) of SA in kidney was 11.8 μg g?1, which was higher than in other tissues and plasma. The terminal half‐life in fish tissue and plasma was as follows: 42.3 h (kidney) > 37.2 h (liver) > 20.1 h (gill) > 18.8 h (muscle) > 10.9 h (spleen) > 10.0 h (plasma). Additionally, we determined the bacterial loads in tissues of common carp infected with Aeromonas hydrophila after i.p. administration of SA at 0, 5, 10 and 20 mg kg?1 BW. The results showed that i.p. administration of SA at 10 mg kg?1 BW significantly enhanced antibacterial efficacy against A. hydrophila, where the antibacterial ratio in the gill, kidney, spleen and liver on day 5 was 95.13%, 93.33%, 90.09% and 92.82%, respectively. Overall, these results suggested the potential of SA to treat A. hydrophila infection in common carp farming industry.  相似文献

This study aimed to test the efficacy of 17 α‐methyl testosterone (17 α‐MT) alone and in combination with letrozole, an aromatase inhibitor, for the induction of sex reversal in protogynous greasy grouper, Epinephelus tauvina. Further, the long‐lasting effects of these treatments and spawning performance of sex‐reversed males were also investigated. Greasy grouper with oocytes in the perinucleolus stage were implanted with 5 mg 17 α‐MT kg?1 body weight (T1), 5 mg 17 α‐MT and 0.2 mg letrozole kg?1 body weight (T2) and 5 mg 17 α‐MT with 0.4 mg letrozole kg?1 body weight (T3) and no androgens/enzyme inhibitor implanted (C). The 17 α‐MT alone and in combination of letrozole‐induced sex reversal in greasy grouper, whereas untreated control fish (C) showed normal ovarian development. However, T2 and T3 group showed 100% sex reversal and completion of spermatogenesis up to functional male phase in 2 and 3 months, respectively, whereas T1 group resulted in only 66.67% functional male with motile spermatozoa after 4 months. Sex‐reversed males successfully fertilized the eggs during induced spawning. There were significant differences on fertilization and hatching rates between T2 group (79.00 ± 4.36%; 77.67 ± 2.87%, respectively) and T1 group (57.67 ± 3.17%; 63.87 ± 2.91%, respectively). The result suggested that 17 α‐MT (5.0 mg kg?1 BW) in combination with letrozole (0.2 mg kg?1 BW) has the potential to produce 100% sex‐reversed male in short period in greasy grouper, which might greatly help in seed production of greasy grouper.  相似文献

The pharmacokinetics of florfenicol (FF) in turbot (Scophthalmus maximus) was studied after single intravenous (10 mg kg−1) and oral (100 mg kg−1) administration. The plasma concentration–time data of florfenicol were described by an open one‐compartment model. The elimination half‐life (t1/2) was estimated to be 21.0 h, and the total body clearance, Cl, was determined as 0.028 L kg h−1. The apparent volume distribution (Vd) was calculated to be 0.86 L kg−1 and the mean residence time (MRTiv) was 30.2 h. Following oral administration, the maximum plasma concentration (Cmax) of 55.4 μg mL−1 was reached at 12 h (Tmax). The absorption constant (ka) was 0.158 h−1. The bioavailability was estimated to be 57.1%. The low bioavailability observed at higher doses was explained by the saturation of the mechanisms of absorption. The drug absorption process was limited by its inherent low solubility, which limited the amount of available FF absorbed in the gastrointestinal tract. Based on the pharmacokinetic data, an optimal dosing schedule for FF administration is hereby provided. Based on the minimum inhibitory concentration found for susceptible strains of Aeromonas salmonicida, oral FF administration of first, an initial dose of 30 mg FF kg−1, followed by 6 maintenance doses at 18 mg kg−1/daily could be effective against furunculosis in turbot.  相似文献

Francisella noatunensis subsp. orientalis (Fno) (syn. F. asiatica) is an emergent Gram‐negative facultative intracellular bacterium. Although it is considered one of the most pathogenic bacteria in fish, there are no commercially available treatments or vaccines. The objective of this project was to determine the most efficacious concentration of florfenicol (FFC) [10, 15 or 20 mg FFC kg?1 body weight (bw) per days for 10 days] administered in feed to control experimentally induced infections of Fno in Nile tilapia, Oreochromis niloticus (L.), reared in a recirculating aquaculture system. The cumulative mortality of fish that received 0, 10, 15 or 20 mg FFC kg?1 bw per day was 60, 37, 14 and 16%, respectively. Francisella noatunensis subsp. orientalis genome equivalents were detected in water from all challenged groups with slight reduction in the concentration in the florfenicol‐treated groups 4 days after treatment. The mean LOG of CFU Fno mg?1 spleen was 3–5 and was present in all challenged groups at necropsy 11 days after treatment (21 days after challenge). Results show that florfenicol administered at doses of 15 and 20 mg FFC kg?1 bw per days for 10 days significantly reduced mortality associated with francisellosis in Nile tilapia.  相似文献

An 8‐week feeding trial was conducted to assess dietary protein and lipid levels on growth performance, feed utilization and body composition of juvenile red‐spotted grouper (7.85 ± 0.03 g fish?1). Nine semi‐purified diets were formulated containing varying protein levels (440–520 g kg?1, dry matter) and lipid levels (60–120 g kg?1, dry matter). The weight gain of juvenile Epinephelus akaara was affected by dietary protein (= .005) and its interaction with dietary lipid (= .020). Viscerosomatic index, intraperitoneal fat ratio and whole‐body lipid level increased with increasing dietary lipid level (p < .001). Nitrogen retention was not affected by dietary protein and lipid, while lipid retention decreased with increasing dietary lipid level (p < .001). The plasma blood urea nitrogen increased with increasing dietary protein level (= .003). This study showed that diet with 520 g kg?1 protein and 60 g kg?1 lipid with 30.58 mg kJ?1 P:E provided a maximal growth for this species. Moreover, an increase in dietary lipid levels (from 60 to 90 g kg?1) could reduce the protein requirement (from 520 to 480 g kg?1) without affecting the growth performance, while higher fat deposition was observed in fish fed high‐lipid diets.  相似文献

A growth trial was conducted to estimate the optimum concentration of dietary potassium (K) for grass carp (Ctenopharyngodon idella). Triplicate groups of grass carp (3.96 ± 0.06 g) were fed diets containing graded levels (0.87, 2.90, 5.37, 7.54, 9.87 and 12.4 g kg?1) of K for 8 weeks. Final body weight, weight gain and feed efficiency and gill Na+‐K+ ATPase activity were highest in fish fed with 9.87 g kg?1 dietary K and lowest in fish fed the basal diet (P < 0.05). The K contents in whole body and muscle were linearly increased up to the 9.87 g kg?1 dietary K and then levelled off beyond this level, whereas in scales and vertebrae up to the 7.54 g kg?1 dietary K (P < 0.05). However, dietary K levels had no significant effect on ash, Ca, P and Mg contents in whole body, scales, vertebrae or muscle. Analysis using polynomial regression of weight gain and gill Na+‐K+ ATPase activity and using the broken‐line regression of whole body K concentrations indicated that the adequate dietary K concentration for grass carp is about 9.45–9.99 g kg?1 diet.  相似文献

The administration of antimicrobials to control bacterial pathologies in Chilean scallop hatcheries is a frequent practice, but their effects on these cultures remained unknown. This study was undertaken to obtain information on the effect of the administration of florfenicol and oxytetracycline on the growth, survival and bacterial content of scallop larvae under farming conditions. Florfenicol‐treated cultures exhibited high survival rates (44% after 17 days of culture), whereas cultures not treated or treated with oxytetracycline collapsed after 11 days of culture. Surprisingly, no significant differences in the heterotrophic (Tukey test; = 0.226) and Vibrio (Tukey test; = 0.666) concentrations between the oxytetracycline‐treated and untreated larval cultures were observed. Otherwise, florfenicol administered directly into rearing tanks produced significantly higher larval growth (Tukey test; = 0.0001) and survival (Tukey test; = 0.011) than bath treatment. When 2 and 4 mg L?1 of florfenicol were compared, no significant differences in growth (t‐test; = 0.4596) and survival (Tukey test; = 0.057) were observed, suggesting that a concentration of 2 mg L?1 is sufficient to ensure larval production. The present results demonstrate the efficacy of florfenicol‐based therapy to increase larval survival and growth at commercial scale and prompt the necessity to standardize its use in Chilean scallop hatcheries.  相似文献

This study investigated the effects of levan produced by Bacillus licheniformis FRI MY‐55 on growth performance, intestinal count of viable bacteria, immune status, pathogen resistance and body composition of orange‐spotted grouper (Epinephelus coioides). Orange‐spotted grouper were fed diets supplemented with levan at concentrations of 0 (control), 5.0, 10.0, 25.0 and 50.0 g kg?1 for 12 weeks. The final weight and per cent weight gain were significantly higher in the 25.0 g kg?1 levan‐supplemented group than in the control group (< 0.05). All levan‐supplemented diets significantly decreased the count of total viable aerobic bacteria and Vibrio spp. in the intestines of groupers (< 0.05). Serum total protein levels, globulin levels, lysozyme activity and survival rate of orange‐spotted grouper after challenge with V. harveyi were significantly higher in the 25.0 g kg?1 levan‐supplemented group than in the control group (< 0.05). This study also found that the 10.0, 25.0 and 50.0 g kg?1 levan‐supplemented diets significantly increased the crude protein level in the body composition of orange‐spotted grouper. Overall, the results of this study indicate that dietary levan (25.0 g kg?1) could be an effective method for enhancing the growth performance and disease resistance in orange‐spotted grouper.  相似文献

The effects of salinity on plasma osmolality, branchial chloride cell density, feed consumption and conversion and growth performance of yellowtail kingfish (Seriola lalandi) were evaluated. Fish (11.6 ± 0.6 g) were kept for 29 days at 14, 18, 22, 26 (experimental) and 30 g L?1 (control) salinity in independent, pilot‐scale recirculation aquaculture systems. No differences in plasma osmolality or chloride cell numbers in gills were observed, pointing to a strong osmoregulatory capacity in the juveniles. Fish at 14, 18 and 22 g L?1 (7.61 ± 0.19, 7.61 ± 0.01 and 7.61 ± 0.13% day?1, respectively) had higher growth rates than fish at 26 and 30 g L?1 (7.10 ± 0.05 and 6.97 ± 0.06% day?1 respectively). The higher growth rate at lower salinity resulted from increased feed intake; feed conversion was not different. An evaluation of the impact of salinity on growth rate of on‐growing stages (till market size) seems warranted to assess whether the profitable effects of low salinity persist in later stages of this important aquaculture species.  相似文献

Jatroha protein concentrate (JPC) was prepared by iso‐electric precipitation method. A 45‐day feeding trial was conducted for the nutritional evaluation of JPC and fermented Jatropha protein concentrate (FJPC) in Labeo rohita fingerlings. Seven iso‐nitrogenous (340 g kg?1 CP) and iso‐energetic (17 MJ kg?1) diets such as C (soy protein concentrate), T1 (50 g kg?1 JPC), T2 (100 g kg?1 JPC), T3 (200 g kg?1 JPC), T4 (50 g kg?1 FJPC), T5 (100 g kg?1 FJPC) and T6 (200 g kg?1 FJPC) were fed to respective groups. The weight gain % and protein effciency ratio showed a decreasing trend with higher inclusion of JPC whereas these parameters increased gradually on higher inclusion of FJPC. FCR showed the reverse trend. The protease, GOTand GPT activities of FJPC fed groups were significantly higher than their non‐fermented counterparts whereas a reverse trend was observed for LDH and MDH enzyme activities in muscle and liver. The survival rate was 100% in all the treatment groups except for T2 and T3. Phorbol esters (PEs) in FJPC was non‐detectable. Results suggested that solid state fermentation with Aspergillus niger is an effective method for removal of PEs and FJPC is a promising protein source in aquafeed.  相似文献

Two 8‐week growth trials were conducted in indoor recirculation system to evaluate the protein requirements for juvenile (3.70 ± 0.20 g) and pre‐adult (85.2 ± 0.70 g) gibel carp, Carassius auratus gibelio var. CAS III. Six isoenergetic diets were formulated for each trial using fish meal and casein as protein sources, and protein level was 250–450 g kg?1 in Trial 1 and 200–450 g kg?1 in Trial 2. With the increasing dietary protein, feeding rate (FR) and feed conversion ratio (FCR) significantly decreased (< 0.05). Weight gain (WG) increased first and then reached a plateau in 330–450 g kg?1 in Trial 1 (> 0.05), while decreased after the maximum value in 350 g kg?1 in Trial 2 (< 0.05). Productive protein values (PPVs) were lower in 370–450 g kg?1 in Trial 1 and 400–450 g kg?1 in Trial 2 (< 0.05). Increasing dietary protein level increased protein content and decreased lipid content in whole fish body and white muscle (< 0.05). Apparent digestibility coefficient of dry matters (ADCd) decreased, while apparent digestibility coefficient of protein (ADCp) increased in 370–450 g kg?1 in Trial 1 and 250–450 g kg?1 in Trial 2 (< 0.05). Trypsin activity significantly increased in 370–450 g kg?1 in Trial 1 (< 0.05) and was not affected in Trial 2 (> 0.05). Hepatic alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities in both trials increased when dietary protein was above 400 g kg?1 (< 0.05). Based on quadratic regression of WG, it was estimated that dietary protein requirement for maximum growth was 414 g kg?1 (digestible protein of 376 g kg?1) and 365 g kg?1 (digestible protein of 324 g kg?1) for juvenile (3.70 g) and pre‐adult gibel carp (85.2 g).  相似文献

This study was conducted to determine the effects of dietary cellulase addition on improving the nutritive value of Chlorella for juvenile crucian carp Carassius auratus (initial body weight: 2.99 ± 0.02 g, mean ± SEM). Five isonitrogenous and isoenergetic experimental diets were formulated to contain 0.0 (control), 0.5, 1.0, 1.5 and 2.0 g kg?1 cellulase, respectively. Each experimental diet was randomly assigned to triplicate groups with 25 juvenile fish per fibreglass tank for 8 weeks. The results showed that weight gain, specific growth rate, feed intake and the trypsin activity in the anterior intestine increased with increasing dietary cellulase to 1.5 g kg?1 and then declined with further addition. However, the mRNA expression levels of Mrf4 and Myf5, the apparent digestibility coefficients for dry matter, protein, energy and the majority of amino acids, and the activity of lipase in the anterior intestine were highest in fish fed the 1.0 g kg?1 cellulase diet, and then tended to decline with further cellulase supplementation. In conclusion, the optimal dietary cellulase supplementation level was 1.0–1.5 g kg?1, which can improve growth performance, digestive activities and nutrient digestibility in crucian carp.  相似文献

The effects of dietary calcium chloride (CaCl2) concentration on the growth, survival, moulting and body composition of Astacus leptodactylus were studied. Diets were prepared using supplementation of 0 (control), 30, 60 and 120 g kg?1 calcium chloride commercial trout larvae diet containing 50% protein and 12% lipid. Astacus leptodactylus larvae with an average total length of 22.0 ± 0.05 mm (TL) and weight of 0.53 ± 0.01 g obtained from eighteen ovigerous females collected from Seydisehir Sugla Dam lake in Konya, Turkey, were stocked in 0.2‐m2 aquariums at a rate of 50 crayfish m?2 and reared for 90 days. The experiments included four treatments (diets) with three replicates each. Calcium chloride exerted positive effects on growth, moulting frequency, feed efficiency and survival of narrow‐clawed crayfish. Crayfish fed with 60 g calcium kg?1 calcium chloride‐supplemented diet exhibited the highest weight gain and specific growth rate (SGR) but the lowest survival rate (63.3%) (P < 0.05). There were no significant differences (P > 0.05) in mean moisture (803.5 g kg?1) and protein content (177.0 g kg?1) in crayfish tail meat. However, lipid values were between 3.8 and 11.6 g kg?1 and significantly different among the diets (P < 0.05). Diet with 60 g kg?1 calcium chloride is recommended for the best growth of freshwater crayfish.  相似文献

The uptake of oxolinic acid by the rotifer Brachionus plicatilis, Artemia franciscana nauplii and metanauplii was studied as a function of its concentration in the enrichment medium and the duration of the enrichment period. An emulsion containing 5, 10, 20 or 30% (w/w) oxolinic acid was administered and the enrichment period lasted 4, 8, 12 or 36 h. Highest incorporation of oxolinic acid was achieved using a 20% emulsion and a 12 h enrichment for rotifers (205.05 ± 17.1 μg g?1 dry weight), a 24 h enrichment for nauplii (2528.8 ± 254.6 μg g?1 dry weight), and an 8 h enrichment for metanauplii (1236.58 ± 22.9 μg g?1 dry weight). Higher concentrations of oxolinic acid in the enrichment emulsion or longer enrichment times resulted in decreased survival. Two hours post enrichment the contents of the drug appeared significantly decreased. The concentration data of oxolinic acid were best fit to a two phase exponential elimination model, the first phase elimination half‐life (t1/2α) being 1.86, 1.08 and 1.74 and the terminal phase elimination half‐life (t1/2β) 26.83, 29.67 and 17.48 in rotifers, nauplii and metanauplii correspondingly. Enrichment with an emulsion containing 20% oxolinic acid is recommended employing a duration of 12, 24, or 8 h enrichment for rotifers, nauplii and metanauplii respectively, while enriched carriers should be used shortly after enrichment.  相似文献

With this study, we aimed to assess the safety of using Curcuma longa in fish feed. Juvenile Astyanax aff. bimaculatus (0.83 ± 0.04 g) were kept in 24 80‐L aquaria, at a density of 0.5 fish L?1 for 60 days. Six diets supplemented with 0.0, 20.0, 40.0, 60.0, 80.0 and 100.0 g kg?1 were evaluated. Histopathological analysis of the gills showed that there was an increasing linear effect of turmeric on the frequency of lamellar aneurysm, hypertrophy of mucous cells, oedema and necrosis. There was a quadratic effect of turmeric supplementation on the frequency of fusion and disruption of secondary lamellae, vacuolation and hypertrophy of the epithelial cells, and the estimated values to minimize these variables were 4.63, 25.93 and 24.46 g kg?1 respectively. There was a quadratic effect of turmeric on the gills index (IBRA), and the estimated value to minimize this index was 30.23 g kg?1. No effect of turmeric on the frequency of histopathological changes and organ index in fish liver was observed. Thus, we conclude that turmeric is not hepatotoxic for A. aff. bimaculatus; however, its use is safe at concentrations up to 30.23 g kg?1 diet as a function of its gill toxic effect at higher doses.  相似文献

Twelve experimental diets (D‐1 to D‐12) in a 4 × 3 factorial design (four protein levels: 250, 350, 400 and 450 g kg?1 and three lipid levels: 50, 100 and 150 g kg?1) were formulated. Carbohydrate level was constant at 250 g kg?1. Rohu fingerlings (average wt. 4.3 ± 0.02 g) were fed the experimental diets for 60 days in three replicates at 2% BW  day?1. Weight gain (%), specific growth rate (% day?1) and feed gain ratio (FGR) indicated that diets containing 450 g kg?1 protein and 100 or 150 g kg?1 lipid (diets D‐11 and D‐12) resulted in best performance, although results were not significantly different from those of diet D‐9 (400 g kg?1 protein and 150 g kg?1 lipid). Protein efficiency ratio was highest with diets D‐6 (350 g kg?1 protein and 150 g kg?1 lipid) and D‐9 (400 g kg?1 protein and 150 g kg?1 lipid) (P > 0.05) and declined with higher and lower protein diets at all levels of lipid tested. Elevated lipid level (50, 100 or 150 g kg?1) did not produce better FGR in diets containing 400 and 450 g kg?1 dietary protein (P > 0.05). The combined effects of protein and lipid were evident up to 400 g kg?1 protein. Growth and FGR showed consistent improvement with increased lipid levels from 50 to 150 g kg?1 at each protein level tested except with diets containing 450 g kg?1 protein. Apparent nutrient digestibility (for protein, lipid and energy) did not show significant variation among different dietary groups (P > 0.05). Whole body protein and lipid contents increased significantly (P > 0.05) with dietary protein level. The results of this study indicate that rohu fingerlings are adapted to utilize high protein in diets with varying efficiency. The fish could utilize lipid to spare protein but there is no significant advantage from this beyond the dietary protein level of 350–400 g kg?1 in terms of growth and body composition.  相似文献
