The present case study focused on the problems that affect the nitrification process at three commercial recirculating aquaculture systems (RAS) for salmonids with fixed-bed biofilters operating in Chile, where the main factors were found to be management problems: (1) large variations in daily feeding, which results in unstable nitrogenous compounds (TAN, NO2, NO3) concentration; (2) variable daily water exchange, producing unstable culture conditions (variations in pH and temperature); (3) high densities of culture, which results in overall bad culture conditions (high CO2 concentration, high amount of fine solids, high oxygen consumption). When properly managed, the RAS have proven to tolerate up to 15% of daily variation in feeding, as low as 10% of daily “new” water inlet, and densities as high as 60 kg fish/m3 without showing any nitrification problems. The results from this study demonstrates that maintaining good water quality is essential to secure an efficient growth of both the target species and the nitrifying bacteria, therefore, the production strategies should consider both the target species and the nitrification process requirements.  相似文献

The present research investigated the effect of carbon/nitrogen ratio (C/N ratio) control in ponds with or without substrate addition for periphyton development on production of giant freshwater prawn. C/N ratios of 10, 15 and 20 were investigated in 40 m− 2 ponds stocked with 2 prawn juveniles (5.023 ± 0.02 g) m− 2 with or without added substrates for periphyton development. The various treatment combinations of C/N ratio and periphyton substrate addition are abbreviated as ‘CN10’, ‘CN15’, ‘CN20’, ‘CN10 + P’, ‘CN15 + P’ and ‘CN20 + P’, P representing periphyton substrate. A locally formulated and prepared feed containing 30% crude protein with C/N ratio10 was applied. Tapioca starch was used as carbohydrate source for manipulating C/N ratio and applied to the water column separately from the feed. Increasing the C/N ratio from 10 to 20 reduced (P < 0.001) the total ammonia-nitrogen (TAN), nitrite–nitrogen (NO2–N) and nitrate–nitrogen (NO3–N) in water column and total Kjeldahl nitrogen (TKN) in sediment. The addition of substrates only influenced the NO2–N concentration in the water column (P < 0.001). Increasing the C/N ratio raised the total heterotrophic bacterial (THB) population in the water column, sediment and periphyton (P < 0.001). It also increased the dry matter (DM), ash free dry matter (AFDM), and chlorophyll a content of periphyton (P < 0.001). The lowest specific growth rate (SGR), the highest food conversion ratio (FCR), and the lowest protein efficiency ratio (PER) were recorded in treatment CN10 (P < 0.05). The addition of substrates did not influence size at harvest (P > 0.05) but improved the survival from 62.8 to 72% (P < 0.001). Increasing the C/N ratio from 10 to 20 increased the net yield by 40% and addition of substrate increased the net yield by 23%. The combination of C/N ratio control and substrate addition increased the net yield by 75% from 309 (CN10) to 540 (CN20 + P) kg ha− 1 (120 days)− 1. This 75% higher production concurred with (1) a lower inorganic nitrogen content in the water column, (2) a higher THB abundance supplying additional single cell protein to augment the prawn production, and (3) an improved periphyton productivity and quality.  相似文献

The aim of this study was to evaluate the short‐term effect of probiotic inoculation on the abundance of heterotrophic and ammonia‐oxidizing bacteria in mature biofloc, as well as on total suspended solids (TSS), chlorophyll‐a and nitrogenous compounds in water. A completely randomized design consisting of five treatments (three commercial probiotics, one native consortia and one control) was performed. At the beginning of the experiment (day 1), each treatment was inoculated with the respective probiotic: PondPlus® (PP), Efinol® PT (EF) and Epicin® ponds (EP), native consortia UE, whereas the control was not inoculated. Water parameters and bacterial abundance were evaluated at 0, 2, 4, 6 and 8 days. The addition of probiotics, either native or commercial, did not show any significant effect on the TSS, Chl‐a and colony‐forming unit (CFU) of heterotrophic bacteria when they were added to the systems containing mature biofloc. A significant increase in ammonium oxidizing bacteria was registered with the probiotics PP and EP, although the levels of total ammonia nitrogen, NO3‐N and NO2‐N were statistically similar among all treatments. Modifications on most of the parameters measured were associated with the factor of time, rather than the inclusion of probiotics. Results suggest that the bacterial conglomerates in mature stage contain well‐established bacterial communities that are difficult to be affected by the addition of probiotics.  相似文献

The growth rate of disk abalone, Haliotis discus hannai, energy consumption and changes in water quality were monitored in a pilot-scale recirculating aquaculture system (RAS) for 155 days. Baffles were installed in the RAS culture tanks to enlarge the attachment area and clean out solid waste materials automatically by hydraulic force only. The experimental disk abalones, of shell length 24.5 ± 0.5 mm, were cultured at three stocking densities, 700, 1300 and 1910 individuals/m2 bottom area, in triplicate. The abalones were fed with sea mustard, Undaria pinnatifida, once a week. The abalone feed conversion rates and daily growth rates ranged from 24.5 to 25.9 and 0.32 to 0.36%, respectively. Their daily shell increments and survival rates ranged from 67.7 to 78.6 μm/day and 87.6–92.2%, respectively. The growth in weight tended to decrease at a culture density of 1300 individuals/m2 bottom area, while shell increments and survival rates were acceptable at this density. The total power consumption for heating was 1185.4 kW, comprising 30.2% of the total power consumption, while the average water exchange rate was only 2.9% per day. The total ammonia nitrogen stabilized below 0.07 mg/L, after conditioning of the biofilter. The NO2–N, NO3–N and total suspended solid concentrations were also maintained within acceptable ranges for the normal growth of disk abalone. The use of the RAS with these newly designed culture tanks for disk abalone culture produced 1300 individuals/m2 bottom area with a water exchange rate of only 2.9% per day and used about one-tenth of the heat energy of a conventional flow-through system.  相似文献

Coral reef fish are collected from the wild and exhibited in aquaria worldwide. Some of the fish spawn in captivity; however, the eggs are usually neglected. In this study, we collected the eggs spawned naturally in the exhibit tanks, hatched and cultured them indoor in 2000‐L fibreglass tanks (initial density = 18 000 egg tank?1). We applied an inorganic fertilization method commonly used in freshwater fish culture in raising these coral reef fish larvae. We maintained inorganic phosphorus concentration at 100 μg P L?1 and inorganic nitrogen at 700 μg N L?1 daily in the fertilized group (n = 4), while the control tanks (n = 4) were fed with rotifers (10 ind mL?1). Chlorophyll a at particle sizes of both 0.45–20 μm and >20 μm, as well as NH3‐N, NO3‐N, and PO4‐P concentrations were significantly higher in the fertilized group than the control. Zooplankton in the size groups of 10–50 μm (mainly flagellates) and 50–100 μm (mainly ciliates) were abundant (about 10~60 ind mL?1) during 3–7 days in fertilized tanks. The average larval fish survival rate at 21 day after hatch in fertilized group was consistently higher than the control in two trials. The experiments demonstrated that the inorganic fertilization approach can be successfully adapted for coral reef fish culture in an aquarium to achieve sustainable exhibits.  相似文献

Series of trials in which mackerel (Scomber scombrus L.) were confined in keepnets at different stocking densities are described. From simple confinement trials it was found that 50% of the fish died after 48 h at a stocking density of 30 fish m−3, equivalent to 6.5 kg m−3. Trials in which fish were held at stocking densities, and for a duration, comparable to those experienced in a “dried up” purse seine prior to “slipping”, showed that up to 90% of “slipped” fish died within 48 h of release. The primary cause of death was probably physical damage, particularly skin loss, caused by abrasion, although there is some evidence that mackerel have a healing process which can accommodate minor skin abrasions. A tagging trial showed a small but significant increase in mortality due to the tagging procedure.  相似文献

To study the potential for transmission of Perkinsus karlssoni, an apicomplexan parasite, among bivalve hosts, a long-term laboratory-scale, closed, artificial seawater system for shellfish was designed. Species of shellfish utilised were as follows: bay scallops, Argopecten irradians; blue mussels, Mytilus edulis; eastern oysters, Crassostrea virginica; European oysters, Ostrea edulis; sea scallops, Placopecten megallanicus; softshell clams, Mya arenaria; and quahaugs, Mercenaria mercenaria. All shellfish used were juveniles with shell heights of 10–25 mm. The design was duplicated for control and experimental systems with a combined population of 1600 individual shellfish. The maximum biomass in each system was estimated at < 1 kg/m3. Each system included six, 400 litre rectangular fibreglass tanks, a modular pump and filter unit (particle and activated carbon filters and ultra-violet sterilisers), a biological filter and a refrigeration unit. The total volume of water for each system was 2300 litres of artificial seawater (Instant Ocean®). The mean water temperature of 22°C was achieved by thermostat-controlled room temperature. Salinity was maintained between 27 and 31‰ by addition of either freshwater or artificial seawater. Shellfish were fed daily a mixed diet of carboy-cultured algae and spray-dried algae. Mean values for water quality parameters in both systems were as follows: NH3 < 0·004 mg/litre; NO2 < 0·01 mg/litre; NO3 < 19·16 mg/litre; and pH 8·0–8·4. The system design was adequate in maintaining healthy bivalves for a period of 22 months, with the exception of the sea scallops which succumbed to warm water. Mean monthly shellfish survival rates were 79·9–100% in the control and 74·8–98·9% in the experimental system. A method of controlling water temperature other than via room temperature should reduce slight seasonal temperature fluctuations.  相似文献

The relative contributions of live Artemia metanauplii and an inert diet for growth of Senegalese sole larvae and postlarvae were assessed through the analysis of carbon stable isotopes ratios (δ13C) in both diets and whole larval tissue. Larvae were reared on four dietary regimes: 100% live prey (rotifers and Artemia), 100% inert formulated diet and two co-feeding regimes of 70:30 and 30:70 ratios of Artemia and inert diet, respectively. Larvae from the live food regime and both co-feeding regimes showed a steep increase in δ13C from 10 days after hatching (DAH) as a result of the onset and continuation of Artemia consumption. From 12 DAH fish larvae from all the regimes showed significant isotopic differences as their δ13C increased to final asymptotic values of − 15.1, − 15.6 and − 16.3‰ in the live food, 70:30 and 30:70 regimes, respectively. Carbon turnover rates in larvae from both live food and co-feeding regimes were relatively high (0.071 to 0.116 d− 1) but more than 90% of the observed change in fish tissue isotopic values was accounted for by the retention of carbon in new tissue growth. A two-source, one-isotope mixing model was applied to estimate the nutritional contribution of Artemia and inert diet to postlarvae growth in the co-feeding regimes. At 23 DAH, the relative contribution of live and inert diets to tissue growth in larvae was respectively, 88 and 12% for the 70:30 co-feeding regime and 73 and 27% for the 30:70 co-feeding regime. At 17 DAH, the estimated proportion of tissue carbon derived from the inert diet was higher at 23 and 38% for the 70:30 and 30:70 regimes, respectively. The results suggest that co-feeding regimes in Solea senegalensis larvae may be adjusted to meet ontogenetic changes in the capacity for larvae to utilise inert diets. The contrasting levels of carbon isotope discrimination between diet and tissue in larvae reared on either 100% live feed or 100% inert diet indicate relatively poor utilization of nutrients from the inert diet. The use of isotopic discrimination factors as potential indicators of the digestive physiological performance of a consuming organism in regards to its diet is discussed.  相似文献

  1. Sexual and asexual modes of coral propagation are used to produce materials to actively restore coral cover on degraded reefs. It is important to evaluate growth, survival and cost‐effectiveness prior to any large‐scale intervention.
  2. This study compared the growth, survival and cost of using sexually, compared with asexually, propagated Acropora granulosa at an in situ nursery and, subsequently, on degraded reef patches in the north‐western Philippines.
  3. For sexual propagation, gametes spawned from gravid A. granulosa colonies were collected, fertilized and reared at an outdoor hatchery facility until they were 200‐day‐old juveniles. For asexual propagation, coral nubbins (obtained from colonies used in the sexual propagation experiment) were reared in the hatchery for 1 month before being transferred to an in situ nursery.
  4. Sexually and asexually derived corals were reared at the in situ nursery and subsequently out‐planted to a degraded reef. Cumulative survival and growth of sexually derived corals were significantly higher than those of the asexually derived ones, both at the in situ nursery (for 200 days) and on coral bommies (for 382 days). At the in situ nursery, mean growth rates were 0.04 and 0.12 cm3 day?1 for the asexually produced and sexually produced corals, respectively, while those at the bommies were 0.7 and 1.04 cm3 day?1, respectively.
  5. Production cost (including collection, hatchery‐ and in situ nursery‐rearing) is more expensive in the sexual (US$2.79/coral) than in the asexual (US$2.62/coral) mode of propagation. However, after 13 months post‐out‐planting it was clear that, in the long run, the individual cost of sexually derived corals is cheaper than the asexual counterparts, US$20.01 vs. US$88.15, respectively, owing to better survival rates.
  6. Hence, sexual propagation using Acropora is a cost‐effective method with relatively high survival and growth that can be used in active restoration.

The lipid classes and the fatty acid (FA) compositions of the zooxanthellae, the host tissue, and intact coral were determined for the first time in a soft coral, Sinularia sp. The contents of monoalkyldiacylglycerol (MADAG), triacylglycerol, and polar lipids differed significantly between the zooxanthellae and the host fractions. The zooxanthellae were rich in polar lipids, whereas neutral lipids were concentrated in the host. MADAG comprised 35% of the host lipids and was practically absent in zooxanthellae. Hence, MADAG is only synthesized in animal tissues and serves as a biomarker for the host in the host–zooxanthellae association of these soft corals. Similar to the zooxanthellae of reef-building corals, the main FA in the zooxanthellae of Sinularia sp. were 18:4n-3, 20:5n-3, and 22:6n-3. In addition, 16:3n-4 and 16:4n-1 (8.9% in total) were found in these zooxanthellae. The ratios of 16:3n-4, 16:4n-1, 18:4n-3, 20:5n-3, and 22:6n-3 in the zooxanthellae to those in the host tissue were 4.2, 11.2, 10.1, 11.0, and 9.1, respectively. The proportions of some FA and lipid classes in the intact coral and its fractions showed that zooxanthellate lipids comprised 36 ± 15% of the total lipids in Sinularia sp. Two tetracosapolyenoic acids (24:5n-6 and 24:6n-3) are proposed as a biomarkers of the animal tissue and indicators of the purity of the zooxanthellae fractions from soft corals.  相似文献
