The efficacy and tolerance of a novel microbial 6‐phytase were investigated in rainbow trout, Oncorhynchus mykiss, and Nile tilapia, Oreochromis niloticus. Reference diets were sufficient in available phosphorus (P). The test diet limiting in available P was supplemented with phytase at 500, 1000, or 2000 phytase units/kg feed. The enzyme was effective in increasing total P apparent digestibility coefficient in relation to increasing the dose of phytase in rainbow trout and Nile tilapia. Zinc apparent digestibility improved in relation to phytase supplementation in rainbow trout. P release due to phytase supplementation ranged from 0.06 to 0.18% P/kg feed in rainbow trout and from 0.13 to 0.26% P/kg feed in Nile tilapia. A 58‐d performance trial was conducted to evaluate tolerance of fish to phytase supplementation. Dietary treatments consisted of a basal diet without phytase or supplemented with 2000 and 200,000 phytase units/kg feed. Results indicate that this novel microbial 6‐phytase is well tolerated by fish. Significant improvements for growth as well as feed conversion ratio were observed when the phytase was fed at 2000 phytase units/kg feed. This phytase is proven efficient in releasing P from phytate and could be added when plants are used for fish meal replacement in diets for salmonid and omnivorous fish.  相似文献

Experiments have been carried out aimed at clarifying variations in the digestibility of dietary nutrients in rainbow trout families and studying how differences in digestibility may be related to growth and feed utilisation at various growth rates. The digestibility of protein, lipid, carbohydrates (nitrogen-free extracts, NFE) and dry matter was analysed in two experiments involving eight rainbow trout families [Ab, Ba, Cd, Dc (first study); V, X, Y, Z (second study)]. In the first experiment rainbow trout were reared for 128 days at 13.0°C, and in the second experiment, they were reared for 84 days at 16.8°C. In both experiments, the fish were fed ad libitum and reared from an initial weight of 70–100 g to a final weight of 500–700 g. When the fish reached a weight of approximately 200 g, some individuals were moved to another experimental system in which the digestibility of protein, lipid, nitrogen-free extracts and dry matter was measured. Taken as a whole, our results indicate that selective breeding still offers a large potential for improved growth and feed utilisation in rainbow trout strains. In the first study, family Dc showed a higher specific growth rate (SGR) than the other three families (P < 0.05), and family Ba showed a lower feed conversion ratio (FCR) than family Ab (P < 0.05); there were no observed differences in digestibility despite some differences in growth. In the second study, family Y grew faster than all of the other families (P < 0.05), and family Z grew faster than families V and X (P < 0.01). A comparable pattern was seen for FCR, with family Y utilising feed better than family V (P < 0.05), and families V, Y and Z performing better than family X (P < 0.001). Protein digestibility was higher in the two fastest growing families (Y and Z) than in the slower growing family X (P < 0.05), while lipid digestion was higher in family Y than in family V (P < 0.05). A comparison of the results from both experiments revealed that protein digestibility in particular was closely related to the SGR and the FCR at high growth rates. However, despite the advantageous protein digestibility on fish growth, analysis of the protein retention efficiency (PRE) showed that when protein was ingested in relatively large amounts, as in the fastest growing families, the “excess” nitrogen was excreted and therefore did not contribute to protein deposition in the fish body. Hence, the potential weight gain offered by improved protein digestibility does not materialise when the protein intake is above a certain level. Other factors must therefore explain the positive relation between fast growth and high protein digestibility.  相似文献

The effect of replacing fish meal with soybean meal (SBM) pretreated with phytase on feeds of juvenile rainbow trout was evaluated in a 90-day feeding trial. The rainbow trout (initial body weight, 4.01 ± 0.02 g) were fed five isonitrogenous (crude protein, 44.97%) and isolipidic (crude lipid, 13.42%) feeds. Diets were formulated to contain phytase-pretreated SBM replacing 0, 20, 40, 60 and 80% of fish meal protein, respectively. The results showed that there was no significant difference in weight gain (WG) among fish fed S0, S20, S40 and S60 diets; however, a significant reduction of this variable occurred when 80% of fish meal protein was replaced by phytase-treated SBM (P < 0.05). Similarly, specific growth rate and protein efficiency ratio had a similar trend with WG. Apparent digestibility coefficient (ADC) of protein and lipid in the S80 diet was significantly lower than that of the other diets, and the ADC of phosphorus significantly increased with the increase of dietary phytase-treated SBM level. No significant differences among treatments were detected for moisture, protein, lipid and ash content in whole body and muscle samples. Nitrogen and phosphorus excretion indicated that fish meal replacement by phytase-treated SBM led to an increase in nitrogen excretion, but led to a reduction in total phosphorus excretion. The results of the present study show that 60% of fish meal could be replaced by phytase-treated SBM in diets of juvenile rainbow trout without compromising weight gain or feed efficiency. A quadratic equation according to regression analysis of weight gain against dietary phytase-treated SBM level indicated that the optimal level of dietary phytase-treated SBM replacement for maximum growth was 26.90%.  相似文献

Three experiments were conducted to evaluate the effects of microbial phytase (Ronozyme P(L) and dosage on apparent digestibility coefficients of dry matter, crude protein, amino acids, and minerals in soybean product-based diets for rainbow trout Oncorhynchus mykiss. In experiments 1 and 2, fish were fed soy protein concentrate and soybean meal-based semi-purified diets, respectively. In experiment 3, fish were fed soybean meal-based practical diets. A total of 900 fish (average body weight 100.1 ± 7.4 g) were stocked into 30 150-L fiberglass tanks with 30 fish per tank and two tanks per diet. Phytase was supplemented into soybean product-based diets at 0, 500, 1,000, 2,000, and 4,000 FTU/kg diet. Yttrium oxide (0.01%) was used as inert marker. Feces were collected by hand stripping three times at 3-d intervals. Results of experiments 1 and 2 showed that phytase supplementation significantly increased apparent digestibility coefficients of minerals (except copper and iron), but not dry matter. Experiment 3 showed that phytase supplementation significantly increased apparent digestibility coefficients of dry matter, crude protein, amino acids (except tryptophan and tyrosine), and minerals (except copper and iron). The optimum level of phytase supplementation in rainbow trout diets was approximately 500 FTU/kg diet.  相似文献

Abstract

Two experiments were conducted to evaluate the effects of microbial phytase in corn distiller's dried grain with solubles (DDGS) on apparent digestibility coefficients (ADCs) and growth performance of rainbow trout, Oncorhynchus mykiss. In experiment 1, DDGS was supplemented with phytase at 0,300,600,900, and 1200 FTU/kg diet. A total of 180 fish (129.1±9.3 g) were stocked into six 140-L tanks with 30 fish/tank. Fish were assigned randomly to a reference diet and each of the 5 diets containing DDGS. Yttrium oxide was used as an inert marker. Results showed that ADCs in DDGS supplemented with different dosages of phytase were: dry matter, 49.1-58.6%; crude fat, 78.9-88.9%; crude protein, 80.0-91.9%; gross energy, 50.5-66.6%; minerals, -7.3-99.7%; and amino acids: 73.9 to 96.8%. In experiment 2, abasal diet containing 15% DDGS supplemented with lysine and methionine was used to determine if trace mineral supplemental levels in rainbow trout diets could be reduced if microbial phytase was supplemented. Six diets were made with trace mineral premix supplementation at 0.1, 0.08, 0.06, 0.04, 0.02, and 0%. Phytase was not supplemented in the basal diet, but supplemented at 500 FTU/kg diet in all other diets. Ten-week results showed that there were no significant differences in fish weight gain, feed conversion ratio, and survival (P < 0.05). There were no significant differences in fish body composition and apparent nutrient retention among fish fed all diets, except that fish fed a diet without trace mineral supplementation had the lowest zinc level and the highest manganese retention. Results indicated that phytase was effective in releasing most of minerals, and that trace mineral supplementation level could be reduced when phytase was used in rainbow trout diets.  相似文献

A feeding trial was conducted to study the effect of partial replacement of dietary monocalcium phosphate (MCP) with neutral phytase on growth performance and phosphorus digestibility in gibel carp, Carassius auratus gibelio (Bloch). Control diet was prepared with 2% MCP but without phytase (P0). Other three experimental diets were prepared by replacement of MCP by 25%, 50% and 75% respectively in comparison with control with supplementation of neutral phytase at 500 U kg?1 diet in each and designated as P25, P50 and P75 respectively. Gibel carp (initial body weight of 30.22 ± 1.98 g) were reared in twelve 300‐L cylindrical fibreglass tanks provided with filtered flow‐through tap water at 26–28°C. After 8‐week experiment, gibel carp fed with P50 had no obvious differences from the control group on weight gain (WG), specific growth rate (SGR), feed conversion ratio (FCR), protein efficiency rate (PER) and survival rate. Phytase supplementation did not affect body compositions or muscle compositions. Crude protein and phosphorus (P) contents in the faeces of fish fed with the phytase‐supplemented diets were significantly lower than those of the control group. The apparent digestibility coefficients (ADCs) of crude protein and P in gibel carp were increased when fish fed with the diets in which MCP was replaced by neutral phytase. This study suggested that partial replacement of dietary MCP at 50% with neutral phytase was considered as a recommended dietary supplemental level and increased dietary P and protein availability.  相似文献

采用喷涂法和预处理两种方法,在豆粕型金鳟饲料中添加不同浓度的植酸酶,通过90 d的饲喂试验,研究植酸酶对金鳟(Oncorhynchus mykiss Walbaum)体成分及其椎骨中钙、磷含量的影响。结果显示:喷涂法和预处理法添加植酸酶的处理组与对照组相比,粗蛋白、粗脂肪的含量均有所下降;喷涂法添加植酸酶的处理组的鱼体粗灰分、磷和钙含量均高于对照组,其中,喷涂法添加1500 U/Kg植酸酶组鱼体磷含量、灰分含量和钙含量显著高于对照组(P(0.05),且此组鱼体磷高于其它试验组;喷涂法各组椎骨中粗灰分、磷含量较对照组提高显著(P(0.05),2500 U/kg植酸酶组椎骨磷含量最高,椎骨中钙含量除D1和D8组外,添加植酸酶的处理组均较对照组提高显著(P(0.05)。预处理法添加植酸酶的处理组与对照组相比,鱼体和椎骨中粗灰分、磷和钙含量均有显著提高(P(0.05),但植酸酶试验组间椎骨中粗灰分、磷含量和钙含量差异均不显著(P(0.05)。结果表明,金鳟饲料喷涂法添加植酸酶1500~2500 U/kg浓度最适,预处理法添加植酸酶500 U/kg浓度较为宜。  相似文献

Microbial phytase (Natuphos 5000G) was supplemented into barley, canola meal, wheat and wheat middlings at 500 FTU kg–1 diet to test the effectiveness of the phytase on digestibility of crude protein (CP), gross energy (GE) and minerals for rainbow trout. A total of 180 rainbow trout (initial mean body weight 223.8 ± 17.9 g) were stocked into ten 40-L fibreglass digestibility tanks with 18 fish per tank. Duplicate tanks were assigned to a reference diet and a single tank was assigned randomly to each of the eight diets made from these four ingredients supplemented with and without phytase. The collection of faeces lasted for 2 weeks. Faeces collected in each week represented a replicate and they were analysed separately. The mean apparent digestibility coefficients (average of four ingredients) in phytase supplemented and nonsupplemented ingredients were: CP, 97.3% and 96.5%; GE, 62.8% and 53.1%; calcium (Ca), 38.3% and –5.3%; magnesium (Mg), 72.4% and 50.3%; manganese (Mn), 31.0% and –4.2%; total-phosphorus (total-P), 71.1% and 47.6%; phytate-phosphorus (phytate-P), 87.5% and 22.3%; copper (Cu), 38.3% and 26.4%; potassium (K), 97.1% and 97.0%; sulphur (S), 90.2% and 88.6%; zinc (Zn), 12.3% and –16.6%. A two-tailed t -test indicated that phytase supplementation significantly ( P  < 0.05) improved the digestibility of Ca, Mg, Mn, total-P, phytate-P, and GE. The efficacy of phytase on nutrient digestibility also depended on the type of ingredient as measured in rainbow trout.  相似文献

This study evaluated the effect and phosphorus equivalency value of dietary microbial phytase based on growth performance and nutrient utilization of Channel catfish (Ictalurus punctatus). An all plant‐protein basal diet containing 7.9 g/kg total Phosphorus (P) (Phytate P of 4.5 g/kg) was supplemented with graded levels 0, 300, 500, 1,000, 1,500 and 2,000 U phytase/kg of diet or with 3, 5, 8, 12 g/kg of Ca(H2PO4)2, namely with 0.7, 1.2, 2.0, 3.09 g/kg P as Ca(H2PO4)2. Triplicate groups of Channel catfish (Ictalurus punctatus) juveniles (initial weight, 1.70 ± 0.04 g) were fed the experiment diets for 90 days. The results indicated that dietary phytase supplementation significantly improved growth performance, apparent digestibility coefficient of P and P retention, and decreased feed conversion ratio (FCR), faecal P content and P excretion per gain. Line relation was found between the effects of P and phytase supplementation levels, and the linear response equations of the effects of P and phytase levels were generated and used to calculate the P equivalency values. The average function of P equivalency values (Y, g/kg) of microbial phytase (X, U/kg of diet) was developed: Y = 0.1695 + 0.0021X; R= 0.996. The results showed that about 0.38 g of P/kg diet was released by per 100 U of phytase over the range of 300–2,000 U of phytase/kg diet for Channel catfish (Ictalurus punctatus) fed all plant‐protein diet.  相似文献

In this study, feeding experiment and subsequent digestibility trial were performed to investigate the utilization of extruded soybean meal (SBM) and corn gluten meal (CGM) as feed ingredients for juvenile rainbow trout. Plant ingredients have undergone extrusion at low temperature (100°C, LT) or high temperature (150°C, HT) for 30 s. Four isonitrogenous (44%, crude protein) and isolipidic (14%, crude lipid) diets were formulated. Control diet is fishmeal based while a combined (1:1) non‐extruded SBM and CGM for NE diet, LT SBM and LT CGM for LT diet and HT SBM and HT CGM for HT diet. Two hundred forty rainbow trout juveniles (7.8 g average body weight) were randomly divided into 12 rectangular 60 L glass aquaria and offered four different diets in triplicate. Fish were fed at satiation twice a day, six days a week for 12 weeks. Phosphorus in phytic acid level of extruded ingredients decreased through extrusion cooking. Final weight, weight gain, SGR and PER of fish fed HT diet were significantly (p < .05) higher than those fed with NE diet. Apparent digestibility coefficient for protein of LT and HT diets is significantly higher than of NE diet. The results of this study demonstrated that HT extruded SBM and CGM are suitable feed ingredients for rainbow trout diet without compromising fish growth, feed utilization and fish body composition.  相似文献
