草鱼,链鳙鱼卵受精率计算方法的探讨 总被引:1,自引:1,他引:0
系统地进行了家鱼卵受精卵受精率统计试验,描述了爱卵与未受精卵的发育形态特征。计算受精率的取样时间应为多细胞期到高囊胚期,随机取卵100-200粒,重复两次,求其平均值。本结果可作为青鱼、草鱼、鲢、鳙鱼卵受精率计算方法标准化的依据。 相似文献

用显微注射技术导入外源DNA对鲤鱼和虹鳟鱼受精卵孵化率的影响 总被引:2,自引:0,他引:2
本报道了利用显微注射技术导入外源基因分别对鲤鱼和虹鳟鱼授精卵孵化率影响的研究。从1993年-1996年4年中,我们用显微注射方法,将全鱼生长激素基因导入虹鳟鱼授精卵(包括囊胚期卵)5,676粒,孵出鱼苗364尾,孵化率为6.4%(4年平均,虹鳟鱼正常孵化率为80%);而用这种方法,将全鱼生长激素基因导入鲤鱼授精卵12,196粒,孵出鱼苗4,290,孵化率为34.0%(4年平均,鲤鱼正常孵化率为70%左右)。实验结果:显微注射技术在虹鳟鱼和鲤鱼两种受精卵上的效果明显不同,斑点杂交和SouthernBlot杂交结果相近。 相似文献

广东省兴宁县鱼苗场于今年10月16日(水温24.0~25.5℃),催产同年8月份已经产过卵的草鱼和鲢鱼亲鱼(各计四组),产卵率均为100%。其中,草鱼产卵51.6万粒,受精率82.8%,下塘鱼苗28万尾;鲢鱼产卵41.2万粒,受精率62.5%,下塘鱼苗21万尾。 相似文献

采用双筒解剖镜,对胭脂鱼人工授精的胚胎发育进行观察。胭脂鱼成熟卵桔黄色,卵径1.8~2.0mm。受精后卵膜遇水膨胀,卵径增至3.8~4.0 mm。在水温17~21℃条件下,多数受精卵经163 h 5 min孵出。受精卵在受精后72 min进入胚盘期,受精后2 h 13 min进入2细胞期,受精后7 h 8 min进入囊胚期,受精后14 h 6 min进入原肠期,受精后32 h 40 min进入神经胚期,受精后61 h 50 min进入尾芽期,受精后163h 5 min 50%以上的胚体孵出。刚出膜的仔鱼长约9 mm。 相似文献

选择对草鱼出血病病毒不敏感的草鱼肝细胞进行细胞培养,建立了草鱼肝细胞株,命名为GLA。对GLA进行攻毒,发现其对FRV有抗性。采用电融合结合继代移核的方法,将GLA细胞,移植于草鱼未受精卵内,获得开口前的幼鱼8尾和存活仔鱼1尾,说明此方法能提高细胞的发育功能。 相似文献

Abstract. Laboratory examinations of pond-reared common carp, Cyprinus carpio L., revealed a close correlation between the prevalence of swimbladder inflammation (SBI), renal sphaerosporosis and infection by C-blood-protozoan among the carp fry. In impression smears as well as light and electron microscopic preparations we detected developmental stages of intercellular protozoa, mainly in the loose fibrous tissue of the swimbladder. The parasites multiplied by internal budding so that 20–46 secondary cells were formed in each primary cell and two tertiary cells were formed in each secondary cell. The final stage of development was a unit consisting of a secondary cell enclosing two tertiary cells, i.e. a so-called triple formation, which bore a close resemblance to the early sporogonic stages of the renal sphaerosporan Sphaerospora angulata Fujita, 1912. Certain morphological similarities and the frequent simultaneous presence of the swimbladder protozoan, C-bloodprotozoan, and S. angulata in hosts with clinical SBI have led us to postulate that the former two parasites could represent the hitherto unknown presporogonic stages of S. angulata. In view of the pathological changes caused by the parasites in the hosts with clinical SBI, and negative bacteriological and virological findings we have postulated that the swimbladder protozoan is the primary cause of SBI in carp fry. 相似文献

Grass carp fry ranging in weight from 9 mg to 300 mg were used in experiments to determine their response to diets of coconut meal and a 1:3 mixture, by weight, of coconut meal and zooplankton at 28 and 33°C. The food intake was generally higher in those fry maintained on the mixed diet at 33°C. The rate of growth was always found to be higher in those fry maintained on a mixed diet at 28°C. The rate of weight increase of fry expressed as a percentage of the initial body weight tended to decrease in fry maintained on a diet of coconut meal, whilst the reverse was observed for others. A maximum food conversion ratio of 1.29 was recorded for fry maintained on a mixed diet of coconut meal and zooplankton at 28°C. Fry of grass carp changed from an almost exclusively carnivorous to a herbivorous one at a length of 25–30 mm. The evacuation time was both temperature- and weight-dependent, decreasing with increasing weight at a particular temperature. 相似文献

草鱼呼肠孤病毒(Grass carp reovirus, GCRV)可引发草鱼(Ctenopharyngodon idellus)出血病,导致高死亡率。草鱼吻端成纤维细胞(Grass carp snout fibroblast cells, PSF)是GCRV的敏感细胞系。JAM-A (Junctional adhesion molecule A)为免疫球蛋白超家族成员,是多种病毒的细胞受体。本研究在前期克隆到草鱼3种jam-a基因,命名为gcjam-a1,gcjam-a2和gcjam-a3。在获取ORF序列的基础上,利用qRT-PCR分析了3种gcjam-a在草鱼胚胎及幼鱼不同发育时期及PSF细胞中受GCRV(GD108株)感染前后的表达模式。结果显示,检测的13个胚胎及幼鱼发育时期中,gcjam-a1在未受精卵中高表达,在受精卵至出膜前的胚胎表达水平均较低;从出膜后1~3 d表达量开始上升;出膜6~15 d均呈高水平表达。gcjam-a2与gcjam-a3在草鱼胚胎及幼鱼发育各阶段表达水平较低。在无病毒感染的PSF细胞中,gcjam-a只有少量表达。受GCRV-GD108感染后,病毒S7基因在PSF细胞中的拷贝数随时间呈显著上调趋势,gcjam-a的表达量也有不同程度的上调,mRNA上调水平为gcjam-a1>gcjam-a2>gcjam-a3。本研究证实了3种gcjam-a基因在PSF细胞中的表达均与GCRV-GD108感染相关,其中,gcjam-a1的表达水平受GCRV-GD108感染影响最大,同时,它在孵化出膜后表达上升,推测它可能与病毒的感染更相关。gcjam-a1可作为下一步GCRV与宿主互作研究中的候选分子。 相似文献
