石斑鱼循环水养殖系统微生物群落结构   总被引:3,自引:1,他引:2  
通过16S rRNA基因片段高通量测序研究了褐点石斑鱼(Epinephelus fuscoguttatus)和东星斑(Plectropomus leopardus)循环水养殖系统水体细菌种群结构。结果显示石斑鱼循环水养殖系统中优势细菌类群为变形菌门(Proteobacteria)、拟杆菌门(Bacteroidetes)、蓝藻门(Cyanobacteria)、梭杆菌门(Fusobacteria)、厚壁菌门(Firmicutes)和硝化螺旋菌门(Nitrospira)。其中养殖塘、固液分离池、沉淀池和蛋白分离池中主要优势细菌为γ-变形菌纲(Gammaproteobacteria),紫外消毒池和补氧池中则以a-变形菌纲(Alphaproteobacteria)为主,而生物滤池中两者比例接近,均为优势种群。从养殖塘到补氧池细菌多样性指数先降低后升高,生物滤池中达到最高值,之后再次下降至紫外消毒池中达到最低值,补氧池中有所回升。PCoA分析和聚类分析结果表明,养殖水在紫外消毒前后其细菌群落组成有着明显的差异。水质理化指标检测表明,经过净化处理后,循环系统的养殖水溶解氧含量上升,氨氮和亚硝酸盐含量下降并维持在较低的浓度。环境因子与细菌群落结构相关性分析结果表明,磷酸盐、pH、溶解氧和温度等指标可能对细菌群落结构影响相对较大,但未发现两者间有很显著的相关性,这可能与实验样本较少有关。  相似文献

Mucins are large glycoproteins that cover epithelial surfaces of the body and play important roles in prevention of inflammatory and various infectious diseases. In this study, five membrane‐bound and seven secreted mucin genes in the channel catfish were identified. All these identified mucin genes possess at least one PTS, von Willebrand D (VWD) or SEA domains. The expression of the 12 mucin genes in channel catfish was first studied with infection of Edwardsiella ictaluri and Flavobacterium columnare. Expression difference in MUC13a, MUC13, MUC2 and MUC5b was found in the intestine after E. ictaluri infection. Eight mucin gene expressions (except MUC3a, MUC2, MUC4 and MUC5f) were up‐regulated at 4 hr and down‐regulated after 24 hr in the gill with F. columnare infection. Expression level of MUC2 gene was up‐regulated in the intestine with E. ictaluri infection without no significant change in the gill under the F. columnare infection, which indicate that MUC2 is tissue‐specific gene expression and has different immune respond to two bacterial challenge. Taken together, the study showed mucin from the gill by F. columnare challenge induced an obvious response than mucin from the intestine with E. ictaluri infection.  相似文献

An 86‐day growth trial was conducted to investigate the effect of dietary oxidized fish oil on the growth and cytopathology of juvenile channel catfish (Ictalurus punctatus). Four diets containing 0 g kg?1 (control: fresh fish oil), 30 g kg?1 (low‐oxidized oil group), 60 g kg?1 (medium‐oxidized oil group) and 90 g kg?1 (high‐oxidized oil group) graded oxidized oil levels with the same dietary lipid level (90 g kg?1 diet) were evaluated. The results show that the specific growth rate decreased with increasing dietary oxidized oil level (< 0.05). All examined liver and kidney tissues in all dose groups exhibited what appeared as dose‐dependent cellular modifications. In addition, lipid droplet accumulation in the hepatocytes of fish in all dose groups was increased, and their localizations were distinctly different between all dose groups. The ultrastructural changes suggest the progression of mitochondrial vacuolation, especially in the renal tubules, in all dose groups. These results reveal a previously underappreciated effect of dietary oxidized fish oil on channel catfish kidneys. Overall, a series of nutriphysiological responses were adversely affected by exposure to dietary oxidized fish oil, and the corresponding interference patterns on the metabolism and transport of nutrients within cells were observed.  相似文献

This study investigated the effects of dietary lysolecithin on growth performance, feed utilization and metabolic responses of channel catfish (Ictalurus punctatus). Fish (initial weight: 14.77 ± 0.45 g) were randomly fed one of five practical diets supplemented with graded levels of lysolecithin (0, 125, 250, 375 and 500 mg/kg) in quadruplicate groups for 10 weeks. Results showed that no significant differences in weight gain, condition factor and viscerosomatic index among all the treatments. Fish fed diet supplemented with 250 mg/kg lysolecithin had significantly decreased feed conversion ratio and significantly increased protein efficiency ratio compared to that fed the control diet. The group with 250 mg/kg of dietary lysolecithin had higher protein and lower lipid contents in the whole body, and lower lipid content in the liver than those in the control group. Groups supplemented with 250–500 mg/kg of dietary lysolecithin had significantly higher gastric and intestinal lipase activities than the control group. Dietary lysolecithin at the level of 250 mg/kg significantly increased the activities of Na+, K+‐ATPase, alkaline phosphatase, total antioxidative capacity, total superoxide dismutase, glutathione peroxidase and catalase and significantly decreased the content of malondialdehyde compared to the control diet. In conclusion, dietary supplementation of lysolecithin could confer benefits of feed utilization, body composition and antioxidative capacity of channel catfish.  相似文献

在河蟹青虾混养池塘构建封闭式循环水养殖系统,并对其净化效果和养殖效益进行了初步研究。结果表明:该系统对水体TN、TP、NH4+-N和CODMn的平均净化效率分别为27.33%、56.14%、43.91%和39.59%。经湿地净化后的水质能够达到GB3838-2002地表水Ⅲ类排放标准,平均纯收益达53769元/hm2。  相似文献

研究利用循环水养殖设备和多层抽屉式养殖箱构建一种新型的立体循环水养殖系统,对马粪海胆进行为期90 d的养殖试验.试验结果表明,养殖用水在循环使用的情况下,水质指标处于良好状态:NH+4-N为0.02~0.15 mg/L,NO-2-N为0.01~0.35 mg/L,pH值为7.7~8.2;马粪海胆养殖密度可达1 444.23 g/屉(约20 L),特定生长率(SGR)为0.33,成活率为89.15%.  相似文献

2005—2010年,对天津市天祥水产有限公司封闭循环水养殖系统水质进行了检测。结果显示:养殖池塘是否覆膜、覆膜面积大小,均会使整个养殖系统中的总硬度(TH)、总碱度(AT)、NH3-N和PO4-P在时空上产生差异。时间上,它们年份间差异极显著(P<0.01);空间上,各采样点的AT和NH3-N差异显著(P<0.05)。覆膜使系统的TH和AT稳定性减弱,CV上升了166.5%~241.7%,这种情况在养殖中期尤为突出,但对NH3-N和PO4-P稳定性的影响并不明显。养殖系统中较大面积的覆膜(43.65%)对养殖池塘TH、AT和NH3-N稳定性的影响比非覆膜池塘的大,CV分别高出了75.9%、135.6%和21.6%;而小面积(19.2%)覆膜的养殖池塘相对非覆膜池塘稳定,CV分别降低了71.9%、41.5%和91.4%。导致它们间差异及稳定性变异的原因主要归结于覆膜影响了物质与土壤间的物质交换及其间的生物活性强度。  相似文献

为探究凡纳滨对虾(Litopenaeus vannamei)工厂化循环水养殖系统的养殖水体水质情况以及微生物菌群的组成结构,本研究利用高通量测序技术和生物信息学分析手段,测定凡纳滨对虾工厂化循环水养殖过程一级移动床生物净化、二级固定床生物净化、养殖水体的水质指标、水体和生物净化载体以及对虾肠道微生物菌群的组成。结果显示,水体的氨氮(NH4+-N)和亚硝态氮(NO2-N)质量浓度显著降低,分别为0.85和0.21 mg/L。养殖系统水体、生物净化载体和虾肠道样品中共有的优势菌为变形菌门(Proteobacteria)、拟杆菌门(Bacteroidetes),此外,一级、二级生物净化系统水体中的放线菌门(Actinobacteria)为优势菌,生物净化载体中浮霉菌门(Planctomycetes)和硝化螺旋菌门(Nitrospirae)为优势菌;对虾肠道中的厚壁菌门(Firmicutes)为优势菌。另外,对虾养殖循环水系统中生物净化载体上的细菌物种含量比水样中的细菌物种少,但微生物多样性高于养殖水体,...  相似文献

Recirculating aquaculture systems (RASs) are an efficient method for developing sustainable aquaculture and the development of a feed strategy for RASs could be a key factor for the success of culturing Oreochromis niloticus. The daily protein intake (DPI) mathematical function related to body weight (BW) was designed (DPI = ?3.818 ln (BW) + 30.158) named DPI 1.0 = 100%. Three feeding rates were established, DPI 1.4, 1.2 and 1.0, and were implemented in an RAS production cycle (100 fish/m3) for 34 weeks. The final growth measurements were 908.0 ± 57.9 g (DPI 1.4), 887.0 ± 113.5 g (DPI 1.2) and 702.2 ± 38.1 g (DPI 1.0) (p < .05), with feed conversion rates (FCRs) of 2.3, 1.9 and 1.9, respectively; the survival was ≥ 98.3% (p > .05) for all treatments. DPI levels conditioned the growth of the fish and the proportion of lipid: protein in the bodies of O. niloticus but not in the fillets. The mathematical function DPI 1.2 = 120% (DPI = ?4.582 ln (BW) + 36.19) used in the RAS at the 40% protein level improved fish growth, the protein efficiency ratio and FCRs. The DPI values were estimated for 49 different cases and feeding strategies showed lower values than most of the different DPI mathematical functions used and cited in this work. Therefore, DPI metric is a good tool that provides the specific requirements of protein for O. niloticus growth, which can enable farmers to ration feed and improve energy use (kWh / fish) and facilitate the integration of RAS with horticulture.  相似文献

为探究不同养殖模式下斑点叉尾鮰的生长性能和肌肉品质特性,随机选取传统池塘(TP)、高密度循环水系统(HD-RAS)和水泥池塘(CP)养殖模式下的鲜活斑点叉尾鮰,测定其生长状况,以及肌肉营养成分、颜色、持水力和质构特性。结果显示,TP和HD-RAS的斑点叉尾鮰的增重率和特定生长率差异不显著,但均高于CP;CP的肝体比(2.35%±0.22%)显著高于TP和HD-RAS;与TP相比,HD-RAS和CP肌肉的粗脂肪含量更低,粗蛋白和灰分含量更高;HD-RAS肌肉的亮度值(L*)(58.18±3.56)、红度值(a*)(6.05±1.08)最高,CP最低,分别为48.15±6.04和4.81±0.84;传统池塘养殖的斑点叉尾鮰熟肉率最低,离心损失、滴水损失和失水率最高,HD-RAS和CP之间差异不显著;HD-RAS和CP肌肉的硬度、咀嚼性和弹性显著高于TP,但3处理组间的内聚性和回复性没有显著差异。研究表明,与传统池塘养殖模式相比,水泥池塘养殖模式易造成斑点叉尾鮰生长减慢,而高密度循环水养殖模式对斑点叉尾鮰生长性能没有显著影响,并且在该养殖模式下肌肉具有更高的持水力、质构特性,同时呈现高蛋白、低脂肪的营养特点。  相似文献

The efficacy of copper sulphate (CuSO4) against the early stages of an experimental acute infection of Flavobacterium columnare in channel catfish (Ictalurus punctatus) was evaluated. Fish were challenged, by waterborne exposure to F. columnare in an ultra‐low flow‐through water delivery system, and treated with CuSO4 at 4.2 and 2.1 mg L?1 at 5.5 h post challenge for 4 h. Bacterial challenged non‐treated fish acted as a positive control and non‐challenged non‐treated fish acted as a negative control. Fish challenged with F. columnare and treated with CuSO4 at 4.2 and 2.1 mg L?1 post challenge had mortalities of 6.4% and 18.3%, respectively, compared with the positive control, which had 90.2% mortality. The mortality of challenged fish treated with CuSO4 at 4.2 and 2.1 mg L?1 was significantly different from the positive control. There was no significant difference between the mortalities of the 4.2 and 2.1 mg L?1 treatments. The results suggest that CuSO4 has clear therapeutic value against columnaris infections.  相似文献

The pullet carpet shell Venerupis corrugata is an economically valuable species in several European countries, however, nowadays stocks are under high fishing pressure. Hatchery production of juveniles for release is a major contributor to strengthen the stock and consequently improve the sustainability of the natural stocks. This study aimed to determine the feasibility of rearing V. corrugata larvae with different larval densities (10, 40 and 200 larvae per mL) in a recirculating aquaculture system (RAS), compared with the traditional larval rearing methodology (Batch). The mean survival, growth and metamorphic rate of V. corrugata larvae in RAS was higher (11.1%; 71.3 μm; 21.6% respectively) than in the Batch system, in all tested densities. The larval growth was not affected by the initial density until 40 larvae per mL, however, 200 larvae per mL decreased the larval growth in length nearly 54 μm. The larval rearing time was shortened in 2 days in the RAS system. The physical, chemical and microbiologic parameters suggested that the tested densities were not excessive to disturb the biofilter stability of RAS. The V. corrugata larval rearing performed at high larval stocking densities in RAS system present a reduction in the operating costs to produce this species.  相似文献

为探究温度对拟穴青蟹(Scylla paramamosain)循环养殖水体中微生物群落的影响,深入理解环境因子与养殖水体微生物群落之间的相关性。在水温分别为18℃、22℃、26℃、30℃和32℃的循环养殖系统中进行拟穴青蟹养殖,采用16S rRNA高通量测序技术对不同温度下的养殖水体微生物群落组成进行分析。结果表明:(1) Alpha多样性指数显示5组水样中细菌群落多样性指数大小为t26t22t30t32t18,物种丰度大小为t22t26t32t30t18。温度可以导致养殖水体中微生物群落多样性发生变化,但并未发现二者之间直接的相关性。(2) 5种温度下的养殖系统中细菌群落共有31门、43纲、94目、185科和351属,变形菌门(Proteobacteria)为循环养殖水体中绝对优势菌门。在养殖系统水温为26℃时,养殖水体中氮磷污染物含量相对较高,其水中脱氮除磷菌属的相对丰度也显著高于其他4组。(3)由花瓣图和UPGMA聚类分析图进行相似性分析可知,水体微生物群落对温度的变化有着良好的响应, 5组水样的细菌群落结构总体相似性较低,其中最高温度32℃和最低温度18℃的水体中细菌群落结构最为相似。(4)水体中微生物群落结构和与环境因子的相关性分析显示,氮磷营养盐中的总磷(TP)和硝酸盐(NO3–-N)对水中微生物群落特征具有显著性影响,而温度是导致养殖水体中氮磷营养盐差异化的主要原因。上述研究结果初步解释了温度对拟穴青蟹循环养殖水微生物群落的作用机制,为下一步深入研究拟穴青蟹循环水养殖系统中环境变化与微生物的相互关系提供参考。  相似文献

为了评估全封闭循环水养殖系统中养殖密度对钝吻黄盖鲽生长的影响及水质变化情况,将体质量为(250.00±50.83)g的钝吻黄盖鲽分成8个试验组(放养密度分别为18、22、26、30、34、38、42、46 kg/m3),进行了3个月的饲养试验,检测不同养殖密度下鱼的成活率、体质量增长率及饲料系数,同时对试验期间氨氮、亚硝酸盐和溶解氧等各项水质指标的动态变化进行监测。试验结果显示,各试验组鱼的成活率均达到96%以上,但随着养殖密度的增加,钝吻黄盖鲽的成活率总体呈现降低的趋势;低密度组(18 kg/m3)的体质量增长率最高,为36.1%,高密度组(46 kg/m3)的体质量增长率最低,为24.8%,且体质量增长率随着养殖密度的增加而逐渐降低;随着养殖密度的增加,饲料系数呈逐渐升高的趋势;养殖期间各项水质指标均保持在适宜钝吻黄盖鲽生长的范围内。结果表明,在本试验的循环水养殖系统中,综合考量养殖生长指标及单位面积产量,钝吻黄盖鲽规模化生产的最适养殖密度为42~46 kg/m3。  相似文献

Aquaflor® [50% w w?1 florfenicol (FFC)], is approved for use in freshwater‐reared warmwater finfish which include tilapia Oreochromis spp. in the United States to control mortality from Streptococcus iniae. The depletion of florfenicol amine (FFA), the marker residue of FFC, was evaluated after feeding FFC‐medicated feed to deliver a nominal 20 mg FFC kg?1 BW d?1 dose (1.33× the label use of 15 mg FFC kg?1 BW d?1) to Nile tilapia O. niloticus and hybrid tilapia O. niloticus × O. aureus held in a recirculating aquaculture system (RAS) at production‐scale holding densities. Florfenicol amine concentrations were determined in fillets taken from 10 fish before dosing and from 20 fish at nine time points after dosing (from 1 to 240 h post‐dosing). Water samples were assayed for FFC before, during and after the dosing period. Parameters monitored included daily feed consumption and biofilter function (levels of ammonia, nitrite and nitrate). Mean fillet FFA concentration decreased from 13.77 μg g?1 at 1‐h post dosing to 0.39 μg g?1 at 240‐h post dosing. Water FFC concentration decreased from a maximum of 1400 ng mL?1 at 1 day post‐dosing to 847 ng mL?1 at 240 h post‐dosing. There were no adverse effects noted on fish, feed consumption or biofilter function associated with FFC‐medicated feed administration to tilapia.  相似文献

This study investigated the microbial community, water quality, bacterial densities and growth performance in the industrial aquaculture system of sea cucumber Apostichopus japonicus without water exchange. Six treatments were set including, C (control treatment without sea cucumber culturing), S (small individual treatment), B (big individual treatment), Sd (small individual treatment with high density), Ss (small individual treatment adding carbohydrate source) and Sb (small individual treatment adding effective microorganisms). A total of 27916–32236 optimized reads and 564–742 operational taxonomic units (OTUs) were obtained from each samples. The phylum Proteobacteria, Actinobacteria, Bacteroidetes and Cyanobacteria predominated, representing 69.01–97.21% of the bacterial communities in the water samples. Cluster analysis and principal component analysis (PCA) showed that higher similarity was observed among S, Sd, Ss and Sb. The densities of TB and Vibrio in Ss were significantly higher than those in the other culture treatments after the 7th day. The concentrations of NH4‐N, NO2‐N, NO3‐N and PO4‐P in Ss and Sb were relatively lower than the other treatments. Conclusively, no deterioration was found in the water environment parameters during the 21‐day culture period without water exchange, indicating that low‐level water exchange protocol may be applied to the industrial aquaculture system. Based on the effects of different operations on culture system, industrial aquaculture is proved to be a viable way to rear sea cucumber.  相似文献

The effects of system partition on the water quality were studied using three shrimp ponds (Ts), and each one of the shrimp pond was connected individually with a fish pond by piping and pump. Three other isolated shrimp ponds were designated as the Cs treatment. The results showed that the implementation of the system can improve the water quality in the shrimp ponds. Particulate organic matter (POM), chemical oxygen demand (COD), nitrite nitrogen (NO2–N) and nitrate nitrogen (NO3–N) in the Ts were lower than those in the Cs during the experiment, and in some cases, they were significantly lower (p < 0.05). Throughout most of the experimental period, the soluble reactive phosphorus in the Ts treatment were greater than that in the Cs treatment (p < 0.05), and N/P ratio of the water column was lower in the Ts treatment than in the Cs treatment throughout most of the experimental period (p < 0.05). These results indicate that the partitioned system contributed to the stability of water quality and N/P ratio in the water. Vibrio parahaemolyticus was dominant in the third to seventh weeks in Cs, and the dominant condition was found only in seventh week in Ts. The result indicated that system may contribute to the slowing down of the breakout of V. parahaemolyticus in the shrimp pond. According to the redundancy analysis, COD, chlorophyll a and POM were mainly positively related to the dominance index of V. parahaemolyticus, and maybe POM can serve as an indicator to reflect the trend of V. parahaemolyticus.  相似文献
