In the last 9 years, epizootics of an icterus condition has affected coho salmon, Oncorhynchus kisutch (Walbaum), reared in seawater cages in southern regions of Chile. At necropsy, fish from field cases exhibited signs of jaundice accompanied by pale light-brown livers and dark spleens. Histopathological and haematological results indicated that these fish presented haemolytic anaemia. After microbiological examination no bacterial or viral agents could be identified as aetiological agents of this disease. In an infectivity trial, coho salmon, Atlantic salmon, Salmo salar L., and rainbow trout, Oncorhynchus mykiss (Walbaum), were inoculated intraperitoneally with a filtrate of an organ homogenate (0.45 microm) from a diseased coho salmon and held for 60 days in tanks supplied with fresh water. The disease was only reproduced in coho salmon in which mortalities, beginning at day 23 post-inoculation (p.i.), reached a cumulative value of 24% at day 27 p.i. This condition was transmitted to non-inoculated cohabiting coho salmon suggesting that it is a waterborne disease. Thus, this icteric condition is caused by an infectious form of haemolytic anaemia, probably of viral aetiology, and coho salmon are more susceptible than either Atlantic salmon or rainbow trout.  相似文献

Abstract. The ability of selected strains of Flavobacterium branchiophilum to attach to and colonize the gills of five species of teleosts (four salmonid and one cyprinid) and cause mortality was investigated. In virulence studies with F. branchiophilum strain LAB4a, cumulative mortality was dose-dependent in rainbow trout, Oncorhynchus mykiss (Walbaum), and ranged from 0 to 75%. However, regardless of dose, the relative amount of gill-associated F. branchiophilum antigen increased 1 h after challenge. The gill-associated F. branchiophilum antigen in fish which survived (moribund fish) increased by four to six times compared to that detected on the gills 1 h after challenge. The gill-associated antigen on moribund fish was not significantly different from that on the gills of dead fish. Flavobacterium branchiophilum strain LAB4a also attached to the gills of brook trout, Salvelinus fontinalis (Mitchell), rainbow trout, chinook salmon, Oncorhynchus tshawytscha (Walbaum), Arctic charr, Salvelinus alpinus (L.), and common shiners, Luxilus cornutus (Mitchell), following a 1-h bath exposure, and caused mortality in all species. The virulence of eight strains of F. branchiophilum for rainbow trout was examined. Both virulent and avirulent strains adhered to the gills following bath exposure (fimbriae were observed on all strains), but only virulent strains had the capacity to further colonize the gills and cause mortality.  相似文献

Abstract. A rickettsia-like organism (RLO) was isolated from infected coho salmon, Oncorhynchus kisutch (Walbaum), in cultures from six fish cell lines, but could not be cultured on 41 artificial media. The organism was confirmed by Koch's postulates as the aetiological agent of a systemic disease causing signifieant mortality among coho in Chile. The organism was a Gram-negative, obligate intracellular pathogen frequently observed within intracyto-plasmic vacuoles or inclusions of host cells, and appears to belong to the order Rickettsiales, family Rickettsiaceae and possibly the tribe Ehrlichiae. The organism, as well as the clinical signs and pathology it produces, is described. The disease, originally observed only in coho raised in seawater net pens, was reproduced experimentally in fish in both freshwater and seawater aquaria. Horizontal transmission without parasite vectors was observed in fish in both aquaria. The organism was shown, for the first time, to cause disease and mortality in Atlantic, Salmo salar L., and Chinook, O. tshawytscha (Walbaum), salmon and rainbow trout, O. mykiss (Walbaum). Because of the systemic nature of the disease, it is proposed that it be called 'salmonid rickettsial septicaemia'.  相似文献

Ceratomyxa shasta is a myxozoan parasite of salmonid fish. In natural communities, distinct genotypes of the parasite are associated with different salmonid hosts. To test the hypothesis that genotypes of C. shasta cause differential mortality, the polychaete host was experimentally infected with different parasite genotypes. Genotype I was obtained from Chinook salmon, Oncorhynchus tshawytscha, and genotype II from either coho salmon, O. kisutch, or rainbow trout, O. mykiss, We then challenged four salmonid strains: Chinook and coho salmon that occur in sympatry with the parasite and allopatric Chinook salmon and rainbow trout. Parasite genotype I caused mortality only in Chinook strains, although mortality in the allopatric strain also occurred from exposure to genotype II. A second experiment demonstrated that genotype II could be separated into two biotypes based on differential mortality in rainbow trout and coho salmon. These differential patterns of mortality as a result of infection by certain genotypes of C. shasta support field observations and suggest a co‐evolutionary relationship between these parasites and their hosts.  相似文献

Abstract. A microsporidian parasite, Microsporidium rhabdophilia n. sp. is described from rodlet cells of salmonid fishes. Chinook salmon, Oncorhynchus tshawytscha , coho salmon, Oncorhynchus kisutch , steelhead rainbow trout, Salmo gairdnerii gairdnerii , and various strains of domesticated rainbow trout were found infected. In living wet mount phase-contrast observations of various tissues, 16 mature spores could be seen infecting the nucleus of rodlet cells. The number of spores infecting each nucleus generally numbered 16 and spores were not found in any cell or organelle other than the nucleus of the rodlet cell. All observations were limited to California hatchery-reared fish.  相似文献

The present paper describes, for the first time, clinical signs and pathological findings of pancreas disease (PD) in farmed Atlantic salmon, Salmo salar L., and rainbow trout, Oncorhynchus mykiss (Walbaum), in sea water in Norway. Similarities and differences with reports of PD from Ireland and Scotland are discussed. Samples of 68 rainbow trout from disease outbreaks on 14 farms and from 155 Atlantic salmon from outbreaks on 20 farms collected from 1996 to 2004 were included in the present study. The histopathological findings of PD in Atlantic salmon and rainbow trout in sea water were similar. Acute PD, characterized by acute necrosis of exocrine pancreatic tissues, was detected in nine Atlantic salmon and three rainbow trout. Salmonid alphavirus (SAV) was identified in acute pancreatic necroses by immunohistochemistry. Most fish showed severe loss of exocrine pancreatic tissue combined with chronic myositis. Myocarditis was often but not consistently found. Kidneys from 40% and 64% of the rainbow trout and Atlantic salmon, respectively, had cells along the sinusoids that were packed with cytoplasmic eosinophilic granules. These cells resembled hypertrophied endothelial cells or elongated mast cell analogues. Histochemical staining properties and electron microscopy of these cells are presented. SAV was identified by RT-PCR and neutralizing antibodies against SAV were detected in blood samples.  相似文献

The first alphavirus to be isolated from fish was recorded in 1995 with the isolation of salmon pancreas disease virus from Atlantic salmon, Salmo salar L., in Ireland. Subsequently, the closely related sleeping disease virus was isolated from rainbow trout, Oncorhynchus mykiss (Walbaum), in France. More recently Norwegian salmonid alphavirus (SAV) has been isolated from marine phase production of Atlantic salmon and rainbow trout in Norway. These three viruses are closely related and are now considered to represent three subtypes of SAV, a new member of the genus Alphavirus within the family Togaviridae. SAVs are recognized as serious pathogens of farmed Atlantic salmon and rainbow trout in Europe. This paper aims to draw together both historical and current knowledge of the diseases caused by SAVs, the viruses, their diagnosis and control, and to discuss the differential diagnosis of similar pathologies seen in cardiomyopathy syndrome and heart and skeletal muscle inflammation of Atlantic salmon.  相似文献

Unique-staining Renibacterium salmoninarum (Rs) cells, termed bar forms, first observed in a coho salmon, Oncorhynchus kisutch (Walbaum), in 1983, could not be cultured, making their characterization difficult and significance obscure. They can be detected only by the fluorescent-antibody technique (FAT) and their numbers estimated only by a quantitative FAT (QFAT). Data collected over a 10-year period showed that bar forms were observed only in vivo and appeared associated with a host response. Bar forms were observed in 10 salmonid species from five countries and in fish from < 1 g to spawning adults. They were observed in 50.1% of kidney smears prepared from 10,061 Rs positive chinook, O. tshawytscha (Walbaum), coho, and Atlantic salmon, Salmo salar L. Bar forms were shown to be Rs cells based on absorption studies, their reaction with an Rs-specific FAT and enzyme-linked immunosorbent assay, and a transition from 'typical' Rs cells to bar forms in naturally and experimentally infected fish. Bar forms were determined to be non-virulent, damaged or dead Rs cells, based on fluorescence and electron microscopy observations, the inability to culture them, and mortality data. Bar forms appeared to represent visual markers of recovery from an Rs infection.  相似文献

The microhabitat selection of the ectoparasite Gyrodactylus salaris (Lærdalselva strain, Norway) was investigated concurrently with studies on the parasite population growth on five strains of Atlantic salmon, Salmo salar L., and a strain of Danish rainbow trout, Oncorhynchus mykiss (Walbaum). The salmon used were hatchery‐reared parr of East Atlantic strains [River Conon (Scotland), River Storå (western Denmark) and River Ätran (western Sweden)] and Baltic strains [Lule and Ume (eastern Sweden)]. The location and numbers of parasites were recorded on anaesthetized fish once a week from week 0 to week 8. The mean abundance of G. salaris steadily increased to high levels on the River Conon, Storå and Ätran strains until the end of the experiment. The mean abundance of G. salaris on the two Baltic strains (River Lule älv and River Ume älv) initially increased but after 4–7 weeks the growth of the parasite infrapopulations decreased markedly. The Danish rainbow trout strain showed the lowest abundances of all the fish species and strains. Gyrodactylus salaris preferentially selected the fins and head region when colonising the hosts (all species and strains). Increasing percentages of G. salaris on the tail fins of the East Atlantic strains and rainbow trout were found during the course of infection, whereas the two Baltic salmon strains experienced a decreasing percentage of parasites in this microhabitat.  相似文献

Plasma levels of insulin in rainbow trout,Oncorhynchus mykiss, Atlantic salmon,Salmo salar, and Pacific coho salmon,Oncorhynchus kisutch and plasma circulating levels of glucagon and glucagon-like peptide, in rainbow trout and Atlantic salmon, were measured by homologous radioimmunoassays. Hormonal levels were compared against the average body weight of the same group of fish. Plasma insulin levels were significantly correlated (r=0.56, 0.46 and 0.42 respectively) with body weight in all three salmonid species. Moreover, rainbow trout from fast-growing families had significantly higher (p<0.005) plasma insulin levels than did fish from slow-growing families. Abstract A total of 1239 kidney samples from four species of salmonid fish, Atlantic salmon, Salmo salar L., brown trout, Salmo trutta L., Arctic charr, Salvelinus alpinus (L.), and rainbow trout, Oncorhynchus mykiss (Walbaum), were screened for Renibacterium salmoninarum using double-sandwich ELISA and bacterial isolation. For bacterial isolation, samples were homogenized, washed, plated onto S-KDM and incubated for 12 weeks. Samples for ELISA were kept frozen until tested. After thawing, 25% homogenates in PBS were heated at 100°C for 15 min in the presence (2.5% v/v) of HemoDe solvent (terpene and butylated hydroxyanisole) and then centrifuged. The supernatant was tested with polyclonal antibodies against whole bacterium in a double-sandwich ELISA. In seven out of 12 groups tested, all samples were negative in both tests. Positive ELISA results occurred in five groups. Renibacterium salmoninarum was isolated on SKDM from samples in four out of these five groups. The ELISA test gave significantly higher numbers of positive samples in three out of the four groups showing positive results in both tests.  相似文献

Abstract. The pathogenicity of Renibacterium salmoninarum to carp, Cyprinus carpio L., and rainbow trout, Salmo gairdneri Richardson, was investigated. All carp injected with 4·8 × 108 cells/fish, or 4·8 × 107 cells/fish survived for 38 days. R. salmoninarum was isolated from all moribund fish, but not from the kidney of surviving fish, although R. salmoninarum antigen was detected in several of these fish by the dot blot assay. On the other hand, mortality in rainbow trout was 95% in the fish injected with 4·8 × 108 cells/fish, and 15% in those which received 4·8 × 107 cells/fish. R. salmoninarum antigen was detected by the dot blot assay in all surviving rainbow trout. The number of R. salmoninarum cells was immediately decreased by carp or rainbow trout serum, and the serum bactericidal activity of carp was higher than that of rainbow trout. Carp blood leucocytes had higher phagocytic activity than those of rainbow trout.  相似文献

Rainbow trout, Oncorhynchus mykiss (Walbaum), sea trout, Salmo trutta L., and Atlantic salmon, Salmo salar L., were experimentally infected with Flavobacterium psychrophilum in order to evaluate any species differences in susceptibility to the bacterium. Furthermore, differences in pathological changes and distribution of the bacteria in internal organs were studied. The bacteria were injected intraperitoneally in two doses, high dose (Hd) 1 x 10(7) colony forming units (CFU) fish(-1) and low dose (Ld) 1 x 10(6) CFU fish(-1). The mortalities in the Ld groups varied between 0 and 7.5% and in the Hd groups between 55-70%. No significant differences in mortality between the species were recorded. Clinical signs and pathological findings were similar in the three species and in accordance with those of rainbow trout fry syndrome. Rainbow trout showed more pronounced lesions in the spleen compared with the other species. Necrosis of renal tubular epithelium and haematopoietic tissue was most prominent in rainbow trout and Atlantic salmon. Intracellular eosinophilic droplets in the kidney tubular epithelium were a prominent finding in rainbow trout and sea trout surviving the infection. The distribution of the bacteria in internal organs was similar in the three species, as studied with immunohistochemistry.  相似文献

The present study evaluates if abdominal dis-tension caused by a water-filled stomach in seawater farmed rainbow trout, Oncorhynchus mykiss (Walbaum), and Atlantic salmon, Salmo salar L., could be provoked experimentally by live chilling in sea water (0.5 °C). Fifty rainbow trout and 50 salmon were visually classified, either as normal or suffering from the condition. Prior to chilling, no rainbow trout or salmon suffered from a water-filled stomach. During chilling, 25% of the rainbow trout and 2.5% of the salmon developed water-filled stomach. Affected rainbow trout had significantly higher blood plasma osmolality and significantly lower body weight than normal trout. The frequency of trout with a water-filled stomach increased significantly with increasing chilling time and increasing plasma osmolality. The regression coefficients revealed that the proportion of affected rainbow trout in-creased by 0.82% h−1 of chilling and by 0.46% per unit increase in mosmol L−1. The present study revealed that abdominal distension in seawater farmed rainbow trout was far more severe than in Atlantic salmon and could be provoked experimentally by osmoregulatory stress. This is consistent with the observation that water-filled stomach appear only occasionally in seawater farmed Atlantic salmon.  相似文献

Flavobacterium psychrophilum is the aetiological agent of rainbow trout fry syndrome and bacterial cold water disease. This study examined the genetic diversity of F. psychrophilum isolates retrieved from multiple epizootics at rainbow trout, Oncorhynchus mykiss, rearing facilities and from spawning coho salmon, O. kisutch. A total of 139 isolates were confirmed as F. psychrophilum by PCR assay and were further typed using pulsed-field gel electrophoresis (PFGE). Multiple epizootics at three proximally located rainbow trout rearing facilities were numerically dominated by three PFGE profiles, which accounted for 76% of all trout isolates. In coho salmon, 19 PFGE profiles were differentiated by PFGE and four numerically dominant PFGE profiles represented 56% of all coho salmon isolates. PFGE analysis also indicated that the average similarity of macrorestriction patterns of F. psychrophilum isolates was greater in rainbow trout than in coho salmon (88% vs. 70%). Furthermore, it was not unusual to isolate multiple PFGE profiles from a single coho salmon sample whereas only two PFGE profiles were shared between two sample dates separated by 1 month. It is clear that the domestic rainbow trout aquaculture facilities studied here were primarily affected by a complex of genetically related strains whereas spawning coho salmon supported a much more genetically diverse collection of F. psychrophilum.  相似文献

Abstract. Severe mortalities of up to 60% have occurred in Australian and South African farmed rainbow trout, Oncorhynchus mykiss (Walbaum), infected with Gram-positive cocci. Infected fish develop septicaemia and characteristic exophthalmos. Strains collected from outbreaks of disease appear to have attributes of the genus Enterococcus although no Lancefield group D antigen could be detected. All strains grew at 10°C but not at 45°C, grew in 6·5% NaCI, were bile-aesculin positive and hydrolysed L-pyrrolidonyl-β-naphthylamide. Isolates from both countries have similar phenetic characteristics and whole cell protein profiles.  相似文献

Abstract The survival of small-sized (<50.8 cm) chinook salmon, Oncorhynchus tshawytscha (Walbaum), and coho salmon, Oncorhynchus kitsutch (Walbaum), caught by sport fishing was determined to assess the potential for increasing the size limit for these fish. Fishermen were recently catching smaller salmon than in the 1970s, but salmon growth rates had not changed. To be an effective management option, the survival rate of hooked and returned fish must be high. The overall survival rates were high: 76% for chinook salmon and 70% for coho salmon. There was no significant difference in survival of the coho salmon with size of hook used ( P = 0.31). Any mortality among fish was generally acute; fish hooked deep in the mouth or gills generally bled and died shortly after capture. Fish hooked in the gills had a significantly greater mortality ( P = 0.0002). The overall high survival rate for these species was the result of a small proportion of fish being hooked in the gills or deep in the mouth. Since the survival rate of the salmon was high, the size limit could be increased to allow smaller fish to grow to sizes preferred by sport fishermen.  相似文献

Amoebic gill disease (AGD) of maricultured salmonids, turbot, Scophthalmus maximus (L.), European seabass, Dicentrarchus labrax (L.), and sharpsnout seabream, Diplodus puntazzo (Cetti), caused by Neoparamoeba pemaquidensis has been reported from Australia (Tasmania), Ireland, France, Chile, North America (Washington State and California) and Spain. Of the salmonids, Atlantic salmon, Salmo salar L., appears to be the most susceptible with rainbow trout, Oncorhynchus mykiss (Walbaum), also suffering significant disease. Only minor outbreaks have been reported in coho, O. kisutch (Walbaum), and chinook salmon, O. tshawytscha (Walbaum). The disease now accounts for 10–20% of production costs of Atlantic salmon in Tasmania and has lead to temporary abandonment of culture of this species in parts of Spain. It is of lesser, but still significant, importance in other countries. Much is known about the pathology of AGD but the pathophysiology of the disease is poorly understood. There is evidence that non-specific immunity is involved in fish acquiring resistance to AGD, but no unequivocal evidence exists for protection as a result of specific immune responses. To date, for salmonids, the only effective treatment for AGD is a freshwater bath. Control procedures based on modification of management strategies have been minimal and virtually unresearched.  相似文献

Abstract – Although homing behaviour has been observed in juvenile Atlantic salmon, brown trout and resident cutthroat trout, this behaviour has not been well studied in juvenile Pacific salmon. We examined the site fidelity and homing behaviour of juvenile coho salmon ( Oncorhynchus kisutch ) by marking and relocating them within an off-channel habitat. Over 80% of displaced fish returned to the area from which they were originally collected. The proportion of fish that returned to the original location did not vary significantly among three sampling dates. However, we found that this proportion decreased over time in a brackish lagoon when we statistically analysed the data reported by Day (1966) . Our results suggest that juvenile coho salmon exhibit strong site fidelity and are able to return to their home ranges after displacement. These behaviours are likely to be important for the winter survival of juvenile coho salmon.  相似文献

Out of 19 Vibrio harveyi isolates obtained from a diversity of hosts and geographical locations, 14 were pathogenic to rainbow trout, Oncorhynchus mykiss (Walbaum), and Atlantic salmon, Salmo salar L., with mortalities of up to 100% following intraperitoneal injections of 106 cells fish?1. The extracellular products (ECPs) of only five pathogenic isolates were harmful to fish. Both pathogenic and non‐pathogenic cultures produced ECPs containing caseinase, gelatinase, phospholipase, lipase and haemolysins. Vibrio harveyi VIB 645, which was the most pathogenic isolate, produced ECPs with a maximal effect on salmonids from preparations obtained by using cellophane overlays on tryptone soya agar supplemented with 1% (w/v) sodium chloride with incubation at 28 °C for 24 h. This preparation contained the highest titre of haemolytic activity to Atlantic salmon (1:256) and rainbow trout (1:32) erythrocytes.  相似文献
