由湖南师范大学生命科学学院与中国科学院水生生物研究所协议合作,将草鱼生长激素基因转移到三倍体鲫鱼进行研究。为检测经转移生长激素基因后的三倍体新鱼的养殖特性和生长情况,我们于2000年在长沙市水产科技推广站进行了转基因三倍体湘云鲫与普通三倍体湘云鲫专池对照养殖试验。1试验材料与试验养殖条件1.1试验材料先将草鱼激素全鱼基因移入异源四倍体鲫(鲤),再将已整合了草鱼生长激素的四倍体雄鱼与日本白鲫雌鱼通过人工杂交而获得转基因三倍体湘云鲫夏花鱼种;对照鱼系同源未转基因的三倍体湘云鲫夏花鱼种。试验和对照夏花规格均为3… 相似文献

用人工选育的异源四倍体鲫鲤(♂)与白鲫(♀)杂交获得具有明显生长优势的三倍体湘云鲫。采用差速离心和核酸酶处理等方法,从三倍体、白鲫和异源四倍体鲫鲤肝组织中提取线粒体DNA,并用9种限制性内切酶进行单酶酶切分析。经琼脂糖凝胶电泳后计算出各酶切片段的大小,测得三倍体、白鲫和异源四倍体鲫鲤mtDNA的分子大小分别为16.24kb、16.60kb和16.20kb。根据各单倍型间的酶切片段共享度,估算出3个群体间的遗传距离,说明了mtDNA母系遗传的特性。 相似文献

为探究饲料维生素 E 含量对彭泽鲫(Carassius auratus var. Pengze)亲本生长、卵巢发育和营养成分、抗氧化指标及组织维生素 E 含量的影响, 选取初始体重为(256.30±4.70) g 的彭泽鲫亲本进行养殖试验, 投喂含有 0 mg/kg (实测值为 68.1 mg/kg)、400 mg/kg (实测值为 462.3 mg/kg)和 1600 mg/kg (实测值为 1655.8 mg/kg)维生素 E 的等氮等能饲料, 养殖周期 60 d。结果显示: 饲料中维生素 E 含量对彭泽鲫亲本的生长、存活率和增重率无显著影响(P>0.05), 400 mg/kg 组肥满度显著高于 0 mg/kg 组(P<0.05); 400 mg/kg 组性腺指数显著高于其余两组(P<0.05)。1600 mg/kg 组肝脏中维生素 E 含量显著高于其余两组(P<0.05), 400 mg/kg 和 1600 mg/kg 组卵巢中维生素 E 含量显著高于 0 mg/kg 组(P<0.05); 卵巢必需氨基酸总量显著增加(P<0.05), 400 mg/kg 组的缬氨酸、异亮氨酸、苏氨酸及赖氨酸显著高于 0 mg/kg 组(P<0.05), 1600 mg/kg 组的组氨酸、谷氨酸和总氨基酸含量显著高于其余两组(P<0.05); 脂肪酸含量 1600 mg/kg 组 C18∶3n6 和 ΣPUFA显著低于其余两组(P<0.05), C22∶6n3 (DHA)和Σn-3PUFA在400 mg/kg组达峰值, 显著高于0 mg/kg组(P<0.05)。 血清中, 400 mg/kg 组和 1600 mg/kg 组 CAT、ACP 和 VTG 显著高于 0 mg/kg 组(P<0.05), 添加维生素 E 的两组与 0 mg/kg 组相比 MDA 显著降低(P<0.05), T-AOC 和 LZM 在 400 mg/kg 组达峰值, 显著高于 0 mg/kg 组(P<0.05)。肝脏中, SOD 活性在 400 mg/kg 组达峰值, 显著高于 0 mg/kg 组(P<0.05), 且随着饲料维生素 E 含量增加, MDA 活性显著降低(P<0.05)。 综上所述, 在本研究中, 饲料中 400 mg/kg 的维生素 E 的添加量可显著提高彭泽鲫亲本的性腺指数和卵巢中维生素 E 含量, 有利于氨基酸和脂肪酸的沉积并增加血清和肝脏中免疫和抗氧化酶活力, 从而改善彭泽鲫亲本性腺发育。 相似文献

为了探寻不同倍性鱼生殖育性的表观遗传调控规律,实验基于不同倍性鲫鲤精巢的转录本信息库设计引物克隆了不同倍性鲫鲤的MeCP2基因。序列比对结果发现,在远缘杂交的过程中基因组发生了遗传重组和变异。基于序列公共部分设计引物,通过半定量PCR和实时定量PCR的方法分析了MeCP2基因在不同倍性鱼发育过程中的时空表达特征。结果还发现,MeCP2基因在所有组织中都有表达,但在脑组织中的表达量最高;纵向对比性腺的发育过程,MeCP2基因的表达量会随着性腺发育成熟而下降;横向对比不同倍性鱼的性腺发育,MeCP2基因在三倍体鱼卵巢中的表达量明显高于二倍体和四倍体鱼,但在精巢中的表达量随鱼的倍性增加而增加。研究表明,MeCP2基因的表达与不同倍性鱼的卵巢发育密切相关,这为研究鱼类生殖不育的分子机理及其在水产育种上的应用奠定了坚实的基础。 相似文献

This study determined biometric and weight parameters and relevant indices of diploid and triploid tench. Altogether, 137 siblings of tench were studied. The effect of ploidy level appeared in significantly better growth of triploids (P<0.001) as to biometric [total length (TL), standard length (SL), body height (BH), body width (BW)] and weight [fish weight (FW), carcass weight (CW)] parameters of T3 of both sexes and of T3+ females. The effect of ploidy level also appeared as significantly higher dressing percentage (DP; P<0.001) of triploid T3 females compared with other groups, significantly higher gonad weight (GW) and gonadosomatic index (GSI; P<0.001) of diploid T3 females, as well as GSI and hepatosomatic index of diploid T3+ females. The effect of sex appeared in significantly higher (P<0.001) biometric (TL, SL, BH, BW) and weight (FW, CW) parameters of T3 females of both ploidy levels, as well as of triploid T3+ females. The effect of sex also appeared as significantly higher DP (P<0.001) of males in diploid T3 fish, as well as of males of both ploidy levels in T3+ fish and significantly higher GW and GSI (P<0.001) of females in diploid T3 fish, as well as of females of both ploidy levels in T3+ fish. This study shows evidence for faster somatic growth and bigger final weight of triploid populations of tench compared with diploids in both age categories T3 and T3+. 相似文献

ABSTRACT: This study clarifies the annual reproductive cycle and the lunar-synchronized spawning of the spiny rabbitfish ( Siganus spinus ) that inhabit the Okinawan waters. Annual and weekly changes in the gonadosomatic index (GSI) and the histological features of the ovaries were checked. Gonadosomatic index was high during the months of May to July, and yolk-laden oocytes were observed in the ovaries from March to July. Some of the ovaries collected during June and July contained oocytes at maturation stage or ovulatory follicles. These results suggest that the spiny rabbitfish undergo active vitellogenesis and spawning from May to July. During the reproductive season (May to July), collection of fish according to the lunar phase revealed that a high GSI occurred around the time of the new moon. Cyclic oocyte development with peaks around the time of the same moon phase was also observed, suggesting that, in Okinawan waters, this species is a lunar-synchronized spawner and spawns three times. 相似文献

本文通过气相色谱分析法,对野生银鲫背部肌肉脂类脂肪酸的组成和人工养殖银鲫、白鲫相比较。野生银鲫和人工养殖银鲫、白鲫背部肌肉脂类中饱和脂肪酸的含量基本相同,野生银鲫饱和脂肪酸的含量为24.55~27.30%,人工养殖银鲫为23.35~23.97%,白鲫为23.73%;—烯酸的含量野生银鲫比人工养殖银鲫、白鲫低,野生银鲫—烯酸的含量为23.97~29.53%,人工养殖银鲫为41.81~42.52%,白鲫为46.53%;多烯酸的含量野生银鲫比人工养殖银鲫、白鲫高,野生银鲫多烯酸的含量为41.53~46.77%,人工养殖银鲫为28.91~32.58%,白鲫为27.46%;其中ω_3族脂肪酸的含量野生银鲫亦比人工养殖银鲫、白鲫高,野生银鲫ω_3族脂肪酸的含量为25.74~27.99%,人工养殖银鲫为11.86~12.99%,白鲫为6.43%,而且ω_3/ω_6族脂肪酸的比率,野生银鲫也同样大于人工养殖银鲫、白鲫;野生银鲫ω_2/ω_6族脂肪酸的比率为1.37~1.74.人工养殖银鲫为0.60~0.87,白鲫为0.31. 相似文献

Performance tests of the Pr?erov Scaly Carp (PS), two crossbreds (PS × M72 and PS × ROP) and an internal control group (M2 × M72) were designed to assess their production and dressing out parameters at harvest size for their potential commercial utilization. The fastest growth rate (standard length (SL): 405±21.91 mm, P<0.01) and the highest final live weight (FLW: 2015.70±323.92 g, P<0.01) were recorded in the PS × ROP hybrid. The SL values in this group of carp were 12.5%, 19.12% and 11.57% higher (P<0.01) than those at M2 × M72, PS and PS × M72, respectively, and the FLW values were 21.57%, 62.83% and 24.37% higher (P<0.01) compared with M2 × M72, PS and PS × M72 hybrids respectively. Growth rate parameters of PS × M72 hybrid and control group M2 × M72 were practically identical. The lowest growth rate potential and FLW were recorded in the PS group (P<0.01). The highest (P<0.01) dressing out parameters were found in the control group of mirror carp M2 × M72 with a dressing percentage of 65.64±2.11%. There were practically no differences in this parameter between scaly carp groups (PS: 61.79±2.82%, PS × M72: 61.51±1.72%, PS × ROP: 63.60±1.96%). This study shows evidence for faster somatic growth and bigger final weight in the hybrid line PS × ROP of 3‐year‐old common carp compared with the pure line PS and the crossbreed PS × M72. 相似文献
