In this study, we investigated the potential utility of fish skin, usually discarded during processing, as a cost‐effective protein source. We evaluated the effects on juvenile black rockfish (Sebastes schlegeli) of an 8‐week feeding trial in which 0%, 5%, 10% or 20% of the casein (purified protein) in their diets was replaced with flounder skin meal. The fish that received flounder skin meal, particularly the 20% group, showed significantly higher growth performance and feeding efficiency. They had lower levels of body lipids, elevated levels of high‐density lipoprotein cholesterol, and increased body and muscle protein contents. They recovered more rapidly from stress induced by anaesthesia or exposure to air. Supplementation with flounder skin meal confirms the functionality of flounder skin meal as a high collagen component of fish diets.  相似文献

Cytochrome P450c17 (CYP17, 17α‐hydroxylase/17,20‐lyase) is a critical enzyme in the production of androgens and estrogens in vertebrates. A 2102 bp full‐length cDNA of P450c17‐II (CYP17A2) has been isolated from the ovary of half‐smooth tongue sole, Cynoglossus semilaevis which encodes 524 amino acids. The putative P450c17‐II enzyme shares higher sequence identity with those of teleosts than with P450c17‐I of vertebrate. The similarity between the two types of tongue sole P450c17 was 48%.Semi‐quantitative RT‐PCR analysis of spatial expression showed the enzyme was specifically expressed in the ovary and the head kidney. However, temporal expression shows that P450c17‐II can be found in the brain. Furthermore, temporal expression pattern of P450c17‐II in ovary and brain revealed developmental stage‐dependency, and ovary P450c17‐II expressed remarkably throughout the whole reproductive cycle. Otherwise, the expression pattern of P450c17‐II in head kidney indicated negative ovary development‐dependence. In addition, combined with our data on P450c17‐I, T and E2 levels, the results further endorse the critical role of P450c17‐II during shift in steroidogenesis, suggesting that P450c17‐I and ‐II may act together to this physiological process. Based on the present study, we indicate an important role for P450c17‐II during ovarian development.  相似文献

胡子鲇脑型芳香化酶基因全长cDNA克隆及表达   总被引:1,自引:0,他引:1  
采用RT-PCR和RACE法,克隆了胡子鲇(Clarias fuscus)脑型芳香化酶基因Cyp19a1b,应用荧光实时定量PCR检测其在前脑、下丘脑、脑垂体、肝、精巢和卵巢6种组织,以及性腺分化前后(出膜后12~30 d)全鱼中的mRNA表达。结果表明,Cyp19a1b cDNA全长2 347 bp,5′端非编码区219 bp,3′端[不包括poly(A)]596 bp,开放阅读框(ORF)1 503 bp,编码500个氨基酸,推测其编码蛋白质分子量为56.388 kD。序列分析及分子系统进化树结果表明,胡子鲇Cyp19a1b氨基酸序列与非洲鲇(Clarias gariepinus)Cyp19a1b同源性最高,达95.6%;与南方大口鲇(Silurusmeridionalis)、黄颡鱼(Pelteobagrus fulvidraco)、斑马鱼(Danio rerio)、斑点叉尾(Ictalurus punctatus)、赤点石斑鱼(Epinephelus akaara)、鲤(Cyprinus carpio)、鲫(Carassius auratus)及稀有鲫(Gobiocypris rarus)Cyp19a1b同源性达75%以上,但与上述鱼类的性腺型芳香化酶基因(Cyp19a1a)同源性低于62%,表明其为脑型芳香化酶,同源性分析结果与根据传统形态学和生化特征分类的结果相一致。荧光实时定量PCR结果显示,Cyp19a1b在胡子鲇上述组织中均有表达,其中下丘脑中表达量最高,肝和精巢最低,在脑部的表达存在明显的性别差异(P<0.05)。此外,在胡子鲇性腺分化前(出膜后12 d)Cyp19a1b即开始表达,但分化前后表达量无显著性差异(P>0.05)。结果暗示Cyp19a1b可能不是引起胡子鲇性腺分化的直接因素,但其可能通过"下丘脑垂体性腺轴"对性腺分化过程产生间接影响。  相似文献

The objectives of the present experiment were to determine the effects on growth factors and some haematological parameters in Korean rockfish, Sebastes schlegeil, after dietary 7,12‐dimethylbenz(a)anthracene (DMBA) exposure at 0.6, 1.2, 2.4 and 4.8 mg kg?1 diet for 8 weeks. The specific growth rate of the fish exposed to DMBA (≥1.2 mg kg?1) showed significantly lower performance than the control. Following 8 weeks of exposure, the DMBA‐exposed groups (2.4 and 4.8 mg kg?1) had a significantly higher mean hepatosomatic index. Condition factor of the fish exposed to DMBA did not show any statistically significant deviation from the control (P>0.05), although the mean values were somewhat lower than those of the control group. Observations on haematological parameters indicated DMBA treatment induced a severe anaemia. Total protein, albumin and total cholesterol concentrations decreased following exposure to DMBA at 2.4 and 4.8 mg kg?1. In contrast, serum bilirubin and blood urea nitrogen in fish exposed to DMBA increased. Significant decreases in serum electrolytes, chloride, phosphorus, magnesium and calcium, and osmolality were observed in all DMBA‐treatment groups. All DMBA‐treatment groups showed a significantly higher activity of lactate dehydrogenase and alkaline phosphatase. Serum transaminase activity after the highest level of 4.8 mg kg?1 DMBA was significantly increased. The key finding from this study is that rockfish exposed to dietary DMBA at concentrations of 1.2 and 2.4 mg kg?1 diet are likely to experience adverse impacts in growth and haematological property respectively.  相似文献

The present experiment was conducted to evaluate the effects of trace mineral (Cu, Zn, Mn and Fe) premixes from inorganic and chelated (chelated to 2‐hydroxy‐4‐methylthiobutanoic acid or hydroxy analog of methionine; Mintrex?, Novus International Inc., St. Charles, MO, USA) sources, in rockfish, (Sebastes schlegeli). fed diets containing mineral inhibitor phytic acid. Seven isonitrogenous (46% crude protein/CP) and isocaloric (16.63 KJ available energy per g diet) semi‐purified diets comprising of a Basal control and diets supplemented either with the inorganic (I) or chelated premix (M) at three different levels of 0.75 (I0.75/M0.75), 1.5 (I1.5/M1.5) and 3 (I3/M3) g kg?1 diets were formulated. Twenty‐four fish averaging individual wet body weight of 9 ± 0.2 g (Mean ± SD) were fed one of the experimental diets in quadratic group for 16 weeks. Average weight gain (WG) of fish fed M3 was significantly higher than that of fish fed Basal control and I1.5 diets (< 0.05). Whereas, data for the liver and whole body trace mineral contents showed a higher Cu and Zn saturation among fish fed M3 and M1.5 diets (< 0.05). Furthermore, hepatic Cu‐Zn super oxide dismutase (Cu‐Zn SOD) activity was recorded to be highest for fish fed M3 diet. Whereas, hepatic thiobarbituric acid‐reactive substance (TBARS) value was lowest in fish fed M3 diet (< 0.05). Challenge test with E. tarda showed an improved disease resistance among the fish fed different levels of trace mineral premixes. Therefore, these results demonstrated a higher efficiency of chelated compared to inorganic source of trace mineral premixes in marine rockfish.  相似文献

The effect of the substitution of fishmeal with tuna by‐product meal (TBM) in the diet of Korean rockfish on growth, body composition, plasma chemistry and amino acid profiles was determined. Nine experimental diets were prepared. The control (Con) diet consisted of 550 g kg?1 fishmeal. The 10, 20, 30, 40, 60, 80 and 100 % fishmeal diets were substituted with tuna by‐product meal (TBM), referred to as the TBM10, TBM20, TBM30, TBM40, TBM60, TBM80 and TBM100 diets, respectively. Finally, the fishmeal and soybean meal was completely replaced with TBM in the diet TBM100‐S. There was no significant difference in weight gain and SGR of the fish that were fed the TBM40 and Con diets. The feed conversion ratio (FCR) of the fish that were fed the Con, TBM10, TBM20 and TBM30 diets was lower than that of the fish that were fed all of the other diets. The protein efficiency ratio (PER) of fish that were fed the TBM10, TBM20 and TBM300 diets was higher than that of fish that were fed the TBM40, TBM60, TBM80, TBM100 and TBM100‐S diets. It can be concluded that the substitution of fishmeal with up to 40 and 30 % TBM in the diet of juvenile Korean rockfish could be made without exerting a detrimental effect on their growth (SGR) and feed utilization (FCR and PER), respectively.  相似文献

CYP19 is considered as an important factor affecting reproductive endocrinology in many fishes, and plays an important role in ovarian development, reproductive function and sexual differentiation. In this study, three single nucleotide polymorphisms (SNPs) within CDS of the CYP19a gene were tested and the associations between their genotypes and four reproductive traits were analyzed in 65 Japanese flounder individuals with Polymerase chain reaction and Single-stranded conformational polymorphism (PCR–SSCP). Results indicated that a SNP in the exon7 of CYP19a gene, SNP2, was significantly associated with 17β-estradiol (E2) (P < 0.05) and gonadosomatic index (GSI) (P < 0.05). Individuals with genotype AB of SNP2 had significantly higher serum E2 levels (P < 0.05) and GSI (P < 0.05) than those of genotype AA or BB. In addition, there was significant association between one diplotype based on three SNPs and reproductive trait. The genetic effects for both serum E2 of diplotype D9 and GSI of diplotype D1 were respectively much higher than those of other diplotypes (P < 0.05). The evidence of the associations between genetic variants with serum E2 and GSI may help explain effects of CYP19a gene in reproductive endocrinology of Japanese flounder.  相似文献

Vibrio rotiferianus is an important marine pathogen of various aquatic organisms and can be found widely distributed in the marine environment. To further characterize this pathogen, the pathogenic properties and genome of V. rotiferianus SSVR1601 isolated from Sebastes schlegelii with skin ulcer were analysed. SSVR1601 was shown to be short rod‐shaped cell with a single polar flagellum. Different degrees of pathological changes in fish kidney, intestine, gills and liver were observed after SSVR1601 challenge. The SSVR1601 genome consists of two chromosomes and two plasmids with a total of 5,717,113 bp, 42.04%–44.93% GC content, 5,269 predicted CDSs, 134 tRNAs and 40 rRNAs. The common virulence factors including OMPs, haemolysin, flagellin, DNase, entF, algU, tcpI, acfB and rfaD were found in strain SSVR1601. Furthermore, factors responsible for iron uptake (fur, fepC and ccmC) and types II, IV and VI secretion systems were detected, which are likely responsible for the pathogenicity of SSVR1601. The antimicrobial resistance genes, bacA, tet34 and norM, were detected based on Antibiotic Resistance Genes Database. The phylogenetic analysis revealed SSVR1601 to be most closely related to V. rotiferianus strains CAIM577 and B64D1.  相似文献

β‐catenin gene is a pivotal gene for gonad development and maintenance of ovarian function in mammals. However, little is known about its expression and function in gonad development of fish. In this study, a complete cDNA (3342 bp) sequence of β‐catenin 1 was cloned from the common carp, Cyprinus carpio, by RACE PCR, which encodes a 780‐amino‐acid protein. Quantitative real‐time PCR demonstrated that β‐catenin 1 mRNA expressions were high in the testis and ovary tissue and the expression increased as the testes developed and the early stage ovaries developed. Western blot results revealed a single immunoreactive band with an estimated molecular weight of 90 kDa in testes. Immunohistochemistry analysis revealed that the β‐catenin 1 protein was concentrated mainly in the cytoplasm of early development stage of oocyte cells and in the cytomembrane of developing and mature sperm cells. 17β‐Ethinylestradiol injecting intraperitoneally into the fish decreased the relative β‐catenin 1 mRNA expression level except 1 μg/g 72 hr and 5 μg/g 48 hr of treatments in the ovary by real‐time PCR. These results suggest, for the first time, that β‐catenin 1 is an essential protein in gonad development and might be involved in ovarian early development of C. carpio.  相似文献

Little is known about the population structure of Alaskan rockfishes, including Pacific ocean perch (POP, Sebastes alutus), and how persistent and variable oceanographic features may influence their structures. Moreover, early life history information is sparse for many species. We used data from 14 microsatellite loci to characterize the genetic structure of young‐of‐the‐year Pacific ocean perch collected during 1998–2003 from the Gulf of Alaska and Bering Sea. Broad‐scale geographic variation in genetic structure of the young‐of‐the‐year (FST = 0.005, P < 10?4) had similarities to that observed in a previous adult study. The overall correlation between genetic and geographic distance (isolation by distance) was nearly identical to that observed in the adults. Fine‐scale geographic divergence was also observed and may be the result of oceanographic circulation features within the Gulf of Alaska. Interannual variation (between cohorts) at locations sampled in more than one year is consistent with variable oceanography and fine‐scale population structure rather than the influence of a sweepstakes effect. The similarities of the young‐of‐the‐year with the adults and the pattern of genetic divergence confirm that dispersal of Pacific ocean perch is limited in all life stages.  相似文献

根据NCBI数据库报道的黄鳝芳香化酶基因的gDNA序列,设计了一对特异性引物.用Trizol试剂盒提取黄鳝脑总RNA,反转录获得cDNA第一条链,进行PCR扩增.扩增产物经过回收、连接、测序后得到了一条长1514 bp的芳香化酶基因cDNA序列.同源性比较表明该基因属于脑型P450aromB,与其他鱼类脑型P450aromB的同源性较高(>70%),与性腺型P450aromA的同源性较低(<60%),与自身性腺型芳香化酶同源性为59.5%.采用RT-PCR的方法对该基因在雄性黄鳝的各组织表达情况进行了分析,结果表明,该基因的转录本较高的表达于脑和精巢中,较低的在皮肤中表达,而在肝脏、心脏、小肠、肌肉等组织中没有检测到该基因的表达.  相似文献

表达差异及其在早期发育阶段的表达水平变化。 研究结果表明,  半滑舌鳎Dorsalin-1-like基因cDNA全长为1 961 bp,
包括了 153 bp 的 5′非翻译区(5′UTR)、 编码 419 个氨基酸的 1 260 bp 的开放读码框(ORF),  以及 548 bp  的 3′非翻译
区(3′UTR)。 同源性分析和分子进化聚类分析结果显示,  半滑舌鳎 Dorsalin-1-like 鲀 与红鳍东方 (Taleifugu rubripes)、
尼罗罗非鱼(Oreochromis niloticus)等的同源性最高,  并与之共同聚为鱼类 Dorsalin-1 分支。此外, Dorsalin-1-like
mRNA 在半滑舌鳎成鱼的 13 个组织中具有广泛的分布,  并在心脏组织中表达量最高,  在肝、脊髓等组织中次之。
在半滑舌鳎早期发育阶段(卵期、仔鱼、稚鱼、幼鱼)中, Dorsalin-1-like 在卵期中的胚胎期表达量最高,  其次为前期
仔鱼期。上述实验结果表明, Dorsalin-1 在调控卵期和仔鱼期的早期发育中具有重要作用,  并可能通过调节神经系
统的发育进而参与心脏、骨骼等组织的发育调控。本研究旨在揭示 Dorsalin-1-like 基因在调控半滑舌鳎早期神经
系统发育的重要作用,  并为筛选半滑舌鳎骨骼发育的调控因子奠定理论基础。

Retinoblastoma (Rb) is a multifunctional regulator involved in several key cellular processes, such as cell cycle control, cell differentiation, tumorigenesis and senescence. In this study, an Rb‐like gene, PmRBL, was cloned from black tiger shrimp (Penaeus monodon). The full‐length cDNA sequence of PmRBL is 4,069 bp with an open reading frame of 3,243 bp, which encodes 1,080 amino acids. Quantitative real‐time PCR (qRT‐PCR) analysis indicated that PmRBL was highly expressed in the gills, hepatopancreas and ovaries of P. monodon. The highest PmRBL expression levels were observed in stage III of the ovarian development in P. monodon. RNA interference experiments were conducted to examine the expression profiles of PmRBL, PmCDK2 and PmCyclin E. The knockdown of PmRBL in the ovary and hepatopancreas by dsRNA‐RBL was successful. After dsRNA‐RBL was injected into the shrimp, the relative expression levels of PmCDK2 and PmCyclin E were upregulated at 12–72 hr in the ovaries and hepatopancreas. The localization and level of PmRBL expression in the ovary and hepatopancreas were investigated through in situ hybridization, which revealed consistent results with those of qRT‐PCR. Therefore, PmRBL, PmCDK2 and PmCyclin E may be involved in vitellogenin synthesis and ovarian maturation in P. monodon.  相似文献

Insulin‐like growth factor‐1 (IGF‐1), somatolactin and leptin are involved in growth regulation and energy metabolism in fish. We herein focused on serum IGF‐1 concentration analysed by enzyme‐linked immunosorbent assay in restrictively fed rainbow trout (Oncorhynchus mykiss). The animals were fed a high‐fat/low‐protein diet at daily feed increases (DFI) ranging from 0.5% to 2% of initial body weight (IBW), starting either at 62 or 176 g IBW. In selected groups, growth hormone receptor 1 (GHR1) and leptin mRNA were quantified in liver, and GHR1 mRNA also in visceral adipose tissue. Serum IGF‐1 concentrations in both IBW groups were highest at 2% and 1% DFI and were nonlinearly decreasing with reduced DFI. The low‐IBW groups had mostly lower IGF‐1 concentrations than the high‐IBW groups. Leptin and GHR1 mRNA decreased with feeding intensity in liver, but GHR1 mRNA increased in adipose tissue. IGF‐1 is related to growth and may help to mitigate oxidative stress in consequence of lipid mobilization during restrictive feeding. IGF‐1 secretion associated with stress response in addition to its function in growth and energy metabolism seemed to reach a point of inflection at DFI 1%. Leptin and GHR1 might be linked to lipid metabolism and free fatty acid partitioning towards liver.  相似文献

Cytochrome P450 aromatase, which is encoded by the CYP19a gene, converts androgens to estradiol. Considerable evidence suggests that estrogens play an important role in fish reproductive process. Therefore CYP19a is an excellent candidate gene for reproductive traits. Variants in the promoter of the CYP19a gene might also be involved in the control of aromatase expression and affect regulatory mechanism linking cholesterol metabolism to the synthesis of sex steroids. In this study, nine single-nucleotide polymorphisms (SNPs) were detected with polymerase chain reaction-single stranded conformational polymorphism (PCR-SSCP), namely A-680G, G-672A, AGTAGT-649 inserting or deleting, T-623C, C-410A, T7-454A, T-402C, TTTCCAGACTGA-345 inserting or deleting, and G-297C. Nine SNPs within the promoter of the CYP19a gene were tested for association with four reproductive traits [serum testosterone (T), serum 17β-estradiol (E2), hepatosomatic index (HSI), and gonadosomatic index (GSI)] in a population of 50 female Japanese flounder individuals. A locus, P3 (TTTCCAGACTGA-345 inserting or deleting, G-297C), was significantly associated with 17β-estradiol (E2) level (P < 0.05) in female Japanese flounder. In addition, there was significant association between one diplotype based on nine SNPs and reproductive trait. The genetic effect for E2 level of diplotype D3 was significantly higher than those of other diplotypes (P < 0.05). Results indicate that these genetic effects of those variants on E2 level may help to explain CYP19a gene status in the reproductive endocrinology of Japanese flounder.  相似文献

根据5种硬骨鱼已知CYP3A序列保守区设计兼并引物,以异育银鲫cDNA为模板扩增得到CYP3A基因片段,根据得到片段序列设计特异引物并利用cDNA末端快速扩增技术(RACE)获得全长cDNA。得到异育银鲫CYP3A基因全长为1 769 bp,开放阅读框为1 545个核苷酸,编码514个氨基酸。其预测蛋白质分子量为58.624 ku,理论等电点为6.30。将异育银鲫CYP3A序列理论编码氨基酸序列提交细胞色素P450命名委员会(Cytochrome P450 Nomenclature Committee)并由其命名为CYP3A136。氨基酸序列分析显示,其与稀有鮈鲫、草鱼、鲦同源性较高,并且具有高度保守的血红素结合区域FXXGXXXCXG。异育银鲫CYP3A基因的半定量RT-PCR显示,在肝和肠组织中转录水平最高,在肾和鳃中次之,其余组织较低。  相似文献

蜕皮是甲壳动物重要的生理活动,与其蜕皮激素的合成密切相关,细胞色素P450(CYP)302a1是甲壳动物蜕皮激素合成通路中的关键酶之一。本研究克隆了罗氏沼虾CYP302a1基因(Mr-CYP302a1),cDNA全长1 859 bp,开放阅读框(ORF)为1 629 bp,编码543个氨基酸(aa),分子量大小为61.09 ku,等电点为8.42。氨基酸序列分析显示CYP302a1基因的保守结构域含有5个P450基因家族特征保守区域:heme-binding、helix-K、helixC、helix-I及PERF。系统进化分析结果显示Mr-CYP302a1首先与绿虾CYP302a1聚为一支,然后与凡纳滨对虾及三疣梭子蟹等十足目甲壳动物的CYP302a1聚为一支,与甲壳动物的亲缘关系最近。实时荧光定量PCR(qRT-PCR)检测表明Mr-CYP302a1在罗氏沼虾的多个组织中均有表达,其中在Y器官中的表达量最高,性腺中次之。同时研究发现,MrCYP302a1基因在罗氏沼虾的蜕皮后期(A期和B期)表达量很低,蜕皮间期(C期)表达量开始上升,在蜕皮前期D1亚期达到峰值。对Mr-CYP302a...  相似文献

Clostridium autoethanogenum protein (CAP) is a single‐cell protein derived from ethanol production and may have potential to become a substitute for fishmeal in aquafeeds. A 70‐day feeding trial was conducted with black sea bream (Acanthopagrus schlegelii) (mean initial weight 6.03 ± 0.09 g) to evaluate partial replacement of fishmeal with CAP in diets. Seven isonitrogenous and isoenergetic diets were formulated with graded levels of CAP (0, 4.85, 9.70, 14.55, 19.40, 38.80 and 58.20%) to replace fishmeal. The inclusion of CAP at all dietary levels tested did not significantly affect the growth performance (p > .05). Fish fed the CAP58.20% diet showed a significantly lower feeding rate, with significantly higher protein efficiency ratio and feed efficiency ratio compared with fish fed the other diets (p < .05). No statistical differences were found in dorsal muscle and whole‐body compositions. Total superoxide dismutase in serum of fish fed CAP58.20% diet was significantly lower compared with that of the control. Malondialdehyde, catalase, total antioxidant capacity and digestive enzyme activities revealed no significant differences among dietary treatments. Phosphorus retention efficiency significantly increased, and phosphorus discharge showed a downward trend with increasing CAP inclusion levels. In conclusion, the results indicated that CAP is a safe and effective alternative protein source, which can replace fishmeal in the diet of black sea bream up to 58.20%, without adverse effects on growth performance, antioxidation and digestive enzyme activity. This study has shown the potential of converting industrial waste into a high protein feed ingredient for aquafeeds.  相似文献
