An experiment was conducted to examine carotenoid depletion in the muscle of rainbow trout. After 4 weeks of ingestion of canthaxanthin, fish were divided into two groups: the first was fed the same diet but without the canthaxanthin, the second was starved. Lipid and canthaxanthin levels in epaxial muscle were followed for 38 days. The lipid content increased (× 1.84) in fed fish and remained stable in those which were starved. Fed fish lost more canthaxanthin than starved fish. The ratio of the relative weight gain of canthaxanthin to that of the lipids was higher in starved than in fed fish (0.91 and 0.69 respectively). It is concluded that an internal redistribution of canthaxanthin takes place in the absence of an intake of the pigment. These results also show that the muscle of rainbow trout has a high storage capacity.  相似文献

This study examined how muscle metabolic organization varied during an annual cycle in which rainbow trout (Oncorhynchus mykiss) were held in outdoor holding ponds in which they were exposed to natural changes in temperature (range 0.2 to 15.6°C) and photoperiod. We examined the activities of glycolytic and mitochondrial enzymes in red and white muscle to evaluate whether trout enhance their capacity for lipid and carbohydrate oxidation during cold-acclimization. When assayed at habitat temperature, the enzyme activities generally increased in spring to reach a maximum in summer followed by a decrease in the fall. This led to significantly higher activities at warm than cold periods for all enzymes measured in red muscle and all but one in white muscle. The activities at 10°C provided little evidence for compensatory adjustments of aerobic capacity. Particularly in red muscle, enzyme levels at 10°C were generally lower during cold than warm periods. The variation of enzyme activities throughout the cycle was not due to changes in protein concentration, as the same responses were observed when activities were expressed per g wet mass or per mg protein. Although the aerobic capacity did not increase with cold-acclimatization, the relative capacity for lipid oxidation was higher in winter than in summer trout. In contrast, the relative capacity for aerobic glycolysis was higher in summer than in winter trout. Thus, the metabolic capacities of trout muscle undergo seasonal reorganization.  相似文献

Northern Lake Huron (Ontario, Canada) supports the largest concentration of freshwater salmonid cage culture farms in Canada and receives inputs of domestic‐strain rainbow trout (Oncorhynchus mykiss) through escapement. We assessed the potential effects of these domestic fish on the naturalised rainbow trout of this ecosystem by comparing their respective ecologies. Mature adults were sampled from spawning tributaries, primarily in spring, and to a lesser extent in fall. Fish of domestic origin comprised ~80% of rainbow trout sampled from sites near cage culture facilities but <20% of rainbow trout sampled from more distant sites. Domestic adults in spawning condition (gametes free‐flowing) were present in the spawning tributaries in both spring and fall sampling seasons, whereas naturalised fish in spawning condition were only observed in the spring. Domestic adults were younger and smaller (in length), appeared to have shorter lifespan and had a higher male to female ratio compared with their naturalised counterparts. Growth rates (change in length with age) of naturalised and domestic females in the wild were similar, but domestic males grew more slowly than naturalised males. Domestic females also produced smaller eggs than naturalised females. Food web positions (inferred from δ13C and δ15N) of domestic and naturalised fish were very similar but varied more strongly with body size in the former. Domestic‐strain rainbow trout of cage culture origin can survive, grow and attempt to spawn in northern Lake Huron and have the potential to compete for food, mates and spawning habitat with naturalised rainbow trout.  相似文献

Reproduction related immunoglobulin changes in rainbow trout   总被引:4,自引:1,他引:4  
Annual changes in plasma immunoglobulin (IgM) levels were investigated in three strains of rainbow trout Oncorhynchus mykiss which have different spawning periods, i.e., September–October, November–December, and January, reared under constant water temperature and natural day length. Plasma IgM levels decreased during the spawning season in all strains tested. The IgM changes became reversed in response to significant increases in plasma testosterone (T) and estradiol-17 in females and T and 11-ketotestosterone in males. Though the IgM decline showed a connection with suppressed immunocompetence, since many mature fish caught fungal diseases, no clear differences were observed in the plasma IgM levels between infected and noninfected fish during the spawning season. Incidentally, plasma IgM levels in infection prone fish were higher than in noninfection prone fish prior to the spawning season, whereas coincident differences in the plasma steroid levels were observed. Immature fish reared under lower water temperatures showed lower IgM levels. The effect of water temperature may have to be considered when analyzing the defense mechanism during the spawning season in rainbow trout.  相似文献

Rainbow trout (Oncorhynchus mykiss) were acclimated to 4 and 18°C and fed at rations levels which led to an equal change in mass at these temperatures during the six week acclimation. Thermal acclimation markedly modified the metabolic organization of red and white muscle. Cold-acclimated fish had activities of -hydroxyacyl CoA dehydrogenase in both red and white muscle that were twice those of warm-acclimated fish. By contrast, the activities of cytochrome oxidase were unchanged by thermal acclimation. Thus, the capacity for -oxidation of lipids is specifically enhanced in the muscle of cold-acclimated trout. In white muscle, citrate synthase and phosphofructokinase activities were also enhanced by cold acclimation (increases of 125% and 35% respectively), while cytochrome oxidase levels were unchanged. The non-parallel changes in the activities of mitochondrial enzymes strongly suggest that trout muscle mitochondria undergo qualitative reorganization during cold acclimation. The relative activities of mitochondrial enzymes suggest that mitochondria from red muscle have a threefold greater capacity to oxidize lipids than those from white muscle.  相似文献

Rainbow trout around 743 g were fed four different diets over an 8-month period. Maturing males grew faster in the period before spawning (Sept.–Dec.) than maturing females, and significantly faster than immature fish. Mature fish stagnated in growth while the immature fish surpassed the growth of mature fish during the spawning season. The condition factor was moderately influenced by the maturation process. In the period before spawning (Sept.–Jan.) maturing females had a significantly higher hepatosomatic index than males and immature fish. An increase in dry matter and fat and a decrease of protein were observed in the muscle of maturing males and immature fish towards the spawning season, while maturing females had a slight fall in dry matter and fat and a small increase in protein in the same period. Dry matter and fat in muscle of rainbow trout were positively correlated.  相似文献

This paper reports on the effect of administration of mammalian growth hormone (GH) on muscle protein synthesis as measured in white muscle using the phenylalanine flooding technique. The effect of exogenous GH was compared with that of insulin and prolactin, and with endogenous GH.The rate of protein synthesis in white muscle of rainbow trout 6 h after the injection of bovine GH or bovine insulin was twice (2.6 and 2.9% d–1) that of the control saline-injected fish (1.2% d–1). A metabolic effect of GH, as observed with insulin, is suspected.The rates of change in body weight and body length and the fractional rate of protein synthesis in muscle of rainbow trout were enhanced by mammalian GH administration. The effect of GH on muscle RNA/protein ratios was not significant. An opposite effect of antibodies against salmon GH (Lebailet al. 1989) on growth rate and muscle protein synthesis rate was found in rainbow trout. It is suggested that the effects of exogenous and endogenous GH on capacity and efficiency of muscle protein synthesis were similar.The long-term effects of mammalian GH on presmolt Atlantic salmon was also tested. The same trends were found with ovine prolactin supplementation in Atlantic salmon but not as high as those observed with ovine GH.  相似文献

Adult female rainbow trout (Salmo gairdneri) were fed a commercial trout production diet until approximately 40 days prior to spawning at which time the fish were divided in two groups. One group continued to be fed while the other group was starved until spawning. Egg size, egg number, ovary weight, and egg proximate composition were examined periodically starting 5 months before spawning. Muscle tissues of the fish were sampled at spawning to measure somatic depletion of proximate consituents in the two groups. Egg hatchability was determined by incubating test lots from each spawning female. No significant differences (P > 0.05) were found in egg hatchability between the fed and starved groups. Changes in proximate composition were observed in the eggs during the months prior to spawning, but no differences were detected between the eggs of the starved and the fed groups. Short-term starvation did not significantly alter the proximate composition of muscle tissue of the spawned female trout.  相似文献

SUMMARY: Behavioral experiments concerning a releaser pheromone in the urine of female rainbow trout were performed using immature fish administered orally with 17α-methyltestosterone (MT) during the non-spawning season. The urine was collected by catheter. The frequency of entries of test fish was recorded in each channel scented by test and control solutions in a Y-maze trough. The behavior of both MT-treated and control fish demonstrated that they could not discriminate the differences between distilled and environmental water as control solutions. There was also no difference between MT-treated and control fish when distilled and environmental water were introduced. The MT-treated immature fish were attracted to the channel scented by ovulated female urine. Neither coelomic fluid nor the immature female urine had any effect on the behavioral responses of MT-treated fish, while immature control fish had no preference for the urine of ovulated females. These results suggest in rainbow trout that ovulated female urine contains a releaser pheromone to attract mature males, and that androgens are involved in the sensory mechanisms detecting the releaser pheromone in fish.  相似文献

Adiponectin is one of the adipokines secreted mainly from adipocytes in mammals. In rainbow trout, however, adiponectin is highly expressed in skeletal muscle. Although it has been suggested that fish skeletal muscle contains adipocytes, their endocrine function and distribution are poorly understood. Recently, an EST analysis of rainbow trout found that heart-type fatty acid binding protein (H-FABP), a member of the intracellular fatty acid binding protein family, encodes an adipose-specific gene. In this study, we produced anti-adiponectin and H-FABP antibodies and investigated the distribution of adipocytes and related cells in skeletal muscle of rainbow trout. The adiponectin signal was detected at around 75 kDa in muscle in Western blotting. Since the molecular mass of rainbow trout adiponectin is around 25 kDa, this 75 kDa band would be a trimer. For H-FABP, the signal band was detected at around 15 kDa. Immunohistochemistry of skeletal muscle sections indicated that adiponectin and H-FABP signals were present outside of muscle cells and throughout the muscle tissues, suggesting the existence of adipocyte-related cells in these regions. These results will contribute to our understanding of energy metabolism in fish skeletal muscle.  相似文献

The present work investigates the importance of circadian and circannual rhythms in the photoperiodic control of reproduction in the rainbow trout. Maintenance of groups of 20–30 female trout under continuous light (LL), constant long (18L:6D) or short (6L:18D) days and conditions of constant temperature (8.5–9.0°C) and feeding rates (0.5% body weight, day–1), starting in February, produced markedly different spawning periodicities during the first and subsequent years of treatment. At the end of the first year, spawning was advanced by up to 2 months in the fish under LL and 18L:6D and delayed by up to 5 months in those under 6L:18D when compared with the December spawning of control fish under ambient light. Continued exposure of the fish to the same photoperiod regimes produced cycles of spawning and peak levels of testosterone, oestradiol-17 and calcium (as an index of vitellogenin) at intervals of 150–170 days under LL and 18L:6D and 320–420 days under 6L:18D. The functional importance of these spawning cycles and their relationship to circannual rhythms and the control of reproduction is discussed.Exposure of fish to skeleton (6L:4D:2L:12D, 6L:6D:2L:10D and 6L:8D:2L:8D) and resonance (6L:42D, 6L:48D and 6L:54D) procedures produced ranges of spawning times up to two months in advance of control fish. Results with the resonance regimes, where fish received only a half the light-dark cycles and a quarter of the total daylight hours of those on ambient light cycles show that trout do not measure daylength and time by counting daily cycles or by hour-glass mechanisms. Collectively, these data support the proposition that rhythmic processes of photosensitivity are involved in the modulation of reproductive development in the rainbow trout and that circadian and circannual rhythms cooperate in the timing and entrainment of this cycle.  相似文献

The cytochrome P-450 content in liver cells from rainbow trout was not affected by starvation for 12 weeks whereas the rate of cytochrome P-450-dependent deethylation of 7-ethoxycoumarin in liver cells from 6 or 12 weeks starved fish was 60% of the rate in fed fish. Treatment of fish with -naphthoflavone increased the 7-ethoxycoumarin metabolism several-fold in both starved and fed fish.Optimal cytochrome P-450 monooxygenation in liver cells from fed or starved fish was not affected by addition of glucose or 2-bromooctanoate, an inhibitor of fatty acid -oxidation which is the main source of metabolic fuel in trout liver. The cellular content of NADPH, an obligatory cofactor for cytochrome P-450 monooxygenation, was not affected by addition of substrate to cytochrome P-450, inhibition of fatty acid -oxidation or inhibition of the oxidative phosphorylation. This indicates a great capacity of rainbow trout liver cells to retain high NADPH/NADP+ ratios. These results suggest that the cytochrome P-450 mediated metabolism of xenobiotics in liver cells from fed or starved trout is not limited by the availability of reducing equivalents.  相似文献

Hybridisation with introduced taxa poses a threat to native fish populations. Mechanisms of reproductive isolation can limit or prevent hybridisation between closely related species. Understanding how these mechanisms interact between the same species across geographically distinct occurrences of secondary contact, and how regional factors influence them, can inform our understanding of hybridisation as a threat and management actions to mitigate this threat. We used data collected on adult fish migration timing and approximate emergence timing of subsequent juvenile fish paired with genomic data to assess whether temporal isolation in the timing of spawning exists between Yellowstone cutthroat trout, rainbow trout and hybrids in the North Fork Shoshone River drainage in northwest Wyoming. We found evidence that Yellowstone cutthroat trout spawn, on average, two to four weeks later than rainbow trout and hybrids and two environmental covariates related to water temperature and discharge were associated with differences in spawning migration timing. Despite statistical support for Yellowstone cutthroat trout spawning later, disproportionately high numbers of rainbow trout and hybrids, paired with extended spawning seasons, lead to substantial overlap between all genotypes. Our results provide further evidence of temporal segregation in the timing of spawning as a mechanism of reproductive isolation between closely related species, but substantial spawning overlap suggests temporal segregation alone will not be enough to curtail hybridisation in conservation populations.  相似文献

The role of cortisol in the mobilization and metabolism of amino acids following exhaustive exercise in rainbow trout (Onorhynchus mykiss) was investigated. Plasma cortisol levels were elevated for 4h following exercise and 6, of a total of 22 amino acids, showed significant responses. In white muscle, alanine and glutamine were elevated immediately after exercise through to 4h, whereas glutamate, and the branched-chain amino acids (BCAA), isoleucine, leucine and valine, all decreased over this time. In plasma all of these amino acids increased from 2-4h post-exercise, while in liver, alanine and glutamine increased, glutamate did not change and the BCAAs declined over this time. Blockade of the post-exercise elevation in plasma cortisol with either metyrapone (cortisol synthesis inhibitor) or dexamethasone (cortisol release blocker) prevented the changes in tissue amino acid levels. This study demonstrates that cortisol can act rapidly (within minutes to hours) to alter amino acid metabolism in fish. A model is presented to explain the action of cortisol on amino acid metabolism.  相似文献

Increased use of plant oils with different origins and quality in fish feed needs to be approached from a food safety and fish welfare point of view. Plant oils contain a number of bioactive minor lipid compounds that may affect the fish’s metabolism and taste perception. This study focuses on the effect of replacing fish oil (FO) with different levels of cold‐pressed rapeseed oil (RO) on the lipid composition in muscle and liver as well as on the preference by the fish. Rainbow trout (Oncorhynchus mykiss) were fed diets with a FO : RO ratio of 100 : 0, 75 : 25, 50 : 50 and 25 : 75 until twofold weight increase. In self‐selecting feed trials of single rainbow trout, fish preferred the diet composed of only FO compared with the diets with RO but did not discriminate between different levels of RO. Plant sterols and their metabolites were found in liver of the fish fed RO diets, suggesting an effect on the sterol metabolism different from fish fed a 100% FO diet. The largest effects were seen in the fatty acid composition of the edible tissue of the fish with a decrease in 22:6n‐3 and 20:5n‐3 and an increase in 18:2n‐6 and 18:1n‐9.  相似文献

Supranutritional dietary selenium (Se) has been demonstrated beneficial for fish health, however, its effects on fish flesh quality remain unclear. This study investigated the effects of supranutritional dietary Se on the flesh quality of rainbow trout (Oncorhynchus mykiss) and their potential mechanism. Fish were fed a basal diet supplemented with or without graded Se yeast for 10 weeks. Results showed that Se supplementation significantly increased fillet crude protein and enhanced fillet water‐holding capacity as well as fillet firmness. Supplementing with both 2 and 4 mg/kg Se significantly downregulated the expressions of two autophagy–lysosome‐related genes (autophagy‐related 12‐like and gamma‐aminobutyric acid type A receptor‐associated protein‐like 1) in fish muscle, while supplementing with 4 mg/kg Se also significantly downregulated the expression of two ubiquitin–proteasome‐related genes (muscle RING finger 2 and F‐box protein 25). Correlation analysis indicated that the improved fillet quality parameters were closely correlated with the expressions of these differentially expressed genes. This study revealed that dietary Se was effective for the improvement of rainbow trout flesh quality, and the improved fish flesh quality was associated with the inhibited protein degradation in fish muscle.  相似文献

Two experiments were conducted to compare the patterns of plasma free amino acid concentrations after force-feeding in rainbow trout Oncorhynchus mykiss (Walbaum) with and without dorsal aorta cannulation. In the first experiment, 35 rainbow trout averaging 504 ± 7.8 g (mean ± SD) were divided into seven groups of five fish each. After 48 h starvation, a group of fish was anaesthetized and blood samples were taken at one of the following time periods: 0, 4, 8, 12, 24, 36 and 48 h after feeding. In the second experiment, five dorsal aorta cannulated rainbow trout averaging 511 ± 6.2 g (mean ± SD) were kept in a cage. After 48 h starvation, the fish were anaesthetized and blood samples were taken from the same fish at 0, 4, 8, 12, 24, 36 and 48 h after feeding. In the first experiment, the concentration of all plasma free amino acids except histidine and glycine peaked at 4 h and returned to the basal level 24 h after feeding. In the second experiment, the concentration of all plasma free amino acids except isoleucine, leucine, phenylalanine and tryptophan also peaked at 4 h and returned to the basal level 24 h after feeding. These results showed that the pattern of plasma free amino acid concentrations from fish with and without dorsal aorta cannulation were similar.  相似文献

As part of the investigation into cysteine metabolism in fish, sulfur amino acids and their derivatives were injected intraperitoneally to fingerling rainbow trout (Oncorhynchus mykiss) to examine how the doses of these compounds affect the hepatic cysteine dioxygenase [EC 1.12.11.20] in this species. A dose of 0.25 mmol L-cysteine per 100 g body weight induced the enzyme activity as much as 2.5 times that of the control fish within 4h after the injection. The activity increased proportionally to the increasing dose of cysteine up to the dose of 0.15 mmol per 100 g body weight. The induction was observed to be rather specific to L-cysteine. These findings suggested that the cysteine sulfinate pathway might play an important role in the metabolism of excess cysteine in rainbow trout. The dosage of L-cysteine larger than 0.50 mmol per 100g body weight led to mortality of the fish. The pathway of cysteine catabolism was considered to function to prevent toxic accumulation of cysteine in rainbow trout, as in the case of mammals.  相似文献

Three experiments were conducted in order to evaluate the effects of urea and trimethylamine oxide (TMAO) supplementation in diets for seawater‐farmed rainbow trout Oncorhynchus mykiss. In all experiments, commercial extruded feed was used as basal control diet, whereas experimental diets were the basal diet supplemented with either 10 g kg–1 TMAO, 10 g kg–1 urea or 10 g kg–1 TMAO and 10 g kg–1 urea. The experiments were conducted both in research units and in a commercial fish farm. Quality was evaluated in three different commercial slaughterhouses, sorting the rainbow trout into superior, ordinary or processing grade. Significantly more TMAO was observed in muscle of rainbow trout given dietary supplementation of TMAO. However, the levels were low compared with levels in marine fish in general. Trimethylamine (TMA) was not found. No increase in muscle urea content was observed in trout fed urea in the diet. Significantly reduced fat belching, seen as reduced number of fat droplets surfacing in the net pen, was observed in groups given TMAO‐supplemented feeds. At least twice as many rainbow trout with skin vesicles were observed in the control group as in the group fed urea‐supplemented diet. In addition, skin vesicles developed into open sores in the winter only in rainbow trout fed control diet. In a large‐scale trial, quality grading of almost 200 000 market‐sized rainbow trout showed significantly more (88 vs. 83%) trout of superior grade in the group fed dietary urea compared with fish fed control diet. The main reason for downgrading from superior to ordinary and processing grade was skin lesions, supporting the observations made in the experimental units.  相似文献

Non‐native trout are currently stocked to support recreational fisheries in headwater streams throughout Nebraska. The influence of non‐native trout introductions on native fish populations and their role in structuring fish assemblages in these systems is unknown. The objectives of this study were to determine (i) if the size structure or relative abundance of native fish differs in the presence and absence of non‐native trout, (ii) if native fish‐assemblage structure differs in the presence and absence of non‐native trout and (iii) if native fish‐assemblage structure differs across a gradient in abundances of non‐native trout. Longnose dace Rhinichthys cataractae were larger in the presence of brown trout Salmo trutta and smaller in the presence of rainbow trout Oncorhynchus mykiss compared to sites without trout. There was also a greater proportion of larger white suckers Catostomus commersonii in the presence of brown trout. Creek chub Semotilus atromaculatus and fathead minnow Pimephales promelas size structures were similar in the presence and absence of trout. Relative abundances of longnose dace, white sucker, creek chub and fathead minnow were similar in the presence and absence of trout, but there was greater distinction in native fish‐assemblage structure between sites with trout compared to sites without trout as trout abundances increased. These results suggest increased risk to native fish assemblages in sites with high abundances of trout. However, more research is needed to determine the role of non‐native trout in structuring native fish assemblages in streams, and the mechanisms through which introduced trout may influence native fish populations.  相似文献
