Outbreaks of mass mortality of postlarval abalone, Haliotis diversicolor supertexta, have occurred in south China since 2002 and have forced many abalone farms to close. About 30 representative bacterial strains were isolated from a sample of five diseased postlarval abalone, taken 25 days post-fertilization during an outbreak of postlarval disease in Shenzhen, China, in October 2006. Bacterial challenge tests showed that the predominant strain, designated as strain 22, was highly virulent to postlarvae with an LD50 value of 7.8 × 104 colony forming units (CFU) ml−1, while four of the other isolates were weakly virulent with LD50 values ranging from 1 × 106 to 1 × 107 CFU ml−1, and the remaining 25 isolates were classified as avirulent with LD50 values greater than 1 × 108 CFU ml−1. By means of API 20NE and 16S rDNA and ITS sequencing analyses, strain 22 was identified as Pseudomonas aeruginosa. Antibiotic susceptibility tests showed that strain 22 exhibited around 75% of susceptibility to 16 various antibiotics tested. The results of this study show P. aeruginosa as one of the bacteria involved in the mortality of abalone postlarvae in Shenzhen, China.  相似文献

In this study, β-haemolysin gene (AHTPS30HEM) of Aeromonas hydrophila was cloned from diseased fish in mainland China. AHTPS30HEM gene (AB021152) resulted in a 1589 bp fragment which covers the open reading frame (ORF) in region 5–1486 coding for 493 amino acids. Multiple alignment of AHTPS30HEM with other β-haemolysin amino acid sequences showed 18 amino acid substitutions between AHTPS30HEM and A. hydrophila β-haemolysin. Although the ORF sizes between AHTPS30HEM and Aeromonas species are different, four cysteins and four potential N-glycosylation sites were conserved. To identify the β-haemolysin-producing virulent or pathogenic A. hydrophila, a specific PCR to amplify 208 bp target DNA of β-haemolysin gene was established. Twenty strains containing pathogenic A. hydrophila, Aeromonas sobria, Vibrio anguillarum, Cytophaga columnaris, Pseudomonas fluorescens, and Yersinia yuckeri were investigated by PCR. Based on the cloned β-haemolysin sequences, the specific PCR method for identification of the β-haemolysin gene of A. hydrophila was established, and surveyed on those samples. The results indicated that β-haemolysin-specific PCR might be useful in the detection of pathogenic A. hydrophila.  相似文献

Fisheries-independent survey data are vital to stock assessment and management because they provide reliable information on stock status. Although survey data have been increasingly recognised for their contributions to fisheries management, they have often not been adequately used to provide quantitative management recommendations for China's fisheries that are subject to limited data. In the present study, a framework was proposed to integrate two types of survey-based management procedures (MPs) into China's fisheries management: the slope-based MP and the target-based MP. Specifically, the former generates fishing effort limits based on the trend in recent years’ abundance index, while the latter sets effort limits based on a static abundance index target. To test the performance of these MPs, management strategy evaluation was performed using a simulated fishery based on the small yellow croaker, Larimichthys polyactis (Bleeker) in Haizhou Bay, China. The sensitivity of MPs was investigated under different algorithm parameterisations and historical exploitation levels. Additionally, their robustness was evaluated in the face of survey uncertainty and changes in recruitment success. Both MPs could effectively improve the stock status compared with the status quo management when appropriately parameterised regardless of exploitation history. The target-based MP was more robust to survey uncertainty than the slope-based MP. Non-stationary changes in recruitment success strongly impaired the management effects, while using recruitment-specific instead of the age-aggregated abundance index could slightly enhance their performance. This work indicates that survey-based MPs can address the current challenges in China's fisheries management and reinforce the status quo management practice with limited data and highlights the potential risks.  相似文献

In this study, we investigated the bacterial communities in the shrimp and sea cucumber culture environment, including shrimp ponds (SP), sea cucumber ponds (SCP), mixed-culture ponds (MCP) and the effluent channel (EC) in Qingdao, China. Bacteria cultivation showed that the counts of heterotrophic, nitrate-reducing and sulfate-reducing bacteria in the sediment of SP were higher than that in the sediment of SCP and MCP, varying between 8.7 × 104 and 1.86 × 106, 2.1 × 104 and 1.1 × 105, and 9.3 × 101 and 1.1 × 104 CFU g?1, respectively. In contrast, the counts of ammonium-oxidizing and nitrifying bacteria in the sediment of SP was lower than that in the sediment of SCP and MCP. Denaturing gradient gel electrophoresis (DGGE) of 16S rDNA gene and dendrogram analyses showed that bacterial diversity in the mixed-culture environment was higher than that in the monocultures. The similarity of bacterial community between EC and SCP or MCP was higher than that between EC and SP. These results indicated that sea cucumber culture played a significant role in influencing the environmental bacterial communities that were composed mainly of Flavobacteriaceae (64.3%), Bacteriodetes (21%) and delta proteobacteria (14.7%), including the genera of Croceimarina, Lutibacter, Psychroserpens and so on. The results explained the benefit of sea cucumber culture in shrimp ponds at the level of microbial ecology.  相似文献

The spawning grounds of the chub mackerel (Scomber japonicus) and spotted mackerel (Scomber australasicus) in the East China Sea were estimated based on catch statistics of the Japanese large- and medium-type purse seine fishery from 1992 to 2006. Biometric data were obtained from specimens caught by purse seiners in the East China Sea from 1998 to 2006. Gonadosomatic index (GSI) at 50% sexual maturity of chub mackerel and spotted mackerel females was 2.5 and 2.6, respectively. Using this criterion for GSI, chub mackerel larger than 275 mm and spotted mackerel larger than 310 mm in fork length were considered to be mature. Mature chub mackerel was observed in the area of 15–22°C sea surface temperature (SST), and mature spotted mackerel was observed in the area of 17–25°C SST. The spawning period of chub mackerel ranged from February to June, and that of spotted mackerel ranged from February to May in the East China Sea. The spawning grounds were estimated from the distributions of catch per unit effort (CPUE) of spawners and SST. As a result, the spawning ground of chub mackerel was estimated to be in the central and southern part of the East China Sea and the area west of Kyushu in February, March, and April, and in the central part of the East China Sea, the area west of Kyushu and Tsushima Straight in May, and in Tsushima Straight and western part of the Sea of Japan in June. The spawning ground of spotted mackerel was estimated to be in the central and southern part of the East China Sea and southern coastal area of Kyushu in February, March, and April, and the central and southern part of the East China Sea and the area west of Kyushu in May.  相似文献
