The haematological and biochemical characteristics of two healthy farmed cyprinids, the topmouth culter Culter alburnus and yellowcheek carp Elopichthys bambusa, were investigated in this study. Erythrocytes, thrombocytes, lymphocytes, monocytes and granulocytes (i.e. neutrophils and eosinophils) were observed in these two fish. Every type of these cells (excluding the erythrocyte and lymphocyte) showed similar sizes in the topmouth culter and yellowcheek carp. Thrombocytes and neutrophils were the two most abundant leucocytes in the topmouth culter while thrombocytes and lymphocytes were the two most frequent leucocytes observed in the yellowcheek carp. The erythrocyte counts, haemoglobin concentrations and values of serum glucose in these two fish were high. There were significant differences in the leucocyte counts, haemoglobin concentrations, mean cellular haemoglobin contents, mean cell haemoglobin concentrations and values of serum glucose, triglyceride, total bilirubin, alkaline phosphatase and chlorine between the topmouth culter and the yellowcheek carp. The information of haematology and blood biochemistry obtained here would be useful for the prevention and diagnosis of diseases of farmed topmouth culter and yellowcheek carp.  相似文献

Nine experimental diets at three protein (35%, 40% and 45% crude protein) and lipid (5%, 8% and 11% crude lipid) levels with variable digestible protein to digestible energy (DP/DE) ratios ranged from 21.9 to 27.8 g protein MJ?1 were fed to topmouth culter (Culter alburnus Basilewsky) fingerlings (initial weight 6.5 ± 0.9 g) in triplicated groups (30 fish per replicated) for a period of 10 week to assess the optimum dietary DP/DE ratio and the protein sparing effect by utilizing dietary lipid. 27 cages of 1.5 m3 capacity placed in a lake located in Wuhan were used for rearing the fish. At the end of the experiment, maximum weight gain and thermal‐unit growth coefficient was found in fish fed diet D4 with 45% protein, 8% lipid and P/E ratio of 26.2 g protein MJ?1, but without a significant difference compared to fish fed diet D5 with 40% protein, 8% lipid and DP/DE ratio of 25.3 g protein MJ?1. The best flesh quality evaluated by muscle collagen content was found in fish fed D5. High fat accumulation with increasing dietary lipid levels was observed in whole body but not in muscle tissue. Hence, it may be concluded that the optimum formulation for maximum growth and quality of topmouth culter is a diet containing 40% protein and 8% lipid with a resultant DP/DE ratio of 25.3 g protein MJ?1. In addition, the protein sparing effect by inclusion lipid was observed but limited.  相似文献

釆用MS-222浓度为0(C0组)、5 mg/L(C1组)、10 mg/L(C2组)、15 mg/L(C3组)、20 mg/L(C4组),运输时间为0、2、4、6、8、10 h,运输密度为33 g/L,采用5×6双因素实验,氧气袋运输翘嘴鲌(Culter alburnus)稚鱼,研究了运输时间和MS-222浓度对翘嘴鲌全鱼皮质醇、乳酸含量及氧气袋内水质的影响。结果显示:翘嘴鲌全鱼皮质醇、乳酸水平随运输时间呈现先升高后降低的趋势,随麻醉剂浓度的增加而显著降低;C1-C4组的DO、p H随运输时间呈现先降低后升高的趋势,C0组则随运输时间的增加显著降低,各组的DO和p H随MS-222浓度的增加而显著增加;C1-C4组的氨氮(TAN)和游离CO_2浓度随运输时间呈现先升高后降低的趋势,而C0组则随运输时间的增加显著升高,各组的TAN和游离CO_2浓度随MS-222浓度的增加而显著降低。时间和浓度的交互作用都显著。运输时间T6 h时,MS-222刺激翘嘴鲌,不利于运输,运输时间T6 h时,MS-222能显著降低其应激水平及代谢强度,其中C4组皮质醇、乳酸含量最低,C3、C4组水质条件最好,其适宜的麻醉剂浓度为15~20 mg/L。  相似文献

Growth and survival of 0+ perch were studied in 4 ponds with the topmouth gudgeon (Pseudorasbora parva) and without it. In the end of April, all ponds were stocked by free-swimming perch larvae (120,000 ind ha?1). In June, topmouth gudgeon was introduced as forage fish (40 kg ha?1) into two of the experimental ponds. Topmouth gudgeon significantly influenced neither the total abundance of zooplankton nor the abundance of its groups (Rotifera, Cladocera, Copepoda). The most important food item for perch (TL > 29 mm) was macroinvertebrates (especially Chironomidae). In the ponds with topmouth gudgeon, copepods and cladocerans were more important than in ponds without it. Specific growth rate of perch was 0.01 mm day?1 in all ponds. Final mean total length (TL ± SD) of perch was 73 ± 13 and 70 ± 6 mm in the ponds with topmouth gudgeon and without it, respectively. Only 1 % of the perch reached higher TL than that recorded in the ponds without the topmouth gudgeon. Survival rate of perch varied from 12 to 36 % depending on ponds. Piscivory was recorded in all ponds from the age of 57 days (post-stocking); however, perch siblings were preferred to topmouth gudgeon. The highest asymptotic growth (L∞ = 88 mm) was calculated in the pond stocked with topmouth gudgeon. This corresponded with the highest cannibalism and lowest survival rate (12 %). Perch growth rate increased till 42–53 days of perch age and then started to decrease. There was no significant influence of potential prey fish (topmouth gudgeon) on the growth of 0+ perch; however, two size cohorts were found in the ponds with the topmouth gudgeon. Presence or absence of littoral macrophytes seems to be more substantial for rearing perch in ponds than stocking with the prey fish.  相似文献

为研究新型杂交鱼翘嘴鳊(二倍体鲂鲌F1♀×团头鲂♂)的肌肉营养价值,实验运用生化方法测定和分析了翘嘴鳊、翘嘴红鲌、团头鲂、二倍体鲂鲌F1(团头鲂♀×翘嘴红鲌♂)的肌肉营养成分,包括这些样品的水分、脂肪、蛋白质和灰分含量,以及脂肪酸组成和氨基酸组成。结果表明,翘嘴鳊与其原始亲本相比具有较高的蛋白质和较低的碳水化合物含量;脂肪酸分析发现翘嘴鳊的不饱和脂肪酸,尤其是油酸、DHA等含量显著高于其原始父母本和其他有关鱼类(P0.05);翘嘴鳊肌肉中具有较高呈味氨基酸比例,该比例高于其父母本的相应比例。研究表明,翘嘴鳊是一种营养价值高、口感好的优质鱼类。  相似文献

Three single nucleotide polymorphisms, c.1462C > T, c.1663G > T and c.2023G > A, in smad4 gene were isolated and typed in a normal population (meio‐G0), a first‐generation gynogenetic population (meio‐G1) and a second‐generation gynogenetic population (meio‐G2) in topmouth culter (Culter alburnus) by using the tetra‐primer amplification refractory mutation system (ARMS) PCR. The homozygosity of different loci in the three populations was analysed to study the efficiency of gene purification in this fish. Correlation of the three populations to seven growth traits (body length, body weight, body thickness, body height, total length, caudal peduncle length and caudal peduncle height) was estimated. Results showed that homozygosity and homozygosity rates of three SNP loci from meio‐G0 to meio‐G2 increased obviously, and the average homozygosity of c.1462C > T and c.1663G > T loci was 0.930 and 0.880 in meio‐G2 population, which was significantly higher than that in meio‐G0 population. In the meio‐G1 and meio‐G2 populations, correlation of seven growth traits with the CC genotype was significantly higher than that with the TT and TC genotypes in the c.1462C > T locus (p < .05). These results suggested that the CC genotype in the c.1462C > T locus could be an advantage mutation that stably affects the growth of different generations of this fish. It may be used as a potential molecular marker for gene‐assisted selection to improve a range of growth traits in topmouth culter.  相似文献

The loss of sperm quality in sterlet (Acipenser ruthenus) due to freeze-thaw process in cryopreservation was investigated in the present study. Two antifreeze proteins (AFPI or AFPIII) were used at different concentrations of 0.1, 1, 10, and 100 μg/mL. We compared motility, curvilinear velocity, and plasma membrane integrity of fresh, cryopreserved sperm, and sperm cryopreserved in the presence of antifreeze proteins. Fresh sperm (control) had 85?±?4% motility and 160?±?2 μm/s curvilinear velocity, respectively. After cryopreservation, the motility of frozen-thawed sperm without addition of antifreeze proteins significantly decreased (44?±?9%), compared to the control. The highest motility of frozen-thawed sperm was obtained in cryopreserved sperm with addition of 1 μg/mL of AFPIII (58?±?14%). No significant differences were observed in curvilinear velocity between fresh sperm and cryopreserved sperm with/without addition of AFPI or AFPIII. The flow cytometry analysis revealed that fresh sperm contained 94.5?±?6% live cells, while the cryopreserved sperm only contained 26.6?±?14% live cells. Supplementation of antifreeze proteins has significantly improved the percentage of live cells in frozen-thawed sperm, except 0.1 μg/ml of AFPI group. No significant difference in percentage of live cells was detected in the sperm cryopreserved with 10 μg/mL of AFPI or AFPIII, compared to fresh sperm. Thus, addition of antifreeze proteins to cryopreservation medium could be considered to improve the post-thawed sperm quality of sterlet.  相似文献

2014年7~10月在黑龙江干流抚远江段采集红鳍原鲌Cultrichthys erythropterus样本249尾,研究抚远江段红鳍原鲌种群的年龄结构与生长特性。结果表明,黑龙江抚远江段红鳍原鲌种群的年龄组成为1~7龄,以4~5龄鱼为主,占总样本量的67.8%;红鳍鲌为异速生长型,其体长和体质量的关系为W=0.0038L~(3.403)(n=248,R2=0.99,P0.01),红鳍原鲌体长与体质量的生长符合Von Bertalanffy生长方程:Lt=46.317[1-e~(-0.174(t+0.235))],Wt=1 771.325[1-e~(-0.174(t+0.235))]3.403,其生长拐点为t=6.80龄,对应的体长和体质量分别为32.70cm和541.66g。建议捕捞规格提高到体长300mm或体质量500g以上,以可持续开发利用黑龙江抚远江段红鳍原鲌渔业资源。  相似文献

In this study, the effects of peripheral (intraperitoneal) injections of d-amphetamine on feeding behavior were assessed in goldfish. Compared with the saline-injected group, amphetamine injections decreased food intake at doses ranging from 1 to 75 μg/g, but not 0.5 μg/g, but increased locomotor behavior, as indicated by the increased number of total feeding and non-feeding acts, at doses ranging from 2.5 to 25 μg/g. Amphetamine at high doses inhibited both food intake (at 25, 50 and 75 μg/g) and feeding behavior (at 75 μg/g). In the hypothalamus, the expression of orexin was down-regulated, and both CART 1 and CART 2 expressions were up-regulated in amphetamine-treated fish (50 μg/g) as compared to saline-injected fish, but amphetamine treatment had no effect on either hypothalamic TH or TRH expression. In the telencephalon, amphetamine treatment (50 μg/g) up-regulated CART 1, CART 2 and TH mRNA expressions but had no effect on either orexin or TRH. Our results suggest that, as in mammals, the orexin, CART and TH systems might be involved in amphetamine-induced feeding/locomotor responses in goldfish.  相似文献

对中华倒刺鲃(Spinibarbussinensis)、白甲鱼(Onychostomasimus)和岩原鲤(Procyprisrabaudi)3种鱼的鲜精在不同水溶液和不同浓度梯度的NaCl溶液中的活力以及精子形态、密度和精液pH值、浓度等进行了比较研究。结果表明:3种鱼的精液pH值都偏弱碱性,位于7·2~7·7之间;精液浓度依次为71·1%,76·1%和71%;精子头长依次为(3·89±0·53)μm,(2·98±0·08)μm和(6·42±0·59)μm,全长依次为(41·63±3·66)μm,(65·67±2·97)μm和(58·89±5·25)μm;精子密度依次为1·527×1010尾/mL,1·336×1010尾/mL和1·362×1010尾/mL。分析表明,3种鱼的精子大小与密度无相关性。在不同水溶液中,3种鱼的精子活力均以池塘水中最高;在不同浓度NaCl溶液中,3种鱼精子的最适浓度位于0·45%~0·55%之间,有效运动时间以中华倒刺鲃最高,为(85·23±12·02)s,其次是岩原鲤,为(50·45±6·89)s,白甲鱼最短,为(31·44±5·53)s。3种鱼的精子全长与精子活力存在负相关性,即精子越长,活力越低,精子越短,活力越高。  相似文献

ABSTRACT:   The modori phenomenon is defined as heat-induced myofibrillar degradation caused by endogenous serine protease(s) of fish muscle during Kamaboko fish meat gel production. This study was undertaken to analyze myofibrillar proteolysis of white croaker Argyrosomus argentatus muscle, which is an ingredient of high quality Kamaboko, by myofibril-bound serine protease (MBSP) under conditions corresponding to the modori phenomenon. White croaker MBSP was stable between pH 2–11 and below 65°C, and about 60% of its initial activity remained after incubation for 2 h under the conditions at 65°C and pH 7.5. About 60% of the enzyme activity was suppressed by 0.5 M NaCl. White croaker MBSP degraded various myofibrillar proteins between 40 and 70°C and pH 6.0–9.0, and preferentially degraded myosin heavy chain rather than other myofibrillar proteins. The enzyme degraded the myosin heavy chain most strongly at 55°C and pH 7.0, and a major part of the bands of myosin heavy chain and its degradation products disappeared for a period of 2 h. These degradation characteristics are very similar to those observed during the modori phenomenon, indicating that MBSP could be a modori-inducing protease involved in the modori phenomenon of white croaker Kamaboko production.  相似文献

ABSTRACT

This study assessed effects of high pressure processing on physicochemical quality and endogenous enzyme activities of grass carp fillets stored at 4 ºC for 21-day storage. Fresh fillets were vacuum-packed and subjected to high pressure processing (0.1, 200, 300, 400, 500, and 600 MPa at room temperature for 15 min). Results showed that high pressure processing significantly delayed microbial growth, and reduced total volatile basic nitrogen (TVB-N) and thiobarbituric acid reactive substances (TBARS) levels of grass carp fillets, especially for the 600 MPa group. On the 21st day, the corresponding increases of 6%, 35%, 45%, 43%, and 53% in hardness were observed for 200 MPa, 300 MPa, 400 MPa, 500 MPa, and 600 MPa samples compared with control samples, respectively. In addition, enzyme activities in different groups decreased during storage. Among them, calpain, and myofibril-bound serine proteinases (MBSP) activities were immediately reduced after high pressure treatments, while the activities of collagenase, cathepsin B, and cathepsin D were significantly (< 0.05) inhibited when the pressure exceeded 400 MPa. However, the pressure treatments activated cathepsin L initially, and then the activity gradually decreased in treated samples. Generally, high pressure processing reduced activities of most of the endogenous enzymes and improved the quality and extended the shelf life by at least 4 ~ 6 days of refrigerated grass carp fillets.  相似文献

A 6-week experiment was performed to compare different carbon sources, i.e. sucrose, glycerol and rice bran, to a nitrogen ratio of 15:1 in a biofloc-based African catfish Clarias gariepinus culture system. Catfish survival, growth, whole-body proximate composition, body indices, liver histopathology and glycogen content were measured. Each treatment was triplicated in glass aquaria with each replicate containing 50 fish (500 fish/m3) with an initial weight ± SD of 5.06 ± 0.05 g. Glycerol significantly increased total biofloc production, and both the sucrose and glycerol treatments generally had lower nitrogenous levels, compared to the control. These levels spiked at week 2 in the rice bran treatment, leading to significantly lower survival compared to all other treatments. At both weeks 3 and 6, liver histopathology of fish in the rice bran treatment revealed substantial vacuolation and less glycogen while the highest was in fish from the glycerol treatment. Fish growth was unaffected among the treatments, but survival was highest in the glycerol treatment. Rice bran appears unsuitable for C. gariepinus, likely due to being a slower-releasing carbon source. Instead, glycerol is recommended based on significantly higher biofloc production and subsequently improved water quality and survival of C. gariepinus during their nursery culture.  相似文献

This study aimed to elucidate the changes in Pacific white shrimp (Litopenaeus vannamei) myofibrillar protein as influenced by multiple freeze-thaw cycles as well as the stabilization effects of sucrose and trisodium citrate on shrimp myofibrils. Shrimp myofibrils in 0.1 M NaCl, 20 mM Tris-HCl (pH 7.5) were mixed individually with sucrose and citrate at concentrations of 0.05 M and were evaluated for Ca2+-ATPase activity, salt solubility, total and reactive sulfhydryl, and surface hydrophobicity during three freeze-thaw cycles. Sucrose and citrate had strong cryoprotective effects against freeze denaturation by retaining higher Ca2+-ATPase activity and salt-soluble myosin and actin, by slowing the reduction of reactive sulfhydryl (SH) and by exposing less hydrophobic groups at the surface of the protein compared with the no-additive sample. Results indicated that both cryoprotectants had suppressive effect against protein denaturation and helped stabilize white shrimp myofibrillar protein during the freeze-thaw process. This study suggests that sucrose and citrate stabilized the protein structure by retarding the unfolding of protein; thus, the native protein could be protected during frozen storage.  相似文献

Physicochemical attributes of Chesapeake Bay’s blue catfish and quality of muscle protein isolates (PIs) as influenced by storage condition and extraction methods were investigated. All fish were within the maximum consumable size limit. Those obtained during the autumn/winter and in Year 2 were larger with firmer fillets than their counterparts during the spring/summer and in Year 1. Protein was the predominant nutrient component. Protein extraction yield ranged from 35.3 ± 1.4% to 47.0 ± 5.1%, and β-sheet was the predominant secondary structure for all fish PIs. Salt-extracted PIs were whiter and had higher water holding capacity at neutral and alkali environments. The results of this study support the use of Chesapeake Bay blue catfish and muscle protein for potential value-added application.  相似文献

Sexually mature males (BW?=?1600?±?150 g and TL?=?235?±?30 mm) of northern pike (Esox lucius L.) were randomly selected from a pond to record changes in their sperm quality parameters (spermatozoa morphology, sperm volume, density, and motility parameters) during the spawning season. The morphological and motility parameters changed significantly during the reproductive season with following trends. Only, head width was not changed during the spawning season. The longest spermatozoa and its flagellar length were found at the middle of spawning period (TL?=?38.24?±?0.37 μm and 35.14?±?0.26 μm) and shortest at the beginning of spawning period (TL?=?34.81?±?0.29 μm and 32.53?±?0.18 μm). Other morphological characters were always the lowest at the beginning of spawning period. Sperm volume was changed from 0.33?±?0.3 ml in February, 0.43?±?0.2 ml in March to 0.24?±?0.1 ml in April, and density from 16.2?±?0.2?×?109 spermatozoa ml?1 in February, 19.4?±?0.2?×?109 spermatozoa ml?1 in March to 4.8?±?0.2?×?109 spermatozoa ml?1 in April. Same sperm velocity was observed in all spawning terms at 10 and 20 s after activation. Higher velocity was found at 30 and 40 s after activation in sperm collected at the middle and the end of spawning period. Significantly, higher percentage of motile sperm was observed at 20, 30, and 40 s after activation in sperm sampled at the end of spawning period. This study supports the hypothesis that longer spermatozoa swim faster.  相似文献

This study determined some antioxidative activities and functional properties of hydrolyzed proteins prepared from kilka (Clupeonella sp.) meat (KMH), kilka fishmeal (KFH), and stickwater (SWH). The highest degree of hydrolysis (DH) was 25.02% (p < 0.05) in SWH; however, the difference in DH between KMH and KFH was not significant. KFH had the highest protein content (85.69%) compared with other hydrolysis samples. The Fourier transform infrared spectra of KFH had the highest amide absorption picks (p < 0.05). Solubility at neutral pH was 84.09%, 86.00%, and 93.08% for KMH, KFH, and SWH, respectively. The highest fat absorption capacity was obtained with KFH (204 mg/50 mg). KFH also showed the best foaming properties with 25.66% foaming capacity and proper stability during 60 min (< 0.05). The highest and lowest whiteness values were obtained in KMH (72.33%) and SWH (36.54%), respectively (< 0.05). The highest 2,2-diphenyl-1-picrylhydrazyl (DPPH) (IC50 value of 1.99 mg/mL) and 2,2?-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) (IC50 value of 2.00 mg/mL) radical scavenging activities were measured at the highest concentrations (40 mg/mL) in KFH (< 0.05). KFH had the best functional and antioxidant properties, while SWH had a strong ability to inhibit lipid oxidation. Therefore, stickwater from fishmeal production factories is a potential resource for use in developing value-added products instead of being discarded.  相似文献

ABSTRACT

Tubifex (Tubifex tubifex) was cultured in captivity using three different wastes: rice mill sludge (RMS), dairy sludge (DS), and raw cattle dung (RCD). Three experiments were conducted: 10, 20, and 30 days. A total of 100 g of tubifex at 62.5 g m?2 was inoculated in 1.6 m2 fiberglass-reinforced plastic tanks. Comparing all production parameters, RMS > DS > RCD. Growth rate (g m?2 d?1) did not differ among durations. RMS- and DS-fed tubifex contained higher protein and fat than RCD-fed tubifex. Efficiency on production of g tubifex per kg of waste material was highest at 10 days, declining with time for all waste materials. This experiment suggests that RMS and DS are effective wastes for tubifex culture, with total production increasing with no reduction in growth rate through 30 days, but with efficiency declining after 10 days.  相似文献
