Mycobacteriosis due to infection of Mycobacterium marinum is a common disease in pond-cultured Chinese soft shell turtles, especially in those surviving beyond their first year. The infected turtles independently showed either heterophilic or histiocytic granulomas in various organs such as the spleen, liver, lungs, intestine, kidneys, stomach and pancreas. The heterophilic granuloma contained many acid-fast unbranching bacilli intracellularly in macrophages and extracellularly in the necrotic center. The histiocytic granuloma had only a few bacteria, mainly in the cytoplasm of Langhan's giant cells. The organisms were rarely observed in the advanced lesions of both types. Based on PCR assays for partial hsp65 gene of Mycobacterium spp., all of our strains were identified as M. marinum which can be divided into two groups. The strains of the first group induced heterophilic granulomas and had very high nucleotide sequence identities (99.8%-100%) to the reference strains of M. marinum (AF456471) and M. pseudoshottsii (AY550226). Those strains of the second group caused histiocytic granulomas and also showed very high identities (99.8%-100%) to the reference strains of M. marinum ATCC 927 (AF456470) and M. shottsii (AY550225). However, when we compared the partial sequence of the hsp65 gene from group one and two strains the identities between the two groups range from 98.8% to 99.3%, therefore we can not assert that these two belong to the same species.  相似文献

为了示踪研究拟态弧菌感染草鱼的动态过程,将增强型绿色荧光蛋白编码基因EGFP克隆至质粒pBAD24,并转化到拟态弧菌04-14菌株构建荧光标记重组菌.重组菌经阿拉伯糖诱导后,能高效表达EGFP蛋白;荧光显微镜观察和流式细胞仪检测均发现重组菌能够发出明显的绿色荧光信号,且传至30代后质粒稳定率仍为100%;生物学特性检测结果显示,与野生株相比,重组菌的形态、生长特性和细胞黏附性均未发生明显改变.用标记重组菌浸泡感染草鱼,定点采集鳃、肠道、肌肉、头肾、脾脏和肝脏,借助荧光信号检测4d内细菌在不同组织脏器中的动态分布.结果发现感染4h后即可在肠道和鳃中检测到绿色荧光信号,标记菌检出量分别为3.60×108和2.36×106 CFU/g,直至10 h,其含量无明显变化,12 h后含菌量逐渐下降,但持续存在直至鱼死亡.标记菌在肌肉、头肾、脾脏和肝脏中呈现相似的动力学,感染24 h后才检测到荧光信号,24~ 85 h时间段含菌量呈现先增加后下降的变化,48 h达到峰值,检出量分别为9.58×104(肌肉)、8.75×104(头肾)、1.50×104(脾脏)和4.50×104 CFU/g(肝脏),但均低于肠道中的检出量,结果表明肠道是拟态弧菌黏附定植与繁殖的主要靶器官.  相似文献

A disease outbreak occurred during the Spring of 2006 among rainbow trout, Oncorhynchus mykiss, kept in growout tanks. During the outbreak 35% of the rainbow trout died. Typical clinical signs were ulceration on the dorsal surface of the fish. Based on biochemical characteristics and sequence of 16S rRNA, the causative bacteria isolated from diseased fish were identified as Pseudomonas putida. Histologically, skin ulcer initially was characterized by sloughed off epithelia and epithelial necrosis. In the advanced form of the disease, almost all layers of the skin down to the epidermis were lost. Outside of Japan, this study reports the first P. putida infection in fish.  相似文献

Flavobacterium columnare is an important pathogen of freshwater fishes often causing high mortality. Seven strains of F. columnare have been isolated from gill necrosis, skin lesions and internal organs of Catla catla, Labeo rohita, Cirrhinus mrigala, Carassius auratus, Anabas testudineus and Clarias batrachus and characterized by biochemical reactions and serological tests viz indirect enzyme‐linked immunosorbent assay (ELISA), Dot‐ELISA, agglutination test. All the strains showed binding to Congo red dye as well as hemolysis. All the seven strains were susceptible to amikacin, gentamycin and ofloxacin. In vivo LD50 dose of virulent strain MS2 was found to be 6 × 104 CFU/mL after experimental infection to L. rohita. Serologically all the seven strains showed a positive result to agglutination, Dot‐ELISA and indirect ELISA. The agglutination titer was found to be in the range of 256‐131,072. The lysozyme activity of hyperimmune sera was found to be 39.37 ± 0.80 units/mL. This is the first extensive study that report about the various strains of F. columnare from freshwater fishes of Eastern India and their differentiation by biochemical and serological methods.  相似文献

本研究旨在观察大鲵在大鲵蛙病毒(Chinese giant salamander ranavirus,CGSRV)感染过程中组织的动态病理损伤,同时定量检测CGSRV在组织中的动态分布。对大鲵腹腔注射1.0×106.5 TCID50/m L的CGSRV进行人工感染,在感染的0d,3d,5d,7d,9d,13d,16d随机采集3尾,取肝、肾、脾、肺、肠、皮肤、肌肉、脑、心脏和胃等组织,石蜡切片和HE染色对CGSRV感染大鲵的病理损伤过程进行观察,采用SYBR Green q PCR技术对病毒在组织中的动态分布进行定量研究。组织病理结果表明,CGSRV感染导致大鲵多组织器官损伤,其中肝、脾、肾和皮肤肌肉病变严重,为损伤靶器官,且在一些损伤的细胞内见嗜碱性或嗜酸性包涵体。感染后3d肝细胞与肾小管上皮细胞变性,肾间以及肝小叶中央静脉周围淋巴细胞和嗜酸性粒细胞浸润;感染后5~7d实质性器官变性、坏死,炎症加重,胃肠道呈卡他性炎。感染后9d表皮细胞坏死、脱落,肌纤维变性、坏死。感染后13~16d肝出现广泛变性、坏死与炎症,脾淋巴细胞数量显著减少,肾小球渗出性-坏死性炎,皮肤肌肉呈出血性坏死性炎和实质性心肌炎。SYBR Green qPCR结果显示,整个感染进程中各组织CGSRV含量呈上升趋势,不同组织中病毒量为2.36×103~1.84×109 copy/mg组织,其中肺、肠、肝、脾、肾和皮肤肌肉含量高,表明CGSRV具有广泛的组织分布特征,但肝、脾、肾、皮肤肌肉为其复制和损伤的靶器官,且病毒分布量与病理损伤程度呈正相关。  相似文献

Mycobacteriosis in fish is a chronic progressive ubiquitous disease caused by Mycobacterium marinum, M. gordonae and M. fortuitum in most cases. The aim of this study was to describe the morphology and distribution of lesions in 322 freshwater ornamental fish across 36 species. Granulomatous inflammation was diagnosed by gross examination and histopathology testing in 188 fish (58.4%); acid‐fast rods (AFR) were determined in only 96 (51.1%) fish from 19 species after Ziehl–Neelsen staining. The most often affected organs with AFR were the kidney (81.2%), digestive tract (54.1%), liver (48.2%), spleen (45.9%) and skin (21.2%); sporadically, AFR were found in the branchiae (9.4%) and gonads (4.7%). In 14 randomly selected fish originating from four different fish tanks, the distribution of mycobacterial infection was studied by culture examination of the skin, gills, muscle tissue, digestive tract, liver, spleen and kidney. In 12 fish, the species M. marinum, M. gordonae, M. fortuitum, M. triviale, and M. avium subsp. hominissuis (serotypes 6 and 8 and genotype IS901? and IS1245+) were detected; mixed infection caused by different mycobacterial species was documented in five of them.  相似文献

During the present study, we aimed at providing a first look at the elemental composition of the early stages of cephalopods as an approach to their elemental requirements in culture. Essential and non-essential elemental profiles of the European cuttlefish Sepia officinalis, the European squid Loligo vulgaris and the common octopus Octopus vulgaris laboratory hatchlings and wild juveniles were analysed. In addition, for O. vulgaris we determined elemental profiles of mature ovary, eggs in different stages of development and followed possible effects of four dietary treatments during paralarval rearing, also analyzing elemental content of the live preys Artemia nauplii and Maja brachydactyla hatchling zoeae. Content was determined for essential (As, Ca, Cr, Co, Cu, Fe, K, Mg, Mn, Na, Ni, P, Rb, S, Sr, Zn) and non-essential (Ag, Al, Ba, Cd, Hg, Pb) elements. The content in non-essential elements found in hatchlings and juveniles of the three species analyzed here seems to be far lower in comparison with subadult and adult stages of coastal cephalopods. In the octopus eggs, the non-essential element concentrations remained globally low compared to hatchlings and juveniles indicating the absorption of these elements along the ontogenetic development. The elemental composition of the octopus ovary and of the eggs, hatchlings and juveniles of the three cephalopod species analyzed here showed a high content in S. As expected, the calcified internal shell of the cuttlefish, rich in Ca and Sr, originates the main difference between species. It is remarkable the richness in Cu of hatchling octopus, that may indicate a particular nutritional requirement for this element during the planktonic life. The reared octopus paralarvae feed on Artemia nauplii, a prey with relatively low Cu content, showed nearly half Cu content that the “natural” profile of octopus hatchlings or wild juveniles. This suggests a dietary effect and/or an indication of the poor physiological stage of the Artemia-fed paralarvae. At the present, the percentage of essential element absorption by food or seawater is unknown for cephalopods and should be determined in the future to understand their feeding requirements in culture.  相似文献

以1.0×106CFU·m L~(-1)拟态弧菌(Vibrio mimicus)浸浴感染黄颡鱼(Pelteobagrus fulvidraco),于第0、第4、第8、第16、第24、第36、第48、第60、第72小时剖杀取样,研究病理损伤及病原菌的体内动态分布。结果显示,皮肤肌肉、鳃、肠道出现病变最早,其中皮肤肌肉损伤最严重。第16小时表皮变性、坏死,鳃上皮肿胀,肠绒毛水肿;第24~第48小时表皮坏死、脱落,真皮严重出血,肌纤维变性、坏死,鳃出血,上皮局部坏死,肠上皮变性与灶性坏死;第48~第72小时皮肤肌肉坏死更严重,形成溃疡灶,鳃灶性坏死。肾、肝、脾与心36 h后相继出现不同程度的淤血、出血,变性和灶性坏死。q PCR检测发现,第4小时即在鳃、肠、皮肤肌肉检出病菌,细菌含量分别为1.3×102CFU·mg~(-1)、2.6×102CFU·mg~(-1)、4.7×102CFU·mg~(-1);鳃与皮肤肌肉中菌量随时间延长而增加,第72小时皮肤肌肉中菌量达到4.5×107CFU·mg~(-1);第24小时后相继在肾、肝、脾、心检出病菌,菌量介于1.9×10~4.7×102CFU·mg~(-1)之间。结果表明皮肤、鳃和肠道是病菌的入侵位点,并在体内多组织、器官分布。  相似文献

Betanodaviruses are the causative agents of the disease known as viral nervous necrosis (VNN) or viral encephalopathy and retinopathy (VER) in a variety of marine and freshwater fish species. The aim of this study was to demonstrate experimental infection of an isolate of betanodavirus (RGNNV genotype) in freshwater fish, Gambusia affinis, for elucidation of transmission mechanism and potential use as a laboratory model. Morbidity and mortality rate was significantly higher by injection route of infection as compared to immersion by bath and resembled the natural infection of juvenile marine fish. The fish in disease affected group showed severe neurological disorders accompanied by extensive vacuolar degeneration and mild to moderate neuronal necrosis of the brain in comparison to control. Amplification of ~?427 bp product in the variable region of the coat protein gene of betanodavirus was achieved by RT-PCR with 100% sequence homology to RGNNV genotype.  相似文献

采用RACE方法,克隆了背角无齿蚌抗菌肽theromacin基因的cDNA全序列。结果显示,该cDNA序列全长为870 bp,5'端非翻译区104 bp,3'端非翻译区460 bp,开放阅读框长为306 bp,共编码101个氨基酸,相对分子量为11 166.9 u。同源性分析显示,该基因编码的蛋白与三角帆蚌theromacin基因氨基酸序列的相似度最高,为73%;与贻贝中发现的抗菌肽defensins、mytilins、myticins和mytimycins等4种类型相似度较低,属于Macin家族(登录号:KJ598604)。实时荧光定量PCR分析结果显示,该基因在血液、肝脏、外套膜、鳃、斧足和肠等组织中均有表达,在外套膜中的表达量最高,在其他组织中的表达量较低;经嗜水气单胞菌诱导后,各个组织的表达量出现明显上调的趋势且与对照组显著差异,推测theromacin基因可能在背角无齿蚌的免疫反应中起重要作用。  相似文献
