Arginine vasotocin is a hormone produced in the hypothalamus of teleost fish that has been shown to regulate gonad development and sexual behavior. To study the role of arginine vasotocin in the gonadal cycle of the hermaphrodite gilthead seabream, Sparus aurata, we cloned the seabream arginine vasotocin (avt) complementary DNA (cDNA). We investigated the expression of brain avt throughout the gonad cycle using real-time quantitative PCR and compared its expression levels to the expression levels of two key gonadal steroidogenic enzymes, cyp19a1a and cyp11b2. In July, when the process of sex reversal is thought to begin, avt expression was elevated over the previous 2 months. Avt in the brain remained at or above the level of July until November then peaked again in December. There was no difference between males and females in the expression levels of brain avt throughout the year. However, only in ambisexual fish was the expression of the cyp19a1a gonadal aromatase correlated to the expression of avt in the brain. Cyp11b2 did not show any correlation to brain avt expression. We also found that females had more intense body coloration than males and that this intensity peaked prior to spawning. Avt expression and female coloration were positively correlated. The fact that brain avt expression was lowest during gonad quiescence, together with the observation of a correlation between brain avt with gonadal cyp19a1a and body coloration during that time suggests that avt may play a role during the process of sex reversal and spawning of the gilthead seabream.  相似文献

Long-chain unsaturated n-3 fatty acids are of remarkable significance in human nutrition. They have antiatherosclerotic efficacy and other beneficial health effects too. Aquaculture fish, e.g. cyprinids, such as silver carp (Hypophthalmichthys molitrix), bighead carp (Aristichthys nobilis), common carp (Cyprinus carpio), tench (Tinca tinca), and salmonids like Atlantic salmon (Salmo salar) and rainbow trout (Oncorhynchus mykiss) as well as European catfish (Silurus glanis), Baltic whitefish (Coregonus maraena), European seabass (Dicentrarchus labrax), gilthead seabream (Sparus aurata), and Nile tilapia (Oreochromis niloticus), are rich in these fatty acids. When fed on suitable diets, the fatty acid composition of cultured fish can be influenced advantageously. Several clinical tests proved the effectiveness of the consumption of farmed fish in promoting human health. Thus, aquaculture fish can be recommended as wholesome food.  相似文献

Partial substitutions of fish meal by 5, 15, or 25 % of Gracilaria cornea or Ulva rigida in experimental diets were evaluated to study their effects on biodiversity of intestinal microbiota composition in gilthead seabream (Sparus aurata) juveniles. The diets were offered to duplicate groups of 15 juvenile fish (14.0 ± 0.5 g) for 70 days, and at the end of the experiment the intestinal microbiota from four specimens of each treatment was analysed by denaturing gel gradient electrophoresis. Results showed that the substitution of fish meal by algae meal induced important modifications in the intestinal microbiota community, as a big reduction of the biodiversity when the highest percentage (25 %) of U. rigida was included. On the contrary, an increase on the number of species was detected when a 15 % of algae was included. Various Lactobacillus delbrueckii subspecies were selectively stimulated when G. cornea was included in the feed, and other bacterial species, such as those included in the Vibrio genus, were reduced.  相似文献

In euryhaline teleosts, the hypothalamus-pituitary-thyroid and hypothalamus-pituitary-interrenal axes (HPT and HPI, respectively) are regulated in response to environmental stimuli such as salinity changes. However, the molecular players participating in this physiological process in the gilthead seabream (Sparus aurata), a species of high value for aquaculture, are still not identified and/or fully characterized in terms of gene expression regulation. In this sense, this study identifies and isolates the thyrotropin-releasing hormone (trh) mRNA sequence from S. aurata, encoding prepro-Trh, the putative factor initiating the HPT cascade. In addition, the regulation of trh expression and of key brain genes in the HPI axis, i.e., corticotrophin-releasing hormone (crh) and corticotrophin-releasing hormone-binding protein (crhbp), was studied when the osmoregulatory status of S. aurata was challenged by exposure to different salinities. The deduced amino acid structure of trh showed 65–81% identity with its teleostean orthologs. Analysis of the tissue distribution of gene expression showed that trh mRNA is, though ubiquitously expressed, mainly found in brain. Subsequently, regulation of gene expression of trh, crh, and crhbp was characterized in fish acclimated to 5-, 15-, 40-, and 55-ppt salinities. In this regard, the brain gene expression pattern of trh mRNA was similar to that found for the crh gene, showing an upregulation of gene expression in seabream acclimated to the highest salinity tested. Conversely, crhbp did not change in any of the groups tested. Our results suggest that Trh and Crh play an important role in the acclimation of S. aurata to hypersaline environments.  相似文献

This study compared the efficacy of different dietary algae on the growth and reproduction of the cyclopoid copepod Cyclopina kasignete, a potential live food species for fish larvae in aquaculture. The experimental diets for the copepod consisted of three monoalgal diets (Nannochloropsis oculata, Tisochrysis lutea and dry Melosira sp.) and two mixed algae diets (T. lutea?+?N. oculata, T. lutea?+?dry Melosira sp.). The experiment was carried out for 30 days, and the population growth, survival and reproductive performance (generation time, hatching rate, life spawning times, daily offspring production, eggs per sac, lifespan and sex ratio) were used to assess the responses of C. kasignete to different food types. Population growth, survival and reproductive capacities of C. kasignete were significantly affected by the mono and binary species of algal diets. The results showed that copepods exhibited superior growth, survival and productivity when fed on fresh T. lutea, dry Melosira sp. and a mixture of both species compared to other dietary treatments. Copepods produced comparable growth, survival and productivity when fed on diatoms (dry Melosira sp.) as a single or in combination with other algae. This study indicates that cyclopoid copepod C. kasignete grow fast and have the potential to serve as a live food for aquaculture. The algae T. lutea, dry Melosira sp. and their combination are appropriate food to sustain the growth and reproduction of this copepods in mass culture as a potential live food in fish hatchery.  相似文献

The influence of acclimation of the euryhaline gilthead sea bream (Sparus aurata) larvae/post-larvae to brackish water on growth, energetic contents, and mRNA levels of selected hormones and growth-regulating hypothalamic neurohormones was assessed. Specimens from 49 days post-hatching were acclimated during 28 days to two different environmental salinities: 38 and 20 psu (as brackish water). Both groups were then transferred to 38 psu and acclimated for an additional week. Early juveniles were sampled after 28 days of acclimation to both salinities and one week after transfer to 38 psu. Pituitary adenylate cyclase-activating peptide (adcyap1; pacap), somatostatin-I (sst1), growth hormone (gh1), insulin-like growth factor-I (igf1), and prolactin (prl) mRNA expression were all studied by QPCR. Post-larvae acclimated to 20 psu showed better growth performance and body energetic content than post-larvae maintained at 38 psu. prl, adcyap1, and igf1 mRNA expression levels increased in 20-psu-acclimated post-larvae but decreased upon transfer to 38 psu. GH1 expression did not show significant changes under both experimental conditions. Our results suggested an enhanced general performance for post-larvae in brackish water, supported by the actions of adcyap1, igf1, and prl.  相似文献

In aquaculture, nutrition and supplemented diets have been shown to affect broodstock reproductive performance. In this study, we investigated the effects of dietary supplementation with Cymbopogon flexuosus essential oil (CFEO) microcapsules on reproductive-related parameters in silver catfish (Rhamdia quelen) male broodfish. Adult male broodstocks were separated into three groups according to the concentrations of supplemented CFEO (0.0?=?control; 1.0 or 3.0 mL per kg of diet). After 20 days under experimental conditions, the animals were euthanized and the gonads were harvested for gonadosomatic index, sperm analysis, oxidative stress, and histopathology; testosterone levels were measured in the plasma; gene expression of prl, smtl, pomca, and pomcb was assessed in the pituitary gland by real-time PCR. The results showed no alterations on reproductive parameters in R. quelen males treated with Cymbopogon flexuosus essential oil compared to the control-diet animals. In conclusion, CFEO microcapsules supplied for 20 days in the concentrations of 1.00 or 3.00 mL per kilogram of diet did not affect the reproduction criteria evaluated in this study in male silver catfish.  相似文献

Heat stress is one of the major environmental concerns in global warming regime and rising temperature has resulted in mass mortalities of animals including fishes. Therefore, strategies for high temperature stress tolerance and ameliorating the effects of heat stress are being looked for. In an earlier study, we reported that Nrf-2 (nuclear factor E2-related factor 2) mediated upregulation of antioxidative enzymes and heat shock proteins (Hsps) provide survivability to fish under heat stress. In this study, we have evaluated the ameliorative potential of dietary curcumin, a potential Nrf-2 inducer in heat stressed cyprinid Puntius sophore. Fishes were fed with diet supplemented with 0.5, 1.0, and 1.5% curcumin at the rate 2% of body weight daily in three separate groups (n = 40 in each group) for 60 days. Fishes fed with basal diet (without curcumin) served as the control (n = 40). Critical thermal maxima (CTmax) was determined for all the groups (n = 10, in duplicates) after the feeding trial. Significant increase in the CTmax was observed in the group fed with 1.5% curcumin- supplemented fishes whereas it remained similar in groups fed with 0.5%, and 1% curcumin-supplemented diet, as compared to control. To understand the molecular mechanism of elevated thermotolerance in the 1.5% curcumin supplemented group, fishes were given a sub-lethal heat shock treatment (36 °C) for 6 h and expression analysis of nrf-2, keap-1, sod, catalase, gpx, and hsp27, hsp60, hsp70, hsp90, and hsp110 was carried out using RT-PCR. In the gill, expression of nrf-2, sod, catalase, gpx, and hsp60, hsp70, hsp90, and hsp110 was found to be elevated in the 1.5% curcumin-fed heat-shocked group compared to control and the basal diet-fed, heat-shocked fishes. Similarly, in the liver, upregulation in expression of nrf-2, sod, catalase, and hsp70 and hsp110 was observed in 1.5% curcumin supplemented and heat shocked group. Thus, this study showed that supplementation of curcumin augments tolerance to high temperature stress in P. sophore that could be attributed to nrf-2-induced upregulation of antioxidative enzymes sod, catalase, gpx, and the hsps.  相似文献

The effect of the dietary incorporation of drumstick, Moringa oleifera, leaf meal (MOL; 0, 5, 10 and 15%) on the growth, feed utilization, some skin mucus and systemic immune parameters and intestinal immune-related gene expression in gilthead seabream (Sparus aurata) specimens. The experiment lasted 4 weeks. The results revealed that MOL can be incorporated in S. aurata diet up to 10% with no significant negative effect on growth and feed utilization. However, there was a significant decrease with MOL at a level of 15% after 2 weeks of feeding. The systemic immune status of fish fed with the different levels of MOL showed an improvement in head kidney leucocyte phagocytosis, respiratory burst and peroxidase activities. Also, serum humoral components, including protease, ACH50 and lysozyme activities and IgM level, increased with MOL inclusion especially at the 5% level. MOL at 5% improved skin-mucosal immunity such as protease, antiprotease, peroxidase and lysozyme activities. Moreover, the feeding of MOL revealed an upregulation of the intestinal mucosal immunity genes (lyso and c3), tight junction proteins (occludin and zo-1) and anti-inflammatory cytokines (tgf-β) with a downregulation of pro-inflammatory cytokine (tnf-α). Therefore, it is recommended to incorporate MOL in S. aurata diets at a level of 5% for the best immune status or 10% for the high growth performance and acceptable immune surveillance.

The diets and feeding guild structure of a freshwater reed belt fish assemblage (30 species) were examined in Lake Kitaura, part of the Lake Kasumigaura system, eastern Japan, from June to September in 2009 and 2010. Ontogenetic dietary shifts were recognized in 14 species, including several species (e.g., Cyprinus carpio, Tridentiger brevispinis and Hyporhamphus intermedius) targeted by local fisheries. Juveniles of these species generally fed on zooplankton, later switching to other prey items (e.g., benthic chironomid larvae, gammaridean amphipods, shrimps, juvenile fishes, diatoms and decomposing reeds) with growth. A cluster analysis based on dietary overlaps showed that the reed belt fish assemblage comprised five feeding guilds (plant, zooplankton, benthic invertebrate, terrestrial insect and fish feeders). Of these, zooplankton feeders were the most abundantly represented in terms of species numbers, suggesting that the main food items for the fish assemblage were zooplankton (e.g., cladocerans, and calanoid and cyclopoid copepods). Fish feeders included five species, of which one was an alien species introduced from another region in Japan (Opsariichthys uncirostris) and three were from foreign countries (Micropterus salmoides, Lepomis macrochirus macrochirus and Ictalurus punctatus).  相似文献

Studies considering broodstock conditioning of Ruditapes decussatus mostly used flagellates and diatoms for feeding, mainly Isochrysis galbana and Chaetoceros calcitrans, respectively. The present study evaluated the effect of different microalgal diets on gonadic maturation of R. decussatus including three microalgae species (C. calcitrans, Tetraselmis suecica, and Nannochloropsis oculata) which were used to prepare six diets: three monospecific and three bispecific mixed diets at a proportion of 1:1. Clams were daily fed at a ratio of 1% dry weight of algae/live weight of clam and unfed treatment was used as control. Results showed that clams fed C. calcitrans alone or mixed with T. suecica attained earlier full maturity with the highest response to spawning induction (after 34 conditioning days) and largest proportion in the partial spawning phase for both, followed by T. suecica alone after 48 days. In contrast, N. oculata alone or mixed diets demonstrated poor performance for gonadal maturation. Unfed treatment did not show any sign of maturation. The results indicated that T. suecica has high conditioning potential like C. calcitrans and their mixture gave the highest benefit. Therefore, T. suecica can be recommended for R. decussatus broodstock conditioning.  相似文献

Larval rearing is affected by a wide range of microorganisms that thrive in larviculture systems. Some seaweed species have metabolites capable of reducing the bacterial load. However, no studies have yet tested whether including seaweed metabolites on larval rearing systems has any effects on the larvae development. This work assessed the development of Sparus aurata larvae fed preys treated with an Asparagopsis armata product. Live prey, Brachionus spp. and Artemia sp., were immersed in a solution containing 0.5% of a commercial extract of A. armata (Ysaline 100, YSA) for 30 min, before being fed to seabream larvae (n = 4 each). In the control, the live feed was immersed in clear water. Larval parameters such as growth, survival, digestive capacity (structural-histology and functional-enzymatic activity), stress level (cortisol content), non-specific immune response (lysozyme activity), anti-bacterial activity (disc-diffusion assay) and microbiota quantification (fish larvae gut and rearing water) were monitored. Fish larvae digestive capacity, stress level and non-specific immune response were not affected by the use of YSA. The number of Vibrionaceae was significantly reduced both in water and larval gut when using YSA. Growth was enhanced for YSA treatment, but higher mortality was also observed, especially until 10 days after hatching (DAH). The mortality peak observed at 8 DAH for both treatments, but higher for YSA, indicates larval higher susceptibility at this development stage, suggesting that lower concentrations of YSA should be used until 10 DAH. The application of YSA after 10 DAH onwards promotes a safer rearing environment.  相似文献

Levels of the oxidative stress‐related minerals selenium (Se), zinc (Zn) and manganese (Mn) that should be supplied in microdiets for marine fish larvae depend on the availability of the molecular form of these minerals. The objective of this study was to determine how effectively Se, Zn and Mn in organic, inorganic and nanoparticle forms promote larval performance and bone development. Microdiets supplemented with Se, Zn and Mn were fed for 24 days to 20 dah seabream larvae. Microdiets without Se, Zn and Mn supplementation were associated with poor growth, low bone mineralization and a high incidence of anomalies in the branchial arches. Including Zn, Mn and Se in an amino acid chelate organic form promoted maximum larval growth, increased body lipid reserves, enhanced early mineralization and prevented branchial arches anomalies. In contrast, feeding with inorganic forms of these minerals was less effective than organic minerals in improving larval weight or bone mineralization in comparison to the non‐supplemented diet. Moreover, the larvae were less resistant to stress, and fish showed higher bone anomalies in the pre‐hemal region. Adding Zn, Mn and Se in the form of nanometals did not enhance growth, but improved stress resistance and bone mineralization. The study showed the need to supplement seabream with early weaning diets based on squid meal and krill oil with one or more of the antioxidant minerals, to promote larval growth, bone mineralization and prevention of skeleton anomalies, organic minerals being more effective than inorganic forms and nanometals in promoting mineralization and stress resistance.  相似文献

The effects of galactooligosaccharide (GOS) and a combination of yeast and β-glucan (YβG) supplementation of dietary soybean meal (SBM) on the growth and digestive performance of striped catfish Pangasianodon hypophthalmus were evaluated. Four isonitrogenous (30% protein) and isocaloric (19 MJ/kg diet) diets were formulated to contain 100% fish meal (FM) protein, 55% FM protein/45% SBM protein, FM-SBM supplemented with 1% GOS, and a combination of 1% yeast and 0.1% β-glucan, respectively. Each diet was fed for 12 weeks to three groups of 30 striped catfish fingerlings (average weight 16.45?±?0.07 g) maintained in circular fiberglass tanks (600 l). Growth, feed utilization, and muscle protein composition of fish improved significantly after supplementation with either GOS or YβG compared to the unsupplemented SBM diet, but were similar to those of fish fed the FM diet. Nutrient digestibility, digestive enzyme activities, villi and microvilli length were significantly increased in fish fed the supplemented SBM diets. The gut microbiota ranking profile showed that supplementing the SBM diet with YβG and GOS gave a ranking of Verrucomicrobia, Spirochaetes, Bacteriodetes, and Actinobacteria phyla similar to that of the FM diet. Thus, diet containing 45% protein from soybean supplemented with either GOS or YβG can be recommended to improve the growth and digestive performance of striped catfish.  相似文献

To clarify the recruitment process of sand lance Ammodytes sp., we investigated larval condition factor, relative gut fullness (%GF), prey abundance and oceanographic structure in Mutsu Bay, Japan, during 1999–2001. Ammodytes sp. larvae, which were collected by horizontal hauls of Motoda nets and a ring net at depths of 1, 10, 20, 30 and 40 m, were mainly distributed at 10–30 m. Larvae at the first feeding time until 12 mm in body length (BL) fed predominantly on copepod nauplii, whereas large larvae with BL of 12.1–14.0 mm fed on a mixture of copepod nauplii, copepodites and appendicularians from late February to April. A path analysis showed that difference in water density between 35- and 5-m depths negatively affected naupliar abundance at 10–30-m depth (standardised path coefficient β = ?0.71, p = 0.005 for 3.3–8.0-mm BL larvae and β = ?0.78, p < 0.001 for 8.1–12.0-mm BL larvae). Naupliar abundance positively affected the %GF of Ammodytes sp. larvae (β = 0.75, p < 0.001 for 3.3–8.0-mm BL larvae and β = 0.66, p < 0.001 for 8.1–12.0-mm BL larvae), whereas it was negatively affected by water temperature (β = ?0.45, p = 0.008 for 3.3–8.0-mm BL larvae and β = ?0.56, p = 0.002 for 8.1–12.0-mm BL larvae), and the temperature effect was weak compared with that of naupliar abundance. In turn, %GF positively affected larval somatic weight (β = 0.91, p < 0.001 for 6.0-mm BL larvae and β = 0.70, p = 0.005 for 10.0-mm BL larvae). The recruitment failure in 1999 was likely caused by a reduced condition factor, which resulted from low naupliar abundance. In contrast, the abundance of nauplii and Oithona similis copepodites was high in 2000 and 2001. It is possible that the higher recruitment success in 2001 was because of the higher water temperatures in Mutsu Bay, sustaining faster growth of the larvae than in 2000 under the high-prey abundance conditions.  相似文献

The present study was carried out to evaluate the effect of supplementation of Garlen®, Diamond V XPC®, and Bactozyme® and their combination in the diets of European sea bass, Dicentrarchus labrax larvae development in hatcheries on feed intake, growth performance, feed efficiency, intestinal bacterial, survival rate, and economics analysis. Dicentrarchus labrax juveniles (1.4 ± 0.5 mg) were distributed into eight experimental groups with a density of 2.5 larvae per liter. Eight iso-nitrogenous (55% crude protein) and iso-caloric (19.2 ± 0.2 MJ/kg, DM) weaning diets were formulated to contain the control diet (no feed additives), individually or mixed alternatively with three feed additives (Garlen®; Diamond V XPC®, and Bactozyme®). Each experimental diet was allocated to three tanks of fish and fed for 12 weeks. Growth and survival rate (S %) were improved for larvae fed the diets supplied with either (Diamond V XPC® + Bactozyme®) or (Garlen® + Diamond V XPC® + Bactozyme®), respectively compared to the control larvae group. The best FCR value was recorded for larvae fed a diet supply with (Garlen® + Diamond V XPC® + Bactozyme®), while the control larvae group recorded the worst FCR. The optimum significant (P ≤ 0.05) nutrient utilization values, larvae body crude protein content, activity test (ATV%), and profit index (PI) values were observed for larvae fed a diet containing Diamond V XPC® + Bactozyme® or Garlen® + Diamond V XPC + Bactozyme® compared to other treatments. The opposite trend was observed for total bacterial (TBC) and Vibrio sp. counts. No significant (P ≥ 0.01) difference was recorded in Aeromonas sp. count values in all experimental treatments. The results from this study show that (Garlen®; Diamond V XPC®, and Bactozyme®) individually or mixed alternately as growth promoters and immune stimulants in early weaning larval diets of European sea bass under hatchery conditions led to improve growth performance, feed utilization, survival, lowest intestinal bacterial load, and highest profit index (LE).  相似文献

The characterization of cholinesterase activity in brain and muscle of gilthead seabream was carried out using four specific substrates and three selective inhibitors. In addition, K m and V max were calculated from the Michaelis–Menten equation for ASCh and BSCh substrates. Finally, the in vitro sensitivity of brain and muscle cholinesterases to three organophosphates (OPs) was also investigated by estimating inhibition kinetics. The results indicate that AChE is the enzyme present in the brain, whereas in muscle, a typical AChE form is present along with an atypical form of BChE. Very low ChE activity was found in plasma with all substrates used. The inhibitory potency of the studied OPs on brain and muscle AChEs based on bimolecular inhibition constants (k i ) was: omethoate < dichlorvos < azinphosmethyl-oxon. Furthermore, muscle BChE was found to be several orders of magnitude (from 2 to 4) more sensitive than brain and muscle AChE inhibition by dichlorvos and omethoate.  相似文献

Rhodomyrtus tomentosa is a medicinal plant that shows biological effects including immunomodulatory activity on human and other mammals but not in fish. In this study, we evaluated the in vitro immunomodulatory effects of R. tomentosa leaf extract and its active compound, rhodomyrtone, on the immune responses, using rainbow trout (Oncorhynchus mykiss) head kidney (HK) macrophages as a model. The tested immune functions included the expression of genes involved in innate immune and inflammatory responses and the production of reactive oxygen species (ROS). Gene expression was evaluated after exposure to 10 μg mL?1 of R. tomentosa and 1 μg mL?1 of rhodomyrtone for 4 and 24 h. R. tomentosa and rhodomyrtone induced changes in the expression of pro-inflammatory cytokines (il1β, il8, and tnfα), anti-inflammatory cytokines (il10 and tgfβ), inducible enzymes (inos, cox2, and arginase), and an antioxidant enzyme (gpx1). Co-exposure of R. tomentosa with LPS resulted in a prominent reduction in the expression of genes related to an inflammatory process (il1β, il8, tnfα, inos, saa, hepcidin, and gpx1), suggesting anti-inflammatory effects. Similarly, co-exposure of rhodomyrtone with LPS led to a downregulation of inflammation-related genes (il1β, inos, saa, and hepcidin). In addition, exposure to both natural plant products caused a reduction in cellular ROS levels by HK macrophages. The present results indicate that R. tomentosa and rhodomyrtone exerted immunostimulatory and anti-inflammatory effects on fish macrophages, thus opening up the possibility of using these natural products to further develop immunostimulants for health management in aquaculture.  相似文献
