Postmortem changes in blue shrimp (Litopenaeus stylirostris) muscle were studied on the basis of biochemical, chemical, physical, and microbiological changes during an 18 day storage period at 0°C. Adenosine 5′-triphosphate (ATP) content, breakdown products, K-value, pH, trimethylamine nitrogen (TMA-N), total volatile basic nitrogen (TVB-N), water holding capacity (WHC), color, and texture (shear force) changes were examined. Also, total mesophilic and psychrophilic bacterial counts were measured. K-value increased linearly (r2 = 0.98) from an initial value of 1.37 ± 0.59 to 59.42 ± 6.05% at Day 18. Spoilage indicators TVB-N and TMA-N increased from 29.56 ± 1.33 and 0.69 ± 0.25 to 39.04 and 2.04 ± 0.59 mg of N/100 g at Day 18, respectively; meanwhile, the total viable counts of mesophilic and psychrophilic bacteria increased from 3.48 ± 0.44 and 2.61 ± 0.29 log CFU/g to 6.27 ± 0.21 and 7.14 ± 0.39 log CFU/g, respectively, which indicated spoilage at the end of the storage period. The pH, texture, WHC, and color were affected (p < 0.05) during the storage period. Overall, results indicate that blue shrimp muscle quality was maintained for 12 days of storage in ice. 相似文献

Sutchi catfish (Pangasianodon hypophthalmus) is a highly preferred farmed species that is produced in huge quantities. Meat from ice-stored (4 ± 2°C) whole Sutchi catfish was evaluated for formation of biogenic amines, such as putrescine, cadaverine, histamine (HIM), agmatine, tyramine, spermine, and spermidine, and compared with biochemical, microbial, and sensory attributes during 22 days. Analysis of content of biogenic amines in the meat was carried out by liquid chromatography with tandem mass spectrometry using a without derivatization method. Three amines, namely, tyramine, spermidine, and spermine, were only present on 0th day of storage. Presence of cadaverine was noticed from 9th day onwards, where the aerobic plate count (APC) reached 4.85 log CFU/g. Putrescine was detected on the 22nd day of storage, where the APC crossed 7 log CFU/g. HIM was detected in lower quantities from 3rd day onwards. A shelf life of 15 days was determined based on sensory and microbiological evaluation. Although the samples were biochemically acceptable throughout the storage period, APC exceeded the limit on day 19, and the gradual increase of H2S-producing bacteria, Brochothrix thermosphacta, Aeromonas, and Enterobacteriaceae, was observed during storage. 相似文献

Fertilizability of cryopreserved and cadaveric fish spermatozoa was attempted in the freshwater catfish Pangasius sutchi. Cryopreservation of spermatozoa was done with three cryoprotectants for short time storage (30 days). Whereas the spermatozoa obtained from the cadaveric fish were stored at ?20°C (30 days) without any cryoprotectants. Cryoprotectant toxicity assay showed maximum motility of 88.53 ± 2.01% and viability of spermatozoa (96.19 ± 4.92%) with 15% of Dimethyl acetamide (DMA) at 15 min equilibration time. Whereas Dimethyl sulfoxide (DMSO) (15%) registered moderate level of motility and viability 79.23 ± 2.02% and 80.89 ± 2.1%, respectively. However, the methanol (MeOH) (20%) resulted in low percentage of motility (58.6 ± 0.9%) and viability (68.6 ± 0.9%). Scanning electron micrographs further showed no significant deformity on the surface topography of spermatozoa of cadaveric fish as well as cryopreserved with DMA (15%). The results indicated that 15% of DMA with hanks balanced salt solution (HBSS) extender at a dilution ratio of 1:10 at ?80°C proved to be suitable for cryopreservation of spermatozoa in P. sutchi. This may be due to the osmolality of HBSS similar to seminal plasma of P. sutchi. Further studies on motility, viability and fertility potential of spermatozoa revealed 73.62 ± 1.61%, 88.34 ± 1.05% and 54 ± 2.2%, respectively, with DMA (15%). On the other hand, cadaveric fish sperm registered 57.12 ± 2.32%, 63.45 ± 0.94% and 25.33 ± 1.53% of motility, viability and fertilizability respectively. Thus, this study augments the feasibility of using cryopreserved as well as cadaveric fish spermatozoa for the seedling production in the fresh water catfish P. sutchi. 相似文献

This study aimed to elaborate a convenience food – sous vide – and establish its shelf life by monitoring physicochemical, microbiological, and sensorial characteristics. Sous vide fillets were prepared from tambaqui (Colossoma macropomum) with two types of sauces: soy sauce (SS) and basil sauce (BS), at a pasteurization temperature of 65°C for 12.5 min, a binomial established according to a 22 central composite rotational design. The packed samples were kept under refrigeration in the range of 1 ± 1°C, and then they underwent analyses at 7, 14, 21, 28, 35, 42, 49, and 56 days of storage. The results of the microbiological analyses indicated that microorganisms remained below acceptable limits either in the raw material or in the sous vide over the storage period. The characteristics that sous vide SS and BS showed on the 42nd and 49th days of shelf life, respectively, indicate that these products, when appropriate processing and storage are used, can preserve their fresh quality. 相似文献

ABSTRACT Cuphea meal is a new, alternative feedstuff that has potential as a sustainable, economical replacement for wheat, rice, and corn ingredients in channel catfish, Ictalurus punctatus, diets. Channel catfish fingerlings were fed a control diet containing wheat or two experimental diets containing 7.5% cuphea meal, or 12.5% cuphea meal for eight weeks to determine if cuphea meal could replace wheat products in catfish diets. Mean (±SE) weight gains were 317.8 ± 28.8 g, 407.0 ± 36.9 g, and 372.8 ± 29.8 g for fish fed the control diet, the 7.5% cuphea meal diet, and the 15% cuphea meal diet, respectively, and there were no significant differences (P < 0.05) among treatments. Mean (±SE) whole-body protein of fish fed the cuphea diets (13.3 ± 0.66 and 14.5 ± 0.21%) was significantly (P < 0.05) higher than that of fish fed the wheat diet (12.7 ± 0.44%). Cuphea meal enhanced body composition of juvenile channel catfish without affecting growth or survival. Therefore, cuphea meal is a promising candidate for replacement of wheat bran at the levels tested. 相似文献

ABSTRACT Physicochemical attributes of Chesapeake Bay’s blue catfish and quality of muscle protein isolates (PIs) as influenced by storage condition and extraction methods were investigated. All fish were within the maximum consumable size limit. Those obtained during the autumn/winter and in Year 2 were larger with firmer fillets than their counterparts during the spring/summer and in Year 1. Protein was the predominant nutrient component. Protein extraction yield ranged from 35.3 ± 1.4% to 47.0 ± 5.1%, and β-sheet was the predominant secondary structure for all fish PIs. Salt-extracted PIs were whiter and had higher water holding capacity at neutral and alkali environments. The results of this study support the use of Chesapeake Bay blue catfish and muscle protein for potential value-added application. 相似文献

Abstract This study was conducted to determine if channel catfish could be cultured at an increased density in the Partitioned Aquaculture System (PAS) without significantly affecting performance. Channel catfish fingerlings (36.53±6.76 g; mean ±SD) were initially stocked into six 9.15 m3 sections at 3,461±317 fish per section, twice the designed carrying capacity, and fed twice daily to satiation. After 75 days, the density of three units was reduced by approximately 50% and all six units were fed for another 97 days. There was no significant difference (P > 0.05) in mean growth rate, feed conversion ratio, or production between fish grown at the low- and high-density treatments. Also, length variation (CV) and condition factors (K) were similar (P > 0.05) between fish grown at the low and high-density. Increasing density by twice the designed carrying capacity did not affect performance of channel catfish in the PAS. 相似文献

This study investigated the composition of milt of the South American silver catfish (Rhamdia quelen) or jundiá. The semen was taken from jundiá in different periods during the four seasons. The biochemical composition of
seminal fluid and the characteristics of sperm were analyzed. The semen quantity which can be extracted per fish in one day
was 0.95 ± 0.08 ml during spring (maximum) and 0.24 ± 0.03 ml during winter (minimum). Sperm density (spermatocrit) showed
higher values in the spring (75.1 ± 1.3%) decreasing slightly afterwards and reaching 63.0 ± 2.4% to 65.0 ± 2.2% in the fall
and winter. Immediately after water dilution, 90–100% of the spermatozoa presented vigorous straightforward motility that
remained for at least 20 s. The total duration of the motility was 47.9 ± 1.3 s in the spring and 38.6 ± 0.6 s in the other
seasons (P < 0.05). This pattern of motility is maintained for more than 2 h after storage of the milt at room temperature.
The pH from 5 to 10 of the water dilution does not influence the sperm motility. The mean seminal pH and osmolality values
were 8.7 ± 0.07 and 274.8 ± 11.2 (mOsm/kg), respectively. The ion concentration was: Na 153.7 ± 2.4, K 10.7 ± 2.4, Cl 139.4
± 2.1, Ca 4.2 ± 0.2, Mg 0.9 ± 0.05, P 0.9 ± 0.08 (mEq/l). The total protein was 0.6 ± 0.05 mg/dl and cholesterol concentration

ABSTRACTThe effects of different atmospheric conditions on the quality and the shelf life of pasteurized oyster meat in chilled storage were investigated. The alternatives tested for pasteurized oyster meat were normal air (Control), 75% CO2/25% N2 (MAP-1), 75% CO2/25% O2 (MAP-2), 75% CO2/5% O2/20% N2 (MAP-3), and vacuum packing (VP). The various observations of microbiological (total viable count (TVC), Enterobacteriaceae, Pseudomonas spp., lactic acid bacteria (LAB), and hydrogen sulfide–producing bacteria), physical (color coordinates L*, a*, and b*, and cutting strength), chemical (moisture, pH, total volatile basic nitrogen (TVB-N), peroxide value (PV), and thiobarbituric acid reactive substances (TBARS)) and sensory properties were performed every 3 days over a total of 27 days. During storage across all the treatments, L*, a*, cutting strength, pH, moisture content, and sensory scores decreased; whereas, b*, TVB-N, PV, TBARS, and microorganisms increased. However, among the various alternatives, MAP-1 had the best retention of oyster quality during storage. Based on sensory and microbiological qualities, the shelf lives in chilled storage were 9 days for control, 15 days for VP, 21 days for MAP-2 and MAP-3, and 24 days for MAP-1-treated cases. 相似文献

Silver catfish, Rhamdia quelen (Quoy & Gaimard, 1824), is an endemic species from Latin America that is raised in cultivation ponds, and consequently may be exposed to low oxygen levels. Therefore, the objective of this study was to verify the lethal concentration (CL50?96 h) of dissolved oxygen levels for silver catfish juveniles. In addition, the effects of different dissolved oxygen levels (1.96±0.08, 3.10±0.10, 4.14±0.09, 5.20±0.07 and 6.16±0.03 mg L?1) on growth and metabolic parameters (glycogen, glucose, protein, lactate levels and catalase activity) were also investigated. CL50?96 h was 0.52 mg L?1 (CI 0.42–0.61 mg L?1) or 6.7% oxygen saturation. After exposure of silver catfish to hypoxia for 30 days, there were no changes in biochemical parameters indicating the use of an anaerobic pathway by the fish. However, the dissolved oxygen levels influenced silver catfish juvenile behaviour, survival and growth, and under the experimental conditions 5.2 mg L?1 (or 65.6% oxygen saturation) is the minimum oxygen level recommended for the growth of this species. 相似文献

To find out whether or not the locomotor activity of the Japanese sea catfish, Plotosus lineatus, shows a circadian rhythmicity, the locomotor activity of this catfish was recorded in a laboratory. All of the catfish recorded
(n = 20) showed a nocturnal locomotor activity rhythm under a light–dark (LD) cycle (LD 12:12). The locomotor activity started
after the light was turned off, and declined during the period of darkness. In five of the six catfish we examined, locomotor
activity shifted gradually over the course of 3–4 days in order to synchronize to the LD cycle after it was advanced by 6 h.
Locomotor activity persisted within a constant darkness (DD) in all eight of the catfish. The average free-running period
of locomotor activity under DD lasted for 24.2 ± 0.4 h (mean ± SD). Thus, the present results demonstrate that the Japanese

This study evaluated the efficacy of an oral live attenuated Edwardsiella ictaluri vaccine against enteric septicemia of catfish in 20 full‐sib fingerling channel catfish families. The vaccine was delivered orally by feeding fish a diet coated with an attenuated E. ictaluri isolate. Sixty‐nine days postvaccination, control and vaccinated fish were challenged with virulent E. ictaluri and mortality was examined for 21 d postchallenge. Vaccinated fish had significantly lower mortality than nonvaccinated fish following challenge (P < 0.001). Mortality of vaccinated fish was 1.7 ± 1.4% as opposed to 47.8 ± 28.7% in nonvaccinated fish. Relative percent survival ranged from 87.7 to 100% with an average of 95.2 ± 4.0% (±SE) among the 20 families of fish. There were significant differences in mortality among families in nonvaccinated fish (P < 0.01) while there were no differences among vaccinated families of fish. Results indicate that the live attenuated E. ictaluri vaccine is effective at reducing mortality in channel catfish exposed to virulent E. ictaluri. These data demonstrate that genetic differences among healthy families of channel catfish are not major considerations in developing an effective vaccination program utilizing the oral vaccination platform described in this study. 相似文献

ABSTRACTIn the present study, the effect of fish gelatin coating (FG; 4%, w/v) containing oregano (Origanum vulgare L.) essential oil (OEO; 1.2% w/v) on shelf-life extension of rainbow trout (Oncorhynchus mykiss) fillet stored under refrigerated storage (4 ± 1ºC) was investigated. The control and the treated fish samples were analyzed periodically for chemical (pH; total volatile basic nitrogen, TVB-N; peroxide value, PV; thiobarbituric acid, TBA) and microbiological (total viable count, TVC; psychrotrophic count) characteristics. The FG + OEO treatment produced significantly lower (p < 0.05) TVB-N values as compared to control and FG-coated samples after Day 6 and until the end of the storage period. The TBA values remained low and were found to fluctuate during storage. Results showed that TVC exceeded 6 log CFU g?1 on Day 8 of storage for control and FG-coated samples and Day 12 for samples coated with FG + OEO. The reduction of 1.05 and 0.83 log CFU g?1 of psychrotrophic counts was achieved by FG + OEO coating in trout fillet (p < 0.05) as compared with control and FG-coated samples, respectively. Therefore, the incorporation of OEO into FG coating may be a promising method of maintaining the storage quality of the rainbow trout fillet stored at refrigerated temperature. 相似文献

ABSTRACT Fingerling channel catfish Ictalurus punctatus were exposed to Ichthyophthirius multifiliis-infested fish until immature trophonts developed. The fish were transferred to individual static fiberglass tanks filled with 600 L of pond water (total alkalinity and total hardness was 220 mg/L and 101 mg/L, respectively) and were treated with 0, 1.1, 2.2, 3.3, or 4.4 mg/L copper sulfate (CuSO4 · 5H2O) every other day for four treatments to evaluate its effectiveness to control mortality associated with ichthyophthiriasis. Water temperature was maintained at 18 ± 1°C. Fish were observed for ten days post-treatment and mortalities were recorded. Results indicate that half of the recommended dose (1.1 mg/L CuSO4) is needed to effectively control an occurrence of ichthyophthiriasis under the conditions of this study. However, fish culturists should be aware that effective CuSO4 treatment of ichthyophthiriasis on channel catfish raised in ponds may be influenced by water chemistry characteristics and suspended materials such as pond sediments. 相似文献

The aim of the study was to quantify the shelf life and quality characteristics of puffer fish, Lagocephalus guentheri, fillets on chilled storage at 2 ± 1°C. The puffer fish muscle contains 16.14% protein, 80.76% moisture, 0.99% ash, and 0.17% fat. The mouse bioassay proved the absence of tetradotoxin in muscle, liver, skin, and gonads of the fish. The polyunsaturated fatty acid to saturated fatty acid (P/S) ratio of puffer fish was found to be 1.3, with n-3 to n-6 ratio of 3.27. The puffer fish had hypocholesterolemic/hypercholesterolemic ratio (H/H) value of 2.68, artherogenicity index of 0.43, and thrombogenicity index of 0.29. Beheaded and deskinned fishes were filleted and were studied for 10 days of chilled storage. The physical, chemical, sensory, and microbiological parameters showed significant (p < 0.05) changes during the entire storage period. While the hydrolytic rancidity was significantly correlated to springiness, gumminess, and chewiness of fillets, the peroxide value (PV) correlated with the color attributes and pH. The hardness of the samples decreased significantly and was evident in both sensory and instrumental analysis. With storage time, the yellowness of the fillets increased steadily. The results of this study can help in improving processing, preservation, and marketing aspects and enhance the utility of puffer fish. 相似文献

The effect of sublethal hypoxia exposure on stress and immune responses and susceptibility to Edwardsiella ictaluri infection in juvenile channel catfish, Ictalurus punctatus, was investigated. Fish were monitored for temporal changes in glucose and cortisol concentrations before, during, and after 2 h exposure to sublethal hypoxia (<2 mg/L dissolved oxygen [DO]) and when maintained under normoxic conditions (6.0 ± 0.3 mg/L DO). Both blood glucose and plasma cortisol increased significantly in response to hypoxic conditions. Fish exposed to hypoxic or normoxic conditions were challenged with a high dose (1.3 × 107 colony‐forming units [CFU]/mL) or a low dose (1.3 × 105 CFU/mL) of E. ictaluri or sterile culture broth by 30‐min immersion bath. Approximately 1% of fish in both the normoxic and the hypoxic groups died when challenged with the low dose of E. ictaluri. However, when challenged with the high dose of E. ictaluri, catfish exposed to hypoxic conditions had significantly higher cumulative mortality (36 ± 12.1%) than those maintained under normoxic conditions (12 ± 1.1%). Total hemolytic complement and bactericidal activities and antibody response were lower in hypoxia‐exposed channel catfish, indicating that increased susceptibility of channel catfish to E. ictaluri may be the result of the immunosuppressive effects of the stress response to hypoxia. 相似文献

Abstract Biological condition factors and proximate composition of hoki (Macruronus novaezelandaie) white muscle, dark muscle and liver vary considerably between individual fish and with season. Seasonal changes in the gonads and liver were correlated with concomitant changes in proximate composition of white muscle, dark muscle and liver. In white muscle there was an inverse relationship between protein and moisture content, while in dark muscle and the liver the inverse relationship was between lipid and moisture. These seasonal changes in proximate composition can be related to hoki's breeding cycle and have a direct effect on yields, quality and storage stability of products made from hoki. 相似文献

An 8‐week feeding trial was conducted in a recycling water system at 28 ± 1 °C to investigate compensatory growth and body composition in African catfish Clarias gariepinus (13.05 ± 0.05 g). A fishmeal‐based diet containing 350 g kg?1 protein and 17.5 kJ g?1 gross energy was fed to triplicate groups of 20 fish (per 30‐L tank). Fish were fed the diet either to satiation or feed restricted in six feeding schedules as satiation 56 days; restricted 28 days + satiation 28 days; restricted 14 days + satiation 14 days; restricted 7 days +satiation 7 days; restricted 3 days + satiation 4 days; and restricted 2 days + satiation 2 days. The restricted regime was achieved by offering fish 1% (maintenance ration) of their body weight per day adjusted after fortnightly weighing. African catfish showed partial compensatory growth under alternating periods of feed restriction and satiation feeding with significantly different values (P < 0.05) from fish fed in satiation throughout. However, significantly indifferent (P > 0.05) values of feed, protein, lipid and energy utilization were found under alternating periods of feed restriction and satiation feeding. Significantly higher (P < 0.05) feed intake was observed in treatment with satiation throughout than those in other treatments. All the feeding schedules showed no significant differences (P > 0.05) on body composition, organ indices, eviscerated carcass composition, viscera lipid and liver lipid. These studies reveals that C. gariepinus showed partial compensatory growth responses at alternating periods of restricted and satiation feeding. 相似文献

ABSTRACTIn the present study, aqueous solutions of sodium caseinate (SC) 8% (w/w) were provided in the form of four treatments—including SC without any essential oil (EO) as well as SC with 0.25, 0.5, and 1% (v/w) concentrations of Zataria moltiflora EO. Fish samples were treated with aqueous solutions and stored at 4 ± 1°C for 20 days. The control and treated fish samples were analyzed for chemical (total volatile basic nitrogen, pH, thiobarbituric acid, peroxide value), microbiological (total viable count and psychrotrophic count), and sensory analyses every 4 days. Based on results, SC edible coating had no antimicrobial effect but acted like a barrier against the fish flesh contamination. Moreover, incorporating Zataria moltiflora EO with SC edible coating reduces peroxide value and thiobarbituric acid-reactive substances values as well as the chemical and microbiological spoilage of refrigerated ?sh samples. Furthermore, sensory evaluation results showed that the presence of thyme oil improved the sensory quality of the rainbow trout. 相似文献
