Two size groups (0.5 and 3 kg) of Atlantic salmon, Salmo salar L., were stressed by netting and transport from sea cages into land-based tanks. Handling lasted for 1 h. Before onset of stress, basal concentrations of haematocrit and haemoglobin, plasma cortisol and glucose, and liver and muscle glycogen were measured. All levels were within ranges reported for salmon. After handling and at chosen time intervals during recovery, samples were taken to judge the impact of handling and whether secondary changes in carbohydrate metabolism were related of fish size. A low cortisol peak indicated a mild stress reaction after handling independent to fish size. Plasma glucose peaked as cortisol declined, and returned to basal levels within 48 h. Liver glycogen seemed to be the main source of plasma glucose. No changes were measured in muscle glycogen concentrations. The results indicate a high tolerance to handling stress in Atlantic salmon independent of fish size.  相似文献

This study examined the effects of boat noise pollution on the stress indices of gilthead sea bream (Sparus aurata, Linnaeus 1758). To assess the stress response in these fish, biometric values and plasma parameters such as ACTH, cortisol, glucose, lactate, haematocrit, Hsp70, total protein, cholesterol, triglycerides and osmolarity were analysed. After acclimatization of the animals, the experiment was carried out in a tank fitted with underwater speakers where the fish were exposed to sound treatments (in duplicate) consisting of: 10 days of no sound (control treatment; the animals were only exposed to the experimental tank’s background noise) and 10 days of noise derived from original recordings of motor boats, including recreational boats, hydrofoil, fishing boat and ferry boat (vessel noise treatment). The exposure to noise produced significant variations in almost all the plasma parameters assessed, but no differences were observed in weights and fork lengths. A PERMANOVA analysis highlighted significantly increased values (p < 0.05) of ACTH, cortisol, glucose, lactate, haematocrit, Hsp70, cholesterol, triglycerides and osmolarity in the fish exposed to vessel noise for 10 days. This study clearly highlights that anthropogenic noise negatively affects fish, and they are valuable targets for detailed investigations into the effects of this global pollutant. Finally, these experimental studies could represent part of the science that is able to improve the quality of the policies related to management plans for maritime spaces (Marine Strategy Framework Directive 56/2008 CE) that are aimed at stemming this pollutant phenomenon.  相似文献

The effects of tank color on the growth, stress responses, and skin color of snakeskin gourami (Trichogaster pectoralis) were investigated in this study. Fish with initial body weights of 5.03±0.00 g were reared in five experimental tank colors (white, red, green, blue, and black) for 8 weeks. Each tank color was tested in triplicate with an initial stocking density of 15 fish per tank. Fish were fed with commercial sinking pellets at 4% of the average body weight per day. Growth performance, feed utilization efficiency, stress indicators (hematocrit, blood glucose, plasma cortisol levels), and skin color parameters were investigated. The fish reared in blue tanks had a significantly higher average final body weight (9.73?±?0.14 g) and significantly lower average feed conversion ratio (3.42?±?0.12) than the fish reared in black tanks (P?<?0.05). The fish reared in black tanks exhibited higher average hematocrit (36.63?±?1.11%), blood glucose (48.33?±?1.45 mg dL?1), and plasma cortisol (9.00?±?0.56 μg dL?1) levels than those reared in the other tank colors. However, the blood glucose levels in only the fish reared in black tanks were significantly higher than those in the fish reared in the other tank colors. The fish skin color ranged from very pale (high skin lightness) in the white tanks to very dark (low skin lightness) in the black tanks, and 80% of the variation in skin lightness were explained by the tank lightness. The use of a blue tank resulted in normal skin color; hence, blue tanks will not affect the customer acceptance of the fish. Our study revealed that blue is the most appropriate tank color for culturing snakeskin gourami.  相似文献

The purpose of this study was to determine the cortisol response of spotted wolffish (Anarhichas minor) after exposure to air immersion and netting (acute stress) under a range of increasing densities (chronic stressor). In addition, the presence of a cumulative negative impact from chronic stress was assessed by the application of an additional acute stressor following rearing at various densities, and proxies of behavior were evaluated (feeding and aggression data). The stress response to air immersion and netting led to increasing plasma cortisol values at 0.5, 13, and 37 h post-stress, with the highest value achieved after 13 h. At 168 h, cortisol concentrations returned to similar pre-stress value. Changes in cortisol concentrations were relatively low after acute stress (five to six fold increases) and recovery time long lasting (>37 h). Prolonged rearing at 20, 30, and 40 kg m?2 showed a non-significant trend of increasing cortisol values with increasing density, and the cortisol response after the application of an additional acute stressor was significantly altered, i.e., higher cortisol levels were measured in unstressed (chronic stress only) compared to stressed fish (chronic stress + acute stress). Our results suggest that (1) spotted wolffish is a low responder that additionally displays a long-lasting cortisol elevation following an acute stressor; (2) fixed densities did not lead to significant differences of plasma cortisol concentration or aggression levels, although involved marked changes in daily feed intake and food conversion efficiency were observed; and (3) cortisol levels reached acute stress values after prolonged chronic stress. Although this species displays indications of being relatively resistant to handling disturbances during aquaculture practices, it is nevertheless responsive to chronic stress factors that could alter the physiological response to subsequent acute stressor.  相似文献

分别在拥挤胁迫后第3、10、20、30天对鲫鱼(Carassiusauratus)血液皮质醇水平和血液溶菌酶水平进行了检测,并在每次采集血样后用嗜水气单胞菌(Aeromonashydrophila)进行人工感染以评估鱼体抵抗力。结果显示,血液皮质醇水平在胁迫后几天内显著升高(P<0.005),随后虽有所下降,但至第30天高密度组仍保持较高水平(216.32±11.91)ng/mL。实验初期,血液溶菌酶水平小幅升高,但经过长时间的拥挤后,高密度组血液溶菌酶活性受到极大抑制,显著低于同期对照组(P<0.01)。攻毒后的死亡率与胁迫时间成正相关,在最后一次采样期,高密度组死亡率高达58.3%,而同期对照组仅为16.7%,两者具统计学差异(P<0.01)。实验表明,经过拥挤胁迫后,鲫鱼血液皮质醇水平和溶菌酶水平均发生了变化,短期内,两者成正相关,而长期胁迫则表现为负相关。另外,拥挤胁迫也引起了鱼体抵抗力的下降。  相似文献

The physiological stress response of cutthroat trout, Oncorhynchus clarki, to three methods of loading into truck tanks for transport was evaluated. All loading methods caused a significant decrease in chloride and increase in plasma cortisol levels, relative to the baseline sample of unstressed fish. Based on plasma chloride changes, the conveyor method appeared the least stressful to the fish, while fish pump and dip netting were not significantly different from each other. Differences in plasma cortisol responses suggested that the conveyor and dip net method were less stressful than the fish pump. Plasma glucose concentrations in the fish were not elevated after loading and did not differ among loading methods. Overall, the conveyor method was the least stressful method of loading, but the differences were not sufficient magnitude to preferentially use one method over another.  相似文献

In this study, groups of diploid (control) and triploid brook trout were subjected to a 5 min acute handling/confinement stress. Blood samples were collected from equal numbers of diploid and triploid fish at regular predetermined intervals, beginning prior to confinement and continuing for up to 3h after confinement. These blood samples were used for the measurement of hematocrit and plasma cortisol and glucose concentrations. The effects of cohort sampling and the diel cycle on these variables were also analysed. Except for minor differences, both groups responded similarly to the acute stress, with short-term elevations in plasma cortisol and hematocrit levels, and a more sustained increase in plasma glucose levels. No significant differences were found between diploid and triploid fish in their reactions to cohort sampling. However, the act of removing an individual from a tank caused significant changes in plasma cortisol levels in cohorts remaining in that tank (p < 0.001). The Atlantic salmon (Salmo salar) stress response was examined by measuring plasma cortisol, glucose and chloride in fish after water level reduction within rearing tanks. Maximum plasma cortisol levels (366±43 (SD) nM and 534±280 nM for Groups 1 and 2, respectively) were observed 20 min after application of the stressor. Cortisol levels were down to control levels 24 h later. The pattern for cortisol changes observed within these two groups were comparable. Plasma glucose and chloride concentrations did not change significantly in either of the experimental groups. The fish in group 2 were then repeatedly subjected to the same stressor every third day. After the fifth exposure to the stressor, blood was obtained from fish at times corresponding to sampling after the first exposure. Their maximum plasma cortisol level only reached 223 (±96) nM, and was down to prestress levels within 2 h. Plasma glucose and chloride concentrations did not change significantly in this second experiment. This challenge test revealed an acute primary response in Atlantic salmon without any apparent harmful secondary responses, that may thus serve as a standardized reference stressor using other fish groups under comparable conditions.  相似文献

We evaluated whether bearing tetrodotoxin (TTX) affects salinity stress in the juvenile tiger puffer Takifugu rubripes. Juveniles of hatchery-reared non-toxic T. rubripes [body weight (BW): 1.7?±?0.2 g, n?=?120] were divided into six tanks and acclimatized to salinity (8.5 ppt) that is equivalent to blood osmolality. Fish in three tanks were fed non-toxic diet, and those in the other three tanks were fed a TTX-containing diet (356 ng/g diet) three times a day until satiation. In each diet treatment, salinity of one tank was kept at 8.5 ppt, and the other two tanks were adjusted to either 1.7 or 34.0 ppt, and fish were reared for another 33 days. Then, we compared survival, growth, TTX accumulation, plasma osmolality, plasma cortisol, and glucose levels among treatments. We detected TTX only in the fish in the TTX-diet groups. Survival was highest at 8.5 ppt (70%) and lowest at 1.7 ppt in the TTX-diet group (20%). The BW was greater at 8.5 ppt, and plasma osmolality was significantly higher at 34.0 ppt than at any other salinities. Plasma cortisol level was significantly higher but glucose level was lower at 1.7 ppt. Possessing TTX at a low salinity may be lethal to tiger puffer juveniles.

The influence of salinity and dietary composition on blood parameter values (haematocrit, leucocrit, immature lymphocytes, mature lymphocytes, granulocytes, plasma osmolarity and total plasma protein) of red hybrid tilapia, Oreochromis niloticus (Linnaeus) x O. mossambicus (Peters), was studied. Two groups of tilapia were fed a high-protein diet while kept in fresh or brackish water, respectively, and compared with two groups fed a low-protein diet under similar environmental conditions. Treatments were executed in duplo. Results show that salinity influenced all cellular blood parameters except the haematocrit. Dietary composition influenced the total plasma protein and haematocrit, while all parameters changed as time progressed during the experiment. It was concluded that the environmental parameters investigated in the present study should be considered when estimating fish health based on blood parameter values.  相似文献

In this study, we investigated the toxic effects of ammonia‐N on the plasma and haematological parameters and histological structure of blunt snout bream (Megalobrama amblycephala) juveniles. The fish (initial weight, 14.79 ± 0.01 g) were randomly sorted into six tanks (200 L), and each tank was stocked with 40 fish for culture. The juveniles were exposed to two ammonia‐N levels—0 mg/L (control group) and 25 mg/L (experimental group)—and sampled at 0, 6, 12, 24, 48 and 72 hr, and then they underwent 96 hr of post‐exposure recovery. The results showed that ammonia‐N had significant effects on the plasma and haematological parameters. The treatment group showed increased cortisol, plasma ammonia and haematocrit levels and white blood cell count with increasing exposure time, up to 24 hr, and then the levels and count decreased. A significantly higher plasma glucose level was observed in the treatment group at 12 hr. After 96 hr of post‐exposure recovery, all parameter levels decreased to the control levels. The fish displayed histopathological alterations in the gills, liver and kidney. The results indicate that the severity of the lesions clearly differed among the organs, with the liver showing the most extensive damage, followed by the gills and kidney. Adverse effects to physiological indicators and histological structure increased with increasing exposure time before 24 hr. The fish showed self‐regulation; however, the histological structure could not recover fully, the gill tissue showed irreversible changes and the kidney tissue exhibited the worst recovery ability.  相似文献

The effects of clove oil anaesthetic on mitigating the physiological responses to air exposure, a stressful and routine situation in fish farming, laboratory conditions and sport fishing (catch and release), were evaluated in lambari (A. altiparanae). Adult females (n = 80) were randomly sorted to receive one of four treatments: control, anaesthesia (clove oil 50 mg/L), stress (5 min air exposure) and pre‐anaesthesia associated to stress. Their cortisol, glucose, lactate and haematocrit levels, the hepatosomatic index (HSI), liver and muscle glycogen, lipid peroxidation and the enzymatic activity of lactate dehydrogenase (LDH), catalase (CAT) and glutathione reductase (GR), were recorded. Glucose levels increased (53.9%) after anaesthesia and/or stress. The stress situation increased plasma cortisol (146.6%), lactate (294.6%) and lipid peroxidation in white muscle (45%) and decreased glycogen in white muscle (40.1%). The haematocrit increased after stress or anaesthesia (7.9%) while the liver glycogen and HSI did not change. Anaesthesia or stress did not affect the LDH but reduced the activity of CAT (46.1%) and GR (30.3%). We concluded that the anaesthetic clove oil in the concentration 50 mg/L modulates the physiological responses to air exposure stress improving the welfare; air exposure and clove oil affect the antioxidant defences of lambari; the activity of CAT and GR and the concentration of MDA can be used as biomarkers of stress in A. altiparanae.  相似文献

Plasma cortisol is the most commonly used indicator of stress in fish but, as the blood sampling procedure itself can be a source of stress, it would be helpful to measure cortisol using less invasive matrices. It is also necessary to find alternative matrices as stress indicators in dead fish in which blood sampling is impossible. In the present study, we investigated transport stress in three aquaculture species, European sea bass (Dicentrarchus labrax L.), common carp (Cyprinus carpio L.) and rainbow trout (Oncorhynchus mykiss Walbaum), by cortisol determination (radioimmunoassay) in plasma and other matrices (skin mucus, gut content, lateral muscle and caudal fin). Cortisol significantly increased after transport in all species and matrices, except in the sea bass gut content, where it remained unchanged. The three species responded to transport stress by producing different cortisol levels. In conclusion, the significant correlation found between plasma cortisol and most of the other matrices opens up the possibility of using them to evaluate stress in fish: mucus sampling is a less invasive method than blood sampling, and in addition to muscle and fin sampling, it can be used in postmortem fish.  相似文献

The existence of daily rhythms under light/dark (LD) cycles in plasma cortisol, blood glucose and locomotor and self-feeding activities, as well as their persistence (circadian nature) under constant light (LL), was investigated in Senegalese sole (Solea senegalensis). For the cortisol and glucose rhythms study, 48 soles were equally distributed in 8 tanks and exposed to a 12:12 LD cycle and natural water temperature (experiment 1). After an acclimation period, blood was sampled every 3 h until a 24-h cycle was completed. Blood glucose levels were measured immediately after sampling, while plasma cortisol was measured later by ELISA. In experiment 2, the fish were exposed to LL for 11 days, and after this period, the same sampling procedure was repeated. For the study of locomotor and self-feeding rhythms (experiment 3), two groups of sole were used: one exposed to LD and the other to LL. Each group was distributed within 3 tanks equipped with infrared photocells for the record of locomotor activity, and self-feeders for feeding behavior characterization. The results revealed a marked oscillation in cortisol concentrations during the daily cycle under LD, with a peak (35.65 ± 3.14 ng/ml) in the afternoon (15:00 h) and very low levels during the night (5.30 ± 1.09 ng/ml). This cortisol rhythm persisted under LL conditions, with lower values (mean cortisol concentration = 7.12 ± 1.11 ng/ml) and with the peak shifted by 3 h. Both rhythms were confirmed by COSINOR analysis (p < 0.05). The synchronizing role of temperature and feeding schedule, in addition to light, is also discussed. Diel rhythms of glucose were not evident in LD or LL. As to locomotor and self-feeding activity, a very marked rhythm was observed under LD, with higher activity observed during the night, with acrophases located at 2:14 and 3:37 h, respectively. The statistical significance of daily rhythms was confirmed by COSINOR analysis. Under LL, both feeding and locomotor rhythms persisted, with an endogenous period (τ) around 22.5 h. In short, our findings described for the first time the existence of circadian cortisol and behavioral circadian rhythms in flat fish. Such results revealed the importance of taking into account the time of day when assessing stress responses and evaluating physiological indicators of stress in fish.  相似文献

Hormone‐induced spawning of channel catfish held communally in tanks is a reliable method to produce channel catfish, Ictalurus punctatus ♀ × blue catfish, Ictalurus furcatus ♂, F1 hybrid catfish fry. However, mature catfish are crowded, and repeatedly handled during the process of induced ovulation. Repeated handling of gravid females is stressful and may impair ovulation, egg quality, and reproductive performance. Three trials were conducted to evaluate the effects of two methods of confining post‐hormone‐injected female channel catfish on stress response (cortisol concentrations) and reproductive performance: fish were either held individually while suspended in soft, nylon‐mesh bags or communally in a concrete tank. Percent of females ovulated to hormone treatment, relative fecundity, percent egg viability, and latency of channel catfish did not differ for fish in the two treatments. However, percent hatch and fry/kg of females was higher (P < 0.05) for fish held in bags that for fish held communally in tanks. Mean plasma cortisol response immediately prior to the first hormone injection (0 h) did not differ among fish groups in the two treatments. However, mean plasma cortisol concentrations were significantly lower (P < 0.05) for fish in the bag treatment at 16 and 36 h compared to fish held communally in tanks. Plasma estradiol levels (measure of oocyte maturation) were assessed at 0, 16, and 36 h after hormone injection; concentrations were (P < 0.05) higher at 16 h compared to 0 and 36 h; however, estradiol concentrations did not differ for fish held in the two treatments (P > 0.05). Suspending hormone‐injected broodfish individually in soft bags reduced stress response, improved egg hatching rate, and increased hybrid fry produced per kg weight of female broodfish. Using this simple technology, farmers can improve the efficiency of hatcheries producing hybrid catfish fry.  相似文献

The aim of this study was to examine species‐specific differences in pre‐ and post‐stress concentrations of haematological, metabolic and hormonal parameters in two Mediterranean fish species, one with relatively high (European sea bass, Dicentrarchus labrax) and the other with low (meagre, Argyrosomus regius) basal and post‐stress blood cortisol concentrations, in relation to different water temperatures. Fish were reared in net‐pen sea cages and exposed to an identical acute stressor (crowding and chasing with a net for 5 min) in three different periods of the year. Results indicated that inter‐specific differences occurred in most of the examined parameters. In addition, within each species, differences between sampling periods existed in plasma cortisol, glucose, osmolarity, blood pH and muscle glycogen concentration. Glucose, lactate and osmotic pressure showed a constant pattern of change, with maxima at half an hour post stress in both species, while cortisol, and blood and muscle pH, were significantly altered only in E. sea bass. No alterations were observed in liver and muscle glycogen concentrations in E. sea bass, while a significant reduction was evident only in the coldest temperature in meagre. It is concluded that there are important species‐specific differences in the magnitude of hormonal and metabolic response to acute stress and that both basal and post‐stress blood cortisol concentrations are affected by the sampling period.  相似文献

The effects of 60‐mg L?1 clove oil and 60‐mg L?1 tricaine methanesulphonate (MS‐222) on the blood chemistry of rainbow trout were compared after exposure to handling stress via caudal puncture blood sampling. Fish sampled by caudal puncture and subsequently exposed to anaesthetics showed a typical handling stress response over a 48‐h period. There were no significant differences between the responses of fish exposed to equal concentrations of clove oil and MS‐222, with the following exceptions: the blood glucose at full anaesthesia, and lactate at full recovery increased significantly in the clove oil‐exposed fish. In a subsequent experiment, the stress response observed in fish sampled by caudal puncture and exposed to clove oil and MS‐222 was compared with a non‐anaesthetized control group. The increases in plasma cortisol levels were significantly lower at recovery in fish treated with either anaesthetic compared with the control fish. Fish exposed to MS‐222 had significantly higher cortisol levels at 1 h. These findings show that few differences exist between the anaesthetic effects of clove oil and MS‐222 on the physiological response of fish to stress. However, clove oil is more effective at reducing the short‐term stress response induced by handling and blood sampling, and is recommended as an alternative fish anaesthetic.  相似文献

The parasite, Anguillicola crassus is a non-native species that infects naive European eels, Anguilla anguilla, and causes pathological damage to the swimbladder, potentially compromising their ability to cope with hypoxic conditions. This study aimed to elucidate whether anguillicolosis exacerbates the stress responses to exposure to hypoxic water, conditions that have been implicated in mass mortalities of wild infected European eels. Blood parameters in infected and uninfected eels were measured during exposure to severe hypoxia over an 8-h period. Infected fish showed significantly higher levels of plasma cortisol compared with uninfected eels after 4 h of hypoxia. Uninfected fish showed an almost twofold increase in plasma glucose after 8-h exposure to hypoxia but infected fish showed no significant change, so that the plasma glucose concentration was significantly higher in uninfected eels than in infected eels. Both groups showed similar elevations in blood haematocrit, suggesting a similar catecholamine response in infected and uninfected eels. The lack of a hyperglycaemic response in infected eels, despite indirect evidence of a catecholamine response to hypoxia, may reflect an increase in glucose turnover. The data suggest that anguillicolosis results in a significantly greater corticosteroid stress response to hypoxia accompanied by a higher metabolic cost.  相似文献

Three experiments were conducted in flow‐through tanks at 24, 27 and 30 C to evaluate growth, food conversion, morbidity, survival, and the cortisol stress response of surubim Pseudoplatystoma sp. at different temperatures. In Experiment 1, fish (mean initial weight, 33.3 ± 7.2 g) reared at 27 and 30 C for 60 d grew significantly faster than fish reared at 24 C (P < 0.05). Fish at the lowest temperature showed the poorest feed conversion ratio (FCR, 5:1), while fish at 27 and 30 C had FCRs of 2.5:1 and 3.5:1, respectively. Mortality and morbidity rates were inversely proportional to the temperatures tested. In Experiment 2, serum cortisol levels following an acute handling stressor peaked at 30 min after stress, returned to near‐resting levels at 1 h after stress, and completely returned to resting levels at 3 h after stress in fish at all temperatures tested. Peak serum cortisol levels were higher in fish at 30 C (321.1 ± 38.8 ng/mL) than those in fish at 27 and 24 C (143.3 ± 37.4 ng/mL and 104.2 ± 37.2 ng/mL, respectively). In Experiment 3, fish (mean initial weight, 67 ± 8.58 g) reared at 27 C for 137 d grew significantly faster than fish at 30 and 24 C. Virtually, all fish reared in 30 C had some sign of morbidity (skin lesions and fin abrasion) compared to only a small percentage of fish in the other two temperatures. Again, the highest cortisol peak level was measured in fish reared at 30 C (117.6 ± 16.7 ng/mL), as compared to fish at 24 and 27 C (99.2 ± 15.5 ng/mL and 80.2 ± 12.8 ng/mL, respectively). Our findings indicate that the optimum temperature for rearing surubim in tanks is 27 C. At 30 C, these fish may suffer from stress, as suggested by the elevated peak of cortisol and morbidity and mortality rates. The relatively rapid cortisol stress response of surubim suggests that they may be more tolerant to handling stressors than other fish species.  相似文献

Exposure of fish to stressors can elicit biochemical and organismal changes at multiple levels of biological organization collectively known as stress responses. The organismal (plasma glucose and cortisol levels) and cellular (hepatic hsp70) stress responses in fish have been studied in several species, but little is known about sex-related differences in these responses. In this study, we exposed sexually immature juvenile chinook salmon (Oncorhynchus tshawytscha) to bleached kraft mill effluent (BKME: 0%, 1%, and 10% v/v) for 30 days and then measured components of their organismal and cellular stress responses. Males exposed to 1% BKME had higher levels of plasma glucose than females. Plasma cortisol levels were unaffected in females exposed to BKME, but males exposed to 10% BKME had significantly higher levels of plasma cortisol relative to non-exposed males. While exposure to BKME did not affect hsp70 levels in males, females exposed to 1% BKME had higher levels of hsp70 relative to non-exposed and 10% BKME groups. Within any given treatment, females had higher levels of hsp70 relative to males. This study demonstrates that sex-related differences exist in commonly used indicators of stress in fish, and points out the importance of considering the sex of the fish in stress research.
