Aphid parasitoids are common in Brazilian wheat fields, and parasitize aphids at the wheat tillering stage. However, there is little information available about when this natural enemy occurs in wheat crops. This study investigated the initial occurrence of aphid parasitoids in four commercial wheat crops in northern Paraná during the 2009 crop season. We installed two Malaise traps at each wheat farm, and 400 tillers were assessed weekly in each field for aphid abundance. During this study, we captured 4,355 aphid parasitoids and 197 aphids. Three species of braconid parasitoids were identified, including Aphidius colemani (Viereck 1912), Lysiphlebus testaceipes (Cresson 1880), and Diaeretiella rapae (McIntosh 1855). The aphids species identified were Rhopalosiphum padi (Linnaeus 1758) and Sitobion avenae (Fabricius 1775). This study showed that aphid parasitoids are present in wheat crops even when aphid densities are low, and in one farm, occurred before the aphids colonization. These reports can justified the high efficiency of these natural enemies against aphids in wheat fields.  相似文献

One of the largest threats to biodiversity is land use change and habitat loss. Hollow oaks (Quercus spp. L.) are well-defined patches that are hotspots for biodiversity and red-listed species, but they are often rare and fragmented in the landscape. We investigated the effect of patch size, habitat quality, and isolation on functional groups and red-listed saproxylic beetles in hollow oaks (n = 40) in Norway. The groups were defined by host tree association, trophic grouping, and red-listed status. Habitat quality, represented by tree form was most important in explaining species richness for most groups. Patch size, represented by circumference and amount of dead branches, was most important in explaining abundance. Isolation, that is single oaks compared with oaks in groups, had a negative effect on the abundance of beetles feeding both on wood and fungi (xylomycethopagous), as well as on species associated with broadleaved trees (oak semi-specialists), but did not affect species richness. This indicates that at this scale and in this landscape, isolated oaks are as species rich and valuable for conservation as other oaks, although some functional groups may be more vulnerable to isolation than others. The red-listed species only responded to patch size, indicating that oaks with large circumference and many dead branches are especially important for red-listed species and for conservation.  相似文献

Increased nitrogen applications to crops influence plant-insect interactions and potentially increase herbivore population growth. The effect of nitrogen applications on the interactions between a wheat host plant, Triticum aestivum, and its herbivores, Sitobion avenae (Fabricius, 1775) and Rhopalosiphum padi (Linnaeus, 1758) was investigated. Adult body weight, fecundity, maturity rate and longevity of the aphids were measured under different nitrogen application on four wheat varieties, Solstice, Einstein, Deben and Alchemy. Adult body weight of R. padi on Alchemy and of S. avenae on Deben was lowest. Fecundity of R. padi was lesser on Deben and Alchemy while fecundity of S. avenae was least on Alchemy. Days to reach maturity were same on all varieties in S. avenae but in R. padi, Deben positively affected the days to reach maturity. Longevity of S. avenae and R. padi was the same on all varieties. N-fertilizer had a positive effect on the adult weight achieved by S. avenae and R. padi. Fecundity and longevity of both species were also positively correlated with N-fertilizer application. In R. padi, N-fertilizer reduced the time to reach maturity but not in S. avenae.  相似文献

Venomics, the study of biological venoms, could potentially provide a new source of therapeutic compounds, yet information on the venoms from marine organisms, including cnidarians (sea anemones, corals, and jellyfish), is limited. This study identified the putative toxins of two species of jellyfish—edible jellyfish Rhopilema esculentum Kishinouye, 1891, also known as flame jellyfish, and Amuska jellyfish Sanderia malayensis Goette, 1886. Utilizing nano-flow liquid chromatography tandem mass spectrometry (nLC–MS/MS), 3000 proteins were identified from the nematocysts in each of the above two jellyfish species. Forty and fifty-one putative toxins were identified in R. esculentum and S. malayensis, respectively, which were further classified into eight toxin families according to their predicted functions. Amongst the identified putative toxins, hemostasis-impairing toxins and proteases were found to be the most dominant members (>60%). The present study demonstrates the first proteomes of nematocysts from two jellyfish species with economic and environmental importance, and expands the foundation and understanding of cnidarian toxins.  相似文献

Polygraphus proximus Blandford (Coleoptera: Curculionidae: Scolytinae) has caused mass mortality of fir (Abies spp. (Pinaceae)) forests across large areas of Russia in the past decade. More recently, mass mortality of A. veitchii  Lindl. due to P. proximus infestation has been reported in Japan. This bark beetle species traditionally has been considered to be polygynous because their galleries have multiple gallery arms, and because harem-polygyny is common in the tribe Polygraphini. Although the mating system(s) potentially could have a marked effect on their reproductive success and population dynamics, the reproductive behavior of the tree-killing bark beetle P. proximus has not been investigated in detail in a natural setting in Japan. We, therefore, investigated the number of males and females in a gallery and the number of gallery arms in Abies species in Japan. None of the galleries examined contained more than one male, and 57.2% of the galleries had multiple gallery arms, even though only 2.8% of the galleries contained two females. The findings showed that the typical mating system employed by P. proximus is monogyny and that this species constructs multiple gallery arms in each gallery. In addition, 70.4% of galleries in which the sex of adult beetles could be determined contained no males, and 26.6% contained no females, suggesting that P. proximus males and females re-emerge.  相似文献

Conotoxins are small peptides present in the venom of cone snails. The snail uses this venom to paralyze and capture prey. The constituent conopeptides display a high level of chemical diversity and are of particular interest for scientists as tools employed in neurological studies and for drug development, because they target with exquisite specificity membrane receptors, transporters, and various ion channels in the nervous system. However, these peptides are known to contain a high frequency and variability of post-translational modifications—including sometimes O-glycosylation—which are of importance for biological activity. The potential application of specific conotoxins as neuropharmalogical agents and chemical probes requires a full characterization of the relevant peptides, including the structure of the carbohydrate part. In this review, the currently existing knowledge of O-glycosylation of conotoxins is described.  相似文献

Field experiments were conducted to investigate the mechanism of the underlying patterns (abundance, species richness, diversity and similarity) of rove beetles in transgenic Bt (MON810) and in near isogenic maize stands in Hungary. During the three-year (2001–2003) survey, 1538 individuals and 21 species were sampled with pitfall traps. The Cry1Ab protein expressed by the MON810 maize hybrid did not influence the overall community structure. After grouping staphylinids into guilds we found no significant differences for non-aphidophagous predators and parasitoids, whereas there were significantly and marginally significantly higher abundances for predators with aphids in their diet in isogenic maize stands in 2002 and 2003 respectively. The abundance of the prey Rhopalosiphum padi (L.) showed a high fluctuation between stands and years and was numerically higher only in isogenic stands in the second half of the maize-growing season. The abundance of predatory guilds including aphids in their diet did not correlate with the total annual number of R. padi in the same year, but there was a linear correlation in successive years.  相似文献

Microalgae are well known for their biotechnological potential, namely with regard to bioactive lipidic components—especially carotenoids and polyunsaturated fatty acids (PUFA), well-known for therapeutic applications based on their antioxidant capacity. The aim of this work was to evaluate the influence of four distinct food-grade solvents upon extractability of specific lipidic components, and on the antioxidant capacity exhibited against both synthetic (2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS+•)) and biological reactive species (O2 and NO). A eukaryotic microalga (Scenedesmus obliquus (M2-1)) and a prokaryotic one (Gloeothece sp.) were used as case studies. Concerning total antioxidant capacity, the hexane:isopropanol (3:2) and acetone extracts of Sc. obliquus (M2-1) were the most effective against DPPH and ABTS+•, respectively. Gloeothece sp. ethanol extracts were the most interesting scavengers of O2, probably due the high content of linolenic acid. On the other hand, acetone and hexane:isopropanol (3:2) extracts were the most interesting ones in NO assay. Acetone extract exhibited the best results for the ABTS assay, likely associated to its content of carotenoids, in both microalgae. Otherwise, ethanol stood out in PUFA extraction. Therefore, profiles of lipidic components extracted are critical for evaluating the antioxidant performance—which appears to hinge, in particular, on the balance between carotenoids and PUFAs.  相似文献

Hollows of veteran trees (i.e., rot holes) provide habitat for many rare and threatened saproxylic invertebrates. Rot holes are highly heterogeneous, particularly in terms of substrate and microclimate conditions. There is, however, a dearth of information regarding the differences in biological communities inhabiting rot holes with different environmental conditions. Invertebrates were sampled from European beech (Fagus sylvatica) rot holes in Windsor, Savernake, and Epping Forests (United Kingdom). For each rot hole, physical and environmental conditions were measured, including tree diameter, rot hole dimensions, rot hole height, substrate density, water content, and water potential. These parameters were used to assess the influence of environmental conditions and habitat characteristics on invertebrate communities. Rot hole invertebrate communities were extremely diverse, containing both woodland generalist and saproxylic specialist taxa. Large variation in community structure was observed between rot holes and across woodlands; all sites supported threatened and endangered taxa. Environmental conditions in rot holes were highly variable within and between woodland sites, and communities were predominantly structured by these environmental conditions. In particular, turnover between invertebrate communities was linked to high β-diversity. The linked heterogeneity of environmental conditions and invertebrate communities in rot holes suggests that management of deadwood habitats in woodlands should strive to generate environmental heterogeneity to promote invertebrate diversity. Additional research is required to define how management and conservation activities can further promote enhanced biodiversity across rot holes.  相似文献

The mucus produced by many marine organisms is a complex mixture of proteins and polysaccharides forming a weak watery gel. It is essential for vital processes including locomotion, navigation, structural support, heterotrophic feeding and defence against a multitude of environmental stresses, predators, parasites, and pathogens. In the present study we focused on mucus produced by a benthic cnidarian, the sea anemone Actinia equina (Linnaeus, 1758) for preventing burial by excess sedimentation and for protection. We investigated some of the physico-chemical properties of this matrix such as viscosity, osmolarity, electrical conductivity, protein, carbohydrate, and total lipid contents. Some biological activities such as hemolytic, cytotoxic, and antibacterial lysozyme-like activities were also studied. The A. equina mucus is mainly composed by water (96.2% ± 0.3%), whereas its dry weight is made of 24.2% ± 1.3% proteins and 7.8% ± 0.2% carbohydrates, with the smallest and largest components referable to lipids (0.9%) and inorganic matter (67.1%). The A. equina mucus matrix exhibited hemolytic activity on rabbit erythrocytes, cytotoxic activity against the tumor cell line K562 (human erythromyeloblastoid leukemia) and antibacterial lysozyme-like activity. The findings from this study improve the available information on the mucus composition in invertebrates and have implications for future investigations related to exploitation of A. equina and other sea anemones’ mucus as a source of bioactive compounds of high pharmaceutical and biotechnological interest.  相似文献

Five new polyoxygenated marine steroids—punicinols A–E (1–5)—were isolated from the gorgonian Leptogorgia punicea and characterized by spectroscopic methods (IR, MS, 1H, 13C and 2-D NMR). The five compounds induced in vitro cytotoxic effects against lung cancer A549 cells, while punicinols A and B were the most active, with IC50 values of 9.7 μM and 9.6 μM, respectively. The synergistic effects of these compounds with paclitaxel, as well as their effects on cell cycle distribution and their performance in the clonogenic assay, were also evaluated. Both compounds demonstrated significant synergistic effects with paclitaxel.  相似文献

Bark beetles (Coleoptera: Scolytinae) are well-recognized vectors of Ophiostoma species. Three non-native bark beetle species infest various Pinus species in South Africa, and they are known to carry at least 12 different species of ophiostomatoid fungi. Some of these fungi have not been identified to species level. The aim of this study was to determine or confirm the identities of Ophiostoma species associated with bark beetles in South Africa using comparisons of DNA sequence data. Identities of Ophiostoma ips, O. floccosum, O. pluriannulatum, O. quercus and O. stenoceras were confirmed. Ophiostoma abietinum, O. piliferum and Pesotum fragrans are recognised for the first time and the new species, O. aurorae sp. nov., is described from pine-infesting bark beetles in South Africa.Taxonomic novelty: Ophiostoma aurorae X.D. The sampling of living insects should be avoided in highly endangered species when the sampling would further increase the risk of population extinction. Nonlethal sampling (wing clips or leg removals) can be an alternative to obtain DNA of individuals for population genetic studies. However, nonlethal sampling may not be possible for all insect species. We examined whether remnants of traffic-killed specimens of the endangered and protected flighless longhorn beetle Iberodorcadion fuliginator (L., 1758) can be used as a resource for population genetic analyses. Using insect fragments of traffic-killed specimens collected over 15 yr, we determined the most efficient DNA extraction method in relation to the state of the specimens (crushed, fragment, or intact), preservation (dried, airtight, or in ethanol), storage duration, and weight of the sample by assessing the quantity and quality of genomic DNA. A modified cetyltrimethyl ammonium bromide method provided the highest recovery rate of genomic DNA and the largest yield and highest quality of DNA. We further used traffic-killed specimens to evaluate two DNA amplification techniques (quantitative polymerase chain reaction [qPCR] and microsatellites). Both qPCR and microsatellites revealed successful DNA amplification in all degraded specimens or beetle fragments examined. However, relative qPCR concentration and peak height of microsatellites were affected by the state of specimen and storage duration but not by specimen weight. Our investigation demonstrates that degraded remnants of traffic-killed beetle specimens can serve as a source of high-quality genomic DNA, which allows to address conservation genetic issues.  相似文献

Worldwide, two of the most harmful invasive ants typical of disturbed sites are Solenopsis geminata (Fabricius) and Wasmannia auropunctata (Roger). Both are natives of the Neotropics and are widely distributed all over the tropics. Within its original geographic range, there are few data on its abundance and potential damage to natural ecosystems. In this study, we recorded their abundance and relationships to diversity and richness of soil ant communities in two localities with different amount of forested area (López Mateos, LM 77% and Venustiano Carranza, VC 27%), at Los Tuxtlas reserve. In each locality, four land use systems (LUS) were sampled: tropical rain forests, agroforestry plantations, annual crops, and pastures. Data were gathered from 360 ant samples obtained from litter squares, pitfall traps, and soil monoliths in 40 sampling points (20 per locality, and five per LUS). Solenopsis geminata was more abundant in LM than in VC; the opposite trend was observed for W. auropunctata. In LM, S. geminata was more abundant in crops than in the other LUS, whereas W. auropunctata tended to have higher abundances in less managed sites of both localities. Abundance and species richness of ant communities were higher in LM than in VC. At regional and local levels, we found negative relationships between the abundance of S. geminata and species richness; the inverse pattern was found for W. auropunctata. We conclude that at Los Tuxtlas, W. auropunctata can be considered as a typical dominant native species, whereas S. geminata is the common exotic invasive ant.  相似文献

Understanding collection methodologies and their limitations are essential when targeting specific arthropods for use in habitat restoration, conservation, laboratory colony formation, or when holistically representing local populations using ecological surveys. For dung beetles, the most popular collection methodology is baited traps, followed by light traps and unbaited flight-intercept traps during diversity surveys. A less common collection method, flotation, is assumed to be laborious and messy, and so only a handful of papers exist on its refinement and strengths. Our purpose was threefold: First, we tested the recovery and survival rates of Labarrus (=Aphodius) pseudolividus (Balthasar) and Onthophagus taurus (Schreber) when floating beetle-seeded dung pats to determine potential collection and safety issues. We collected 72.4 and 78% of the seeded L. pseudolividus and O. taurus, respectively, with >95% survival rating. Second, we developed a flotation-sieving technique that enables users to rapidly collect and passively sort dung beetles with less time and effort. Specifically, we often collected 50–100 g of wild dung beetles within a couple of hours of gathering dung and sorted them in a couple more by allowing dung beetles to sort themselves by size within a series of sieves; Third, we reviewed flotation-based advantages and disadvantages in comparison to other methodologies.  相似文献

Microlepidopterous leafminers are a potential threat to Spanish cider-apple orchards. For pest management purposes it is important 1) to know the species composition, their incidence and their spatial distribution and 2) to have an efficient and accurate sampling method. To fill this gap of knowledge, the occurrence of leafminers was followed in four cider-apple orchards at Asturias (NW Spain) from summer to harvest during 2003-2005. A total of 5120 mines from six different leafminer species were identified. Phyllonorycter blancardella (Fabricius) mines were the most abundant, followed by those of Leucoptera scitella (Zeller). These two species accounted for more than half of the total mines counted (58%). The other four species detected were Stigmella malella (Stainton), Lyonetia clerkella (L.), Phyllonorycter corylifoliella (Hübner) and Callisto denticulella (Thunberg). Abundance and incidence of all leafminer species taken as a whole were similar among orchards, although year-to-year oscillations were recorded. Based on the low densities observed (maximum abundance for a given orchard and sampling date was 0.548 mines per leaf), leafminers could not be considered as a key pest in our apple orchards. Taylor’s coefficient b obtained for the different leafminer species were indicative of aggregated distributions of both infested leaves and mines within the tree. Enumerative and binomial sampling plans for apple leafminers were developed. Although binomial plans are usually faster than enumerative ones, the latter was recommended because of the lower size required for practical usage (the sampling of 20 leaves would be enough for estimating populations around the economic threshold of one mine per leaf).  相似文献

The gut microbiota of insects usually plays an important role in the development and reproduction of their hosts. The fecundity of Henosepilachna vigintioctopunctata (Fabricius) varies greatly when they develop on different host plants. Whether and how the gut microbiota regulates the fecundity of H. vigintioctopunctata was unknown. To address this question, we used 16S rRNA sequencing to analyze the gut microbiomes of H. vigintioctopunctata adults fed on two host plant species (Solanum nigrum and Solanum melongena) and one artificial diet. The development of the ovaries and testes was also examined. Our results revealed that the diversity and abundance of gut microorganisms varied significantly in insects reared on different diets. The gut microbiota of H. vigintioctopunctata raised on the two host plants was similar, with Proteobacteria being the dominant phylum in both groups, whereas Firmicutes was the dominant phylum in the group reared on the artificial diet. The predominant microbiota in the S. nigrum group were Acinetobacter soli and Acinetobacter ursingii (Acinetobacter, Moraxellaceae); Moraxella osloensis (Enhydrobacter, Moraxellaceae); and Empedobacter brevis (Empedobacter, Weeksellaceae). The microbiota in this group are associated with high lipid metabolism. In addition, the beetles’ ovaries and testes were more highly developed in the S. nigrum group than in the other two groups. These findings provide valuable information for elucidating the complex roles the gut microbiota play in the fecundity of H. vigintioctopunctata, and may also contribute to developing future novel control strategies involving this economically important pest.  相似文献

Iron, as inorganic ion or as oxide, is widely used by biological systems in a myriad of biological functions (e.g., enzymatic, gene activation and/or regulation). In particular, marine organisms containing silica structures—diatoms and sponges—grow preferentially in the presence of iron. Using primary sponge cell culture from S. domuncula–primmorphs—as an in vitro model to study the Demospongiae spiculogenesis, we found the presence of agglomerates 50 nm in diameter exclusively inside sponge specialized cells called sclerocytes. A clear phase/material separation is observed between the agglomerates and the initial stages of intracellular spicule formation. STEM-HRTEM-EDX analysis of the agglomerates (30–100 nm) showed that they are composed of pseudohexagonal nanoparticles between 5 and 15 nm in size, displaying lattice parameters corresponding to hematite (Fe2O3) and mixed iron oxide phases typically attributed to ferritin. Further analysis, using western blotting, inductively coupled plasma mass spectrometry (ICP-MS), sequence alignment analysis, immunostaining and magnetic resonance imaging (MRI), of mature spicule filaments confirm the presence of ferritin within these organic structures. We suggest that S. domuncula can be classified as a dual biomineralizating organism, i.e., within the same cellular structure two distinct biomineralizing processes can occur as a result of the same cellular/metabolic function, spiculogenesis.  相似文献

Empedopeptins—eight amino acid cyclic lipopeptides—are calcium-dependent antibiotics that act against Gram-positive bacteria such as Staphylococcus aureus by inhibiting cell wall biosynthesis. However, to date, the biosynthetic mechanism of the empedopeptins has not been well identified. Through comparative genomics and metabolomics analysis, we identified empedopeptin and its new analogs from a marine bacterium, Massilia sp. YMA4. We then unveiled the empedopeptin biosynthetic gene cluster. The core nonribosomal peptide gene null-mutant strains (ΔempC, ΔempD, and ΔempE) could not produce empedopeptin, while dioxygenase gene null-mutant strains (ΔempA and ΔempB) produced several unique empedopeptin analogs. However, the antibiotic activity of ΔempA and ΔempB was significantly reduced compared with the wild-type, demonstrating that the hydroxylated amino acid residues of empedopeptin and its analogs are important to their antibiotic activity. Furthermore, we found seven bacterial strains that could produce empedopeptin-like cyclic lipopeptides using a genome mining approach. In summary, this study demonstrated that an integrated omics strategy can facilitate the discovery of potential bioactive metabolites from microbial sources without further isolation and purification.  相似文献
