Bombyx mori (B. mori) silk was modified with the nano-TiO2 and chitosan dispersion system by the crosslinking reactions of citric acid (CA) and maleic anhydride (MA). The average size of the nano-TiO2 particles in the aqueous dispersion system was 36.7 nm. The scanning electron microscopy (SEM) micrographs showed that the nano-TiO2 particles were spherical and homogeneously dispersed in the dispersion system, and the surface ofB. mori silk fiber treated with the nano-TiO2 and chitosan dispersion system was rougher than that of the untreated one. X-ray diffraction (XRD) and Fourier transform infrared (FT-IR) Spectrometry indicated that the crystallinity of theB. mori silk fiber increased after treatment. It was also found that the nano-TiO2 and chitosan contributed to significantly enhance the mechanical properties including breaking strength, breaking elongation, initial modulus, rupture work, and elastic recovery property of theB. mori silk fiber. The wrinkle-resistant performance of the treatedB. mori silk fabrics was also greatly improved.  相似文献

Platycodon grandiflorum (PG), a species of herbaceous flowering perennial plant of the family Campanulaceae, has been used as a traditional oriental medicine for bronchitis, asthma, pulmonary tuberculosis, diabetes, hepatic fibrosis, bone disorders and many others similar diseases and as a food supplement. For the primary profiling of PG gas chromatography coupled with high resolution – time of flight mass spectrometry (GC/HR-TOF MS) was used as an analytical tool. A comparison of optimal extraction of metabolites was carried out with a number of solvents [hexane, methylene chloride, methanol, ethanol, methanol: ethanol (70:30, v:v)]. In extracts with methanol: ethanol (70:30 v:v) were detected higher amounts of metabolites than with other solvents. Principal component analysis (PCA) and partial least-squares discriminant analysis (PLS-DA) plots showed significant differences between the diploid and tetraploid metabolite profiles. Extracts of tetraploid showed higher amounts of amino acids, while extracts of diploid contained more organic acids and sugars.

The Journal of Biomedical Discovery and Collaboration was created to provide, for the first time, a unified forum to consider all factors that affect scientific practice and scientific discovery – with an emphasis on the changing face of contemporary biomedical science. In this endeavor we are bringing together three different groups of scholars: a) laboratory investigators, who make the discoveries that are the currency of the scientific enterprise; b) computer science and informatics investigators, who devise tools for data analysis, mining, visualization and integration; and c) social scientists, including sociologists, historians, and philosophers, who study scientific practice, collaboration, and information needs. We will publish original research articles, case studies, focus pieces, reviews, and software articles. All articles in the Journal of Biomedical Discovery and Collaboration will be peer reviewed, published immediately upon acceptance, freely available online via open access, and archived in PubMed Central and other international full-text repositories.  相似文献

The precise level of environmental control in vitro may aid in identifying genetically superior plant germplasm for rooting characteristics (RC) linked to increased foraging for plant nitrogen (N). The objectives of this research were to determine the phenotypic variation in root morphological responses of 49 Solanum chacoense (chc), 30 Solanum tuberosum Group Phureja – Solanum tuberosum Group Stenotomum (phu-stn), and three Solanum tuberosum (tbr) genotypes to 1.0 and 0.5 N rate in vitro for 28 d, and identify genotypes with superior RC. The 0.5 N significantly increased density of root length, surface area, and tips. All RC were significantly greater in chc than in either phu-stn or tbr. Based upon clustering on root length, surface area, and volume, the cluster with the greatest rooting values consisted of eight chc genotypes that may be utilized to initiate a breeding program to improve RC in potato.  相似文献

N-halamines serve as important antimicrobial agents. Development of this class of compounds has been shown to provide benefits especially from a biocidal point of view. A novels-triazine-basedN-heterocycle, dichloro-m-aminophenyl-hydantoinyl-s-triazine (DAPHT), which could be rendered antimicrobial through exposure to diluted chlorine bleach, was synthesized and characterized by1H NMR,13C NMR, and FT-IR. A finishing method was used to apply theN-halamine precursor onto cotton fabric, and the optimum conditions for finishing were investigated. The DAPHT-treated cotton fabric had durable antimicrobial properties up to 50 standard washing cycles and was rechargeable under normal laundry/bleaching conditions. The antimicrobial efficacy against Gram-positive and Gram-negative bacteria was demonstrated.  相似文献

Background

The drivers of species co-existence in local communities are especially enigmatic for assemblages of morphologically cryptic species. Here we characterize the colonization dynamics and abundance of nine species of Caenorhabditis nematodes in neotropical French Guiana, the most speciose known assemblage of this genus, with resource use overlap and notoriously similar external morphology despite deep genomic divergence.

Methods

To characterize the dynamics and specificity of colonization and exploitation of ephemeral resource patches, we conducted manipulative field experiments and the largest sampling effort to date for Caenorhabditis outside of Europe. This effort provides the first in-depth quantitative analysis of substrate specificity for Caenorhabditis in natural, unperturbed habitats.

Results

We amassed a total of 626 strain isolates from nine species of Caenorhabditis among 2865 substrate samples. With the two new species described here (C. astrocarya and C. dolens), we estimate that our sampling procedures will discover few additional species of these microbivorous animals in this tropical rainforest system. We demonstrate experimentally that the two most prevalent species (C. nouraguensis and C. tropicalis) rapidly colonize fresh resource patches, whereas at least one rarer species shows specialist micro-habitat fidelity.

Conclusion

Despite the potential to colonize rapidly, these ephemeral patchy resources of rotting fruits and flowers are likely to often remain uncolonized by Caenorhabditis prior to their complete decay, implying dispersal-limited resource exploitation. We hypothesize that a combination of rapid colonization, high ephemerality of resource patches, and species heterogeneity in degree of specialization on micro-habitats and life histories enables a dynamic co-existence of so many morphologically cryptic species of Caenorhabditis.

Background

The brown planthopper (BPH) has become the most destructive and a serious threat to the rice production in Asia. Breeding the resistant varieties with improved host resistance is the most effective and ecosystem-friendly strategy of BPH biological management. As host resistance was always broken down by the presence of the upgrading BPH biotype, the more resistant varieties with novel resistance genes or pyramiding known identified BPH resistance genes would be needed urgently for higher resistant level and more durability of resistance.

Results

Here, we developed near isogenic lines of Bph9 (NIL-Bph9) by backcrossing elite cultivar 93–11 with Pokkali (harboring Bph9) using marker-assisted selection (MAS). Subsequently, we pyramided Bph6 and Bph9 in 93–11 genetic background through MAS. The resulting Bph6 and Bph9 pyramided line LuoYang69 had stronger antixenotic and antibiosis effects on BPH and exhibited significantly enhanced resistance to BPH than near isogenic lines NIL-Bph6 and NIL-Bph9. LuoYang69 derived hybrids, harboring heterozygous Bph6 and Bph9 genes, also conferred high level of resistance to BPH. Furthermore, LuoYang69 did not affect the elite agronomic traits and rice grain quality of 93–11. The current study also developed functional markers for Bph9. Using functional dominant marker, we screened and evaluated worldwide accessions of rice germplasm. Of the 673 varieties tested, 8 cultivars were identified to harbor functional Bph9 gene.

Conclusion

The development of Bph6 and Bph9 pyramided line LuoYang69 provides valuable resource to develop hybrid rice with highly and durable BPH resistance. The development of functional markers will promote MAS of Bph9. The identified Bph9 containing cultivars can be used as new sources for BPH resistance breeding programs.

Low molecular weight (LMW-GS) and high molecular weight glutenin subunits (HMW-GS) were added to a base flour using both «addition» and «incorporation» protocols. «Incorporation» of glutenin subunits into the glutenin network of the base flour was performed by partial (reversible) reduction and subsequent reoxidation of the glutenin network in the presence of the added glutenin subunits whereas, in the «addition» protocol, glutenin subunits were added without reduction/oxidation. The effects of both «addition» and «incorporation» of alkylated and unalkylated LMW-GS and HMW-GS on dough extension parameters maximum resistance (MR) and extensibility (EX) were compared and thoroughly discussed. HMW-GS and LMW-GS had totally different effects on dough extensibility. «Addition» of LMW-GS significantly decreased both MR and EX whereas HMW-GS caused a significant increase in MR. «Incorporation» of LMW-GS caused a decrease in MR whereas HMW-GS clearly increased MR. The similarity in effects obtained with «addition» and «incorporation» of glutenin subunits indicated that, even with «addition», glutenin subunits can be partially incorporated into the glutenin network in the presence of oxygen. Alkylated and unalkylated glutenin subunits had different effects. This was probably caused by the effect of free sulphydryl groups in unalkylated subunits (possibility of SS/SH exchanges and/or incorporation) and/or the effect induced by introduction of alkylated derived substituents. A protocol for «incorporation with excess KIO3» was developed to exclude the possible effect of a lowering of the available oxidant concentration by oxidation of free sulphydryl groups in glutenin subunits. However, the use of high levels of oxidant in the «incorporation with excess KIO3» protocol seems to overrule the effects of added glutenin subunits or may force glutenin subunits to incorporate differently from what can be observed under gentle oxidation conditions. Therefore, «incorporation with excess KIO3» is not suitable for studying the effects of incorporation of glutenin subunits on dough extensibility.  相似文献

Background  

Acacia tortilis is a keystone species across arid ecosystems in Africa and the Middle East. Yet, its life-history, longevity and growth are poorly known, and consequently ongoing changes in tree populations cannot be managed in an appropriate manner. In other arid areas parenchymatic bands marking growth zones in the wood have made dendrochronological studies possible. The possibilities for using pre- and post-bomb 14C content in wood samples along with the presence of narrow marginal parenchymatic bands in the wood is therefore tested to gain further insight into the age, growth and growth conditions of A. tortilis in the hyper-arid Eastern Desert of Egypt.  相似文献

Turmeric extracts, among which curcumin and bis-demethoxycurcumin, are by far known for their therapeutic activities. In this study we propose easy and low cost synthetic pathways in order to obtain glucosyl-curcuminoids, safe and water soluble potential drugs and dyes, which may be implied in different fields ranging from pharmacology to food chemistry. The prebiotic effect of different concentrations of inulin (0, 1 and 2%) on the growth and survival of Lactobacillus plantarum (LP) CECT 220 in blended carrot and orange juices was investigated after 24 h of fermentation, during 30 days of storage at 4 °C and through the phases of gastrointestinal digestion after different storage periods. Microbiological and chemical determinations were also carried out in all juices. The lactic fermentation increased the shelf life of the fermented juices with inulin. The hygienic-sanitary quality in fermented juices was better than the control juices. During storage, the inulin improved the viability of LP and the monosaccharide concentration remained higher with respect to the juice without inulin (40% lower). At 30 days, the fermented juices with 2% inulin after in vitro digestion presented the highest survival of L. plantarum.  相似文献

Vietnam is one of the world’s top two rice exporting countries. However, rice cultivation is the primary source of agriculture’s greenhouse gas (GHG) emissions in Vietnam. In particular, strategies are required to reduce GHG emissions associated with the application of organic and inorganic fertilisers. The objective of this study was to assess the effects of various combinations of biochar (BIOC), compost (COMP) and slow-release urea (SRU) on methane (CH4) and nitrous oxide (N2O) emissions. In total, 1170 gas samples were collected from closed gas chambers in rice paddies at Thinh Long commune and Rang Dong farm in northern Vietnam between June and October 2014. The gas samples were analysed for CH4-C and N2O-N fluxes using gas chromatography. The application of BIOC alone resulted in the lowest CH4 emissions (4.8–59 mg C m?2 h?1) and lowest N2O emissions (0.15–0.26 µg N m?2 h?1). The combined application of nitrogen–phosphorus–potassium (NPK) + COMP emitted the highest CH4 (14–72 mg C m?2 h?1), while ½NPK + BIOC emitted the highest N2O (1.03 µg N m?2 h?1 in the TL commune), but it was the second lowest (0.495 µg N m?2 h?1) in the RD farm. Green urea and orange urea reduced N2O emissions significantly (p < 0.05) compared to white urea, but no significant differences were observed with respect to CH4 emissions. SRU fertilisers and BIOC alone measured the lowest greenhouse gas intensity, i.e. <2.5 and 3 kg CO2 eq. kg?1 rice grain, respectively. Based on these results, application of fertilisers in the form of BIOC and/or orange or green urea could be a viable option to reduce both CH4 and N2O emissions from rice paddy soils.  相似文献

Present paper reports a method of preparing polymer composite electrolyte nanofiber mat using polyvinyl alcohol (PVA), ammonium thiocynate (NH4SCN) salt, and aluminium oxide (Al2O3) nano particles based on electrospinning technique. Two-stage process of preparation of nanofibers, namely, preparation of nano particles filled PVA electrolyte gel solution followed by its electrospinning has been used. The so obtained nanofibers have been characterized by XRD, DSC, SEM, and Conductivity measurements. XRD patterns affirm the formation of nanocomposite while SEM pictures reveal formation of fibers on a nano scale format (300–800 nm). Fibers of the electrolytes are seen to be thermally stable. Ionic conductivity of electrolyte fiber is seen to improve in the presence of nano filler at room temperature with a maximum at 5.31×10−3 Scm−1 for 4 wt% filler concentration, which is comparable to that for corresponding dried gel electrolyte films.  相似文献

This study, we synthesized graphite-nanoflakes (GNFs) by acid treatment and thermal shock and then using the ultrasonic irradiation technique to exfoliate flake-carbon. The SrFe12O19 nanoparticles (NPs) were coated by co-precipitate method on GNFs after by alkaline treatment. Finally nanocomposite (GNF/SrFe12O19/PTh) was prepared by in-situ oxidative polymerization method in presence of thiophene (Th) as monomer. The magnetic and electrical conducting properties of the resulting nanocomposites were measured by using vibrating sample magnetometer and standard four-point-probe method, respectively. The synthesized nanocomposites were characterized by X-ray diffraction (XRD) and fourier transform infrared spectra (FTIR). In addition, morphological analyses were investigated by scanning electron microscopy (SEM). A minimum reflection loss (RL) of GNFs/SrFe12O19/PTh with 50 % wt GNFs/SrFe12O19 as core were observed ?28 and ?39 dB at 9.7 and 12 GHz for a 1.5 mm thickness. The results indicated that we can perform good microwave shielding in X-band (8–12 GHz) by these nanocomposites.  相似文献

Pollen protoplasts provide a sexual and haploid system for haploid production, cell fusion and mutation studies used in plant improvement. Due to the multiploidy, heterozygosity, and often self-incompatibility in tetraploid genotypes, haploid potatoes are desirable for breeding schemes via ploidy manipulations. In this study, two tetraploid varieties and two dihaploid lines of potato were used for pollen tetrad protoplast isolation and culture. The meiotic tetrad buds were first pre-treated at 5 °C for 0–12 days, then the tetrads were transferred into enzyme solutions containing different concentrations of snailase (0.5–1.5%), 0.3 M osmolites (sucrose, mannitol, glucose or sorbitol), 1.0% Cellulose, 0.5% Hemicellulase, 0.5% Pectolyase, 0.3% Sucrose, 3 mM 2-(N-Morpholino) ethane sulfonic acid, 1% polyvinyl pyrrolidone, 0.01% casein hydrolysate and K3 medium compositions. Among the four donor materials, tetraploid cv. Gannongshu No. 3 (‘GNS No.3’) showed the greatest protoplast yield (74.6 ± 2.4%). In this variety, most of the tetrad protoplasts regenerated a cell wall and continued cell divisions were observed when they were inoculated in K3 basic medium supplemented with (0.5–1.0) mg/L 2,4-D + (0.1–0.5) mg/L KT + 0.4 mg/L 6-BA +800 mg/L glutamine +100 mg/L serine. ‘GNS No.3’ also showed the greatest first division frequency (21.6 ± 1.5%) and sustained division to form multicellular structures. The study findings suggested that cultured tetrad pollen protoplasts could reverse the gametophytic developmental pattern programmed in vivo to a sporophytic pathway leading to multicellular microspore-derived colonies.  相似文献

Betacyanins are reddish to violet pigments that can be found in red pitahaya (Hylocereus polyrhizus) and red spinach (Amaranthus dubius). This study investigated the impact of sub-fractionation (solvent partitioning) on betacyanin content in both plants. Characterization of betacyanins and evaluation of their antimicrobial activities were also carried out. Betanin was found in both plants. In addition, isobetanin, phyllocactin and hylocerenin were found in red pitahaya whereas amaranthine and decarboxy-amaranthine were found in red spinach. Sub-fractionated red pitahaya and red spinach had 23.5 and 121.5 % more betacyanin content, respectively, than those without sub-fractionation. Sub-fractionation increased the betanin and decarboxy-amaranthine content in red pitahaya and red spinach, respectively. The betacyanin fraction from red spinach (minimum inhibitory concentration [MIC] values: 0.78–3.13 mg/mL) demonstrated a better antimicrobial activity profile than that of red pitahaya (MIC values: 3.13–6.25 mg/mL) against nine Gram-positive bacterial strains. Similarly, the red spinach fraction (MIC values: 1.56–3.13 mg/mL) was more active than the red pitahaya fraction (MIC values: 3.13–6.25 mg/mL) against five Gram-negative bacterial strains. This could be because of a higher amount of betacyanin, particularly amaranthine in the red spinach.  相似文献

Precedence of microbial colorants can be seen in almost all the industrial sectors viz. food, textile, paper, agriculture, pharmaceutical, and cosmetics. These colorants are gaining popularity due to their salient advantages over synthetic and natural dyes. This study deals with the optimization and extraction of such colorants from Penicillium minioluteum for the purpose of dyeing different protein fibers. Penicillium minioluteum was cultured under different growth conditions to optimize the extracellular color production. The extracellular colorant grown under optimized fermentation conditions (medium-sabouraud; pH-5.6; temperature-15 °C; time-20 days; incubation-static) in 3000 ml haffkine flask showed maximum color or highest optical density of 1.014 at λ max=490 nm. Later on, this colorant was extracted using nbutanol. The remaining mycelial mat obtained after filtration was extracted using different chemical and mechanical procedures to get the intracellular colorant. Highest O.D of 0.897 was recorded at λ max=490 nm when desiccated powdered mycelial mat was extracted with methanol. Dyed silk and leather with the extracellular colorant showed color strength (K/S) as 3.88 and 3.81, respectively, whereas silk and leather dyed with the intracellular colorant showed K/S as 0.56 and 0.55, respectively. The color strength of extracellular dyed samples was found to be three times higher as compared to the samples dyed with the intracellular aqueous colorant of the same optical density. After dyeing, two different shades were obtained viz. deep red with the extracellular colorant and beige with the intracellular colorant on mulberry silk and wet blue goat nappa skin leather. Fastness towards rubbing was found to be good for both the samples. Wash fastness was excellent on silk. Fastness towards light was poor for both silk and leather. Furthermore, the color yield of the extracellular colorant (0.62 %) was found to be approximately five times more than the color yield of the intracellular colorant (0.14 %).  相似文献

The antioxidative activities of water extract (WE) and crude hot-water soluble polysaccharide (PS) from Ficus carica L. fruit were investigated using various assays in vitro, including scavenging abilities on DPPH, superoxide and hydroxyl radicals and reducing power. The immunity activities of PS were evaluated using the carbon clearance test and serum hemolysin analysis in mice. In addition, total phenolics and flavonoids contents were also determined. Both WE and PS have notable scavenging activities on DPPH with the EC50 values of 0.72 and 0.61 mg/ml, respectively. The PS showed higher scavenging activity than WE on superoxide radical (EC50, 0.95 mg/ml) and hydroxyl anion radical (scavenging rate 43.4% at concentration of 4 mg/ml). The PS (500 mg/kg) also has a significant increase in the clearance rate of carbon particles and serum hemolysin level of normal mice. The results indicate that both WE and PS might be applicable in healthy medicine and food industry.  相似文献

We report on the in situ synthesis and stabilization of manganese dioxide (MnO2) onto four different silk yarns (mulberry, tasar, muga and eri silks). A new ultrasound-assisted procedure was used to reduce permanganate (MnO4 ?) and yielded MnO2 nanoparticles (NPs) on/in the different silk fibers. Using a factorial design we assessed the influence of the silk type, manganese precursor concentration, sonication time, and temperature. The results indicated no measurable effect of the process parameters on the silk structures, but significant correlation with the rate of degradation of methylene blue (MB) and the fraction of permanganate consumed. Further optimization of the factorial model identified the optimal process conditions for each silks: mulberry (150 min sonication, 20 mM permanganate), eri (360 min, 10 mM), tasar (150 min, 10 mM) and Muga (20 min, 10 mM). The operational stability (successive catalysis) of the optimum hybrids showed good performance over 5 cycles and most importantly reduced direct dye absorption relatively to dye oxidation. Overall, we found that all silks could template the formation and stabilization of different MnO2 polymorphs and yielded catalytic instead of stoichiometric hybrid fibers.  相似文献

Fresh button mushrooms (Agaricus bisporus) were harvested and treated with a solution of 1.5% CaCl2 + 0.5% citric acid and stored for 16 days at 12 °C. The effects of this treatment on firmness, weight, color, cell wall compositions (cellulose and chitin) and cell wall degrading enzymes (cel1ulase, beta-1, 3 glucanase, chitinase and phenylalanine ammonialyase) were investigated during post-harvest storage. The expressions of major genes (Cel1, Glu1, Chi1 and PAL1) involved in cell wall degradation during post-harvest storage were also monitored. The results revealed that the post-harvest chemical treatment maintained better firmness, weight, color and inhibited cellulase, beta-1, 3 glucanase, chitinase and phenylalanine ammonialyase activities. These findings showed that the down-regulation of cell wall degrading enzymes is a possible mechanism that delays the softening of button mushrooms by the application of combined chemical treatment.  相似文献
