改良环介导等温扩增技术快速检测转基因大豆 总被引:6,自引:0,他引:6
采用改良环介导等温扩增(LAMP)技术,建立一套准确、快速、可靠的用于转基因大豆检测及DNA提取的方法.以CP4-EPSPS外源基因为检测的目的片段,设计内、外引物和环引物,并采用改进的方法提取DNA,进行LAMP扩增,实际检测已知转基因大豆和市售大豆,通过肉眼观察白色沉淀,判断检测结果.改进的DNA提取方法与经典的CTAB方法相比,提取时间至少缩短了1 h,降低了提取成本.环介导等温扩增(LAMP)技术对转基因大豆的检出限为0.01%,是普通PCR方法的20倍.因此,改良的LAMP检测转基因大豆方法灵敏度高,耗时短,方法简便. 相似文献

柑橘黄龙病可视化LAMP检测技术的建立及应用 总被引:2,自引:0,他引:2
建立柑橘黄龙病(HLB)可视化环介导等温扩增(LAMP)检测方法,为HLB的快速检测提供有力的技术支持。根据LAMP技术原理,针对柑橘黄龙病菌16S rDNA靶序列的6个位点设计4条特异性引物,通过对反应体系和反应条件的优化,并在反应前的体系中加入1∶10的钙黄绿素(calcein)和氯化锰(MnCl2)混合液作为荧光显色剂,建立可视化LAMP检测技术,同时对该技术的特异性、灵敏度和样品检测进行了测试。该检测方法具有很高的特异性,反应体系中除了HLB具有特异性的扩增,产生黄绿色荧光扩增产物,其他供试菌株均无特异扩增。对柑橘黄龙病菌总DNA的检测限为1.51×10-3 ng/μL,而对含有目的基因的CG-pMD18-T质粒DNA的检测限为2.12×10-6 ng/μL,与定量PCR灵敏度相当,而比常规PCR灵敏度高1 000倍。利用可视化LAMP技术和定量PCR对来自漳州、南平、福州等地的样品进行检测,结果 2种技术的检出率均为76.7%,符合率达到100%。 相似文献

以柑桔黄龙病(Citrus Huanglongbing,HLB)病原菌16s rDNA保守区域为靶标设计LAMP引物,建立柑桔黄龙病的环介导等温核酸扩增(loop-mediated isothermal amplification assay,LAMP)检测技术。LAMP检测方法具有极高的检测特异性和灵敏性,其检测下限约为1 pg/μL质粒DNA,是PCR检测灵敏度的100倍,能快速、准确地对田间疑似样品进行检测。建立的柑桔黄龙病LAMP检测方法是对黄龙病检测方法的拓展和延伸,可以很好地应用于田间检测,可满足基层以及科研单位对该病害检测的需要。 相似文献

We have evaluated 145 covered and 29 hulless barley genotypes artificially infected with Fusarium graminearum Schw. in a field experiment. Significant genetic variation for Fusarium head blight severity measured by the area under the disease progress curve and for mycotoxin contamination was found among covered and hulless barley lines. The overall mean area under the disease progress curve of covered and hulless barley did not differ significantly. Twenty-nine hulless-covered pairs with approximately the same disease severity were chosen for analysis of mycotoxins (B-trichothecenes) in the harvested grain. The average content of deoxynivalenol of the 29 covered and 29 hulless barley samples was 15,520 and 12,964 μg kg−1, respectively. The mean deoxynivalenol, 3-acetyl-deoxynivalenol and 15-acetyl-deoxynivalenol content of covered barley was significantly higher than that of hulless barleys (p<0.01), whereas for nivalenol there was no significant difference between the mean values of covered and hulless barley. Hulless barley appears to be less prone to accumulation of mycotoxins in the harvested crop than covered barley most likely because a considerable proportion of the mycotoxin resides in the barley hulls. 相似文献

为了探明不同类型大麦品种籽粒中酚酸类化合物含量的差异,采用超声辅助提取、高效液相色谱测定的方法对15个大麦品种(系)籽粒中的13种酚酸含量进行了比较分析。结果表明,大麦籽粒中的酚酸类化合物主要是羟基苯甲酸衍生物,羟基肉桂酸衍生物含量较少。不同类型大麦品种籽粒中酚酸类化合物的含量存在极显著的差异,黑皮大麦籽粒中总羟基苯甲酸衍生物(THBA)、总羟基肉桂酸衍生物(THCA)和总酚酸(TPA)的含量均高于非黑皮大麦;二棱大麦籽粒中THBA、THCA和TPA的含量均高于多棱大麦;皮大麦籽粒中THCA和TPA的含量高于裸大麦,但THBA的含量与裸大麦无显著差异。另外,与裸大麦相比,皮大麦籽粒中含有更为丰富的酚酸种类。 相似文献

利用高通量测序分析明确玉米黑粉菌菌瘤内菌粉培养液中内生细菌种类,结合16SrDNA分析,分离并鉴定在培养液中占比最高的菌株。以溶菌酶及Amp处理玉米黑粉菌冬孢子为对照,显微镜下观察菌瘤内内生细菌对玉米黑粉菌冬孢子萌发的影响。结果表明,在4份样品的菌粉培养液中,肠杆菌属细菌的占比在90%以上。当肠杆菌大量增殖时对黑粉菌的冬孢子萌发产生影响,可引起黑粉菌的冬孢子发生质壁分离,释放原生质体,部分原生质体再生成菌体;也可以使冬孢子壁直接被降解,原生质体萎缩。研究发现,玉米黑粉菌菌瘤内的内生细菌以肠杆菌为主,该内生细菌对黑粉菌的冬孢子萌发产生影响。 相似文献

为筛选特色淀粉青稞材料,采用双波长法测定和分析了来自西藏、青海、甘肃、四川、国外的栽培青稞以及野生裸大麦共469个青稞材料的直链和支链淀粉含量。结果表明,这些青稞材料的直链淀粉含量平均为24.65%,变异范围为4.09%(昆仑8号)~39.72%(长芒红四棱),其中西藏材料最高(26.87%),甘肃材料最低(21.23%);支链淀粉含量平均为31.76%,变异范围为15.24%(康青1号)~60.09%(喜玛拉雅2号),其中野生裸大麦最高(35.51%),西藏材料最低(30.17%);总淀粉含量平均为56.00%,变异范围为31.28%(阿青5号)~74.41%(阿里当地青稞),以野生裸大麦最高(58.11%),青海材料最低(54.19%)。根据淀粉含量分布特征将不同来源青稞材料间直、支链及总淀粉含量都分成4个级别,这些青稞材料的直链淀粉含量主要分布在第3级,支链和总淀粉含量主要分布在第2级。从中筛选出18份具有特殊淀粉含量的青稞材料,可用于专用青稞的培育。 相似文献

为了明确产地和品种对西藏青稞营养品质的影响,对采自西藏6个地(市)、26个县的7个普通青稞品种和3个有色青稞品种(257份样品)的5个品质指标和8种矿质元素含量进行了分析。结果表明,(1)产地生态条件是影响青稞品质和矿质元素含量的首要因素,产地对普通青稞的10个被测指标有显著影响;品种对普通青稞的5种被测指标有显著影响;产地和品种的交互作用显著影响5个被测矿质元素含量;产地显著影响有色青稞的黄酮、粗蛋白和磷含量。(2)青稞不同品质指标和矿质元素含量变异程度不同,变异系数最高为52.3%。供试青稞粗淀粉含量为46.64%~53.76%,粗蛋白含量为6.51%~14.21%,粗纤维含量为1.66%~3.4%,β-葡聚糖含量为3.63%~5.13%,黄酮含量为0.18%~0.40%;钾含量为4 224.8~7 646.5mg·kg~(-1),磷含量为2 243.1~6 288.7mg·kg~(-1),镁含量为731.6~1 572.0mg·kg~(-1),钙含量为246.4~789.0mg·kg~(-1),铁含量为27.59~173.99mg·kg~(-1),锌含量为17.56~85.22mg·kg~(-1),锰含量为10.42~22.23mg·kg~(-1),铜含量为2.12~11.84mg·kg~(-1)。(3)有色青稞的粗淀粉和黄酮含量显著高于普通青稞,而粗蛋白和粗纤维含量显著低于普通青稞,β-葡聚糖含量二者间无显著差异。 相似文献

Four newly developed huless barley cultivars with altered carbohydrate traits were developed at the Crop Development Centre (CDC), University of Saskatchewan which varied for amylose (1–40% DM), and β-glucan (5–10% DM) content. The four hulless barley cultivars include zero-amylose waxy, CDC Fibar; 5%-amylose waxy, CDC Rattan; normal-amylose, CDC McGwire and high-amylose, HB08302. CDC Copeland was also included as a hulled barley control. Our previous study revealed the altered carbohydrate traits in barley affected nutrition availability and rumen degradation kinetics. These biological differences may be caused by differences in their molecular structure features. The objectives of this study were to (1) reveal molecular structure features in the four hulless barley cultivars with altered carbohydrate traits, and (2) quantify the molecular structural features in relation to rumen degradation kinetics, intestinal nutrient digestion and predicted protein supply to dairy cattle. Conventional Molecular Infrared-vibration Spectroscopy was applied to detect biochemical characteristics of functional group bands including protein, non-starch carbohydrate (β-glucan and cellulosic compounds) and total carbohydrate. Spectral data were further analyzed using univariate analysis of recording absorption peak parameters (baseline, region, relative height and area) and also multivariate analysis with agglomerative hierarchical cluster analysis and principal components analysis. Although correlation results showed weak correlation (P < 0.05) between identified functional group bands and ruminal degradation kinetics and estimated protein supply from both models, molecular structure differences of hulless barley cultivars can still be detected by the Molecular Infrared-vibration Spectroscopy technique and potential truly protein supply (MP) was significantly affected (P < 0.05) by protein molecular structure characteristics in hulless barley cultivars. 相似文献

为了解西藏青稞的染色体多样性,从西藏六个地区共采集124个青稞品种,每个品种取3个单株,基于南京农业大学开发的寡核苷酸探针ONPM#8(Oligonecleotide probe multiplex#8),利用荧光原位杂交(fluoresence in situ hybridization, FISH)技术对所有品种进行染色体鉴定,并建立可以区分全部染色体的青稞参考核型。结果表明,采集到的青稞植株有26种染色体多态类型,其中5种多态类型(1H-1、2H-2、4H-1、5H-1和7H-4)的频率大于50%。染色体多态性信息含量(PIC)分析发现,3H染色体最多(PIC=0.75),5H染色体最少(PIC=0.00)。372个单株存在明显的核型变异,涉及染色体多态性、杂合性和异质性,其中261个单株为纯合类型,其余111个单株为杂合类型,平均每株包含1.15对杂合染色体,说明新采集的青稞涉及染色体多样性,可通过后续个体选择和后代鉴定选育出纯系品种。 相似文献
