Microscopic examination showed the cell wall decay pattern produced by the brown-rot fungus Coniophora puteana to be different from the degradation pattern known to be typical for brown-rot fungi. Erosion and thinning of cell walls in patterns considered to be characteristic of white-rot decay were observed. In particular, the fungal strain COP 20242 degraded secondary cell wall layers extensively, and also degraded lignin-rich middle lamellae. Some strains of C. puteana produced soft-rot type cavities in the S2 layer. All strains of C. puteana employed in the present work showed a positive reaction to tannic acid in the Bavendamm test, indicating the production of laccase. Microscopic and enzymatic studies provided evidence to suggest that the wood decay by C. puteana is unique both in terms of micromorphological and enzymatic patterns of cell wall degradation. This is because brown-rot fungi are not generally known to form cavities in the cell walls or to produce lignin-degrading enzymes. These observations suggest that lignin degradation capacity of brown-rot fungi may be greater than previously considered.  相似文献

 Conditions for cell growth of suspension cultures of Cupressus lusitanica, which has high β-thujaplicin productivity, were studied. The medium that provided the highest growth rate was IS-1 medium (pH 5.5), modified from Gamborg B5 medium containing 32 mM of total nitrogen. Its NO3-N/NH4-N ratio was 30 : 2. The maximum growth represented a 25-fold increase over the initial biomass on a fresh weight basis after 30 days of culture in this medium. The highest cell growth was obtained with an initial pH of 3.5–5.5, but the pH of the medium settled to about pH 4.0 from any of the initial pH values in this report. The cells cultured under this condition were able to produce a high level of β-thujaplicin. To investigate the linkage types between carbohydrates and lignin, residual lignins were isolated from three different unbleached pulps [kraft, alkaline sulfite anthraquinone methanol (ASAM), and soda with anthraquinone (AQ) and methanol] of spruce and beech wood and then characterized by oxidation with 2,3-Dichloro-5,6-dicyanobenzoquinone and followed by Prehm’s methylation. In residual lignins, sugar moieties were bound to lignins via benzyl ether bonds. In particular, galactose and mannose are predominantly linked to lignin fragments in residual lignins of spruce wood, while xylose and galactose are favored in the formation of LC bonds in the residual lignins of beech wood. In the case of hexoses, primary hydroxyl groups (C6 position) preferentially take part in benzyl ether linkages. Hydroxyl groups in the C2 and C3 positions of xylose participate in LC bonds and a small portion of arabinose was notably connected to lignin via the C5 position. Approximately seven or eight sugars were connected in soda/AQ/methanol residual lignin per 100 C9 lignin units, while the frequencies of LC bonds in kraft and ASAM residual lignins were distinctively less at one to three sugars per 100 C9 lignin units. Any means helpful for the promotion of termite feeding activity has potential for use in a matrix in termite bait application. Therefore, energy transfer by gamma irradiation is worthy of consideration for converting wood into termite-accessible material. Wood specimens gamma-irradiated at 100 kGy and at lower levels were tested for their degrees of polymerization (DP) of cellulose and biological resistance. The DP of cellulose adversely decreased with increased doses of gamma irradiation. Termite wood consumption rates, which were determined by laboratory tests using undifferentiated larvae (workers) of Coptotermes formosanus Shiraki, were significantly higher at 100 kGy than at other doses. On the other hand, the decay resistance of gamma-irradiated wood against the fungi Fomitopsis palustris (Berkeley et Curtis) Murrill and Trametes versicolor (L. ex Fr.) Quel did not vary by irradiation dose.  相似文献

A new approach is proposed for the evaluation of the brittleness of heat-treated Styrax tonkinensis wood. Heat treatment made wood more brittle when wood was heated at a higher temperature or for a longer time. The brittleness increased to four times that of the control when wood was heated at 200°C for 12 h. For treatment at 160°C, the increase in brittleness without any change in weight is thought to be possibly caused by the relocation of lignin molecules. At higher temperatures, loss of amorphous polysaccharides due to degradation is thought to become the main factor affecting brittleness. The crystallites that were newly formed after 2 h of treatment showed brittleness that was different from that of the inherent crystallites remaining after 12 h of heat treatment. This inherent crystalline cellulose possibly plays a role in brittleness. There is also the possibility of using color to predict the brittleness of heat-treated wood.  相似文献

We examined the relationships between the absorptional characteristics in the near infrared region and the chemical changes of decomposing beech (Fagus crenata) and pine (Pinus densiflora) litters. Spectra as well as the concentrations of chemical substances approached each other and converged with decomposition, although both initial characteristics differed markedly between beech and pine. This indicated that the fundamental chemical structures were almost the same, although their organochemical composition differed. Specific absorption bands for lignin, polysaccharide, and protein were identified at 2,140 and 1,670 nm, 2,270, 1,720, 1,590, and 1,216 nm, and 2,350 nm, respectively. Absorbance at 1,670 nm, peculiar band of aromatics, showed a positive correlation with lignin concentration, which suggested the relative increment of aromatics due to condensed lignin in decomposing litters. Absorbance at 2,140 nm, characterized as the C–H bond in HRC = CHR, showed a negative correlation with lignin concentration, which suggested the decrements of some structures such as side-chains in lignin polymers unrelated to aromatics. Absorbance at 2,270, 1,720, and 1,216 nm, specified to O–H/C–O/C–H bonds in saccharide, might reflect the change of polysaccharide during decomposition because they showed a positive correlation to polysaccharide concentration. In the same way, absorbance at 2,350 nm, identified to the C–H/CH2 bonds in protein, showed a negative correlation to nitrogen concentration in decomposing litters, which might indicate that the C–H/CH2 bonds in protein decreased with decomposition due to microbial consumption of carbon in protein. Our findings suggested the possibility that the spectral changes indicate the litter digestibility during decomposition and that also explain the compositional change in decomposing litters.  相似文献

Heat treatment of Pinus pinaster and Eucalyptus globulus woods, two important species in Portugal, was performed in the absence of air by steaming, inside an autoclave, for 2–12 h at 190–210°C. Mass losses increased with treatment time and temperature reaching 7.3% for pine and 14.5% for eucalypt wood. The wood behaviour with moisture was improved. The equilibrium moisture content decreased by 46% for pine and 61% for eucalypt, the dimensional stability increased (maximum anti-shrinking efficiency in the radial direction of 57 and 90% for pine and eucalypt, respectively) and the surface wettability was lowered. In relation to mechanical properties, the modulus of elasticity was little affected (maximum decrease of 5% for pine and 15% for eucalypt) but the bending strength was reduced (by 40% at 8% mass loss for pine and 50% at 9% mass loss for eucalypt wood). The variation of properties was related to treatment intensity and mass loss but significant improvements could already be obtained for a 3–4% mass loss without impairing the mechanical resistance. The response of eucalypt was higher than that of pinewood. Heat treatment of eucalypt wood shows an interesting potential to improve the wood quality for solid timber products.  相似文献

To compare the responses to repeated flooding and drought of Salix gracilistyla, which grows on coarse gravel substrates, and Salix subfragilis, which grows on fine silt or clay substrates, we measured pre-dawn leaf water potential (Ψw pd), osmotic adjustment (Ψw tlp, Ψo sat), and biomass production of cuttings under greenhouse conditions. The experimental design involved a control and four treatments that crossed 1 or 3-week flooding (F) with 1 or 2-week droughts (D). Ψw pd was reduced after 2 weeks of drought when preceded by 1 week of flooding. Neither species increased osmotic adjustment in response to increased duration of drought between repeated 3-week flooding. Moreover, a decrease in the ratio of leaf biomass to total biomass or an increase in the ratio of root biomass to total biomass with longer drought repetitions was not observed for either species. The root ratio of S. gracilistyla was more strongly inhibited by flooding than that of S. subfragilis. The shoot-to-root ratio of S. subfragilis was higher than that of S. gracilistyla in all F combinations. The hypertrophied lenticel ratio of S. gracilistyla after 1 week of flooding was nearly the same as that after 3 weeks of flooding, whereas values for S. subfragilis after 1 week of flooding were lower than those after 3 weeks of flooding. The low allocation to roots and the generation of hypertrophied lenticels by S. gracilistyla in response to flooding, as compared with S. subfragilis, seem to be related to the different habitat substrate conditions of the two species.  相似文献

We examined the saccharification and fermentation of meals from Acacia mangium wood, Paraserianthes falcataria wood, and Elaeis guineensis trunk. The levels of enzymatic hydrolysis of cellulose and ethanol production were highest for P. falcataria wood and lowest for A. mangium wood. Ultrasonication pretreatment of meal further increased the rates of hydrolysis and ethanol production in meal from P. falcataria wood. Through this pretreatment, hemicelluloses (xylan and xyloglucan) and cellulose were released in the meal from P. falcataria wood. Loosening of hemicellulose associations can be expected to make P. falcataria wood more useful for bioethanol production.  相似文献

UGPase gene related with wood cellulose synthesis was transferred into C. acuminata using the method of Agrobacte- rium-mediated genetic transformation, and an efficient transformation system was developed for C. acuminata on the basis of evaluations of several factors affecting Agrobacterium-mediated DNA transfer rate. The highest transformation rate was achieved when pre-cultttred leaf explants were infected with an Agrobacterium culture corresponding to OD600 (0.5) for 10 min, and cultured on explant regeneration medium for three days. The results of Southern hybridization showed that genomic DNA of the kanamycin-resistant shoots to an UGPase gene probe substantiated the integration of the transgene. Transformation efficiency (6%) was achieved under the optimized transformation procedure, This system should facilitate the introduction of important useful genes into C, acuminata.  相似文献

The fresh pollen vitality,the effect of different storage conditions on the pollen vitality,and the difference of vitality among the species of seven species of Syringa were determined in Shenyang,China.The results indicated that the pollen vitality (81.5%) of Syringa villosa was the highest among the seven tested species,followed by S.microphylla and S.meyeri,and that of S.oblata var.affinis was the lowest.The low temperature was the best condition for storage of pollen of Syringa,and the most proper temperature for the storage was 0-2℃.The storability of S.microphylla was the best of all,and it could be stored over 60 days at the temperature of 0-2℃,next was S.villosa and S.meyeri.  相似文献

The aim of this study was to determine changes in crystallinity and crystalline unit cell type of heat-treated Scots pine (Pinus sylvestris L.) and Uludağ fir (Abies nordmanniana stev. subsp. bornmuelleriana Mattf.) wood samples by means of FT-IR spectroscopic method. Heat treatment was applied on the test samples in an oven at three different temperatures (120, 150, and 180°C) and for two different periods of time (6 and 10 h) under atmospheric pressure. It was designated that crystallinity of both Scots pine and Uludağ fir wood samples increased during heat treatment depending on the duration. However, monoclinic structure in crystalline unit cells of Scots pine and Uludağ fir wood samples converted to triclinic structure when heat treated. It was estimated that monoclinic structure was dominant in the crystalline unit cell. It was established that the crystalline structure of Scots pine wood samples was more affected by heat treatment than that of Uludağ fir wood samples.  相似文献

The release of mass propagated egg parasitoids could be one option to control the lepidopteran pests. The efficiency of the parasitoid Trichogramma evanescens (Westwood) reared on eggs of three different factitious hosts; Sitotroga cerealella, Ephestia kuehniella and Galleria mellonella was studied for controlling bollworm Helicoverpa armigera. Efficiency of Trichogramma was studied by measuring parasitism rates, emergence rates, longevity and sex ratio. Wasps reared from each source were tested on the source host and on the target host, H. armigera under laboratory conditions. Rates of parasitism on H. armigera eggs, emergence rates of parasitoids and their longevity were the highest for wasps reared on H. armigera. Wasps reared on S. cerealella gave comparable rates. However, wasps from E. kuehniella gave the lower rates and G. mellonella gave the lowest ones. Parasitized eggs of H. armigera and S. cerealella produced more parasitoid females than eggs of E. kuehniella and G. mellonella. Results are discussed for magnifying efficiency of the parasitoid in controlling H. armigera in the field.  相似文献

Lignins are cell wall phenolic heteropolymers that result from the oxidative coupling of three monolignols bearing p-coumaryl (H), coniferyl (G), and sinapyl (S) units, in a reaction mediated by peroxidases. Here, we report the existence of a relationship between the Σβ-O-4/ΣO-4 end monomer ratio of the linear lignin fraction, released through the specific cleavage of the alkyl ether linkages by thioacidolysis, and the G/S ratio of lignins, when this was estimated in differentially evolved vascular land plants. Most importantly, in the case of angiosperms, Gnetales, and lycopods, the Σβ-O-4/ΣO-4 end monomer ratio was apparently predictable from the proportions at which the G and S units were mixed. In the case of G lignins (present in basal gymnosperms and ferns), the Σβ-O-4/ΣO-4 end monomer ratio decayed exponentially to increase the O-4-linked dihydroconiferyl alcohol (DHCA) content. The results obtained suggest that the Σβ-O-4/ΣO-4 end monomer ratio of the linear lignin fraction depends intimately on the lignin monomer composition, and, therefore, on the chemical nature of the radicals derived from three monolignols (coniferyl, dihydroconiferyl, and sinapyl alcohols), whose gain have been finely tuned during land plant evolution.  相似文献

 Spatial distribution of sporocarps of Suillus pictus A.H. Smith and Thiers was studied in a plot of 6 × 12 m in size established in a stand of Pinus koraiensis Sieb. et Zucc. in Kyoto, Japan for 4 years, and the biomass of mycorrhizas was examined in the last year. S. pictus was dominant in both sporocarp and ectomycorrhizal community in the study plot. The number of S. pictus sporocarps ranged from 0.94 to 1.26 m−2 (surface area) in the study plot and did not vary very much during the study period. Sporocarps of S. pictus occurred in clumps and the distributions of clumps were generally random. As the spots of sporocarp occurrence changed gradually from year to year, the distributions of sporocarps that occurred successively in 2-year periods overlapped, especially when analyzed in 9-m2 unit size using the m * –m method. Mycorrhizas of S. pictus were distributed in more subplots than its sporocarps. Distribution of mycorrhizas and sporocarps of S. pictus generally overlapped well. The biomass of mycorrhizas and mycelia in the mycorrhizas of S. pictus was estimated at 15.5 g DW m−2 and 6.2 g DW m−2 (surface area) in this plot, respectively. The biomass of mycorrhizas and mycelia in the mycorrhizas supporting the production of one sporocarp (average dry weight was 0.86 g) of S. pictus was evaluated as about 16.4 and 7.3 g DW, respectively, in this plot. The decay pattern in bamboo fibers caused by a brown rot fungus, Gloeophyllum trabeum, was examined by microscopy. The inner part of the polylaminate secondary wall was degraded, while the outer part of the secondary wall remained essentially intact. Degradation in bamboo fiber walls without direct contact with the fungal hyphae was similar to wood decay caused by brown rot fungi. Degradation in polylaminate walls was almost confined to the broad layers whereas the narrow layers appeared resistant. The p-hydroxylphenyl unit lignin in middle lamella, particularly in the cell corner regions, was also degraded. The degradation of lignin in bamboo fibers was evidenced by Fourier transform infrared spectra. The present work suggests that the decay of bamboo fiber walls by G. trabeum was influenced by lignin distribution in the fiber walls as well as the polylaminate structures.  相似文献

To determine the effect of shade on morphology, growth and biomass allocation in Picea sitchensis, Larix × eurolepis and Thuja plicata, seedlings were grown in the open or under shadehouses providing 25%, 50% and 75% reductions of full-light for two growing seasons. For most of the characteristics assessed there was no significant interaction between species and shade indicating that the morphological responses to changing shade treatments were not species-dependent. After two growing seasons the mean height increment for the three species was significantly greater in 25% (76.1 cm) and 50% shade (74.9 cm) than in the open (69.5 cm). Root collar diameter increment, shoot, root and total biomass declined significantly with increasing shade while the opposite was true for the height:diameter ratio. In both western red cedar and hybrid larch the shoot:root ratio was significantly greater in the shade while in Sitka spruce this characteristic was not influenced by shade. While all species had significantly greater specific shoot areas in 75% shade than in 0% shade, this trend was particularly pronounced in hybrid larch. In hybrid larch and western red cedar, the normalised specific projected shoot area increased significantly with increasing shade. The opposite trend was observed for Sitka spruce. We conclude that in the main the species studied demonstrated similar shade acclimation responses despite their reported differences in shade tolerance.  相似文献

In Mediterranean pine forests, truffles and mushrooms generate greater profits than any other woodland products. However, there are no studies on Tuber melanosporum Vittad. associated with pines. For this reason, we have carried out a study of this truffle in mountain woods with Pinus sylvestris L. and P. nigra Arnold subsp. salzmannii (Dunal) Franco, in central Spain. Two hundred and eight Tuber melanosporum burns were monitored for 7 years in five different habitats within the same geographical area. An ANOVA test confirmed significant differences in carpophore production. In higher producing habitats, pines were less abundant. We also confirmed that in 433 burns, T. melanosporum was always unequivocally associated with the root base of Quercus or Corylus trees. Similarly, 14 truffle collectors confirmed that they had never found a single burn with carpophore production associated exclusively with pines. Nevertheless, soil analyses indicated that the soil of these pine woods was very favourable to Tuber melanosporum. We therefore conclude that at present Pinus nigra salzmannii and P. sylvestris are of little interest to Tuber melanosporum culture, as they hinder carpophore production. However, this study has also confirmed that Pinus nigra salzmannii and P. sylvestris mycorrhize easily with Tuber melanosporum, both in the laboratory and in natural environments. On this basis, we propose that pines may act as transmitters of T. melanosporum, although they do not induce fruiting. As a result, the commercial cultivation of Pinus nigra salzmannii and P. sylvestris seedlings mycorrhized with Tuber melanosporum is not recommended in truffle culture at the present time.  相似文献

To determine the lifespan of Siebold’s beech (Fagus crenata), a dominant canopy tree species of the cool temperate forests of Japan, we estimated the age of 33 mature beech trees felled by a large typhoon in September 2004 at Karibayama, in southern Hokkaido. The study site lies within one of the northernmost distributions of Siebold’s beech populations in Japan. We counted tree rings from tree disks at a height of 1 m, and diameter growth was measured in annual increments. The sample age revealed two major peaks at 240 and 175 years. The diameter growth was good between 40 and 60 years of age and gradually decreased after 70 years of age. Annual diameter increments were noticeably small in good fruiting years, indicating a large allocation of annual carbon gains to the reproductive organs. Bud scale counts of juvenile trees revealed that the age at which the height reached 1 m was approximately 6–13 years. The lifespan of Siebold’s beech in the Karibayama population was therefore estimated to be approximately 250 years, an estimation that is higher than that previously reported for this species in the northernmost populations.  相似文献

The diversity, spatial patterns and temporal dynamics of dead wood were examined within the near-natural beech forests (Fagus sylvatica) of Serrahn (North-eastern Germany). Data were collected in an 8 ha sample plot and in two permanent plots (0.36 and 0.25 ha) that had been established at the end of the 1960s. The mean volume of dead wood was 94 m3 ha−1, amounting to 14% of the total volume of all trees. The dead wood displayed a large variation in dead wood type, tree size and decay class. Standing dead wood accounted for about one-third of the total dead wood volume. The densities of standing dead trees were about 10% of the densities of the living trees over a wide range of diameters. The overall spatial distribution of dead trees exhibited a random pattern. Among the different dead wood types, standing entire dead trees and uprooted trees deviated from this pattern by displaying a significantly aggregated pattern. In the permanent plots a high mortality of overstorey trees was observed (1.3% year−1) and the average amount of dead wood increased greatly from 2.9 to 111.6 m3 ha−1 over the 35-year observation period. The near-natural beech forests of Serrahn have experienced a long period of low human interference. Nevertheless, our results suggest that the structure and dynamics of dead wood are strongly affected by the last major disturbance events that took place at the end of the Middle Ages. Information about the forest history is therefore a basic requirement when interpreting the results obtained in near-natural forests.  相似文献
