Summary Random amplified polymorphic DNA (RAPD) analysis was carried out to evaluate polymorphism and genetic similarity between 39 Mediterranean mandarin genotypes. One hundred eleven amplification products were identified using 21 random primers. An average of 2.2 RAPD markers was obtained for each primer, corresponding to 42% of the amplification products. Genotype-specific RAPD markers were also found, mainly in known hybrids. UPGMA cluster analysis revealed the low level of genetic variation between accessions of Mediterranean mandarins, whereas their hybrids with other Citrus species showed greater genetic dissimilarity. Twenty accessions yielded very similar patterns, suggesting either that they could be a single clone, or that the technique was not able to detect genomic variation. However, for the other specimens genetic polymorphism can easily be detected by RAPD, although the genetic variation between accessions was quite low. The large number of hybrids and the low polymorphism between accessions support the hypothesis that Mediterranean mandarins are all true hybrid of Common mandarins (Citrus reticulata Blanco).  相似文献

RAPD markers were used to evaluate genetic similarity among 35 mandarin accessions, including 10 species and 7 hybrids. One octamer and twenty-two decamer primers produced 109 RAPDs, 45 of which were polymorphic. Jaccard coefficient was used to calculate genetic similarity, and UPGMA to generate the phenogram. The RAPDs obtained were sufficient to generate some accession-specific markers, and to separate these accessions by clustering them into several groups, many of them according to Tanaka's or Webber's systematic units. The genetic similarity within the mandarin group is high (GJ = 0.77), and suggests that cultivated mandarins have a narrow genetic base. The genetic similarity of mandarins to other true citrus species (Citron [C. medica L.] and Pummelo [C. grandis Osbeck]) was much lower (minimum GJ = 0.27). We propose that the mandarin group is a single species, C. reticulata Blanco, composed of several genetically different individuals and a great number of hybrids, rather than a large number of species as proposed by some taxonomic studies. Mandarins suffer from short ‘flavor-life’ compared with other citrus species. The recommended minimum safe temperature for mandarin storage is 5-8 °C. However, because of continuing reductions in permitted chemical residues and increasing concern regarding decay development, mandarins are often shipped at much lower temperatures of 3-4 °C. In the last few years we noticed wide differences in responsiveness of mandarin varieties to chilling, and that the earliest indication of damage was a decrease in flavor acceptability. In the present study, we evaluated changes in flavor and quality of chilling-tolerant ‘Or’ and chilling-sensitive ‘Odem’ mandarins after 4 weeks of storage at 2, 5, or 8 °C followed by 3 days at 20 °C. Low storage temperatures resulted in loss of orange peel color in fruit of both varieties, which became paler and yellowish. The flavor of ‘Or’ mandarins was not affected by different storage temperatures, whereas ‘Odem’ showed severe flavor loss at low storage temperatures. GC-MS analysis of aroma volatiles revealed that changes of storage temperatures had no major effects on aroma volatile contents in ‘Or’ mandarins. However, in ‘Odem’ mandarins, storage at 2 °C caused accumulation of 13 volatiles, mainly terpenes and their derivates, whereas storage at 8 °C resulted in decreases of six volatiles, comprising five terpenes and one terpene derivative. Overall, we conclude that storage temperature is a fundamental factor affecting color and flavor of mandarins, and therefore it is crucial to define the optimal minimum safe temperature for each mandarin variety. Furthermore, massive accumulation of terpenes is most likely the cause for the decrease in flavor acceptability of ‘Odem’ mandarins after storage at low chilling temperatures.  相似文献

Citrus is one of the most cultivated fruits in the world, and satsuma mandarin (Citrus unshiu Marc.) is a major cultivated citrus in Japan. Many excellent cultivars derived from satsuma mandarin have been released through the improvement of mandarins using a conventional breeding method. The citrus breeding program is a lengthy process owing to the long juvenility, and it is predicted that marker-assisted selection (MAS) will overcome the obstacle and improve the efficiency of conventional breeding methods. To promote citrus molecular breeding in Japan, a genetic mapping was initiated in 1987, and the experimental tools and resources necessary for citrus functional genomics have been developed in relation to the physiological analysis of satsuma mandarin. In this paper, we review the progress of citrus breeding and genome researches in Japan and report the studies on genetic mapping, expression sequence tag cataloguing, and molecular characterization of breeding characteristics, mainly in terms of the metabolism of bio-functional substances as well as factors relating to, for example, fruit quality, disease resistance, polyembryony, and flowering.  相似文献

Fluorochrome staining with chromomycin A3(CMA) was used to characterize and compare the CMA banding patterns of chromosomes in 17 accessions of mandarins (Citrus spp.). All accessions had 2n = 18 chromosomes. These chromosomes were classified into six types based on the number and position of CMA positive bands; A: two telomeric and one proximal band, B: one telomeric and one proximal band, C: two telomeric bands, D: one telomeric band, E: without bands, F: one proximal band. Each accession possessed two to five types of chromosomes, and showed unique CMA banding patterns. Type D and E chromosomes were predominant in all accessions. Their numbers were generally higher in Chinese and Indian mandarins. Type F chromosome was present only in some mandarins originating in Japan, which could thus be distinguished from mandarins originating in other areas. This study is the first report of type F chromosome in citrus. The Vietnamese mandarins we studied were the only one shaving type A, B, C, D and E chromosomes. Summary A hypothetical scheme is given for the evolution of the different types of cauliflower (Brassica oleracea var. botrytis L.). This has been useful in identifying sources of reduced susceptibility to cabbage root fly (Delia radicum (L.)), and may also be useful in the search for reduced susceptibility to clubroot (Plasmodiophora brassicae Woron.). Green mold (Penicillium digitatum) and blue mold (Penicillium italicum) are important sources of postharvest decay affecting the commercialization of mandarins. These fungi infections produce enormous economic losses in mandarin production if early detection is not carried out. Nowadays, this detection is performed manually in dark chambers, where the fruit is illuminated by ultraviolet light to produce fluorescence, which is potentially dangerous for humans. This paper documents a new methodology based on hyperspectral imaging and advanced machine-learning techniques (artificial neural networks and classification and regression trees) for the segmentation and classification of images of citrus free of damage and affected by green mold and blue mold. Feature selection methods are used in order to reduce the dimensionality of the hyperspectral images and determine the 10 most relevant. Neural Networks were used to segment the hyperspectral images. Results achieved using classifiers based on decision trees show an accuracy of around 93% in the problem of decay classification.  相似文献

Summary Atylosia scarabaeoides Benth. and A. platycarpa Benth., close relatives of the species Cajanus cajan (L.) Millsp., were screened for photoperiodic response. Four photoperiods ranging from 12 h 45 min to 19 h were studied in three environments. A. scarabaeoides flowered freely only in the first photoperiod. A. platycarpa not only flowered early (39 to 63 days after planting) in all four photoperiods, but also exhibited a relatively constant vegetative phase up to 16 h of illumination. Cajanus cajan, in which most cultivars-if not all-exhibit a response to photoperiod, was successfully hybridized with the two Atylosia species. It is suggested that genes for earliness and insensitivity to day-length could be transferred from A. platycarpa to C. cajan.  相似文献

Summary Two theories for the origin of the Bolivian weed potato species Solanum sucrense Hawkes have been tested. The first was that it had an allopolyploid origin as a hybrid of the cultivated tetraploid S. tuberosum ssp. andigena (Juz. et Buk.) Hawkes with the wild/weed tetraploid cytotype of S. oplocense Hawkes. The second hypothesis postulated that it was a cross of the weed diploid species S. sparsipilum (Bitt.) Juz. et Buk. with S. oplocense.Synthetic hybrids of S. tuberosum ssp. andigena x S. oplocense and of S. sparsipilum x S. oplocense have been compared morphologically with S. sucrense accessions. Their crossability has also been investigated.The crossability and morphological studies strongly suggested that the former hypothesis was the more likely. This was borne out by observations made on the 1974 University of Birmingham Expedition when segregating populations of S. sucrense and S. oplocense were collected growing with feral S. tuberosum ssp. andigena.The evidence from this study indicates that the S. tuberosum ssp. andigena x S. oplocense hybrid has formed the basis of the S. sucrense gene pool. It is suggested that this hybridogenic taxon be maintained under the name Solanum x sucrense.The clarification of the taxonomic relationships of these three species will be of interest to Globodera resistance breeders, in view of the fact that these taxa are resistant to various pathotypes of the golden nematode.  相似文献

Summary Evidence is provided that hybridization of the tetraploid (2n=4x=48), self-fertile tuber-bearing species Solanum acaule Bitt. with the diploid (2n=2x=24), self-incompatible, tuber-bearing S. megistacrolobum Bitt. takes place in several localities of the province of Jujuy in the high, cold plateaux of the Argentine Puna. The triploid hybrids (2n=3x=36) closely resemble S. acaule in growth habit, leaf morphology and floral structures and for this reason they can be easily overlooked for that species in the field. Experimental data show that S. acaule can be crossed with S. megistacrolobum though the crossability is rather low and variable according to the particular cross considered. The artificial hybrid obtained compares well with the natural hybrid in morphology and chromosome number. The hybrids, though almost completely male sterile, are successful colonizers of disturbed areas around farmers' dwellings, cattle enclosures and other areas where the soil is rich in organic matter.There is some evidence that the Tilcara material of S. acaule subsp. aemulans and the hybrids of S. acaule x S. megistacrolobum have some characters in common, which can be interpreted as having a similar origin.It is postulated that S. acaule subsp. aemulans, in Jujuy at any rate, is not a primitive form of S. acaule as thought by Hawkes and Hjerting, but rather a fertile hybrid derivative of S. acaule x S. megistacrolobum through the functioning of 2n gametes.We also provide evidence that S. bruecheri Correll should not be considered a hybrid of S. acaulle x S. megistacrolobum but a synonym for S. gourlayi Hawkes. The new name, S. x indunii Okada et Clausen, is proposed to designate this hybrid.  相似文献

Summary Fifty-two accessions of the section Patellares wild beet (including 26 Beta patellaris Moq., 13 B. procumbens Chr. Sm and 13 B. webbiana Moq.) and 14 progeny families were selected and tested against sugarbeet cyst nematode, Heterodera schachtii Schm. All Patellares species tested were highly resistant, but not immune, to the development of H. schachtii, after infection. This is the first report that mature female nematodes developed in the roots of B. webbiana plants. Summary Available evidences drawn from biosystematics, evolutionary biology, biogeography, archaeology, history, anthropology, paleo-geology and paleo-meteorology are pooled to reconstruct the series of events that led to the cosmopolitan cultivation of the Asian cultivated rice (O. sativa) and the regionalized planting of the African cultigen (O. glaberrima) in West Africa. The genus Oryza originated in the Gondwanaland continents and, following the fracture of the supercontinent, became widely distributed in the humid tropics of Africa, South America, South and Southeast Asia, and Oceania. The two cultivated species have had a common progenitor in the distant past. Parallel and independent evolutionary processes occurred in Africa and in Asia, following the sequence of: wild perennialwild annualcultivated annual. The weed races also contributed to the differentiation of the cultivated annuals. The corresponding members of the above series are O. longistaminata Chev. et Roehr., O. barthii A. Chev., O. glaberrima Steud., and the stapfii forms of O. glaberrima in Africa; O. rufipogon Griff., O. nivara Sharma et Shastry, O. sativa L., and the spontanea forms of O. sativa in Asia.The differentiation and diversification of the annuals in South Asia were accelerated by marked climatic changes following the last glacial age, dispersal of plants over latitude or altitude, human selection, and manipulation of the cultural environment.Cultivation of rice began in many parts of South and Southeast Asia, probably first in Ancient India. Cultural techniques such as puddling and transplanting were first developed in north and central China and later transmitted to Southeast Asia. Wetland culture preceded dryland culture in China, but in hilly areas of Southeast Asia, dryland cultivation is older than lowland culture. The planting method progressed from shifting cultivation to direct sowing in permanent fields, then to transplanting in bunded fields.Widespread dispersal of the Asian cultigen led to the formation of three eco-geographic races (Indica. Sinica or Japonica, and Javanica) and distinct cultural types in monsoon Asia (upland, lowland, and deep water). Varietal types changed readily within the span of a millenium, largely due to cultivators' preferences, socio-religious traditions, and population pressure. Genetic differentiation developed parallel to the ecologic diversification process.The African cultigen developed later than the Asian cultigen and has undergone less diversification. The wild races in South America and Oceania retain their primitive features mainly due to lack of cultivation pressure or dispersal.Both the African and Asian rices are still undergoing evolutionary changes at habitats where the wild, weed, and cultivated races co-exist.  相似文献

Summary Triticum aestivum L. em Thell. (2n=42; AABBDD), and T. durum Desf. (2n=28; AABB) genomes were substituted into the cytoplasms of Aegilops mutica Boiss. (2n=14; MtMt), Ae. heldreichii Holzm. (2n=14; MM), Ae. uniaristata Vis. (2n=14; MuMu), and Ae. ovata L. (2n=28; CuCuMoMo), to identify the M-genome diploid cytoplasm donor of Ae. ovata. Substitution of the T. durum genome into Ae. uniaristata cytoplasm resulted in a large proportion of shriveled inviable seeds. A few plump viable seeds were obtained all of which produced male-sterile plants having one univalent or telocentric chromosome from Ae. uniaristata. The T. aestivum plants having Ae. uniaristata or Ae. mutica cytoplasms were fertile. However, Ae. mutica was similar to Ae. ovata in the induction of delayed maturity and tall robust growth habit to the T. durum and T. aestivum plants. Cytoplasms of the other C- and M-genome diploids Ae. umbellulata Zhuk. (2n=14; CuCu) and, Ae. heldreichii (2n=14; MM) earlier had been shown to differ from that of Ae. ovata. Therefore, Ae. mutica is the most likely cytoplasm and M-genome donor to Ae. ovata.  相似文献

Summary Cultivars of common wheat (Triticum aestivum L. em. Thell.) of high wheat-rye (Secale cereale L.) crossability set more seed with pollen of other related species than did wheats of low wheat-rye crossability. Summary Cytogenetic evidence has shown that teosinte (Zea mays ssp. mexicana (Schrad.) Iltes) and maize (Zea mays L. ssp. mays) are conspecific. They hybridize readily and their offspring are generally fertile. Teosinte could not have originated as a byproduct of maize-Tripsacum hybridization. Such introgression gave rise to plants that are phenotypically maize or Tripsacum, depending on which parent was used as a pollen donor. Summary The objective of the present paper is to provide information on the morphology and cytology of an intersub-generic hybrid (2n=59) between Glycine tomentella Hayata (2n=78) and G. max (L.) Merr. (2n=40) obtained through in vitro immature seed culture. The hybrid plant was slow in vegetative growth and twinning like the female parent but morphologically was intermediate between both parents for several traits. At metaphase I, the average chromosome associations and ranges for 25 cells were 44.0 I (37–51)+7.5 II (3–11). The plant was completely pollen and seed sterile. The present investigation suggests that wild perennial Glycine species can be exploited as either the male or female parent in wide hybridization programs with the soybean, G. max.  相似文献

Summary The two most serious diseases of cassava (Manihot esculenta Crantz) are cassava mosaic disease (CMD) and cassava bacterial blight (CBB) (Xanthomonas manihotis Starr). Clone 58308, derived from the third backcross of the interspecific cross of cassava (M. esculenta) x ceara rubber (M. glaziovii), showed a high level of resistance to both diseases. Crosses of 58308 with several other clones which varied from susceptible to moderately susceptible to both diseases gave progenies with a significant genotypic correlation between resistance to both diseases (r=0.90), apparently due to linkage. The heritabilities of resistance to the diseases were estimated at 50–70% for CMD and 25–65% for CBB. Resistance to both diseases is assumed to be polygenic. The correlated response to selection for CMD and for CBB was estimated.  相似文献

Summary In black currant progenies derived from parents heterozygous for resistance to American gooseberry mildew. Sphaerotheca mors-uvae (Schw.) Berk., and to gall mite, Cecidophyopsis ribis Westw. (genes Sph 2from the Swedish cultivar Öjebyn and Ce from Ribes grossularia L., respectively), there is invariably a defieit of mildew resistant plants. Linkage in repulsion between Sph 2and Ce would account for the significantly greater deficit of mildew resistants in the gall mite resistant class. In contrast with published data for the mildew resistance gene Sph 3, there is no evidence of linkage of Sph 2with the leafing out gene Lf 1, suggesting that the gene order is Sph 2-Ce-Lf 1-Sph 3.  相似文献

Summary Introgression of germplasm from diploid wild Arachis species to A. hypogaea has great potential for improving pest resistance in cultivated peanuts. This investigation evaluated methods for incorporating exotic germplasm into cultivated peanuts, especially for Cercospora arachidicola Hori resistance. Interspecific hybrids between A. hypogaea (cvs. NC 2 and NC 5) and the wild species A. cardenasii Krap. et Greg. nom. nud. and A. chacoense Krap. et Greg. nom. nud. were analyzed cytologically and for leafspot resistance. All F1 hybrids were sterile, had irregular meiosis, and very few multivalents. They were highly resistant to C. arachidicola in field tests and had a 10-fold reduction of conidia per lesion in the greenhouse as compared to A. hypogaea cultivars. After colchicine treatments of F1 hybrids, hexaploids (2n=60) and aneuploids (2n=54, 56, 63) were observed. The hexaploids had up to 18 univalents per pollen mother cell and very few multivalents, indicating a low frequency of intergenomic chromosome pairing. For C. arachidicola resistance, significant differences were not found among wild species parents, F1 hybrids and two generations of hexaploids. Most hexaploids were stable at 2n=60 and embryos aborted when backcrosses with the respective wild species were attempted. However, when hexaploids were backcrossed to A. hypogaea, several fertile pentaploid (2n=50) offspring were obtained. Use of self-pollinating pentaploids is believed to be the quickest method to recover 40-chromosome hybrid derivatives in these hybrids.  相似文献
