Faba bean has the potential to become a key food and feed protein crop in many areas of the world. The presence of tannins in its seed coat has limited the deployment of this crop as feed and food. The expression of either of the two recessive genes, zt1 and zt2, causes a great reduction of tannins from the seed coat and results in a white flower phenotype. Molecular markers linked to these loci are fundamental tools for speeding up the breeding of low-tannin varieties. The main aim of this study was to develop and validate a robust molecular marker linked to the zt2 locus. We used 176 recombinant inbred lines of the Disco/2 × ILB 938/2 cross at F6 and genotyped those using 257 SNP (single nucleotide polymorphism) markers. An SNP marker associated with zt2 locus was found on faba bean chromosome 3 and was used to develop a high-throughput low-cost KASP (kompetitive allele-specific PCR) marker. The KASP marker can successfully discriminate low-tannin faba beans carrying zt2 from those carrying zt1 and wild-type alleles.  相似文献

Genetic mapping for faba bean lags far behind other major crops. Density enhancement of the faba bean genetic linkage map was carried out by screening 5,325 genomic SSR primers and 2033 expressed sequence tag (EST)‐SSR primers on the parental cultivars '91825' and 'K1563'. Two hundred and fifteen genomic SSR and 133 EST‐SSR primer pairs that detected polymorphisms in the parents were used to screen 129 F2 individuals. This study added 337 more SSR markers and extended the previous linkage map by 2928.45 cM to a total of 4516.75 cM. The number of SSR markers in the linkage groups varied from 12 to 136 while the length of each linkage group ranged from 129.35 to 1180.21 cM. The average distance between adjacent loci in the enhanced genetic linkage map was 9.71 cM, which is 2.79 cM shorter than the first linkage map of faba bean. The density‐enhanced genetic map of faba bean will be useful for marker‐assisted selection and breeding in this important legume crop.  相似文献

Parental lines, F1 and F2 hybrids from a 7×7-diallel of topless faba beans and eight standard cultivars were grown in single-rows for two years at four sowing dates on a highly fertile loam soil near Göttingen. In grain yield, F1 hybrids showed 21 to 54% advantage over the higher yielding parent. Compared with the midparental value an average heterosis of 50% was observed for grain yield. Corresponding heterosis mean values ranged from 3 to 37% for yield components and up to 11% for phenological traits. The grain yield of the highest yielding topless F1 hybrid was equal to that of Alfred, the highest yielding indeterminate standard cultivar. High grain yield in the topless hybrids was closely correlated with high biomass yield, late ripening and tall plants generally having a high number of podded nodes and pods on the main stem. Due to their agronomic advantages, i.e., lower plant length, reduced lodging sensitivity and earlier ripening, efforts at breeding topless hybrids seemed to be worth-while in faba beans. However, their yielding ability must be improved further before they can compete with normal, semi-dwarf and stiff-strawed cultivars.  相似文献

Wild Lens taxa are invaluable sources of useful traits for broadening genetic base of cultivated lentil. Nine inter‐sub‐specific and interspecific crosses were made successfully between cultivated (Lens culinaris ssp. culinaris) and wild lentils (L. culinaris ssp. orientalis, odemensis, lamottei and ervoides). The effect of species groups, day length and temperature on crossability in lentils was evident under normal winter sowing in New Delhi and in summer Himalayan nursery at Sangla in Himachal Pradesh, India, although pollen fertility assessed in all the cross‐combinations showed no significant variation. True hybridity of nine inter‐sub‐specific and interspecific crosses was confirmed through morphological and molecular (ISSR) markers, in which three of 120 primers could confirm the hybridity of all the crosses. All cross‐combinations were also studied for important quantitative traits related to yield. The range, mean and coefficient of variation were estimated in parental lines, F1 and F2 generations to determine the extent of variability generated in cultivated lentils through the introgression of genes from wild L. taxa. A high level of heterosis was observed in F1 crosses for important traits studied. Substantially higher variations for seed yield and its attributing traits were exhibited in F2 generations indicating transgressive segregation. The results of the present investigation revealed that wild L. taxa can be successfully exploited for lentil improvement programmes, and the variations generated could be easily utilized for broadening the genetic base of cultivated lentil gene pool for improving the yield as well as wider adaptation.  相似文献

The genetics of resistance to Ascochyta blight (Ascochyta fabae f. sp. fabae) was studied in two populations of faba bean (Vicia faba). Plants of a resistant population, ILB 752, and a susceptible one, NEB 463, were screened for their reaction to the pathogen and the results were quantified on a scale of 0–5. Crosses were made between plants both within and between accessions and the F1 and F2 generations assessed in a field trial 21 and 45 days after inoculation. Disease scores were greater at 45 days than at 21 days and they were not significantly affected by the presence of susceptible spreader rows in part of the trial. ILB 752 carried a major dominant gene conferring resistance while NEB 463 carried the recessive allele for susceptibility. Furthermore, a minority of plants of NEB 463 appeared to carry at least one pair of complementary recessive genes, also conferring resistance. Most of the plants of ILB 752 were homozygous for the dominant resistance gene and a few were heterozygous. Reciprocal crosses behaved identically, indicating the absence of maternal effects in the expression of Ascochyta blight resistance in faba beans. The results show that it is important to confirm the level of heterozygosity for the resistance genes in this partially outbreeding species before crossing is commenced. The major dominant gene for resistance, identified in ILB 752, has clear potential for use in breeding for Ascochyta blight resistance. The minor genes identified in NEB 463 also show the potential for accumulating resistance through mass selection. Chickpea shows a distinct domestication trajectory vis‐a‐vis pod dehiscence and growth cycle mediated by vernalization insensitivity compared with its companion Near Eastern legumes. Our objectives were: (i) to map the quantitative trait loci (QTLs) associated with vernalization response and seed free tryptophan in domesticated × wild chickpea progeny and (ii) estimate the genetic correlation between vernalization response and free tryptophan content. A domesticated × wild chickpea cross was used to document phenotypic segregation in both traits and to construct a skeletal genetic map for QTL detection. A number of vernalization response and seed free tryptophan content QTLs were documented in both F2 and F3 generations. No significant genetic correlation between these two traits was observed. Epistatic relationship between two free tryptophan loci was documented. It is evident that selection for high seed tryptophan is easier to accomplish relative to selection for vernalization insensitivity. This suggests that the two traits were selected independently in antiquity, thereby corroborating earlier claims for conscious selection processes associated with chickpea domestication.  相似文献

Simple sequence repeat (SSR) marker is a powerful tool for construction of genetic linkage map which can be applied for quantitative trait loci (QTL) and marker‐assisted selection (MAS). In this study, a genetic map of faba bean was constructed with SSR markers using a 129 F2 individuals population derived from the cross of Chinese native variety 91825 (large seed) and K1563 (small seed). By screening 11 551 SSR primers between two parents, 149 primer pairs were detected polymorphic and used for F2 population analysis. This SSR‐based genetic linkage map consisted of 15 linkage groups with 128 SSR. The map encompassed 1587 cM with an average genetic distance of 12.4 cM. The genetic map generated in this study will be beneficial for genetic studies of faba bean for identification of marker‐locus‐trait associations as well as comparative mapping among faba bean, pea and grasspea.  相似文献

Groundnut (Arachis hypogaea L.) is an important oilseed crop grown in more than 100 countries across wide range of environments. Frequent occurrence of drought is one of the limiting factors adversely affecting groundnut productivity, especially in rainfed areas, and hence genotypes having high water use efficiency (WUE) under limited available water need to be developed. In groundnut, WUE is correlated with SPAD chlorophyll meter reading (SCMR) and specific leaf area (SLA). These two traits, SCMR and SLA, can be used as surrogate traits for selecting for WUE. In order to improve SCMR and SLA, and in turn WUE in groundnut, a good knowledge of the genetic system controlling the expressions of these traits is essential for the selection of the most appropriate and efficient breeding procedure. The present investigation was conducted to determine the gene action controlling the inheritance of SCMR and SLA in two crosses, ICG 7243 × ICG 9418 and ICG 6766 × Chico, and their reciprocals. Six generations of each cross (P1, P2, F1, F2, BC1P1, and BC1P2) were evaluated for SCMR and SLA at two stages of the crop growth viz., 60 and 80 days after sowing (DAS). For SCMR at 80 DAS, additive effects were important in both the crosses whereas predominance of dominance effects with duplicate epistasis was observed for SCMR at 60 DAS and SLA at both stages in both the crosses. Predominance of additive effect for SCMR at 80 DAS suggested effective selection could be practiced even in early generations whereas for SCMR at 60 DAS and SLA at both stages in both crosses, it would be better to defer selection to later generations. Further, recording of SCMR and SLA should be done between 60 and 80 DAS for screening the germplasm lines for drought tolerance.  相似文献

Wide crosses were made to identify new cytoplasmic male sterility (CMS) systems in faba beans, based on the interaction of cytoplasm with restorer and maintainer alleles. A total of 330 F1 hybrids were produced in both reciprocal forms. Male sterile segregates were observed in one reciprocal version in the F2 generation of six crosses. Two of these crosses with female parents originating from Afghanistan and Egypt expressed stable male sterility in subsequent backcross generations. Based on the female parents of the two crosses, these two CMS systems were designated CMS 199 and CMS 297. CMS 199 was more stable than CMS 297 during backcross generations and across different environments. Maintainer and restorer lines for both CMS systems were identified. Lower expression of male sterility occurred in CMS 297 in the greenhouse during the winter generations than in isolation cages during the summer generations, which may be utilized to maintain male sterile lines by selfing. Regarding practical applications, the CMS 199 shows great promise for hybrid breeding in faba beans.  相似文献

Temperature stress including low and high temperature adversely affect the growth, development and productivity of crops. Faba bean (Vicia faba L.) is an important crop as both human food source and animal feed, which contains a range of varieties that are sensitive to cold and heat stresses. In this study, 127 faba bean genotypes were collected from gene banks based on differences in geographical origin. The 127 genotypes were treated by single cold stress (2/2 °C day/night temperature (DT/NT)) and 42 genotypes were treated by either single episode of cold or heat (38/30 °C DT/NT) stress, or a combination of both at photosynthetic photon flux density of 250 µmol m?2 s?1. Chlorophyll fluorescence was used to detect the tolerance of faba beans to low and high temperatures. The maximum quantum efficiency of photosystem II (PSII), Fv/Fm, revealed pronounced differences in cold tolerance among the faba bean genotypes. The 42 genotypes were clustered into four groups according to cold and heat stresses, respectively, and the susceptibilities of faba beans under temperature stress could be distinguished. The combination of cold and heat stresses could aggravate the damage on reproductive organs, but not on the leaves, as indicated by the Fv/Fm. These results confirm that the use of Fv/Fm is a useful approach for detecting low and high temperature damage to photosystem II and to identify tolerant faba bean genotypes, however the results also indicate that the geographical origin of the genotypes could not directly be used to predict climate resilience. These sources of cold- and heat-tolerance could improve the temperature tolerance of faba bean in breeding programs.  相似文献

Heterosis, or hybrid vigour, has been used to improve seed yield in several important crops for decades and it has potential applications in soybean. The discovery of over‐dominant quantitative trait loci (QTL) underlying yield‐related traits, such as seed weight, will facilitate hybrid soybean breeding via marker‐assisted selection. In this study, F2 and F2 : 3 populations derived from the crosses of ‘Jidou 12’ (Glycine max) × ‘ZYD2738’ (Glycine soja) and ‘Jidou 9’ (G. max) × ‘ZYD2738’ were used to identify over‐dominant QTL associated with seed weight. A total of seven QTL were identified. Among them, qSWT_13_1, mapped on chromosome 13 and linked with Satt114, showed an over‐dominant effect in two populations for two successive generations. This over‐dominant effect was further examined by six subpopulations derived from ‘Jidou12’ × ‘ZYD2738’. The seed weight for heterozygous individuals was 1.1‐ to 1.6‐fold higher than that of homozygous individuals among the six validation populations examined in different locations and years. Therefore, qSWT_13_1 may be a useful locus to improve the yield of hybrid soybean and to understand the molecular mechanism of heterosis in soybean.  相似文献

To develop a high density linkage map in faba bean, a total of 1,363 FBES (Faba bean expressed sequence tag [EST]-derived simple sequence repeat [SSR]) markers were designed based on 5,090 non-redundant ESTs developed in this study. A total of 109 plants of a ‘Nubaria 2’ × ‘Misr 3’ F2 mapping population were used for map construction. Because the parents were not pure homozygous lines, the 109 F2 plants were divided into three subpopulations according to the original F1 plants. Linkage groups (LGs) generated in each subpopulation were integrated by commonly mapped markers. The integrated ‘Nubaria 2’ × ‘Misr 3’ map consisted of six LGs, representing a total length of 684.7 cM, with 552 loci. Of the mapped loci, 47% were generated from multi-loci diagnostic (MLD) markers. Alignment of homologous sequence pairs along each linkage group revealed obvious syntenic relationships between LGs in faba bean and the genomes of two model legumes, Lotus japonicus and Medicago truncatula. In a polymorphic analysis with ten Egyptian faba bean varieties, 78.9% (384/487) of the FBES markers showed polymorphisms. Along with the EST-SSR markers, the dense map developed in this study is expected to accelerate marker assisted breeding in faba bean.  相似文献

The effectiveness of early generation selection under low and high plant density was evaluated in faba bean (Vicia faba L.). For this, the faba bean cv. 'Polycarpe' was pollinated by eight higher yielding cultivars of different origin. The F1's and F2's together with 'Polycarpe' and the pollinator with the highest yielding ability (cv. 'Effe') were evaluated under high plant density with adjacent controls and under low plant density in a honeycomb design. It was found that the eight hybrids had a different ranking at low than at high plant density for both generations (F1and F2). Hybrid Pol.x A-58 had the best combined yield performance in the F1and F2under high and hybrid Pol.x A-37 under low plant density. Application of individual plant selection in the F2and F3generation of the two crosses (Pol.x A-58 and Pol.x A-37), under both low plant density according to UNR-O honeycomb design and high plant density using a moving average, resulted in the selection of four groups with 20 selected F3plants each. Within each group, equal numbers of seeds from each plant were mixed to establish four mixtures. These mixtures together with two controls ('Polycarpe' and 'Effe') were evaluated in a randomized complete block design (22 plants/m2) in one location for two years. It was found that the material selected continuously under low plant density had a significantly higher yield than the material selected under high plant density. In addition, the material selected from Pol.x A-37 (H1R2) under high plant density had a lower mean yield than that selected continuously under low plant density (H1H2). Their average yield over years was significantly higher than the yield of the material selected from Pol.x A-58 under both plant densities. Furthermore, the material selected from Pol.x A-58 under low plant density, had a significantly higher mean yield over years than its sibs selected under high plant density. It was concluded that honeycomb selection at low plant density can be used effectively for early generation selection in faba bean.  相似文献

The fungal disease cercospora leaf spot CLS (Cercospora zonata) has affected major faba bean (Vicia faba) production regions in southern Australian in the last several years. This study offers the first report of sources of resistance to CLS in faba bean and describes techniques to evaluate resistance to C. zonata in faba bean genotypes within a controlled environment. The method was rapid (43 days), repeatable (R 2 > 0.74) and demonstrated positive correlations (R 2 > 0.45–0.80) to data collected from field disease nurseries under naturally established CLS epiphytotics. All faba bean cultivars currently adopted by the Australian industry were found to be susceptible to CLS and defoliation was found to be an important component of disease expression. Genetic analysis of segregation patterns in F 2 derived F 3 families of 1322/2*Farah (resistant*susceptible) showed the mode of inheritance of resistance to C. zonata was monogenic dominant. F 3 families were shown to segregate in the ratio of 1:2:1 for homozygous resistant: heterozygous: homozygous susceptible (χ22 = 2.78; P > 0.05) and individual plants within heterozygous F 3 families segregated in the ratio of 3:1 for resistant: susceptible responses (χ12 = 2.93; P > 0.05). Monogenic dominant inheritance also explained the change in frequency of resistant and susceptible plants within a population of cv. Cairo following one generation of self-pollination (χ2 = 0.88, 0.3 < P < 0.5). The sources of resistance identified in this study are being used to transfer CLS resistance to adapted faba bean genotypes for future cultivar releases to the southern Australian industry.  相似文献

Efficient management of legumes in order to maximize benefits depends on a correct field assessment of N2 fixation. A field experiment was conducted during a 6-year period (2001–2002 to 2006–2007) in Córdoba (Southern Spain) on a rainfed Vertisol within the wheat-chickpea and wheat-faba bean rotation framework of a long-term experiment started in 1986. The aim was to determine the effect of tillage systems [no tillage (NT) and conventional tillage (CT)] on chickpea and faba bean N2 fixation. Fixation was calculated using the 15N isotopic dilution (ID) and 15N natural abundance (NA) methods with the reference being the wheat crop. The strong inter-annual rain variation caused great differences in the behaviour of both leguminous plants with regard to grain yield, nodule biomass and N2 fixation. The NT system showed more nodule biomass than the CT system in both legumes. The ID method was more accurate than the NA method in determining N2 fixation. The average amount of fixed N in faba bean (80 kg ha?1 year?1) was much greater than that in chickpea (31 kg ha?1 year?1). The Vertisol under the NT system offered more favourable conditions for the stimulation of the N2 fixation, with fixed N values that were significantly higher than under CT. The N added to the system through N2 fixation was low in faba bean and virtually nonexistent in chickpea, only in terms of above-ground biomass.  相似文献

The objectives of this study were to determine genetics of Al tolerance and whether the Al tolerance observed is governed by the same gene. The lines ‘L‐7903’ and ‘L‐4602’ have been developed through breeding programme as Al‐tolerant lines. These lines showed maximum root regrowth and minimum accumulation of Al and callose as compared to sensitive genotypes (‘BM‐4’ and ‘L‐4147’). Al tolerance in the parents, F1, F2 and backcross generations was estimated using the regrowth of the primary root after staining and scoring of fluorescent signals. The F1 hybrids responded similarly to the tolerant parents, indicating dominance of Al tolerance over sensitivity. The segregation ratios obtained for Al tolerance and sensitivity in the F2 and backcross generations were 3 : 1 and 1 : 1, respectively. Test of allelism confirmed the same gene was conferring Al tolerance in both genotypes (‘L‐7903’ and ‘L‐4602’) as the F1 was also tolerant and no segregation of tolerant : sensitive was recorded. These results indicated that Al tolerance is a monogenic dominant trait that can be easily transferred to agronomic bases through backcross breeding technique.  相似文献

Cultivated lentil (Lens culinaris Medik.) is susceptible to aphanomyces root rot (ARR), whereas partial resistance is present in wild lentil including Lens ervoides (Brign.) Grande. Approximately six generations of selfing are required to fix a desired trait in a population, which usually requires 2 years in a breeding programme, so the primary objective was to develop a rapid generation cycling (RGC) technique that achieves this goal in 1 year. Rapid generation cycling was then tested on an F2 population (LR‐59) derived from a L. culinaris × L. ervoides cross in combination with a reliable ARR screening technique, which generates a wide range of disease severities conducive to selection. Phenotyping of an F2 population of more than 1,200 plants resulted in scores ranging from 2.4 to 4.0 on a scale from zero to five. Plants with scores lower than 4.0 were selected for advancement for five generations using a modified single‐seed descent method, optimum growing conditions, 20‐hr photoperiod and harvest of immature seeds. Seeds were germinated in a 100 μM gibberellin solution. Average generation length after phenotyping was 56 days resulting in five generations within approximately 300 days. Using a modified inoculation protocol, ARR phenotyping of the F7 population resulted in scores ranging from 1.4 to 4.0. This inexpensive, nonsterile speed breeding protocol saves 1 year in the development of lentil varieties with improved ARR resistance.  相似文献

Crosses were performed to introgress genes for productivity and other desirable traits from ricebean (Vigna umbellata) into black gram (Vigna mungo). Crossability was very poor in black gram × ricebean crosses, and only two to nine true hybrid plants were obtained. Plant fertility was very poor in initial generations, but was improved gradually from F2 onwards. Twenty‐four uniform progenies, bulked in F7, were evaluated for yield potential. The percentage increase/decrease in yield ranged from ?35.48 to 50.31 over the check cultivar (‘Mash338’, female parent). All the progenies were found resistant to Mungbean yellow mosaic virus, Cercospora leaf spot and Bacterial leaf spot diseases. Overall, it was found that desirable traits such as high pod number, seed weight, productivity and resistance to diseases have been introgressed successfully into black gram from ricebean. A derivative line, KUG114, recorded yield superiority of 39.45% over the check cultivar ‘Mash338’ on the average of 14 multilocation research trials. It was released under the name ‘Mash114’ for cultivation in the Punjab state.  相似文献

Leptosphaeria maculans causes blackleg disease on Brassica napus, an economically important oilseed crop. Brassica juncea has high resistance to blackleg and is a source for the development of resistant B. napus varieties. To transfer the Rlm6 resistance gene from B. juncea into B. napus, an interspecific cross between B. napus “Topas DH16516” and B. juncea “Forge” was produced, followed by the development of F2 and F3 generations. Sequence characterized amplified region (SCAR) and cleaved amplified polymorphic sequence (CAPS) markers linked to the L. maculans resistance gene Rlm6 were developed. Segregation of SCAR and CAPS markers linked to Rlm6 were confirmed by genotyping of F2 and F3 progeny. Segregation of CAPS markers and phenotypes for blackleg disease severity in F2 plants had a Mendelian ratio of 3:1 in resistant vs. susceptible plants, respectively, supporting the assumption that genetic control of resistance was by a single dominant gene. The molecular markers developed in this study, which show linkage with the L. maculans resistance gene Rlm6, would facilitate marker‐assisted backcross breeding in a variety development programme.  相似文献

The use of resistant cultivars is one of the best methods for nematode control and reduction of economic losses caused by these pathogens. Studies of inheritance of nematode resistance in common bean (Phaseolus vulgaris L.) are nonetheless scarce. The present paper reports on the estimation of genetic parameters associated with resistance to the root nematode Meloidogyne incognita race 1 in common beans. Two contrasting bean lines, ‘Aporé’ (P1 = nematode resistant) e ‘Macarr?o Rasteiro Conquista’ (P2 = susceptible), and the generations F1 (P1 × P2), F2 (P1 × P2), BC1(P1) = (F1 × P1) and BC1(P2) = (F1 × P2), were assessed 45 days after nematode inoculation, through a scale related to the number of eggs per gram of root tissue. Dominant genetic effects were inferior in magnitude to additive effects, indicating incomplete dominance of nematode resistance. Dominance was in the direction of increased nematode resistance (i.e., lower number of eggs per g root). Resistance to Meloidogyne incognita race 1 in common bean is under control of a single gene locus, with incomplete dominance of the resistance allele present in ‘Aporé’, but modifier genes affecting its expression appear to be present in the susceptible parent ‘Macarr?o Rasteiro Conquista’.  相似文献
