苎麻基因组SRAP扩增体系的优化研究   总被引:21,自引:0,他引:21  
以苎麻自交系品种为材料,研究了苎麻SRAP分析过程中的影响因素,包括10xPCR Buffer、模板浓度、Mg^2+、dNTP、引物、Taq酶、循环次数、退火温度等,建立了适于苎麻SRAP分析的PCR反应体系:即在201.1反应体系中,引物浓度为0.3μmol/L;模板DNA的用量为60~100ng;Mg^2+浓度为2.0mmol/L;dNTP浓度为0.2mmol/L;砌酶用量为1U。适宜的扩增程序为先94℃变性1min,前5个循环以94℃变性1min、33℃复性1min、72℃延伸1min进行,接下来将复性温度提高到52℃,其它条件不变,进行30个循环,最后72℃延伸5min,4℃保存。本文讨论了SRAP分子标记在苎麻上的应用前景,提出了应用SRAP分子标记构建苎麻遗传图谱的可行性。  相似文献

以萝卜基因组DNA为模板,对其SRAP-PCR反应体系的各个主要影响因子进行了系统的优化,建立了重复性好、多态性丰富的萝卜SRAP-PCR反应体系:20 μL反应体系中,DNA 50 ng、Mg2+2.25 mmol/L、dNTP0.2 mmol/L、Taq1 U、Primer 0.7μmol/L.扩增程序为:94℃预变性5 min;94℃变性1 min、35℃复性1 min、72℃延伸1 min,5个循环;94℃变性1 min、50℃复性1 min、72℃延伸1 min,35个循环,72℃延伸10 min,4℃保温.该反应体系在不同萝卜种质资源遗传多样性分析、遗传图谱构建、分子标记辅助选择育种等方面都有重要作用.  相似文献

本研究从Mg2+、dNTPs、Taq DNA聚合酶、引物、模板5种因素对蚕豆序列相关扩增多态性(sequencerelated amplified polymorphism, SRAP)体系进行了优化。结果表明,蚕豆最佳PCR反应体系(25μL)为:1.0 mmol/L d NTPs,0.5 mmol//L Mg2+,0.4 U Taq酶,4.0 mmol/L引物,2.0 mmol/L的模板。反应程序为:94℃预变性5 min;94℃变性1 min,35℃复性1 min,72℃延伸1 min,5个循环;94℃变性1 min,50℃复性1 min,72℃延伸1 min,35个循环;72℃延伸5 min,4℃保存。利用优化的SRAP标记对12份蚕豆材料进行遗传多样性分析,研究结果表明12个材料明显聚为2个类群,类群中又有亚类。12个蚕豆品种中有些虽然来自同一地区,但亲缘关系却较远,而来自不同地区的蚕豆也显示出来很大差异,说明蚕豆材料具有丰富的遗传多样性。  相似文献

以西瓜品种中华拳王为试材,探索了西瓜SRAP-PCR反应程序,并对西瓜SRAP-PCR反应体系的各影响因子进行了梯度设置试验,筛选和建立了可扩增多态性高、重复性好、带型清晰的最佳SRAP-PCR反应程序和体系。反应程序为:94℃预变性5 min;94℃变性1 min,35℃退火1 min,72℃延伸1 min,共5个循环;94℃变性1 min,50℃退火1min,72℃延伸1 min,共35个循环;72℃延伸5 min;4℃保存。反应体系(Total为15μL):DNA 100 ng,Mg2 2.0 mmol/L,dNTPs 0.3 mmol/L,Primer 60 ng,Taqpolymerase 0.75 U。结果表明,该程序和体系能很好地满足西瓜基因组SRAP扩增的要求,SRAP标记应用于西瓜遗传研究是可行的。  相似文献

建立适宜桃基因组DNA的SRAP-PCR扩增体系,为桃基因图谱的构建和分子标记打下基础。以桃基因组DNA为模板,通过正交试验设计,从dNTPs、Mg2+、Taq酶、引物、模板5种因素4个水平对桃SRAP-PCR反应体系进行优化,所建立的体系为25μL:dNTPs为0.12 mmol/L,Mg2+为4 mmol/L,Taq酶2 U,引物为0.3 mmol/L,模板DNA50ng。PCR反应程序为:94℃预变性5 min;94℃变性l min,35℃复性l min,72℃延伸l min,5个循环;94℃变性l min,50℃复性l min,72℃延伸l min,35个循环,72℃延伸10 min。  相似文献

以大粒无核葡萄‘皇家秋天’的基因组DNA为模板,对无核葡萄SRAP-PCR反应体系的主要成分及反应退火温度进行优化。获得的最优SRAP-PCR反应程序为94℃预变性5 min,94℃变性1 min,38℃复性1 min,72℃延伸1 min,5个循环;94℃变性1 min,56℃复性1 min,72℃延伸1 min,35个循环;72℃终延伸10 min。25 μL体系中,模板DNA 40 ng,Mg2+ 1.5 mmol/L,dNTPs 0.2 mmol/L,引物0.2 μmol/L,Taq DNA聚合酶0.6 U。实验结果表明,优化后的SRAP-PCR反应体系扩增多态性高,带型清晰,稳定性好。  相似文献

为建立草莓SRAP-PCR适宜的反应体系,以草莓品种‘丰香’为实验材料,采用单因素实验设计,对Mg2+、dNTPs、Taq DNA聚合酶及引物浓度4个因素4水平进行优化,并在此基础上对模板DNA的浓度和退火温度进行优化。结果表明,草莓SRAP-PCR最佳反应体系为:20μL的反应体系中含10×PCR buffer 2μL,Mg2+2.0 mmol/L,dNTPs 0.3 mmol/L,正反向引物各为0.6μmol/L,Taq DNA聚合酶1.0 U,模板DNA为100 ng。扩增程序为:94℃预变性5 min;94℃变性1 min,35℃退火1 min,72℃延伸1 min,共5个循环;94℃变性1 min,54℃退火1 min,72℃延伸1 min,共35个循环;72℃延伸5 min;4℃保存。利用该优化体系筛选引物,从110对SRAP引物组合中筛选出29对条带清晰丰富、多态性好的引物,证明了此优化体系稳定可靠,能够用于草莓种质资源的鉴定、分子标记辅助育种等研究。  相似文献

以烤烟品种为材料,研究了烟草RAPD分析过程中的影响因素,包括模板浓度、Mg2+、dNTP、引物、Taq 酶、循环次数、退火温度等,建立了适于烟草RAPD分析的PCR反应体系:即在25μl反应体系中,模板用量为40ng;引物浓度为0.4μM;Mg2+浓度为2.5mM;dNTP浓度为0.2mM;Taq DNA聚合酶用量为1U。扩增程序为94℃预变性5min;然后94℃变性1min,38℃复性1min,72℃延伸1.5min,39个循环;最后72℃延伸5min。  相似文献

本研究以芦笋基因组总DNA为模板,通过对芦笋SRAP反应体系的重要参数进行优化,建立了一套适用于芦笋的SRAP反应体系:25μL的反应体系中,模板DNA量80ng、Mg2+浓度3.0mmol/L、上下游引物各0.2μmol/L、dNTPs0.3mmol/L、Taq DNA聚合酶1U以及1×Buffer。扩增程序为:94℃预变性3min;94℃30s、35℃30s、72℃1.5min,5个循环;然后退火温度提高到50℃,35个循环;最后72℃延伸10min。比较琼脂糖凝胶和变性聚丙烯酰胺凝胶电泳检测SRAP扩增产物的多态性,结果发现:6%变性聚丙烯酰胺凝胶电泳检测扩增产物比琼脂糖的效果好。利用该优化体系筛选引物,从256个SRAP引物组合中筛选出239个,它们具有扩增条带清晰、丰富、重复性好的优点,证明了此优化体系稳定可靠。  相似文献

本研究利用L16(54)正交试验设计研究了影响冬瓜SSR-PCR的五个因子(10×Buffer(含Mg2+),d NTP,DNA,引物,r Taq),研究结果表明Buffer(含Mg2+)和r Taq对PCR影响最明显,最终筛选出最佳反应体系:10×Buffer 2.0μL(1×Buffer),d NTP 2.0μL(200μmol/μL),DNA 0.6μL(6 ng/μL),引物0.4μL(0.2μmol/L)。体系体积为20μL,循环数为35。引物筛选程序为:94℃预变性3 min,94℃变性45 s,58℃~68℃复性30 s,5个循环,每个循环退火温度降2℃,72℃延伸1 min,94℃变性30 s,50℃~58℃复性1 min,8个循环,每个循环退火温度降低1℃,72℃延伸1 min,94℃变性30 s,50℃复性30 s,72℃延伸1 min,共30个循环,72℃延伸7 min。本研究建立了适合冬瓜的SSR反应体系和引物多态性筛选程序。  相似文献

Autotoxicity restricts reseeding of alfalfa (Medicago sativa L.) after alfalfa until autotoxic chemical(s) breaks down or is dispersed into external environments. A series of aqueous extracts from leaves, stems, roots and seeds of alfalfa ‘Vernal’ were bioassayed against alfalfa seedlings of the same cultivar to determine their autotoxicity. The highest inhibition was found in the extracts from the leaves. Extracts at 40 g dry tissue l?1 from alfalfa leaves were 15.4, 17.5 and 28.7 times more toxic to alfalfa root growth than were those from roots, stems and seeds, respectively. A high‐performance liquid chromatography (HPLC) analysis with nine standard compounds showed that the concentrations and compositions of allelopathic compounds depended on the plant parts. In leaf extracts that showed the most inhibitory effect on root growth, the highest amounts of allelochemicals were detected. Among nine phenolic compounds assayed for their phytotoxicity on root growth of alfalfa, coumarin, trans‐cinnamic acid and o‐coumaric acid at 10?3 m were most inhibitory. The type and amount of causative allelochemicals found in alfalfa plant parts were highly correlated with the results of the bioassay, indicating that the autotoxic effects of alfalfa plant parts significantly differed.  相似文献

Summary The high-lysine gene in Risø mutant 1508 conditions an increased lysine content in the endosperm via a changed protein composition, a decreased seed size, and several other characters of the seed. The designation lys3a, lys3b, and lys3c, is proposed for the allelic high-lysine genes in three Risø mutants, nos 1508, 18, and 19. Linkage studies with translocations locate the lys3 locus in the centromere region of chromosome 7. A linkage study involving the loci lys3 and ddt (resistance to DDT) together with the marker loci fs (fragile stem), s (short rachilla hairs), and r (smooth awn) show that the order of the five loci on chromosome 7 from the long to the short chromosome arm is r, s, fs, lys3, ddt. The distance from locus r to locus ddt is about 100 centimorgans.  相似文献

Development of onion (Allium cepa L., cv. ‘Early Cream Gold’) seed under cool climate conditions in Tasmania, Australia occurred over a longer duration than previously reported, but similar patterns of change in yield components were recorded. In contrast to previous studies, umbel moisture content declined from 85 to 67 % over 57 days while seed moisture content decreased from 85 to 31 %. Seed yield continued to increase over the duration of crop development, with increasing seed weight compensating for seed loss resulting from capsule dehiscence in the later stages of maturation. Germination percentage was high and did not vary significantly from 53 to 77 days after full bloom (DAF), but mean germination time declined and uniformity of germination increased significantly over the same time period. The percentage abnormal seedlings declined with later harvest date, resulting in highest seed quality at 77 DAF. The results of this study suggest that the decision to harvest cool climate onion seed crops before capsule dehiscence will result in a loss of potential seed yield and quality.  相似文献

Summary Avoidance of rust fungi that was based on poor appressorium induction was previously found in Hordeum chilense. In the present study 95 accessions of Triticeae were screened for avoidance of Puccinia hordei. The percentage of appressorium formation per germinated spore ranged from 6 to 90%. On none of the 41 accessions of Aegilops, Agropyron, Elymus, Secale, Thinopyrum or Triticum studied was the rate of appressorium formation lower than 25%. Lower rates of appressorium formation were, however, found on accessions of wild barley species Hordeum brachyantherum, H. marinum, H. parodii and H. secalinum. Progress is being made, mainly by ICARDA but also elsewhere, in breeding for resistance to Botrytis, AScochyta, Uromyces, and Orobanche; and some lines have resistance to more than one pathogen. The strategy is to extend multiple resistance but also to seek new and durable forms of resistance. Internationally coordinated programs are needed to maintain the momentum of this work.Tolerance of abiotic stresses leads to types suited to dry or cold environments rather than broad adaptability, but in this cross-pollinated species, the more hybrid vigor expressed by a cultivar, the more it is likely to tolerate various stresses.  相似文献

Summary Cytoplasmic male sterility (cms) is described in the F1 hybrids Ribes × carrierei (R. glutinosum albidum × R. nigrum) and R. sanguineum × R. nigrum. In backcrosses to R. nigrum, progenies with R. glutinosum cytoplasm were either all male sterile, or segregated for full male fertility (F) and complete (S) and partial (I) male sterility. Ratios of F:I+S suggested that two linked genes controlled cms, F plants being dominant for one (Rf 1) and recessive for the other (Rf 2).Segregation for cms in relation to three linded genes, Ce (resistance to the gall mite, Cecidophyopsis ribes), Sph 3(resistance to American gooseberry mildew, Sphaerotheca mors-uvae) and Lf 1(one of two dominant additive genes controlling early season leafing out) indicated that Rf 1and Rf 2were in this linkage group.
