This experiment was conducted in maize field plots to study the effects of controlled release and application depth of urea on soil microbial biomass and activities at two depths of surface soil of a Japanese Andisol from June to September, 2001. Three N amendment treatments and a Control were included in this experiment: deep application (8 cm) of controlled release urea; deep application (8 cm) of conventional urea; surface application of conventional urea; Control, without N application. Prior to this experiment, the field plots received the same N fertilizer treatments for two consecutive years under maize/barley rotation. Soil microbial biomass, dehydrogenase and nitrification activities exhibited great vertical and temporal variations during the maize growth season, and the microbial biomass was significantly correlated to soil water-filled pore space (p<0.01). N fertilization did not significantly affect the microbial biomass, but greatly increased the dehydrogenase and nitrification activities. The increase in the microbial activities following N fertilization was not attributed to the increase in microbial biomass but to the increase in intrinsic microbial activities. Controlled release urea was found to continuously affect the dehydrogenase activity over a shorter distance, while conventional urea could greatly increase the enzyme activity for a shorter period of time. Both controlled release and deep application of urea had potentials to reduce the nitrification activity and suggested that the nitrate production might be decreased in 0–10 cm surface soil. Deep application of urea increased aboveground N uptake by maize and then the recovery rate of N fertilizer, whereas controlled release of urea greatly increased grain yield and N uptake by grain.

A better understanding of soil microbial processes is required to improve the synchrony between nutrient release from plant residues and crop demand. Phospholipid fatty acid analysis was used to investigate the effect of two crop rotations (continuous maize and maize-crotalaria rotation) and P fertilization (0 and 50 kg P ha−1 yr−1, applied as triple superphosphate) on microbial community composition in a highly weathered soil from western Kenya. Microbial substrate use in soils from the field experiment was compared in incubation experiments. Higher levels of soil organic matter and microbial biomass in the maize-crotalaria rotation were connected with higher total amounts of phospholipid fatty acids and an increase in the relative abundances of indicators for fungi and gram-negative bacteria. P fertilization changed the community profile only within the continuous maize treatment. The decomposition of glucose, cellulose and three plant residues (all added at 2.5 g C kg−1 soil) proceeded faster in soil from the maize-crotalaria rotation, but differences were mostly transient. Microbial P and N uptake within one week increased with the water-soluble carbon content of added plant residues. More P and N were taken up by the greater microbial biomass in soil from the maize-crotalaria rotation than from continuous maize. Re-mineralization of nutrients during the decline of the microbial biomass increased also with the initial biological activity of the soil, but occurred only for a high quality plant residue within the half year incubation period. Compared to the effect of crop rotation, P fertilization had a minor effect on microbial community composition and substrate use.

Data from a 49-year-long organic–mineral fertilization field experiment with a potato–maize–maize–wheat–wheat crop rotation were used to analyse the impact of different fertilizer variations on yield ability, soil organic carbon content (SOC), N and C balances, as well as on some characteristic energy balance parameters. Among the treatments, the fertilization variant with 87 kg ha?1 year?1 N proved to be economically optimal (94% of the maximum). Approximately 40 years after initiation of the experiment, supposed steady-state SOC content has been reached, with a value of 0.81% in the upper soil layer of the unfertilized control plot. Farmyard manure (FYM) treatments resulted in 10% higher SOC content compared with equivalent NPK fertilizer doses. The best C balances were obtained with exclusive mineral fertilization variants (?3.8 and ?3.7 t ha?1 year?1, respectively). N uptake in the unfertilized control plot suggested an airborne N input of 48 kg ha?1 year?1. The optimum fertilizer variant (70 t ha?1 FYM-equivalent NPK) proved favourable with a view to energy. The energy gain by exclusive FYM treatments was lower than with sole NPK fertilization. Best energy intensity values were obtained with lower mineral fertilization and FYM variants. The order of energy conversion according to the different crops was maize, wheat and potato.

Increasing nutrient inputs into terrestrial ecosystems affect not only plant communities but also associated soil microbial communities. Studies carried out in predominantly unmanaged ecosystems have found that increasing nitrogen (N) inputs generally decrease soil microbial biomass; less is known about long-term impacts in managed systems such as agroecosystems. The objective of this paper was to analyze the responses of soil microorganisms to mineral fertilizer using data from long-term fertilization trials in cropping systems. A meta-analysis based on 107 datasets from 64 long-term trials from around the world revealed that mineral fertilizer application led to a 15.1% increase in the microbial biomass (Cmic) above levels in unfertilized control treatments. Mineral fertilization also increased soil organic carbon (Corg) content and our results suggest that Corg is a major factor contributing to the overall increase in Cmic with mineral fertilization. The magnitude of the effect of fertilization on Cmic was pH dependent. While fertilization tended to reduce Cmic in soils with a pH below 5 in the fertilized treatment, it had a significantly positive effect at higher soil pH values. Duration of the trial also affected the response of Cmic to fertilization, with increases in Cmic most pronounced in studies with a duration of at least 20 years. The input of N per se does not seem to negatively affect Cmic in cropping systems. The application of urea and ammonia fertilizers, however, can temporarily increase pH, osmotic potential and ammonia concentrations to levels inhibitory to microbial communities. Even though impacts of fertilizers are spatially limited, they may strongly affect soil microbial biomass and community composition in the short term. Long-term repeated mineral N applications may alter microbial community composition even when pH changes are small. How specific microbial groups respond to repeated applications of mineral fertilizers, however, varies considerably and seems to depend on environmental and crop management related factors.

The influence of two experimental soil treatments, Z93 and W91, on nitrogen transformations, microbial activity and plant growth was investigated in soil microcosms. These compounds are commercially marketed fermentation products (Agspectrum) that are sold to be added to field soils in small amounts to promote nitrogen and other nutrient uptake by crops in USA. In laboratory microcosm experiments, soils were amended with finely ground alfalfa-leaves or wheat straw, or left unamended, in an attempt to alter patterns of soil nitrogen mineralization and immobilization. Soils were treated in the microcosms with Z93 and W91 at rates equivalent to the recommended field application rates, that range from 0.2 to 1.1 l ha−1, (0.005-0.03 μl g−1 soil). We measured their effects on soil microbial activity (substrate-induced respiration (SIR), dehydrogenase activity (DHA) and acid phosphatase activity (PHOS)), soil nitrogen pools (microbial biomass N, mineral N, dissolved organic N), and transformations (net N mineralization and nitrification, 15N dilution of the mineral N pool, and accumulation of mineral N on ion-exchange resins), and on wheat plant germination and growth (shoot and root biomass, shoot length, N uptake and 15N enrichment of shoot tissues), for up to 56 days after treatment. To follow the movement of nitrogen from inorganic fertilizer into plant biomass we used a 15N isotopic tracer. Most of the soil and plant responses to treatment with Z93 or W91 differed according to the type of organic amendment that was used. Soil treatment with either Z93 or W91 influenced phosphatase activity strongly but did not have much effect on SIR or DHA. Both chemicals altered the rates of decomposition and mineralization of organic materials in the soil, which was evidenced by significant increases in the rates of the decomposition of buried wheat straw, and by the acceleration of net, rates of N mineralization, relative to those of the controls. Soil nitrate availability increased at the end of the experiment in response to both chemical treatments. In alfalfa-amended soils, the final plant biomass was decreased significantly by treatment with W91. Increased plant growth and N-use efficiency in straw-amended soil, resulting from treatments with Z93 or W91, was linked to increased rates of N mineralization from indigenous soil organic materials. This supports the marketing of these compounds as promoters of N uptake at these low dosage inputs.

Because soil biota is influenced by a number of factors, including land use and management techniques, changing management practices could have significant effects on the soil microbial properties and processes. An experiment was conducted to investigate differences in soil microbiological properties caused by long- and short-term management practices. Intact monolith lysimeters (0.2 m2 surface area) were taken from two sites of the same soil type that had been under long-term organic or conventional crop management and were then subjected to the same 2.5-year crop rotation [winter barley (Hordeum vulgare L.), maize (Zea mais L.), lupin (Lupinus angustifolius L.), and rape (Brassica napus L. ssp. oleifera)] and two fertilizer regimes (following common organic and conventional practices). Soil samples were taken after crop harvest and analyzed for microbial biomass C and N, microbial activity (fluorescein diacetate hydrolysis, arginine deaminase activity, and dehydrogenase activity), and total C and N. The incorporation of the green manure stimulated growth and activity of the microbial communities in soils of both management histories. Soil microbial properties did not show any differences between organically and conventionally fertilized soils, indicating that crop rotation and plant type had a larger influence on the microbial biomass and enzyme activities than fertilization. Initial differences in microbial biomass declined, while the effects of farm management history were still evident in enzyme activities and total C and N. Links between enzyme activities and microbial biomass C varied depending on treatment, indicating differences in microbial community composition.

Investigations on the adjustment of nitrogen supply from different N sources to the N uptake of maize A two years field experiment was conducted to study the effects of the nitrogen fertilizers: wheat swill, BASAMMON (NH4 + nitrification inhibitor DCD) and calcium ammonium nitrate (CAN) applied at N rates of 0.60.120 or 180 kg N/ha on yield, N uptake and soil mineral N residues of maize for silage or grain production. CAN was either broadcast or placed in the maize rows, with or without the addition of an Azospirillum-biopreparation (AZOGREEN). Due to initially high soil mineral N contents (50–70 kg N/ha) and a high mineralization from the soil (unfertilized: 100–170 kg N/ha), neither effects of fertilizer type, nor interactions between fertilizer type and N rate on biomass production and N uptake of maize were observed. The economically optimum total dry matter production and grain yield were obtained with a fertilization of 60–120 kg N/ha. In contrast to the biomass production the soil mineral N was considerably influenced by fertilizer type and N rate. The nitrate content of the soil increased during the early developmental stages of maize after an early application of swill or BASAMMON, and also during the ripening period after a late high dose of CAN. Though the placement of CAN was reflected in the soil, the soil nitrogen content per unit area could not be much reduced by row fertilization. The effects of AZOGREEN were only small. The results were greatly influenced by the high mineralization potential at the experimental site.

The interactions between soil P availability and mycorrhizal fungi could potentially impact the activity of soil microorganisms and enzymes involved in nutrient turnover and cycling, and subsequent plant growth. However, much remains to be known of the possible interactions among phosphorus availability and mycorrhizal fungi in the rhizosphere of berseem clover (Trifolium alexandrinum L.) grown in calcareous soils deficient in available P. The primary purpose of this study was to look at the interaction between P availability and an arbuscular mycorrhizal (AM) fungus (Glomus intraradices) on the growth of berseem clover and on soil microbial activity associated with plant growth. Berseem clover was grown in P unfertilized soil (−P) and P fertilized soil (+P), inoculated (+M) and non-inoculated (−M) with the mycorrhizal fungus for 70 days under greenhouse conditions. We found an increased biomass production of shoot and root for AM fungus-inoculated berseem relative to uninoculated berseem grown at low P levels. AM fungus inoculation led to an improvement of P and N uptake. Soil respiration (SR) responded positively to P addition, but negatively to AM fungus inoculation, suggesting that P limitation may be responsible for stimulating effects on microbial activity by P fertilization. Results showed decreases in microbial respiration and biomass C in mycorrhizal treatments, implying that reduced availability of C may account for the suppressive effects of AM fungus inoculation on microbial activity. However, both AM fungus inoculation and P fertilization affected neither substrate-induced respiration (SIR) nor microbial metabolic quotients (qCO2). So, both P and C availability may concurrently limit the microbial activity in these calcareous P-fixing soils. On the contrary, the activities of alkaline phosphatase (ALP) and acid phosphatase (ACP) enzymes responded negatively to P addition, but positively to AM fungus inoculation, indicating that AM fungus may only contribute to plant P nutrition without a significant contribution from the total microbial activity in the rhizosphere. Therefore, the contrasting effects of P and AM fungus on the soil microbial activity and biomass C and enzymes may have a positive or negative feedback to C dynamics and decomposition, and subsequently to nutrient cycling in these calcareous soils. In conclusion, soil microbial activity depended on the addition of P and/or the presence of AM fungus, which could affect either P or C availability.

Summary The soil microbial biomass contains important labile pools of C, N, P, and S, and fluctuations in its size and activity can significantly influence crop productivity. In cropping systems where fertilizer use is reduced or eliminated and green-manure legumes are used, nutrient availability is more directly linked to C-cycle dynamics. We observed the fluctuations in microbial biomass C and P, and in microbial biomass activity over three cropping seasons in continuous maize and 2-year maize-wheat-soybean rotation agroecosystems under no-till and reduced-chemical-input management. We estimated the concentrations of microbial C and P using fumigation-incubation and fumigation-extraction techniques for the surface 20 cm of Cecil and Appling series soils (clayey, kaolinitic, thermic, Typic Kanhapludults). There were significant seasonal fluctuations in microbial C and P under all cropping systems. Generally, the magnitude of fluxes and the quantity of microbial C and P tended to be higher in reduced-chemical-input systems due to tillage and incorporation of crop, weed, and legume residues. Over 3 years, the means for microbial C were 435 under reduced-input maize; 289 under no-till maize; 374 und the reduced-input crop rotation; and 288 mg kg-1 soil under the no-till rotation. The means for microbial P were 5.2 under reduced-input maize; 3.5 under no-till maize; 5.0 under the reduced-input rotation; and 3.5 mg kg-1 soil under the no-till rotation. Estimates of microbial activity, derived from CO2–C evolution and specific respiratory activity (mg CO2–C per mg biomass C), suggest that reduced-chemical-input management may cause a larger fraction of the biomass to be relatively inactive but may also increase the activity of the remaining fraction over that in no-till. Thus in these specific systems, the turnover of C and P through the microbial biomass with a reduced chemical input to the soil may be higher than under a no-till system.

An incubation experiment was carried out with maize (Zea mays L.) leaf straw to analyze the effects of mixing the residues with soil and N amendment on the decomposition process. In order to distinguish between soil effects and nitrogen effects for both the phyllospheric microorganisms already present on the surface of maize straw and soil microorganisms the N amendment was applied in two different placements: directly to the straw or to the soil. The experiment was performed in dynamic, automated microcosms for 22 days at 15 °C with 7 treatments: (1) untreated soil, (2) non-amended maize leaf straw without soil, (3) N amended maize leaf straw without soil, (4) soil mixed with maize leaf straw, (5) N amended soil, (6) N amended soil mixed with maize leaf straw, and (7) soil mixed with N amended maize leaf straw. 15NH415NO3 (5 at%) was added. Gas emissions (CO2, 13CO2 and N2O) were continuously recorded throughout the experiment. Microbial biomass C, biomass N, ergosterol, δ13C of soil organic C and of microbial biomass C as well as 15N in soil total N, mineral N and microbial biomass N were determined in soil samples at the end of the incubation. The CO2 evolution rate showed a lag-phase of two days in the non-amended maize leaf straw treatment without soil, which was completely eliminated when mineral N was added. The addition of N generally increased the CO2 evolution rate during the initial stages of maize leaf straw decomposition, but not the cumulative CO2 production. The presence of soil caused roughly a 50% increase in cumulative CO2 production within 22 days in the maize straw treatments due to a slower decrease of CO2 evolution after the initial activity peak. Since there are no limitations of water or N, we suggest that soil provides a microbial community ensuring an effective succession of straw decomposing microorganisms. In the treatments where maize and soil was mixed, 75% of microbial biomass C was derived from maize. We concluded that this high contribution of maize using microbiota indicates a strong influence of organisms of phyllospheric origin to the microbial community in the soil after plant residues enter the soil.

Gross rates of soil processes and microbial activity were measured in two grazed permanent pasture soils which had recently been amended with N fertilizer or dung. 15N studies of rates of soil organic matter turnover showed gross N mineralization was higher, and gross N immobilization was lower, in a long-term fertilized soil than in a soil which had never received fertilizer N. Net mineralization was also found to be higher in the fertilized soil: a consequence of the difference between the opposing N turnover processes of N mineralization and immobilization. In both soils without amendments the soil microbial biomass contents were similar, but biomass activity (specific respiration) was higher in the fertilized soil. Short-term manipulation of fertilizer N input, i.e. adding N to unfertilized soil, or witholding N from previously fertilized soil, for one growing season, did not affect gross mineralization, immobilization or biomass size and activity. Amendments of dung had little effect on gross mineralization, but there was an increase in immobilization in both soils. Total biomass also increased under dung in the unfertilized soil, but specific respiration was reduced, suggesting changes in the composition of the biomass. Dung had a direct effect on the microbial biomass by temporarily increasing available soil C. Prolonged input of fertilizer N increases soil C indirectly as a result of enhanced plant growth, the effect of which may not become evident within one seasonal cycle. There is a growing recognition for the need to develop sensitive indicators of soil quality that reflect the effects of land management on soil and assist land users in promoting sustainability of agro-ecosystems. Three soil enzymes (dehydrogenase, phosphatase and invertase) microbial biomass as biological variables and soil organic matter content (SOM) were investigated relative to fertilization and soil fertility (estimated by crop yield) at a long-term fertilization trial (Keszthely, Hungary). 0-34.7-69.4-104.1t farmyard manure (FYM) ha m 1 5 yr m 1 and the corresponding amount of mineral fertilizers (NPK) were applied in two different crop rotation systems. There were four straw and/or stalk incorporating treatments in the second crop rotation 'B'. Enzyme activities, microbial biomass and the amount of SOM were generally higher in the fertilized soils than in the unfertilized soils. The type of amendments (mineral, FYM or mixed) had significant effects only on the amount of SOM. The correlations among the biological variables and the crop yield were generally low (r < 0.250. The differences in field management resulted only in the invertase activity.

The effect of long-term (45 years) mineral and organic fertilization on soil organic matter (SOM) quantity (organic C and N content) and quality (hot-water-soluble C content, microbial biomass C content, hydrophobic organic components of SOM, soil enzyme activities) was determined in a field experiment established in Trutnov (North Bohemia, sandy loam, Eutric Cambisol). Six treatments were chosen for investigation: unfertilized control, mineral fertilization (NPK), straw N, farmyard manure (FYM) and straw and FYM completed with mineral NPK. Soil samples were taken from the arable layer (0–20 cm) in spring over the period of 2004–2010. The positive effect of FYM on the total organic C and N content, hot-water-soluble C content and hydrophobic organic components of SOM was more than 50% higher than that of straw and mineral N fertilization. Application of straw N increased microbial biomass C content in soil and generated invertase activity above the level of FYM. Hot-water-soluble C content, hydrophobic organic components of SOM and urease activity were positively correlated with total organic C and N content (R = 0.58–0.98; p < 0.05). Addition of mineral NPK to both the straw and FYM emphasized the effect of organic fertilization in most of monitored characteristics.

As labile organic pools, soluble organic matter and soil microbial biomass are sensitive to changes in soil management and therefore good indicators of soil quality. Effects of a 17-year long-term fertilization on soil microbial biomass C (SMBC) and N (SMBN), soluble organic C, and soluble organic N during the maize growing season were evaluated in a loess soil (Eum-Orthic Anthrosol) in northwest China. The fertilization treatments included no fertilizer (CK), inorganic N, P, and K fertilizer (NPK), cattle manure plus NPK fertilizer (MNPK), and straw plus NPK fertilizer (SNPK). Our results showed that C storage in the 0–20 cm soil layer was 28% to 81% higher in the fertilized treatments compared to the unfertilized treatment. In the 0–10 cm soil layer, SMBC and SMBN in the three fertilized treatments were higher than in the unfertilized treatment on all sampling dates, while microbial biomass C and N in the 0−10 cm soil layers were the highest at grain filling. In the same soil layer, soil-soluble organic C generally decreased in the order MNPK > SNPK > NPK > CK, while soluble organic N was the highest in the MNPK followed by the SNPK treatment. There was no significant difference in soluble organic N in the NPK and CK treatments throughout most of the maize growing season. Changes in soluble organic N occurred along the growing season and were more significant than those for soluble organic C. Soluble organic N was the highest at grain filling and the lowest at harvest. Overall, our results indicated that microbial biomass and soluble organic N in the surface soil were generally the highest at grain filling when maize growth was most vigorous. Significant positive relationships were found between soluble organic C and SMBC and between soluble organic N and SMBN.

Silvicultural treatments of fertilization (F) and competing vegetation suppression (H) have continued to increase as demands for forest products have grown. The effects of intensive annual F and H treatments on soil C, N, microbial biomass, and CO2 efflux were examined in a two-way factorial experiment (control, F, H, FxH) in late-rotation (20+ years) loblolly pine stands. This study is unique in testing the cumulative effects of continual H and repeated F treatments for the first 20 years of stand growth, an uncommon operational practice, and in having treatments replicated upon four different soil types in the state of Georgia, USA. Annual fertilization included applications of N, P, K and periodic additions of micronutrients while competing vegetation suppression was maintained for all non-pine vegetation with herbicides throughout the rotation. Measurements included total O-horizon (forest floor) organic matter, C, and N, and 0-10 cm mineral soil pH, C, N, microbial biomass C and N, and surface CO2 efflux. Sample collections and analyses were conducted seasonally for 1.5 yrs. Competing vegetation suppression was associated with a decrease of total soil C, soil microbial biomass C and N, and soil surface CO2 efflux, while increasing O-horizon C:N. The fertilization treatment greatly reduced soil microbial biomass C and N, soil pH, and O-horizon C:N, while increasing O-horizon mass, N content, and soil carbon. No significant interactions between F and H were found. The combination of F and H treatments acted additively to achieve the greatest loss of soil microbial biomass, which may possibly have negative implications for long-term soil fertility.
