莲藕的酶促褐变及其贮藏中褐变的控制   总被引:4,自引:0,他引:4       下载免费PDF全文
为有效控制莲藕褐变,试验研究了温度、pH值、底物浓度以及抑制剂对莲藕多酚氧化酶(PPO)的影响。结果表明:莲藕PPO最适pH值为7.5,最适温度为35℃,以邻苯二酚为底物,米氏常数Km为0.0273 mol/L。亚硫酸钠、抗坏血酸(VC)对莲藕PPO活性具有较好抑制作用,乙酸、柠檬酸、植酸、木瓜蛋白酶、半胱氨酸也对莲藕PPO活性具有一定抑制作用。0.2%VC+2%乙酸+0.03%亚硫酸钠抑制剂处理对莲藕褐变的抑制效果最佳。可以通过浸泡亚硫酸钠和抗坏血酸(VC)、调节pH值、低温贮藏等方法来抑制莲藕PPO活性,控制莲藕褐变。  相似文献

该文通过研究花魔芋和白魔芋球茎中酚类物质含量、多酚氧化酶(Polyphenol Oxidase, PPO)活性、褐变底物和干燥脱水过程中褐变的机理,探讨了无公害酶促褐变抑制剂植酸、抗坏血酸、柠檬酸、L-半胱氨酸对魔芋褐变的抑制效果。结果表明:从11月至次年1月,白魔芋球茎的总酚含量始终高于花魔芋,花魔芋的PPO活力始终强于白魔芋,白魔芋的褐变强度始终弱于花魔芋的褐变强度,褐变强度跟魔芋球茎PPO活力呈正相关。引起魔芋褐变的主要底物可能是多巴胺。引起白魔芋和花魔芋褐变的酶均为多酚氧化酶。对白魔芋采用0.01% L-半胱氨酸单因子抑制剂浸泡5 min,并在60℃下烘烤,所得白魔芋干片颜色白,效果最好。对花魔芋采用0.15% L-半胱氨酸+1%柠檬酸+1%植酸+0.01%抗坏血酸组合抑制剂浸泡5 min,并在60℃下烘烤,所得魔芋干片颜色白,效果最好。  相似文献

CaCl2和6-BA溶液处理对枣果软化褐变及相关酶活性的影响   总被引:4,自引:1,他引:4  
试验探讨了鲜枣贮藏过程中果肉软化褐变与PG活性、PPO活性的关系以及采前CaCl2和6-BA溶液处理对采后枣果果肉软化褐变、PG活性、PPO活性的影响。结果表明,枣果采后硬度逐渐下降且与贮藏时间呈极显著负相关,PG活性在贮藏期间呈起伏变化,与枣果软化无显著相关性。 随贮藏时间延长果肉褐变指数逐渐增大,PPO活性逐渐升高,PPO活性与果肉软化褐变的发生相一致。采前用1% CaCl2、15 mg/kg 6-BA以及1% CaCl2+15 mg/kg 6-BA对枣果进行喷雾处理,能降低采后枣果的PG活性和PPO活性,抑制果肉硬度的下降和果肉的褐变进程。1% CaCl2处理可使赞皇大枣贮藏末期的果肉硬度比对照增加1.2 kg/cm2;1% CaCl2+15 mg/kg 6-BA处理可使赞皇大枣贮藏末期的果肉硬度增加1.5 kg/cm2。15 mg/kg 6-BA处理对枣果软化褐变及PG活性、PPO活性有一定影响,但效果不及CaCl2处理。  相似文献

试验探讨了鲜枣贮藏过程中果肉软化褐变与 PG活性、PPO活性的关系以及采前 Ca Cl2 和 6 - BA溶液处理对采后枣果果肉软化褐变、PG活性、PPO活性的影响。结果表明 ,枣果采后硬度逐渐下降且与贮藏时间呈极显著负相关 ,PG活性在贮藏期间呈起伏变化 ,与枣果软化无显著相关性。随贮藏时间延长果肉褐变指数逐渐增大 ,PPO活性逐渐升高 ,PPO活性与果肉软化褐变的发生相一致。采前用 1% Ca Cl2 、15 mg/kg6 - BA以及 1% Ca Cl2 +15mg/kg6 - BA对枣果进行喷雾处理 ,能降低采后枣果的 PG活性和 PPO活性 ,抑制果肉硬度的下降和果肉的褐变进程。 1% Ca Cl2 处理可使赞皇大枣贮藏末期的果肉硬度比对照增加 1.2 kg/cm2 ;1% Ca Cl2 +15 m g/kg6 - BA处理可使赞皇大枣贮藏末期的果肉硬度增加 1.5 kg/cm2 。 15 m g/kg 6 - BA处理对枣果软化褐变及 PG活性、PPO活性有一定影响 ,但效果不及 Ca Cl2 处理  相似文献

在温度4℃和10℃(冷库贮藏)条件下贮存的紫薯、红薯、白薯,通过检测贮存过程中水分、淀粉、还原糖含量、PPO活性和质构等指标,研究贮藏温度对其品质的影响,为甘薯贮存及加工提供参考。结果表明:贮藏温度对甘薯水分、淀粉和还原糖含量,PPO活性,及硬度、咀嚼性等指标影响显著(P0.05),对弹性、粘结性影响无显著差异(P0.05)。贮藏温度4℃比10℃能够更好地保持甘薯的品质。本研究为甘薯贮存及加工提供参考。  相似文献

为探明石榴果实发育期果皮褐变及其相关酶活性、总酚和抗氧化能力的变化规律及相互关系,以泰山红和泰山三白甜石榴为试材,研究各发育期果皮褐变度、多酚氧化酶(PPO)活性、过氧化物酶(POD)活性、苯丙氨酸解氨酶(PAL)活性、总酚含量和DPPH自由基清除率的变化。结果表明,2个品种果皮褐变度、PPO活性、POD活性、总酚含量和DPPH自由基清除率随着发育天数的增加逐渐降低,均在7月15日达到最高。2个品种果实发育期果皮PAL活性呈先降低后升高再降低的变化趋势,分别在7月15日和9月13日出现峰值。泰山红抗褐变能力高于泰山三白甜,二者分别在9月23日和9月13日采收能减轻褐变的发生。相关性分析表明,PAL、PPO、POD、总酚和DPPH自由基清除率均与石榴果实酶促褐变密切相关,石榴果实酚类物质含量高低与其抗氧化能力极显著正相关。本研究结果为深入揭示石榴果实褐变机理提供了理论依据。  相似文献

快速脱水抑制葡萄干制过程中膜脂过氧化及褐变   总被引:2,自引:1,他引:1  
为探索脱水速度对无核白葡萄干制过程中膜脂过氧化作用及褐变的影响,以新疆无核白葡萄为试验材料,经过葡萄促干剂处理后,采用快速脱水和缓慢脱水2种处理,质量每减轻10%进行取样,测定脱水过程中果实干基含水率、干燥速率、褐变度、细胞膜透性、丙二醛(malonaldehyde,MDA)含量、脂氧合酶(lipoxygenase,LOX)、超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)、过氧化物酶(peroxidase,POD)、多酚氧化酶(polyphenol oxidase,PPO)活性以及总酚含量的变化。结果表明:与缓慢脱水相比,快速脱水处理显著(P0.05)降低了无核白葡萄褐变度的上升,减少MDA生成量以及膜透性的增加,抑制LOX活性的升高,保持较高的活性氧清除酶SOD、CAT及POD活性及较高的总酚含量,且使PPO活性保持在一个较低的水平。因此认为,快速脱水可以有效地抑制无核白葡萄的膜脂过氧化作用对细胞膜的破坏,保持细胞的完整性,且PPO活性较低,从而减少无核白葡萄脱水褐变的发生。研究结果为快速脱水在无核白葡萄干制中的应用提供参考。  相似文献

为探索湿度对甘薯愈伤的影响,本试验以普薯32号甘薯为原料,人工模拟机械损伤后于25℃、不同相对湿度(RH50%和RH80%)条件下愈伤,通过比较甘薯木质素生成、失重率、呼吸强度、愈伤过程关键酶活性、总酚和类黄酮含量的差异,评价湿度对甘薯愈伤的影响。结果表明,RH80%条件下较RH50%更有利于甘薯愈伤木栓组织的形成,在愈伤4 d时甘薯木质素积累达到最高。与RH50%组相比,RH80%条件处理能显著提高甘薯愈伤组织苯丙氨酸解氨酶(PAL)、过氧化物酶(POD)和多酚氧化酶(PPO)活性(P<0.05),提升总酚、类黄酮含量,促进呼吸高峰提前,降低甘薯重量损失。相关性分析表明,RH80%条件下愈伤,甘薯木质素含量与PAL、POD、PPO活性及总酚、类黄酮含量呈显著正相关(P<0.05)。本研究结果为甘薯采后愈伤提供了一定的理论依据。  相似文献

小麦籽粒多酚氧化酶(PPO)活性是影响面团褐变的主要原因,检索NCBI网站上注册的小麦PPO基因并对其进行分类及相关变异的研究,对于弄清PPO基因与籽粒PPO活性的关系,开发分子标记选育出含有低PPO活性基因的品种,改良我国面制食品的外观品质有重要作用。本研究通过对NCBI上注册的小麦PPO基因序列的搜索与比对后发现,现有的小麦PPO基因按表达方式可分为两大类(I、II),其中第II大类的PPO基因与小麦籽粒PPO活性密切相关,第II大类第i小类中的PPO基因可能位于小麦2A、2D以外的染色体上,可做为改良面团色泽的侯选基因。通过对具有完整开放阅读框(ORF)的4条PPO基因比对后发现,位于小麦2D染色体长臂上的PPO基因(PPO-2D)存在丰富的等位变异,等位基因间有94个单核苷酸变异(SNP),其中发生在编码区的有80个(cSNP),这些cSNP中有36个影响到基因编码的氨基酸序列,属非同义cSNP。在非同义cSNP处,设计引物(STS-H),对130个已连续测得两年PPO活性的小麦品种进行PCR扩增,结果发现STS-H在大部分低PPO活性品种中没有扩增出目标片段(a),而大部分高PPO活性品种可以扩增出460bp的目标片段(b)。方差分析表明,a、b两种类型品种的PPO活性均值差异达极显著水平(p<0.01),说明非同义cSNP对小麦籽粒PPO活性有重要影响。与STS01(低PPO活性显性标记)比较后发现,STS-H与STS01是一对互补标记。为提高单显性分子标记的使用效率,根据STS01和STS-H引物各自的特点,研究了能同时扩增两对引物的多重PCR反应体系。  相似文献

为寻求适应分散的小规模生产食用菌的保鲜技术,以鸡腿菇为试验材料,研究了短波紫外线(UV-C)照射对鸡腿菇低温(4℃)和常温(20℃)贮藏保鲜的影响。研究结果表明,UV-C照射处理能够显著延长鸡腿菇的保鲜时间,鸡腿菇在4℃贮藏时的保鲜时间由处理前的6~7 d延长至10~11 d,在常温下的保鲜时间由2 d延长至4 d以上。UV-C照射能够抑制鸡腿菇在贮藏期间的后熟作用,降低多酚氧化酶(PPO)活性,从而降低鸡腿菇的呼吸强度,减小细胞膜透性,延缓褐变反应,尤其能有效地抑制菌柄伸长,从而延长鸡腿菇的保鲜时间。UV-C的照射时间越长,抑制后熟生长、降低PPO活性的作用越明显,但带给鸡腿菇的损伤(如褐变、呼吸增强等)越强烈,因此UV-C的照射时间不宜过长。  相似文献

The effect of minimal processing on polyphenol oxidase (PPO), peroxidase (POD), phenylalanine ammonia-lyase (PAL), and phenolic compounds was studied in five potato cultivars (Agria, Cara, Liseta, Monalisa, and Spunta). Minimal processing caused an overall increase in PPO, POD, and PAL activities. The isoform pattern of PPO was the same for all of the cultivars before and after processing. No latent PPO was detected. The isoperoxidase pattern was approximately the same among cultivars. An increase in POD activity was related to the specific induction of an acidic isoperoxidase. PAL showed an induction pattern characterized by the presence of a maximum peak of activity after 4 days of processing for all of the cultivars. The sequence of browning susceptibility of potato cultivars was as follows: Monalisa > Spunta > Liseta > Cara > Agria. Browning development was only partially correlated to PAL activity (only during the first 4 days after wounding). However, this correlation could not explain the above sequence of browning susceptibility. Minimal processing caused an increase of chlorogenic acid, whereas tyrosine content remained unchanged. In summary, no significant correlation was found between either rate or degree of browning and any other biochemical and physiological attribute investigated (PPO, POD, hydrogen peroxide, ascorbic acid content, and initial phenolics content as well as total and individual phenolics accumulation).  相似文献

Polyphenol oxidase (PPO) activity of Russet Burbank potato was inhibited by sense and antisense PPO RNAs expressed from a tomato PPO cDNA under the control of the 35S promoter from the cauliflower mosaic virus. Transgenic Russet Burbank potato plants from 37 different lines were grown in the field. PPO activity and the level of enzymatic browning were measured in the tubers harvested from the field. Of the tubers from 28 transgenic lines that were sampled, tubers from 5 lines exhibited reduced browning. The level of PPO activity correlated with the reduction in enzymatic browning in these lines. These results indicate that expression of tomato PPO RNA in sense or antisense orientation inhibits PPO activity and enzymatic browning in the major commercial potato cultivar. Expression of tomato PPO RNA in sense orientation led to the greatest decrease in PPO activity and enzymatic browning, possibly due to cosuppression. These results suggest that expression of closely related heterologous genes can be used to prevent enzymatic browning in a wide variety of food crops without the application of various food additives.  相似文献

多酚氧化酶(PPO)是生物体内黑色素合成的关键酶。研究了甜柿叶乙醇提取物、涩柿叶乙醇提取物、甜柿叶水提取物、涩柿叶水提取物对马铃薯多酚氧化酶活性的抑制作用。结果表明,上述4种柿叶提取物对马铃薯多酚氧化酶具有明显的抑制作用,使该酶活力下降50%所需的浓度(IG50)分别为0.21、0.26、0.37、0.45mg/mL。乙醇提取物较水提取物的作用效果更强,甜柿叶提取物的抑制作用比涩柿叶提取物的强,4种柿叶提取物对酶的抑制作用均为非竞争性可逆抑制,其抑制常数(KI=KIS)分别为0.18、0.23、0.34、0.45mg/mL。  相似文献

钾素对食用型甘薯糖代谢相关酶活性的影响   总被引:10,自引:6,他引:4  
为了探讨钾素提高甘薯块根可溶性糖含量的生理基础。选用典型的食用型甘薯品种北京553,设置不同施钾处理,于2009~2010年2个生长季在山东农业大学农学试验站进行试验。采用甘薯块根膨大过程中定期取样的方法,测定块根可溶性糖和淀粉含量及相关酶活性、功能叶蔗糖含量及相关酶活性。结果表明,与对照比较,施用钾肥能显著提高块根产量、可溶性糖及各糖组分含量,其中K2O用量为24 g/m2处理增幅最大,为最适用量。进一步研究发现,适宜供钾处理显著提高了功能叶磷酸蔗糖合成酶活性和蔗糖含量,生育期内平均增幅分别为10.31%和34.13%,同时提高了块根中蔗糖合成酶、不溶性酸性转化酶的活性,生育期内平均增幅为16.47%和3.66%,在提高源端光合产物供应的同时促进蔗糖在库端的卸载,促进块根中淀粉和可溶性糖的积累;适宜供钾处理还提高了块根中-和-淀粉酶的活性,生育期内平均增幅分别为26.06%和14.64%,促进淀粉向可溶性糖转化。此外,适宜供钾处理还显著提高了生长前期和后期块根中可溶性酸性转化酶活性、以及生长后期块根中蔗糖-蔗糖果糖基转移酶活性,促进了葡萄糖、果糖和果聚糖在块根中的积累。在甘薯收获期,块根可溶性糖和淀粉含量分别提高了13.52%和3.02%。即钾肥能够增加块根中蔗糖的供应量、促进块根对蔗糖的吸收、促进淀粉水解,是其提高块根可溶性糖含量的生理原因。  相似文献

Honeys from different floral sources were evaluated for their antioxidant content and for their ability to inhibit enzymatic browning in fruits and vegetables. Antioxidant contents of honeys vary widely from different floral sources, as do their abilities to protect against enzymatic browning. Polyphenol oxidase (PPO) activity was reduced over a range of approximately 2-45% in fruit and vegetable homogenates, corresponding to a reduction in browning index by 2.5-12 units. Soy honey was particularly effective when compared to clover honey, which had a similar antioxidant content. When compared to commercial inhibitors of browning, honeys were less effective; however, in combination they added to the effectiveness of metabisulfite and ascorbic acid. Honey has great potential to be used as a natural source of antioxidants to reduce the negative effects of PPO browning in fruit and vegetable processing.  相似文献

甘薯淀粉产量及相关性状的遗传多样性和关联度分析   总被引:1,自引:0,他引:1  
甘薯[Ipomoea batatas(L.)Lam.]是加工淀粉和燃料乙醇的重要原料,是目前我国最具开发前景的非粮食类新型能源作物。选育高淀粉产量的能源型甘薯新品种是甘薯育种的重要目标。为了获得准确筛选高淀粉产量育种材料的性状指标,提高甘薯高淀粉产量育种效率,缩短育种周期,本研究利用不同甘薯品种(系)的自然变异,根据淀粉产量、不同生长发育阶段的5个主要农艺性状和3个淀粉合成关键酶活性测定结果,利用相似系数和遗传距离矩阵,以类平均法对国内48份不同淀粉产量甘薯种质资源进行了遗传多样性分析,通过关联度分析研究了淀粉产量与不同时期农艺性状、淀粉合成关键酶活性的相关性。结果表明:48份甘薯种质资源材料在不同时期农艺特征差异较大;不同时期农艺性状的聚类结果中,栽后100 d的农艺性状与淀粉产量关联度最大,淀粉产量与该时期的基部分枝数呈极显著负相关(r=0.428),与干率呈极显著正相关(r=0.423),而与最长蔓长、单株结薯数和单株鲜薯重相关性不显著。48份甘薯种质材料在不同时期的酶活聚类结果差异明显。不同时期的甘薯淀粉合成关键酶活性聚类结果中,栽后50 d酶活聚类与淀粉产量聚类结果关联度最大,淀粉产量与该时期测得的ADPG焦磷酸化酶(ADPG-PPase)活性呈负相关关系(r=0.163),与蔗糖合成酶(SS)活性(r=0.101)、蔗糖磷酸合成酶(SPS)活性(r=0.016)呈正相关,但相关性均未达到显著水平。加之淀粉合成关键酶活性测定步骤繁琐,不适宜作为甘薯高淀粉产量育种早期选择的生理指标。在高淀粉产量育种材料筛选时可于栽后100 d对农艺性状进行综合考察,重点考虑干率较高及分枝数较少的品系。本研究可为甘薯高淀粉产量育种提供一定的理论依据。  相似文献

Polyphenol oxidase (PPO) was extracted from five Sicilian varieties of tomato fruit [Pizzutello, Naomi (Hazera), F1 PS212 (Peto seed), Rosa Maletto, and PO228] and assayed with a method using 3-methylbenzothyazolinone hydrazone (MBTH) as chromophore coupling agent. 3,4-Dihydroxyphenylacetic acid was chosen for tomato PPO activity determination. The tomato PPO had maximum activity at pH 4.8. The pH of juice in ripe fruits is between 4.1 and 4.4, a range in which PPO relative activity is between 74 and 87%. The optimum temperature of activity for tomato PPO was 40 degrees C; the enzyme showed a good relative activity (55% of the maximum) at cold-storage temperature (4 degrees C). PPO retained 82% relative activity at an NaCl concentration of 0.1 M; at higher concentrations the PPO became gradually inactivated. The commercial variety Naomi is more susceptible to enzymatic browning than the local varieties Pizzutello, Rosa Maletto and PO228, due to higher PPO activity levels. This result confirms the suitability of these local tomato varieties to national markets. Results from storage tests seem to relate PPO activity with color changes associated with browning and lycopene degradation, because lycopene is an antioxidant agent that reconstitutes the polyphenols oxidized by the action of PPO.  相似文献

Inhibition of polyphenol oxidases activity by various dipeptides   总被引:1,自引:0,他引:1  
In an effort to develop natural and nontoxic inhibitors on the activity of mushroom polyphenol oxidase (PPO) the effect of various glycyl-dipeptides (GlyAsp, GlyGly, GlyHis, GlyLeu, GlyLys, GlyPhe, GlyPro, GlyTyr) was investigated. The inhibition study with dihydroxyphenylalanine (DOPA) as substrate is based on separation of the enzymatic reaction components by reversed phase HPLC and the UV detection of the dopachrome formed. The results have evidenced that several of tested dipeptides inhibited PPO activity in the range of 20-40% while GlyPro and GlyLeu had no effect. The study has also permitted the characterization of the following kinetic pattern: a linear-mixed-type mechanism for GlyAsp, GlyGly, GlyLys, and GlyPhe and a hyperbolic-mixed-type for GlyTyr. It was not possible to identify the inhibition mechanism for GlyHis, although it affects PPO activity. In addition the effects of GlyAsp, GlyLys and GlyHis were evaluated for lessening the browning of fresh Golden Delicious apple and Irish White Skinned potato. The effectiveness of such inhibitors was determined by the difference between the colors observed in the dipeptide-treated sample and the controls using the color space CIE-Lab system. The % browning inhibition on potato (20-50%) was greater than of apple (20-30%) by the all tested dipeptides. Only GlyLys presented the significant value of 50%.  相似文献

探讨制备高品质甘薯全粉的简便方法,及以甘薯全粉为主要原料,通过挤压膨化技术生产甘薯营养食品的可能性,并对挤压膨化过程中营养成分及理化性质的变化进行了研究分析。结果表明,经挤压膨化后,原料中淀粉、蛋白质含量略有下降,总糖含量上升,吸水性,水溶性指数增大,色泽略有加深,并有浓郁的特征香味产生。  相似文献
