Soil organic matter level, mineralizable C and N, microbial biomass C and dehydrogenase, urease and alkaline phosphatase activities were studied in soils from a field experiment under a pearl millet-wheat cropping sequence receiving inorganic fertilizers and a combination of inorganic fertilizers and organic amendments for the last 11 years. The amounts of soil organic matter and mineralizable C and N increased with the application of inorganic fertilizers. However, there were greater increases of these parameters when farmyard manure, wheat straw or Sesbania bispinosa green manure was applied along with inorganic fertilizers. Microbial biomass C increased from 147 mg kg–1 soil in unfertilized soil to 423 mg kg–1 soil in soil amended with wheat straw and inorganic fertilizers. The urease and alkaline phosphatase activities of soils increased significantly with a combination of inorganic fertilizers and organic amendments. The results indicate that soil organic matter level and soil microbial activities, vital for the nutrient turnover and long-term productivity of the soil, are enhanced by use of organic amendments along with inorganic fertilizers.  Long-term experiments on different crop management systems provide essential information about turnover of soil organic matter and changes in microbial properties over a period of time. A long-term field site trial, which was established in 1967 near Vienna, Austria, to document the fate of 14C-labelled manure (straw and farmyard) under different crop management systems (crop rotation, spring wheat and bare fallow), was investigated. Soil samples were taken in 1997 and separated into size fractions (>250 μm, 250–63 μm, 63–2 μm, 2–0.1 μm and <0.1 μm) after aggregate dispersion using low-energy sonication. Organic C, total N and 14C content were measured in the bulk soil and the size fractions and microbial properties were analysed in the bulk soil. Additionally, C mineralization in bulk soil samples was monitored at 20 °C over a period of 28 days, and subsequently 14C-CO2 content was analysed. The distribution of organic C and N within the size fractions was similar between crop rotation and spring wheat; the highest amounts of organic C and N were found in the clay-sized fraction. The amounts of C and N were significantly smaller in the bare fallow, which was depleted of organic matter in the coarse-sized fractions. 14C distribution differed significantly from unlabelled C distribution, labelled C was accumulated in the silt-sized fraction, indicating weak humification of the applied manure C. The highest rate of C mineralization was measured in the crop rotation and spring wheat, whereas the emission rate of the bare fallow was about 40% lower. The higher 14C:C ratio of the bulk soil in comparison to the emitted CO2 indicated that labelled C compounds still remained mineralizable after a period of 30 years. Microbial properties showed a great difference between crop management systems and bare fallow, particularly regarding urease and xylanase activity.

We investigated the soil organic C and N stocks, storage profiles and microbial biomass as influenced by different crop management systems in a tropical agricultural ecosystem. The different crop management systems significantly affected the C and N stocks and microbial biomass C and N at different soil depths. Amongst the systems evaluated, the rice-wheat system maintained a higher soil organic C content. Inclusion of legumes in the system improved the soil organic matter level and also soil microbial biomass activity, vital for the nutrient turnover and long-term productivity of the soil. Irrespective of the cropping system, approximately 58.4%, 25.7% and 15.9% of the C was distributed in 0–15, 15–30 and 30–60 cm depths, respectively.

One of the challenges in organic farming systems is to match nitrogen (N) mineralization from organic fertilizers and crop demand for N. The mineralization rate of organic N is mainly determined by the chemical composition of the organic matter being decomposed and the activity of the soil microflora. It has been shown that long-term organic fertilization can affect soil microbial biomass (MB), the microbial community structure, and the activity of enzymes involved in the decomposition of organic matter, but whether this has an impact on short-term N mineralization from recently applied organic substances is not yet clear. Here, we sampled soils from a long-term field experiment, which had either not been fertilized, or fertilized with 30 or 60 t ha−1 year−1 of farmyard manure (FYM) since 1989. These soil samples were used in a 10-week pot experiment with or without addition of FYM before starting (recent fertilization). At the start and end of this experiment, soil MB, microbial basal respiration, total plant N, and mineral soil N content were measured, and a simplified N balance was calculated. Although the different treatments used in the long-term experiment induced significant differences in soil MB, as well as total soil C and N contents, the total N mineralization from FYM was not significantly affected by soil fertilization history. The amount of N released from FYM and not immobilized by soil microflora was about twice as high in the soil that had been fertilized with 60 t ha−1 year−1 of FYM as compared with the non-fertilized soil (p < 0.05).

Soil organic carbon (SOC), microbial biomass carbon (MBC), their ratio (MBC/SOC) which is also known as microbial quotient, soil respiration, dehydrogenase and phosphatase activities were evaluated in a long-term (31 years) field experiment involving fertility treatments (manure and inorganic fertilizers) and a maize (Zea mays L.)-wheat (Triticum aestivum L.)-cowpea (Vigna unguiculata L.) rotation at the Indian Agricultural Research Institute near New Delhi, India. Applying farmyard manure (FYM) plus NPK fertilizer significantly increased SOC (4.5-7.5 g kg−1), microbial biomass (124-291 mg kg−1) and microbial quotient from 2.88 to 3.87. Soil respiration, dehydrogenase and phosphatase activities were also increased by FYM applications. The MBC response to FYM+100% NPK compared to 100% NPK (193 vs. 291 mg kg−1) was much greater than that for soil respiration (6.24 vs. 6.93 μl O2 g−1 h−1) indicating a considerable portion of MBC in FYM plots was inactive. Dehydrogenase activity increased slightly as NPK rates were increased from 50% to 100%, but excessive fertilization (150% NPK) decreased it. Acid phosphatase activity (31.1 vs. 51.8 μg PNP g−1 h−1) was much lower than alkali phosphatase activity (289 vs. 366 μg PNP g−1 h−1) in all treatments. Phosphatase activity was influenced more by season or crop (e.g. tilling wheat residue) than fertilizer treatment, although both MBC and phosphatase activity were increased with optimum or balanced fertilization. SOC, MBC, soil respiration and acid phosphatase activity in control (no NPK, no manure) treatment was lower than uncultivated reference soil, and soil respiration was limiting at N alone or NP alone treatments.

A long-term experiment, which started in 1971 near Perugia, central Italy, was performed to investigate the effect of different crop residue management practices and rotation systems on some soil properties. Twenty years after the beginning of the experiment, chemical (organic C, total N, humified organic C, humic and fulvic acids), microbiological and biochemical parameters (microbial biomass, global hydrolase activity, dehydrogenase and catalase activities) were investigated. Two crop residue management practices were used in the experiment, i.e. removal (RCR soils) and burial (BCR soils). These treatments were factorially combined with eight rotation systems, i.e. five maize-wheat rotations of different lengths (M-1W, M-2W, M-3W, M-4W and M-5W) and three continuous wheat systems with different fertilization inputs, from 150 to 250 kg N ha–1. Soil samples were collected in the spring of 1991 for chemical determinations, and in the spring and autumn of 1992, 1993 and 1994, for microbiological and biochemical determinations. All soil chemical, microbiological and biochemical parameters investigated showed significant differences depending on the management of the crop residues. The BCR soils showed more favourable characteristics. In contrast, few significant effects were observed in relation to rotation and N-fertilization treatment. Significant correlations were found between organic-C content and all microbiological and biochemical parameters, as well as between the microbiological and biochemical parameters themselves, indicating that organic-C content plays an important role in determining the level of soil enzyme activity and, consequently, of soil fertility. This experiment showed that burying crop residues in soil can be considered good agronomic practice, which may help limit the gradual depletion of soil organic matter and improve the chemical properties of the soil.  In arable soils in Schleswig-Holstein (Northwest Germany) nearly 30% of the total organic C (TOC) stored in former times in the soil has been mineralized in the last 20 years. Microbial biomass, enzyme activities and the soil organic matter (SOM) composition were investigated in order to elucidate if a low TOC level affects microbial parameters, SOM quality and crop yield. Microbial biomass C (Cmic) and enzyme activities decreased in soils with a low TOC level compared to soils with a typical TOC level. The decrease in the Cmic/TOC ratio suggested low-level, steady-state microbial activity. The SOM quality changed with respect to an enrichment of initial litter compounds in the top soil layers with a low TOC level. Recent management of the soils had not maintained a desirable level of humic compounds. However, we found no significant decrease in crop yield. We suggest that microbial biomass and dehydrogenase and alkaline phosphatase activities are not necessarily indicators of soil fertility in soils with a high fertilization level without forage production and manure application.  The effects on soil condition of increasing periods under intensive cultivation for vegetable production on a Typic Haplohumult were compared with those of pastoral management using soil biological, physical and chemical indices of soil quality. The majority of the soils studied had reasonably high pH, exchangeable cation and extractable P levels reflecting the high fertilizer rates applied to dairy pasture and more particularly vegetable-producing soils. Soil organic C (Corg) content under long-term pasture (>60 years) was in the range of 55 g C kg–1 to 65 g C kg–1. With increasing periods under vegetable production soil organic matter declined until a new equilibrium level was attained at about 15–20 g C kg–1 after 60–80 years. The loss of soil organic matter resulted in a linear decline in microbial biomass C (Cmic) and basal respiratory rate. The microbial quotient (Cmic/Corg) decreased from 2.3% to 1.1% as soil organic matter content declined from 65 g C kg–1 to 15 g C kg–1 but the microbial metabolic quotient (basal respiration/Cmic ratio) remained unaffected. With decreasing soil organic matter content, the decline in arginine ammonification rate, fluorescein diacetate hydrolytic activity, earthworm numbers, soil aggregate stability and total clod porosity was curvilinear and little affected until soil organic C content fell below about 45 g C kg–1. Soils with an organic C content above 45 g C kg–1 had been under pasture for at least 30 years. At the same Corg content, soil biological activity and soil physical conditions were markedly improved when soils were under grass rather than vegetables. It was concluded that for soils under continuous vegetable production, practices that add organic residues to the soil should be promoted and that extending routine soil testing procedures to include key physical and biological properties will be an important future step in promoting sustainable management practices in the area.  An incubation experiment lasting 120 days was carried out to ascertain the effect on the soil microbial activity and organic matter mineralization of adding a sewage sludge compost contaminated with two different levels of Cd to an arid soil. Two composts, with a low (2 mg kg–1) and high (815 mg kg–1) Cd content, respectively, were used in this experiment. Both composts increased the total organic C, humic substance and water-soluble C contents, the beneficial effects still being noticeable after 120 days of incubation. The most labile C fraction (water-soluble C) was the most sensitive to the high Cd content. The high Cd concentration decreased soil microbial biomass C and stimulated the metabolic activity of the microbial biomass, the metabolic quotient (qCO2) revealing itself to be a very sensitive index of the stress that the incorporation of a Cd-contaminated sewage sludge compost causes in a soil. The effect of Cd contamination on enzyme activities (urease, protease that hydrolyse N-α-benzoil-l-arginamide, phosphatase, and β-glucosidase) depended on the enzyme studied. To assess the effect of long-term fertilization on labile organic matter fractions, we analyzed the C and N mineralization and C and N content in soil, particulate organic matter (POM), light fraction organic matter (LFOM), and microbial biomass. Results showed that fertilizer N decreased or did not affect the C and N amounts in soil fractions, except N mineralization and soil total N. The C and N amounts in soil and its fractions increased with the application of fertilizer PK and rice straw. Generally, there was no significant difference between fertilizer PK and rice straw. Furthermore, application of manure was most effective in maintaining soil organic matter and labile organic matter fractions. Soils treated with manure alone had the highest microbial biomass C and C and N mineralization. A significant correlation was observed between the C content and N content in soil, POM, LFOM, microbial biomass, or the readily mineralized organic matter. The amounts of POM–N, LFOM–N, POM–C, and LFOM–C closely correlated with soil organic C or total N content. Microbial biomass N was closely related to the amounts of POM–N, LFOM–N, POM–C, and LFOM–C, while microbial biomass C was closely related to the amounts of POM–N, POM–C, and soil total N. These results suggested that microbial biomass C and N closely correlated with POM rather than SOM. Carbon mineralization was closely related to the amounts of POM–N, POM–C, microbial biomass C, and soil organic C, but no significant correlation was detected between N mineralization with C or N amounts in soil and its fractions.

Effects of conventional tillage on biochemical properties of soils
Modification of soil environment by different farming practices can significantly affect crop growth. Tillage causes soil disturbance, altering the vertical distribution of soil organic matter and plant nutrient supplies in the soil surface, and it may affect the enzyme activity and microbial biomass which are responsible for transformation and cycling of organic matter and plant nutrients. In this study, the influence of three conventional tillage systems (shallow plowing, deep plowing and scarification) at different depths on the distribution and activity of enzymes, microbial biomass and nucleic acids in a cropped soil was investigated. Analysis of variance for depth and tillage showed the influence of the different tillage practices on the activity of some enzymes and on the nucleic acids. Glucosidase, galactosidase, nitrate reductase and dehydrogenase activity were significantly affected by the three tillage modalities. Activity in the upper layer (0–20 cm) was higher in the plots tilled by shallow plowing and scarification than in those tilled by deep plowing. Positive relationships were observed between the soil enzymes themselves, with the exception of urease and pyrophosphatase activity. Moreover, significant correlations were found between DNA and β-galactosidase, and between RNA and β-glucosidase, β-galactosidase, alkaline phosphatase and phosphodiesterase. α-Glucosidase, β-galactosidase, alkaline phosphatase and phosphodiesterase were highly correlated with biomass C determined by the fumigation-extraction method.

We studied the influence of soil compaction in a loamy sand soil on C and N mineralization and nitrification of soil organic matter and added crop residues. Samples of unamended soil, and soil amended with leek residues, at six bulk densities ranging from 1.2 to 1.6 Mg m–3 and 75% field capacity, were incubated. In the unamended soil, bulk density within the range studied did not influence any measure of microbial activity significantly. A small (but insignificant) decrease in nitrification rate at the highest bulk density was the only evidence for possible effects of compaction on microbial activity. In the amended soil the amounts of mineralized N at the end of the incubation were equal at all bulk densities, but first-order N mineralization rates tended to increase with increasing compaction, although the increase was not significant. Nitrification in the amended soils was more affected by compaction, and NO3 -N contents after 3 weeks of incubation at bulk densities of 1.5 and 1.6 Mg m–3 were significantly lower (by about 8% and 16% of total added N, respectively), than those of the less compacted treatments. The C mineralization rate was strongly depressed at a bulk density of 1.6 Mg m–3, compared with the other treatments. The depression of C mineralization in compacted soils can lead to higher organic matter accumulation. Since N mineralization was not affected by compaction (within the range used here) the accumulated organic matter would have had higher C : N ratios than in the uncompacted soils, and hence would have been of a lower quality. In general, increasing soil compaction in this soil, starting at a bulk density of 1.5 Mg m–3, will affect some microbially driven processes.

Little information is available on soil microbial and biochemical properties, important for understanding nutrient cycling and organic matter dynamics, as affected by different peanut cropping systems and how they relate to soil functioning. Thus, we studied a Tifton loamy sand (fine-loamy, kaolinitic, thermic Plinthic Kandiudults) in Georgia, which is first in peanut production in USA, after 5 and 8 years under continuous cotton (Gossypium hirsutum, L) (CtCtCt), cotton–cotton–peanut (CtCtPt), corn (Zea mays L.)–peanut–cotton (CrPtCt), peanut–peanut–cotton (PtPtCt), and continuous peanut (PtPtPt). Soil organic carbon (OC) at 0–20 cm was already higher under PtPtPt (average, 8.7 g C kg−1 soil), PtPtCt (average, 7.7 g C kg−1 soil), and CrPtCt (average, 7.8 g C kg−1 soil) compared with CtCtPt (average, 4.7 g C kg−1 soil) and CtCtCt (average, 3.3 g C kg−1 soil). Similarly, alkaline phosphatase, acid phosphatase, and phosphodiesterase as a group showed higher activities under PtPtPt, PtPtCt, and CrPtCt than under CtCtPt and CtCtCt. The activities of glycosidases (α-galactosidase, β-glucosidase, and β-glucosaminidase) as a group were more sensitive to the cropping systems than phosphastases and showed a distinctive cropping system separation as follows: PtPtPt = CrPtCt > PtPtCt > CtCtPt > CtCtCt. Similar to OC and microbial biomass C trends, distinctive differences were found in the microbial community structure of this sandy soil after 8 years between peanut-based cropping systems (CrPtCt, PtPtCt, and PtPtPt) and cotton-based cropping systems (CtCtCt and CtCtPt) as indicated by the fatty acid methyl esters profiles. Trade names and company names are included for the benefit of the reader and do not infer any endorsement or preferential treatment of the product by USDA-ARS.

The effects of growing trees in combination with field crops on soil organic matter, microbial biomass C, basal respiration and dehydrogenase and alkaline phosphatase activities were studied in soils under a 12-year-old Dalbergia sissoo (a N2-fixing tree) plantation intercropped with a wheat (Triticum aestivum) – cowpea (Vigna sinensis) cropping sequence. The inputs of organic matter through D. sissoo leaf litter increased and crop roots decreased with the increase in tree density. Higher organic C and total N, microbial biomass C, basal soil respiration and activities of dehydrogenase and alkaline phosphatase were observed in treatments with tree-crop combination than in the treatment without trees. Soil organic matter, microbial biomass C and soil enzyme activities increased with the decrease in the spacing of the D. sissoo plantation. The results indicate that adoption of the agroforestry practices led to an improved organic matter status of the soil, which is also reflected in the increased nutrient pool and microbial activities necessary for long-term productivity of the soil. However, tree spacing should be properly maintained to minimize the effects of shading on the intercrops.

Short-term effects of tillage systems on active soil microbial biomass
Conservation tillage, and especially no-tillage, induce changes in the distribution of organic pools in the soil profile. In long-term field experiments, marked stratification of the total soil microbial biomass and its activity have been observed as consequence of the application of no-tillage to previously tilled soils. Our objective was to study the evolution of the total and active soil microbial biomass and mineralized C in vitro during the first crop after the introduction of no-tillage to an agricultural soil. The experiment was performed on a Typic Hapludoll from the Argentinean Pampa. Remaining plant residues, total and active microbial biomass and mineralized C were determined at 0–5 cm and 5–15 cm depths, at three sampling times: wheat tilling, silking and maturity. The introduction of no-tillage produced an accumulation of plant residues in the soil surface layer (0–5 cm), showing stratification with depth at all sampling dates. Active microbial biomass and C mineralization were higher under no-tillage than under conventional tillage in the top 5 cm of the profile. The total soil microbial biomass did not differ between treatments. The active soil biomass was highly and positive correlated with plant residues (r 2=0.617;P<0.01) and with mineralized C (r 2=0.732;P<0.01). Consequently, the active microbial biomass and mineralized C reflected immediately the changes in residue management, whereas the total microbial biomass seemed not to be an early indicator of the introduction of a new form of soil management in our experiment. A better understanding of soil microbial processes is required to improve the synchrony between nutrient release from plant residues and crop demand. Phospholipid fatty acid analysis was used to investigate the effect of two crop rotations (continuous maize and maize-crotalaria rotation) and P fertilization (0 and 50 kg P ha−1 yr−1, applied as triple superphosphate) on microbial community composition in a highly weathered soil from western Kenya. Microbial substrate use in soils from the field experiment was compared in incubation experiments. Higher levels of soil organic matter and microbial biomass in the maize-crotalaria rotation were connected with higher total amounts of phospholipid fatty acids and an increase in the relative abundances of indicators for fungi and gram-negative bacteria. P fertilization changed the community profile only within the continuous maize treatment. The decomposition of glucose, cellulose and three plant residues (all added at 2.5 g C kg−1 soil) proceeded faster in soil from the maize-crotalaria rotation, but differences were mostly transient. Microbial P and N uptake within one week increased with the water-soluble carbon content of added plant residues. More P and N were taken up by the greater microbial biomass in soil from the maize-crotalaria rotation than from continuous maize. Re-mineralization of nutrients during the decline of the microbial biomass increased also with the initial biological activity of the soil, but occurred only for a high quality plant residue within the half year incubation period. Compared to the effect of crop rotation, P fertilization had a minor effect on microbial community composition and substrate use.

Nitrogen and carbon mineralization of cattle manure (N=6 g kg–1; C:N=35), pressmud (N=17.4 g kg–1; C:N=22), green manure (N=26.8 g kg–1; C:N=14) and poultry manure (N=19.5 g kg–1; C:N=12) and their influence on gaseous N losses via denitrification (using the acetylene inhibition technique) in a semiarid subtropical soil (Typic Ustochrepts) were investigated in a growth chamber simulating upland, nearly saturated, and flooded conditions. Mineralization of N started quickly in all manures, except pressmud where immobilization of soil mineral N was observed for an initial 4 days. Accumulation of mineral N in upland soil plus denitrified N revealed that mineralization of cattle manure-, pressmud-, poultry manure- and green manure-N over 16 days was 12, 20, 29 and 44%, respectively, and was inversely related to C:N ratio (R 2=0.703, P=0.05) and directly to N content of organic manure (R 2=0.964, P=0.01). Manure-C mineralized over 16 days ranged from 6% to 50% in different manures added to soil under different moisture regimes and was, in general, inversely related to initial C:N ratio of manure (R 2=0.690, P=0.05). Cumulative denitrification losses over 16 days in control soils (without manure) under upland, nearly saturated, and flooded conditions were 5, 23, and 24 mg N kg–1, respectively. Incorporation of manures enhanced denitrification losses by 60-82% in upland, 52–163% in nearly saturated, and 26–107% in flooded soil conditions over a 16-day period, demonstrating that mineralized N and C from added manures could result in 2- to 3-fold higher rate of denitrification. Cumulative denitrification losses were maximal with green manure, followed by poultry manure, pressmud and cattle manure showing an increase in denitrification with increasing N content and decreasing C:N ratio of manure. Manure-amended nearly saturated soils supported 14–35% greater denitrification than flooded soils due to greater mineralization and supply of C.

Effects of increased soil temperature on soil microbial biomass and dehydrogenase activity were examined on organic (O) horizon material in a low-elevation spruce-fir ecosystem. Soil temperature was maintained at 5  °C above ambient during the growing season in the experimental plots, and soil temperature, moisture, microbial biomass, and dehydrogenase activity were measured during the experiment. An incubation study was also conducted under three temperature regimes, 5, 15, and 25  °C, and under four moisture regimes of 20, 120, 220, and 320% to further evaluate these environmental factors on dehydrogenase activity and microbial biomass. Soil moisture content and microbial biomass controls were significantly lower (30% and 2 μg g–1 soil, respectively) in the heated plots during the treatment period, suggesting that moisture content was important in controlling microbial biomass. In the incubation study, temperature appeared more important than moisture in controlling microbial biomass and dehydrogenase activity. Increasing temperature between 5  °C and 25  °C resulted in significant decreases in microbial biomass and dehydrogenase activity.
