The organic acids and phenolics compositions of nine wild edible mushrooms species (Suillus bellini, Tricholomopsis rutilans, Hygrophorus agathosmus, Amanita rubescens, Russula cyanoxantha, Boletus edulis, Tricholoma equestre, Suillus luteus, and Suillus granulatus) were determined by HPLC-UV and HPLC-DAD, respectively. The antioxidant potential of these species was also assessed by using the DPPH* scavenging assay. The results showed that all of the species presented a profile composed of at least five organic acids: oxalic, citric, malic, quinic, and fumaric acids. In a general way, the pair of malic plus quinic acids were the major compounds. Only very small amounts of two phenolic compounds were found in some of the analyzed species: p-hydroxybenzoic acid (in A. rubescens, R. cyanoxantha, and T. equestre) and quercetin (in S. luteus and S. granulatus). All of the species exhibited a concentration-dependent scavenging ability against DPPH*. T. rutilans revealed the highest antioxidant capacity.  相似文献

Quantitation of nine organic acids in wild mushrooms   总被引:3,自引:0,他引:3  
The organic acids composition of six wild edible mushroom species (Amanita caesarea, Boletus edulis, Gyroporus castaneus, Lactarius deliciosus, Suillus collinitus, and Xerocomus chrysenteron) was determined by an HPLC-UV detector method. The results showed that all of the samples presented a profile composed of at least five organic acids: citric, ketoglutaric, malic, succinic, and fumaric acids. Several samples also contained oxalic, ascorbic, quinic, and shikimic acids. In a general way, the quantitation of the identified compounds indicated that malic acid, followed by the pair citric plus ketoglutaric acids, were the main compounds in the analyzed species, with the exception of A. caesarea, in which malic and ascorbic acids were the most abundant compounds. The relative amounts and the presence/absence of each identified compound may be useful for the differentiation of the species.  相似文献

The relationship between the organic nitrogen status of Egyptian broomrape and one of its hosts, carrot, was studied by comparing amino acid profiles of leaf and root tissues of nonparasitized and broomrape-parasitized carrot plants and by analyzing amino acid profiles of broomrape at different growth stages. Total N concentrations, expressed as a percentage of the dry weight of the tissues, were similar in leaves of nonparasitized and parasitized carrot plants but were lower in parasitized roots than in nonparasitized roots. In both dry and germinated broomrape seeds, N concentrations were lower than or similar to those in broomrape tubercles, shoots, or callus. Individual amino acid concentrations in hydrolysates of leaves of parasitized carrot plants tended to be similar to or greater than those in hydrolysates of nonparasitized carrot plants. Roots of parasitized plants tended to have similar or lower amino acid concentrations than roots of nonparasitized plants. Dry and germinated broomrape seeds had similar amino acid profiles, but individual amino acid concentrations were lower than in the other broomrape tissues examined. The broomrape shoot tended to have lower amino acid concentrations than the tubercle and callus. Free amino acid profiles of leaves and roots of parasitized plants paralleled those of nonparasitized plants, respectively. Individual free amino acids tended to occur at similar or lower levels in dry and germinated broomrape seeds than in the tubercle, shoot, or callus. Free amino acid composition of the broomrape tubercle was similar to that of the parasitized root. Arginine and alanine concentrations in broomrape callus were dramatically higher than those of other amino acids in this or other tissues investigated. These results indicate that changes in the composition of both free and bound amino acids in carrot are associated with broomrape parasitism.  相似文献

This work reports the results for the composition of robusta and arabica coffee species in terms of their amino acid enantiomers in the green and roasted states. The analyses were conducted for the free amino acids, as well as for the amino acids obtained after acid hydrolysis. The amino acids were extracted/hydrolyzed and isolated by SPE on strong cation exchange columns, derivatized to their N-ethoxycarbonylheptafluorobutyl esters, and analyzed by gas chromatography/FID on a Chirasil l-Val column. Multivariate analyses applied to the results showed that the free amino acids can be used as a tool for discrimination between coffee species, with a special reference to l-glutamic acid, l-tryptophan, and pipecolic acid. There is also some evidence that these compounds can be used for discrimination between green coffees subjected to different postharvest processes. It is also shown that the amino acid levels observed after acid hydrolysis can be used for the same purposes, although displaying less discriminatory power.  相似文献

The changes in amino acid composition that occur with maturity of the Noble cultivar of the Vitis rotundifolia Michx. (muscadine) grape were determined by HPLC. Eighteen amino acids were identified. Histidine was the most prominent amino acid followed by alanine. The concentrations of most of the major amino acids (alanine, glycine, histidine, valine, isoleucine, aspartic acid, and serine) were highest at verasion. Glutamine and threonine contents dropped sharply after fruit set, while those of arginine and proline increased gradually with maturity and ripening. Tyrosine content increased gradually with maturity and ripening following a slight drop after fruit set. In ripe grapes, seeds contained most of the amino acids in mature grapes (50%) followed by the pulp (23%), the juice (15%), and the skin (11%). Alanine, histidine, and arginine were the principal amino acids identified in the juice. Alanine, histidine, arginine, valine, glutamine, aspartic acid, proline, serine, and threonine accounted for about 90% of the amino acids in the pulp. In seeds, alanine, proline, asparagine, and histidine accounted for over 55% of the amino acids, while alanine and histidine were found to be the predominant free amino acids in the skin. The profile indicates some differences in the changes in amino acid composition with berry maturity and relative amounts of amino acids present in muscadine compared to those in nonmuscadine grape species.  相似文献

This paper presents a large amount of data on the composition of quince fruit with regard to phenolic compounds, organic acids, and free amino acids. Subsequently, principal component analysis (PCA) is carried out to characterize this fruit. The main purposes of this study were (i) the clarification of the interactions among three factors-quince fruit part, geographical origin of the fruits, and harvesting year-and the phenolic, organic acid, and free amino acid profiles; (ii) the classification of the possible differences; and (iii) the possible correlation among the contents of phenolics, organic acids, and free amino acids in quince fruit. With these aims, quince pulp and peel from nine geographical origins of Portugal, harvested in three consecutive years, for a total of 48 samples, were studied. PCA was performed to assess the relationship among the different components of quince fruit phenolics, organic acids, and free amino acids. Phenolics determination was the most interesting. The difference between pulp and peel phenolic profiles was more apparent during PCA. Two PCs accounted for 81.29% of the total variability, PC1 (74.14%) and PC2 (7.15%). PC1 described the difference between the contents of caffeoylquinic acids (3-O-, 4-O-, and 5-O-caffeoylquinic acids and 3,5-O-dicaffeoylquinic acid) and flavonoids (quercetin 3-galactoside, rutin, kaempferol glycoside, kaempferol 3-glucoside, kaempferol 3-rutinoside, quercetin glycosides acylated with p-coumaric acid, and kaempferol glycosides acylated with p-coumaric acid). PC2 related the content of 4-O-caffeoylquinic acid with the contents of 5-O-caffeoylquinic and 3,5-O-dicaffeoylquinic acids. PCA of phenolic compounds enables a clear distinction between the two parts of the fruit. The data presented herein may serve as a database for the detection of adulteration in quince derivatives.  相似文献

Whole cell fatty acid (WCFA) compositions of three different structures of ectomycorrhizal (ECM) fungi: sporocarps, pure culture mycelia and ectomycorrhizas were analysed to evaluate the potential use of fatty acid profiles as biomarkers for ECM fungi and ectomycorrhiza-associated bacteria. Sporocarps of Amanita muscaria, Amanita rubescens, Lactarius rufus, Lactarius thejogalus, Leccinum scabrum, Paxillus involutus, Russula foetens, Russula rosea, Russula vesca, Suillus grevillei, Tylopilus felleus, Xerocomus badius, Xerocomus subtomentosus, pure cultures of A. muscaria, P. involutus, X. badius, X. subtomentosus, Suillus bovinus Suillus luteus and seven ectomycorrhizal morphotypes of Norway spruce were examined. Our results revealed species-specific composition of fatty acids of fungal sporocarps and pure culture mycelia. Ectomycorrhizal morphotypes distinguished and identified by morphological and molecular methods (PCR-RLFP and sequencing) created specific fatty acid profiles. The dominating fatty acids in pure cultures and sporocarps were 18:2ω6,9, 18:1ω9 and 16:0, whereas ectomycorrhizas also contained plant and bacterial specific fatty acids. Especially, fatty acids specific to Gram-positive bacteria 15:0 anteiso and 17:0 anteiso were present in relatively high amounts and suggested that these bacteria are dominating in the examined Norway spruce mycorrhizosphere. In conclusion, our results show that fatty acid based methods can be useful in studies of ectomycorrhizal fungi, both as a quick method for differentiation of fungal species and also in studies of mycorrhiza-associated microorganisms in the field.  相似文献

The effects of processing and cooking practices on the chemical composition and antioxidant activity of Portuguese wild edible mushroom species (Lactarius deliciosus, Macrolepiota mastoidea, Macrolepiota procera, and Sarcodon imbricatus) were investigated. Dried, frozen, and cooked samples were analyzed for proximate constituents (moisture, fat, crude protein, ash, and carbohydrates) and nutritional value. Fatty acid and sugar profiles were also obtained by gas-liquid chromatography/flame ionization detection and high-performance liquid chromatography/refraction index, respectively. The antioxidant properties were evaluated by several biochemical assays: 2,2-diphenyl-1-picrylhydrazyl radical scavenging capacity, reducing power, inhibition of beta-carotene bleaching, and inhibition of lipid peroxidation in brain tissue using thiobarbituric acid reactive substances. Results of this study show that mushroom species and processing and cooking practices are all effective determinants for either chemical composition or antioxidant properties. Cooked samples proved to have lower nutrient concentrations and lower antioxidant activities than either dried or frozen samples. In what concerns fatty acids and sugar individual profiles, only cooking proved to be relevant: The cooked samples presented higher monounsaturated fatty acid and lower polyunsaturated fatty acid and sugars contents.  相似文献

Twenty-one free amino acids present in several samples of quince fruit (pulp and peel) and quince jam (homemade and industrially manufactured) were analyzed by GC/FID. The analyses showed some differences between quince pulps and peels. Generally, the highest content in total free amino acids and in glycine was found in peels. As a general rule, the three major free amino acids detected in pulps were aspartic acid, asparagine, and hydroxyproline. For quince peels, usually, the three most abundant amino acids were glycine, aspartic acid, and asparagine. Similarly, for quince jams the most important free amino acids were aspartic acid, asparagine, and glycine or hydroxyproline. This study suggests that the free amino acid analysis can be useful for the evaluation of quince jam authenticity. It seems that glycine percentage can be used for the detection of quince peel addition while high alanine content can be related to pear addition.  相似文献

The importance of fresh seedlings for human consumption on European markets continues to increase. Although the contents of free amino acids and potentially toxic free nonprotein amino acids in these fresh and supposedly healthy seedlings is very different from those of the seeds, the crude composition is never mentioned on commercial packages. A commercial product containing seven different kinds of fresh seedlings including kamut, adzuki bean, chickpea, mungbean, pinto bean, garden pea, and lentil has been analyzed by HPLC. Per 100 g of fresh product, 548.2 mg of total free amino acids was found, of which 56.7 mg is free nonprotein amino acids including beta-(isoxazolin-5-on-2-yl)alanine, homoserine, and isowillardiine and the plant hormone trigonelline (N-methylnicotinic acid). The highest amounts of free nonprotein amino acids and trigonelline are found in garden pea (28.3 mg/100 g), mungbean (9.59 mg/100 g), and lentil (7.50 mg/100 g) seedlings. Trigonelline is present in all legume seedlings examined.  相似文献

Effect of potassium nutrition on the content and the spectrum of soluble amino compounds in Red Clover 1. A better K nutrition of Red Clover (Trifolium pratense) resulted in higher contents of soluble amino acids of the upper plant parts. The plants of the treatment with the highest K application showed nearly double as high contents of glutamic acid, aspartic acid, alanine and γ-aminobutyric acid than the plants of the K0 treatment. The content of asparagine was not as much affected by the K nutrition and the content of glutamine was even lowered. 2. Red Clover responded on the K treatment with its content of soluble amino compounds quite different than non leguminous plant species which show decreasing contents of soluble amino acids with an increasing K nutrition. Therefore it is concluded, that the K nutrition of legumes affects the N2-fixation of Rhizobium leguminosarum. 3. The higher contents of soluble amino compounds did not influence the protein content of the upper plant parts. But with the better K supply the yields were increased considerably and therefore also higher yields of proteins were harvested. Very low K supply resulted in a high protein content, due to an inhibition of plant growth.  相似文献

Only a minor part of Maillard reaction studies in the literature focused on the reaction between carbohydrates and peptides. Therefore, in continuation of a previous study in which the influence of the peptide C-terminal amino acid was investigated, this study focused on the influence of the peptide N-terminal amino acid on the production of pyrazines in model reactions of glucose, methylglyoxal, or glyoxal. Nine different dipeptides and three tripeptides were selected. It was shown that the structure of the N-terminal amino acid is determinative for the overall pyrazine production. Especially, the production of 2,5(6)-dimethylpyrazine and trimethylpyrazine was low in the case of proline, valine, or leucine at the N-terminus, whereas it was very high for glycine, alanine, or serine. In contrast to the alkyl-substituted pyrazines, unsubstituted pyrazine was always produced more in the case of experiments with free amino acids. It is clear that different mechanisms must be responsible for this observation. This study clearly illustrates the capability of peptides to produce flavor compounds such as pyrazines.  相似文献

The Organization for Economic Co-operation and Development (OECD) recommends the measurement of specific plant components for compositional assessments of new biotechnology-derived crops. These components include proximates, nutrients, antinutrients, and certain crop-specific secondary metabolites. A considerable literature on the natural variability of these components in conventional and biotechnology-derived crops now exists. Yet the OECD consensus also suggests measurements of any metabolites that may be directly associated with a newly introduced trait. Therefore, steps have been initiated to assess natural variation in metabolites not typically included in the OECD consensus but which might reasonably be expected to be affected by new traits addressing, for example, nutritional enhancement or improved stress tolerance. The compositional study reported here extended across a diverse genetic range of maize hybrids derived from 48 inbreds crossed against two different testers. These were grown at three different, but geographically similar, locations in the United States. In addition to OECD analytes such as proximates, total amino acids and free fatty acids, the levels of free amino acids, sugars, organic acids, and selected stress metabolites in harvested grain were assessed. The major free amino acids identified were asparagine, aspartate, glutamate, and proline. The major sugars were sucrose, glucose, and fructose. The most predominant organic acid was citric acid, with only minor amounts of other organic acids detected. The impact of genetic background and location was assessed for all components. Overall, natural variation in free amino acids, sugars, and organic acids appeared to be markedly higher than that observed for the OECD analytes.  相似文献

为探究不同栽培基质对侧耳属食用菌风味成分的影响以及不同品种食用菌间的风味成分差异,本研究采用顶空固相微萃取结合气相色谱-质谱联用(HS-SPME-GC-MS)技术对桑枝、杂木屑和棉籽壳基质上栽培的6种侧耳属食用菌风味成分进行了分析。结果表明,18个样品中共检出47种挥发性化合物,包括13种醛类化合物、9种醇类化合物、9种酮类化合物、2种呋喃类化合物、2种酯类化合物等。18个样品中的共有成分有17种,包括正己醛、壬醛、1-辛烯-3-醇、3-辛酮、1-辛烯-3-酮、2-正戊基呋喃等。18个样品中挥发性化合物的种类和相对含量差异较大,但棉籽壳基质上栽培的6个菌株子实体中均出现了较高含量的酯类化合物。所有样品中的关键风味成分(ROAV≥1)均包括1-辛烯-3-酮和1-辛烯-3-醇,桑枝基质和杂木屑基质栽培的部分菌株中关键风味成分还包括正己醛、壬醛和2-壬烯醛等。18个样品中总5'-核苷酸含量和总游离氨基酸含量分别为7.86~43.86 mg·g-1 DW和8.54~25.34 mg·g-1 DW, 且杂木屑基质栽培的所有菌株中总游离氨基酸含量均高于桑枝基质和棉籽壳基质。综上所述,棉籽壳基质有利于侧耳属食用菌中酯类化合物的生成,而杂木屑基质有利于游离氨基酸的生成。本研究结果为侧耳属食用菌栽培及风味成分的开发利用提供了科学依据。  相似文献

Extraction of free amino acids from fungal spores (Phycornyces blakesleeanus) Because extraction conditions are crucial for the reliability of biochemical analysis of fungal spore materia1, comparative investigations on methods were conducted, using the amount of free amino acids as a parameter of exhaustive extraction. It was found that optimal conditions are: Extraction in a Soxhlet apparatus with methanol as extracting medium (for 8 hours under reduced pressure and 40°C boiling temperature) or treatment with ice-cold perchloric acid (10 % for 30 minutes) or trichloroacetic acid (10 % for 30 minutes). A pretreatment of the material by desintegration was not necessary. Extraction with hot water (70 or l00T) or with ethanol, propanol, butanol, 3,3 %perchloric or trichloroacetic acid proved to be inappropriate. All of the common amino acids were detected in the extracts except proline and phenylalanine. A few amino acids make up the majority of the pool (glutamic acid, glutamine, aspartic acid, asparagine, alanine and y-butyric acid). These amino acids comprised about 80% of the total intracellular pool (ca. 12000 moles/l00mg spores) and account for nearly 2% of the spore dry weight. Sacrosine, probably being present in considerable amounts, could not be determined quantitatively.  相似文献

To check the influence of the conservation procedure in the chemical composition of chanterelle mushroom, phenolic compounds and organic acids of samples preserved under four different conditions (drying, freezing, conservation in olive oil and in vinegar) were determined. Phenolics and organic acids were analyzed by HPLC-DAD and HPLC-UV, respectively. The results showed that chanterelle is characterized by the presence of six phenolic compounds (3-, 4-, and 5-O-caffeoylquinic acid, caffeic acid, p-coumaric acid, and rutin) and five organic acids (citric, ascorbic, malic, shikimic, and fumaric acids). Samples preserved in olive oil also exhibited hydroxytyrosol, tyrosol, luteolin, and apigenin, whereas conservation in vinegar led to the detection of hydroxytyrosol, tyrosol, and tartaric acid in the analyzed samples. The conservation procedures to which chanterelle samples were subjected seem to affect the qualitative and quantitative phenolics and organic acids profiles.  相似文献

Substantial amounts of low molecular weight organic compounds (LMWs) such as sugars and amino acids are transferred from plant roots into soil. These substances are released due to decomposition processes or leaching (exudation). Afterwards they can be metabolized by soil microorganisms into different compounds, or they can be partially re‐absorbed by the plants. The aim of this study was to clarify the influence of five wild plant species on the composition and pool sizes of LMWs extractable from three different soils. Four of the five species caused significant changes in soil LMW pools. In Chernozem, the sugar concentrations of soil with plants were up to 60 % higher than those of the bulk reference soil, and amino acids increased by as much as 207 %. The relative abundance of free amino acids in roots did not correlate with the relative abundance of amino acids in soil after six weeks of plant growth. The relative abundance of soil amino acids, that increased after plant growth, was strongly dependent on the type of soil and on the plant species present. We suggest that rather than rhizodeposition being dependent on soil type, it reflects differential microbial metabolization of amino acids in the respective soils.  相似文献
