Anchote (Coccinia abyssinica) is plant endemic in Ethiopia with a high calcium content grown for its edible tuberous roots. In spite of its importance as food security crop, there is no information available on molecular genetic diversity of this crop. Therefore, the aim of this study was to assess genetic diversity within and among 12 populations of anchote using ISSR markers. Using nine ISSR primers, a total of 87 scorable bands was generated of which 74 were polymorphic. Within population diversity based on polymorphic bands ranged from 13.8 to 43.53 % with a mean of 33.05 %, Nei’s genetic diversity of 0.04–0.156 with a mean of 0.12, Shannon information index of 0.07–0.23 with a mean of 0.175 and analysis of molecular variation (AMOVA) of 51.4 % were detected. With all diversity parameters, the highest diversity was obtained from Gimbi, Bedele and Ale populations, whilst the lowest was from Manna. AMOVA showed a 48.56 % between populations variability and significantly lower than that of within population variation. Population differentiation with FST was 48.56 %. From Jaccard’s pairwise similarity coefficient, Decha and Nedjo were most related populations exhibiting 0.76 similarity and Manna and Nedjo were the most distantly related populations with similarity of 0.52. The only pentanucliotide primer used in the study, Primer 880 (GGAGA)3, showed a unique band in some individuals that appeared to be associated with morphological quantitative traits (lowest seed number, high protein content, largest fruit size and smallest vine length). Illubabor and Gimbi populations exhibited highest genetic diversity so that the populations should be considered as the primary sites in designing conservation areas for this crop.  相似文献

Local patterns of seed regeneration and trade that occur outside the formal breeding sector (seed exchange networks) can have a strong influence on the genetic diversity and evolution of traditional crop varieties. Despite this, little is known about the extent to which seed exchange networks influence gene flow and genetic structure in traditional crop varieties. Here we study barley (Hordeum vulgare subsp. vulgare) in rural communes of Northern Morocco in 2008 and 2009. We quantified seed regeneration and exchange by farmers within the seed exchange network using structured interviews. Using SSR markers, we also quantified the neutral genetic diversity and structure of a complex of traditional varieties referred to as Beldi that is managed in this exchange network. The majority of farmers (>88 %) report cultivating Beldi. Most seeds of Beldi (70–90 %) are maintained on-farm, while the remainder of seeds are obtained from local markets within the commune. Beldi has high genetic diversity and there is weak but significant genetic structure between communes (FST = 0.031). From SSR marker data there is evidence of a high level of gene flow between communes not reported in interviews. Seeds purchased in local markets likely represent seeds from a larger geographic region, leading to lower genetic structure among communes than expected based on the reported level of on-farm seed regeneration and local sourcing of seed. We discuss the implications of this seed exchange network for the conservation of traditional barley varieties in the study region.  相似文献

‘Microcenters of diversity’ are restricted geographical areas within which a significant diversity of genetic resources is accumulated. Maize genetic variability, one of the largest among cultivated species, particularly in far western Santa Catarina, southern Brazil, has shown a rich store of landraces, conserved on farm by small-scale farmers. Thus, this study aimed to characterize the diversity of Zea mays L. landraces in two municipalities of this micro-region in Santa Catarina, as well as its geographic distribution, developing the census of diversity as a methodological proposal. Diagnosis was made according to the farmers’ knowledge. The field survey was conducted in 70 rural communities involving 2049 farms. The methodology allowed identifying 136 populations of wild relatives, of which some belong to the species Zea luxurians (Durieu and Ascherson) Bird, and 1513 populations of maize landraces, comprising 1078 of popcorn, 337 of common maize, 61 of sweet maize and 37 of flour maize. The identification of 59 morphological groups, the high use, adaptive and agronomic values, and the Shannon index (H′), estimated according to the grain shape (0.79), endosperm type (0.73), size (0.87), color (1.40), and morphological group (3.16), showed at set a considerable diversity distribution conserved on farm in this micro-region. Landrace richness and the presence of wild relative species, associated with the local human activity and sociocultural aspects, allowed to characterize the far western Santa Catarina as a ‘microcenter of diversity’ of Zea mays L.  相似文献

Different conservation measures including domestication approaches are needed to preserve rare and extensively used plant species and to satisfy future market demands. Snow lotus (a common name used for a number of high altitude species in central Asia) species in Mongolia are important medicinal plants, mostly endangered at the regional level due to the extensive use for medicinal purposes and naturally limited distribution ranges. In order to develop effective conservation and cultivation approaches we studied the morphological and genetic diversity as well as the seed germination behavior of three populations of Saussurea involucrata in the Altay Mountains of Western Mongolia. Plant height and leaf length were measured on 41–53 randomly selected individuals in each population. Enhancement of seed germination was studied using either mechanical or chemical scarification with potassium nitrate and gibberellic acid. Genetic diversity within and among three populations of the species was analyzed using Amplified Fragment Length Polymorphisms. Plants in three populations differed in height and leaf length (P < 0.001). Plant size tended to be negatively correlated with habitat altitude. Seed germination rate was low and increased with seed soaking in gibberellic acid. However, the response of seeds to different treatments was rather population-specific. Most treatments did not significantly affect mean days to germination and the ratio of shoot to root dry weight of 2-weeks old seedlings. The level of genetic diversity in the studied populations was lower than the average value for other long-lived perennial herbs. Analysis of molecular variance revealed a high differentiation within populations (92 % of total variation) indicating sufficient gene flow among populations.  相似文献

Chili peppers play a significant role in the world diet and can contribute to the improvement of horticultural producers’ incomes. In Benin, chili pepper production is hampered by many challenges that need to be characterized, prioritized and addressed. Moreover, the existing diversity, as well as the agronomic potential, of the varieties being cultivated in Benin is still not well understood. In order to define the scientific basis for the improvement of chili pepper production and the preservation of its diversity, one hundred villages were randomly selected and surveyed in Northern and Central Benin by using participatory research appraisal tools and techniques. Twelve production constraints were identified, of which low productivity (19 %), soil infertility (18.5 %), drought (18.3 %), pests and disease susceptibility (17.9 %), early fall of plant organs (15 %) appear as the most important. In terms of diversity, 77 chili pepper varieties (40 from the frutescens group, 24 from the annuum group and 13 from the chinense group) including 72 local and five introduced varieties (Tataché, Yèyèkouka, Yèyèkouokourè, Côte d’ivoire and MC) were found. The number of varieties varies from three to seven (four on average) per village and from one to five by household. The proportion of loss of diversity per village varies from 0 to 75 % with an average of 26.61 %. Susceptibility to pests and diseases (37.1 % of responses), early fall of plant organs (11.6 % of responses) and susceptibility to drought (8.5 %), were the most important reasons given in attempts to explain the loss of diversity. Farmers’ varietal preference criteria are essentially agronomic (90.4 % of responses). The participative agronomic evaluation revealed that the varieties of frutescens group are significantly higher with respect to the evaluated variables, but they are less appreciated by the consumers.  相似文献

Climate change and other anthropogenic disturbances can lead to the loss of genetic variation and thereby affect evolutionary potential and survival of plant populations in the wild. We examined these predictions in the primary wild relative of barley, Hordeum vulgare L. subsp. spontaneum (K. Koch) Thell., within its center of diversity, in Jordan. Changes in genotypic and phenotypic diversity were assessed using seed samples collected in 1981 and 2012 from the same 18 sites across Jordan. The overall population structure was conserved, but we observed an increase of within population genetic diversity and a reduction in population differentiation. Phenotypic variation differed among years and sites but the magnitude and direction of change variated among sites. While the sampled region became significantly hotter and drier during this period, simple correlation models did not support association between measures of climate change and the observed genetic and phenotypic changes. Agricultural activities that promote disturbance and demographic fluctuations may affect crop wild relatives that grow in agricultural landscapes, in unexpected ways. The observed increase in genetic diversity within populations might be explained by increased migration or by an advantage of increased genetic variation in the face of variable environmental conditions. This study provides a new perspective on the range of possible responses of crop wild relatives to environmental pressures.  相似文献

Twenty-three populations of Lotus ornithopodioides L., collected from different regions of the Mediterranean basin, were investigated for their ecological and agronomic traits in Western Australia. Great variability was found between and within populations for flowering time, forage and seed yield. Flowering time ranged between 75 and 120 days, dry matter production from 2.8 to 4.3 t ha?1 and seed yield from 284 to 684 kg ha?1. Other important traits such as non-shattering pods and hard seed were taken into account during the selection to assure an easy seed harvesting and legume persistence in the targeted environments. The high level of hard seed recorded in early winter, associated to the low seedling regeneration, indicates that L. ornithopodioides is best suited to ley cropping systems. Elite lines of L. ornithopodioides characterized by early flowering time, high seed yield and non-shattering pods were selected. Two of them, LOR02.1 and LOR03.2, showed dry matter higher than 4.0 t ha?1 and seed yield around 700 kg ha?1 resulting the lines with most potential for Mediterranean farming systems. The results encourage the exploitation of L. ornithopodioides germplasm to develop a new annual self-reseeding legume resource for Mediterranean farming systems for both forage production and crop rotation uses.  相似文献

Fifty grass pea (Lathyrus sativus L.) populations collected from different administrative regions and altitude classes in Ethiopia, were evaluated for variations of five morphological traits. Shannon-Weaver diversity index was calculated for traits, populations and altitude classes. Monomorphism was recorded in flower color (blue) for most of the populations. Olive seed colour was dominantly distributed in all regions with mean frequency of 50% while black seed colour is rare with mean frequency of 4% in Ethiopia. Much variability was observed within populations. High diversity values for pattern of testa colour and leaflet size were recorded in populations of highland origin (>2550 m.a.s.l.). Shannon's diversity index is highest in Gondar (H = 0.65) followed by Tigray region (H = 0.64) even though there was no significant variation in H values between regions. These regions with high diversity are recommended for immediate in situ and ex situ conservation.  相似文献

Silybum marianum (L.) Gaertn. (milk thistle), grown as a medicinal plant in several countries, is considered as a weed in pastures and cereal crops but also as an interesting plant for biomass production. As an additional contribution to the full exploitation of a such promising species, two Sardinian populations of S. marianum were investigated for chemical composition, bioactive compounds and antioxidant properties at vegetative and reproductive stages. Dry matter yield was affected by the phenogical stage and differed between populations, ranging from 148 to 246 g plant?1. Chemical composition did not differ between populations. Antioxidant capacity detected by means of ABTS [(2,2′-azinobis (3-ethylbenzothiazoline-6-sulphonic acid) diammonium salt)] and by DPPH (1,1-diphenyl-2-picrylhydrazyl) methods ranged from 3.45 to 5.42 and 3.83 to 6.32 mmol/100 g dry weight of Trolox equivalent antioxidant capacity, respectively. Differences in antioxidant capacity and bioactive compound contents in the different plant organs were found and also a significant linear correlation between antioxidant capacity and total phenolics and flavonoids, at flowering compared to vegetative stage. Research highlights antioxidant capacity in different organs of milk thistle and encourages the exploitation of biomass also as functional food, source of natural antioxidants and as a complementary fodder.  相似文献

Bambara groundnut (Vigna subterranea (L.) Verdc.) is a neglected and less exploited species in Benin. However, the rational use of the different landraces available involves an appropriate knowledge of their characteristics. This study examined the characteristics of 52 local landraces of Bambara groundnut traditionally cultivated in Benin based on 12 qualitative and 11 quantitative variables, in order to expose those of good qualities. After a classification based on the morphological variables of the seeds, the experimental field was installed in a completely randomized device of blocks repeated three times. The collected data was analyzed with Statistica 7.1 and NTSYS PC 2.2. softwares using qualitative characteristics (Growth’s type, Stem’s hairiness, Flower’s colour, Leave’s shape etc.). As result, the number of morphotypes increased from 18 (out of classification) to 47. Therefore, it appears that the Benin collection of Bambara groundnut is broad in morphological variability. The analysis of the principal component of the quantitative variables revealed three specific classes within the collection. The most efficient variety which belonged to the second class, had a high field yield per hectare (> 1 t/ha). The Souhouloumankpa variety yielded the earliest (103 days after sowing) in addition to the high field yield. The study also showed a positive relationship between 100 seed’s weight, their length, the number of seeds per plant and the seed’s field yield in kg/ha. For a sustainable preservation and an improvement of the available resources, a molecular characterization of the Bambara groundnut landraces in Benin Republic needs to be done.  相似文献

American ginseng (Panax quinquefolius) is a native North American medicinal plant that is becoming increasingly vulnerable despite government harvest restrictions. To better understand the genetic diversity and gene flow of American ginseng, we studied RAPD variation in cultivated and wild populations. Classical and Bayesian analogues of genetic diversity statistics were estimated in seven wild and two cultivated populations. The wild populations were more highly structured (G stβ  = 0.41) than the cultivated populations (G stβ  = 0.24). The genetic diversity within populations ranged from H  = 0.05 to 0.38. Based on genetic pairwise distances, six of the wild populations clustered with the locally-derived cultivated population, while one wild population was more similar to the non-local cultivated population than the local populations. This wild population was highly diverse (P = 1.0; U = 1.0) suggesting that it was supplemented from exotic seed. A set of eight RAPD markers was identified that differentiated plants of local and non-local origin. As a conservation strategy, we recommend that regional gene banks be established based on molecular and geographic diversity to preserve the locally adapted germplasm. These regional gene banks would serve as a conservation tool and also provide a source of genes for genetic improvement of cultivated ginseng.  相似文献

Vitellaria paradoxa C. F. Gaertn., commonly known as shea tree or Vitellaria, is ranked the most important tree species of the savannah regions in the most African countries due to its ecological and economic importance for livelihoods and national economies. However, the savannah regions are the most vulnerable areas to the global climate change. Moreover, the Vitellaria populations on farmlands are threatened by the dominance of old trees with low or lack of regeneration. In this study both morphological and genetic diversity were assessed using several phenotypic traits and 10 microsatellite markers, respectively, to assess the impact of land use and agro-ecozone types on Vitellaria in Ghana. The land use types were forests and farmlands, and the agro-ecozone types included the Transitional, Guinea, and Sudan savannah zones. The mean values of morphological traits, such as diameter at breast height (DBH) and canopy diameter (CD), were statistically different between forest (DBH = 22.20, CD = 5.37) and farmland (DBH = 39.85 CD = 7.49) populations (P < 0.00001). The Sudan savannah zone with mean petiole length of 4.96 cm showed significant difference from the other zones, likely as a result of adaptation to drier climate conditions. Genetic data analysis was based on 10 microsatellite markers and revealed high genetic diversity of Vitellaria in Ghana: mean expected heterozygosity, H e was 0.667, and allelic richness, measured as number of effective alleles A e , was 4.066. Both farmlands and forests were very diverse indicating lack of negative influence of farmer’s selection on genetic diversity. Fixation index was positive for all populations (mean F IS = 0.136) with farmlands recording relatively higher values than forests in all ecological zone types studied, probably indicating less gene flow in the farmlands. Moderate differentiation (F′ ST = 0.113) was comparable to other similar tree species. Both land use and ecological zone types influenced genetic differentiation of Vitellaria at varying levels. The species was spatially structured across three ecozones and following climatic gradient. The forest reserves are used in situ conservation for Vitellaria in Ghana. High diversity observed in the most arid zones provides opportunity to find and use appropriate plant materials for breeding climate change resilient trees.  相似文献

We investigated the genetic variation and relationships among 35 melon landraces collected from the Xinjiang Uygur Autonomous Region in northwestern China by using 19 polymorphic simple sequence repeat markers (SSRs). A total of 55 polymorphic alleles were amplified. The number of alleles per SSR locus ranged from 2 to 5 with an average of 2.89 alleles per locus. The average gene diversity (GD) was 0.42 with a range of 0.06–0.71, and the average observed heterozygosity was 0.22 with a range of 0.06–0.97, indicating that the genetic diversity among the Xinjiang melon landraces was abundant. Genetic variation was also detected between the landrace populations in different regions in Xinjiang. The most abundant genetic diversity was observed among the landraces in Eastern Xinjiang, with the highest GD of 0.45 and PIC of 0.39. Eleven alleles (20 %) were found exclusively in the landraces from Eastern Xinjiang, and two alleles (3.6 %) were unique to the landraces from Southern and Northern Xinjiang. The genetic similarity matrix was defined on the basis of Jaccard coefficient to determine the genetic relationships among Xinjiang landraces. Cluster analysis was performed using the unweighted pair group method with arithmetic means, showing that the ‘wild Hami’ (XJ-34) landrace was distinct from the 34 other landraces that were divided into three clusters. Therefore, the genetic background of XJ-34 differed from that of the other landraces. The landraces were not precisely separated on the basis of their geographic origins, although most of these landraces were likely grouped near one another, as visualized through principal coordinate analysis. Thus, western China is one of the primary or secondary centers of melon diversity because of the relatively higher genetic variation detected among Xinjiang landraces. Except the ‘wild Hami’ landrace, Xinjiang melon landraces could be classified into two botanical varieties, namely, var. inodorus and var. cantalupensis. Saponins occur in numerous plants, including agaves, determining benefic and harmful properties to humans; their presence may favor using plants as soap and other products, but also they may cause caustic effects producing contact dermatitis. In domestication, favorable and unfavorable properties of saponins may cause an increase or decrease of their content, respectively. This study quantified and identified saponins among wild and managed populations of three agave species: A. cupreata Trel. et Berger, A. inaequidens Koch with wild and cultivated populations used for mescal production, and A. hookeri Jacobi, existing exclusively cultivated, used for production of the fermented beverage pulque. We studied 272 plants from 19 populations, quantifying contents of crude saponins through spectrometry. In 12 populations, the saponins types were identified by High Performance Liquid Chromatography–Mass-Spectrography-Time-of-Flight HPLC-MS-TOF. The highest crude saponins content was recorded in A. hookeri (26.09 mg/g), followed by A. cupreata (19.85 and 15.17 mg/g in wild and cultivated populations, respectively). For A. inaequidens, we recorded 14.21, 12.95, and 10.48 mg/g in wild, silvicultural managed and cultivated populations, respectively. We identified 18 saponins types, A. inaequidens showing all of them. A hecogenin glycoside (HG1) is found in high amounts in A. hookeri but in low quantities in A. inaequidens and A. cupreata. A. inaequidens had the greatest diversity of saponins. The contents of crude saponins in A. inaequidens and A. cupreata decrease with management intensity, but contrarily to what we expected, it was the highest in A. hookeri. We hypothesize that such high amount could be due to some saponins, probably HG1, may be precursors of sugars.  相似文献

Perception of farmers’ about the use of pesticides and genetic erosion of tetraploid wheat landraces of Ethiopia was assessed through focus group discussions with farmers, on-farm observations, personal interviews with farmers, by using structured questionnaires of temporal and spatial methods. A total of 1496 farmers from seven provinces in the country were interviewed. Farmers’ knowledge about pesticide increases suggests that they are not happy on using chemicals because of their negative impact on farm land. About 75 % of the farmers believe that, although the use of pesticides may increase the production of wheat, it has a negative impact on (human) health and environment. Women showed a higher concern for pesticides’ harmfulness than men. Farmers’ valuation of genetic erosion was estimated as reduced importance of landraces, as shown by a the lower proportion of landraces either grown or sold on the market. The four most important factors cited for loss of landraces were reduction in land area per capita, displacement by released/modern varieties of hexaploid wheat and teff, reduced benefit from landraces, and displacement by other crops and chat. Genetic erosion of 100 % was observed for Triticum dicoccon in the provinces of Gojam and Gonder and for T. polonicum in Tigray and Gojam. Overall, genetic erosion in the country was 32.0, 35.3, 55.9, 84.4 and 84.4 % for T. durum Desf., T. turgidum L., T. aethiopicum Jakubz., T. polonicum L. and T. dicoccon Schrank, respectively.  相似文献

Significant genetic diversity was observed in 218 out of a total of 1309 accessions of amaranth (Amaranthus hypochondriacus L.) and its seven wild relatives, A. spinosus L., A. dubius Mart. ex Thell., A. hybridus L., A. tricolor L., A. cruentus L., A. caudatus L., A. retroflexus L. for 24 nutritional parameters including total oil content, fatty acid profile, total protein content and amino acid profile. Diversity for total oil content (6.42–12.53%), linoleic acid (25.68–54.34%), oleic acid (21.97–42.01%) of the total fatty acids, total protein content (7.84–18.01%), among important essential amino acids; lysine content (0.66–11.12 g/16 g N), methionine (0.35–4.80 g/16 g N) and half cystine and (0.12–8.32 g/16 g N) was reported. The un-weighted pair-group method using arithmetic average cluster analysis based on pair wise Euclidean genetic distance grouped the accessions into seven major clusters. Histidine, half cystine, tyrosine, essential amino acids, total oil content, linoleic acid and oleic acid content were the major parameters contributing significantly to genetic diversity. Present findings indicate that significant diversity exists for nutritional parameters in amaranth germplasm. The promising accessions with higher multiple nutritive traits; protein content (>16%), oil content (>11%), lysine content (>7.5 g/16 g N) and EAA higher than the FAO reported values, were identified. This is the first report on detailed nutritional analysis of diversity collected worldwide. These could be used as potential breeding material for nutritional enhancement through genetic improvement. This will help in overcoming the “triple burden” of malnourishment, hidden hunger, and obesity.  相似文献

The modern plant breeding is generally considered to be a practice that leads to a narrowing in genetic diversity of crops. The objective of the present study was to assess whether this practice has led to the reduction of genetic diversity in modern Chinese wheat cultivars. A set of 80 dominant Chinese wheat cultivars released from 1942 to 2011 was used to describe the genetic diversity based on 137 simple sequence repeat (SSR) and 52 functional markers. Several important properties about the genetic diversity were revealed. First, relative low genetic diversity level was detected on a genome-wide scale. A total of 752 alleles were detected with a range from 1 to 15, and the mean polymorphic information content value was 0.53 with a range from 0.00 to 0.87. Second, the genetic diversity significantly decreased from 2001 to 2011 at the genome-wide level. More importantly, significant differences of genetic diversity among the three different genomes were observed by the analysis of variance (ANOVA). The three genomes had clearly different changing trends over time: the A genome displayed a decreasing trend (regression coefficient (b) = ?0.01); in contrast, the other two genomes, B and D, showed the increasing trends (b = 0.01 for the B genome, P = 0.05; b = 0.01 for the D genome, P = 0.05). Third, the analysis of qualitative variations in allelic composition over time indicated that, the more recent the cultivars were, the more similar they were to each other. Finally, the frequencies of favorable alleles related to important agronomic traits had been increasing over time or maintained high frequencies in all seven temporal groups. These findings indicate that modern wheat breeding results in not only a qualitative, but also a quantitative change in genetic diversity in the dominant Chinese wheat cultivars. A special attention should be paid to broaden the genetic base in the A genome.  相似文献

Xanthosoma sagittifolium (L.) Schott originated from the American tropics. Domestication may have occurred in various places as this Araceae species is an important food source. It has been cultivated for many decades. In this study, Amplified Fragments Length Polymorphism (AFLP) markers were used to analyze the genetic relationships among 78 Ethiopian X. sagittifolium accessions, for conservation purpose. Cormels were collected from Bench-Maji, Kefa, Dawuro and Wolaita zones, representing four populations. The accessions belonged to either green (G) or purple (P) colored leaf and petiole accessions. Three different AFLP primer combinations resulted in 478 scorable bands, of which 99.2% were polymorphic. The mean Nei’s gene diversity (He) within populations was 0.35 while the G accessions featured higher He (0.38) than the P ones (0.35). The Nei’s gene diversity (He) at entire collection level was 0.38. The detected high genetic diversity may indicate the X. sagittifolium plants growing in the country may derive from diverse parental genotype stock elsewhere and/or there may be multiple introductions to the country. Low levels of genetic differentiation were detected among populations (Gst?=?0.07) and between the G and P accessions (Gst?=?0.02). Insignificant genetic and geographic correlation was revealed by Mantel test. Clustering analysis grouped 91% of the accessions together. Conservation and management of X. sagittifolium in the country should concentrate on maintaining high level genetic diversity within each population as well as at entire collection level through both ex situ and in situ conservation actions.  相似文献

Little is known regarding the effect of fragmentation and human agricultural management on the genetic variation and gene flow of Cucurbita pepo L., 1753 in moderate fragmented areas in central Guatemala. We hypothesize that the genetic variation of C. pepo is affected by forest fragmentation and by traditional agricultural management. Therefore, we aim to determine: (1) the genetic diversity and genetic structure of C. pepo in the Cloud Forest Corridor (CFC) (2) the extent of genetic admixture between commercial variety (CV) and traditional landraces (TL) of C. pepo, (3) the effect of habitat fragmentation in the population genetics of C. pepo with a landscape approach, and (4) the potential relationship between traditional management practices and genetic diversity of C. pepo in the CFC. We detected the existence of high level of genetic diversity (AR = 3.43; He = 0.50), inbreeding (Fis = 0.25) and moderate population structure of C. pepo in the CFC (Fst = 0.16). No correlation between landscape and genetics was found. Also, we found high genetic admixture between CV and TL. It seems that human practices, mainly related with seed exchange patterns, could affect genetic diversity of C. pepo in the CFC. C. pepo populations in the CFC are structured, with inbreeding, and show admixture with the CV, an aspect that could affect its genetic diversity. The agricultural management influenced the population genetics of C. pepo in the CFC, but the landscape did not. We suggest that special efforts should be made to preserve the diversity of this important indigenous food source for Guatemalan people as well as their management practices.  相似文献

Yam (Dioscorea spp.) is widely cultivated in China and many landraces are maintained by local farmers. However, there is little information available about their diversity and species identity. In this study, inter simple sequence repeat (ISSR) and sequence related amplified polymorphism (SRAP) techniques were used to assess genetic diversity within 21 yam landraces from seven cultivated populations. We observed high level of polymorphism among these landraces, specifically, 95.3 % for ISSR and 93.5 % for SRAP. Analysis of molecular variance revealed a significantly greater variation among the four yam species (40.39 %) and their populations (35.78 %) than within the populations (23.83 %). The unweighted pair group method arithmetic averages clusters and principal component analysis for 21 landraces formed four well-separated groups containing landraces of each of the four species, namely, Dioscorea opposita Thunb., Dioscorea alata L., Dioscorea persimilis Prain et Burkill, and Dioscorea fordii Prain et Burkill. The ISSR and SRAP primers were highly discriminatory among the 21 landraces; all 21 landraces could be easily differentiated using these primers. The average mean of gene flow (Nm = 0.1081) estimated from high genetic differentiation (Gst = 0.8222) suggested that gene flow among the populations was relatively restricted. The lack of genetic diversity within individual yam species suggests that it is critical to develop long-term strategies for enhancing genetic diversity within various yam species.  相似文献
