The aim of the present work was to determine the influence of different CaCl(2) dosages on foliar biomass and quality of tobacco plants (Nicotiana tabacum L. var. Tennesse 86). Plants were grown under controlled conditions and submitted to regular fertilization with macro- and micronutrients. The CaCl(2) was applied to the nutrient solution at 1.25 mM (T1), 2.5 mM (T2), and 5 mM (T3). The results indicated that, under the experimental conditions of this work, the application of 1.25 mM of CaCl(2) favored the growth and development of the leaves, this leading to improved biomass production in tobacco leaves. The increase in foliar biomass in treatment T1 could largely be a result of the stimulation of NO(3)(-) assimilation. However, this situation has negative consequences for tobacco quality, given that the T1 treatment augmented the NO(3)(-) concentration and the foliar concentration of nicotine (both effects being harmful for human consumption) and decreased the concentration of reducing sugars in leaves of tobacco plants compared with those of T2 and T3. Finally, a close and directly proportional relationship was found in our experiment between the parameters of foliar-biomass production, total N concentration, chlorophyll concentration, and decline in quality.  相似文献

【目的】生长素响应因子(ARF)在介导生长素信号传递和调控下游生长素响应基因的表达中发挥着重要功能。本文旨在以在富集丰磷特异表达基因的小麦根系cDNA差减文库中鉴定的1个ARF类别的家族成员TaARF6为基础,对该基因cDNA序列、分子特征、不同供磷水平下该基因在根、叶中表达模式及遗传转化TaARF6对丰磷和缺磷条件下植株形态的影响进行较全面研究,阐明该小麦生长素响应因子基因介导不同供磷水平下对植株生长特性的影响。【方法】采用生物信息学工具预测TaARF6编码蛋白特征; 采用溶液培养法培养丰、缺磷处理小麦幼苗,采用半定量RT-PCR技术鉴定TaARF6在丰、缺磷处理下的表达特征。采用DNA重组技术构建将TaARF6编码阅读框融合至表达载体中的表达质粒,利用农杆菌介导的遗传转化法建立超表达TaARF6转基因烟草植株。采用琼脂培养和溶液培养法,培养丰、缺磷不同供磷水平下野生型植株和转基因烟草植株,进而利用常规分析方法鉴定不同磷水平下植株长势、根系和茎叶生物量和植株根叶形态及性状。【结果】1)TaARF6编码生长素响应因子(ARF)型转录因子,编码蛋白中含有ARF家族成员具有的保守结构域。该基因在氨基酸水平上与源于短柄草BdARF6和源于水稻的OsARF6具有高度同源特征。表达分析表明,TaARF6在根、叶中均呈典型低磷下表达下调、复磷后表达再度回升模式,表明该基因表达受到外界供磷水平的调节。2)遗传转化结果表明,在正常生长和低磷逆境下,与野生型植株相比,转基因烟草株系幼苗和植株形态明显增大。3)丰、缺磷不同供磷水平下,与未转化的野生型(WT)对照植株相比,转基因系(Line 3 和Line 5)植株幼苗和植株根系、茎叶和单株鲜、干重均较野生型显著增加。此外,与WT相比,转基因系植株根系数量增多、主侧根长度、根体积、叶面积和根冠比增加。【结论】TaARF6编码典型的生长素响应因子,其编码蛋白具有生长素响应因子特有结构域。TaARF6对环境中的低磷胁迫逆境能产生明显应答。上调表达TaARF6基因,具有增加植株根、叶鲜、干重和改善根叶及植株形态的生物学功能。本研究表明,通过对植株体内生长素响应基因的转录调控,TaARF6在介导植株不同供磷水平下的根叶形态建成和干物质累积过程中发挥着重要作用。  相似文献

养分是烤烟生长与烟叶品质形成的基础,为探究烤烟大田生育期间土壤养分供应、烟株养分吸收以及两者匹配情况及其与烟叶品质问题的关系,本研究测定和分析了湘南目前施肥模式下典型植烟土壤速效养分含量(铵态氮、硝态氮、有效磷和速效钾)、表观肥料养分利用率、烟株养分吸收以及有关烟叶品质指标,并监测了烤烟大田生育期间的气候条件及土壤温湿度变化状况。结果表明:①施氮总量较高(183 kg/hm2),且基施氮量占比高(48.4%),烤烟大田生育前期(0~36 d)土壤速效养分丰富,但烟株生长较慢,导致当季肥料养分利用率不高;②最后一次追肥时间(43 d)偏晚,使得后期烟株脱氮困难,造成顶叶和上二棚叶烟碱含量过高,还原糖含量偏低;③早春低温是烤烟大田生育前期烟株生长缓慢的重要影响因子。针对目前施肥模式与烟株养分需求不匹配问题及其主要影响因素,提出减施基肥、适时适量追肥的优化施肥模式以及应用控缓释肥料、促生微生物、高效多功能叶面肥和施加外源碳等对策。  相似文献

Soybean vegetative storage proteins (S-VSPs) are lysine-rich and, hence, are potentially of high nutritive value for high productive ruminants. Using S-VSPs from wild-type soybean and from transgenic tobacco plants expressing either one of the two S-VSPs subunits (S-VSP alpha or S-VSP beta) or both, we tested their stability in cow rumen fluid under in situ conditions, using SDS-polyacrylamide gel electrophoresis. Proteolysis and degradation pattern of S-VSPs from transgenic tobacco leaves occurred relatively fast compared with that of wild-type (WT) soybean plants. Comparing the two S-VSPs subunits expressed in transgenic plants, we found that S-VSP alpha was degraded much faster than S-VSP beta. The degradation pattern of S-VSPs in transgenic tobacco plants expressing both subunits resembled that of WT soybean. In contrast, the degradation pattern of transgenic tobacco plants expressing a single subunit was different. These finding suggest that the quaternary structure of S-VSPs may be an important factor determining their resistance to rumen degradation. Our results also suggest that the stability to rumen proteolysis of a given protein, when expressed in a transgenic plant, may not always be predictable and has to be verified.  相似文献

机械损伤对烤烟植株氮素吸收及体内烟碱含量的影响   总被引:7,自引:0,他引:7  
采用砂培和营养液培养方法,研究了在营养生长阶段切除顶芽和叶片损伤对烟株生长、体内烟碱浓度、氮浓度及吸氮量的影响。结果表明,切除顶芽导致烟株体内烟碱浓度和含量显著增加。叶片损伤也能增加体内的烟碱浓度,且损伤二次比损伤一次的效果更为明显,说明叶面损伤对烟株体内烟碱合成的影响具有累积效应。但叶面损伤对烟株体内烟碱合成的影响小于打顶处理。切除顶芽和叶面损伤对烟株体内的氮浓度及吸氮量均无显著影响,但显著提高了烟株体内单位氮素产生烟碱的能力。试验结果还表明,机械损伤刺激烟株体内烟碱的合成,烟碱增加与氮素吸收无直接关系。  相似文献

以烤烟(Nicotiana tabacum L.K 326)为试验材料.在大田条件下,利用~(15)N示踪法,研究了土壤氮对烤烟氮素累积和烟碱合成的影响。结果表明。对烤烟氮素累积和烟碱合成的影响。土壤氮比例均随叶位上升而上升,且打顶后上、中、下三个叶位和根茎中土壤氮比例均随生育时期推进而显著增加;打顶后烤烟各器官的土壤氮占总氮比例和占总烟碱氮比例分别为50%和60%。显著高于肥料氮。  相似文献

于黑钙土上进行3个施氮量(0、52.5、67.5 kg·hm~(-2))试验,观察了土壤氮素矿化动态、烟碱积累与分配,以及烟叶的评吸质量.全生育期内,土壤氮素矿化量分别为93.53、65.54、51.66 kg·hm~(-2);烟碱积累总量分别为87.56、128.31、138.91 kg·hm~(-2);成熟采收时下部、中部和上部烟叶烟碱含量分别为1.56%~2.27%、2.01%~2.75%和2.45%~3.13%,烟叶烟碱含量随施氮量的增加而增加.黑钙土在不施氮肥的条件下,中下部烟叶烟碱含量较适中,烟叶品质相对较好,说明黑钙土本身有较强的供氮能力;生产中烤烟氮肥用量控制在52.5~67.5 kg·hm~(-2)为宜,可获得品质较好的烟叶.  相似文献

用盆栽试验研究了不同氮形态和不同氮浓度对烟草钾、氯、总糖和烟碱等品质指标的影响。两种氮形态均有助于叶片钾和氯的积累,铵态氮有助于增加氯的积累,硝态氮更有利于钾的积累。铵态氮有利于总糖提高,其比例超过硝态氮时不利于烟碱含量的提高。任何铵/硝比下,氮浓度增加均会降低糖/烟碱比。下部叶对氮的反应不同于上部和中部叶,也是其烟叶品质不同于上部和中部叶的内在原因。  相似文献

采用水培实验,研究了5个氮(N)浓度下(0.01~5 mmol L-1)水稻的生物量、体内氮浓度、根系发育、体内生长素浓度以及生长素外流蛋白OsPIN家族基因的表达情况。结果表明,与正常供氮水平(2.5mmol L-1)相比,低氮(0.01 mmol L-1)胁迫下水稻根冠比增加28%,地上部全氮浓度降低约20%,根系全氮浓度降低约33%,种子根长度增加25%,种子根上的侧根密度降低26%,倒一叶中的生长素含量增加140%,而根茎结合处和根系的生长素浓度分别下降22%和60%;RT-PCR的结果表明,低氮(0.01 mmol L-1)胁迫下水稻根系中OsPIN1a-b、OsPIN2、OsPIN5a-b和OsPIN9基因表达显著下调;而外源生长素α-萘乙酸(NAA)和生长素极性运输抑制剂1-萘氨甲酰苯甲酸(NPA)的施加均能影响到水稻种子根长和种子根上的侧根密度。由此推论,低氮胁迫下水稻体内生长素从倒一叶到根系极性运输减少是水稻根系对低氮胁迫响应的生理机制之一。  相似文献

采用35S启动子控制Arabidopsis General Regulatory Factor 9 (AtGRF9)在两个转基因番茄株系(E2,E7)中高效表达,以野生型番茄WT、转基因番茄E2和E7三个株系为试验材料,在水培条件下用20%聚乙二醇(PEG6000)模拟干旱胁迫,探究了拟南芥14-3-3蛋白GRF9能否增强番茄根系响应水分胁迫的能力。结果表明:①在干旱胁迫下,野生型番茄和转基因番茄的根系形态指标均受到不同程度的抑制,WT、E2和E7三个番茄材料相对总根长的受抑制程度分别为43%、28%、30%,相对根表面积的受抑制程度分别为46%、33%、35%,相对根体积的受抑制程度分别为47%、32%、29%,相对根直径的受抑制程度分别为29%、21%、22%。②在响应干旱胁迫时,转基因番茄根系蔗糖含量比野生型番茄高20%,根系干物质量比野生型番茄高23%。③在干旱胁迫时,转基因番茄根系质膜H+-ATPase酶活性较高,具有较强的分泌质子的能力,其根系泌酸量比野生型番茄高35%。因此,GRF9能够促进番茄根系蔗糖含量的增加和干物质的累积、增强根系分泌质子的能力,这对于转基因番茄根系在总根长以及根表面积、根体积和根直径的生长以响应干旱胁迫的过程中具有重要作用。  相似文献

采用盆栽试验研究了干旱胁迫下氮用量对烤烟(Nicotiana.Tabacum.L.)养分的积累与分配及烟叶产量和品质的影响。结果表明,在干旱胁迫下随氮用量增加,烟株体内N、P、K、Mn含量显著增加,Ca、Mg、Fe含量明显减少,烟株对各种养分的积累量都显著增大。氮用量还影响烟株体内养分的分配,随施氮量增大,伸根期干旱烟株体内大多数养分在根和茎中的分配比例增加,在叶中的分配比例减少;旺长期和成熟期干旱时则相反。在干旱条件下增加氮用量可以提高烟叶产量和总氮、烟碱含量,降低烟叶还原糖含量。  相似文献

The present study is based on the hypothesis that different methods of supplying boron (B) may have different effects on the yield and quality performance of tobacco crop. A field experiment was conducted to study the effect of different methods of B fertilization on the yield, quality, and leaf composition of flue-cured Virginia tobacco (cv. K-399) in 2007 at the Pakistan Tobacco Research Station, Mansehra. Three different methods (i.e., foliar spray, soil application, and root dipping at the rates of 0.25 kg ha–1, 1 kg ha–1, and 0.5 mg B L–1, respectively) in the form of boric acid along with a control (where no B was applied) were used in a randomized complete block design and replicated four times. Results revealed that different methods of B application significantly affected tobacco yield, quality, and nutrient uptake compared to the control. Maximum leaf area of 707 cm2, green leaf yield of 18553 kg ha–1, cured leaf yield of 2314 kg ha–1, grade index of 79%, nicotine content of 2.54%, and sugar content of 18.35% were noted in the treatment plot where B was applied as a foliar spray; however, in a few cases, there was no significant difference found among different methods of B application. Chloride and potassium contents were not significantly affected by any method of B fertilization. The B fertilization increased the concentration of this element in tobacco leaf, and a maximum concentration of B 48.55 mg kg–1 was noted in the foliar spray treatment. Moreover, fertilizer-use efficiency for different methods of B application revealed that foliar spray is more efficient as compared to soil application and root-dipping methods. Nutrient ratios to B such as potassium (K) / B and chloride (Cl) / B considerably decreased in tobacco leaf with increases in the concentration of B in leaves. These ratios provided some indication of the interrelationship of B with these nutrients in tobacco plants. Results also revealed that K/B and Cl/B ratios closely correlated with the grade index of tobacco leaf, and good grades of tobacco were found to be at a K/B ratio of 682 and a Cl/B ratio of 148 under the experimental conditions. The overall results indicated that the B foliar spray at the rate of 0.25 kg ha–1 significantly increased the yield, quality, and the nutrient uptake by the tobacco crop under the prevailing conditions and was more effective than other methods of B application.  相似文献

烤烟氮素营养诊断及精准施肥模式研究   总被引:6,自引:3,他引:3  
为解决烤烟生产中存在过高施氮或不合理的施肥技术措施致使烟叶产质量下降和氮素利用效率低下等突出问题,该研究于2008-2010年在广东韶关烟草产区以烤烟品种K326为材料,研究不同施氮水平下不同叶位及叶片不同位置上的叶绿素仪测定值(SPAD值)、叶绿素含量、总氮含量的变化特征及其与产质量的相互关系,分析实时氮肥管理(RTNM)模式下不同SPAD施肥阈值对烟草产质量及氮肥利用率的影响,结果表明:顶三叶的叶中位置SPAD值与叶片叶绿素含量、总氮含量的相关性最好,且变异系数小,是较为理想的指示叶或参照叶;获得较好产质量烤烟时的SPAD值在伸根期和旺长期的阈值范围为38.3~47.2。在设置不同SPAD阈值的RTNM模式下,SPAD阈值40.5~43范围内(氮肥用量为75~110kg/hm2)能获得较高的烟叶产量、产值,最高产量和产值分别为2844.09kg/hm2和26989.24元/hm2,烟叶化学成分较为协调。此外,通过与农民习惯施肥模式(167.8kg/hm2)相比较,在SPAD设定阈值为43的RTNM模式下,相应的氮肥用量为110kg/hm2,氮肥农学利用率和吸收利用率分别提高53.0%、46.5%,产值提高了7.4%,调制后烟叶的淀粉、总氮、烟碱含量显著下降,烟叶化学成分更为协调,内在质量较好。  相似文献

烤烟烟碱合成及其氮素来源与移栽期和氮肥的关系研究   总被引:2,自引:0,他引:2  
采用田间试验与15N同位素示踪微区试验,在湖北襄樊植烟生态区老湾村(N 31°27,′E 111°14′,海拔1130 m)研究了3个不同移栽期和施用氮肥对烤烟烟碱含量和烟碱氮素来源的影响。研究结果表明,与5月5日移栽相比,推迟移栽期至5月15日2~5日,烟碱含量、烟碱氮占总氮比例以及烟碱肥料氮比例分别平均增加10%8~5%、5%1~10%和21%5~6%;与不施氮相比,施用氮肥提高烟碱含量12%5~9%,提高烟碱氮占总氮比例5%~127%。烤烟烟碱氮占总氮的比例随生育进程逐渐增加,而各部位烟叶烟碱肥料氮比例随生育进程和叶位上升逐渐下降。土壤氮是烟碱氮的主要来源,对烟叶烟碱含量有决定性影响。结果说明在当地条件下,控施氮肥和适当提前移栽期更有利于降低烟叶(尤其是中、上部叶)中的烟碱含量。  相似文献

采用大田试验研究了不同施氮条件对烤烟打顶前后碳氮代谢、苯丙烷类代谢关键酶活性及其相关物质积累的影响。结果表明: 不施氮下(N0), 打顶前烟叶碳氮代谢关键酶活性较低, 而苯丙氨酸解氨酶活性却较高, 这使得光合固定的碳优先向蔗糖、淀粉、多酚类积累; 单施化肥下(N1), 烟叶碳氮代谢关键酶活性高于N0处理, 但氮代谢强度大, 烟叶固定的碳较早地向烟碱积累, 色素含量高; 加施饼肥下(N2), 烟叶碳氮代谢关键酶活性高, 但代谢协调, 淀粉积累推迟, 色素含量较高, 而多酚类、烟碱积累量均处于N0与N1处理之间。烟株打顶后, 烟叶在短期内各关键酶活性都增强, 但其改变程度不一致, 烟叶内含物重新分配, 游离氨、色素含量递减, 而烟碱含量逐渐积累, N2处理烟碱含量最高。这说明施氮条件影响烟株碳氮代谢的协调程度, 影响烟叶物质积累的流向。因此, 通过控制施氮条件可以改变烟叶的代谢流向以至改变烟叶的质量风格特征。  相似文献

The effects of nitrogen (N‐) and phosphorus (P‐) deficiency, isolatedly or in combination, on growth, nitrogenous fraction, and inorganic phosphate in xylem exudade, and photosynthesis of common bean (Phaseolus vulgaris L. cv. Negrito) were investigated. Plants were grown in nutrient solution adjusted daily to pH 5.5 and aerated continuously. Ten days after emergence mineral deficiency was imposed. Plants were then supplied with high N (7.5 mol m‐3) or low N (0.5 mol m‐3), and also with high P (0.5 mol m‐3) or low P (0.005 mol m‐3). All sampling and measurements were made 28 days after emergence. N‐ or P‐deprivation brought about large decreases in total leaf area by inhibiting the emergence of new leaves and primarily the expansion of the leaves. The specific leaf area did not change under N‐ but decreased under P‐limitation. The decreased shoot to root ratio in all deficiency treatments was a consequence of a lowering mass of above‐ground organs, especially of leaves.

The content of chlorophylls declined significantly only under N‐deficiency alone; carotenoids declined under both N‐ and combined N‐ and P‐limitation. No alteration in amino acid concentration in xylem exudate occurred in plants experiencing N‐starvation, while ureides increased by 79%, and nitrate and inorganic phosphate decreased greatly. Under P‐deprivation, amino acids and nitrate in xylem sap dropped by about half; ureides were held relatively constant, and phosphate was severely depressed. Total upward translocation of N through xylem was estimated to be about 16% higher in N‐deficient plants than in plants without mineral limitation, but leaf N levels in the former were lower as compared to control plants. The net carbon (C) assimilation decreased similarly regardless of the imposed deficiency treatment. Such a decrease was mainly determined by non‐stomatal factors. In general, no additive effect between N‐ and P‐limitation on any of measured parameters was observed.  相似文献

【目的】探究油菜NO3-的吸收、分配和对低氮胁迫的响应及其氮利用效率,为理解油菜在不同低氮胁迫下相关生理变化及其氮素利用效率提供科学依据。【方法】以常规油菜品种814为研究材料,采用砂培试验,在正常供氮水平(10 mmol/L)和低氮胁迫水平(3 mmol/L、1 mmol/L)下,研究油菜的根系特性、蒸腾作用对低氮胁迫的响应及其氮素吸收效率,并研究油菜NO3-的运输分配与同化对低氮胁迫的响应及其氮素利用效率。【结果】与正常供氮处理(10 mmol/L)相比,低氮胁迫处理(3 mmol/L、1 mmol/L)的油菜NO3-含量、全氮含量均显著下降,但(NO3-)叶/根、(全氮(%))叶/根显著升高,植株根系干物质重、根系吸收面积均显著下降,但根冠比显著升高。油菜植株在低氮胁迫下气孔导度和蒸腾速率显著增加,一方面促进植株对NO3-的捕获,另一方面也促使更大比例的NO3-分配在植物的地上部分,但植株的水分散失加剧,水分利用效率显著下降。低氮胁迫处理油菜根和叶中NR、GS活性与正常供氮处理之间的差异不显著或有增加,其叶绿素含量、光合速率均显著下降,但光合氮素利用率显著升高。【结论】在低氮胁迫条件下,油菜植株的氮素和干物质累积均显著下降,但NO3-在植株的地上部分分配比例的增加以及光合氮素利用率的升高促使植株的氮素利用效率显著提高。  相似文献

为了探索韶关烟区适宜的播种期和移栽期,以6个优质烤烟品种(品系)为材料,进行分期播种,塑造不同的光温气候条件,以优质中部烟叶化学成分为参考样,用指标赋值法对烤烟(等级为C3F)化学品质进行综合评分,依品质优劣将不同品种排序。结果表明:播期显著影响烤烟品种的产量和品质的形成。产量高低决定产值高低,适当推迟播期有利于提高烤烟产量和产值;但播种过迟会严重降低产量,降低烤后烟叶中上等烟比例,导致产值下降。烟叶化学品质中烟碱含量、总氮含量、淀粉含量、钾含量均受播期影响,随着播期延迟C3F烟叶的烟碱含量、总氮含量、钾含量减小,淀粉含量明显增加,影响烤烟品质;但3个播期中各品种的氮碱比、糖碱比、钾氯比变化因品种而异。在3个播期试验品种中,各品种的C3F品质平均综合评分表现为"云烟100"、"NX208"、"K326"的评分相对较高,C3F烟叶品质较好。适当的播种期使得烟叶化学成分的协调性好,利于提高烤烟的产量和品质。不同播期试验表明,韶关烟区适宜播种期为12月1日—12月10日,适宜移栽期为次年2月25日—3月5日。  相似文献

通过在云南省玉溪市赵桅村6组烟田(A)和赵桅实验基地(B)进行的大田15N示踪试验,研究了在不施氮和施氮90 kg/hm2条件下,水稻土烤烟干物质累积特征、肥料氮和土壤氮的吸收和分配及肥料氮和土壤氮对烟叶品质的影响。结果表明,施氮能显著提高烟株干物重,增加烟株吸氮量,但在生育后期施氮与不施氮处理,烟株均存在干物质和氮素累积过多问题。施氮90 kg/hm2情况下,烟株整个生育期中吸收的氮素主要来自于土壤氮,而且烟株吸收的土壤氮及其占总吸氮量的比例随生育期延长和烟叶着生部位的升高显著增加;到采收结束时,两试验田烟株吸收的土壤氮占总吸氮量的比例分别达69%和74%。不同施氮水平烟叶品质差异较大,不施氮处理各部位烟叶总氮和烟碱含量均偏低,糖碱比不协调;施氮处理各部位烟叶总氮含量均在适宜范围内,但由于烟株生育后期土壤供氮较多,上部烟叶存在烟碱含量偏高的问题。选择有机质含量适宜的土壤,控制烟株生育后期土壤供氮量,对降低上部烟叶总氮和烟碱含量,提高我国烟叶整体质量至关重要。  相似文献
