Abstract

Yacon (Polymnia sonchifolia) plants originating from Andean highlands are known to accumulate a large amount of oligofructans in their tubers and tuberous roots (Ohyama et al. 1990; Asami et al. 1992). Asami et al. (1992) have reported that tubers and tuberous roots which were harvested in late fall contained about 57 and 66% respectively (on a dry matter basis) of oligofructans. However, the tubers, tuberous roots and shoots of the yacon contained only a small amount of starch and inulin. The tuberous roots of yacon which resemble those of sweet potato in appearance are usually eaten raw and are expected to become commercially valuable as a source of oligofructans.  相似文献

Protein is the second most abundant constituent in the rice grain next to starch. Association analysis for protein concentration in brown rice was performed using a “mini‐core” collection, which represents the germplasm diversity found in the USDA rice world collection. Protein concentration was determined in replicated trials conducted in two southern U.S. locations, and association mapping was performed by using 157 genomewide DNA markers. Protein concentration ranged from 5.4 to 11.9% among the 202 accessions. Protein variation owing to accession and accession × location interaction were highly significant. Ample variation was seen within each subpopulation by ancestry, as well as within the 14 geographic regions where the accessions originated. Accessions from Eastern Europe had the highest level of protein. Ten markers on eight chromosomes were significantly associated with protein concentration. Five of these markers occurred near known protein precursor genes or quantitative trait loci, and the other five markers were novel for the association with protein concentration in rice. The germplasm and genetic markers identified in this study will assist breeders in developing cultivars tailored for applications requiring specific protein concentration in the rice grain. The research results contribute to the potential discovery of novel rice storage protein pathways in the endosperm.  相似文献

To assess the genetic variation within and among accessions of Russian wildrye (Psathyrostachys juncea), 88 individuals of 11 accessions originated from geographically diverse locations were analyzed using randomly amplified polymorphic DNA (RAPD). Under the optimized condition, 56% of the 200 tested primers produced polymorphic RAPDs among accessions. High level RAPD marker variations existed both within and among accessions. About 56% and 44% of the amplified fragments were polymorphic within and among accessions, respectively. Accessions from neighboring geographical regions are more similar than those from distant regions. These results are comparable with those obtained from studies on chromosome C-banding and isozyme electrophoresis.  相似文献

The collection, identification and maintenance of genebank accessions of the genus Trifolium is a major task because of the large number of genera and their occasional morphological similarity. We investigated whether the measurement of nuclear DNA content can serve as an additional criterion for identification of mislabeled accessions. Relative nuclear DNA content was determined by flow cytometry measurements for a total of 151 genebank accessions of 23 Trifolium species with notable agronomical value. Among 23 species analyzed, 15 were found to possess a uniform relative nuclear DNA content, with intraspecific variability of the majority of analyzed species lower than 5%. Within six Trifolium species, 1–2 atypical accessions with outstanding differences in relative DNA content, chromosome number and morphological features were found. In T. hybridum and partially in T. ambiguum these outstanding differences could be ascribed to variations in accession ploidy level. For the remaining atypical accessions, the determined nuclear DNA content, chromosome number and morphological features were not related to those characteristic of the species. Additionally taxonomic identity of atypical accessions was determined using ITS region sequencing and morphological observations. We propose flow cytometric measurements of nuclear DNA content as simple and reliable technique which can be used at seedling stage to verify identity and genetic stability of Trifolium genebank accessions.  相似文献

An experiment was conducted in 2001 at Holetta and Kulumsa, Ethiopia, to study the extent and pattern of genetic diversity in Ethiopian field pea (Pisum sativum L.) landraces. One hundred forty-eight germplasm accessions were grown in an alpha lattice design with 2 replications. Data on 12 traits were collected and analyzed. Differences among the accessions were significant for most of the traits (except number of seeds/ pod) at each location even though differences pooled over location were mostly non-significant. The accessions were grouped into five clusters of different sizes. Accessions from the southern part of the country (Arsi) distributed overall clusters while those from the northern half (North and South Wello, North Gonder and North Shewa) fell into clusters C1 to C3. Cumulative effects of a number of characters dictated differentiation of the accessions into clusters. There was no definite relationship between geographic diversity and genetic diversity as overlapping was encountered in clustering pattern among accessions from different parts of the country. Accessions from different regions might have similar genetic background and those from the same origin might also have different genetic background. Therefore, geographic diversity should not necessarily be used as an index of genetic diversity and parental selection should be based on a systematic study of genetic diversity in a specific population. Genetic distances among most of the clusters were significant that crosses between parents selected out of them are expected to generate desirable genetic recombination. Selection should also consider the special advantages of each cluster and each accession within a cluster. Future germplasm collection, conservation and breeding efforts should focus not only on inter-regional diversity but also on intra-regional diversity.  相似文献

Seedlings of Pistacia vera L. developed from seeds of two separate populations in Turkmenistan, Kepele and Agachli, were evaluated for their growth potential and genetic polymorphism. Plant growth rate as well as random amplified polymorphic DNA (RAPD) analysis showed distinct differences between the two populations. In plant height growth rate, 17 Agachli accessions were 1.3 times higher on average than that of 10 accessions of Kepele (significant at p = 0.046) and 1.2 times higher for trunk diameter growth rate (p = 0.062). Cluster analysis divided most accessions into two main genetic groups according to their geographic origin. The Agachli group was further divided into two subgroups. One Kepele accession (K4), was genetically different from the rest and clustered on a separate outgroup. Two Agachli accessions (A12 and A17) were outside the two main populations clusters. Accessions K9 and K10 from Kepele were exceptional and were clustered in each of the two Agachli's subgroups, indicating a close genetic relationship between the two populations. In addition, high similarity values (0.58–1.00) and small genetic distances reflect plausible gene flow between Kepele and Agachli, which are 100 Km apart. Mantel test revealed significant relationship between the RAPD and the morphological traits matrices, pointing to the genetic basis for the measured differences in the growth rate. Growing the accessions on the same plot, under similar conditions enabled the evaluation of genotypic differences. The combination of morphological traits and molecular markers will further assist in preservation of genetic variability and cultivation of useful genotypes of P. vera L.  相似文献

Summary As part of a project to assess the U.S. Asiatic Cotton Germplasm Collection as a source of genes for pest resistance, forty-three accessions of Gossypium arboreum L. were evaluated for resistance to thrips. Thrips, Thysanoptera spp., are a minor, but widespread pest on cotton, G. hirsutum L. and G. barbadense L. Accessions were planted in free-choice field tests in 1988, 1989, and 1990 at the Cotton Branch Experiment Station, Marianna, Arkansas. Damage ratings, based on a scale of 0 (no damage) to 7 (severe damage), were made approximately one month after planting. Relative score was computed by dividing the rating of each accession by the rating of the reference, G. hirsutum Stoneville 506. The relative score averaged over all G. arboreum accessions by year varied with the year, but was less than 100 (relative score for Stoneville 506) in each case. When data were combined for all three years, twenty-five accessions were significantly better than Stoneville 506. A collection of 10 accessions of fenugreek (Trigonella foenum-graecum L.), an annual legume, was grown during two summers at three plot locations in western Canada to assess whether genetic (accession) and environmental factors (site and year of production) influenced levels of diosgenin, a steroidal sapogenin. The 60 harvested seed samples, each analyzed by single determinations on three subsamples of defatted and dried seed material, were hydrolyzed by a microscale procedure in water containing 2-propanol (70%) and sulfuric acid (1 M). The extracts were analyzed by capillary gas chromatography with 6-methyldiosgenin as internal standard. Diosgenin levels from mature seeds ranged from 0.28 to 0.92% (28-92 microg/10 mg). Analysis of variance on combined diosgenin levels from the three sites and two years revealed that accession, accession x year, and site x year effects were significant for diosgenin content, whereas site, year, and site x accession effects were not. Four accessions, CN 19062, CN 19067, CN 19070, and CN 19071, were identified with high levels of diosgenin on the basis of the 2-year data set. In these accessions, mean levels of diosgenin plus yamogenin from seven site years were estimated at 0.70, 0.98, 0.84, and 0.87%, respectively.  相似文献

This investigation was aimed at exploring the genetic diversity among nine typical accessions of Chinese mustard crops using random amplified polymorphic DNA (RAPD) markers and morphological comparison. Totally, 111 reproducible DNA bands were generated by 16 arbitrary primers, of which 91 bands were proved to be polymorphic. Based on pair-wise comparisons of the amplified bands, genetic similarities were obtained using Nei & Li's similarity coefficients and a dendrogram reflecting their relationships was made using the unweighted pair–group method with arithmetic averages (UPGMA). The result of cluster analysis indicated that the nine accessions were capable of being classified into two primary groups, one including accession 2 with expanded root (root mustard), accession 3 with entirely expanded whole stem (long-stem mustard), accession 6 with edible leaves (leaf mustard), accession 8 with edible seed stalk (seed stalk mustard) and another one including accession 4 with expanded basal stem (short-stem mustard), accession 5 with bulgy petiole (leafy bulgy mustard), and accession 9 with mustard-rich seed (seed mustard). Besides, accession 1 with expanded root (root mustard) and accession 7 with edible leaves and seed stalk (seed stalk mustard) were independent of other accessions in the dendrogram. Additionally, by cluster analysis based on highly reproducible RAPD markers, the accessions with similar edible parts of leaves or roots were not actually in the same phylogenetic groups. This implied that they were probably derived from different geographical origins with dissimilar genetic background and possessed higher genetic diversification. Furthermore, the results indicated that the traditional method for classifying Chinese mustard crops was not much reliable as it was largely dependent on phenotypic behaviors. Meanwhile, the phenotypic differences among individuals did not necessarily mean they must have sharp difference in genetic background as they met in the same group. Undoubtedly, these results aforementioned make this crop quite interesting to researchers for further investigating the molecular evolution of this special AABB group.  相似文献

A set of 99 common bean germplasm collected from central Himalaya was investigated for their genetic variability using random amplified polymorphic DNA (RAPD) markers. Ten oligonucleotide primers, selected from 60 initially screened, generated 123 amplicon products. Of these, two amplicons were shared by all the accessions whereas 112 were polymorphic at least in two pair wise comparison. Nine unique bands identified were as low as 0.32 kb M.W. to as high as 3.5 kb and were confined to eight collections. All primers produced polymorphic amplicons though the extent of polymorphism varied with each primer. The primer OPF-17 was found to be most powerful and efficient as it generated a total of 17 bands of which 15 were polymorphic. RAPD markers data were analysed statistically using NTSYSpc.2.02e software and a dendrogram was generated using Jaccard’s similarity coefficient. The similarity coefficient values varied from 0.19 to 0.91. Grouping analysis revealed the categorization of 99 germplasm into 12 major branches with different level of similarity. Three branches namely branch 2, 3 and 5 out of 12 had only one accession. Branch 1 which consisted of three accessions was the most divergent as revealed by Jaccard’s similarity coefficient. Branching pattern of the accessions did not show any correlation with morphological data or altitudinal alignment of the accessions.  相似文献

Guar [Cyamopsis tetragonoloba (L.) Taub.] is an important commercial crop of India, mainly cultivated in the northwestern part and neighboring areas of Pakistan. High morphological diversity is reported in guar. The present study attempts to analyze the genetic diversity in cultivated guar using allozyme polymorphism and compare it with reported morphological diversity. Accessions for the study were selected from a set of guar germplasm conserved at the National Bureau of Plant Genetic Resources, New Delhi. The morphological data of 108 selected accessions was subjected to UPGMA clustering. Fifty-five accessions were then picked randomly from the clusters generated. Tris-borate system (pH 8.3) was found to be most effective for detection of allozyme polymorphism in guar. Ten enzymes were selected for the study out of 18 systems tried, namely, malate dehydrogenase, malic enzyme, phosphogluco isomerase, glutamate oxaloacetate transaminase, esterase, peroxidase, alcohol dehydrogenase, glutamate dehydrogenase, glucose 6-phosphate dehydrogenase and shikimate dehydrogenase. A total of 20 loci were detected with 34 alleles. The number of polymorphic loci in different accessions was observed to be highly variable (0–9). An average of 1.27 allozyme per polymorphic locus was observed. The allele frequency data of the 55 accessions was used to determine genetic diversity parameters. The dendrogram generated on genetic distance showed three clusters in the distance range of 0–0.28. Most of the accessions were in the first clusters in a narrow distance range of 0–0.08. The second cluster consisted of improved varieties and accession from Pakistan. The third cluster of accessions was genetically more distant but there was no definite segregation of accessions according to the place of collection. The accessions from south west Rajasthan were scattered in all the three clusters showing higher variability in this region, also supported by higher morphological variation. The genetic distance was low among accessions of guar, indicating low diversity in India.  相似文献

ABSTRACT

Indian mustard (Brassica juncea Czern.) has the potential to extract zinc (Zn) and other metals from contaminated soils, but the potential to accumulate metals at different levels of exposure is not well documented. The objectives of this research were to assess plant growth and Zn accumulation for different metal-accumulating accessions of Indian mustard grown with various Zn concentrations. In the experiment, three accessions of Indian mustard (426308, 182921, and 211000) were supplied with 12 levels of Zn (ranging from 0.0 to 7.0 mg L?1) for three weeks in solution culture. Accession 426308 had a greater capacity for dry-mass accumulation than the others, but differences among accessions lessened as the concentration of Zn in solution increased. Accessions did not differ in Zn concentrations in shoots, but accession 426308 had a greater potential to accumulate Zn than the other accessions. Elevating the Zn supply in solutions had a limited effect on increasing the total Zn accumulation of shoots. Plants suffered Zn-induced iron (Fe) deficiency if the Zn concentration in solution exceeded 2.0 mg Zn L?1. The level of Zn tolerance of Indian mustard accessions was: 211000 > 182921 > 426308. Maximum Zn accumulation in shoots was approximately 5.0 mg Zn per plant. The phytoextraction potential of Indian mustard may be limited under Zn-contaminated conditions by nutrient disorders and toxic effects of Zn that suppress growth.  相似文献

Germplasm evaluation of ex situ collections is needed to document collection characteristics, enhance utilization, and to determine collection needs. The objectives of this study were to (1) provide oil and meal evaluation information for a major portion of the United States Department of Agriculture (USDA) safflower (Carthamus tinctorius L.) collection, (2) compare ranges, variances and means between 203 core and 797 non-core accessions, and (3) determine if region of origin could be differentiated based on accession oil and meal characteristics. Means of the core and non-core accessions differed for % oil, palmitic acid, stearic acid, -tocopherols, and phenolic glucosides (both bitter and cathartic) (P<0.05). Differences between linoleic acid, oleic acid and -tocopherol means were not significant, and the variance between core and non-core accessions differed only for palmitic acid. Thus the core was not fully representative of the non-core accessions, but did capture a large fraction of the diversity in oil and meal factors present. Accessions from the Americas, China, South-West Asia, and South-Central Asia were differentiated using canonical discriminate analysis, but these regions overlapped to varying degrees with the E. European, Mediterranean, and E. African regions. Variation in % oil and fatty acids were generally more important than tocopherols and phenolic glucosides in differentiating accessions on a regional basis. The check cultivars Montola 2001 (high oleic) and Morlin (high linoleic) had oil and fatty acid content comparable to the maximums found in the collection. The ranges for tocopherols and phenolic glucosides among collection accessions, however, exceeded those of the cultivars, suggesting that collection accessions could be useful for genetic manipulation of these factors.  相似文献

A polymerase chain reaction (PCR) based approach involving the directed amplification of minisatellite DNA region (DAMD-PCR) was used to identify accession specific DNA markers and study genetic relationships between and within 15 accessions corresponding to 11 species in genus Capsicum. A touch down PCR profile and unique chemical concentration of ingredients resulted in reproducible and reliable DNA amplifications. The number of amplified products varied from 1 to 12 fragments depending on the template DNA and the primers. The DAMD-PCR technique provided a total of 38 accession specific DNA markers (diagnostic DAMD-PCR) which can be utilized in accession identification, preservation and genetic studies of Capsicum germplasm. Based on 1,292 polymorphic and monomorphic DNA markers directed with 22 minisatellite specific primers, accessions were divided into four major groups, three of which corresponded to the three distinct Capsicum complexes. Genetic diversity among 26 cucumber (Cucumis sativus L. var. sativus) accessions from five African countries [Algeria (1), Egypt (21), Ethiopia (2), Kenya (1), and Libya (1)] present in the U.S. National Plant Germplasm System (NPGS) were examined by assessing variation at 71 polymorphic random amplified polymorphic DNA (RAPD) loci. Genetic distances (GD; simple matching coefficient) were estimated among these African accessions and a reference array (RA) of 21 accessions representative of the genetic variation in cucumber. GD among African accessions ranged between 0.41 and 0.97. GD among accessions in the reference array ranged between 0.36 and 0.88. Multivariate analysis identified three distinct groupings (1–3) of African accessions; Group 1 contained 21 accessions (Egypt, Ethiopia and Libya), Group 2 consisted of two accessions (Kenya, Algeria), and Group 3 possessed three accessions (Egypt). These groupings were distinct from each other (P > 0.001). Accessions in Group 1 differed genetically from all other accessions examined (P > 0.01), and accessions in Groups 2 and 3 were uniquely associated with several RA accessions. While GD among accessions in Group 1 ranged between 0.52 and 0.90, distances among Group 2 accessions varied between 0.93 and 0.97. The GD between the two accessions in Group 3 was 0.65. An accession from Syria (PI 181874) and from one Turkey (PI 199383) were genetically more similar to accessions in Group 1 than to other accessions in the RA. Likewise, accessions in Group 2 were genetically similar to two RA accessions from China and a European glasshouse cucumber line, and Group 3 accessions showed genetic affinities with the U.S. market class cultivar Dasher II. Data suggest that some Egyptian accessions (Group 1) possess unique genetic variation, that this germplasm has potential for broadening the genetic base of commerical cucumber, and that further collection of African germplasm is likely to enhance genetic diversity of cucumber in NPGS.  相似文献

Kentucky bluegrass (Poa pratensis L.) is an important turf and forage grass species with a facultative apomictic breeding behavior. In this study, mature seed and leaf tissue from 38 accessions of a USDA core collection of Kentucky bluegrass were analyzed with flow cytometry to characterize the reproductive mode and DNA content for each accession. Major reproductive pathways for each accession were determined based upon the presence and the position of the peaks observed and the known methods of reproduction for Kentucky bluegrass. While the majority of the accessions exhibited facultative apomictic reproductive behavior with a combination of reduced, zygotic and unreduced, parthenogenic embryo production, obligate sexual or obligate apomictic accessions were also found to be present in this core collection. In addition, reduced, parthenogenic and unreduced zygotic embryos were also detected in several accessions. Flow cytometric analysis of somatic tissue revealed a large range of DNA variation within this core collection. We also examined the sensitivity of flow cytometry in analyzing bulked samples containing a large number of plants with varied DNA content and determined that flow cytometry can effectively detect a plant having a different DNA content within a 15-plant bulk sample. Overall the combination of mature seed and somatic tissue analysis generated important information for the Kentucky bluegrass core collection and can be an effective and affordable tool to characterize even greater numbers of Kentucky bluegrass accessions.  相似文献

The sesquiterpene lactones (STLs) content of glandular trichomes from the leaves of twenty-five yacon (Smallanthus sonchifolius, Asteraceae) accessions, obtained along a latitudinal gradient from Ecuador to northwest Argentina, was characterized by gas chromatography/mass spectroscopy (GC/MS). While accessions from Ecuador, Bolivia and Argentina proved to be very chemoconsistent, significant variation was found in quantitative composition of STLs from accessions in central Peru, the probable region of origin for the species.  相似文献

A field experiment was conducted to assess yield, oil content, and composition of 38 genotypes of sweet basil ( Ocimum basilicum L.). Overall, biomass yields were high and comparable to those reported in the literature. However, basil genotypes differed significantly with respect to oil content and composition. Oil content of the tested accessions varied from 0.07% to 1.92% in dry herbage. On the basis of the oil composition, basil accessions were divided into seven groups: (1) high-linalool chemotype [19-73% (-)-linalool], (2) linalool-eugenol chemotype [six chemotypes with 28-66% (-)-linalool and 5-29% eugenol], (3) methyl chavicol chemotype [six accessions with 20-72% methyl chavicol and no (-)-linalool], (4) methyl chavicol-linalool chemotype [six accessions with 8-29% methyl chavicol and 8-53% (-)-linalool], (5) methyl eugenol-linalool chemotype [two accessions with 37% and 91% methyl eugenol and 60% and 15% (-)-linalool], (6) methyl cinnamate-linalool chemotype [one accession with 9.7% methyl cinnamate and 31% (-)-linalool], and (7) bergamotene chemotype [one accession with bergamotene as major constituent, 5% eucalyptol, and <1% (-)-linalool]. Our results demonstrated that basil could be a viable essential oil crop in Mississippi. The availability of various chemotypes offers the opportunity for production of basil to meet the market requirements of specific basil oils or individual compounds such as (-)-linalool, eugenol, methyl chavicol, methyl cinnamate, or methyl eugenol.  相似文献
