The effects of soil disturbance and residue retention on the functionality of the symbiosis between medic (Medicago truncatula L.) and arbuscular mycorrhizal fungi (AMF) were assessed in a two-stage experiment simulating a crop rotation of wheat (Triticum aestivum L.) followed by medic. Plants were inoculated or not with the AMF, Glomus intraradices and Gigaspora margarita, separately or together. The contribution of the arbuscular mycorrhizal (AM) pathway for P uptake was determined using 32P-labeled soil in a small hyphal compartment accessible only to hyphae of AMF. In general AM colonization was not affected by soil disturbance or residue application and disturbance did not affect hyphal length densities (HLDs) in soil. At 4 weeks disturbance had a negative effect on growth and phosphorus (P) uptake of plants inoculated with G. margarita, but not G. intraradices. By 7 weeks disturbance reduced growth of plants inoculated with G. margarita or AMF mix and total P uptake in all inoculated plants. With the exception of plants inoculated with G. margarita in disturbed soil at 4 weeks, the AM pathway made a significant contribution to P uptake in all AM plants at both harvests. Inoculation with both AMF together eliminated the negative effects of disturbance on AM P uptake and growth, showing that a fungus insensitive to disturbance can compensate for loss of contribution of a sensitive one. Application of residue increased growth and total P uptake of plants but decreased 32P in plants inoculated with the AMF mix in disturbed soil, compared with plants receiving no residue. The AMF responded differently to disturbance and G. intraradices, which was insensitive to disturbance, compensated for lack of contribution by the sensitive G. margarita when they were inoculated together. Colonization of roots and HLDs in soil were not good predictors of the outcomes of AM symbioses on plant growth, P uptake or P delivery via the AM pathway.  相似文献

The capacities of two arbuscular mycorrhizal (AM) fungi, Glomus mosseae and Glomus versderme, tomineralize added organic P were studied in a sterilized calcareous soil. Mycorrhizal (inoculated with either of the AM fungi) and non-mycorrhizal red clover (Trghlium pmtense L.) plants were grown for eight weeksin pots with upper root, central hyphal and lower soil compartments. The hyphal and soil compartmentsreceived either organic P (as Na-phytate) or inorganic P (as KH2PO4) at tbe rate of 50 mg P kg-1. No P wasadded to the root compartments. Control pots received no added P. Yields were higher in mycorrhizal than innon-mycorrhizal clover. Mycorrhizal inoculation doubled shoot P concentration and more than doubled total P uptake of plaflts in P-amended soil, irrespective of the form of applied P. The mycorrhizal contributionto inorganic P uptake was 80% or 76% in plants inoculated with G. mosseae or G. verefforme, respectively. Corresponding values were 74% and 82% when Na-phytate was applied. In the root compartments of the mycorrhizal treatments, the proportion of root length infected, hyphal length density and phosphatase activity were all higher when organic P was applied than when inorganic P was added.  相似文献

以红三叶草为材料 ,利用三室隔网培养方法 ,施用不同有机磷源 :植酸钠 (Na -Phytate)、核糖核酸 (RNA)和卵磷脂 (Lecithin) ,研究接种菌根真菌Glmous versiforme对土壤及外加有机磷源的利用效率 ,另设无机磷及不施磷作为对照。结果表明 ,接种菌根真菌能明显增加植株干物重、含磷量和吸磷总量。与各有机磷处理相比 ,无机磷处理前期的生长效应较好 ,施用有机磷各处理在不同生长时期均明显促进了植株生长 ,但不同有机磷源之间没有显著差异。在植株吸磷量上 ,植株生长 7周以前 ,磷酸二氢钾处理高于其它处理 ,而植株生长 10周时 ,植酸钠处理高于磷酸二氢钾处理。接种菌根处理由于丛枝菌根活化了土壤有机磷 ,到植株生长 10周时其吸收有机磷的量已占吸磷总量的 76 .7%。  相似文献

The external hypha of arbuscular mycorrhizal (AM) fungi, extending from roots out into soil, is an important structure in the uptake of phosphate from the depletion zone around each root. In this paper, we analysed some phospholipid fatty acids (PLFAs) derived from external hyphae of four AM fungi (Glomus etunicatum, Glomus clarum, Gigaspora margarita and Gigaspora rosea) to find fatty acids which may be useful as specific markers for identifying and quantify the external hyphae of Gigaspora species. Leek (Allium porrum L.) seedlings inoculated with each AM fungus were grown in river sand. Sand samples were collected and four PLFAs (16:1ω5, 18:1ω9, 20:1ω9 and 20:4) in the sand were analysed. In addition, the hyphal biomass in the sand was determined by the direct microscopic method. PLFAs 18:1ω9 and 20:4 were found in all the AM-inoculated and non-inoculated sand samples. PLFA 16:1ω5 was detected in the sand inoculated with G. etunicatum, G. clarum and Gi. rosea. PLFA 20:1ω9 was detected only in the sand inoculated with Gi. rosea. PLFAs 16:1ω5 and 20:1ω9 were not found in the sand inoculated with Gi. margarita. The amount of PLFA 20:1ω9 was closely correlated with the amount of biomass of external hyphae of Gi. rosea (r=0.937, P<0.001), whereas no correlation was observed for PLFA 16:1ω5. The 20:1ω9 content of Gi. rosea was approximately 6.56 nmol mg−1 hyphal biomass. We suggest that PLFA 20:1ω9 can be used as a specific marker for identifying and quantifying the external hyphae of Gi. rosea, at least in controlled experimental systems.  相似文献

Exudates of Rhodotorula mucilaginosa, a yeast commonly found in the rhizosphere, increased hyphal length of the arbuscular mycorrhizal (AM) fungi Gigaspora rosea and Gigaspora margarita. Rhodotorulic acid (RA), a siderophore compound obtained from R. mucilaginosa exudates, increased hyphal length and branching. Thus, the increase in the number of entry points and the higher AM root colonization of tomato plants in the presence of RA can at least partially be explained by the positive effect of RA on the pre-symbiotic stages of the AM fungi.  相似文献

A glasshouse pot experiment investigated the uptake by arbuscular mycorrhizal (AM) fungi associated with red clover of three organic sources of P added to a sterilized calcareous soil of low P availability. Each pot was separated into a central compartment for plant growth and two outer compartments for external mycelium using 30-μm nylon mesh to restrict the roots but allow hyphal penetration. Plants in the central compartments were inoculated with the AM fungus Glomus versiforme and uninoculated controls were included. Plants were harvested on three occasions: 5, 7 and 10 weeks after sowing. Application of each of the three organic P sources (lecithin, RNA and sodium phytate) or inorganic P (KH2PO4) at 50 mg P kg−1 to the outer compartments of mycorrhizal and uninoculated pots increased the yield, P concentration and total P uptake of red clover compared with pots to which no P was applied, with no differences among P sources in non-mycorrhizal plants but differences observed in mycorrhizal plants both 7 and 10 weeks after sowing suggesting differences in availability of the four P sources to AM mycelium. The contribution of external mycelium to plant uptake of applied P increased with time. The three organic P sources made smaller contributions to plant P nutrition than KH2PO4 at the first and second harvests. At the third harvest, the contribution from KH2PO4 was 23%, while those from lecithin, RNA and sodium phytate were 23, 17 and 31%, respectively. This suggests that with the mediation of AM fungi, soil organic P sources can make a contribution to host plant P nutrition comparable to that of soluble orthophosphate.  相似文献

A greenhouse experiment was conducted to examine the favorable effects of sod culture system with bahiagrass (Paspalum notatum Flügge.) and Vulpia myuros (L.) C. C. Gmel. intercropped with citrus trees on the establishment of the network of arbuscular mycorrhizal (AM) fungus hyphae in their rhizospheres. Special acrylic root boxes with three compartments were used for the experiment. Four types of citrus rootstock seedlings, trifoliate orange (Poncirus trifoliata Raf.), sour orange (Citrus aurantium L.), rough lemon (Citrus jambhiri Lush.), and Citrus natsudaidai Hayata, were separately transplanted into one outer compartment in each box, and the seedlings of bahiagrass and V.myuros were separately transplanted into the other outer compartment. An AM fungus, Gigaspora margarita Becker and Hall, was inoculated in the center compartment of each box. Some boxes with both outer compartments without plants and with some plants in only one outer compartment were also prepared. The box with bare × bare had very low density of AM hyphae. There were a few hyphae in bare compartments in the boxes of trifoliate orange × bare, sour orange × bare, rough lemon × bare, and C. natsudaidai × bare. The density of hyphae in the compartments with citrus seedlings and grasses, however, was significantly higher than in every bare compartment, and the hyphae in the compartments with plants penetrated deeply into the sand. In particular, the density in the compartments of citrus seedlings increased when bahiagrass or V. myuros was transplanted as a neighboring plant. The percentage of AM fungus colonization in every plant root was high. New spore formation was observed in compartments with plants, whereas there were few spores in every bare compartment. In particular, the spore formation in bahiagrass compartments was superior to that in other compartments with plants. Our results suggest that the network system by AM hyphae is easily discernible in the rhizospheres between citrus rootstocks and bahiagrass or V. myuros, but bare ground severely inhibits the formation and development of AM hyphal network and reduces the number of AM spores in the soil.  相似文献

In terrestrial ecosystems, plants are frequently in symbiosis with arbuscular mycorrhizal fungi (AMF) with mineral nutrients and photosynthesis carbon exchanges in between. This research sought to identify the effects of phosphorus (P) levels on the nitrogen (N) uptake via extraradical mycelium (ERM) and the mycorrhizal growth response (MGR) of maize plants within the AMF symbiosis. Pots were separated into root compartments and hyphae compartments (HCs) with two layers of a 30‐μm mesh membrane and an air gap in between, where only hyphae could pass through, to avoid both N diffusion and root growth effects. Maize plants were inoculated with Rhizophagus irregularis with different N fertilization in HCs under two different P fertilization levels. Our results indicated that a strong increase in MGR with low‐P fertilization. The same tendency was not observed with high‐P fertilization, although both had a large increase in P concentration as a potential source of growth in shoot tissue of mycorrhizal plants. Substantial effects (10.5% more N) were observed in the case of high‐P availability for the host plants from ERM fed with N, whereas under low‐P conditions ERM may prioritize P uptake rather than N uptake. The AM fungi increase the uptake of N and P, which are most limiting in the soil with fewer forces from soil resources. In addition, there was still more P accumulated than N due to the high N for ERM with high‐P supply. Low N in HCs corresponded with a lower colonization rate in roots but with high hyphae density in HCs; this result suggest that N and P availability might change the ratio of extraradical to intraradical hyphae length.  相似文献

Lonicera confusa, a traditional Chinese medicine herb for treating cold, flu, acute fever, and so forth, is often grown artificially in acidic soils and suffers from phosphorus (P) deficiency. A five-year field experiment was carried out to study the colonization rate, growth, nutrition, and chlorogenic acid content of Lonicera confusa seedlings inoculated with arbuscular mycorrhizal (AM) fungi, Glomus etunicatum and Glomus intraradices. Before transplanting into a field, both AM-inoculated and uninoculated control plants were cultured in nursery beds. In the plants inoculated with the AM fungi, the colonization rate decreased linearly with time and a greater decrease was observed in the plants inoculated with G. intraradices than with G. etunicatum, while the AM colonization increased from 0% to 12.1% in the uninoculated control plants 5 years after transplanting. Plant height, crown diameter, number of new branches, and flower yield increased significantly by AM inoculation as compared to the uninoculated control. Phosphorus concentrations in leaves and flowers increased, and plant uptake of nutrients, e.g., nitrogen (N), P, and potassium (K), was also enhanced significantly by AM inoculation. The Lonicera confusa seedlings had a better response to inoculation of G. intraradices than G. etunicatum in both growth and chlorogenic acid content in flowers. In contrast, both plant P uptake and P concentrations in leaves and flowers were similar between two fungal inoculations. The positive responses of Lonicera confusa to AM inoculation in growth, nutrient uptake, flowering, and chlorogenic acid content in flowers suggested that AM inoculation in nursery beds could promote the plant growth and increase chlorogenic acid content in flowers of Lonicera confusa when grown on acidic and P-deficient soils.  相似文献

We describe a simple technique for the germination of arbuscular mycorrhizal (AM)–fungal spores and their multiplication in pots. Glomus fasciculatum, G. mosseae, and Gigaspora margarita were used. A single wheat seedling was tied to a glass slide, previously covered with filter paper with the help of thread. One single surface‐sterilized AM‐fungal spore was placed on the middle portion of the root of the wheat seedling using a sterilized syringe. The slide was placed vertically in a 100 mL glass beaker filled with 25 mL of root exudates–water (1:4, v/v) solution, which was collected by growing twenty wheat seedlings in a 150 mL beaker filled with 100 mL sterilized distilled water for 7 d. The slide was observed daily using a compound microscope to follow the time course of germination. In this technique, the spore is directly in contact with the host root, and a visualization of spore germination, hyphal development, and appressorium formation is possible without disrupting fungal growth or the establishment of the symbiosis. The method allows to document the germination events and to assess hyphal‐elongation rates by photographing the same spore on consecutive days. The inoculated seedling was used to initiate single‐spore multiplication in a sterilized (autoclave on 3 alternate days at 120°C for 120 min at 1.05 kg cm–2 pressure) potted sandy soil (150 mL volume) into which the slide with the inoculated seedling was inserted carefully through a previously made slit. The wheat seedlings in all pots (4 treatments and 15 replications) became colonized by mycorrhiza, confirming that the establishment of the AM‐fungal symbiosis is highly reproducible. Our technique permits the relatively undisturbed growth of the symbiotic partners, the visualization of germinating AM‐fungal spores, and their multiplication in pots. This simple and low‐cost method facilitates the production of pure lines of AM fungi from single spores, allowing for the study of intraspecific variation and potentiality for cytological, biochemical, physiological, and taxonomical studies.  相似文献

以三叶草为材料,利用三室隔网培养方法,探讨了取自肥料长期定位试验中多年施用与不施用有机肥的田间小区土壤上,接种菌根菌(G.mosseae)对根际土壤酸性和碱性磷酸酶活性的影响。植物生长9周后,收获测定菌丝生长室土壤酸性磷酸酶和碱性磷酸酶活性,并对磷酸酶产生位点进行细胞化学定位。结果表明,接种丛技菌根菌对根际土壤酸性和碱性磷酸酶活性均有增强作用,但作用程度在有机服小区土壤上要大于无机肥小区土壤。根际土壤酸性磷酸酶原位化学定位结果表明,菌丝周围有明显的酸性磷酸酶的反应产物,说明报外丛枝菌根菌丝能直接向外分泌磷酸酶。  相似文献

A pot experiment was conducted to investigate the action mechanisms of arbuscular mycorrhizal (AM) fungi in phosphorus (P) uptake of Capsicum annuum L.in a sterilized fossil Oxisol.Three P levels of 0,10 and 200 mg kg-1 soil (P0,P10 and P200,respectively) without and with AM fungal inoculation were applied as Ca(H2PO4)2·H2O.Shoot dry matter yields and shoot P uptake increased significantly (P > 0.05) by the inoculation of AM fungi at P0 and P10.Root length and P concentration in soil solution increased with the inoculation of AM fungi but the root:shoot ratio decreased or remained constant.Around 50% roots of inoculated plants were infected by AM and the external hyphae amounted to 20 m g-1 soil at P10 and P200.The hyphae surface area of the infected root cylinder amounted to 11 and 2 cm-2 cm-2 root at P0 and P10,respectively.The increased P uptake of inoculated plants was mainly because of an up to 5 times higher P influx of the infected root.Model calculations showed that the root alone could not have achieved the measured P influx in both infected and non-infected roots.But the P influx for hyphae calculated by the model was even much higher than the measured one.The P uptake capacity of hyphae introduced in the model was too high.Model calculations further showed that the depletion zone around roots or hyphae was very narrow.In the case of the root only 7% of the soil volume would contribute P to the plant,while in the case of hyphae it would be 100%.The results together with the model calculations showed that the increased P uptake of AM inoculated plants could be explained partly by the increased P concentration in the soil solution and by the increased P absorbing surface area coming from the external hyphae.  相似文献

A pot experiment was conducted to investigate the effect of epigeic earthworm (Eisenia fetida) and arbuscular mycorrhizal (AM) fungi (Glomus intraradices) on soil enzyme activities and nutrient uptake by maize, which was grown on a mixture of sterilized soil and sand. Maize plants were grown in pots inoculated or not inoculated with AMF, treated or not treated with earthworms. Wheat straw was added as a feed source for earthworms. Mycorrhizal colonization of maize was markedly increased in AM fungi inoculated pots and further increased by addition of epigeic earthworms. AM fungi and epigeic earthworms increased maize shoot and root biomass, respectively. Soil acid phosphatase activity was increased by both earthworms and mycorrhiza, while urease and cellulase activities were only affected by earthworms. Inoculation with AM fungi significantly (p?<?0.001) increased the activity of soil acid phosphatase but decreased soil available phosphorus (P) and potassium (K) concentrations at harvest. Addition of earthworms alone significantly (p?<?0.05) increased soil ammonium-N content, but decreased soil available P and K contents. AM fungi increased maize shoot weight and root P content, while earthworms improved N, P, and K contents in shoots. AM fungi and earthworm interactively increased maize shoot and root biomass through their regulation of soil enzyme activities and on the content of available soil N, P, and K.  相似文献

采用中间隔网的土培根箱试验,对旱作水稻或/和西瓜接种丛枝菌根真菌(简称AM真菌)幼套球囊霉(Glomus etunicatum Becker&Gerdemann),研究了旱作水稻/西瓜间形成菌丝桥并诱导水稻磷酸盐转运蛋白OsPT11的表达和对磷吸收的影响。结果表明:(1)根箱两侧均未接种AM真菌时,旱作水稻和西瓜根系均不形成菌根,水稻根系的磷酸盐转运蛋白OsPT11也不表达。(2)西瓜侧接种AM真菌时,西瓜与水稻间形成的菌丝桥引起水稻菌根的形成,并诱导水稻根系磷酸盐转运蛋白OsPT11表达。(3)菌丝桥侵染和直接接种侵染对旱作水稻和西瓜形成丛枝菌根能达到相同的效果,旱作水稻和西瓜的菌根侵染率分别为80%以上和70%以上。(4)在旱作水稻/西瓜间作系统中,当接种AM真菌时,水稻和西瓜根际有效磷含量显著高于对照处理,水稻地上部全磷含量降低,而西瓜地上部全磷含量升高。  相似文献

在接种G intraradices和G etunicatum的玉米植株盆栽钵中 ,按大田常规用量 ,施用乙草胺、丁草胺、灵达、骠马、百草敌和使它隆 6种除草剂 ,对玉米植株的生物量、AM真菌菌丝根段侵染率、菌丝碱性磷酸酶 (ALP)和琥珀酸脱氢酶 (SDH)活性及土壤中AM真菌菌丝的总量均有显著影响。第 1 0周取样 ,对玉米植株生物量影响较大的除草剂为骠马 (植株生物量为 2 1 1~ 31 3g) ,影响较小的为灵达 (植株生物量为 5 9 7~ 6 5 5g) ,而对照的生物量为 84 4~ 95 7g ;对AM真菌侵染率影响较大的为丁草胺 (AM真菌侵染率为 1 3 7%~1 8 1 %) ,影响较少的为使它隆 (AM真菌侵染率为 30 4 %~ 36 6 %) ,而对照侵染率为 6 3 0 %~ 88 2 %;对AM真菌菌丝ALP和SDH酶活性影响较大的为丁草胺 (ALP为 8 1 %~ 1 0 4 %,SDH为 1 0 6 %~ 1 4 6 %) ,影响较小的为使它隆 (ALP为 1 7 0 %~ 2 0 6 %,SDH为 2 3 1 %~ 2 8 2 %) ,而对照的ALP为 38 0 %~ 5 0 3%,SDH为4 9 7%~ 72 4 %;此外 ,除草剂对土壤中AM真菌菌丝的总量也有显著影响。  相似文献

Arbuscular mycorrhizal (AM) fungi are key organisms of the soil/plant system, influencing soil fertility and plant nutrition, and contributing to soil aggregation and soil structure stability by the combined action of extraradical hyphae and of an insoluble, hydrophobic proteinaceous substance named glomalin-related soil protein (GRSP). Since the GRSP extraction procedures have recently revealed problems related to co-extracting substances, the relationship between GRSP and AM fungi still remains to be verified. In this work the hypothesis that GRSP concentration is positively correlated with the occurrence of AM fungi was tested by using Medicago sativa plants inoculated with different isolates of Glomus mosseae and Glomus intraradices in a microcosm experiment. Our results show that (i) mycorrhizal establishment produced an increase in GRSP concentration - compared to initial values - in contrast with non-mycorrhizal plants, which did not produce any change; (ii) aggregate stability, evaluated as mean weight diameter (MWD) of macroaggregates of 1-2 mm diameter, was significantly higher in mycorrhizal soils compared to non-mycorrhizal soil; (iii) GRSP concentration and soil aggregate stability were positively correlated with mycorrhizal root volume and weakly correlated with total root volume; (iv) MWD values of soil aggregates were positively correlated with values of total hyphal length and hyphal density of the AM fungi utilized.The different ability of AM fungal isolates to affect GRSP concentration and to form extensive and dense mycelial networks, which may directly affect soil aggregates stability by hyphal enmeshment of soil particles, suggests the possibility of selecting the most efficient isolates to be utilized for soil quality improvement and land restoration programs.  相似文献

Previous greenhouse and field studies have shown arbuscular mycorrhizal (AM) plants usually have greater P uptake and growth when raised in undisturbed soil compared to soil disturbed between plantings, such as by tillage. We report here for the first time that AM fungi able to stimulate shoot P uptake in experimental comparisons to non-mycorrhizal plants differ in their ability to bring about similar responses in undisturbed soil compared to disturbed soil. This outcome indicates a difference in functional character between the two stimulation processes. Three isolates of AM fungi were tested for growth promotion of maize (Zea mays L.) in pots in a soil disturbance experiment that included non-mycorrhizal controls. All three fungi colonized roots well and promoted shoot P uptake compared to non-inoculated controls, but only Glomus mosseae was able to stimulate growth in undisturbed soil compared to disturbed soil. This effect was seen when Glomus mosseae was alone or in combination with Gigaspora margarita. However, the presence of Glomus aggregatum in combination with Glomus mosseae prevented any stimulation, presumably due to domination by Glomus aggregatum. The ability of AM fungi to be beneficial to plants in comparison to non-mycorrhizal situations likely relates to the spread of mycelium in the soil and the capacity for nutrient transfer to the root. The ability of an AM fungus to promote growth in undisturbed soil appears to be related to these features and, in addition, a capacity for persistence and retention of functional capacity of the extraradical mycelium from one plant generation to the next.  相似文献

The effects of three commonly used fungicides on the colonization and sporulation by a mixture of three arbuscular mycorrhizal (AM) fungi consisting of Glomus etunicatum (Becker & Gerd.), Glomus mosseae (Nicol. & Gerd.) Gerd. & Trappe, and Gigaspora rosea (Nicol. & Schenck) in symbiosis with pea plants and the resulting response of the host-plant were examined. Benomyl, PCNB, and captan were applied as soil drenches at a rate of 20 mg active ingredient kg-1 soil 2 weeks after transplanting pea seedlings in a silty clay-loam soil containing the mixed inocula of AM fungi (AM plants). Effects of fungicides were compared to untreated plants that were inoculated with fungi (AM control). The effect of mycorrhizal inoculation on plant growth was also examined by including nonmycorrhizal, non-fungicide-treated plants (non-AM control). Fungicides or inoculation with AM fungi had only a small effect on the final shoot weights of pea plants, but had greater effects on root length and seed yield. AM control plants had higher seed yields and lower root lengths than the corresponding non-AM plants, and the fungicide-treated AM plants had intermediate yields and root lengths. Seed N and P contents were likewise highest in AM control plants, lowest in non-AM plants, and intermediate in fungicide-treated AM plants. All three fungicides depressed the proportion (%) of root length colonized by AM fungi, but these differences did not translate to reductions in the total root length that was colonized, since roots were longer in the fungicide-treated AM plants. Pea plants apparently compensated for the reduction in AM-fungal metabolism due to fungicides by increasing root growth. Fungicides affected the population of the three fungi as determined by sporulation at the final harvest. Captan significantly reduced the number, relative abundance, and relative volume of G. rosea spores in the final population relative to the controls. The relative volume of G. etunicatum spores was greater in all the fungicide-treated soils, while G. mosseae relative volumes were only greater in the captan-treated soil. These findings show that fungicides can alter the species composition of an AM-fungal community. The results also show that AM fungi can increase seed yield without enhancing the vegetative shoot growth of host plants.  相似文献

【目的】 探究酸性土壤玉米丛枝菌根侵染对植物磷素吸收的促进作用,以加深理解根外菌丝对局部磷养分的获取如何受丛枝真菌侵染和环境磷养分的影响。 【方法】 以玉米为宿主植物,进行盆栽试验。在低磷酸性土壤上设置供P 0、50、500 mg/kg 3个水平 (P0、P50、P500),供试磷肥为磷酸二氢钾。每个处理再设置局部养分处理,即在每个重复中埋置两个各装有120 g灭菌土 (提前加 P 50 mg/kg) 的塑料小管,分别用孔径为0.45 μm(根系、菌丝均不能进入,以“–H”表示处理) 和50 μm(根系不能进入,菌丝可以进入,以“+H”表示处理) 的尼龙膜封住管口。测定了玉米的生长与磷吸收、土著丛枝菌根真菌的侵染和根外菌丝密度以及菌丝对局部磷养分的获取。 【结果】 1) 玉米株高、叶片SPAD值、全株干重、磷浓度及吸收量都随供磷水平升高而增加,以P50处理的根系干重最高,根冠比随供磷水平上升而降低。3个供磷水平下玉米根系均有不同程度的丛枝菌根真菌侵染。以P50处理的丛枝菌根侵染率、丛枝和孢囊结构发育最好;P0处理的丛枝菌根侵染率、丛枝丰度与P50处理没有显著差异,但孢囊丰度明显下降;P500处理虽然87.2%的根系具有侵染点,但整个根系形成的真菌结构、丛枝和孢囊比例远低于P0和P50处理,丛枝菌根的发育受到严重抑制。2) 土体土 (除塑料管之外的土) 菌丝密度随供磷水平升高而降低,但P0和P50处理差异不显著。–H处理塑料管中的菌丝密度在3个供磷水平下基本不变,保持在极低水平,而+H处理塑料管中的菌丝密度随供磷水平升高而下降。在相同供磷水平下,土体土的菌丝密度最高,其次是+H处理,–H处理的菌丝密度最低。根外菌丝从+H处理塑料管中获取的磷随环境供磷水平的升高而减少。 【结论】 酸性土壤条件下,适当地供磷可以促进玉米根系生长和丛枝菌根真菌的侵染。根外菌丝对局部磷养分的获取受环境磷养分的调控,在环境磷养分较低而局部磷养分高于环境磷养分时,较多的菌丝会进入局部区域获取磷。   相似文献

Biochar application to soils has potential to simultaneously improve soil fertility and store carbon to aid climate change mitigation. While many studies have shown positive effects on plant yields, much less is known about the synergies between biochar and plant growth promoting microbes, such as mycorrhizal fungi. We present the first evidence that arbuscular mycorrhizal (AM) fungi can use biochar as a physical growth matrix and nutrient source. We used monoxenic cultures of the AM fungus Rhizophagus irregularis in symbiosis with carrot roots. Using scanning electron microscopy we observed that AM fungal hyphae grow on and into two contrasting types of biochar particles, strongly attaching to inner and outer surfaces. Loading a nutrient-poor biochar surface with nutrients stimulated hyphal colonization. We labeled biochar surfaces with 33P radiotracer and found that hyphal contact to the biochar surfaces permitted uptake of 33P and its subsequent translocation to the associated host roots. Direct access of fungal hyphae to biochar surfaces resulted in six times more 33P translocation to the host roots than in systems where a mesh prevented hyphal contact with the biochar.We conclude that AM fungal hyphae access microsites within biochar, that are too small for most plant roots to enter (<10 μm), and can hence mediate plant phosphorus uptake from the biochar. Thus, combined management of biochar and AM fungi could contribute to sustainable soil and climate management by providing both a carbon-stable nutrient reservoir and a symbiont that facilitates nutrient uptake from it.  相似文献
