The effect of heat treatment of germinated soybean, lupin, and black bean on chemical composition and protein utilization in rats was evaluated. Heat treatment caused complete inactivation of trypsin inhibitors whereas it did not affect phytic acid levels. Proximate components, minerals, and amino acids did not change, but low molecular weight sugars were affected by heat treatment differently for each germinated legume. The sugar digestibility ratio (total digestible sugars/total nondigestible sugars) in germinated black beans doubled after heat treatment. True protein digestibility (TD) increased with heat treatment only in germinated soybean. Net protein utilization was markedly improved (20%) with heat treatment in germinated soybean and lupin. Utilizable protein of heat-treated germinated legumes was 289, 236, and 132 g/kg of legume dry weight for soybean, lupin, and black bean, respectively. Supplementation with methionine did not alter TD but improved all other indices of protein utilization in the germinated legumes, particularly in black bean. All three germinated legumes become equivalent in protein quality when heating and supplementation with methionine are combined with germination. The use of germinated heat-treated soybean, lupin, and black bean on their own and/or as food ingredients is nutritionally advantageous due to the low content of nondigestible oligosaccharides and the high protein utilization.  相似文献

The composition and contents of nutritional factors such as proteins, lipids, carbohydrates, fibers, amino acids, and antinutritional factors such as trypsin inhibitors, phytic acid, and tannins were compared in soybean and fababean seeds with emphasis placed on the nutritional improvement of the seeds by cortex removal. Protein hydrolysis analysis for both whole seeds and seed with cortex removed revealed the presence of a large amount of lysine, arginine, aspartic acid, glutamic acid, glycine, and leucine while these seeds contained a low level of tryptophan, cystine, and methionine. Some antinutritional factors such as trypsin inhibitors, phytic acid, and tannins were detected in soybean and fababean seeds: phytic acid content and trypsin inhibitor activity were higher in soybean seeds than in fababean seeds while the difference in the tannin content was less pronounced. It was found that most of the tannins occurred in the cortex of the soybean and fababean seeds. Tannins are polyphenolic compounds that readily form indigestible complexes with proteins and other macromolecules under specific environmental conditions. By removal of the cortex, tannins were almost completely eliminated without changes in the protein composition and amino acids. From these results, it is assumed that since soybean and fababean seeds contained a high concentration of antinutritional factors in the cortex such as tannins, the utilization of the legume seeds after removal of all of the cortex is suitable for human diet or industrial products.  相似文献

The basic composition (moisture, total carbohydrates, dietary fiber, crude fat, ash, nitrogen, and protein) and amino acid contents were determined in the cultivated mushrooms Agaricus bisporus/white, Agaricus bisporus/brown, Lentinula edodes, and Pleurotus ostreatus. In addition, nitrogen-to-protein conversion factors were calculated for each species by dividing the sums of amino acid residues with total (Kjeldahl) nitrogen contents. The dry matter contents of mushrooms varied from 7.7% to 8.4%. The dry matter of mushrooms contained large amounts of carbohydrates, from 4.5 (A. bisporus/white) to 5.8 g/100 g fresh weight (L. edodes). L. edodes proved to be an especially good source of dietary fiber (3.3 g/100 g fresh weight); the other mushrooms contained 1.5-2.4 g/100 g fresh weight. Crude fat, ash, and protein (based on amino acid analysis) contents of the mushrooms varied 0.31-0.35, 0.49-0.78, and 1.8-2.09 g/100 g fresh weight, respectively. Mushrooms proved to be good sources of almost all essential amino acids when compared with common vegetables. The mean nitrogen-to-protein conversion factor analyzed in the present study was 4.7 +/- 0.21. When using this factor, a very good estimation of protein contents could be obtained for the main species of mushrooms cultivated in Finland.  相似文献

Proteins dispersed in the continuous phase of oil-in-water emulsions are capable of inhibiting lipid oxidation reactions. The antioxidant activity of these proteins is thought to encompass both free radical scavenging by amino acid residues and chelation of prooxidative transition metals; however, the precise mechanism by which this occurs remains unclear. In this study, the oxidative stability of cysteine, tryptophan, and methionine residues in continuous phase beta-lactoglobulin (beta-Lg) in a Brij-stabilized menhaden oil-in-water emulsion was determined. The presence of low concentrations of continuous phase beta-Lg (250 and 750 microg/mL) significantly inhibited lipid oxidation as determined by lipid hydroperoxides and thiobarbituric acid reactive substances analysis. It was observed that cysteine oxidized before tryptophan in beta-Lg, and both residues oxidized before lipid oxidation could be detected. No oxidation of the methionine residues of beta-Lg was observed despite its reported high oxidative susceptibility. It is conceivable that surface exposure of amino acid residues greatly affects their oxidation kinetics, which may explain why some residues are preferentially oxidized relative to others. Further elucidation of the mechanisms governing free radical scavenging of amino acids could lead to more effective applications of proteins as antioxidants within oil-in-water food emulsions.  相似文献

Soybeans in general contain 35-40% protein. Efforts are underway to increase further this protein content, thus enhancing their nutritive value. Even though higher protein is a desirable characteristic, whether such an increase will be accompanied by enhanced protein quality is not known. Soybean protein quality could be significantly improved by increasing the concentration of the sulfur-containing amino acids, cysteine and methionine. To ascertain if a correlation existed between protein quantity and quality, a comparison of the amino acids of soybeans differing in protein content was made. Soybeans with higher protein content had a significantly lower percentage of sulfur amino acids, while those with lower protein exhibited a higher content of cysteine and methionine. Nitrogen application elevated the protein content but lowered that of the sulfur amino acids. Transmission electron microscopy examination of thin sections of low protein soybean seeds revealed several protein storage vacuoles that were partially filled with storage proteins. Fluorescence two-dimensional difference gel electrophoresis of soybean seed proteins revealed that nitrogen application favored the accumulation of the beta-subunit of beta-conglycinin while decreasing the accumulation of Bowman-Birk protease inhibitor (BBI), a protein rich in cysteine. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis of 60% 2-propanol-extracted proteins showed a drastic reduction in the accumulation of BBI with increasing protein content. Northern blot analysis indicated that nitrogen had a negative influence on the expression of the BBI gene. Our results indicate that the negative correlation between total protein and sulfur amino acid content is mostly mediated by the differential accumulation of BBI.  相似文献

Soy protein is a major component of the diet of food-producing animals and is increasingly important in the human diet. However, soy protein is not an ideal protein because it is deficient in the essential amino acid methionine. Methionine supplementation benefits soy infant formulas, but apparently not food intended for adults with an adequate nitrogen intake. Soy protein content of another essential amino acid, lysine, although higher than that of wheat proteins, is still lower than that of the milk protein casein. Adverse nutritional and other effects following consumption of raw soybean meal have been attributed to the presence of endogenous inhibitors of digestive enzymes and lectins and to poor digestibility. To improve the nutritional quality of soy foods, inhibitors and lectins are generally inactivated by heat treatment or eliminated by fractionation during food processing. Although lectins are heat-labile, the inhibitors are more heat-stable than the lectins. Most commercially heated meals retain up to 20% of the Bowman-Birk (BBI) inhibitor of chymotrypsin and trypsin and the Kunitz inhibitor of trypsin (KTI). To enhance the value of soybeans in human nutrition and health, a better understanding is needed of the factors that impact the nutrition and health-promoting aspects of soy proteins. This paper discusses the composition in relation to properties of soy proteins. Also described are possible beneficial and adverse effects of soy-containing diets. The former include soy-induced lowering of cholesterol, anticarcinogenic effects of BBI, and protective effects against obesity, diabetes, irritants of the digestive tract, bone, and kidney diseases, whereas the latter include poor digestibility and allergy to soy proteins. Approaches to reduce the concentration of soybean inhibitors by rearrangement of protein disulfide bonds, immunoassays of inhibitors in processed soy foods and soybean germplasm, the roles of phytoestrogenic isoflavones and lectins, and research needs in all of these areas are also discussed. This integrated overview of the widely scattered literature emphasizes general concepts based on our own studies as well as recent studies by others. It is intended to stimulate interest in further research to optimize beneficial effects of soy proteins. The payoff will be healthier humans and improved animal feeds.  相似文献

Heat treatment of dairy products leads to structural changes of proteins, which can severely decrease the nutritional value [Mauron, J. J. Nutr. Sci. Vitaminol. (Tokyo) 1990, 36 (Suppl. 1), S57-69]. In this study, model solutions of the two main whey proteins, alpha-lactalbumin and beta-lactoglobulin, respectively, were incubated with lactose, and modifications were monitored by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS). Lactulosyl residues were the most abundant modifications of alpha-lactalbumin and beta-lactoglobulin. Up to four of these adducts were identified on the proteins. Enzymatical digest with endoproteinase AspN prior to mass spectrometric analysis allowed the detection of further modifications and their localization in the amino acid sequence. Most prominent modifications were lactulosyllysine, Nepsilon-carboxymethyllysine, oxidation of lysine to aminoadipic semialdehyde, oxidation of methionine to methionine sulfoxide, cyclization of N-terminal glutamic acid to a pyrrolidone, and oxidation of cysteine or tryptophan. The presence of methionine oxidation was deduced from a control protein that had been oxidized by hydrogen peroxide. These studies establish MALDI-TOF-MS as a reliable tool to monitor chemical modifications of nutritional proteins during food processing.  相似文献

Total protein content and level of 14 amino acid in mature acorns of 20 Quercus taxa from Turkey were studied. The range of total protein amounts between 2.75 and 8.44% were detected among taxa. Similar values in related species and variety level were observed. The amino acid profiles for each taxon were characteristically different and high variability of individual amino acid concentration was present at variety, subspecies and species levels. Different amino acid concentrations, relative percentages and critical values of some amino acids are thought to be additional considerable parameters for diagnosis of Quercus. Generally higher total quantity of amino acids in section Quercus and lower values in section Ilex were observed. When the essential and non-essential amino acids are expressed as a relative percent of total protein indicating quality of proteins, the ratios varied significantly among taxa (p < 0.05). But, no significant difference at section level were detected. Major amino acids were aspartic acid and glutamic acid showing the largest variations and the lowest levels were detected for methionine. Amino acid concentrations ranged from 1665 for aspartic acid to 13 mg/100 g dry wt. for methionine. All taxa had relatively higher amounts of leucin, lysin and valine than other essential amino acids. The highest quantity of protein and amino acids was recorded for Q. infectoria ssp. boissieri and the lowest for Q. pontica. The level of all essential amino acids in examined taxa was not sufficient compared to FAO scoring pattern for children (1985). But, threonine and valine in Q. infectoria ssp. boissieri and isoleucine in Q. petraea ssp. iberica show remarkable concentrations to the requirements. All examined essential amino acids among taxa generally provide adequate levels for adults according to FAO standard.  相似文献

【目的】氨基酸能被作物吸收利用,影响作物的养分吸收和品质形成。采用盆栽试验, 通过三种氨基酸组氨酸(His)、 甘氨酸(Gly)和甲硫氨酸(Met)喷施,研究三种氨基酸对菜心生长及品质的影响。【方法】试验设置六个浓度梯度为0、 50、 100、 200、 400、 800 mg/kg。氨基酸喷施时间为34叶期、 78叶期和抽苔期。喷施5天后测定株高、 倒4叶叶长、 叶宽等生长指标。收获期地上部测重,测定Vc含量、 可溶性糖含量、 蛋白质含量,离子色谱法测定硝酸盐和草酸含量。【结果】三种氨基酸对菜心生长、 产量、 地上部蛋白质、 硝酸盐和草酸的影响如下, 1) 50~400 mg/kg的组氨酸和甲硫氨酸有增产效果,其中喷施100 mg/kg甲硫氨酸菜心产量增幅最大,达17.3%,氨基酸高浓度(800 mg/kg)喷施时,产量下降。2)三种氨基酸处理的菜心地上部蛋白质含量明显升高,且氨基酸喷施浓度和蛋白质含量之间呈极显著正相关关系(r=0.4026)。3)菜心地上部硝酸盐含量随氨基酸喷施浓度的提高先下降后上升,其中喷施100 mg/kg的甘氨酸对降低菜心地上部硝酸盐含量的效果最显著,比对照降低30.7%; 喷施高浓度氨基酸(800 mg/kg)其硝酸盐含量反而比对照高。4)草酸在植物体中普遍存在会影响蔬菜的品质,喷施氨基酸后菜心地上部草酸含量均下降,甲硫氨酸处理与对照相比可降低54.5%~68.3%。【结论】50~200 mg/kg的组氨酸、 甘氨酸和甲硫氨酸喷施能在不同程度上促进菜心生长和增产,提高菜心地上部蛋白质含量,降低硝酸盐和草酸含量。该研究结果可为氨基酸在蔬菜生产上的应用,尤其是如何降低草酸含量、 提高蔬菜品质提供依据。  相似文献

The suitability of a one-step derivatization procedure using N-methyl-N-(tert-butyldimethylsilyl)trifluoroacetamide for the simultaneous assay of 22 free amino acids and its application for their analysis in six animal source foods (pork, dry cured ham, chicken stock, fresh cheese, ripened cheese, and dry salted sardine) by GC-MS were studied. All 22 free amino acid derivatives were correctly detected and resolved. Reproducibility (%RSD) of the method was in the range of 1.9-12.2%. Detection and quantitation limits of the analytical procedure ranged from 0.01 to 0.46 mg/100 g dry weight and from 0.02 to 1.55 mg/100 g dry weight, respectively. The calibration curves were linear within the range 0.1-15.0 mg/100 g with correlation coefficient values (R(2)) from 0.9891 to 0.9983. All analyzed food products showed free amino acid contents similar to those found in the scientific literature. The proposed GC-MS method for the determination of free amino acids in animal source food can be used in routine for both analytical and research purposes.  相似文献

Effects of lipoxygenase I-B (LOX)-catalyzed oxidation of linoleic acid on soybean proteins was evaluated by electron spin resonance (ESR) and fluorescence spectroscopy in different model systems in the presence or absence of antioxidants. A strong central singlet signal was detected by ESR spectroscopy and identified as the carbon radical (g value range 2.0041-2.0054). A downfield shoulder attributed to the sulfur radical (g value 2.019-2.028) was also observed. The changes in soybean proteins were accompanied by an increase in fluorescence, indicating the formation of cross-links. Natural antioxidants such as ascorbic acid and alpha-tocopherol as well as synthetic antioxidants butyl hydroxytoluene (BHT) inhibited the development of both the free radical signal and the fluorescence when added to soybean proteins prior to incubation with linoleic acid and lipoxygenase I-B; the central singlet signal attributed to the carbon radical was reduced by 35-65%. This paper clearly indicates direct free radical transfer from oxidizing linoleic acid catalyzed by LOX to soybean proteins.  相似文献

Accurate and detailed amino acid determinations were carried out on nine northern adapted soybean cultivars to ascertain whether their amino acid profiles could be used as potentially useful indices for assessing their protein quality. The cultivars were Maple Amber, Maple Donovan, Maple Glen, Maple Isle, Maple Presto, Maple Ridge, and three brown seed coat near-isogenic lines, Maple Presto Brown, Maple Ridge Brown, and Maple Arrow Brown. Their total protein and amino acid composition were compared with those of an established cultivar, Maple Arrow. Mean protein values for the new cultivars ranged from 30.1 to 33.1% compared to Maple Arrow, which was 33.2%. The total nitrogen content was also variable among these cultivars, ranging from 5.0 to 5.4%. All nine Maple series soybean cultivars were higher in their essential amino acid (EAA) content, that is, EAA(9) = 45.2-46.5%, than the FAO/WHO reference protein pattern value of EAA(9) = 33.9%, for a 2-5-year-old child. Each of the nine new soybean cultivars was limited only in methionine and to a lesser extent in valine and isoleucine and had a protein digestibility corrected amino acid score of 91% for all cultivars, compared to the value of egg protein (97%). These results suggest that the most accurate evaluation of protein quality in soybeans, and possibly other legumes and cereals, is by the protein digestibility-corrected amino acid score.  相似文献

Finger millet (Eleusine coracana), kidney beans (Phaseolus vulgaris), peanuts (Arachis hypogoea), and mango (Mangifera indica) were processed separately and then combined, on the basis of their amino acid scores and energy content, into a complementary food for children of weaning age. The finger millet and kidney beans were processed by germination, autoclaving, and lactic acid fermentation. A mixture containing, on a dry matter basis, 65.2, 19.1, 8.0, and 7.7% of the processed finger millet, kidney beans, peanuts, and mango, respectively, gave a composite protein with an in vitro protein digestibility of 90.2% and an amino acid chemical score of 0.84. This mixture had an energy density of 16.3 kJ.g(-1) of dry matter and a decreased antinutrient content and showed a measurable improvement in the in vitro extractability for calcium, iron, and zinc. A 33% (w/v) pap made from a mix of the processed ingredients had an energy density of 5.4 kJ.g(-1) of pap, which is sufficient to meet the energy requirements of well-nourished children of 6-24 months of age at three servings a day and at the FAO average breast-feeding frequency.  相似文献

Zabrotes subfasciatus (Boheman) is the main pest of common beans ( Phaselous vulgaris ). Wild legume seeds from Olneya tesota contain a lectin, PF2, that shows insecticidal activities against this insect. The binding of PF2 to midgut glycoproteins of 20-day-old larvae was evaluated using PF2 affinity chromatography. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) analysis of the proteins retained on the gel revealed several putative glycoproteins, ranging in mass from 17 to 97 kDa. Subsequent protein digestion and analysis by liquid chromatography-tandem mass spectrometry (LC-MS/MS) provided amino acid fragments that identified an α-tubulin, cytochrome c oxidase subunit I, an odorant receptor, and a lysozyme from available insect sequence databases. The potential of these proteins to serve as part of the mechanisms involved in the insecticidal activity of PF2 to Z. subfasciatus is discussed.  相似文献

The interactions taking place in composite dough containing rice flour and soybean proteins (5% w/w) in the presence of transglutaminase, an enzyme with cross‐linking activity, were studied using different electrophoretic analyses. The interaction between rice proteins and soybean proteins was intensified by the formation of new intermolecular covalent bonds catalyzed by transglutaminase and the indirect formation of disulfide bonds among proteins. The main protein fractions involved in those interactions were both β‐conglycinin and glycinin of soybean and the glutelins of the rice flour, although albumins and globulins were also cross‐linked. The addition of soybean proteins to rice flour improves the amino acid balance and they also might play an important role on the rice dough properties because soybean proteins interact with rice proteins, yielding protein aggregates of high molecular weight.  相似文献

通过田间试验研究不同品种蚕豆枯萎病病情指数的差异,并通过水培试验鉴定蚕豆根系分泌物中氨基酸组分并测定氨基酸的含量,分析各氨基酸组分与蚕豆枯萎病病情指数的相关性。结果表明:根系分泌物中氨基酸总量随着蚕豆枯萎病抗性的降低而升高。感病品种和中抗品种中检出15种氨基酸,而抗病品种中检出14种,组氨酸只存在于中抗品种中,脯氨酸仅在感病品种中检测到,3个蚕豆品种根系分泌物中均未检出精氨酸。丝氨酸(Ser)、蛋氨酸(Met)和赖氨酸(Lys)与枯萎病病指呈负相关关系,以Ser的相关系数最高,其他13种氨基酸含量与蚕豆枯萎病的病情指数呈正相关。蚕豆根系分泌物中Ser、Met和Lys含量及Ser/Gly、Ser/Ala比值高能抑制枯萎病的发生与发展,而天门冬氨酸(Asp)、苏氨酸(Thr)、甘氨酸(Gly)、丙氨酸(Ala)、缬氨酸(Val)、酪氨酸(Tyr)、苯丙氨酸(Phe)含量高时能促进枯萎病的发生。不同蚕豆品种根系分泌的氨基酸含量与组分的差异是影响蚕豆对枯萎病抗性差异的重要原因之一。  相似文献

Seeds of finger millet (Eleucine coracan (L.) Gaertner) and kidney beans (Phaseolus vulgaris L.) were sprouted, autoclaved, and fermented during the processing of a weaning (complementary) food for children. Relative changes in individual amino acids with processing were evaluated. Finger millet and kidney beans both showed a good percentage of essential to total amino acids, with 44. 2-44.9% in finger millet and 44.2-45.1% in kidney beans, when compared to 33.9% for the FAO/WHO reference protein for 2-5 year old children. Sprouting resulted in a significant decrease in lysine in kidney beans. Autoclaving caused significant decreases in histidine, while fermentation significantly decreased phenylalanine and increased tryptophan in finger millet. The leucine-to-lysine ratio, which is an indicator of the pellagragenic character of a protein, was significantly improved in finger millet by both sprouting and fermentation.  相似文献

A high-performance anion exchange chromatographic method was adapted for the quantitative determination of phytic acid and inositol pentakisphosphate isomers (excluding enantiomers) in foods. Because of the cost and limited availability of inositol phosphate standards, a phytic acid sodium salt standard was used for the calculation of an average relative response factor for the quantification of inositol pentakisphosphate isomers, and the purity of phytic acid sodium salt standard was also accurately established. The detection limits (S/N = 3) for phytic acid and inositol pentakisphosphates were in the range of 1.5-3.4 microM (0.1-0.2 microg/100 microL). This method has been successfully applied to the determination of phytic acid and inositol pentakisphosphates in a variety of beans and nuts after extraction with 0.5 M HCl and cleanup with solid phase extraction cartridges. The results demonstrated that there was a strong correlation between either the phytic acid content or the total content of phytic acid together with inositol pentakisphosphates and the total dietary fiber content in the group of all raw dry beans and in the group of raw dry black beans but not in the group of raw dry red kidney beans, which was probably due to the insufficient number of the raw dry red kidney bean samples.  相似文献

There have been prior investigations concerning the environmental effects and especially soil conditions upon the proximate analysis and mineral content of grains. However, the studies are not complete and have not involved beans grown in the northwestern regions of Argentina. For this reason, this study was initiated to determine the concentration of protein, moisture, ash, fat, and minerals of various bean samples grown in northwestern Argentina. Six varieties of beans were taken from seven different regions. AOAC standard methods were used for chemical analysis. The elements analyzed for all bean samples show that moisture varies from 12 to 14%, proteins from 18 to 22%, fat from 0.7 to 1.20%, copper from 0.8 to 1.20 mg/100 g, iron from 9 to 18 mg/100 g, zinc from 2.5 to 4 mg/100 g, and phosphorus from 295 to 542 mg/100 g. No arsenic was detected in the bean samples. Different analyzed bean varieties were significantly different for proximate composition and mineral content, and each variety from different regions of northwestern Argentina present significant differences.  相似文献
