Purpose

Nitrification and denitrification in the N cycle are affected by various ammonia oxidizers and denitrifying microbes in intensive vegetable cultivation soils, but our current understanding of the effect these microbes have on N2O emissions is limited. The nitrification inhibitor, 3,4-dimethylpyrazole phosphate (DMPP), acts by slowing nitrification and is used to improve fertilizer use efficiency and reduce N losses from agricultural systems; however, its effects on nitrifier and denitrifier activities in intensive vegetable cultivation soils are unknown.

Materials and methods

In this study, we measured the impacts of DMPP on N2O emissions, ammonia oxidizers, and denitrifying microbes in two intensive vegetable cultivation soils: one that had been cultivated for a short term (1 year) and one that had been cultivated over a longer term (29 years). The quantitative PCR technique was used in this study. Three treatments, including control (no fertilizer), urea alone, and urea with DMPP, were included for each soil. The application rates of urea and DMPP were 1800 kg ha?1 and 0.5% of the urea-N application rate.

Results and discussion

The application of N significantly increased N2O emissions in both soils. The abundance of ammonia-oxidizing bacteria (AOB) increased significantly with high rate of N fertilizer application in both soils. Conversely, there was no change in the growth rate of ammonia-oxidizing archaea (AOA) in response to the applied urea despite the presence of larger numbers of AOA in these soils. This suggests AOB may play a greater role than AOA in the nitrification process, and N2O emission in intensive vegetable cultivation soils. The application of DMPP significantly reduced soil NO3?-N content and N2O emission, and delayed ammonia oxidation. It greatly reduced AOB abundance, but not AOA abundance. Moreover, the presence of DMPP was correlated with a significant decrease in the abundance of nitrite reductase (nirS and nirK) genes.

Conclusions

Long-term intensive vegetable cultivation with heavy N fertilization altered AOB and nirS abundance. In vegetable cultivation soils with high N levels, DMPP can be effective in mitigating N2O emissions by directly inhibiting both ammonia oxidizing and denitrifying microbes.

Agricultural soil is a major source of nitrous oxide (N2O), and the application of nitrogen and soil drainage are important factors affecting N2O emissions. This study tested the use of polymer-coated urea (PCU) and polymer-coated urea with the nitrification inhibitor dicyandiamide (PCUD) as potential mitigation options for N2O emissions in an imperfectly drained, upland converted paddy field. Fluxes of N2O and methane (CH4), ammonia oxidation potential, and ammonia-oxidizing bacteria (AOB) and ammonia-oxidizing archaea (AOA) abundances were monitored after the application of PCU, PCUD, and urea to upland soil. The results showed that urea application increased the ammonia oxidation potential and AOB and AOA abundances; however, the increase rate of AOB (4.6 times) was much greater than that of AOA (1.8 times). These results suggested that both AOB and AOA contributed to ammonia oxidation after fertilizer application, but the response of AOB was greater than AOA. Although PCU and PCUD had lower ammonia oxidation potential compared to urea treatment, they were not effective in reducing N2O emissions. Large episodic N2O emissions (up to 1.59 kg N ha?1 day?1) were observed following heavy rainfall 2 months after basal fertilizer application. The episodic N2O emissions accounted for 55–80 % of total N2O emissions over the entire monitoring period. The episodic N2O emissions following heavy rainfall would be a major source of N2O in poorly drained agricultural fields. Cumulative CH4 emissions ranged from ?0.017 to ?0.07 kg CH4 ha?1, and fertilizer and nitrification inhibitor application did not affect CH4 oxidation.  相似文献

Purpose

Yellow clay paddy soil (Oxisols) is a low-yield soil with low nitrogen use efficiency (NUE) in southern China. The nitrification inhibitor nitrapyrin (2-chloro-6- (tricholoromethyl)-pyridine, CP) has been applied to improve NUE and reduce environmental pollution in paddy soil. However, the effects of nitrapyrin combined with nitrogen fertilizers on ammonia oxidizers in yellow clay paddy soil have not been examined.

Materials and methods

A randomized complete block design was set with three treatments: (1) without nitrogen fertilizer (CK), (2) common prilled urea (PU), and (3) prilled urea with nitrapyrin (NPU). Soil samples were collected from three treatments where CK, PU, and NPU had been repeatedly applied over 5 years. Soil samples were analyzed by quantitative PCR and 454 high-throughput pyrosequencing of the amoA gene to investigate the influence of nitrapyrin combined with nitrogen on the abundance and community structure of ammonia oxidizers in yellow clay paddy soil.

Results and discussion

The potential nitrification rate (PNR) of the soil was significantly correlated with the abundances of both ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB). Application of urea significantly stimulated AOA and AOB growth, whereas nitrapyrin exhibited inhibitory effects on AOA. Phylogenetic analysis showed that the most dominant operational taxonomic units (OTUs) of AOA and AOB were affiliated with the Nitrosotalea cluster and Nitrosospira cluster 12, respectively. AOA and AOB community structures were not altered by urea and nitrapyrin application.

Conclusions

Nitrogen fertilization stimulated nitrification and increased the population sizes of AOA and AOB. Nitrapyrin affected the abundance, but not community structure of ammonia oxidizers in yellow clay soil. Our results suggested that nitrapyrin improving NUE and inhibiting PNR was attributable to the inhibition of AOA growth.

Nitrous oxide (N2O) is a potent greenhouse gas and, in grazed grassland systems where animals graze outdoor pastures, most of the N2O is emitted from animal urine nitrogen (N) deposited during grazing. Recently, ammonia-oxidizing archaea (AOA) were found to be present in large numbers in soils as well in the ocean, suggesting a potentially important role for AOA, in addition to ammonia-oxidizing bacteria (AOB), in the nitrogen cycle. The relationship between N2O emissions and AOB and AOA populations is unknown. Soil moisture and nitrogen (N) are two important factors influencing N2O emissions and the growth of microorganisms. Here, we carried out a microcosm experiment to evaluate effects of soil moisture level and N fertilizer type on N2O emissions and abundances and composition of associated microbial communities in the two typical arable soils. The abundances and community composition of functional microbes involved in nitrification and denitrification were determined via quantitative PCR (qPCR) and terminal restriction length fragment polymorphism (T-RFLP), respectively. Results showed that N2O production was higher at 90% water-filled pore (WFPS) than at 50% WFPS. The N2O emissions in the two soils amended with ammonium were higher than those amended with nitrate, especially at relatively high moisture level. In both soils, increased soil moisture stimulated the growth of ammonia-oxidizing bacteria (AOB) and nitrite reducer (nirK). Ammonium fertilizer treatment increased the population size of AOB and nirK genes in the alluvial soil, while reduced the abundances of ammonia-oxidizing archaea (AOA) and denitrifiers (nirK and nosZ) in the red soil. Nitrate addition had a negative effect on AOA abundance in the red soil. Total N2O emissions were positively correlated to AOB abundance, but not to other functional genes in the two soils. Changed soil moisture significantly affected AOA rather than AOB community composition in both soils. The way and extent of N fertilizers impacted on nitrifier and denitrifier community composition varied with N form and soil type. These results indicate that N2O emissions and the succession of nitrifying and denitrifying communities are selectively affected by soil moisture and N fertilizer form in the two contrasting types of soil.  相似文献

Impacts of biochar addition on nitrous oxide (N2O) and carbon dioxide (CO2) emissions from paddy soils are not well documented. Here, we have hypothesized that N2O emissions from paddy soils could be depressed by biochar incorporation during the upland crop season without any effect on CO2 emissions. Therefore, we have carried out the 60-day aerobic incubation experiment to investigate the influences of rice husk biochar incorporation (50 t ha−1) into two typical paddy soils with or without nitrogen (N) fertilizer on N2O and CO2 evolution from soil. Biochar addition significantly decreased N2O emissions during the 60-day period by 73.1% as an average value while the inhibition ranged from 51.4% to 93.5% (P < 0.05–0.01) in terms of cumulative emissions. Significant interactions were observed between biochar, N fertilizer, and soil type indicating that the effect of biochar addition on N2O emissions was influenced by soil type. Moreover, biochar addition did not increase CO2 emissions from both paddy soils (P > 0.05) in terms of cumulative emissions. Therefore, biochar can be added to paddy fields during the upland crop growing season to mitigate N2O evolution and thus global warming.  相似文献

Literature reports on N2O and NO emissions from organic and mineral agricultural soil amended with N-containing fertilizers have reached contradictory conclusions. To understand the influence of organic manure (OM) and chemical fertilizer application on N2O and NO emissions, we conducted laboratory incubation experiments on an agricultural sandy loam soil exposed to different long-term fertilization practices. The fertilizer treatments were initiated in 1989 at the Fengqiu State Key Agro-ecological Experimental Station and included a control without fertilizer (CK), OM, mineral NPK fertilizer (NPK), mineral NP fertilizer (NP), and mineral NK fertilizer (NK). The proportion of N emitted as NO and N2O varied considerably among fertilizer treatments, ranging from 0.83% to 2.50% as NO and from 0.08% to 0.36% as N2O. Cumulative NO emission was highest in the CK treatment after NH 4 + -N was added at a rate of 200 mg N kg?1 soil during the 612-h incubation period, whereas the long-term application of fertilizers significantly reduced NO emission by 54–67%. In contrast, the long-term application of NPK fertilizer and OM significantly enhanced N2O emission by 95.6% and 253%, respectively, compared to CK conditions. The addition of NP fertilizer (no K) significantly reduced N2O emission by 25.5%, whereas applications of NK fertilizer (no P) had no effect. The difference among the N-fertilized treatments was due probably to discrepancies in the N2O production potential of the dominant ammonia-oxidizing bacteria (AOB) species rather than AOB abundance. The ratio of NO/N2O was approximately 24 in the CK treatment, significantly higher than those in the N-fertilized treatments (3–11), and it decreased with increasing N2O production potential in N-fertilized treatments. Our data suggests that the shift in the dominant AOB species might produce reciprocal change in cumulative NO and N2O emissions.  相似文献

[目的]在农业生产中,脲酶抑制剂(urease inhibitor,UI)与硝化抑制剂(nitrification inhibitor,NI)常作为氮肥增效剂来提高肥料利用率。本文研究了在我国南方红壤稻田施用脲酶抑制剂与硝化抑制剂后,土壤中氨氧化细菌(ammonia oxidizing bacteria,AOB)、氨氧化古菌(ammonia-oxidizing archaea,AOA)以及反硝化细菌的丰度以及群落结构的变化特征,旨在揭示抑制剂的作用机理及其对土壤环境的影响。[方法]试验在我国南方红壤稻田进行,共设5个处理:1)不施氮肥(CK);2)尿素(U);3)尿素+脲酶抑制剂(U+UI);4)尿素+硝化抑制剂(U+NI);5)尿素+脲酶抑制剂+硝化抑制剂(U+UI+NI),3次重复。脲酶抑制剂与硝化抑制剂分别为NBPT[N-(n-butyl)thiophosphrictriamide,N-丁基硫代磷酰三胺]和DMPP(3,4-dimethylpyrazole phosphate,3,4-二甲基吡唑磷酸盐)。通过荧光定量PCR(Real-time PCR)研究水稻分蘖期与孕穗期抑制剂对三类微生物标记基因拷贝数的影响,并分析土壤铵态氮、硝态氮与三种菌群丰度的相关性;利用变性梯度凝胶电泳(DenaturingGradient Gel Electrophoresis,DGGE)分析抑制剂对土壤AOB、AOA以及反硝化细菌群落结构的影响,并对优势菌群进行系统发育分析。[结果]1)荧光定量PCR结果表明,施用氮肥对两个时期土壤中AOB的amoA基因与反硝化细菌nirK基因的拷贝数均有显著提高,而对AOA的amoA基因始终没有明显影响;AOB与nirK反硝化细菌的丰度与两个时期的铵态氮含量、分蘖期的硝态氮含量呈极显著正相关,与孕穗期的硝态氮含量相关性不显著;DMPP仅在分蘖期显著减少了AOB的amoA基因拷贝数,表明DMPP主要通过限制AOB的生长来抑制稻田土壤硝化过程;NBPT对三类微生物的丰度无明显影响;2)DGGE图谱表明,在分蘖期与孕穗期,施用氮肥均明显增加了图谱中AOB的条带数,而对AOA却没有明显影响;氮肥明显增加了孕穗期反硝化细菌的条带数;与氮肥的影响相比,抑制剂NBPT与DMPP对AOA、AOB以及反硝化菌的群落结构影响甚微;系统发育分析结果表明,与土壤中AOB的优势菌群序列较为接近的有亚硝化单胞菌和亚硝化螺菌。[结论]在南方红壤稻田中,施入氮肥可显著提高AOB与反硝化细菌的丰度,明显影响两种菌群的群落结构,而AOA较为稳定;NBPT对三类微生物的群落结构丰度无明显影响;硝化抑制剂DMPP可抑制AOB的生长但仅表现在分蘖期,这可能是其缓解硝化反应的主要途径;这也说明二者对土壤生态环境均安全可靠。  相似文献

【目的】本研究旨在明确硝化抑制剂对稻田土壤氮素周转的影响,探讨抑制剂提高氮肥利用率及微生物响应机理。【方法】以草甸黑土发育的水稻土为研究对象,进行了两组培养试验 (25℃),培养周期均为150天。共设4个处理:1) 不施肥 (CK);2) 单施尿素 (Urea);3) 尿素 + 双氰胺 (Urea + DCD);4) 尿素 + 3, 4-二甲基吡唑磷酸盐 (Urea + DMPP)。一组试验从培养第1天起,抽取气体样品,用气相色谱法测定N2O排放量。另一组试验从培养第1天直到结束,取土样测定氨氧化细菌和氨氧化古菌数量,采用荧光定量PCR等技术测定nirK基因和nirS基因拷贝数,用常规方法测定土壤无机氮含量。【结果】施用尿素显著增加了N2O排放量,其中85%的N2O排放发生在培养开始后的前两周内。Urea + DMPP处理土壤NH4+浓度在前23天稳定在较高水平,与Urea处理相比,N2O减排率为78.3%,Urea + DCD处理为21.6%。Urea + DMPP处理排放系数为0.05%,Urea + DCD为0.18%,Urea + DMPP处理显著低于Urea + DCD处理。施用尿素培养,土壤氨氧化细菌 (AOB) 数量显著增加而氨氧化古菌 (AOA) 数量则显著减少。添加DCD和DMPP能显著抑制AOB的数量,但对AOA没有影响。培养第3、30和90天,Urea + DMPP处理土壤中的AOB数量显著低于Urea + DCD处理的30%、56%和60%。对于反硝化细菌来说,所有处理中的nirK基因拷贝数均显著高于nirS基因拷贝数。添加DMPP在培养第3和30天显著减少了含nirK和nirS基因的反硝化细菌数量,而添加DCD对两类反硝化细菌数量无明显作用。【结论】东北黑土水稻生产中,硝化抑制剂DMPP降低N2O排放量和排放系数的效果显著好于DCD,因为DMPP在培养后的30天内,可以显著抑制氨氧化细菌繁衍,降低反硝化细菌数量,从而起到减少N2O排放、提高肥料利用率的作用。  相似文献

Nitrification is essential to the nitrogen cycle in paddy soils. However, it is still not clear which group of ammonia-oxidizing microorganisms plays more important roles in nitrification in the paddy soils. The changes in the abundance and composition of ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB) were investigated by real-time PCR, terminal restriction fragment length polymorphism, and clone library approaches in an acid red paddy soil subjected to long-term fertilization treatments, including treatment without fertilizers (CT); chemical fertilizer nitrogen (N); N and potassium (NK); N and phosphorus (NP); N, P, and K (NPK); and NPK plus recycled crop residues (NPK+C). The AOA population size in NPK+C was higher than those in CT, while minor changes in AOB population sizes were detected among the treatments. There were also some changes in AOA community composition responding to different fertilization treatments. Still few differences were detected in AOB community composition among the treatments. Phylogenetic analysis showed that the AOA sequences fell into two main clusters: cluster A and cluster soil/sediment. The AOB composition in this paddy soil was dominated by Nitrosospira cluster 12. These results suggested that the AOA were more sensitive than AOB to different fertilization treatments in the acid red paddy soil.  相似文献

Purpose

Microbial nitrification plays an important role in nitrogen cycling in ecosystems. Nitrification is performed by ammonia-oxidizing archaea (AOA), ammonia-oxidizing bacteria (AOB), and nitrite-oxidizing bacteria (NOB) including complete ammonia oxidizers. However, the relative importance of nitrifiers in autotrophic nitrification in relation to soil pH is still unclear.

Materials and methods

Combining DNA-based stable isotope probing (SIP) and molecular biological techniques, we investigated the abundance, structure, and activity of AOA, AOB, and NOB along a pH-gradient (3.97–7.04) in a vegetable cropped soil.

Results and discussion

We found that AOA abundance outnumbered AOB abundance and had a significantly negative relationship with soil pH. The abundances of NOB Nitrospira 16S rRNA, nxrB gene, and Nitrobacter nxrA gene were affected by soil pH. Incubation of soil with 13CO2 and DNA-SIP analysis demonstrated that significant 13CO2 assimilation by AOA rather than by AOB occurred in the acidic soils, whereas the labeled 13C level of AOA was much less in the neutral soil than in the acidic soils. There was no evidence of 13CO2 assimilation by NOB except for Nitrobacter with NxrB gene at pH 3.97. Phylogenetic analysis of AOA amoA gene in the 13C- and 12C-labeled treatments showed that the active AOA mainly belonged to Nitrososphaera in the acidic soils.

Conclusions

These results suggested that the main performer of nitrification was AOA in the acidic soils, but both AOA and AOB participated in nitrification in the neutral soil with low nitrification activity. NOB Nitrospira and Nitrobacter did not grow in the soils with pH 4.82–7.04 and other populations of NOB were probably involved in nitrite oxidation in the vegetable cropped soil.

Taking two important agricultural soils with different pH, brown soil (Hap-Udic Luvisol) and cinnamon soil (Hap-Ustic Luvisol), from Northeast China, a pot culture experiment with spring maize (Zea mays L.) was conducted to study the dynamic changes in the abundance and diversity of soil ammonia-oxidizing bacteria (AOB) and ammonia-oxidizing archaea (AOA) populations during maize growth period in response to the additions of nitrification inhibitors dicyandiamide (DCD) and 3,4-dimethylpyrazole phosphate (DMPP) by the methods of real-time polymerase chain reaction (PCR) assay, PCR-denaturing gradient gel electrophoresis (DGGE), and construction of clone library targeting the amoA gene. Four treatments were established, i.e., no urea (control), urea, urea plus DCD, and urea plus DMPP. Both DCD and DMPP inhibited growth of AOB significantly, compared to applying urea alone. Soil bacterial amoA gene copies had a significant positive linear correlation with soil nitrate content, but soil archaeal amoA gene copies did not. In both soils, all AOB sequences fell within Nitrosospira or Nitrosospira-like groups, and all AOA sequences belonged to group 1.1b crenaxchaea. With the application of DCD or DMPP, community composition of AOB and AOA in the two soils had less change except that the AOB community composition in Hap-Udic Luvisol changed at the last two growth stages of maize under the application of DCD. AOB rather than AOA likely dominated soil ammonia oxidation in these two agricultural soils.  相似文献

Disparities in the substrate affinity and tolerance threshold for ammonia have been believed to play a key role in driving niche differentiation between ammonia-oxidizing archaea (AOA) and bacteria (AOB); however, recent surveys argue that direct competition between AOA and AOB is also important in this phenomenon. Accordingly, it is reasonable to predict that diverse AOA lineages would grow in ammonium (NH4+)-rich alkaline arable soils if AOB growth is suppressed. To test this hypothesis, a microcosm study was established using three different types of alkaline arable soils, in which a high NH4+ concentration (200 μg N g-1 dry soil) was maintained by routinely replenishing urea and the activities of AOB were selectively inhibited by 1-octyne or 3,4-dimethylpyrazole phosphate (DMPP). Compared with amendment with urea alone, 1-octyne partially retarded AOB growth, while DMPP completely inhibited AOB. Both inhibitors accelerated the growth of AOA, with significantly higher ratios of abundance of AOA to AOB observed with DMPP amendment across soils. Nonmetric multidimensional scaling analysis (NMDS) indicated that different treatments significantly altered the community structures of both AOA and AOB and AOA OTUs enriched by high-NH4+ amendment were taxonomically constrained across the soils tested and closely related to Nitrososphaera viennensis EN76 and N. garnensis. Given that these representative strains have been demonstrated to be sensitive to high ammonia concentrations, our results suggest that it is the competitiveness for ammonia, rather than disparities in substrate affinity and tolerance threshold for ammonia, that drives niche differentiation between these phylotypes and AOB in NH4+-rich alkaline soils.  相似文献

采用厌氧培养-乙炔抑制法测定了4种不同利用方式红壤的反硝化势和气态产物N2O和N2的排放速率。结果表明,不同利用方式红壤反硝化势和N2O和N2的排放速率差异明显,土壤反硝化势强弱顺序依次为:竹林>茶园>林地>旱地。反硝化势与土壤有机碳(P<0.05)、厌氧培养期间土壤CO2累积排放量(P<0.01)、nirS基因丰度( P<0.05)和nirK基因丰度(P<0.05) 呈显著正相关关系。逐步回归分析结果表明,CO2累积排放量表征的易矿化碳是造成不同利用方式红壤反硝化势差异的主要原因,可以解释反硝化势变化的66%(P<0.01)。不同利用方式红壤N2O和N2排放速率差异明显,旱地红壤N2O和N2排放速率均最低,表明土壤pH的提升并没有增加旱地红壤的反硝化损失风险和N2O排放速率。土壤易矿化有机碳含量也是影响不同利用方式红壤N2O和N2排放速率的主要因素。反硝化功能基因nirS、nirK和nosZ的丰度均与CO2累积排放量呈显著正相关关系,进一步支持了土壤易矿化有机碳含量是影响不同利用方式红壤反硝化势和气态产物排放的主要因子。土壤pH是影响不同利用方式红壤反硝化气态产物N2/N2O的主要因素,但是pH影响红壤N2/N2O的微生物机制仍需要进一步研究。  相似文献

Biochar is an efficacious amendment for mitigating nitrous oxide (N2O) emissions in soils. Nevertheless, the underlying mechanisms responsible for reduced N2O emissions by biochar in paddy soils remain inadequately elucidated. Here, using two typical paddy soils with contrasting pH values (5.40 and 7.56), the N2 and N2O fluxes and the associated functional genes were investigated in soil amended with varying amounts of biochar (0%, 0.5%, and 5%, weight/weight) via soil slurry incubation integrated with the N2/Ar technique and qPCR analysis. The results showed that N2O fluxes were significantly (p < 0.05) reduced by 0.65–3.64 times following biochar amendment, concomitant with a significant (p < 0.05) increase in N2 fluxes (5.47–46.14%) in both acidic and alkaline paddy soils. As a result, the N2O/(N2O + N2) ratios were significantly (p < 0.05) reduced by 1.53–4.65 fold in both soil types. In acidic paddy soils, the enhanced denitrification rates and the decreased N2O/(N2O + N2) ratios exhibited a strong correlation with increased pH values. In alkaline paddy soil, these changes were ascribed to the enhanced nosZ Clade I gene abundance and nosZ/(nirS + nirK) ratio. Our findings reveal that biochar primarily mitigates N2O emissions in paddy soils by promoting its reduction to N2.  相似文献

Biochar can reduce N2O emissions and it can be added to the soil once, whereas fertilizers are often applied every cultivation season. The aging of biochar in soil affects its functioning but it is unclear whether palm shell biochar (PSB) could still mitigate N2O emissions even when additional basal N fertilizers are applied 1 year after the initial biochar application. We studied the impact of fresh and aged PSB (0%, 6%, 12%, and 18% w/w of dry soil) on N2O emissions, soil properties, nutrient content and yield of Komatsuna (Brassica rapa var. perviridis) under sandy soil conditions. The aged PSB non-significantly reduced N2O emissions but significantly offset soil acidification, and maintained a high soil nutrient status. Biochar application with fertilizer significantly increased plant tissue K and Ca content but decreased N, P and Mg content compared to the treatments without biochar. At higher application rates, biochar had negative effects on crop yield but as it aged, the negative effects were offset as a result of the similar variation in plant N uptake. Since seasonal N fertilizer application seems to be inevitable in Komatsuna cultivation, addition of biochar could be a possible way of counteracting the effects of excessive fertilizer use. Further research is needed to assess the feasible biochar application rates for Komatsuna fields in various soil types under field conditions.  相似文献

Purpose

Nitrous oxide (N2O) is a potent greenhouse gas which is mainly produced from agricultural soils through the processes of nitrification and denitrification. Although denitrification is usually the major process responsible for N2O emissions, N2O production from nitrification can increase under some soil conditions. Soil pH can affect N2O emissions by altering N transformations and microbial communities. Bacterial (AOB) and archaeal (AOA) ammonia oxidisers are important for N2O production as they carry out the rate-limiting step of the nitrification process.

Material and methods

A field study was conducted to investigate the effect of soil pH changes on N2O emissions, AOB and AOA community abundance, and the efficacy of a nitrification inhibitor, dicyandiamide (DCD), at reducing N2O emissions from animal urine applied to soil. The effect of three pH treatments, namely alkaline treatment (CaO/NaOH), acid treatment (HCl) and native (water) and four urine and DCD treatments as control (no urine or DCD), urine-only, DCD-only and urine + DCD were assessed in terms of their effect on N2O emissions and ammonia oxidiser community growth.

Results and discussion

Results showed that total N2O emissions were increased when the soil was acidified by the acid treatment. This was probably due to incomplete denitrification caused by the inhibition of the assembly of the N2O reductase enzyme under acidic conditions. AOB population abundance increased when the pH was increased in the alkaline treatment, particularly when animal urine was applied. In contrast, AOA grew in the acid treatment, once the initial inhibitory effect of the urine had subsided. The addition of DCD decreased total N2O emissions significantly in the acid treatment and decreased peak N2O emissions in all pH treatments. DCD also inhibited AOB growth in both the alkaline and native pH treatments and inhibited AOA growth in the acid treatment.

Conclusions

These results show that N2O emissions increase when soil pH decreases. AOB and AOA prefer different soil pH environments to grow: AOB growth is favoured in an alkaline pH and AOA growth favoured in more acidic soils. DCD was effective in inhibiting AOB and AOA when they were actively growing under the different soil pH conditions.  相似文献

Studies about ammonia-oxidizing bacteria (AOB) and archaea (AOA) are often focused on topsoil, but little is known about their activity and distribution in subsoil. A long-term fertilizer experiment was conducted to assess the effects of different fertilizer treatments on AOB and AOA in vertical soil profiles of paddy soil plots that received no nitrogen fertilizer control (CK), NPK chemical fertilizers (CF), organic–inorganic mixed fertilizer (OIMF) and organic fertilizer (OF). Soil properties, potential nitrification rate (PNR) and amoA gene abundance of AOB and AOA were measured and analyzed by two-way ANOVA and correlation analysis. Quantitative PCR analysis of amoA genes showed that AOA were more abundant than AOB in all the soil samples. AOB declined sharply with soil depth. Compared with CK and OF treatments, CF and OIMF treatments had higher abundance of AOB throughout the soil profiles. However, AOA tend less responsive to soil depth and fertilizers compared to AOB. This caused the AOA/AOB ratios in subsoil higher than in topsoil, and in CK and OF higher than in CF and OIMF treatments. These results suggest that AOA are more abundant and can be better adapted to nutrient-poor subsoils than AOB, and autotrophic nitrification could likely be determined by a complex suite of environmental factors in vertical profiles of the paddy soil tested.  相似文献
