Mesembryanthemum edule L. (sourfig, Aizoaceae) has long been used as food and in traditional medicine. This study was intended to characterize the antioxidant properties and the phenolic compounds of M. edule leaf, stem and root. The approach consisted to evaluate these organs for their antioxidant activities through several in vitro tests, to determine tissue contents in total phenolics, flavonoids and proanthocyanidins and to establish the phenolic composition through RP-HPLC analysis. All studied organs showed a high antioxidant activity as compared to positive control BHT, with maximal efficiency for stems followed by leaves and roots. The highest polyphenolic levels were found in stems and leaves (86.5 and 68.7 mg GAE g−1 DW, respectively), suggesting that their strong antioxidant activity could be attributed to these phytochemicals. The HPLC analysis revealed that the main phenolic compounds were quercitrin and avicularin (1.4 and 1.15 mg g−1 DW, respectively) in the leaves, while catechin and procyanidin B2 (1.66 and 1.54 mg g−1 DW, respectively) were the most abundant phenolics in the stems. Overall, the strong antioxidant activity and richness of M. edule aerial tissues suggest that it could be advantageously used as a functional or nutraceutical food, to prevent or moderate oxidative stress-related diseases.  相似文献

Withania somnifera (L.) Dunal. (Indian ginseng) is an important medicinal plant which yields pharmaceutically active compounds called withanolides. The present work deals with optimization of parameters of hairy root culture of W. somnifera for the production of biomass and withanolide A. We also investigated the effects of carbon source [sucrose, glucose, fructose, maltose, glucose + fructose (1:1), fructose + sucrose (1:1) and sucrose + glucose (1:1)], sucrose concentration (1%, 2%, 3%, 4%, 6% and 8%) and the initial medium pH (4.0, 4.5, 5.0, 5.5, 5.8, 6.0 and 6.5) on growth and production of withanolide A in hairy root cultures of W. somnifera. We found that biomass accumulation and production of withanolide A was highest when sucrose was used as the carbon source (11.92 g l−1 DW and 11.96 mg g−1 DW of withanolide A). Further 3% sucrose concentration was found to be optimal for biomass accumulation (11.92 g l−1 DW) and 4% sucrose favoured the production of withanolide A (13.28 mg g−1 DW) in the tested range of concentrations (1-8%). The biomass of hairy roots was optimal when the initial medium pH was 5.8 (12.1 g l−1 DW) and the withanolide A production was highest in the medium pH set at 6.0 (13.84 mg g−1 DW).  相似文献

Castilleja tenuiflora is a highly valued medicinal plant that grows in pine-oak woods in Mexico. In this study, we identified for the first time verbascoside and isoverbascoside as the major phenylethanoid glycosides (PhGs) in C. tenuiflora. These compounds have proven biological activities, including anti-inflammatory, antioxidant, and cytotoxic activities, which may be related to the traditional uses of this plant. We developed a reverse-phase high-performance liquid chromatography (RP-HPLC) procedure to analyze PhGs, and determined their concentrations in various different tissues of wild plants. Verbascoside accumulated mainly in roots and inflorescences (9.23 and 7.88 mg g−1 dry biomass, respectively), while isoverbascoside accumulated mainly in the roots (7.13 mg g−1 dry biomass). To provide an alternative source of material for production of bioactive compounds, we established in vitro adventitious root cultures in which roots were grown in B5 medium containing either 10 μM indole 3-acetic acid (IAA) or 10 μM α-naphthaleneacetic acid (NAA). The greatest dry biomass yield (30 g L−1) was achieved at 30 days after transfer of roots into IAA-containing medium. The highest specific yields of PhGs were also obtained using this auxin; the maximum level of verbascoside was 14.62 mg g−1 dry root biomass (438.6 mg L−1) at 30 days after root transfer, and the maximum yield of isoverbascoside was 37.32 mg g−1 dry root biomass (522.48 mg L−1) at 23 days after root transfer. Adventitious root cultures of C. tenuiflora are a promising system for further studies on scale-up and phenylethanoid glycosides biosynthesis.  相似文献

Gymnema sylvestre is an important medicinal plant which bears bioactive compound namely gymnemic acids. The present work deals with optimization of cell suspension culture system of G. sylvestre for the production of biomass and gymnemic acid and we investigated effects of macro elements (NH4NO3, KNO3, CaCl2, MgSO4 and KH2PO4 - 0.0, 0.5, 1.0, 1.5 and 2.0× strength) and nitrogen source [NH4+/NO3 ratio of: 0.00/18.80, 7.19/18.80, 14.38/18.80, 21.57/18.80, 28.75/18.80, 14.38/0.00, 14.38/9.40, 14.38/18.80, 14.38/28.20 and 14.38/37.60 (mM)] of Murashige and Skoog medium on accumulation of biomass and gymnemic acid content. The highest accumulation of biomass (165.00 g l−1 FW and 15.42 g l−1 DW) was recorded in the medium with 0.5× concentration of NH4NO3 and the highest production of gymnemic acid content was recorded in the medium with 2.0× KH2PO4 (11.32 mg g−1 DW). The NH4+/NO3 ratio also influenced cell growth and gymnemic acid production; both parameters were greater when the NO3 concentration was higher than that of NH4+. Maximum biomass growth (159.72 g l−1 of FW and 14.95 g l−1 of DW) was achieved at an NH4+/NO3 ratio of 7.19/18.80, and gymnemic acid production was also greatest at the same concentration of NH4+/NO3 ratio (11.35 mg g−1 DW).  相似文献

In the present investigation, seed methanolic extracts of three Vitis vinifera (Muscat d’Italie, Syrah and Carignan) were assayed for their antioxidant activities. Results showed that there are strong variations in the contents of total phenols (440.97–121.94 mg GAE g−1 DW), flavonoids (48.07–16.81 mg EC g−1 DW) and tanins (37.15–14.9 mg EC g−1 DW) from the studied seeds. The phenolic composition of these extracts was determined by RP–HPLC after acid hydrolysis. The main phenolic compound was quercetin with 27.2% in Muscat d’Italie, 48.8% in Syrah and 28.4% in Carignan. Besides, all seed extracts showed remarkable DPPH radical scavenging activity with IC50 values ranged from 1.8 to 30 μg ml−1. EC50 values of reducing power activity ranged from 100 to 120 μg ml−1. The high phenolic content and the considerable antioxidant activities of vitis seed extracts could potentially be considered as an expensive source of natural antioxidants.  相似文献

The demand for food is increasing, not only to meet food security for growing populations, but also to provide more nutritious food, rich in good quality proteins and nutraceutical compounds. The amaranth (Amaranthus hypochondriacus) plant, in addition to its high nutritive and nutraceutical characteristics, has excellent agronomic features. The objective of the present study was to analyze some physical and proximal-nutritional properties of amaranth seeds obtained from different varieties grown in arid zones and characterize their phenolic acids and flavonoids. Two commercial (Tulyehualco and Nutrisol) and two new (DGETA and Gabriela) varieties of A. hypochondriacus were grown at the Mexican Highlands zone. Tulyehualco and DGETA varieties had higher seed yield of 1475 and 1422 kg ha−1, respectively, comparable to corn and soybean production in agricultural areas. Gabriela had the highest protein content of 17.3%, but all varieties had an adequate balance of essential amino acids. Polyphenols as rutin (4.0–10.2 μg g−1 flour) and nicotiflorin (7.2–4.8 μg g−1 flour) were detected. Amaranth can be cultivated in arid zones where commercial crops cannot be grown; the seeds besides their well known nutritive characteristics could be a source of phenolic compounds of high antioxidant properties.  相似文献

Changes in the contents and composition of polyphenolics and resulting antioxidant activities of S. verbenaca by-products were investigated at three phenological stages (flowering, early fruiting and late fruiting stages). The highest accumulation of total phenolics was detected at the flowering stage (58.36 mg GAE/g DW). HPLC analysis of methanolic extracts showed the prevalence of methyl carnosate (821.45–919.82 μg/g DW) and rosmarinic acid (544.51–649.26 μg/g DW). Phenolic diterpenes (1056.90–1148.42 μg/g DW) was the most represented class of compounds. Three complementary tests namely, DPPH? (IC50 value, 49.22 μg/mL) and ABTS?+-radical scavenging assays (146.86 μM TE/mg) and FRAP reducing power test (188.93 mM Fe(II)/mg) were used to evaluate the antioxidant capacity and showed the best performance at the early fruiting period. The current study evidenced the significant effect of phenophase on antioxidants and contributed to valorize S. verbenaca extracts as a source of functional phenolic compounds.

In this study, Nigella sativa L. oil was extracted using supercritical carbon dioxide with full factorial design to determine the best extraction condition (pressure, temperature and dynamic extraction time) for obtaining an extract with high yield, antioxidant activity and thymoquinone (TQ) quantity. The maximum thymoquinone content in the highest overall yield was achieved through SC-CO2 extraction condition of 150 bar, 40 °C, 120 min with the value of 4.09 mg/ml. The highest SC-CO2 extraction yield was 23.20% which obtained through extraction condition of 350 bar, 60 °C and 120 min. The extraction conducted at 350 bar, 50 °C, 60 min showed the lowest IC50 value (highest antioxidant activity) of 2.59 mg/ml using DPPH radical scavenging activity method. Fatty acid composition of the extracted oil with highest radical scavenging activity was obtained by gas chromatographic analysis.  相似文献

The study deals with evaluation of antifungal and antiaflatoxigenic Caesulia axillaris Roxb. essential oil (EO) against herbal raw materials deteriorating fungi and its free radical scavenging activity. During mycoflora analysis these herbal raw materials were found to be severely contaminated by different fungi and aflatoxins. A total of nine different fungal species were isolated from three herbal raw materials. Aspergillus flavus LHPtc was recorded as the highest aflatoxin B1 producing strain. EOs of some plants were tested for their fungitoxicity against the toxigenic strain A. flavus LHPtc, and C. axillaris EO was found as potent fungitoxicant. C. axillaris EO was chemically characterized through GC-MS analysis which depicted the presence of 18 compounds, dl-limonene and Euasarone being the major components. The EO exhibited broad spectrum of fungitoxicity against fungi causing postharvest deterioration of herbal raw materials. At 1.0 μl ml−1 the oil showed complete inhibition of fungal growth and aflatoxin B1 production was inhibited at 0.8 μl ml−1. Free radical scavenging activity of the oil was also recorded by 2,2-diphenyl-1-picrylhydrazyl assay, and its IC50 value was found 18 μl ml−1. The safety limit of the EO was determined in terms of LD50 on mice, which was 9166.6 μl kg−1, suggesting its non mammalian toxicity. The EO of C. axillaris may be recommended as a plant based preservative in enhancement of shelf life of herbal raw materials by preventing their lipid peroxidation as well as biodeterioration due to fungal and aflatoxin contamination.  相似文献

CIMMYT hexaploid spring wheat (Triticum aestivum L.) germplasm has played a global role in assisting wheat improvement. This study evaluated four classes of CIMMYT germplasm (encompassing a total of 273 lines), along with 15 Australian cultivars (Oz lines) for grain yield, yield components and physiological traits in up to 27 environments in Australia's north-eastern region, where terminal drought frequently reduces grain yield and grain size.Broadly-adapted CIMMYT germplasm selected for grain yield had greater yield potential and improved performance under drought stress, being up to 5% greater yielding in High-yielding (mean yield 429 g m−2) and 4-10% greater yielding than adapted Oz lines in Low-yielding environments (mean yield 185 g m−2). Whilst maintaining statistically similar harvest index and spikes m−2 compared to broadly-adapted Oz lines across all environments, sets of selected CIMMYT lines had greater canopy temperature depression (0.18-0.27 °C), dry weight stem−1 (0.20-0.37 g), increased grains spike−1 (0.8-3.4 grains), grain number m−2 (ca. 20-800 grains), and maturity biomass (56-83 g m−2). Compared to selected Oz lines, broadly-adapted CIMMYT lines had a smaller reduction in Low compared to High-yielding environments for these traits, especially dry weight stem−1, such that CIMMYT lines had ca. 25% and 10% greater dry weight stem−1 than the Oz lines in Low- and High-yielding environment groups, respectively. Broadly-adapted CIMMYT germplasm also had slightly higher stem water soluble carbohydrate concentration at anthesis (ca. 6 mg g−1), which contributed to their higher grain weight (ca. 0.5 mg grain−1), and maintained an agronomically appropriate time to anthesis and plant height. Thus current CIMMYT germplasm should be useful donor sources of traits to enrich breeding programs targeting variable production environments where there is a high probability of water deficit during grain filling. However, as multiple traits were important, efficient introgression of these traits in breeding programs will be complex.  相似文献

In separate experiments conducted in 2007 and 2008, growth and accumulation of selected caffeic acid derivatives (CADs; i.e., caftaric acid, chlorogenic acid, echinacoside, caffeic acid, cynarin, p-coumaric acid, ferulic acid and cichoric acid) were determined in Echinacea angustifolia DC. var. angustifolia seedlings grown in hydroponic culture (floating raft system) at a density of 122 plant m−2 (at planting). Plants were harvested 11 (2007) or 16 (2008) weeks after transplanting (i.e., 15 or 20 weeks after sowing). In both years, plants grew vigorously and at harvest approximately half of the plants under observation had developed one to three inflorescences. In 2008, the root yield (2940 kg ha−1) harvested in nearly eight months from two consecutive hydroponic cultures was within the yield reported in the literature for field cultivations lasting two to four years. None of the selected CADs was found in the leaves, while the inflorescences (stem and capitulum) contained only caftaric acid and echinacoside at concentrations higher than the detection limits (0.05 mg g−1 dry weight). Echinacoside, cynarin and chlorogenic acid were found in root tissues at concentrations ranging from 0.36 to 5.25 mg g−1 dry weight. In all plant samples, echinacoside, which is the marker compound for E. angustifolia material, did not reach the minimum quality standard (10 mg g−1 dry weight) for the production of standardized extract. We concluded that short-cycle, high-density greenhouse hydroponic culture stimulates plant growth and root production in E. angustifolia, but it does not ensure sufficient CADs accumulation in dried roots.  相似文献

Field and laboratory experiments were conducted to evaluate the productivity and essential oil composition of lavender (Lavandula angustifolia Mill.) and hyssop (Hyssopus officinalis L.) as functions of year, harvest time, and drying. Lavender essential oil content ranged from 0.71 to 1.3% (overall average of 0.89%) and hyssop oil content ranged from 0.13 to 0.26% (overall average of 0.19%). Lavender and hyssop essential oil yields increased with time. Hyssop oil yields varied from 7.3 kg ha−1 to 19.6 kg ha−1, and lavender oil yields varied from 7.8 kg ha−1 to 55.5 kg ha−1. The major constituents of lavender oil were linalool (23.3-43.4%) and linalylacetate (20.2-39.6%), while the major constituents of hyssop oil were pinocamphene + isopinocamphene (57-75%) and β-pinene (5-15%). Lavender oil extracted from dry material had higher concentrations of linalyl acetate and caryophyllene but lower concentrations of myrcene than the oil from the fresh material. Delayed harvest of hyssop increased the concentrations of β-pinene, myrcene, and limonene + cineole but reduced pinocamphone + isopinocamphone. The chemical composition of the lavender and hyssop oil produced in Mississippi was similar to commercial oils from Bulgaria, Canada, France, and US. Lavender and hyssop can be established as essential oil crops in areas of the southeastern United States. Lavender and hyssop essential oils did not show significant antimicrobial, antileishmanial, antimalarial activity, and did not alter ruminal fermentation. However, commercial oil from L. latifolia reduced methane production in an in vitro digestibility study. The antioxidant activity of hyssop essential oil was 2039 μmol of TE L−1, whereas the antioxidant activity of lavender essential oil was 328 μmol of TE L−1.  相似文献

Physic nut (Jatropha curcas L.) is a promising seed oil source for biodiesel production. Natural antioxidants play a major role in maintaining oxidative stability of oils and they also have important food and industrial applications. Among them, tocochromanols are the most abundant in seeds. The objective of this research was to evaluate the variation for tocochromanol content and profile in a germplasm collection of 52 accessions of J. curcas. Seeds collected in two different periods, August and November of 2009, were analysed for tocochromanol content. Additionally, the dynamics of tocochromanol accumulation in developing seeds was studied. Total seed tocochromanol content averaged 307.2 mg kg−1 in August and 303.7 mg kg−1 in November, whereas total oil tocochromanol content averaged 507.4 mg kg−1 in August and 500.8 mg kg−1 in November. The tocochromanol fraction was made up of 15.4% gamma-tocopherol, 83.8% gamma-tocotrienol, and 0.8% delta-tocotrienol in August and 18.0% gamma-tocopherol, 80.4% gamma-tocotrienol, and 1.6% delta-tocotrienol in November. Genotype × environment effects were identified for tocochromanol content but not for the proportion of major tocochromanol homologues, which showed a high positive correlation between both environments. Developing seeds contained primarily alpha-tocopherol and gamma-tocopherol at early stages of development, with gamma-tocotrienol and delta-tocotrienol being practically undetectable. Gamma-tocotrienol content remained practically undetectable till 66 DAP and then increased pronouncedly to final levels of 177.1 mg kg−1 (74.8% of the total tocochromanol content). The powerful antioxidant and health-promoting properties of gamma-tocotrienol encourages further studies on selection for the tocopherol/tocotrienol ratio in Jatropha and on the potential of tocochromanols as high added-value products derived from Jatropha seed oil production.  相似文献

Interest in the consumption of fresh fruits is, to a large extent, due to its content of bioactive nutrients and their importance as dietary antioxidants. There is a growing demand for delivery of antioxidants through functional foods with the related challenge of protecting their bioactivity during food processing and subsequent passage through the gastrointestinal tract. This study focuses on the evaluation of concentration of bioactive compounds in black currant berries (Ribes nigrum L.) and retention of black currant polyphenol compounds and their antioxidant activity after microencapsulation by spray-drying. The polyphenol contents and antioxidant activities were assessed for 5 black currant cultivars grown in Canada in fresh and stored fruits at −20 °C for 9 months. The Ben Alder cultivar was the richest in anthocyanins and flavonols content with high antioxidant potential measured with ABTS and DPPH radicals (4.5 and 2.4 mM/100 g). Black currant polyphenols were stable during 9 month of storage at −20 °C. The results of this study indicated that in black currants storage did not affect negatively the antioxidant capacity. The good stability of bioactive compounds of black currants during freezer storage allows prolonging the storage time of these berries. Maltodextrins with different DE (dextrose equivalent): DE11, DE18, DE21 and inulin were selected as wall material. The stability of encapsulated polyphenols during 12 months storage at 8 °C and 25 °C was evaluated. Compared to maltodextrins DE18 and DE21, maltodextrin DE11 had not only higher drying yield but also offered better protection for phenolics during storage. Radical scavenging activity studies demonstrated significant antioxidant activity of microencapsulated powders before and after storage. The black currant polyphenol microcapsules described in this study represent a promising food additive for incorporation into functional foods due to antioxidant content.  相似文献

Differentiated tissue in Panax ginseng cultures was found to be very efficacious for saponin production. In order to increase the yield of saponins and preserve culture stability we were testing different plant growth regulators (PGR) and auxin/cytokinin combinations to regulate a level of tissue differentiation. For this purpose we used transverse thin cell layers (tTCLs) of adventitious roots of Panax ginseng. Adventitious roots were cultivated in Shenk and Hildebrand (SH) liquid medium supplemented with IBA (24.6 μM). Callus formation and root multiplication of adventitious root tTCLs was evaluated after 4 and following 12 weeks of cultivation, respectively, on SH basal medium containing various auxins (3 mg l−1) or cytokinins (0.2 or 0.02 mg l−1) or their combinations. We found that kinetin (Kin) in combination with auxin benzo[b]selenienyl acetic acid (BSAA), naphthalene acetic acid or indole-3-butric acidis the best for biomass production and following root multiplication. These combinations were tested in previously selected most suitable large-scale system—a temporary immersion system RITA. The best saponin production (15.94 ± 1.89 mg g−1 dry weight) and growth value (5.62 ± 0.34) was reached on medium containing BSAA and Kin combination.  相似文献

Jackfruit is an underutilized edible fruit in the tropics and subtropics. The aim of this study was to produce wine from jackfruit pulp and to evaluate the total phenolic and flavonoid contents and antioxidant properties of the wine. The ability of scavenging free radicals was measured using 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant assay (FRAP), N, N-dimethyl-p-phenylendiamine (DMPD) and nitric oxide (NO) scavenging assays. Experimental results indicated that jackfruit wine was effective in DPPH radical scavenging (69.44 ± 0.34%), FRAP (0.358 optical density value, O.D.), DMPD (78.45 ± 0.05%) and NO (62.46 ± 0.45%) capacity. By the analysis of the high performance liquid chromatography coupled to diode array detector (HPLC-DAD), two phenolic compounds namely gallic acid and protocatechuic acid were identified. The jackfruit wine was also able to protect H2O2 + UV radiation and γ-radiation (100 Gy) induced DNA damage in pBR322 plasmid DNA. The antioxidant and DNA damage protecting properties of jackfruit wine confirmed health benefits when consumed and could become a valuable source of antioxidant rich neutraceuticals. Additionally, the wine could be a commercially valuable by-product for the jackfruit growers.  相似文献

The essential oil content of the oil-bearing rose (Rosa damascene Mill.) is relatively low, around 0.3-0.4 mL kg−1 in fresh flowers. There is a need to increase essential oil yield of oil-bearing rose. The objective was to examine the effect of Tween 20 (polyoxyethylene sorbitan monolaurate) applied with, or without, maceration of flowers on oil content and composition of oil-bearing rose harvested at beginning of flowering, full bloom, and end of flowering. Addition of Tween 20 at 1000 mL L−1 and 2500 mL L−1 increased essential oil yield by 26% (to 0.44 mL kg−1) and 54% (to 0.54 mL kg−1) respectively relative to the untreated control that gave 0.35 mL kg−1 yield. Maceration, in combination with the addition of Tween 20 at 1000 mL L−1 and Tween 20 at 2500 mL L−1, increased oil yield by 69% (to 0.59 mL kg−1) and 94% (to 0.68 mL kg−1) respectively. Among the three phenological phases of harvest, harvesting at the beginning of flowering gave the highest yield followed by the full bloom and then by the end of flowering phases. Since the interaction effect was not significant, the differences obtained among the treatments were regardless of the phase, and vice versa. Treatments did not significantly alter composition of the essential oil. Postharvest pre-extraction application of Tween 20 in combination with maceration could be used in the rose industry for increasing the essential oil yield.  相似文献

The present work is designed to evaluate the bioactive properties of the crude methanolic extract of Jatropha curcas oil and its solvent fractions. The crude methanolic extract obtained was fractionated using a hydrophilic lipophilic balanced (HLB) cartridge and then eluted with different solvents in the order of hexane (F1), dichloromethane (F2), chloroform (F3), ethyl acetate (F4) and methanol (F5), respectively. Total phenolic content of the crude methanolic extract and its fractions was in the range of 0.19-4.5 mg/g as gallic acid equivalent. Antioxidant activity of the crude methanolic extract and its fractions were determined by two complementary test methods, namely, phenanthroline method and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging method. All samples demonstrated weak antioxidant activity (150-851 μmol Fe/100 g of the extract and IC50 of 1.05-13.5 mg/mL). When compared to butylated hydroxytoluene (BHT), a reference synthetic antioxidant, both showed weaker antioxidative potential. The evaluation of antimicrobial activity of the extracts was performed using a disc diffusion method and a micro-well dilution method against six economic plant disease bacteria. The results showed that all extracts possessed strong to moderate antibacterial activity with varying degrees of growth inhibition against the test bacteria. The minimum inhibitory concentrations (MIC) were in the range of 14.92-428.6 μg/mL. In addition, the chemical constituents in each fraction of the extract were subjected to analyze by gas chromatography-mass spectrometry (GC-MS). The eleven constituents were identified. Among them, 2,4-di-tert-butylphenol, methyl 3-(3,5-di-tert-butyl-4-hydroxyphenyl)propionate and linoleic acid may be the main cause of its strong antibacterial activity. Therefore, this oil present in the methanolic extract had great potential as effective antibacterial sources.  相似文献

Heterogeneous crop stands require locally adapted nitrogen fertilizer application based on rapid and precise measurements of the local crop nitrogen status. In the present study, we validated a promising technique for the latter, namely a tractor-mounted field spectrometer with an oblique quadrilateral-view measuring optic, measuring solar radiation and canopy reflectance in four directions simultaneously. Dry matter yield (kg ha−1), total N content (g N g−1 dry matter) and total aerial N (aboveground N-uptake) (kg N ha−1) in maize were determined in 10 m2 calibration areas in 60 plots differing in their N treatment and seeding density three times in each of three years under field conditions. Results show that the sensor used can reliably determine total aerial N ranging from as little as 5 kg N to 150 kg N ha−1 with R2-values ≥0.81 in 2002 and 2004, and with R2-values ranging from ≥0.57 to 0.84 in 2003. Dry matter yields from as low as 0.3–4.2 t ha−1 could be determined with R2-values ranging from 0.67 to 0.91 in 2002 to 2004. The capacity to ascertain DM yield spectrally was drastically reduced in the higher yield range (>6 t ha−1) probably due to decreased sensitivity of the spectral signal. N-contents were generally not well determined. Taken together there is a good potential to determine reliably differences in total aerial N or DM yield from the five leaf stages unfolded to the five node stage where typically nitrogen applications are carried out.  相似文献

The study revealed, for the first time, accumulation of spilanthol, an antiseptic alkylamide, in in vitro cultures of Spilanthes acmella Murr., a medicinal plant of immense commercial value. To achieve this, in vitro shoots were regenerated via direct organogenesis from leaf-disc explants of Spilanthes. Shoots were induced in the presence of N6-benzylaminopurine (BAP) alone or in combination with either α-naphthalene acetic acid (NAA) or Indole-3-acetic acid (IAA) in Murashige and Skoog medium. The best treatment for shoot regeneration was MS + BAP (5.0 μM) + IAA (5.0 μM), which promoted adventitious shoot proliferation in >82% cultures with an average of 5.3 shoots per explant. Regenerated shoots rooted spontaneously with a frequency of 100% on half strength MS medium (major salts reduced to half strength) containing 50 g l−1 sucrose. The plantlets were acclimatized successfully with 90% survival rate. Additionally, ploidy stability of the regenerated plants was assessed by flow cytometry which showed that all investigated plants had the similar ploidy as that of the mother plant. For spilanthol identification, peaks eluted from HPLC were analyzed by mass spectrometry with its characteristic fragmentation pattern. For quantification studies, calibration curve was generated, which revealed a higher amount of spilanthol content (3294.36 ± 12.4 μg/g DW) in the leaves of in vitro plants compare to those of in vivo plants (2703.66 ± 9.6 μg/g DW of spilanthol). An efficient multiplication frequency, ploidy stability and enhanced spilanthol accumulation ensure the efficacy of the protocol developed for this industrially important medicinal plant.  相似文献
