Summary The intercellular volume within tuber tissue increases with tuber maturation. At the same time, both tuber injury after mechanical load and lentical perforation after water uptake by tubers in wet peat decreases. The degree of tuber injury is correlated with intercellular space (r=−0.667,n=16) and with water saturation of tissue (r=0.697,n=16). The tubers lose water during storage. In wet soil wilted tubers take up significantly higher amounts of water than turgid tubers, and lenticel perforation is much more frequent. Tuber maturation in autumn and low water loss during storage enhance the cushioning effect of the intercellular gas volume of tuber tissue against outside mechanical load and internal tension after water uptake.

Summary In a glasshouse experiment three potato cultivars were grown to maturity in wet or dry compost. The number of lenticels per tuber was greater with larger tubers but was unaffected by the cultivar or soil moisture regime. The frequency of lenticel penetration by a water soluble stain, safranin O, remained high as tubers developed to maturity in wet soil. In dry soil penetration declined markedly as the tubers matured. When tubers, harvested just before the onset of haulm senescence, were inoculated withErwinia carotovora ssp.atroseptica, the frequency of lenticel rotting was less than the rate of stain penetration, indicating that additional factors were involved in the bacterial infection process. Inoculating mature tubers with bacteria and incubating them in anaerobic conditions frequently resulted in rotting of the lenticels. Comparable tubers incubated in aerobic conditions showed no rotting or invasion by bacteria. The results are discussed in relation to the biological control of blackleg.

Summary In eight experiments in 4 years, inoculating seed tubers withR. solani caused stem canker and in seven experiments decreased the total yield and the yield of tubers 40–70 mm. Yields of larger tubers were usually increased. In most experiments inoculating increased the reducing sugar content of the tubers and darkened the colour of crisps, but did not consistently affect the amounts of sucrose. Treating soil with aldicarb or oxamyl before planting also slightly increased the total reducing sugars.

Summary Field-grown tubers of 22 progenies ofSolanum tuberosum L. generated in a crossing programme involving seven parents differing in resistance toFusarium coeruleum Lib. ex Sacc. andF. sulphureum Schlect. (=F. sambucinum Fuckel, teleomorphGibberella pulicaris (Fr.) Sacc.) were wound-inoculated with a cornmeal + sand culture of each pathogen. Parental genotypes were also included. The mean lesion size of each progeny was compared in 2 years of tests, as well as with published data on glasshouse-grown tubers. ForF. coeruleum there was a high correlation between years as well as with the glasshouse data, but no such correlations were apparent withG. pulicaris. Furthermore, parental and GCA values, as well as progeny means and mid-parent scores, also correlated highly forF. coeruleum but not forG. pulicaris. Glasshouse-grown tubers of 11 wildSolanum spp. were also inoculated with both pathogens. Some resistance to one or other, or both, was apparent, particularly inS. chacoense.

Summary In 1983 and 1984 there were reports in Norway of more severe late blight infection than usual on tubers of cv. Pimpernel, a cultivar without R-genes known to be somewhat resistant to both haulm and tuber infection. Results from routine tests confirmed earlier experience that cv. Pimpernel shows stable resistance to tuber late blight, although the ranking of cultivars may vary between years. In experiments during three harvest seasons tubers of different cultivars were inoculated with isolates ofP. infestans obtained from stocks of cv. Pimpernel with severe tuber late blight and with other isolates. In these experiments the isolates from cv. Pimpernel caused more rot on tubers of Pimpernel than did other isolates.

Summary Using the fluorescent lipid stain fluorol yellow 088 to detect suberin in tubers, the numbers of layers and the thickness of the suberized cells in the periderm and within the lenticels showed changes after the early stage of tuber formation. These changes as tubers matured were affected by cultivar and soil moisture conditions. Penetration of the surface tissues of tubers by the water soluble stain safranin O was confined to the lenticles. Although the zone of suberized cells in lenticels acted to some extent as a barrier to penetration, there was not a clear relationship between permeability and suberization. Stored tubers showed thicker suberin berriers in lenticels than freshly harvested tubers. When lenticels of stored tubers proliferated the suberin barrier was disrupted but a further thick suberin barrier formed after exposure to air for two weeks. The findings are discussed in relation to biological control ofErwinia by antagonistic bacteria.

Summary Variation in aggressiveness to tubers among isolates ofPhytophthora infestans sampled from three potato growing regions in the Netherlands was compared. Variation in the ability to infect tubers of cv. Bintje was found between isolates of each of the three regional populations. The most aggressive isolate of the old population matched the average level of the new population in its ability to infect tubers. As a consequence, the commonly used reference isolate VK 6C can no longer be considered to be representative of the present population ofP. infestans. Therefore it is recommended that testing tuber resistance for the official list of potato cultivars with this isolate should be discontinued. Tuber infection and subsequent spread of the fungus in the tuber tissues were not found to be correlated. The components of tuber pathogenicity studied were not correlated to pathogenicity factors in the foliage, as measured under growth chamber conditions.

Summary The effects of the leaves of five plant species, one from each of the generaAmbrosia, Anemone, Eupatorium, Eucalyptus andLantana, on potato tuber moth were investigated under indigenous storage conditions at the Central Potato Research Station, Shillong (1800 m above sea level). Their action was compared with that of a biological insecticide (spores ofBacillus thuringiensis), a chemical insecticide (carbaryl), and an untreated control. The data collected after six months storage on tuber damage, sprout damage and the rotting indicated that the leaves ofLantana aculeata provided most protection to the tubers, reducing damage from over 70% in the check to below 5%, and sprout damage from over 45% to below 3%. Next best wasEucalyptus globulus followed byB. thuringiensis. They may be used on tubers stored for table use or for seed as they had no adverse effect on germination or on the yield of a subsequent crop.

Summary Increasing doses of nitrogenous fertilizer increased storage rots in six potato cultivars of which cvs Kufri Sindhuri and Kufri Lalima rotted less and were found to contain high amounts of total phenols compared to the more susceptible cultivars Kufri Jyoti and Kufri Badshah. Nine phenolic acids were identified in the peel and the pulp of tubers and four of these and of other phenolic extracts from tubers suppressed the growth ofErwinia carotovora ssp.carotovora. Summary Erwinia carotovora in potato stocks can be detected more efficiently by the tuber incubation method in which the test tubers are first induced to rot under anaerobic conditions (followed by detection isolation of the bacteria on a selective growth medium or by serological techniques) than by direct lenticel sampling. However, the efficiency of detection is affected by both the extent of decay at sampling time and the incubation temperature. Detection ofE. carotovora is poor from extensive lesions developed at temperature of 25 C or above. Furthermore, detection ofE. carotovora var.atroseptica is more frequent than that ofE. carotovora var.carotovora at temperatures below 22 C in tubers contaminated by both organisms, whereas at higher temperatures the reverse is true. Only within a narrow temperature range, close to 22 C is there an equal chance of detecting both varieties.

Summary The method hitherto used in Norway for testing for tuber resistance to late blight, inoculating newly harvested unwounded tubers, is described. Superficial wounding of the tubers prior to inoculation increased the level of infection. Tests on wounded and unwounded tubers ranged the cultivars in the same order, but the significance level was higher for wounded tubers. When the tubers were harvested 16 days before inoculation instead of the same day, unwounded tubers of susceptible genotypes were less infected, but using the mean for wounded and unwounded tubers, the distinction between susceptible and resistant genotypes was still clear. Tubers inoculated 7 days after harvest were most infected when wounded shortly before inoculation, but tubers wounded two days before inoculation were infected more than unwounded tubers, and the separation between susceptible and resistant genotypes was more distinct than for unwounded tubers.

Summary Glasshouse-grown seedling tubers of 22 progenies from parents differing in resistance toFusarium coeruleum andF. sulphureum (Gibberella cyanogena) were wound-inoculated with a cornmeal + sand culture of one or other of these dry rot pathogens. WithF. coeruleum, differences between progenies were due entirely to differences in the general combining abilities (gca) of the parents. Parental and gca values were highly correlated, as were the mean resistance of a progeny and that of its parents. WithG. cyanogena the agreement between replicates was poor and differences between progenies were less clear. It is suggested that different resistance mechanisms operate against these two pathogens.

Summary In each of the five years 1978 to 1982, seed lots of cv. Bintje, variously infested byPhoma foveata Foister andFusarium solani var.coeruleum (Sacc.) Booth, as found by wound and cut tests, respectively, were planted in a series of field experiments at six sites in Sweden. A statistically significant relationship between seed and progeny infestation was found forP. foveata each year, and in three years forF. solani. Benomyl and thiabendazole mist treatments of seed tubers at grading in spring controlled the development of gangrene and dry rot in treated seed tubers, but did not consistently reduce the levels ofP. foveata andF. solani infestation in progeny from treated seed tubers. The site of cultivation markedly influenced mean levels ofP. foveata infestation, which were found to be related to temperature and rainfall during a 40 day pre-harvest period at the trail sites; however, mean levels ofF. solani infestation were not so related.

Summary Potato plants, after inoculation with a South African strain of biovar 2 or a strain of biovar 3 ofPseudomonas solanacearum were grown in growth chambers at temperatures ranging from 14/16°C (dark/light) up to 25/30°C. When grown at 14/16°C or higher, plants inoculated with the biovar 2 strain developed wilting symptoms and internal symptoms on progeny tubers and the disease was transmitted by the progeny tubers. When the plants were inoculated with the biovar 3 strain, wilting symptoms developed from 18/20°C, internal symptoms on progeny tubers from 20/22°C and the disease was transmitted by progeny tubers from plants grown at 16/18°C or higher.

Effect of saline irrigation and water deficit on tuber quality
Summary The effect of salinity and water dificit on the quality of tubers for processing was investigated. Total tuber yield was not affected by the treatments, while the percentage of non-marketable tubers was significantly reduced by high salinity (ECi=6dS m−1) and by water reduction. Accumulation of dry matter in the tubers was increased by all the treatments, that of proline by salinity only and the content of reducing sugars was increased only by water deficit. The colour of the french fries was similar in tubers from the various treatments.

Summary In artificial inoculations it was found thatFusarium oxysporum f.sp.tuberosi is able to penetrate through potato leaves. The fungus was isolated from the stem of infected plants from all inoculated cultivars. Tubers were completely destroyed if they were inoculated before sprouting. Not any destruction was observed if inoculation was made before young sprouts are longer than 1–3 cm; plants from these sprouts were infected. Lesioned tubers were more sensitive than unlesioned ones, and tuber rot and sprout damage were increased significantly.

Tubers of Cassava (Manihot esculenta), yams (Dioscorea esculenta),aroids (Amorphophallus campanulatus, Colocasia esculenta, Xanthosoma sagittifolium) and Coleus (Solenostemon rotundifolius) were screened for inhibitory activities against amylase, trypsin and chymotrypsin. Coleus tuber possessed the highest anti-amylase activity, whereas Colocasia tuber was the most potent source of anti-tryptic and anti-chymotryptic activity. Xanthosoma tubers exhibited amylase inhibitory activity and Amorphophallus tubers antiprotease activity. Dioscorea esculenta had low levels of amylase and chymotrypsin inhibitors, while Cassava tubers were totally free of inhibitors. When tubers were processed by pressure cooking, there was significant reduction/complete elimination in inhibitory activity. Partial retention of inhibition was observed in the case of amylase inhibitor in Dioscorea, chymotrypsin inhibitor in Colocasia and trypsin inhibitor in Colocasia, Coleus and Amorphophallus. In vitro experiments on heat stability of the different inhibitors revealed almost similar pattern of inactivation.

Summary Offspring were produced from a cross between two long-day-adaptedSolanum phureja clones which carried resistance to tuber soft rot (Erwinia carotovora subsp.atroseptica). In tests carried out on the produce of field-grown plants raised from tubers, over fifty per cent of the 173 offspring were found to be highly resistant. Assessments were also carried out of tuber yield, mean tuber weight, tuber number, shape, regularity, flesh colour, texture of the steamed flesh, fry colour, after-cooking blackening, sprout length after storage and overall dormancy. There were statistically significant differences between clones for all characters (P<0.001). Twelve of the clones were selected on the basis of high resistance, yield, tuber weight, regularity of shape and absence of after-cooking blackening. The value of resistant long-day-adapted diploid material for commercial breeding is discussed.

Summary Five experiments studied the effects of shading, temperature and stolon growth medium on stolon and tuber formation. The lowest numbers of stolons and tubers were produced at the higher and lower nodes. Numbers of primary stolons and tubers were relatively stable across nodes and treatments and variation in total numbers was largely due to changes in numbers of lateral and branch stolons. In the field, 70% shading reduced numbers of stolons and tubers by reducing lateral and branch stolons, while in hydroponics, 45% shading and temperatures at initiation had no effect on tuber numbers. In both environments later planting tended to reduce numbers of stolons and tubers. There were large effects of the physical environment, with compost producing three times as many tubers as dry vermiculite. The potential number of tubers may be determined by the number of stolons and the environmental conditions affecting stolon formation and development.

Summary In three experiments seed tubers of cvs Jaerla, Bintje and Alpha, stored at 12 °C in the dark, were planted in glasshouses on various dates at soil temperatures varying from 8 °C to 28 °C to study the phenomenon of ‘non-emergence’ with ageing seed tubers. In general the tubers were desprouted before planting. At all plantings cv. Alpha produced normal plants. The cvs Jaerla and Bintje, however, showed high proportions of the ‘little potato’ phenomenon when planted in cold soil, and high proportions of decayed tubers more frequently in warm soil. When using physiologically older seed at later planting dates, both the range of soil temperatures became wider and the proportions of seed tubers experiencing both ‘little potato’ and decay increased.
