Surveys were conducted in the five southern rice-producing states of Arkansas, Louisiana, Mississippi, Missouri and Texas in the United States during the 2018 and 2019 cropping seasons to determine the distribution and pathogenicity of fungal pathogens associated with seedling blight in rice. A total of 349 pathogenic fungal isolates were collected and identified as belonging to four genera: Rhizoctonia solani, Fusarium spp., Sclerotium rolfsii and Marasmius graminum based on morphological characteristics, molecular analysis and Koch's postulates. R. solani (252 out of 349 pathogenic isolates) was the most prevalent fungus isolated from diseased samples. Of the 252 pathogenic R. solani isolates, 245 were further classified as anastomosis group 11 (AG-11) and 7 as AG-4. Isolates of R. solani AG-4 and M. graminum were the most aggressive, with the highest stand loss (63% to 100%) and median disease rating (DR; 5.0), followed by isolates of R. solani AG-11 (stand loss = 4% to 100% and DR = 0.6 to 5.0), Fusarium spp. (stand loss = 26% to 48% and DR = 2.0 to 5.0) and S. rolfsii (stand loss = 33% to 48% and DR = 2.0 to 3.0) in causing seedling blight in rice. R. solani (62% to 83% of total pathogenic isolates) and Fusarium spp. (10% to 24% of total pathogenic isolates) were predominant in all the five states surveyed. S. rolfsii and M. graminum were present only in Louisiana and Texas. The results of this first systematic survey of rice seedling diseases in the southern United States will help develop effective fungicide seed treatment strategies for control of stand loss caused by seedling blight, one of the major factors limiting rice production.  相似文献

BACKGROUND: In the mid‐southern region of the United States, sugarcane borer, Diatraea saccharalis (F.), is a major target pest of transgenic maize expressing Bacillus thuringiensis (Bt) proteins. Novel transgenic maize technologies containing two or more pyramided Bt genes for controlling lepidopteran pests have recently become commercially available. Insect resistance management (IRM) is an important issue in the sustainable use of Bt crop technologies. The objective of this study was to determine the frequency of resistance alleles in field populations of D. saccharalis to the new pyramided Bt maize technologies. RESULTS: A total of 382 F2 family lines derived from 735 feral larvae/pupae of D. saccharalis collected from six locations in Louisiana and Mississippi during 2008 and 2009 were screened for resistance to three new Bt maize technologies: MON 89034, Genuity? VT Triple Pro? and SmartStax?. None of the 382 F2 isoline families survived on the Bt maize leaf tissue for ≥ 12 days in the F2 screen. The joint frequency for two‐ or three‐gene resistance models with 95% probability in these populations was estimated to be < 0.0063 to MON 89034 and < 0.003 to VT Triple Pro? and SmartStax?. CONCLUSION: These results suggest that the resistance allele frequency in D. saccharalis to the three pyramided Bt maize technologies is low in the mid‐southern region of the United States, which should meet the rare resistance assumption of the currently used IRM strategy for Bt maize. BACKGROUND: Research has shown that the family of grapefruit flavors called nootkatones have significant repellant and toxic effects to Formosan subterranean termites (Coptotermes formosanus Shiraki). RESULTS: Nineteen synthetic nootkatone derivatives, along with three commercially available nootkatone derivatives, were tested for repellent activity against C. formosanus by a choice assay in a petri dish with a two‐step triage procedure. Based on the repellency threshold value, the relationships between structure and activity are discussed. CONCLUSION: Four derivatives of nootkatone have very high repellency and toxicity to C. formosanus, 9 times the potency of the primary compound nootkatone. Four other compounds have between 2 and 3 times the repellency of nootkatones, and three compounds are equal in their repellency to nootkatone. Copyright © 2010 Society of Chemical Industry  相似文献

BACKGROUND: Among the termite infestations in the United States, the Formosan subterranean termite, Coptotermes formosanus Shiraki (Isoptera: Rhinotermitidae), is considered to be the most devastating termite pest. This pest most likely invaded North America as a result of the disembarkation of wooden military cargo at the port of New Orleans that arrived from Asia during and after World War II. It has now spread over other states, including Texas, Florida, South Carolina and California. Devastation caused by C. formosanus in North America has been estimated to cost $ US 1 billion a year. Over the past decades, organochlorines and organophosphates, the two prominent classes of termite control agents, have been banned owing to environmental and human health concerns. At the present time, phenylpyrazoles, pyrethroids, chloronicotinyls and pyrroles are being used as termite control agents. Mammalian toxicity and seeping of these compounds into groundwater are some of the drawbacks associated with these treatments. The instruction for the application of these termiticides indicate ground water advisory. Hence, with the increasing spread of termite infestation there is an increased need to discover effective, environmentally friendly and safe termite control agents with minimal mammalian toxicity. RESULTS: Chromene analogs derived from a natural‐product‐based chromene amide isolated from Amyris texana were tested in a collaborative discovery program for effective, environmentally friendly termite control agents. Several chromene derivatives were synthesized and characterized as a novel class of potential termiticides, followed by bioassays. These compounds exhibited significantly higher mortalities compared with untreated controls in laboratory bioassays. ABSTRACT Seventy-six populations of Heterodera glycines were collected from 33 counties in 10 states of the United States along the Mississippi and Missouri Rivers in 1992 and 1993. A sterile hyphomycete fungus of an unnamed taxon, designated ARF18 and shown to be a parasite of eggs of H. glycines, was isolated from eggs and cysts of 10 of the populations from Kentucky, Louisiana, Mississippi, and Tennessee. Ten isolates of ARF18 obtained in this study and seven isolates obtained in earlier studies were characterized for cultural morphology on several growth media, the ability to produce sclerotium-like structures (SLS) on cornmeal agar, growth rates, pathogenicity to eggs of H. glycines in vitro, and mitochondrial (mt) DNA restriction fragment length polymorphisms (RFLPs). All 17 isolates of ARF18 readily grew on potato dextrose agar, cornmeal agar, and nutrient agar. Based on colony morphology and SLS appearance on cornmeal agar, the isolates could be grouped into two morphological phenotypes. Isolates that produced SLS that were composed of a compact mass of hyphae were designated ARF18-C, whereas isolates that produced SLS composed of a mass of loosely clumped hyphae were designated ARF18-L. Only minor differences in growth rates were detected among the ARF18-C and ARF18-L isolates. All 10 ARF18-C isolates, which were from Arkansas, Louisiana, Mississippi, and Tennessee, belonged to a single mtDNA RFLP haplotype. The seven ARF18-L isolates shared many comigrating mtDNA restriction fragments with one another, but belonged to three distinct mtDNA RFLP haplotypes. Ability to infect eggs of H. glycines in vitro varied considerably among the various isolates of ARF18. In particular, several of the ARF18-C isolates were consistently able to infect over 50% (mean = 70.0%, standard deviation = 16%) of the eggs of H. glycines, whereas ARF18-L infected eggs to a lesser degree (mean = 25%, standard deviation = 27%). ARF18-C was isolated only from H. glycines populations from below 37 degrees N latitude. The presence of ARF18 was associated with soils with Mg levels <314 kg/ha, cyst numbers >4.5 per 100 cm(3), and iron levels >203.5 kg/ha; or with Mg levels >314 kg/ha and Na levels <121 kg/ha. The widespread distribution of ARF18 and the ability of some isolates to aggressively colonize eggs of H. glycines are indications that it has potential as a biological control agent for H. glycines.  相似文献

Scale insects were observed in neem trees (Azadirachta indica A. Juss) planted in Shambatarea near the bank of the Nile in Khartoum State and along Blue Nile Street in Wad Medani, Gezira State, Sudan in October 2014. Specimens of neem leaves and twigs, which were highly infested with scale insects and showing signs of wilting, were collected from different parts of the tree canopies and sent to the Insect Collection Unit at the Agricultural Research Corporation, Gezira Research Station, Sudan for identification. The pest was morphologically identified as Aonidiella orientalis (Newstead) (Hemiptera: Diaspididae). An endoparasitoid was observed to emerge from the scale insect and was identified as an encyrtid wasp. Aonidiella orientalis had been recorded previously in Shambatarea in 1969 and this paper presents the results of surveys carried out following the initial observations in Khartoum and Gezira States. This was a major outbreak, affecting the majority of the 1685 surveyed neem trees and causing damage on 62.7% of them.  相似文献

BACKGROUND: Drosophila suzukii is an oriental species first reported outside Asia from Hawaii in 1980. The first confirmed records for the continental United States were made in 2008 in California. The identification of this pest is difficult because very few published resources exist. RESULTS: It has since been recorded in Oregon, Washington, British Columbia, Alberta, Manitoba, Ontario, Quebec, Utah, Michigan, Wisconsin, Louisiana, North Carolina, South Carolina and Florida. Males are relatively easy to identify by the black apical wing spots and the single row of combs on the first and second tarsal segment of the fore leg. The male genitalia are also very characteristic and will aid in identifying teneral specimens. Females can be identified by the large ovipositor, which is 6–7 times as long as the diameter of the spermatheca. Immature stages can only be identified by molecular techniques. CONCLUSION: Although this species has been recorded from many US states and Canadian provinces, it has not been established in all of these places, and the main economic damage is restricted to the western part of North America. BACKGROUND: Imidacloprid is the primary insecticide for controlling the tobacco‐adapted form of the green peach aphid (TGPA), Myzus persicae (Sulzer), a major pest of tobacco worldwide. This study used leaf‐dip bioassays to assess TGPA resistance to imidacloprid in the eastern United States from 2004 through 2007. RESULTS: When combined over the 4 year study, 18, 14 and 3% of the TGPA had imidacloprid resistance ratios (RRs) of 10–20‐fold, 20–30‐fold and 30–90‐fold, respectively, compared with the most susceptible colony tested. This indicates that some colonies have developed moderate levels of resistance to imidacloprid. A colony collected near Clayton, North Carolina, had the highest RR of 91 (LC50 value = 31 mg L?1). This resistance declined for six tests over a 3 year period in the laboratory culture from >130‐fold RR (LC50 = 48 mg L?1) to 40‐fold RR (LC50 = 15 mg L?1). Over the same period, the most susceptible colony and a standard colony not exposed to imidacloprid for over 7 years had consistently low LC50 values. CONCLUSION: Moderate levels of resistance to imidacloprid are noticed among TGPA colonies from the eastern United States. The variation in resistance indicates that the factors responsible are present in the populations at low frequencies and are just not enough to cause field failures yet. In this study, 18 microsatellite loci were used to examine the genetic structure and mode of reproduction of Phaeomoniella chlamydospora, the fungus that causes Petri disease of grapevine and is involved in another grapevine disease, esca. A total of 60 southeastern Australian isolates were tested and compared with 64 isolates from southern France. The French population possessed relatively high genotypic diversity (G = 29·2) whilst the Australian population showed low genotypic diversity (G = 11·1), consistent with a limited founder population. Haplotypes from four different grapevine cultivars were not significantly differentiated (Φpt values effectively zero). Likewise, genetic differentiation of haplotypes from different regions within each country was not significant, although small but significant genetic differentiation (9%) was identified between Australia and France. Based on bootstrapped cluster analysis, there did not appear to be any genetic groups within the overall sample of isolates. Significant linkage disequilibrium identified in both countries, together with overrepresented, widespread identical haplotypes, indicated limited genetic recombination and a largely clonal structure consistent with the absence of an observed sexual cycle in this species.  相似文献

ABSTRACT Both typical late season stem infections and atypical early season rosette infections of canola, a relatively new crop in the southeastern United States, were caused by Sclerotinia sclerotiorum. The 51 DNA fingerprints (from 71 isolates) did not match any fingerprints from previous studies of canola or other crops. Single locus haplotypes from nuclear DNA sequences included 18 in the intergenic spacer (IGS) of the rRNA repeat, four in 44.11, six in translation elongation factor 1alpha, three in calmodulin (CAL), and two in chitin synthase 1. Contingency permutation testing for associations of infection type with DNA fingerprint, single- or multilocus haplotype, or hierarchically nested clades based on single locus haplotypes found significant association of haplotype with mycelial compatibility group and DNA fingerprint for all loci except CAL. Significant association of IGS haplotypes with symptom type was detected in one pathogen population. Southeastern U.S. canola was infected by both recently evolved, geographically dispersed pathogen genotypes and older, indigenous genotypes (Carbone and Kohn, 2001. Mol. Ecol. 10:947-964). Indigenous haplotypes are infection-type generalists, and the most frequently isolated from rosette infections. In contrast, haplotypes from the most recently evolved, dispersed population were associated one-to-one with infection type, with only the most recently evolved haplotypes infecting rosettes.  相似文献

Although considerable information exists regarding the importance of moisture in the development of soybean rust, little is known about the influence of temperature. The purpose of our study was to determine whether temperature might be a significant limiting factor in the development of soybean rust in the southeastern United States. Soybean plants infected with Phakopsora pachyrhizi were incubated in temperature-controlled growth chambers simulating day and night diurnal temperature patterns representative of the southeastern United States during the growing season. At 3-day intervals beginning 12 days after inoculation, urediniospores were collected from each plant and counted. The highest numbers of urediniospores were produced when day temperatures peaked at 21 or 25°C and night temperatures dipped to 8 or 12°C. When day temperatures peaked at 29, 33, or 37°C for a minimum of 1 h/day, urediniospore production was reduced to 36, 19, and 0%, respectively, compared with urediniospore production at the optimum diurnal temperature conditions. Essentially, no lesions developed when the daily temperature high was 37°C or above. Temperature data obtained from the National Climatic Data Center showed that temperature highs during July and August in several southeastern states were too high for significant urediniospore production on 55 to 77% of days. The inhibition of temperature highs on soybean rust development in southeastern states not only limits disease locally but also has implications pertaining to spread of soybean rust into and development of disease in the major soybean-producing regions of the Midwestern and northern states. We concluded from our results that temperature highs common to southeastern states are a factor in the delay or absence of soybean rust in much of the United States.  相似文献

Selective media without pentachloronitrobenzene were developed for quantitative assays of Fusarium oxysporum in soils. Media Fo-G1 and Fo-G2 were effective for naturally infested soils, Fo-W1 and Fo-W2 for wild-type isolates in soils containing a nitrate-nonutilizing (nit) mutant, and Fo-N1 and Fo-N2 for nit mutants. Selective media were made using ammonium citrate dibasic, l-sorbose, econazole nitrate, 25% iminoctadine triacetate solution and 50% tolclofos-methyl wettable powder for soil dilutions of 100-fold or more (Fo-G1, FoW1 and Fo-N1) and 10-fold (Fo-G2, Fo-W2 and Fo-N2). Potassium chlorate was added to Fo-N1 and Fo-N2. The efficacy for selectively isolating F. oxysporum was confirmed using six soils naturally infested with one of six formae speciales of F. oxysporum and with soil dilutions containing conidia of wild-type strains or nit mutants from the six formae speciales. On Fo-G1 and Fo-G2, most colonies of F. oxysporum were compact and round with purplish or reddish pigment in the reverse. Cylindrocarpon sp. formed colonies as large as those of F. oxysporum but were distinguishable by their colony morphology. Other contaminants such as F. solani, F. moniliforme, and Trichoderma were suppressed by medium ingredients and colonies of F. oxysporum. On Fo-W1 and Fo-W2, colony morphology of F. oxysporum and contaminants corresponded to that on Fo-G1 and Fo-G2, although F. oxysporum failed to produce the pigment. On Fo-N1 and Fo-N2, nit mutants formed clear colonies from 100- and 10-fold soil dilutions, respectively, and contaminants seldom formed large colonies.  相似文献

Candidatus Liberibacter solanacearum’ is associated with the Zebra Chip (ZC) disorder of potatoes. A panel of eight simple sequence repeat (SSR) markers was developed and used to genetically characterize ‘Ca. L. solanacearum’ strains obtained from ZC-affected potato plants in the United States and Mexico. The multilocus SSR markers in this study effectively differentiated genotypes and estimated genetic diversity of ‘Ca. L. solanacearum’ strains. Genotype assignment analyses identified two major lineages of ‘Ca. L. solanacearum’ in the North American populations while only one lineage type was identified in Mexican population. No clear genetic structure was found among haplotypes based on geographical proximity or host. The high resolution power of the SSR marker system developed in this study provides a useful tool for genotyping closely related strains and tracking sources of the pathogen. Genotype information combined with epidemiological data will advance knowledge of ZC disease and will facilitate development of effective disease management.  相似文献

ABSTRACT Morphological characteristics and vegetative compatibility groups (VCGs) of 486 isolates of Glomerella cingulata, Colletotrichum gloeosporioides, and C. acutatum collected from apple leaves with Glomerella leaf spot (GLS) symptoms and fruit with bitter rot symptoms in the United States and Brazil were studied. From this collection, 155 isolates of G. cingulata (93 from fruit, 61 from leaves, and 1 from buds), 42 isolates of C. gloeosporioides from fruit, and 14 isolates of C. acutatum (10 from fruit and 4 from leaves) were studied using mitochondrial (mt)DNA restriction fragment length polymorphism (RFLP) haplotypes. A subset of 24 isolates was studied by examining the sequence of a 200-bp intron of the glyceraldehyde 3-phosphate dehydrogenase (GDPH) nuclear gene. In addition, 98 isolates were tested for pathogenicity on leaves of cvs. Gala and Golden Delicious in the greenhouse, and 24 isolates were tested for pathogenicity on fruit of cv. Gala in growth chambers. In total, 238 and 225 isolates of G. cingulata were separated into four distinct morphological types and six VCGs, respectively. Five morphological types and six VCGs were identified among 74 and 36 isolates of C. gloeosporioides, respectively. Three morphological types and four VCGs were identified among 74 and 23 isolates of C. acutatum, respectively. Seven different mtDNA RFLP haplotypes were observed within isolates of G. cingulata, two within isolates of C. gloeosporioides, and two within isolates of C. acutatum. Phylogenetic trees, inferred based on maximum likelihood and maximum parsimony methods using the intron sequence, produced similar topologies. Each species was separated into distinct groups. All isolates tested were pathogenic on fruit, though only isolates with specific VCGs and haplotypes were pathogenic to leaves. Vegetative compatibility was a better tool than molecular characters for distinguishing isolates of G. cingulata pathogenic on both leaves and fruit from the ones pathogenic only on fruit. Isolates of G. cingulata capable of causing both GLS and bitter rot were included in haplotypes and groups based on the sequence analysis of the 200-bp intron that also included isolates capable of causing bitter rot only. Additionally, isolates of G. cingulata from the United States and Brazil which cause GLS were included in different haplotypes and sequence analysis groups. Therefore, one hypothesis is that isolates of G. cingulata from the United States capable of causing both GLS on foliage and bitter rot on fruit may have arisen independently of Brazilian isolates of G. cingulata capable of causing both GLS and bitter rot, and the two groups of isolates may represent distinct populations.  相似文献

Summary. The response of tubers to low temperatures was investigated to gain insight into a physiological basis for the differential distribution of Cyperus esculentus L. (yellow nutsedge) and C. rotundus L. (purple nutsedge) in the United States. Only C. esculentus tubers survived the winter of 1968–69 in the field at Urbana, Illinois. Less than 10% of C. rotundus tubers survived at 2°C for 12 weeks, whereas more than 95% of C. esculentus tubers survived this treatment. Exposures to -2°C for 4 h or longer killed 50% of C. rotundus tubers, whereas - 6·5°C was required to kill 50% of C esculentus tubers. C. rotundus distribution is restricted to regions where the soil seldom freezes, whereas C. esculentus is distributed in regions where the soil temperatures often get below freezing. Survival of C. esculentus tubers in soil which frequently freezes may account for its wide distribution. Death of C. rotundus tubers in soils which freeze apparently is the reason why the species is restricted to the southeastern and southwestern regions of the U.S.A. Monilinia fructicola is a quarantine fungal pathogen in Europe, but many major stone fruit growing countries in Europe have reported its presence recently. In Switzerland, the fungus was first found in a single apricot orchard in 2008. This study confirms the presence of M. fructicola in nine out of 22 commercial orchards in Canton Valais, Switzerland. Five simple sequence repeat markers (SSRs) were developed for M. fructicola and samples from Switzerland, Spain, Italy, France and the United States were analysed and compared in order to assess the genetic diversity of the pathogen, identify the origin of the disease, and verify if the fungus reproduces sexually in Europe. In the 119 European samples analysed, 12 different haplotypes were found, indicating a relatively high genetic diversity of the pathogen considering that the first report in Europe was 10 years ago. Three haplotypes found in Europe were identical to those found in the American samples (two from the east coast and one from the west coast). Population structure analysis suggests that the European population is derived from at least two ‘invasion’ events probably originating from the US (one from the east coast, the other from the west coast). Preliminary evidence of sexual reproduction of M. fructicola in Europe is reported.  相似文献

Infestations of aphids(Macrosiphum rosae L.) and of twospotted spider mites(Tetranychus urticae Koch) were examined in relation to growth and sporulation ofClonostachys rosea andBotrytis cinerea, and to suppression of the pathogen by the agent, in green rose leaves. Leaves were infested artificially with 10 aphids/leaflet for 3 h, or naturally with 15-30 aphids/leaflet for 7-12 days or with undetermined numbers of mites for 10-12 days. Leaves that had or had not been infested were inoculated withC. rosea, withB. cinerea, or withC. rosea plusB. cinerea. Germination incidence and germ tube growth ofC. rosea andB. cinerea on the phylloplane in most instances were much greater in leaves previously infested with aphids or mites compared with noninfested leaves. After combined inoculation,C. rosea suppressed germination ofB. cinerea from 47% to 19% in noninfested leaves, but in leaves that had been infested the agent was ineffective and germination incidence of the pathogen increased to 75-93%. Previous infestation with naturally introduced aphids or mites, but not brief infestations of artificially introduced aphids, markedly increased sporulation ofC. rosea after the leaves died during an initial 7-15 days of incubation on a paraquat agar medium, regardless of whether or notB. cinerea was present. Sporulation ofB. cinerea was similarly increased when inoculated alone. After 15-20 days, however, conidiophores of the agent or pathogen covered most of the leaf surface in these treatments. In leaves inoculated withC. rosea plusB. cinerea, the agent suppressed sporulation of the pathogen almost completely in both previously infested and noninfested leaves. Thus, aphid and mite infestations did not compromise the ability ofC. rosea to suppress inoculum production byB. cinerea in the leaves. Increased nutrient availability on the phylloplane through exudation or as honeydew or frass is proposed as a basis to explain effects of the pest infestations onC. rosea andB. cinerea.  相似文献

BACKGROUND: Neonicotinoid insecticides were first used commercially for Colorado potato beetle [Leptinotarsa decemlineata (Say), Coleoptera: Chrysomelidae] control in the United States in 1995, and since then have been critical for management of this pest. Field populations from the northeastern and midwestern United States were tested from 1998 to 2010 for susceptibility to imidacloprid and thiamethoxam using standard topical dose assays with adults. RESULTS: From 1998 to 2001, imidacloprid resistance was present in only a few locations in the eastern United States. By 2003, imidacloprid resistance was common in the northeastern Unites States. In 2004, imidacloprid resistance in Colorado potato beetle was detected for the first time in the midwestern United States. In 2003, the first case of resistance to thiamethoxam was found in a population from Massachusetts. Neonicotinoid resistance in summer‐generation adults was higher than in overwintered adults from the same locations. By 2009, 95% of the populations tested from the northeastern and midwestern United States had significantly higher LD50 values for imidacloprid than the susceptible population. CONCLUSIONS: The increasing resistance to neonicotinoid insecticides raises concerns for the continued effective management of Colorado potato beetles in potatoes and highlights the need for more rigorous practice of integrated pest management methods. Formosan subterranean termites, Coptotermes formosanus Shiraki, were exposed to ? 95% or 50% carbon dioxide atmospheres for intervals of 24-120 h at 26(±3)°C. A 24-h exposure to ? 95% carbon dioxide caused significant termite mortality, but 60 h were required for complete mortality. Exposure to 50% carbon dioxide for 60 h resulted in approximately 70% termite mortality, while complete mortality was recorded after 120 h. When termites were sealed in wooden blocks (90 × 90 × 152 mm), 72-96 h exposure to ? 95% carbon dioxide was necessary for complete control. A limited study with Cryptotermes brevis (Walker) suggested that this drywood termite is also susceptible to carbon dioxide fumigation, although slightly longer exposures may be required than with C. formosanus. Carbon dioxide-modified atmospheres are a viable alternative to conventional fumigants for vault fumigation of termite-infested materials, and may also be applicable to larger-scale fumigations to control structural pests.  相似文献
