The high prevalence of the great resistance to antimicrobials shown by the aetiological agents of infectious bovine mastitis, often leads to chronic recurrent mastitis, one of the main causes of early culling of dairy cows. The purpose of this study was to determine the in vitro susceptibility pattern of Staphylococcus spp. isolated from mammary parenchymas of slaughtered dairy cows, to different anti-microbials. A total of 45 Staphylococcus spp. strains [33 coagulase-negative Staphylococcus (CNS) and 12 Staphylococcus aureus (SA)] were used. These strains were tested with 12 different antimicrobials by the Kirby and Bauer standardized disc diffusion method; 84.44% and 86.66% of the 45 strains were resistant to ampicillin and penicillin, respectively. The highest sensitivity was to cephalothin (84.44%), gentamicin (80%) and to sulphazotrin (77.77%). CNS showed higher resistance (P < 0.05) than SA. The in vitro susceptibility pattern of Staphylococcus spp. strains isolated from mammary parenchymas studied in the present investigation was similar to that observed in recent studies of in vitro and in vivo susceptibility patterns of Staphylococcus spp. isolated from cases of bovine mastitis.  相似文献

In a dairy herd with a low incidence of intrammary infections due to Streptococcus agalactiae and Staphylococcus aureus, clinical mastitis remained a serious problem despite good control of nonclinical mastitis through postmilking teat disinfection and antibiotic therapy of known infected quarters at the end of lactation. During the 2-year study, the incidence of clinical mastitis was 0.88 cases/cow-year; 32.2% were caused by streptococcal species other than Str agalactiae and 33.5% by gram-negative organisms. Among all new infections detected, 54.1% were caused by streptococcal species other than Str agalactiae and 25.7% by gram-negative bacteria. Among new infections, 41.6% occurred during the nonlactating period or within a few days of calving. Incidence of clinical mastitis was highest in the 1st month of lactation. Among 84 gram-negative infections, 42.8% were caused by Klebsiella pneumoniae, 20.2% by Escherichia coli, and 23.8% by Enterobacter spp. Among the many serotypes of K pneumoniae and E coli, none was predominant.  相似文献

乳房炎是奶牛最常见的疾病,随着奶牛产业的蓬勃发展,乳房炎已经成为制约该产业的瓶颈之一。从甘肃省武威市凉州区5个奶牛养殖场采集了1 000头荷斯坦奶牛共计3 991份奶样,对临床型和非临床型(隐性)乳房炎分别通过临床症状和兰州隐性乳房炎诊断液(LMT)进行诊断。结果显示:该地区奶牛临床型乳房炎发病率18.20%,亚临床型乳房炎发病率36.50%,总阳性率54.70%,总阳性乳区率54.72%。从阳性奶样中分离纯化得到的主要病原菌为链球菌属、金黄色葡萄球菌和大肠杆菌,且在不同类型乳房炎中的分布情况不同,临床型中链球菌属最多(54.37%),亚临床型中金黄色葡萄球菌最多(48.56%)。对主要病原菌进行药敏试验,结果表明治疗凉州区奶牛乳房炎的最佳药物应为喹诺酮类和庆大霉素类。  相似文献

奶牛临床型乳房炎的细菌分离鉴定与耐药性分析   总被引:1,自引:0,他引:1  
2011年山西省多个奶牛场发生了较严重的乳房炎,对76份采集的奶样进行细菌分离鉴定并采用药敏纸片法检测主要分离菌的抗生素耐药情况。所分离细菌以革兰氏阳性菌为主,革兰氏阳性球菌和其他革兰氏阳性菌分别占60.67%和23.59%。分离出多种病原菌和机会致病菌,主要的病原菌有链球菌、金黄色葡萄球菌、大肠杆菌等,检出率为2.24%~11.24%,其中无乳链球菌的检出率最高;机会致病菌有粪链球菌、微球菌、克雷伯菌、凝固酶阴性葡萄球菌等,检出率为1.12%~11.24%,其中粪链球菌和微球菌的检出率较大,分别为11.24%和6.74%。药敏试验检测结果显示,在所选的15种药物中,主要分离菌均对丁胺卡那霉素、氟哌酸和恩诺沙星3种药物敏感;大肠杆菌、克雷伯菌、凝固酶阴性葡萄球菌对青霉素类和β-内酰胺/β-内酰胺酶抑制剂类药物产生了极强的耐药性,耐药率均为100%;乳房链球菌对该类药物也产生不同程度的耐药,耐药率为20%~100%;对链霉素产生100%耐药的细菌有大肠杆菌、克雷伯菌、无乳链球菌、乳房链球菌、停乳链球菌等细菌;部分分离菌对卡那霉素、庆大霉素、链霉素、四环素、红霉素、先锋霉素Ⅴ、复方新诺明等药物产生不同程度耐药。被检奶牛场混合感染较为严重,应进一步加强环境卫生管理,临床治疗应合理有效用药。  相似文献

Coagulase-negative staphylococci (CNS) are often associated with bovine mastitis. Knowledge about the relative importance of specific CNS species in different types of mastitis, and differences in antimicrobial resistance among CNS species is, however, scarce. Therefore, the aims of this study were to compare prevalence and antimicrobial susceptibility of CNS species in clinical and subclinical mastitis using material from two national surveys. Overall, Staphylococcus chromogenes and Staphylococcus epidermidis were the most common CNS species found followed by Staphylococcus simulans and Staphylococcus haemolyticus. S. epidermidis was significantly more prevalent in subclinical than in clinical mastitis, and a similar trend was observed for Staphylococcus saprophyticus, while Staphylococcus hyicus was significantly more common in clinical mastitis. The prevalence of β-lactamase producing isolates varied markedly between CNS species, and was significantly higher in S. epidermidis and S. haemolyticus (~ 40%), than in S. simulans and S. chromogenes where none or a few of the isolates produced β-lactamase. Resistance to more than one antimicrobial substance occurred in 9% and 7% of the clinical and subclinical isolates, respectively. In conclusion, the distribution of CNS species differed between clinical and subclinical mastitis indicating inter-species variation of pathogenicity and epidemiology. Overall, the prevalence of antimicrobial resistance was low, but some variation between CNS species was observed.  相似文献

This study was carried out to determine the prevalence of coagulase-negative staphylococci in clinical and subclinical mastitis in commercial and small-scale farms in Zimbabwe. Thirty five quarter milk samples from clinical mastitis cases and 371 quarter milk samples from cows with subclinical mastitis were cultured for bacterial pathogens. The most frequent pathogens isolated in clinical mastitis were the enteric bacteria (31.4%), followed by coagulase negative staphylococci (22.9%) and then Staphylococcus aureus (17.1%), whereas in subclinical mastitis S. aureus (34.2%) and coagulase-negative staphylococci were (33.2%) the most common. Bacillus species were only isolated in milk samples from subclinical mastitis. Coagulase-negative staphylococci were observed in mixed infections with other bacteria in only 2.2 of the 406 milk samples from clinical and subclinical mastitis where they were isolated together with Bacillus species in 6 of the 9 mixed infection cases. About 95% of the milk samples from which 131 coagulase-negative staphylococci were isolated had correspondingly high somatic cell counts. The coagulase-negative staphylococci isolated most frequently were S. chromogenes (7.9%), S. epidermidis (7.4%) and S. hominis (5.9%). They were all associated with high somatic cell counts. All the coagulase-negative staphylococci isolates were susceptible to cloxacillin and erythromycin, and more than 90% of the isolates were susceptible to neomycin, penicillin and streptomycin. The highest resistance was to tetracycline (17.6%), followed by lincomycin (13.7%). About 8% of the isolates were resistant to both penicillin and streptomycin.  相似文献

Six hundred and thirty clinically-normal milk samples from dairy goat flocks comprising a mixed population of German Alpine, Toggenburg, Saanen and Galla crosses were examined over a 3-month period to determine the prevalence of bacterial organisms. Bacteria were isolated in 28.7% of the milk samples (181/630) either singly (92.8%) or in combination (7.2%). The most prevalent bacterial organisms were Staphylococcus spp. (60.3%), followed by Micrococcus spp. (17.7%), Acinetobacter spp. (5%), Actinomyces spp. (5%) and Streptococcus spp. (1.1%). The Staphylococcus spp. were mainly coagulase negative (64.3%). Coagulase-negative staphylococci and coagulase-positive staphylococci accounted for 37.5% and 22.7% respectively of the total bacteria isolated. The isolation of bacteria, some of which are important in clinical and subclinical mastitis, in apparently normal caprine milk, indicates that particular attention should be given to the management of these dairy goat flocks in order to avoid the development of cases of clinical mastitis.  相似文献

The objective of this study was to describe the frequency of occurrence of clinical mastitis in dairy herds in Ontario. The study group consisted of 65 dairy farms involved in a 2-year observational study, which included recording all clinical mastitis cases and milk sampling of quarters with clinical mastitis. Lactational incidence risks of 9.8% for abnormal milk only, 8.2% for abnormal milk with a hard or swollen udder, and 4.4% for abnormal milk plus systemic signs of illness related to mastitis were calculated for 2840 cows and heifers. Overall, 19.8% of cows experienced one or more cases of clinical mastitis during location. Teat injuries occurred in 2.1% of lactations. Standard bacteriology was performed on pretreatment milk samples from 834 cows with clinical mastitis. The bacteria isolated were Staphylococcus aureus (6.7%), Streptococcus agalactiae (0.7%), other Streptococcus spp. (14.1%), coliforms (17.2%), gram-positive bacilli (5.5%), Corynebacterium bovis (1.7%), and other Staphylococcus spp. (28.7%). There was no growth in 17.7% of samples, and 8.3% of samples were contaminated. Clinical mastitis is a common disease in dairy cows in Ontario; approximately 1 in 5 cow lactations have at lease one episode of clinical mastitis. There is, however, considerable variation in the incidence of clinical mastitis among farms. The majority of 1st cases of clinical mastitis occur early in lactation, and the risk of clinical mastitis increases with increasing parity. Environmental, contagious, and minor pathogens were all associated with cases of clinical mastitis.  相似文献

为查明甘肃省某奶牛场发生临床型奶牛乳房炎的主要病原菌并选择治疗用敏感药物,随机无菌采集了6份临床型病牛乳样和1份大罐乳样,采用划线分离、菌落形态观察、革兰染色镜检、生化鉴定及16S rDNA序列分析等方法进行细菌分离鉴定,同时采用K-B法测定主要分离菌株对抗菌药物的敏感性。结果表明,从临床型乳房炎乳样中分离鉴定出大肠埃希菌、芽胞杆菌、凝固酶阴性葡萄球菌(腐生葡萄球菌)3种细菌,其中大肠埃希菌的分离率最高,为66.67%;从大罐奶样中也分离出大肠埃希菌。药敏试验结果表明,大肠埃希菌对氨苄青霉素、阿莫西林、头孢噻吩、杆菌肽、红霉素均存在不同程度的耐药,耐药率为29%~100%;对诺氟沙星、恩诺沙星、氟苯尼考、头孢哌酮等药物敏感。说明此次牛场临床型乳房炎的主要病原菌是大肠埃希菌、葡萄球菌和芽胞杆菌,而且分离的大肠埃希菌具有多重耐药性。根据检测分析结果,给奶牛场提出针对性的防控措施,临床型乳房炎的发病率下降,病情得以控制。  相似文献

In this review of the literature, mastitis-causing coagulase-negative staphylococci (CNS) and Staphylococcus aureus are compared. Staphylococci are the bacteria most commonly isolated from bovine mastitis, and CNS are now predominant over S. aureus in most countries. CNS include various species, but only a few prevail in bovine mastitis. S. aureus can cause clinical mastitis, but often causes subclinical mastitis, which remains persistent and increases milk somatic cell count. CNS, traditionally regarded as minor pathogens, seem to lack the ability to cause severe mastitis. CNS can, however, persist in the mammary gland and moderately increase milk somatic cell count. Resistance to various antimicrobials is more common in CNS than in S. aureus, but CNS mastitis responds much better to antimicrobial treatment than S. aureus mastitis.  相似文献

本研究对江苏部分奶牛场致隐性乳房炎病原进行流行病学调查,采样并进行细菌分离培养、革兰氏染色镜检、生化鉴定,共分离到35株凝固酶阴性葡萄球菌(CNS)。用常规药敏纸片法对CNS分离菌株的抗生素耐药情况进行分析,结果显示分离培养到的35株CNS对临床常用抗生素万古霉素、左氟沙星、克拉霉素、苯唑西林、诺氟沙星、氯霉素、环丙沙星、头孢噻肟、磷霉素和呋喃妥因等高度敏感,高敏率在60%~80%之间;而对青霉素G、复方新诺明和多黏菌素B有一定的耐药性,耐药率为70%~100%。分离菌株表现了较强的多重耐药性,耐20种药物的细菌有3株,耐15种药物的细菌有1株,耐14种药物的细菌有1株,62.9%的CNS分离菌株能耐5种以上抗生素,表明导致奶牛隐性乳房炎CNS菌株耐药情况不容小视。  相似文献

One hundred and forty eight strains of coagulase-negative staphylococci (CNS) from bovine clinical and subclinical mastitis were classified by conventional biochemical testing. Staphylococcus simulans was the most frequently isolated species. The distribution of CNS species in the two groups was similar regardless of whether the cow suffered from clinical or subclinical mastitis. There was no significant difference between the grades of severity of the clinical mastitis associated with the various CNS. The small difference in pathogenicity observed between species in this study, and the similarity of distribution in the various species of clinical and subclinical mastitis could indicate that the species per se is of minor importance in the development of mastitis.  相似文献

Coagulase-negative staphylococci (CNS) have become the most common bovine mastitis isolate in many countries and could therefore be described as emerging mastitis pathogens. The prevalence of CNS mastitis is higher in primiparous cows than in older cows. CNS are not as pathogenic as the other principal mastitis pathogens and infection mostly remains subclinical. However, CNS can cause persistent infections, which result in increased milk somatic cell count (SCC) and decreased milk quality. CNS infection can damage udder tissue and lead to decreased milk production. Staphylococcus simulans and Staphylococcus chromogenes are currently the predominant CNS species in bovine mastitis. S. chromogenes is the major CNS species affecting nulliparous and primiparous cows whereas S. simulans has been isolated more frequently from older cows. Multiparous cows generally become infected with CNS during later lactation whereas primiparous cows develop infection before or shortly after calving. CNS mastitis is not a therapeutic problem as cure rates after antimicrobial treatment are usually high. Based on current knowledge, it is difficult to determine whether CNS species behave as contagious or environmental pathogens. Control measures against contagious mastitis pathogens, such as post-milking teat disinfection, reduce CNS infections in the herd. Phenotypic methods for identification of CNS are not sufficiently reliable, and molecular methods may soon replace them. Knowledge of the CNS species involved in bovine mastitis is limited. The dairy industry would benefit from more research on the epidemiology of CNS mastitis and more reliable methods for species identification.  相似文献

Somatic cell count (SCC), bacteriological findings and electrical conductivity (EC) were measured in milk from 216 quarters of 54 Murrah crossbred buffaloes for characterization of buffalo milk. Mean SCC in normal, subclinical and clinical mastitic milk was 171, 799 and 6039 × 103/mL, respectively. Coagulase negative Staphylococci (CNS) were the major organisms associated with subclinical mastitis, whereas CNS and coliforms were the predominant organisms in clinical mastitis. EC values were found to be significantly higher in buffalo milk with mammary infection. The best trade-off between sensitivity and specificity for diagnosing mastitis in buffaloes by EC score was found at the cut-off value of 3.7 mS/cm. The direct measures such as SCC, EC, bacteriological findings, and appearance of milk were found to be useful for the evaluation of udder health of buffaloes and categorization of milk.  相似文献

The concentrations of haptoglobin (Hp) and serum amyloid A (SAA) and the activity of N-acetyl-β-D-glucosaminidase (NAGase) in milk from 234 cows with spontaneous mastitis caused by different pathogens were measured to assess whether they corresponded with the clinical signs of mastitis and whether there were any differences between pathogens. Ninety-eight of the cows had clinical mastitis and 136 had subclinical mastitis. There were statistically significant positive correlations between the concentrations of SAA and Hp and the activity of NAGase. Significant differences in the concentrations of acute phase proteins and NAGase activity were found in milk from cows with mastitis caused by different pathogens. The highest concentrations of Hp and NAGase were found in cases of mastitis caused by Escherichia coli and Arcanobacterium pyogenes, and the lowest concentrations were from cases of mastitis caused by coagulase-negative staphylococci. Very low SAA concentrations were found in milk from the cases caused by A pyogenes, in contrast to cases caused by other major mastitis pathogens. The median concentration of SAA was over 10 times higher in cases of mastitis caused by E coli than in mastitis caused by other pathogens. There were significant differences in the mean Hp concentration and NAGase activity between clinical and subclinical mastitis. In approximately one-third of the samples, the Hp concentration was below the detection limit, potentially compromising the use of Hp as a mastitis marker.  相似文献

AIM: To identify and enumerate colony forming units (cfu) of mastitis pathogens in bulk tank milk (BTM) from pasture-fed New Zealand dairy cows in the Waikato region. METHODS: BTM samples from seven seasonal-calving dairy herds in the Waikato region were collected monthly from August to December 2004 (cows calved during July-September). Milk samples were cultured on blood aesculin and MacConkey agar plates for 24 h, and the number of mastitis pathogens identified and counted. RESULTS: Colonies identified in BTM included aesculin-positive streptococci, Staphylococcus aureus, coagulase-negative staphylococci (CNS), and coliforms; counts ranged from zero to >1,000 cfu/ml. Counts>1,000 cfu/ml for total aesculin-positive streptococci, CNS and coliforms were present in 48%, 51% and 11% of BTM samples, respectively. Counts of Staph. aureus ranged from zero to 1,000 cfu/ml, but first appeared in BTM samples only in October. Staphylococcus aureus was repeatedly isolated in BTM from 4/7 farms during the testing period. CONCLUSIONS: Counts of mastitis pathogens in this study appeared high relative to interpretive criteria set by other workers, which may indicate a high prevalence of mastitis risk factors on these farms. Interpretation of results is difficult as aesculin-positive streptococci, CNS and coliforms can be isolated from the environment as well as from cows with clinical or subclinical mastitis. Furthermore, Staph. aureus is inconsistently excreted from infected bovine mammary glands. More extensive study of this method is required in New Zealand to attempt to further validate the interpretation of results of bacterial culture of BTM. CLINICAL RELEVANCE: This method may be used to monitor challenge from mastitis pathogens over time as part of milk quality control programmes. The technique may be of use as a screening test to provide information to veterinarians, affording them the opportunity to have an input into mastitis control on dairy farms in New Zealand.  相似文献

Mammary gland quarters of 139 lactating dairy cows from small-scale dairy herds were examined visually and by palpation for teat lesions and by California mastitis test (CMT) and bacterial culture for subclinical mastitis. Teat lesions were observed in 97 teats. These included teat chaps (39.2%), teat papillomas (23.7%), teat erosions (22.7%), teat fistulae (5.1%), inverted teats (5.1%) and blocked teats (4.2%). According to the CMT, the prevalence of subclinical mastitis was 33.4% in all the mammary gland quarters, 71.0% in quarters with teat lesions and 24.5% in quarters without teat lesions. There was a significant (P < 0.01) association between teat lesions and the prevalence of subclinical mastitis. The mammary gland quarters with teat lesions were 7.2 times more likely to have a positive CMT (P < 0.01) and 5.6 times more likely to have bacterial organisms (P < 0.01) isolated from them than those without any teat lesions. The bacterial organisms most frequently isolated from the CMT-positive milk samples from both the mammary gland quarters with teat lesions and those without teat lesions were Staphylococcus aureus (50.0%), Streptococcus spp. (34.8%) and Arcanobacterium pyogenes (6.2%).  相似文献

CONCLUSIONS: Counts of mastitis pathogens in this study appeared high relative to interpretive criteria set by other workers, which may indicate a high prevalence of mastitis risk factors on these farms. Interpretation of results is difficult as aesculinpositive streptococci, CNS and coliforms can be isolated from the environment as well as from cows with clinical or subclinical mastitis. Furthermore, Staph. aureus is inconsistently excreted from infected bovine mammary glands. More extensive study of this method is required in New Zealand to attempt to further validate the interpretation of results of bacterial culture of BTM.

CLINICAL RELEVANCE: This method may be used to monitor challenge from mastitis pathogens over time as part of milk quality control programmes. The coagulase-negative staphylococcal species causing mastitis in lactating cattle were identified and possible differences in the clinical characteristics or persistence of mastitis caused by different CNS were evaluated. The effect of antimicrobial treatment was also assessed. In addition, AFLP-typing of CNS was compared with the phenotypic identification. A total of 133 clinical or subclinical quarter cases of intramammary infection caused by CNS from the practice area of the Ambulatory Clinic of the University of Helsinki were studied. Bacteriological diagnosis was based on biochemical (API) testing. Staphylococcus simulans (43.6%) followed by S. chromogenes (23.3%) were the most common CNS species isolated from the milk samples. Ninety-nine isolates were genotyped using AFLP-analysis. Only 75.0% of S. chromogenes and S. simulans isolates identified with API test were clustered with the type strains of these species. Approximately half of the mastitis cases were clinical, and in the majority clinical signs were mild. The severity and persistence of intramammary infection were unaffected by CNS species. Fifty-nine percent of the quarter cases were treated with antimicrobials, and the rest were left without treatment. Mastitis due to beta-lactamase-negative CNS was treated with penicillin G and that due to beta-lactamase-positive CNS with cloxacillin. Nineteen percent of the isolates were beta-lactamase-positive. The bacterial cure rate for quarters treated with antimicrobials was high, 85.9%, as opposed to only 45.5% for untreated quarters. Bacterial cure rates for the most common CNS species or AFLP clusters were not statistically different. Further studies on identification of CNS species are needed.  相似文献

The aim of the present study was to assess the haemolytic and proteolytic activity of coagulase-negative staphylococci (CNS) isolated from cows with mastitis. The study was conducted on 100 CNS strains: S. xylosus (n=28), S. chromogenes (n=26), S. haemolyticus (n=25), S. sciuri (n=14), S. warneri (n=4), S. hominis (n=2), S. saprophyticus (n=1); 22 CNS were isolated from cows with clinical mastitis and 78 from those with subclinical mastitis. The CNS studied showed the ability to produce only alpha-haemolysin and belonged to one strain - S. haemolyticus (21.0% of isolated CNS strains). Haemolysin-positive CNS were responsible for both clinical and subclinical mastitis (22.7% and 20.5%, respectively). The ability to produce protease was found in 31.0% of CNS belonging to two strains: S. chromogenes and S. sciuri. Protease-positive CNS were the etiological factor of both clinical and subclinical mastitis (31.8% and 30.8%, respectively). All S. xylosus, S. warneri, S. hominis, and S. saprophyticus strains were found protease-negative and haemolysin-negative, irrespective of whether they caused clinical or subclinical mastitis in cows.  相似文献
