The isolated infected hair model is a commonly used technique to test the fungicidal efficacy of topical therapies against Microsporum canis. The most commonly used model uses mats of infective hairs, and results from various laboratories have differed. The objectives of this study were to develop a method to produce spores for testing when only mycelial forms were available and to develop a semiquantitative testing method that used only infective spores from hairs, and not pooled hair samples for testing. Ten isolates of M. canis were used in this study. Juvenile guinea pigs were easily infected using mycelial forms of M. canis and large numbers of spores were easily harvested for testing. Eight dilutions of disinfectants were tested. Fungal culture data were evaluated using an endpoint dilution at which there was 100% fungicidal activity, i.e. no growth on the plates. The 10 samples showed identical results. Chlorhexidine and Virkon(R) S were ineffective even when used at x4 the manufacturer's recommended dilution. Lime sulphur (1 : 33), enilconazole (20 microL mL(-1)), and bleach (1 : 10) were consistently effective when used at the recommended dilution. In addition, lime sulphur and enilconazole were 100% fungicidal even when the recommended concentration was diluted 1 : 4 or x4 as dilute as recommended.  相似文献

Six Yorkshire terrier dogs with generalized, chronic dermatophytosis caused by Microsporum canis were seen over a 3-year period. Specific tests showed that they also had concurrent leishmaniosis (four cases), leishmaniosis and ehrlichiosis (one case) or diabetes mellitus (one case). Although specific therapy for these infectious diseases was instituted and the dogs were treated systemically and topically with appropriate antifungal drugs, only partial clinical resolution of the dermatophytosis was achieved. M. canis infection resolved in the dog with diabetes mellitus after stabilizing the diabetes mellitus. Although immunological studies were not performed in these cases, it is theorized that the immune disregulation caused by leishmaniosis, ehrlichiosis or diabetes mellitus may have favoured generalization of the infection and prevented favourable responses to appropriate treatment of the M. canis infection.  相似文献

Oral lufenuron is reportedly an effective treatment for some cats with dermatophytosis. The purpose of this study was to determine if lufenuron, when used as a pre-treatment prior to challenge exposure, would be protective against the development of infection after the direct topical application of fungal macrocondia (Microsporum canis spores). Three groups (n = 6/group) of juvenile cats were treated with either monthly oral lufenuron (30 or 133 mg/kg) or placebo. After 2 months of treatment, kittens were challenged using 10(5)Microsporum canis spores applied to the skin under occlusion. Cats were examined weekly and the following data collected: Wood's lamp examination; scoring for scale/crust, erythema and induration; lesion size; and the development of satellite lesions. Fungal cultures were performed bi-weekly. All cats became infected; the infections progressed, and then regressed, in a similar fashion in all groups. There were no consistent statistically significant differences in weekly infection scores between treated and untreated cats throughout the study. Treated cats did not recover faster than untreated cats. We conclude that oral lufenuron at the dosing schedule and conditions used in this study did not prevent dermatophytosis or alter the course of infection by direct topical challenge.  相似文献

Multiple cutaneous xanthomas, associated with fasting hyperlipidaemia, are described in a 9-month-old domestic long-haired cat. A severely pruritic, papular, and crusting dermatitis affecting the head and neck, initially diagnosed as lesions of the eosinophilic granuloma complex, progressively developed on the head and pinnae. Pruritus was controlled with administration of prednisolone and chlorambucil. Repeat histological examination confirmed the diagnosis of cutaneous xanthoma and concurrent mild demodicosis. Marked fasting hypercholesterolaemia, hypertriglyceridaemia and transient hyperglycaemia were subsequently confirmed. Treatment for hyperlipidaemia and xanthomas with a low-fat diet (Hill's Feline r/d) and the previously unreported treatment for feline demodicosis of daily oral milbemycin were commenced. Multiple pink, alopecic plaques and papules gradually regressed, however pruritus recurred if immunosuppressive treatment was reduced, and well-demarcated areas of alopecia developed on the head, limbs and trunk, despite negative skin scrapings for demodex mites. Fungal culture of hair samples yielded Microsporum canis. All cutaneous lesions resolved with the addition of griseofulvin to the treatment regimen. Concurrent corneal ulceration and keratoconjunctivitis sicca ultimately resolved with treatment, including topical cyclosporin. Diabetes mellitus developed 6 months after resolution of skin lesions. No cutaneous or ocular abnormalities were present 6 months later with continued low-fat diet and insulin administration, although transient recurrence of papules and pruritus occurred after inadvertent access to a fatty meal. An underlying primary hyperlipidaemia was suspected, causing pruritic xanthomas. This may represent the first report of concurrent cutaneous xanthomas, demodicosis and dermatophytosis in a cat.  相似文献

Abstract— Fourteen sera from dogs naturally or experimentally infected with Microsporum canis were examined by enzyme-linked immunosorbent assay (ELISA) in order to study the humoral response to infection. A group of 12 animals (7 laboratory dogs and 5 dogs from the natural environment) was used as controls. ELISA optical density (OD) values in twofold serial dilutions from 1:100 to 1:51200 were compared between the groups. Significant antibody response to infection with M. canis was observed in naturally and experimentally infected animals and confirmed by threshold determination. Specificity for the dermatophyte fungi was tested by performing cross-reactivity assays with three fungal antigens, Malassezia pachydermatis, Candida albicans and Aspergillus fumigatus. There were no statistically significant indications of cross-reactions, except with anti-A. fumigatus IgG, where differences were noticed within the groups of naturally infected animals and controls. The use of ELISA technique in sero-diagnosis of dermatophytosis in dogs is discussed. Résumé— 14 sérums chiens infectés naturellement ou expérimentalement par Microsporum canis ont été testés par une technique Immunoenzymatique (ELISA) pour étudier la réponse immunitaire humorale à cette infection. Le groupe témoin était composé de 12 animaux (7 chiens de laboratoire et 5 chiens d'un milieu naturel). Les valeurs de densité optique (DO) obtenues avec des dilutions sériées de raison 2 des sérums (1:100 à 1:51,500) ont été comparées entre chaque groupe. Des taux significativement élevés d'anticorps spécifiques de Microsporum canis ont été retrouvés chez les animaux infectés naturellement et expérimentalement et confirmés par l'établissement d'un seull de positivité. La spécificité de la technique vis-à-vis des dermatophytes a été appréciée en effectuant des tests d'antigénicité croisée avec des Malassezia pachydermatis, Candida albicans et Aspergillus fumigatus. II n'y avait pas de réactions croisées significatives, exception faite des IgG spécifiques d'A. fumigatus, pour lequel il existait des differences de réponse entre les animaux infectés naturellement et le groups témoin. L'utilisation d'une technique ELISA dans le diagnostic des dermatophyties canines est discutées. Zusammenfassung— Vierzehn Seren von Hunden, die natürlich oder experimentell mit Microsporum canis infiziert waren, wurde mit dem ELISA-Verfahren (Enzyme Linked Immunosorbent Assay) untersucht, um die humorale Antwort auf die lnfektion zu verfolgen. Eine Gruppe von 12 Tieren (7 Laborhunde und 5 Hunde aus normaler Haltung) dienten als Kontrolle. ELISA-optische Dichtewerte (OD) in zweifacher serieller Verdünnung von 1:100 bis 1:51200 wurden zwischen beiden Gruppen verglichen. Bei den natürlich und experimentell infizierten Tieren konnte eine signifikante Antikörperreaktion auf die lnfektion mit M. canis beobachtet und durch Schwellenwertbestimmung bestätigt werden. Die Spezifität für Dermatophyten wurde überprüft, indem Kreuzreaktionsproben mit drei Pilzantigenen, Malassezia pachydermatis, Canidida albicans und Aspergillus fumigatus, durchgeführt wurden. Statistisch signifikante Anzeichen für Kreuzreaktionen konnten nicht nachgewiesen werden, außer bei Anti-A. fumigatus-IgG, bei dem Unterschiede innerhalb der Gruppe der natürlich infizierten und der Kontrolltiere beobachtet wurden. Es wird über die Brauchbarkeit der ELISA-Technik für die Serodiagnose von Dermatophyten bei Hunden diskutiert. Resumen Se examinaron un total de 14 sueros procedentes de perros infectados de forma natural y artificial con Microsporum canis por el método de enzima ligado de inmunoabsorbencia, (ELISA), con el fin de estudiar la respuesta humoral a la infección. Como controles, se usaron un grupo de 12 animales (7 perros de laboratorio y 5 perros que vivían en condiciones naturales). Los valores de densidad óptica ELISA, (OD), fueron comparados entre los grupos en dos diluciones seriadas de 1:100 a 1:51,200. Una respuesta significativa de anticuerpos a la infección con Microsporum canis fue observada en perros infectados de forma natural y artificial, y confirmada por la determinación del punto crítico. La especificidad del dermatofito füngico se estudió por medio de ensayos de reactividad cruzada con otros tres antigenos fúngicos, Malassezia pachydermatis, Candida albicans y Aspergillus fumigatus. No se encontraron indicaciones de reacciones cruzadas estadísticamente significativas, excepto con la IgG de A. fumigatus, donde se advirtieron diferencias en el group de animales infectados naturalmente y en el grupo control. An agar dilution technique was used to assess the minimum inhibitory concentrations (MIC) of miconazole, chlorhexidine and a 1:1 combination of both agents for 10 isolates of Microsporum canis. For nine of 10 of the isolates, a combination of miconazole and chlorhexidine was more effective than either agent alone; fractional inhibitory concentration indices indicated a synergistic effect for five isolates and an additive effect for four. These results illustrate the potent antimycotic effect of miconazole and chlorhexidine against M. canis and are in accordance with previous clinical studies that showed the value of using miconazole and chlorhexidine shampoo in association with oral griseofulvin in the treatment of feline dermatophytosis caused by M. canis.  相似文献

A 2-year-old intact female Australian Cattle Dog presented with a 1-cm diameter nonexudative dermal nodule on the medial aspect of the right thigh. Fine-needle aspiration revealed pyogranulomatous inflammation and many ovoid, 2-4 microm diameter, thin-capsulated, basophilic bodies that appeared to be fungal spores or yeast. Results of CBC, serum chemistry panel, lymph node palpation, and radiographs were unremarkable. Excisional biopsy and histopathology revealed pyogranulomatous folliculitis, furunculosis, and perifolliculitis. Rare fungal hyphae and spore forms were intimately associated with, and occasionally within, hair shafts. A morphologic diagnosis of dermatophytosis was made and Microsporum canis infection was confirmed by fungal culture. M canis is a common infectious agent found within the follicles and superficial keratin layers of canine skin. The kerion-type clinical presentation observed in the dog of this case is uncommonly observed with M canis. Additionally, the cytologic finding of multiple arthroconidia without hyphae is unusual. In the absence of hyphae, M canis arthroconidia may be confused with other fungal yeast bodies; therefore close scrutiny of a cytologic sample for arthroconidia associated with keratin, hair fragments, or hyphal structures is recommended.  相似文献

A Microsporum canis recombinant 31.5 kDa keratinase and a M. canis crude exo-antigen were tested as vaccines in an experimental infection model in guinea pigs. Animals were vaccinated subcutaneously three times at two-week intervals with either the keratinase, the exo-antigen or the adjuvant alone. Cutaneous challenge was performed blindly. Both humoral and cellular-specific immune responses to M. canis antigens were evaluated every 14 days, while a blind evaluation of clinical lesion development and fungal persistency in skin were monitored weekly. Vaccination induced very high and significant (P < 0.01) antibody responses towards both antigens. High cell-mediated immune responses to both immunogens were also induced by vaccination. After challenge, however, scores reflecting the severity of dermatophytic lesions did not differ significantly between vaccinated and control groups at any time after challenge. These results suggest that, in the guinea pig, the induction of specific immune responses against the M. canis-secreted antigens used in this study are not protective against challenge exposure.  相似文献

Multiple, dermal and subcutaneous nodules developed in a young female Manchester Terrier dog that had a chronic history of superficial dermatophytosis. Skin biopsy specimens of the nodules revealed granulomatous inflammation in the deep dermis and subcutis with branching fungal organisms. Cultures of multiple biopsy specimens from the nodules all yielded Trichophyton mentagrophytes. The lesions in this dog were similar to granulomatous dermatophytosis, a skin disease that has been reported in Persian cats and one Yorkshire Terrier dog.  相似文献

A male ferret, which was purchased from abroad at 9 months of age, had shown significant weight loss starting at 13 months of age. The ferret subsequently showed decreasing motor activity and recumbency and was euthanized at 14 months of age. At necropsy, a white, quail egg-sized mass was found in the mesentery. Histopathologically, multifocal granulomas consisting of necrotic foci, macrophages, fibroblasts and plentiful fibrous connective tissues were observed in the mesenteric mass. Surrounding the granulomas, inflammatory cell infiltration consisting of neutrophils, lymphocytes and plasmacytes was observed diffusely and significantly. Immunohistochemistry revealed small numbers of macrophages around necrotic foci that were positively stained for anti-mouse feline coronavirus. Electron microscopically, the cytoplasm of the macrophages contained viral particles, which were identified as coronavirus. The histopathological features in this ferret were similar to those in cats with feline infectious peritonitis (FIP). This was the first case in ferrets in Japan.  相似文献

The performance of the dermatophyte test medium (DTM) RapidVet-D was assessed using hair samples collected from experimentally infected guinea pigs. Three dermatophyte species were included in the study: Microsporum canis , Trichophyton mentagrophytes and Trichophyton equinum . DTM substrates were inoculated with infected hairs and scales, incubated at 18, 21, 24, 27 or 37 °C and examined daily for 15 days. The rapidity of colour change was clearly related to the incubation temperature and to the number of infected hairs deposited on the reactive substrates. With the optimum incubation temperature 27 °C, a systematic colour change could be observed only a few days post-inoculation: 3 days with M. canis infected hairs, 4 days with T. equinum and 5 days with T. mentagrophytes .  相似文献

This report describes a case of an unusually extensive and invasive fungal granuloma within the right guttural pouch and its surrounding area of a 2-year-old Noriker gelding, causing diverse neurogenic deficits and tissue destruction. The gelding was initially presented with cachexia, unilateral nasal discharge, intermittent fever, swelling of the right side of the head, facial nerve paralysis and dysphagia. The right guttural pouch was not accessible to endoscopic examination initially, but after anti-inflammatory medical management, empyema and an extensive, solid mass appeared within the guttural pouch. Surgical exploration and excision were attempted but were unsuccessful. Histopathological examination identified Aspergillus fumigatus as the causative infective agent of the extensive fungal granuloma, and the consequent invasion and destruction of surrounding bone and soft tissue structures.  相似文献

A 6-year-old Persian cat was examined for constipation, anorexia, and vomiting that was subsequently found to be due to a pseudomycetoma originating from the descending colon and sublumbar region, and causing mechanical obstruction of the colon and rectum. Multiple discrete hyperechoic foci likely representing fungal grains within the lesion gave the mass a coarse echotexture on ultrasound and was supportive of the diagnosis and computed tomography allowed delineation the extent of the mass. A pseudomycetoma is a granulomatous/pyogranulomatous reaction that surrounds dermatophytic fungal hyphae. Definitive diagnosis of a dermatophytic pseudomycetoma requires identification of the etiologic agent by cultivation or immunohistochemical staining. A pseudomycetoma should be included in the differential diagnosis for an abdominal mass in a Persian cat, especially is accompanied by the sonographic findings noted above.  相似文献

The recent literature on the treatment of dermatophytosis in dogs and cats was reviewed. Based upon in vitro studies using isolated infected hairs and controlled or field in vivo studies, the following topical treatments were consistently found to be antifungal (i.e. antidermatophyte): lime sulfur (1:16), 0.2% enilconazole rinses, and a combined 2% miconazole/chlorhexidine shampoo. Animals or hairs were either bathed or rinsed once or twice weekly. Itraconazole, griseofulvin and terbinafine were evaluated in controlled or field studies, most commonly involving cats. Griseofulvin (50 mg kg(-1)) was reported to cure infected animals in 41-70 days. Itraconazole (10 mg kg(-1) once daily or in a combined daily/pulse therapy 10 mg kg(-1) once daily for 28 days and then week on/week off) was reported to cure infected animals in 56-70 days. Low-dose itraconazole (1.5-3.0 mg kg(-1)) in 15-day cycles required 1-3 cycles (15-45 days). Various doses of terbinafine (5-40 mg kg(-1)) were reportedly used to treat dogs or cats. The higher doses of terbinafine (> 20 mg kg(-1)) were required to achieve a mycological cure; the number of treatment days to cure varied from 21 to > 126 days. Lufenuron was reported anecdotally to be an effective cure, however, this was not substantiated in controlled studies. Finally, fungal vaccines were not found to be effective against challenge exposure, however, there is evidence that they may be useful in treatment protocols.  相似文献

Résumé— Le but de cette étude était de déterminer la prévalence des isolements de Microsporum canisà partir de chats apparemment sains dans des refuges de deux régions de climats différents aux Etats-Unis. Deux cents chats (chats sans signes apparent de maladie) provenant d'une région froide et séche et d'une région chaude et humide ont étéétudiés et la technique de la brosse à dents utilisée pour la mise en culture et la rechehe de Dermatophytes. Cinq espèces de Dermatophytes communément associées aux dermatophyties humaines et animales ont été isolées: M. canis (n= 8), Trichophyton verrucosum (n= 5), T. mentagrophytes (n - 1), T. rubrum (n= 16), et Epidermophyton spp. (n= 5). Microsporum canis n'a été isolé que chez les chats vivant dans le Sud et principalement sur des chatons. Trichophyton rubrum a surtout été isolé chez les chats du nord. La prévalence de M. canis chez les chats sains était dans cet étude de 4 pourcent et la prévalence totale de tous les pathogénes de 17,5 pour cent. Zwei hundert Katzen (Katzen ohne makroskopische oder offenscichtliche physische Anzeichen von Krankheit) aus Tierheimen in einer relativ kalt-trockenen geographischen Region und einer feucht-war-men Region der Vereinigten Staaten wurden untersucht und aus dem Haarkleid mittels Zahnbürstentechnik Pilzkulturen angelegt. Es wurden fünf Arten von Dermatophyten identifiziert, die häufig mit menschlichen oder tierischen Hautpilzerkrankungen verbunden sind: M. cams (n= 8), Trichophyton verruscosum (n= 5), T. mentagrophytes (n= 1), T. rubrum (n= 16) und Epidermophyton spp. (n= 5). Microsporum canis wurde nur von Katzen aus dem Süden isoliert, vorwiegend von Katzenwelpen. Trichophyton rubrum wurde häufiger von Katzen aus dem Norden isoliert. Five species of dermatophytes commonly associated with either animal or human dermatophytosis were identified: M. canis (n= 8), Trichophyton verruscosum (n= 5), T. mentagrophytes (n= 1), T. rubrum (n= 16), and Epidermophyton spp. (n = 5). M. canis was isolated only from cats in the south, predominantly from kittens. T. rubrum was isolated more commonly from cats from the north. The prevalence of M. canis in apparently healthy cats in this study was 4 per cent and the overall prevalence of all pathogens was 17.5 per cent.  相似文献

Objective  To describe a case of a conjunctivo-corneal mass in a cat associated with acid-fast bacilli.
Methods  A 2-year-old female black European Short-Hair cat, living outdoors in a suburban environment in Italy, was referred for evaluation of a nodular, vascularized mass of 2 weeks duration. The mass involved the dorsal bulbar conjunctiva at the temporal canthus of OS and invaded the sclera and cornea. Routine ophthalmic and systemic examination, serologic testing, cytology and histology of the mass were performed. Mycobacterium specific polymerase chain reaction (PCR) of variable regions 1, 2 and 3 of the 16S ribosomal RNA (rRNA) gene was also performed.
Results  Neutrophils, lymphocytes, macrophages and giant cells with intracytoplasmic acid-fast bacilli were seen on cytological examination. The histological examination confirmed the presence of a granulomatous lesion with acid-fast bacilli within macrophages. Bacteriological culture of the material from the lesion was negative for Mycobacterium spp. Mycobacterium 16S rRNA gene specific PCR was positive.
A diagnosis of feline leprosy was made. The owners refused any treatment, and 1 year later the lesion was still present.
Conclusions  Veterinary ophthalmologists should be aware of conjunctivo-corneal leproma as an unusual symptom of leprosy.  相似文献
