<正>褐条病是甘蔗生长中后期叶部主要真菌病害之一。近年来因多雨高湿及感病品种大面积种植导致褐条病在我国第二大糖料基地云南暴发为害成灾,减产减糖严重[1],严重制约云南蔗糖产业的高质量发展。以病原菌研究为基础的种植和选育抗病品种是防治这种病害最经济有效的措施。有关云南蔗区褐条病病原菌研究甚少,有效防控依据缺乏。本研究从云南不同蔗区采集褐条病病样进行分离与鉴定,旨在明确病原菌种类,以期为甘蔗褐条病的流行预测及综合防治提供科学依据。  相似文献

Sugarcane orange rust, a disease caused by Puccinia kuehnii, was first reported in Brazil in 2009. There are no studies comparing the Brazilian P. kuehnii collections and the reaction of important sugarcane varieties under controlled conditions. This work compared the reaction of seven sugarcane varieties inoculated with six different P. kuehnii isolates from Brazilian sugarcane areas and verified the pathogenic and genetic variability of these isolates. The incubation (I) and latency (L) disease periods, disease severity (SEV), total number of lesions (TNL), total number of sporulating lesions (TNSL), and percentage of sporulating lesions (%SL) were evaluated. Furthermore, ITS1 and IGS ribosomal sequences of all P. kuehnii isolates used in this study were compared with pathogen sequences from 13 different countries. The disease incubation ranged from 7 to 10 days and the latency ranged from 10 to 21 days. SEV and TNL showed large variations and few significant differences between the reaction of the varieties to P. kuehnii, in contrast with the variables TNSL and %SL. The P. kuehnii isolates did not compose different virulent races, but the isolate from one site (Araras) was a more aggressive race. The ITS1 and IGS ribosomal sequences of six P. kuehnii isolates were identical with each other and to most P. kuehnii American sequences deposited at GenBank. The studied sequences of P. kuehnii isolates differed from the sequences from Asia, Tahiti and Oceania.  相似文献

Puccinia kuehnii and P. melanocephala cause orange and brown rust of sugarcane, respectively. Puccinia kuehnii has been confirmed in Asia, Australia and recently, the Caribbean basin, whereas P. melanocephala is distributed among the majority of sugarcane growing regions. Differentiating these two economically significant pathogens visually is problematic and limited to material exhibiting mature disease symptoms or spores. Partial ITS1, ITS2 and complete 5·8S sequences were generated from P. kuehnii and P. melanocephala isolates from around the world. PCR primers and dual labelled hydrolysis probes were designed for each pathogen for use in real‐time PCR and optimized using locked nucleic acids (LNA). The primers amplified DNA from their target pathogens and not from other species of Puccinia or fungal species isolated from sugarcane leaves. Optimized real‐time PCR conditions allowed the detection of 0·19 pg of P. kuehnii or P. melanocephala genomic DNA and differentiated the pathogens on sugarcane leaves prior to observing typical symptoms in the field. Primer‐introduced restriction analysis‐PCR (PIRA‐PCR) was used to detect a single nucleotide polymorphism (Pk ITS1 183A>G) in ITS1 of P. kuehnii. Allele 183A was observed in all samples, whereas 183G was detected in 52% of samples from Asia and Australia yet absent from all Caribbean basin samples. Long distance spore dispersal, dispersal through an intermediate location or improper movement of contaminated material could explain the introduction of P. kuehnii to the Western hemisphere. However, the current proliferation of the pathogen in the Americas is limited to isolates which contain only the 183A allele.  相似文献

为明确我国热带和亚热带地区蚕豆Vicia faba和豌豆Pisum sativum锈病的病原菌种类,通过致病性测定和ITS序列系统发育分析对来自我国云南省玉溪市的4份豌豆锈菌分离物及云南、广西、重庆和四川省(区、市)的5份蚕豆锈菌分离物进行系统鉴定。结果显示,分离自豌豆的锈菌WX1分离物对蚕豆和豌豆均具有高致病性,在侵染叶片上产生大量锈子器;分离自蚕豆的锈菌CX3分离物仅对蚕豆具有高致病性,能在叶片上产生大量夏孢子,而对豌豆的致病性相对较低,仅产生少量的夏孢子堆;分离物WX1和CX3对小扁豆和鹰嘴豆不具有致病性。基于ITS序列系统发育分析表明,所有不同寄主来源的蚕豆单胞锈菌分离物均聚类于一个系统发育组,但分离自蚕豆和豌豆的分离物分别聚类在不同的亚组。表明分离自云南省玉溪市豌豆上的蚕豆单胞锈菌Uromyces viciae-fabae应为豌豆专化型,定名为U. viciae-fabae ex P. sativaum,而来源于云南、广西、重庆和四川省(区、市)的蚕豆锈病病原菌为蚕豆专化型U. viciae-fabae ex V. faba。  相似文献

In 2016-2019, sixty new varieties and 31 main cultivated varieties were evaluated for their resistance to sugarcane brown stripe disease under field condition. The results showed that 32 (53.33%) of 60 new sugarcane varieties were highly to moderately resistant, and 28 (46.67%) were susceptible to highly susceptible. Twenty-one (67.74%) of 31 main cultivated sugarcane varieties were highly to moderately resistant, and ten (32.26%) were susceptible to highly susceptible. This work may facilitate brown stripe resistance breeding and provide new elite resistant varieties for effectively control of sugarcane brown stripe.  相似文献

Wheat stripe rust, caused by Puccinia striiformis f. sp. tritici, is one of the most destructive wheat diseases in China. Yunnan Province, located in south-western China, possesses unique features of geography, climate, wheat growth and stripe rust epidemics, different from main epidemic regions in China. The isolates of this pathogen were collected from nine counties in Yunnan Province during February to May of 2008. Used as a comparison, isolates were also collected from five counties of Gansu Province, the province important in inter-regional stripe rust epidemics in China. Amplified fragment length polymorphism (AFLP) method was applied to study the population genetics of the pathogen among different populations in these two provinces. Forty one AFLP genotypes were obtained from 150 isolates and the genotype qj3 showed the highest frequency in Yunnan Province. While 22 genotypes were detected from 40 isolates, no genotype showing as predominant was identified in Gansu Province. Genotypic diversity in Gansu Province was higher than that in Yunnan Province. A free recombination signature was detected in Gansu Province but not in Yunnan Province. We concluded that the population of P. striiformis in Yunnan Province can be considered as a clonal population.  相似文献

Isolates of an unidentified Rhizoctonia sp. (UR isolates) were obtained from creeping bentgrass and Kentucky bluegrass with reddish brown sheath and foliar rots. Because the UR isolates anastomosed with isolates of three varieties of Waitea circinata (var. oryzae, var. zeae, and var. circinata), colony morphology, hyphal growth rate at different temperatures, pathogenicity, sequence analysis of the internal transcribed spacers (ITS) region of ribosomal RNA genes (rDNA) were compared. The colony color of mature UR isolates was distinct from isolates of the other three varieties of W. circinata. In pathogenicity tests on creeping bentgrass, the severity of the disease caused by UR isolates was significantly higher than that caused by the three varieties of W. circinata. Sequence similarities of the rDNA-ITS region between UR isolates and between isolates within each variety were high (97–100%), but they were lower among isolates from UR and the varieties of W. circinata (88–94%). In a phylogenetic tree based on the rDNA-ITS sequences, UR isolates formed a cluster separate from each of the clusters formed by the three varieties of W. circinata. These results indicate that the UR isolates clearly differ from the three varieties of W. circinata. We therefore propose that the UR isolates be classified as new Rhizoctonia sp. that are closely related to W. circinata and that the disease on creeping bentgrass should be called Waitea reddish-brown patch disease (Sekikasshoku-hagusare-byo in Japanese).  相似文献

Red rot is an important disease of sugarcane, reported from the main producing countries of the crop. The main causal agent is Colletotrichum falcatum, which induces reddish internal stalk rot, resulting in loss of quality and quantity of sugars. While the occurrence of this disease has been reported in plantations in Brazil, the aetiology of the disease is not yet fully understood. We isolated and identified the fungi associated with sugarcane plants showing symptoms of red rot. Thirty isolates were recovered and grouped by morphology within the genera Colletotrichum and Fusarium. Based on phylogenetic analyses of DNA sequences from ITS, ACT, SOD, GAPDH, and EF-, eight isolates were identified as C. falcatum (n = 6) and C. plurivorum (n = 2). Twenty-two isolates belonged to the Fusarium fujikuroi species complex and were identified as F. sacchari (n = 8), F. proliferatum (n = 3), and F. madaense (n = 11). Coinfection with C. falcatum and Fusarium species were frequently observed. C. falcatum and Fusarium strains induced reddening of internal tissues and stalk rot in sugarcane plants, symptoms of red rot, while Fusarium species also induced symptoms of pokkah boeng. C. plurivorum, whose sexual stage was observed on the surface of stems, did not induce stalk rot or leaf symptoms. The results obtained in this study clarify the aetiological agents of red rot of sugarcane in Brazil. The conditions that lead to the development of specific symptoms of red rot or pokkah boeng are under investigation.  相似文献

为明确近年来云南省严重发生的玉米白斑病病原菌及不同玉米品种对玉米白斑病的抗性差异,2021年于云南省昆明市阿子营乡和曲靖市罗平县九龙镇采集病样,进行病斑组织分离纯化,通过形态学特征和分子生物学方法对病原菌进行鉴定,并在云南省曲靖市罗平县九龙镇采用田间自然发病方法对622份玉米品种的抗性进行鉴定。从2个采样地点的病叶中各分离出5株和9株菌株,经柯赫氏法则验证,确定了菌株WS-1、WS-191为白斑病致病菌;经形态学观察以及基于ITS、rpb2、tub2和LSU基因序列构建的系统发育树分析结果确定其分类学种名为宽颈附球菌Epicoccum latusicollum。田间抗病性试验结果表明,在622份区域试验品种中对白斑病表现高抗、抗病、中抗、感病和高感的品种分别为4、184、216、186和32份,分别占品种总数的0.64%、29.58%、34.73%、29.90%和5.14%。  相似文献

为进一步了解田间稻瘟病菌Magnaporthe oryzae群体中AVR-Pia基因的分布及变异,利用水稻单基因系IRBLa-C水稻品种对自云南省13个市(州)采集分离得到的471株稻瘟病菌菌株进行抗性基因Pia有效性测定;利用无毒基因AVR-Pia特异性标记对471株稻瘟病菌菌株进行PCR检测和测序,并分析稻瘟病菌群体中无毒基因AVR-Pia的分布及DNA结构变异;利用有效性结果和PCR检测结果对471株菌株进行反应型划分,筛选鉴定菌株;利用鉴定菌株对云南省112份地方稻种进行Pia基因鉴定。结果表明,在471株稻瘟病菌菌株中,对含有Pia基因的水稻单基因系IRBLa-C表现为抗病和感病的菌株数分别为139株和332株,所占比例分别为29.5%和70.5%;在471株稻瘟病菌菌株中,分别有244株和227株菌株含有无毒基因AVR-Pia和不含有无毒基因AVR-Pia,所占比例分别为51.8%和48.2%,无毒基因AVR-Pia主要为完全缺失变异;在471株稻瘟病菌菌株中,A-和V+反应型菌株数分别为56株和161株,共217株,占总菌株数的46.1%,在13个市(州)稻瘟病菌群体中,A-和V+反应型菌株所占比例差异较大,其中在普洱市、红河哈尼族彝族自治州、昭通市、玉溪市4个市(州)的比例较大,分别为77.8%、57.1%、52.1%和50.0%;在112份云南省地方稻种质资源中,有20份地方稻品种含有抗性基因Pia,主要分布在9个市(州)中。表明云南省13个市(州)绝大部分水稻产区水稻Pia基因已丧抗性,含Pia基因的水稻种质在云南省分布较广。  相似文献

Red stripe is a bacterial disease of sugarcane causing important economic losses in Argentina that affects 30 % of the milling stems and consequently the juice quality. In this study, sugarcane leaves exhibiting red stripe symptoms were sampled in the 2008–09 growing season from 13 different sugarcane producing areas of Tucumán and Salta (northwest of Argentina). To achieve the identification and characterization of the causal agent of red stripe, bacterial isolation was performed. Species-specific PCR using Oaf1/Oar1 primers allowed the amplification of a fragment of 550 bp from approximately 50 % of the isolates; 16S rDNA sequences analysis displayed a similarity greater than 99 % with Acidovorax avenae subsp. avenae. By means of RAPD-PCR the presence of at least four different biotypes among the analyzed isolates was detected. Results of pathogenicity test allowed us to confirm A. avenae subsp. avenae as the pathogenic agent for red stripe. This study constitutes the first report on the identification and molecular characterization of this plant pathogen from the Argentina sugarcane production areas. The genetic diversity observed among A. avenae is an important factor to be considered to improve an accurate diagnosis and/or the selection of sugarcane tolerant clones.  相似文献

为明确云南省紫茎泽兰根结线虫病的病原种类,于2019年2月在云南省澜沧县林下三七种植区采集根部带有明显根结的紫茎泽兰根系进行根结线虫分离,通过观察所分离根结线虫的2龄幼虫、雌成虫、会阴花纹特征对其进行形态学鉴定,并利用序列比对、系统发育树分析、序列特异性扩增区段(sequence characterized amplified region,SCAR)对其进行分子生物学鉴定。结果表明,该病原线虫雌成虫会阴花纹呈圆形至卵圆形,背弓中等高或低平,侧区一侧或两侧延伸形成翼状,尾区有刻点,2龄幼虫、雌成虫形态特征及形态测量指标与北方根结线虫Meloidogyne hapla相似;该病原线虫rDNA的ITS序列和mtDNA的COI序列与NCBI数据库中已登录的北方根结线虫相应序列相似度较高,分别达99.35%和98.05%以上;该病原线虫rDNA的ITS序列、mtDNA的COI序列分别以99%、100%的支持率与北方根结线虫聚为同一分支;利用SCAR特异性引物,该病原线虫均能扩增出大小约1 500 bp的基因特异性条带。综合形态学和分子生物学鉴定结果将云南省紫茎泽兰根结线虫病病原种类鉴定为北方根结线虫。  相似文献

Drechslera leaf spot (DLS) is a serious leaf disease of oats chiefly caused by Pyrenophora chaetomioides in the main oat-producing areas of the world. However, other species of Pyrenophora that may cause DLS have not been investigated in China. From 2018 to 2020, a total of 256 strains of Pyrenophora were isolated from nine oat-producing areas in three provinces in western China. Using a combination of morphological characteristics and phylogenetic analysis of the rDNA internal transcribed spacer (ITS), 28S ribosomal RNA gene (LSU), RNA polymerase second largest subunit region (RPB2), and glyceraldehyde-3-phosphate dehydrogenase region (GAPDH), 66% of the isolates were identified as P. chaetomioides and 34% of isolates belonged to P. avenicola. Pathogenicity tests confirmed that P. avenicola also infected oat and caused DLS, but it was less virulent than P. chaetomioides. From our survey, DLS caused by P. avenicola generally occurred in warm areas at lower altitude and the annual average temperature was above 0°C (Huan and Zhenyuan county in Gansu, Huize county in Yunnan), compared to high-altitude areas (Tianzhu county, Shandan county, Haiyan county). The combination of optimal and maximal temperature indicated that P. avenicola had greater tolerance to high temperature, which was also an important factor for its distribution on oats. Precise identification and understanding of virulence, distribution, and occurrence of these pathogens has significance for control of oat DLS in the future.  相似文献

为了明确我国北方稻区水稻条纹病毒(Rice stripe virus,RSV)群体的分子变异和水稻品种的抗性情况,测定了2004年和2005年从我国6省9个地区采集的34个RSV分离物的外壳蛋白(coat protein,CP)基因的序列,并对其进行了分析,同时用强致病性江苏分离物(RSV-JS)对河南、安徽、山东、河北等省的25个推广品种进行了抗性研究。结果表明,供试RSV分离物间的核苷酸序列一致性和氨基酸序列的一致性分别在93.9%~100%和96.3%~100.0%之间。根据CP基因核苷酸序列一致性,所有分离物可以分为2组。云南4个分离物为一组,其余为一组。在第2组中各分离物CP基因的核苷酸序列和氨基酸序列的一致性与地域无必然联系。且在2年之内,RSVCP基因变异不大。抗病性鉴定表明同一分离物在不同水稻品种上表现不同症状。表现高抗(HR)的品种占供试品种的24%;60%以上的品种表现为感病,且不同水稻品种上表现不同症状。因此,我国北方稻区RSV的CP基因非常保守,但同一分离物在不同水稻品种上可能表现不同症状,不同水稻品种对RSV抗性有显著差异。这些结果为我国北方稻区水稻条纹叶枯病防治和抗病毒基因工程提供了理论依据。  相似文献

Dothistroma pini is one of two pathogens causing Dothistroma needle blight (DNB), a foliar disease of pines. The species was redefined in 2004 and subsequently recorded in several European countries. In Slovakia, the first report of the pathogen was in 2013. In this study, the population structure, genetic diversity, and reproductive mode of 105 isolates collected from 10 localities and seven hosts were determined in Slovakia. Species-specific mating type markers, ITS haplotype determination, and 16 microsatellite markers were used to characterize and genotype the isolates. Overall, 15 unique multilocus haplotypes (MLHs) based on microsatellite markers and three ITS haplotypes were identified. Three independent methods (DAPC, STRUCTURE, EDENetwork) separated the isolates into two distinct population clusters corresponding with ITS haplotypes. A high level of clonality was recorded suggesting that conidia are the primary source of pathogen dispersal. The low genetic diversity, predominantly asexual reproductive mode of the pathogen, and the fact that most isolates were collected from introduced tree species and native species in artificially planted urban greenery, supports the hypothesis that D. pini has been recently introduced into Slovakia.  相似文献

Phylogenetic analysis of sugarcane rusts based on sequences of ITS and the 5.8 S rDNA revealed two highly divergent ITS groups among isolates of Puccinia sp. sensu Muta, 1987 and P. kuehnii specimens. Although there is sufficient divergence (exceeding normal intraspecific variation) between the ITS regions of the two groups to support separation into different species, unusually high homology of the ITS group I sequences with those of members of Cronartium and identical sequences of the D1/D2 regions of the LSU rDNA for all the isolates of “Puccinia sp.” and P. kuehnii that otherwise exhibited different ITS sequences, suggest that the two highly divergent sequences may have resulted from abnormal genetic events leading to non-orthologous, intraspeciflc polymorphisms. The other sugarcane rust, P. melanocephala and the grass rusts, P. miscanthi and P. rufipes, were separated from “Puccinia sp.” and P. kuehnii and from each other in D1/D2 region analyses, indicating that D1/D2 region sequences may more correctly reflect phylogenetic relationships in these rusts than do the ITS regions. Further studies to examine differences in patho-genicity or finer morphological features within P. kuehnii that may be correlated with the high divergence in ITS sequences and experiments to determine if these two sequence types represent intraspeciflc polymorphism are necessary. The Bursaphelenchus genus (Nematoda: Parasitaphelenchidae) comprises mostly wood‐inhabiting nematodes that feed on various tree‐colonizing fungi. One species of the genus, Bxylophilus, has been proven as an agent causing pine wilt disease (PWD). However, involvement of other Bursaphelenchus species in the PWD remains enigmatic. In the current paper, comparative molecular analysis is performed based on nuclear ribosomal DNA (rDNA) of Bvallesianus, a species that was recently isolated from pine trees (Pinus sylvestris) exhibiting wilting and declining symptoms in the Czech Republic. Sequencing of the nuclear‐encoded ITS1–5·8S–ITS2 rDNA region confirmed previous taxonomic conclusions based on morphology. Evolutionary reconstructions resulted in a phylogenetic tree, where the Czech isolate of Bvallesianus occupied a common clade together with other species belonging to the so‐called Bsexdentati group. Unexpectedly, comprehensive analysis of the sequence data revealed a genetic variation distinguishing the Czech isolate of Bvallesianus from all other species of the Bsexdentati group. This dissimilarity consists of the presence of a four nucleotide exchange found in the 5·8S rRNA‐coding gene. The newly identified genetic variation appears to affect the 5·8S rRNA folding, as deduced from secondary structure models. Additionally, it is shown that for the first time, to the authors’ knowledge, both bursaphelenchid internal transcribed spacers (ITS1 and ITS2) fold into the multibranched closed loops. While the ITS2 closed loop is formed with help of canonical 5·8S‐28S rRNA pairing, the ITS1 forms the thermodynamically stable closed loop with no support of flanking rRNA sequences. The current information on bursaphelenchid ITS rDNA sequence diversity and structure is further discussed.  相似文献

In 2010, the outbreak of a disease with symptoms similar to bacterial brown stripe was observed in rice seedlings planted in a perlite culture system in China. The causal bacterium was identified as Acidovorax avenae subsp. avenae based on its biochemical and physiological characteristics, cellular fatty acid composition, Biolog data, specific PCR detection and 16S rRNA gene sequence analysis. The bacterial isolates caused similar symptoms after inoculation of rice seedlings. This report is the first of bacterial brown stripe of rice in a soil-less culture system caused by A. avenae subsp. avenae in China.  相似文献

甘蔗褐锈病是目前中国蔗区发生最普遍、危害最严重的病害之一,选育和种植抗病品种是防治该病最为经济有效的措施。为明确近年国家甘蔗体系育成的50个新品种(系)对甘蔗褐锈病的抗性,确定其应用潜力,本研究通过在甘蔗褐锈病高发蔗区云南德宏、保山2个区域化试验站,采用田间自然抗性调查与分子标记辅助鉴定抗性基因的方法,于2014年和2015年对中国近年选育的50个优良新品种(系)及2个主栽品种进行抗褐锈病基因Bru1的分子检测及自然抗性评价。田间自然发病调查结果表明,50个优良新品种(系)及2个主栽品种中,高抗至中抗的有34个,占65.38%。其中15个材料表现高抗,占28.85%,16个材料表现抗病,占30.77%,3个材料表现中抗,占5.77%;分子检测结果显示,共29个抗病材料含有抗褐锈病基因Bru1,出现频率为55.77%,表明中国近年选育的优良新品种(系)中抗褐锈病性主要由Bru1控制;其余5个抗病材料均不含抗褐锈病基因Bru1,暗示除了Bru1外,可能还有其他抗褐锈病基因存在。不同系列品种田间感病品种的频率和含抗褐锈病基因Bru1的频率不同,粤糖系列品种感病品种的频率最高达到60%,含Bru1的频率最低,只有30%,抗性最弱;云蔗系列品种感锈病品种的频率最低只有12.5%,含Bru1的频率最高,达到81.25%,抗性最好。本研究结果为深入开展甘蔗抗褐锈病育种,选育和推广优良抗病品种,有效防控甘蔗褐锈病提供了科学依据和优良抗性新品种(系)。  相似文献
