In order to determine skeletal muscle and serum enzyme activities following exercise, six adult Landrace pigs were submitted to 10 min running on a treadmill (0.5 m/s, on a 12% gradient) and compared to six controls. Blood samples were obtained just before the exercise, immediately after, and 24 h, 48 h and 144 h after exercise. Muscle biopsies were taken from the longissimus dorsi and biceps femoris 24 h after exercise. Total lactic dehydrogenase (LDH) and LDH isoenzymes in muscle and serum were unchanged. In muscle homogenates, there was no difference in total creatine phosphokinase (CK) activity between the two groups. Total CK activity in the biceps femoris muscle represented only 59% of that observed in the longissimus dorsi muscle. A mean CK-MB value of 2.5% was found in the control group for both muscles but after exercise it was 9.5% (p<0.05) for the biceps femoris and 12.2% (p<0.01) of the longissimus dorsi muscle. In serum, the total CK (p<0.05), CK-MM (p<0.05) and CK-BB (p<0.05) increased immediately after the exercise, followed by a progressive decrease.  相似文献

Ten male lambs of each of Rahmani (R) and Ossimi (O) were used to study the growth and carcass characteristics within their first year of age (360 days). After suckling period (4 months), lambs were fed on the regular ration. During the course of the experiment, body weight (BW) was recorded monthly accompanied by blood sampling to determine IGF-I. Four lambs from each breed were slaughtered at 270 and 360 days of age, and Longissimus lumbarum (L. lumbarum) at the 9th, 10th and 11th ribs and Biceps femoris (B. femoris) muscles were separated to be used for chemical analysis and histological study.Breed had no effect on the growth curve of the two studied breeds. Chemical analysis indicated that (O) had higher fat (0.9%, P < 0.0001) and lower (0.3%, P < 0.01) moisture compared to (R), with no difference in protein and ash contents. The concentration of IGF-I was almost similar in both breeds up to day 270 of age, and then it was higher in (R). Number of fibers per bundle was higher (P < 0.05) in (O) than in (R) by 6.1%, while, the other histological traits were similar in the two studied breeds.Chemical analysis indicated that lambs slaughtered at 360 days of age had lower (0.7%, P < 0.0006) moisture and higher fat (17.3%, P < 0.0001) compared to day 270. Up to day 270 of age, IGF-I concentration was < 500 ng/ml before increasing to > 500 ng/ml up to the end of the experiment. No effect of age was observed on the studied histological traits.L. lumbarum muscle contained higher protein (4.2%, P < 0.0001), fat (104%, P < 0.0001) and ash (10%, P < 0.001), while moisture was less (5.6%, P < 0.0001) compared to B. femoris muscle. B. femoris muscle was higher (P < 0.0001) in number and diameter of fibers, bundle and stroma sectional areas relative to L. lumbarum by 38.2, 78.8, 53.2 and 23.8%, respectively.  相似文献

This study was conducted to evaluate the effect of reduced dietary protein level on growth performance, muscle mass weight, free amino acids (FAA) and gene expression profile of selected amino acid transceptors in different fibre type of skeletal muscle tissues (longissimus dorsi, psoas major, biceps femoris) of growing pigs. A total of 18 cross‐bred growing pigs (Large White × Landrace × Duroc) with initial body weight (9.57 ± 0.67 kg) were assigned into three dietary treatments: 20% crude protein (CP) diet (normal recommended, NP), 17% CP diet (low protein, LP) and 14% CP diet (very low protein, VLP). The results indicated improved feed‐to‐gain ratio was obtained for pigs fed LP and NP diets (p < 0.01), while the pigs fed VLP diet showed the worst growth performance (p < 0.01). There was no significant difference in the weights of longissimus dorsi and psoas major muscle between LP and NP groups (p > 0.05). Majority of the determined FAA concentration of LP group were greater than or equal to those of NP group in both longissimus dorsi and psoas major muscle (p < 0.01). Further, the mRNA expression levels of sodium‐coupled neutral amino acid transceptor 2, L‐type amino acid transceptor 1 and proton‐assisted amino acid transceptors 2 were higher in skeletal muscle tissue in LP group compared to those of the pigs fed NP or VLP diet. These results suggested that reduced dietary protein level (3 points of percentage less than recommended level) would upregulate the mRNA expression of amino acid transceptors to enhance the absorption of FAA in skeletal muscle of growing pigs. There seems to be a relationship between response of AA transceptors to the dietary protein level in skeletal muscle tissue of different fibre type. To illustrate the underlying mechanisms will be beneficial to animal nutrition.  相似文献

The aim of the study was to investigate the effect of duration of feeding linseed (rich in n-3 PUFA) on peroxisome proliferator-activated receptor γ (PPARγ) and tumor necrosis factor (TNF-α) gene expression, and muscle mass of growing–finishing barrows. Two isoenergetic and isonitrogenous diets were formulated, and one of which was the basal diet and another one was the linseed diet including linseed at the level of 10%. Twenty-four Landrace × Yorkshire barrows weighing 35 ± 3.7 kg were randomly assigned to four treatments with six individuals per treatment. Pigs in treatment 1 (T1) fed the control diet throughout the experimental period, while pigs in T2, T3 and T4 fed the control diet except for 30, 60, and 90 d prior to slaughter when the linseed diet were fed. The experiment was conducted for 90 days. The longissimus muscle mass and each muscle mass in the hind leg were weighted. PPARγ and TNF-α mRNA expression levels in muscle, spleen and adipose tissue, and plasma concentrations of TNF-α data were measured and analyzed. The results showed that the longissimus muscle mass, quadriceps femoris muscle mass and semitendinosus muscle mass increased linearly (< 0.01) as prolonged the time of feeding linseed diet. The expression of PPARγ in longissimus muscle and spleen increased (< 0.01) linearly as prolonged the time of feeding linseed diet, while the expression of PPARγ in adipose tissue were not affected (= 0.095). Duration of linseed addition linearly decreased (< 0.01) TNF-α gene expression levels in the longissimus dorsi muscle, adipose and spleen, and serum concentration of TNF-α as well. The expression levels of PPARγ negatively correlated with the expression of TNF-α in muscle (R2 = 0.70, < 0.001) and spleen (R2R2 = 0.77, < 0.001) respectively. Likewise, PPARγ expression level in spleen (R2R2 = 0.59, < 0.01) or muscle (R2R2 = 0.52, < 0.05) negative correlated with serum TNF-α concentration, while there were significant quadratic relation between muscular PPARγ (R2R2 = 0.80, < 0.01) or muscular TNF-α (R2R= 0.87, < 0.01) expression and the longissimus dorsi muscle mass. These data suggested that duration of feeding linseed diet lead to a linear decrease of TNF-α gene expression, which may increase the muscle mass in growing–finishing barrows, at least in part, through a PPARγ-dependent mechanism.  相似文献

We investigated the effects of long‐term microalgae supplementation (7% in a piglet diet and 5% in a fattening diet) on muscle microstructure and meat quality, including fatty acid composition in female Landrace pigs (n = 31). The major effects were muscle‐specific increases in n‐3 and n‐6 polyunsaturated fatty acids (PUFA) concentrations, resulting in increased accumulation of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA). Carcass traits and meat quality of longissimus thoracis muscle were not affected by the microalgae diet with the exception of reduced drip loss (p = 0.01) and increased protein proportion (p = 0.04). In addition, the microalgae diet resulted in a shift to a more oxidative myofibre type composition in semitendinosus but not longissimus thoracis muscle. In conclusion, microalgae supplementation offers a unique opportunity to enhance essential n‐3 PUFA contents in pig meat. The results support small but coordinated changes in skeletal muscle phenotypic appearance and functionality.  相似文献

The viability of a fungal formulation using the nematode-trapping fungus Duddingtonia flagrans was assessed for the biological control of horse cyathostomin. Two groups (fungus-treated and control without fungus treatment), consisting of eight crossbred mares (3–18 years of age) were fed on Cynodon sp. pasture naturally infected with equine cyathostome larvae. Each animal of the treated group received oral doses of sodium alginate mycelial pellets (1 g/(10 kg live weight week)), during 6 months. Significant reduction (p < 0.01) in the number of eggs per gram of feces and coprocultures was found for animals of the fungus-treated group compared with the control group. There was difference (p < 0.01) of 78.5% reduction in herbage samples collected up to (0–20 cm) between the fungus-treated group and the control group, during the experimental period (May–October). Difference of 82.5% (p < 0.01) was found between the fungus-treated group and the control group in the sampling distance (20–40 cm) from fecal pats. During the last 3 months of the experimental period (August, September and October), fungus-treated mares had significant weight gain (p < 0.01) compared with the control group, an increment of 38 kg. The treatment with sodium alginate pellets containing the nematode-trapping fungus D. flagrans reduced cyathostomin in tropical southeastern Brazil and could be an effective tool for biological control of this parasitic nematode in horses.  相似文献

This study was conducted to evaluate the effect of mulberry leaves as an alternative source of protein on growth performance, carcass traits and meat quality in finishing pigs. A total of 180 Xiangcun Black pigs were randomly assigned to five treatment groups with six pens of six pigs per pen. The pigs were provided with a basal diet or a diet contained 3%, 6%, 9% or 12% of mulberry leaf powder during a 50‐day experiment period. The results showed that dietary mulberry leaf powder had no negative effect on growth performance in Xiangcun Black pigs, except in the 12% mulberry group, where final body weight and average daily gain decreased (p < .05) and feed to gain ratio of the pigs increased (p < .05). Dietary mulberry inclusion decreased (quadratic, p < .05) the back fat thickness, fibre mean cross‐sectional area (CSA) in the longissimus dorsi (LD) muscle and mRNA expression levels of myosin heavy chain (MyHC) IIb in LD and biceps femoris (BF) muscles, while increased (linear or quadratic, p < .05) the plasma concentration of albumin, levels of crude protein (CP), inosine monophosphate (IMP) and several amino acids in muscle tissues. When compared with the other groups, the 9% mulberry diet increased (p < .05) loin‐eye area and contents of CP and IMP in muscles, while decreased (p < .05) plasma activity of cholinesterase and concentrations of uric acid and urea. The 6% mulberry diet had the lowest fibre mean CSA and shear force and increased total fibre number of the LD muscle, when compared with the other groups. These results suggest that including mulberry in the diet at <12% is an effective feed crop to improve meat quality and the chemical composition of muscle without negatively affecting growth performance.  相似文献

The effect of transportation during the hot season (42 °C) and low voltage electrical stimulation on physiological, histochemical and meat quality characteristics of Omani sheep was studied. Forty intact male sheep were divided into two equal groups: 3 h transported or non-transported. The non-transported group remained in holding pens for 48 h prior to slaughter, while the transported group was transported 300 km (approximately 3 h) in an open truck under solar radiation on the day of slaughter. Blood samples were collected from the animals before loading and prior to slaughter. Fifty percent of the carcasses from each group were randomly assigned to low voltage (90 V) at 20 min postmortem. Temperature and pH decline of the left longissimus thoracis muscle were monitored. Ultimate pH, WB-shear force, sarcomere length, myofibrillar fragmentation index (MFI), expressed juice, cooking loss and color L, a, b were measured on samples from both sides muscles collected at 24 h postmortem at 3–4 °C. The transported sheep had significantly higher plasma cortisol (P < 0.01), adrenaline, nor-adrenaline and dopamine concentrations (P < 0.05) than non-stimulated animals. Electrical stimulation resulted in a significantly (P < 0.05) more rapid pH fall in muscle during the first 12 h after slaughter. Muscles from electrically-stimulated carcasses had significantly (P < 0.05) lower pH values, longer sarcomere length, lower shear force value, higher expressed juice, MFI and lighter L than those from non-stimulated ones. The muscle samples from the transported sheep had significantly (P < 0.05) smaller and lower proportion of Types I and IIA fibers than those from the non-transported group. This study indicated that pre-slaughter transport at high ambient temperatures can cause noticeable changes in physiological and muscle metabolism responses in sheep. Electrical stimulation improved meat quality characteristics, which indicate that meat quality of transported sheep can be improved by electrical stimulation.  相似文献

Sixty crossbred (Iberian dam × Duroc sire) females, 80 days of age (17.6 ± 0.13 kg body weight, BW), was used to investigate the effect of castration on productive performance, carcass and meat quality and fatty acid profile of backfat (BF). There were 2 treatments (intact females, IF; castrated females, CF) and 5 replicates of 6 pigs per treatment. Pigs were reared indoor under an intensive production system, ovariectomized at 92 days of age (26.1 ± 0.19 kg BW) and slaughtered at 267 days of age (143.6 ± 6.49 kg BW). Meat samples were taken at longissimus dorsi muscle at the level of the last rib and BF samples were taken at the tail insertion. For the entire experiment (18 to 144 kg BW), IF ate less feed and were more efficient than CF (P < 0.05). Also, IF had less carcass yield (P < 0.01) and fat thickness at the gluteus medius muscle (P < 0.05) and tended to have lower backfat depth (P < 0.10) than CF. However, IF had higher shoulder yield at 2 and at 24 h post mortem (P < 0.05) and after trimmed (P < 0.10) than CF. The pH24 of the semimembranosus muscle tended to be lower for IF than for CF. Also, IF had more moisture (710 vs. 691 g/kg) and less fat (66.4 vs. 91.2 g/kg) in the longissimus dorsi muscle than CF (P < 0.05). Meat from IF was more lightness (higher L value; P < 0.01), redder (higher a value; P < 0.001) and had more intensive color (higher c value; P < 0.001) than meat from CF. Backfat was more saturated in CF than in IF (P < 0.05), mostly because of the higher palmitic acid (P < 0.05) and the lower linolenic acid (P < 0.05) content. We conclude that intact females have better productive performance and shoulder yield but less carcass yield than castrated females and that castration does not improve meat quality. Therefore, when animal welfare, cost of castration, productive performance and carcass and meat quality traits are considered, the use of intact females rather than castrated females is recommended for the production of Iberian pigs reared under intensive management systems.  相似文献

The aim of the present study was to carry out molecular epidemiological investigation on enterotoxigenic Escherichia coli (ETEC) K99 and Salmonella spp. in diarrheic neonatal calves. Fecal samples were obtained from 220 diarrheic calves at 9 farms related to four governorates in central and northern Egypt. E. coli and Salmonella spp. isolates were examined for E. coli K99 and Salmonella spp. using PCR. ETEC K99 was recovered from 20 (10.36 %) out of 193 isolates, whereas Salmonella spp. was recovered from nine calves (4.09%).Multivariable logistic regression was used to evaluate the risk factors associated with both infections. ETEC K99 was significantly affected by age (P < 0.01; OR: 1.812; CI 95%: 0.566–1.769), colostrum feeding practice (P < 0.01; OR: 5.525; CI 95%: 2.025–15.076), rotavirus infection (P < 0.001; OR: 2.220; CI 95%: 0.273–1.251), vaccination of pregnant dams with combined vaccine against rotavirus, coronavirus and E. coli (K99) (P < 0.001; OR: 4.753; CI 95%: 2.124–10.641), and vitamin E and selenium administration to the pregnant dam (P < 0.01; OR: 3.933; CI 95%: 0.703–1.248).Infection with Salmonella spp. was found to be significantly affected by the animal age (P < 0.05; OR: 0.376; CI 95%: 0.511–1.369), Hygiene (P < 0.05; OR: 0.628; CI 95%: 1.729–5.612), and region (P < 0. 01; OR: 0.970; CI 95%: 0.841–1.624).The results of the present study indicate the importance of PCR as rapid, effective and reliable tool for screening of ETEC and Salmonella spp. when confronted with cases of undifferentiated calf diarrhea. Moreover, identification of the risk factors associated with the spreading of bacteria causing diarrhea may be helpful for construction of suitable methods for prevention and control.  相似文献

This experiment was conducted to investigate the effects of different dietary energy levels on growth performance, slaughter traits, meat quality and blood biochemical parameters in fattening male Hu lambs. Sixty lambs were fed five iso-protein diets which contained different levels of metabolizable energy in a completely randomized design for 70 days. At the end of study, fifteen lambs were harvested for further study. With the increase in dietary energy level, the daily weight gain and dry matter intake extremely increased (p < .001), and feed conversion ratio decreased significantly (p < .01). The live weight before slaughter (LWBS) and carcass weight had a significant increase (p < .05), non-carcass fat ratio and routine indexes of meat quality had no significant difference. Almost no effect was observed for the amino acid profile except for glycine concentration decreased (p < .05) in longissimus dorsi (LD) muscle. The concentrations of C17:0, C18:3n-3 and n-3 PUFA (p < .01) significantly decreased with the increasing dietary energy levels, and the ratio of ∑n-6/∑n-3 (p < .01) increased, whereas the concentrations of C18:1n-9t (p < .05) decreased in LD muscle and C18:1 (p < .01) increased in biceps femoris (BF) muscle. There were no obviously differences for diameter, area and density of muscle fibres. The relative expression of MyHC-IIa and MyHC-IIx decreased significantly (p < .05) in BF muscle, and the relative expression of MyHC-IIa displayed an obviously decreasing trend (p < .10) in LD muscle. These results suggest that increasing the dietary energy level can improve the growth performance and slaughter traits, and influence meat quality and fatty acid profiles in different muscle tissues of fattening male Hu lambs. These results provide a theoretical basis for developing Hu sheep nutritional standards and designing feed formulations.  相似文献

The effect of feeding system and sex on intake, growth and carcass and meat characteristics of Assaf fattening lambs was studied. Twenty four weaned Assaf lambs, half males and half females were used. After an adaptation period, lambs were randomly assigned to one of the two feeding systems studied. Control lambs received ad libitum commercial concentrate (70% barley, 22% soybean meal, 4.8% wheat, 0.5% bicarbonate, 2.7% mineral–vitamin premix) and barley straw. Free Choice lambs had ad libitum and separate access to whole barley grain and a protein supplement (73.3% soyabean meal, 16% wheat, 1.7% bicarbonate, 9% mineral–vitamin premix). Lambs were slaughtered at 25 kg LBW. Feed intake was not significantly affected by sex (P > 0.05) but Free Choice lambs showed higher dry mater intake than the Control lambs. Nevertheless, only males from the Free Choice group showed an improvement in feed efficiency rate (P < 0.05) and average daily gain (P < 0.01) relating to the Control lambs. Neither feeding system nor feeding system × sex interaction significantly affected non-carcass weight, fat depots, cold carcass weight, dressing percentage, main proportions of commercial cuts and meat characteristics (P < 0.05). As it was expected, female lambs showed a lower average daily gain and a higher feed conversion efficiency than male lambs (P < 0.001) which was related to a greater fat deposition. Meat characteristics were unaffected by sex, with the exception of longissimus lumborum fat content, which was higher in female lambs.The present results suggest that Free Choice feeding system might be suitable for intensively reared male lambs since improvements in daily weight gain and feed conversion rate were achieved, with no adverse effect on carcass and meat quality. Despite the selected diet of both, males and females, did not differ in the crude protein content, the use of Free Choice feeding system for female lambs did not improve performance compared to the Control group.  相似文献

Vitamin A plays a critical role in many essential life processes. In herbivores, it is either derived from plant β-carotene or directly as a dietary supplement. In cattle, vitamin A has the potential to influence various carcass traits that are sought by specific beef markets. A group of 20 Angus steers was removed from pasture and fed a low β-carotene and vitamin A cereal-based ration on a feedlot for 308 days. Ten of the steers were supplemented with vitamin A (retinyl palmitate, 60 IU of vitamin A/100 kg body weight/day) and the other ten received no supplement. The results demonstrated that restriction of vitamin A intake changed intramuscular fat deposition without changing subcutaneous fat depots. Angus steers that had been depleted of vitamin A showed increased intramuscular fat in the longissimus thoracis et lumborum (LTL) by 35% (P < 0.026) and seam fat area at the quartering site by 33% (P < 0.0273), when compared with cattle supplemented with vitamin A. There were no changes in intramuscular fat in the semitendinosus. Visually assessed marbling scores were also higher (19%; P < 0.094) in the non-supplemented, depleted group. There was no effect of vitamin A depletion on cattle growth and other meat traits (eye muscle area, meat colour, pH, meat cut weight), meat eating attributes (tenderness, cooking loss) or muscle fibre diameter. The only difference (P < 0.0177) among the meat traits was fat colour where depleted animals had whiter fat than the controls. Moreover, the fat from the vitamin A depleted group was softer with a lower melting point. We conclude that the reduced vitamin A consumption, leading to vitamin A depletion, increases intramuscular fat. On the other hand, the vitamin A depletion did not increase subcutaneous fat depth or change other meat quality traits, suggesting that marbling and these other traits are not invariably related.  相似文献

In this experiment the effect of digestible energy (DE) restriction on carcass traits and meat quality of rabbits selected divergently for body fat content was studied. Using a 2 × 3 factorial arrangement, Pannon White rabbits selected for high (HFAT) or low (LFAT) total body fat content were fed isocaloric diets between 4 and 12 weeks of age. Energy restriction was achieved by reducing the daily feed intake by 10% (M) and 20% (L) compared to the ad libitum fed group (H). Proportional to the reduction of feed intake the nutrient density of diets M and L was increased. Thus, the DE intake was reduced while the nutrient intake remained stable among the 3 dietary groups. Effect of genetic group on carcass traits and on meat quality parameters was less marked than DE restriction. Selection for high body fat content improved the dressing out percentage (58.3 and 57.3% in HFAT and LFAT respectively; P < 0.01), and reduced the percentage of the full gastrointestinal tract to slaughter weight (13.7 and 14.3% in HFAT and LFAT respectively; P < 0.05). Reduction of DE intake decreased the body weight and the weight of the chilled and reference carcass (P < 0.001), while it had no effect on dressing out percentage. In group H the percentage of fore part to reference carcass was higher (29.7 vs 29.0%; P < 0.01) while that of the hind part was lower than in group L (37.0 vs 38.2%; P < 0.001). The Longissimus lumborum meat from HFAT rabbits exhibited a lower content of moisture (76.1 and 76.5% in HFAT and LFAT respectively; P < 0.01) as well as lower L (53.5 and 54.4 in HFAT and LFAT respectively; P < 0.05) and b (0.42 and 0.81 in HFAT and LFAT respectively; P < 0.05) colour values. When compared with ad libitum fed rabbits, those restricted at 20% (L) exhibited a higher content of moisture (76.9 vs 75.8%; P < 0.01), as well as higher pHu (5.87 vs 5.73; P < 0.01) associated with lower cooking loss (17.8 vs 19.1%; P < 0.05). L rabbits also produced lower values of redness (a, 2.21 vs 3.35; P < 0.01) and yellowness (b, 0.23 vs 0.91; P < 0.01). No effect of the divergent selection for body fat content on the total lipid percentage and on most of the fatty acids in the meat of the hind leg was found. DE restriction resulted in lower SFA (L: 32.1 vs H: 35.8%; P < 0.01) and MUFA (L: 21.0 vs H: 28.6; P < 0.01) but higher PUFA contents (L: 45.5 vs H: 34.9%; P < 0.01) in the hind leg meat.  相似文献

The effect of extruding the cereal and/or the protein supplement of a compound feed on its nutritive value and on the performance of intensively reared male calves was studied. The compound feed was formulated with 0.65 of a cereal blend (60:40 maize:barley), 0.25 of a protein blend (1/3:1/3:1/3 raw soybeans:peas:lupins), and 0.08 of urea to contain 0.17 of crude protein. It was tested without extruding (NE) and with the cereal blend (CE), the protein blend (PE) or both (CPE) extruded. Another non-extruded compound feed with mainly soybean meal as the protein supplement (NE-SBM) also was studied. The five experimental compound feeds together with barley straw were offered ad libitum to 50 male Friesian calves (112 kg initial live weight) for 13 weeks, recording individual feed intake and live weight gain. Crude protein (CP) solubility and in vitro and in situ CP degradation of experimental compounds, and in vitro gas production kinetics, in vivo digestibility and urinary allantoin excretion of diets were determined. Cereal extrusion promoted a greater gas production at all incubation times (p < 0.001), associated to the increase in starch gelatinisation. PE and NE-SBM compounds showed lower in vitro (p < 0.01) and in situ (p < 0.001) CP degradability than the other feeds. Ingredient extrusion did not affect apparent DM, OM and CP digestibilities, but diet CPE showed a lower NDF digestibility (p < 0.05) than the others (0.293 vs. 0.420, 0.387, 0.390 and 0.407 with CPE vs. NE, CE, PE and NE-SBM, respectively). The apparent EE digestibility increased (p = 0.053) with the inclusion of extruded ingredients, (0.799, 0.749 and 0.794 vs. 0.719 and 0.702 with CE, PE and CPE vs. NE and NE-SBM, respectively). Daily allantoin excretion was lower (p < 0.01) in calves receiving the CPE diet than in those receiving the NE, PE and NE-SBM diets. Treatments did not affect the average daily gains (1.61 ± 0.022 kg d− 1), however calves on the CPE diet showed a lower (p < 0.01) concentrate and total conversion ratios than those fed NE, PE and NE-SBM (2.60 vs. 2.79, 2.85 and 2.98 kg concentrate DM kg− 1 daily gain and 2.93 vs. 3.09, 3.13 and 3.26 kg total DM kg− 1 daily gain, respectively). The CE diet resulted in numerically higher (p > 0.05) concentrate and total DM conversion ratios (2.72 and 3.02) than CPE and lower (p < 0.05) than NE-SBM. Improvement in feed conversion ratio after extrusion would be related to a better starch utilisation and not to changes in microbial or dietary protein flow to the duodenum, although a better utilisation of fat energy cannot be disregarded.  相似文献

The aim of the study was to evaluate the protection generated in mice against Toxoplasma gondii brain cyst burden by vaccination with T. gondii cytoskeleton proteins using Lactobacillus casei as adjuvant. One hundred and sixty-eight NIH mice were randomly allocated into eight groups of 21 mice each. Animals were immunized as follows: in group 1 with Toxoplasma lysate antigen (TLA) in Freund's modified adjuvant, containing L. casei (FMA), in group 2 with Toxoplasma cytoskeleton proteins (TCPs) in FMA, in group 3 with FMA, in group 4 with phosphate buffered saline (PBS), in group 5 with L. casei dead by heath (Lc), in group 6 with Freund's complete adjuvant (FCA), in group 7 with TLA in FCA, and in group 8 with TCP in FCA. Mean brain cyst burden (±S.E.M.) was assessed in mice 8 weeks after challenge with T. gondii Me49 strain (20 cysts per mouse). The percentages of reduction in cyst burden per brain (P < 0.01) as compared with the group 4 (control: mean 3181 ± 97.5) were 77.25% (724 ± 98) in group 1, 88.02% (381 ± 97.5) in group 2, 38.92% (1943 ± 130.3) in group 3, 44.31% (1771.4 ± 102) in group 5, 59.28% (1295.2 ± 99.1) in group 7 and 55.69% (1409.5 ± 89.9) in group 8. In order of importance, the best protection was obtained in groups 2, 1, 7, 8, 5 and 3. Noticeably the mice inoculated with L. casei alone showed a significant reduction in T. gondii brain cysts (P < 0.01), while those animals treated with FCA alone did not. Additionally, IgM anti-T. gondii antibody levels, as determined by ELISA 2 weeks after challenge, were highest in group 2 (P < 0.01) than in the other seven groups. Results suggest that T. gondii cytoskeleton proteins with L. casei as adjuvant constitute a good anti-toxoplasmosis vaccine candidate.  相似文献

Background

AMP-activated protein kinase (AMPK) plays an important role in the regulation of glucose and lipid metabolism in skeletal muscle. Many pigs of Hampshire origin have a naturally occurring dominant mutation in the AMPK γ3 subunit. Pigs carrying this PRKAG3 (R225Q) mutation have, compared to non-carriers, higher muscle glycogen levels and increased oxidative capacity in m. longissimus dorsi, containing mainly type II glycolytic fibres. These metabolic changes resemble those seen when muscles adapt to an increased physical activity level. The aim was to stimulate AMPK by exercise training and study the influence of the PRKAG3 mutation on metabolic and fibre characteristics not only in m. longissimus dorsi, but also in other muscles with different functions.

Methods

Eight pigs, with the PRKAG3 mutation, and eight pigs without the mutation were exercise trained on a treadmill. One week after the training period muscle samples were obtained after euthanisation from m. biceps femoris, m. longissimus dorsi, m. masseter and m. semitendinosus. Glycogen content was analysed in all these muscles. Enzyme activities were analysed on m. biceps femoris, m. longissimus dorsi, and m. semitendinosus to evaluate the capacity for phosphorylation of glucose and the oxidative and glycolytic capacity. Fibre types were identified with the myosin ATPase method and in m. biceps femoris and m. longissimus dorsi, immunohistochemical methods were also used.

Results

The carriers of the PRKAG3 mutation had compared to the non-carriers higher muscle glycogen content, increased capacity for phosphorylation of glucose, increased oxidative and decreased glycolytic capacity in m. longissimus dorsi and increased phosphorylase activity in m. biceps femoris and m. longissimus dorsi. No differences between genotypes were seen when fibre type composition was evaluated with the myosin ATPase method. Immunohistochemical methods showed that the carriers compared to the non-carriers had a higher percentage of type II fibres stained with the antibody identifying type IIA and IIX fibres in m. longissimus dorsi and a lower percentage of type IIB fibres in both m. biceps femoris and m. longissimus dorsi. In these muscles the relative area of type IIB fibres was lower in carriers than in non-carriers.

Conclusions

In exercise-trained pigs, the PRKAG3 mutation influences muscle characteristics and promotes an oxidative phenotype to a varying degree among muscles with different functions.  相似文献

A grazing experiment was undertaken to evaluate the effect of PEG supplementation on the fatty acid composition of milk from Sarda sheep grazing sulla. Twenty-four late-lactating sheep (12 per group), were paired and split into two groups: group control (CON), dosed daily with a quenching gun with 200 ml of water, and group PEG, dosed with 200 ml of a 50/50 w/v water solution of PEG. The sheep grazed two 0.8 ha plots of sulla under a rotational grazing scheme.The contents of c-9, t-11 CLA and t-11 C18:1 in milk fat were on average 40% higher (P < 0.01) in the PEG group than in the CON group. This can be explained by the higher biohydrogenation activity of ruminal bacteria in the PEG group, due to the partial inactivation of the tannins. Odd-branched chain fatty acids (OBCFA) were higher in PEG than in the control group (+ 20%; P < 0.01) and this confirms the hypothesis that tannin in sulla reduced ruminal microbial activity. Both linoleic (C18:2 c-9 c-12) and linolenic (C18:3 c-9 c-12 c-15) fatty acids were lower (P < 0.05) in milk from PEG, than in the CON-group (− 12% and − 30% for linoleic and linolenic acids, respectively). The mitigating effect on tannins of PEG increased the ratio of ω6/ω3 by 24%; (P < 0.01) and total trans FA content in milk by 20% (P < 0.01). In conclusion, condensed tannins in sulla at flowering are conducive to lower c-9, t-11 CLA and t-11 C18:1 but also lower total trans FA, ω6/ω3 ratio and higher linoleic and linolenic acid.  相似文献
