Serum concentrations of thyroxine (T4), 3,5,3'-triiodothyronine (T3), free thyroxine (fT4), and free 3,5,3'-triiodothyronine (fT3) were compared between tumor-bearing dogs (with and without chronic weight loss) and non—tumor-bearing dogs (with and without chronic weight loss) (n = 83). Serum T4, T3, and fT3 concentrations were lower ( P < .05) in dogs with weight loss, whether or not they were tumor-bearing, than in dogs without weight loss. Serum fT4 concentrations did not vary among the groups. Serum albumin concentrations were lower ( P < .05) in cachectic dogs than in dogs not experiencing weight loss, regardless of their tumor-bearing status. Percentage of weight loss was found to be associated ( P < .05) with T4, T3, and fT3 concentrations. It appears that the low thyroid hormone concentrations are related to either an abnormal nutritional state or to the severity of illness, rather than to a tumor-related phenomenon.  相似文献

Background: Neutral Protamine Hagedorn human analogue insulin (Humulin N) is commonly used for treatment of canine diabetes mellitus (DM). However, blood glucose and serum insulin concentrations in Humulin N-treated dogs with naturally occurring DM have not been reported.
Objective: To investigate blood glucose and serum insulin concentrations in the clinical setting of client-owned Humulin N-treated dogs with naturally occurring, well-regulated DM.
Animals: Ten client-owned dogs with naturally occurring, well-regulated DM.
Methods: In this clinical study, blood glucose and serum insulin concentrations were measured when dogs received food and insulin (T0), at approximately every half hour for the next 2 hours, and then approximately every 2 hours for an additional 8 hours. Insulin duration of action was defined as the number of hours from T0 to the lowest blood glucose concentration and until blood glucose concentration returned to an interpolated value of 70% of basal blood glucose concentration (Glucoseb).
Results: Mean percent of insulin-induced blood glucose suppression was 49.9 ± 17.1% (median, 46%; range, 29–78%). Insulin duration of action ranged from 4 to 10 hours. Blood glucose concentration increased initially and returned to Glucoseb within 0.6–2.2 hours after T0 in 5 dogs. This initial blood glucose surge then was followed by blood glucose suppression in all 5 dogs.
Conclusions and Clinical Importance: These results suggest that Humulin N administered SC twice daily is an effective mode of treatment for dogs with naturally occurring DM. Postprandial hyperglycemia is present in some well-regulated diabetic dogs treated with Humulin N.  相似文献

Objective  To evaluate the effect of acepromazine or xylazine on Schirmer tear test 1 results in clinically normal cats.
Animals  Sixteen healthy cross-breed cats.
Procedure  The animals were randomly divided into two groups of eight cats each. The first group was sedated with acepromazine alone (0.2 mg/kg) and the second group received only xylazine (2 mg/kg). All cats had Schirmer tear test (STT) readings taken prior to sedation and at 15 and 25 min postsedation.
Results  Sedation with acepromazine or xylazine in cats with normal pre-sedation STT 1 values caused a statistically significant decrease in mean values of tear production in both groups. In acepromazine group the mean ± SEM STT at T15 and T25 were 4.31 ± 0.98 ( P  < 0.001) and 5.18 ± 1.07 ( P  = 0.002). The post-treatment mean ± SEM values in xylazine group were 2.18 ± 0.97 ( P  < 0.001) and 2.62 ± 1.17 ( P  = 0.001) at 15 and 25 min respectively. Comparison between T15 and T25 in acepromazine group ( P  = 0.49) and xylazine group ( P  = 0.56) revealed no significant differences.
Conclusion  These observations indicate that both acepromazine or xylazine significantly reduced tear production in clinically normal cats. In cats, clinicians should measure STT values prior to utilizing acepromazine or xylazine as sedatives in order to accurately assess the results. Moreover, sterile ocular lubricant or tear replacement should be used as a corneal protectant during sedation with these drugs.  相似文献

Concentrations of prolactin (PRL), LH, testosterone (T), TSH and thyroxine (T4) were determined before and at 20, 120 and 180 min after a single iv injection of thyrotropin-releasing hormone (TRH) in eight Beagles, eight Fox Terriers, six Labrador Retrievers and five Great Danes that were normospermic. Mean basal PRL concentrations were lower in the Fox Terriers compared with the Great Danes (p < 0.05). Mean LH concentrations were higher in the Fox Terriers than in the Beagles, and T was lower in the Fox Terriers at some times but not others (p < 0.05). Thyroid Stimulating Hormone (TSH) concentrations did not differ among breeds, while mean basal T4 values were lower in Fox Terriers compared with Labrador Retrievers and Great Danes (p < 0.05). Stimulation of T4 secretion 120 and 180 min after iv TRH injection was most pronounced in the Beagles and less in the Fox Terriers (p < 0.05). The results of the present study indicate that potential breed differences in circulating concentrations of PRL, LH, T, TSH and T4 in male dogs with apparently normal fertility can be encountered, but further studies are needed to determine whether the observed differences are typical features of these breeds, reflect subsets of dogs within breeds, or are in part because of possible uncontrolled parameters such as sample timing, ambient photoperiod, housing conditions or diet.  相似文献

GASTRIC EMPTYING OF SOLID RADIOPAQUE MARKERS IN HEALTHY DOGS   总被引:1,自引:0,他引:1  
The gastric emptying of 1.5 mm diameter (small) and 5.0 mm diameter (large) radiopaque markers (BIPS) was assessed in 20 dogs. The markers were fed to the dogs in a test meal and abdominal radiographs were made hourly thereafter. Studies were repeated three times in each dog. The variation between two veterinarians interpreting the radiographs was low. The sex, age and day of the study did not have a significant effect on the lag phase or the time taken to empty 25%, 50% and 75% of the markers (T25, T50 and T75 respectively). There was a weak but significant positive correlation between the body weight and T50. There was no significant difference in gastric emptying parameters between the large and small markers.
The mean gastric emptying versus time curve of the small markers on day one was chosen to represent the reference curve for healthy dogs. The lag phase of the small markers on day one was 2.45 ± 2.04 hours, the T25 was 4.85 ± 2.15 hours, the T50 was 6.05 ± 2.99 hours and the T75 was 8.32 ± 2.72 hours (mean ± SD).  相似文献

A 60-days experiment was conducted to study the optimum gelatinized (G) to non-gelatinized (NG) starch ratio in the diet of Labeo rohita juveniles with respect to digestive and metabolic response and on growth. Two-hundred and thirty-four juveniles (avg. wt 2.53 ± 0.04 g) were randomly distributed in six treatment groups with each of three replicates. Six semipurified diets either containing NG and/or G corn starch viz., T1 (100% NG, 0% G starch), T2 (80% NG, 20% G starch), T3 (60% NG, 40% G starch), T4 (40% NG, 60% G starch), T5 (20% NG, 80% G starch) and T6 (0% NG, 100% G starch) were prepared. The dry matter digestibility and carbohydrate digestibility were highest (p < 0.05) in T6 group and lowest in T3 and T4 groups. The amylase activity in intestine increased as G:NG level increased in the diet. Protease activity in intestine was highest in T6 group and lowest in T1 group. Similar trend was recorded for specific growth rate, protein efficiency ratio and apparent net protein utilization. Liver glycogen, hepatosomatic index and blood glucose level increased linearly with the increasing level of G starch in the experimental diet. The results indicate that higher nutrient digestibility and growth was recorded either at low (20% G starch, T2) or high (100% G starch, T6) G starch fed group. But high G starch fed group (T6) exhibits higher liver glycogen and blood glucose level, which may lead to stress due to long-term feeding. Hence, it is suggested that 20% G and 80% NG starch is optimum for better nutrient digestibility and growth in L. rohita juveniles.  相似文献

An endocrinopathy characterised by hyperprolactinaemia and galactorrhoea was found to be associated with primary hypothyroidism in an entire bitch. Clinical diagnosis was confirmed by lowered baseline T4 blood levels, reduced response to the thyroid stimulating hormone (TSH) test, elevated prolactin and TSH values, and echographic and scintigraphic examinations. Levothyroxine treatment resulted in a clinical improvement (especially of the galactorrhoea), and levels of prolactin, thyroxine, TSH, triglycerides and total cholesterol plasma returned to normal.  相似文献

Background: Thyroid hormone concentrations were found to be different in Greyhounds and Whippets compared with nonsight hound dogs.
Hypothesis: In Sloughis, thyroid hormone concentration is lower than in nonsight hounds and comparable to Greyhounds.
Animals: Fifty-one Sloughis with no evidence of disease and a mean age of 4 years (range, 1–12 years).
Methods: Thyroid profiles consisting of total thyroxine (tT4), free thyroxine (fT4), free thyroxine after equilibrium dialysis (fT4 after ED), canine thyroid stimulation hormone (cTSH), and thyroglobulin antibodies as well as CBC and serum biochemistry results of Sloughis were compared with those of normal dogs. In 8 Sloughis, TSH stimulation tests were performed.
Results: In Sloughis, tT4 concentrations and fT4 concentrations measured by chemiluminescence were lower than those of controls (1.13 ± 0.65 μg/dL compared with 2.9 ± 0.8 μg/dL, P < .0001 and 11 ± 4.3 pmol/L compared with 16.7 ± 5.2 pmol/L, P < .0001, respectively). Concentrations of fT4 after ED and TSH were increased in Sloughis, when compared with controls (41.3 ± 26.9 pmol/L compared with 20.98 ± 10.29 pmol/L, P < .0001 and 0.22 ± 0.15 pmol/L compared with 0.15 ± 0.13 pmol/L, P = .0138, respectively). T4 concentration after TSH stimulation increased from 1.5 μg/dL (range, 0.2–2.7 μg/dL) to 2.7 μg/dL (range, 1.2–4.7 μg/dL); the recommended post-TSH T4 concentration was achieved by only 3 of 8 Sloughis. Hemoconcentration was found in 84.3% and hypoglobulinemia in 80.3%.
Conclusions and Clinical Importance: When evaluating Sloughis for hypothyroidism, veterinarians should be aware that these dogs have different thyroid hormone concentrations than nonsight hound dogs.  相似文献

Three cats were diagnosed as hyperthyroid based on clinical signs, historical findings, laboratory abnormalities, and basal serum thyroxine (T4) concentrations, and/or nuclear thyroid scans. Additionally, a presumptive diagnosis of thyroid carcinoma with pulmonary metastasis was made in each cat based on radiographic or scintigraphic evaluation. All three cats had solitary pulmonary nodules 1.5 to 2 cm in diameter on survey thoracic radiographs; one cat also had chylous pleural effusion and pulmonary lobar consolidation. Focal pulmonary accumulation of sodium pertechnetate (99mTcO4 -) and/or radioiodine (131I) corresponding to radiographic lesions were seen in all cats. Two cats were treated with single ablative doses (1111 to 1480 MBq) of131I; the remaining cat was euthanatized.
One of the treated cats died 8 days later; the other cat was euthanatized 22 weeks following treatment. Histopathologic examination of tissue obtained at necropsy confirmed metastatic thyroid carcinoma in one cat and bronchogenic adenocarcinoma in two cats. Our findings indicate that increased radionuclide uptake in focal pulmonary lesions and cytologic evaluation of tissue obtained by fine-needle aspiration are not specific for thyroid tissue. Background: Retinol-binding protein (RBP) is suggested as a clinically useful marker of renal function in cats.
Hypothesis: Serum and urinary RBP concentrations in hyperthyroid (HT) cats differ from those in healthy (H) cats; radioiodine (131I) treatment influences serum and urinary RBP concentrations in HT cats.
Animals: Ten HT and 8 H cats.
Methods: RBP concentration was evaluated in feline serum and urine samples from a prospective study.
Results: There was a significant ( P = .003) difference in the urinary RBP/creatinine (uRBP/c) ratios of H (−) and untreated HT (1.4 ± 1.5 × 10−2 μg/mg) cats. Serum total thyroxine concentration (1.8 ± 1.9 μg/dL, 24 weeks) and uRBP/c (0.6 ± 1.0 × 10−2 μg/mg, 24 weeks) decreased significantly ( P < .001) in HT cats at all time points after treatment with 131I, and these variables were significantly correlated with one another ( r = 0.42, P = .007). Serum RBP concentrations from HT cats (199 ± 86 μg/L) did not differ significantly ( P = .98) from those of H cats (174 ± 60) and did not change after treatment with 131I (182 ± 124 μg/L, P = .80).
Conclusion and Clinical Importance: The presence of urinary RBP in HT cats is a potential marker of tubular dysfunction that is correlated to thyroid status, although it is independent of circulating RBP concentrations. The decreased uRBP/c combined with the absence of changes in serum RBP after treatment suggests that the suspected tubular dysfunction was partly reversible with treatment of 131I.  相似文献

The objectives of this study were to determine if phenylbutazone decreased serum thyroxine (TT4) and free thyroxine (FT4) concentrations using radioimmunoassay and equilibrium dialysis techniques in horses, and, if so, an additional objective was to determine the duration of this decreased concentration once phenylbutazone administration was discontinued. Serum TT4 and FT4 concentrations were determined before and after administration of 4.4 mg/kg of phenylbutazone IV bid for 5 days. Treatment with phenylbutazone caused a significant decrease in TT4 and FT4 concentrations ( P < .05). Serum TT4 concentration significantly decreased after day 4 of treatment and remained significantly below baseline value for 10 days after discontinuing phenylbutazone administration; it returned to a value not different from the baseline value by the 11th day. Serum FT4 concentration significantly decreased after day 4 of treatment and remained significantly below the baseline value for only 1 day after phenylbutazone administration was discontinued; it returned to a value not different from the baseline value by the 3rd day after discontinuation of phenylbutazone. These results indicate that serum TT4 and FT4 should not be used to evaluate thyroid function in horses receiving phenylbutazone. In addition, results should be interpreted cautiously when phenylbutazone has been administered within 2 days (for FT4) or within 10 days (for TT4) of sample collection.  相似文献

Background: Hyperthyroid cats are at risk of developing azotemic chronic kidney disease (CKD) and diagnostic tools currently used to screen for CKD in hyperthyroid cats are either unreliable or impractical.
Hypothesis: Urine N -acetyl-β- d -glucosaminidase index (NAGi) is a good biomarker for azotemic CKD in hyperthyroid cats.
Animals: Twenty-four newly diagnosed nonazotemic hyperthyroid cats and 10 healthy cats.
Methods: All cats were evaluated for hyperthyroidism at baseline. Hyperthyroid cats were treated with methimazole and reevaluated once euthyroid. At the end of the study, cats were divided into 3 groups: healthy cats, nonazotemic, and azotemic euthyroid cats. Baseline group characteristics were compared to predict azotemic CKD. The influence of treatment on NAGi was evaluated.
Results: Baseline NAGi was significantly different among groups ( P = .004). Azotemic cats had a higher median value (13.12 U/g) when compared with healthy cats (1.38 U/g). With NAGi >2.76 U/g, negative and positive predictive values for development of azotemia were 77.7 and 50%, whereas the combination of a urine specific gravity (USG) ≤1.035 and T4 >7.80 μg/dL enhanced predictive values to 88.9 and 83.3%, respectively. NAGi values decreased significantly over time in treated nonazotemic cats.
Conclusions and Clinical Relevance: Baseline NAGi did not differentiate azotemic from nonazotemic euthyroid cats. NAGi could be used to assess renal function during medical therapy allowing the clinician to adjust methimazole dosage accordingly. The combination of USG and T4 could optimize identification of appropriate candidates for permanent treatment of hyperthyroidism.  相似文献

The serum 25-OH-D3, Ca, P, and Mg concentrations in thirty-seven cows that had calved in March April were studied in this trial. Twenty-three had puerperal paresis. Values observed in this group were Ca, 1.27 ± 0.3 mmol/l, P, 1.63 ± 0.82 mmol/l, Mg, 1.14 ± 0.27 mmol/l, and 25-OH-D3, 32.7 ± 22 μg/l. In the fourteen age-matched healthy control cows from the same herd values were Ca, 1.93 ± 0.5 mmol/l, P, 3.09 ± 1.28 mmol/l, Mg, 1.01 ± 0.3 mmol/l and 25-OH-D3, 31.5 ± 19.7 μg/l. The range of values for the cows serum 25-OH-D3 concentration was l't; 3.2 to 76 μg/l. No differences could be established in terms of 25-OH-D3 concentrations between the groups.  相似文献

Acute pelvic limb paralysis occurred in a racing Greyhound after a traumatic incident. During myelography, a dorsal laceration of the dura mater was detected at T12–13. Immediate hemilaminectomy was performed at T12–13, but neurologic status did not improve. At necropsy there was epidural and subdural hemorrhage at the T12–13 interspace, and malacia and swelling of the spinal cord at that level. Intramedullary hemorrhage, malacia, and free fibrocartilaginous material characterized histopathology of the lesion. Possible mechanisms of the traumatic disk extrusion are discussed.  相似文献

Concentrations of the non-steroidal anti-inflammatory drug (NSAID) alclofenac were determined by a sensitive high performance liquid chromatographic procedure in plasma and urine of horses following oral administration of a dose of 3 g. In plasma, alclofenac was present in detectable concentrations for 72 h. The plasma disposition in individual horses was best described by a bi-compartmental model with two successive rate constants ka1= 0.05 ± 0.06 h-1 and ka2= 0.06 ± 0.01 h-l. Alclofenac half-lives t ½ and t 1/2β were 1.0 ± 0.8 h and 6.9 ± 1.5 h, respectively. Maximal concentrations (38.9 ± 16.2 μg/ml) were obtained after 8.5 ± 2.4 h. Alclofenac was detected in urine for at least 48 h after dosing. The percentage of the dose excreted as unchanged alclofenac in 12 h was very low (0.68 ± 0.19%), total (free + conjugated) alclofenac accounted for 2.16 ± 0.55% of the dose.  相似文献

The existence of hypothyroidism in greyhounds remains controversial and its investigation is complicated by the low circulating thyroid hormone concentrations typically found in healthy dogs of this breed. Quantitative measurement of thyroidal technetium-99m pertechnetate (99mTcO4) uptake is known to be useful in assessing thyroid function in other breeds. The aim of this study was to evaluate thyroid scintigraphy as a method of assessing thyroid function in greyhounds suspected of primary hypothyroidism. Twenty greyhounds (eight females, 12 males) were studied. Thirteen had bald thigh syndrome and seven poor performance and low total T4. Total T4 concentrations were decreased in 18 (90%), and free T4 in two (10%) dogs. All canine thyroid stimulating hormone concentrations were within the reference interval. Thyroidal 99mTcO4 uptake values (mean ± SD, 0.76 ± 0.26%) were within the reference limits published for euthyroid dogs (0.39–1.86%) making hypothyroidism highly unlikely. There were no significant differences ( P <0.05) when comparing data between dogs with bald thigh syndrome (13 dogs) and the remaining dogs (seven dogs). Seventeen (85%) dogs had higher uptake in the left thyroid gland than in the right that might reflect an anatomic feature of the greyhound breed. Calculation of percent thyroidal uptake of 99mTcO4 is more accurate than thyroid:salivary gland ratios because of high variability in salivary gland uptake. Percent thyroidal uptake of 99mTcO4 should be used when assessing thyroid function scintigraphically in the greyhound breed.  相似文献

Phenylbutazone was administered intravenously (i.v.) to a group of four lactating cows at a dosage of 6 mg/kg body weight. Whole plasma, protein-free plasma and milk were analysed for phenylbutazone residues. Pharmacokinetic parameters of total and free phenylbutazone in plasma were calculated using a non compartmental method. In regards to whole plasma data, the mean volume of distribution at steady state ( V ss), was 147 mL/kg body weight, with a mean (± SEM) terminal elimination half-life ( t 1/2) of 40 ± 6 h. The mean clearance ( Cl ) was 3 mL/h/kg body weight. The V ss as determined from the protein-free plasma fraction was 50 021 mL/kg body weight. This larger V ss of free phenylbutazone compared to total plasma phenylbutazone was attributed to a high degree of plasma protein binding, as well as the greater penetration of free phenylbutazone into tissues. The mean t 1/2 of free phenylbutazone was 39 ± 5 h. This similarity to the t 1/2 estimated from total plasma phenylbutazone data is attributed to an equilibrium between free and plasma phenylbutazone during the terminal elimination phase. Mean t 1/2 as determined from milk, applying a urinary excretion rate model, was 47 ± 4 h. Milk clearance of phenylbutazone was 0.009 mL/h/kg body weight, or about 0.34% of total body clearance. Furthermore, evidence suggests that phenylbutazone either binds to milk proteins, or is actively transported into milk, as its concentration in milk was greater than that predicted due to a simple partitioning from plasma into milk.  相似文献

Objective  We hypothesized that propofol can produce rapidly-reversible, dose-dependent standing sedation in horses.
Study design  Prospective randomized, blinded, experimental trial.
Animals  Twelve healthy horses aged 12 ± 6 years (mean ± SD), weighing 565 ± 20 kg, and with an equal distribution of mares and geldings.
Methods  Propofol was administered as an intravenous bolus at one of three randomized doses (0.20, 0.35 and 0.50 mg kg−1). Cardiovascular and behavioral measurements were made by a single investigator, who was blinded to treatment dose, at 3 minute intervals until subjective behavior scores returned to pre-sedation baseline values. Continuous data were analyzed over time using repeated-measures anova and noncontinuous data were analyzed using Friedman tests.
Results  There were no significant propofol dose or temporal effects on heart rate, respiratory rate, vertical head height, or jugular venous blood gases (pHv, PvO2, PvCO2). The 0.35 mg kg−1 dose caused mild sedation lasting up to 6 minutes. The 0.50 mg kg−1 dose increased sedation depth and duration, but with increased ataxia and apparent muscle weakness.
Conclusions and clinical relevance  Intravenous 0.35 mg kg−1 propofol provided brief, mild sedation in horses. Caution is warranted at higher doses due to increased risk of ataxia.  相似文献

The pharmacokinetic properties of pradofloxacin and doxycycline were investigated in serum, saliva, and tear fluid of cats. In a crossover study design, six cats were treated orally with a single dose of pradofloxacin (Veraflox® Oral Suspension 2.5%) and doxycycline (Ronaxan® 100 mg) at 5 mg/kg body weight. Following administration, samples of serum, saliva, and tear fluid were taken in regular intervals over a period of 24 h and analysed by turbulent flow chromatography/tandem mass spectrometry. All values are given as mean ± SD. Pradofloxacin reached a mean maximum serum concentration ( C max) of 1.1 ± 0.5 μg/mL after 1.8 ± 1.3 h ( t max). In saliva and tear fluid, mean C max was 6.3 ± 7.0 and 13.4 ± 20.9 μg/mL, respectively, and mean t max was 0.5 ± 0 and 0.8 ± 0.3 h, respectively. Doxycycline reached a mean C max in serum of 4.0 ± 0.8 μg/mL after 4.3 ± 3.2 h. Whilst only at two time-points doxycycline concentrations close to the limit of quantification were determined in tear fluid, no detectable levels were found in saliva. The high concentrations of pradofloxacin in saliva and tear fluid are promising to apply pradofloxacin for the treatment of conjunctivitis and upper respiratory tract infections in cats. As doxycycline is barely secreted into these fluids after oral application the mechanisms of its clinical efficacy remain unclear.  相似文献

Gonadotropin-releasing hormone (GnRH) antagonists are particularly useful when a rapid inhibitory effect on the gonadal axis is required. The aim of this study was to test the efficacy and clinical safety of a low and high dose of the third generation GnRH antagonist, acyline, on pregnancy termination in female dogs. The effect of the antagonist on the progesterone (P4) serum concentration was also described. Twenty-one mid-pregnant bitches were randomly assigned to a single subcutaneous (SC) dose of a placebo (PLACE; n = 7), a low (ACY-L; 110 μg/kg; n = 6) or high (ACY-H; 330 μg/kg; n = 8) dose of acyline. The animals were followed up for 15 days. All ACY treated but no placebo-treated animals terminated their pregnancy by abortion (p < 0.01). The ACY-L and ACY-H groups interrupted their pregnancy 7 ± 1.9 and 6.4 ± 1.3   days after treatment, respectively (p = 0.7). A significant interaction between treatment and day was found (p < 0.01) for P4 serum concentrations when PLACE was compared with both ACY groups. No difference was found for this hormone between both ACY groups (p > 0.05) where P4 diminished throughout the study. The decreasing rate varied among animals and was closely related to the time of abortion when P4 reached basal concentrations. In PLACE animals, gestation progressed normally and P4 did not change throughout the study (p > 0.05). None of the bitches presented side effects. It was concluded that acyline safely terminated mid-pregnancy by permanently decreasing P4 serum concentrations.  相似文献
