A crop rotation field study was conducted in 1981–1989 to assess the effect of six crop rotation sequences on a soil population of potato cyst nematode (PCN), Globodera rostochiensis, and on potato yield. In the plots with potato monoculture, the tuber yield decreased from 35 to 4.6 t ha?1. All other cropping systems maintained the yield at the original or an even higher level. In monoculture, the population density of the nematode was raised during the first 3 years from 0.1 to 265 larvae g?1 soil, and the density fluctuated afterwards between 30 and 136 larvae g?1 soil. Three of the cropping systems, (1) susceptible potato once in 5 years, (2) susceptible potato once in 4 years, and (3) potato once in 3 years using resistant (H1) and susceptible cultivars alternately, decreased the nematode density under the economic threshold or even under the level of detection. The nematicide treatments (oxamyl) did not control nematode multiplication but prevented yield losses. No nematicide is currently registered nor needed for the control of PCN in Finland. The nematode can be controlled effectively with crop rotation, resistant cultivars and early harvesting.  相似文献

Field trials were conducted in a plastic house artificially infested with an avirulent population of Meloidogyne javanica to determine the durability of the resistance mediated by the Mi gene in tomato rootstocks after repeated cultivation for three consecutive years. Treatments included an experimental rootstock cv. PG76 ( Solanum lycopersicum  ×  Solanum sp.), a commercial rootstock cv. Brigeor ( S. lycopersicum  ×  S. habrochaites ), a resistant tomato cv. Monika ( S. lycopersicum ), and a susceptible cv. Durinta ( S. lycopersicum ). Based on the reproduction index (RI: number of eggs per g root on the resistant cultivar divided by number of eggs per g root on the susceptible cultivar × 100), rootstock cv. PG76 responded as highly resistant (RI = 7%) after the first cropping cycle (3·4 nematode generations), showed intermediated resistance (RI = 33%) after the second cropping cycle (3·3 generations), and was fully susceptible (RI = 94%) after the third cycle (3·3 generations). In contrast, rootstock cv. Brigeor and resistant cv. Monika retained intermediate resistance levels (RI = 41 and 25%, respectively) after the third cropping cycle. Virulent nematode populations were rapidly selected from an avirulent one after repeated cultivation of resistant tomatoes under field conditions. Bioassays conducted under controlled conditions confirmed that selection for virulence occurred more rapidly in plots with cv. PG76 followed by Brigeor and Monika. The nematode population in the field not exposed to Mi resistance remained avirulent to Mi genotypes. The genetic background of the resistant rootstocks and the frequency of cropping were critical factors for the appearance of virulent nematode populations. Irrespective of nematode infection, all resistant tomatoes yielded more than the susceptible cultivar.  相似文献

ABSTRACT The hypothesis that host plants exert selection pressure on Heterodera schachtii populations was tested. Host selection of genotypes from three genetically distinct isolates of H. schachtii was assessed using cabbage, sugar beet, oilseed radish (Raphanus sativus), and white mustard (Sinapis alba). The plants represent a range of susceptibility to H. schachtii and included R. sativus and S. alba, because cultivars of those species have been used as trap crops for H. schachtii in Europe. Genotypic differences in amplified fragment length polymorphism (AFLP) and random amplified polymorphic DNA (RAPD) markers were detected among the isolates after they reproduced on the different hosts. The poorest host plant, R. sativus, resulted in the greatest number of changes in both AFLP and RAPD markers. Oilseed radish selected nematode genotypes in less than four nematode generations. The nematode population genotypes detected by RAPD analyses after selection on oilseed radish were observed even after nematode populations were transferred back to the other three hosts. The genetic markers that were detected after selection were influenced by the genotypes of the original nematode isolates. The results indicate the utility of RAPDs and AFLPs for identifying and monitoring intraspecific genetic variability in nematodes and for understanding nematode population responses to host plants. Nematode management practices such as using resistant cultivars may alter gene frequencies, thereby reducing the efficacy of the tactic and exacerbating the nematode's potential to damage subsequent crops.  相似文献

Resistant tomato cultivars are an important tool to control Meloidogyne spp., which cause the highest yield losses attributed to plant‐parasitic nematodes. However, the repeated cultivation of Mi resistant cultivars can select virulent populations. In the present study, the susceptible tomato cv. Durinta and the resistant cv. Monika were cultivated from March to July in a plastic greenhouse for 3 years to determine the maximum multiplication rate, maximum nematode density, equilibrium density, relative susceptibility and population growth rate of M. incognita; these were used as proxy indicators of virulence and yield losses. The values of population dynamics and growth rate on the resistant tomato increased year by year and were higher when it was repeatedly cultivated in the same plot compared to when it was alternated with the susceptible cultivar and the level of resistance decreased from very to moderately resistant. The relationship between the nematode density at transplanting (Pi) and the relative yield of tomato fitted to the Seinhorst damage model for susceptible, but not resistant, cultivars. The tolerance limit and the relative minimum yield were 2–4 J2 per 250 cm3 of soil and 0.44–0.48, respectively. The tomato yield did not differ between cultivars at low Pi, but it did at higher Pi values, at which the resistant yielded 50% more than the susceptible. This study demonstrates the utility of population dynamics parameters for the early detection of selection for virulence in Meloidogyne spp., and that three consecutive years were not sufficient to select for a completely virulent population.  相似文献

Results are reported for several international collaborative experiments which examined methods of assessing degrees of partial resistance in potato and virulence in potato cyst nematode (PCN, Globodera pallida). It was demonstrated that absolute rates of multiplication can be extremely variable on both susceptible and partially resistant clones, even when the same population and test procedures are employed. It was therefore concluded that, on clones with quantitatively inherited resistance (i.e. partially resistant), absolute rates of multiplication cannot be used to separate pathotypes. Expression of these rates as percentages of those on the susceptible controls reduced the absolute differences between tests, but the values obtained were still too variable for statutory use. However, whatever the environment or nematode population used, it was observed that the resistance of the test clones and virulence of the nematode populations were generally ranked in a similar order. The main exceptions to this were: (1) a Petri-dish test where cv. Vantage was less resistant than in canisters or pots and (2) a pot test where cv. Darwina tended to be more interactive with environmental factors than the other test clones. It was also observed that with some populations of PCN in pots the susceptible cv. Bintje was a less good host than cv. Désirée. On the basis of these results it is suggested that certain partially resistant clones should be used as internal references in statutory, recommended list and breeders' assessment tests against which the resistance of the test clones are compared. For international comparability it is necessary that the different centres conducting such tests use the same reference clones and nematode populations, and similar test methods.  相似文献

Control of Globodera rostochiensis in Ukraine depends on the use of resistant potato cultivars. In the last 2 years, the proportion of resistant entries in the official Ukrainian list has increased from 27% to 45%, and the number of Ukrainian-bred resistant cultivars has increased from 3 to 12. It is now suggested that resistant cultivars are classified into the categories‘resistant and tolerant’ and ‘resistant but only relatively tolerant’. The choice of these cultivars should depend on the nematode population of the soil. Control of Ditylenchus destructor is also considered, on the basis of prophylactive and agrotechnical measures.  相似文献

菲利普孢囊线虫Heterodera filipjevi是禾谷类作物上重要的病原线虫之一,严重影响禾谷类作物的产量和品质。本课题组前期研究发现菲利普孢囊线虫许昌群体是一个新的致病型,其在小麦上的致病力强于其他多个群体,危害更为严重。本研究旨在建立菲利普孢囊线虫许昌群体的快速、准确的分子检测体系,为Heterodera filipjevi许昌群体的监测和防控及抗病品种的选育利用奠定基础。本研究采用随机扩增多态性DNA(RAPD)和序列特征扩增区域(SCAR)的方法,对黄淮麦区5个重要小麦孢囊线虫致病型共9个线虫群体进行RAPD分析和SCAR标记转化,并通过增加除黄淮麦区外的线虫群体验证所获得的致病型相关分子标记的特异性和有效性。本研究共筛选了331条RAPD引物,筛选出2个菲利普孢囊线虫许昌群体相关的RAPD标记,引物S86可以扩增出1条约550 bp的多态性片段,引物S178可以扩增出1条约1 200 bp的多态性片段,并将这2个RAPD标记成功转化为SCAR标记。SCAR标记的特异性检测结果表明这两个SCAR标记只在许昌群体上有特异性扩增,可以用于许昌群体的分子检测。  相似文献

The variability of resistance durability in different potato genotypes harbouring the same resistance QTL but differing by their genetic background was explored. The indirect consequences of the resistance adaptation in terms of local (i.e. genotype‐specific) adaptation and cross‐virulence was also investigated. Following the virulence of the potato cyst nematode Globodera pallida in a long‐term experimental evolution protocol, the results showed that nematode populations were able to adapt to the resistance of four potato genotypes carrying the QTL GpaV from Solanum vernei, and that the plant genetic background has an impact upon the durability of resistance. The pattern of local adaptation observed here indicates that divergent selection has occurred during the experimental evolution performed from the same initial nematode population, and revealed a trade‐off between the adaptation to a resistant potato genotype and the adaptation to another resistant genotype differing in its genetic background. In terms of cross‐virulence between potato genotypes derived from different resistance sources (S. sparsipilum and S. spegazzinii), this study shows that the adaptation to resistance QTL GpaVvrn does not necessarily allow the adaptation to collinear GpaV loci. The results presented here could be useful for predicting evolution of nematode populations in natural agro‐ecosystems and identifying durable strategies for resistance deployment.  相似文献

The eggplant Solanum melongena cv. Cristal, either ungrafted or grafted onto the Solanum torvum rootstock cv. Brutus, was cultivated for two consecutive years in the same plots in a plastic greenhouse to assess the level of resistance to Meloidogyne incognita and crop yield. At the end of the second crop, the putative selection for virulence of the nematode subpopulations coming from infected ungrafted and grafted eggplant was assessed in the eggplant and in S. torvum in a pot experiment. Nematode population densities at transplantation in 2017 ranged from 2 to 378 per 100 cm3 of soil and did not differ between ungrafted and grafted eggplant. At the end of each crop, a higher galling index and number of nematodes in soil and in roots was registered in ungrafted compared to grafted eggplant. The grafted eggplant was categorized as resistant in 2017 and as highly resistant in 2018. Eggplant yield did not differ irrespective of grafting in 2017 after being cultivated for 135 days, but it differed after 251 days of cultivation in 2018. In the pot experiment, S. torvum was categorized as resistant to both M. incognita subpopulations. However, the M. incognita subpopulation obtained from roots of S. torvum produced 49.4% fewer egg masses and 56% fewer eggs per plant in the eggplant than the nematode subpopulation obtained from roots of the eggplant cv. Cristal. The results of this study revealed that the infective and reproductive fitness of the nematode decreased without having been selected for virulence.  相似文献

Resistance to anticoagulant rodenticides in brown rats (Rattus norvegicus Berk.) is associated with pleiotropic effects, notably with an increased dietary vitamin K requirement. Owing to this disadvantage, resistance is believed to be selected against if anticoagulant selection is absent. In small experimental populations of wild brown rats, an investigation was carried out to establish whether tolerance to anticoagulant exposure changed over a period of 2 years. In the same populations, DNA microsatellite markers were used to infer parentage, and this made it possible to estimate reproductive success of sensitive and resistant rats and estimate effective population size, Ne. Even though there was evidence for a selection against resistant rats with high vitamin K requirement, anticoagulant tolerance was not seen to be significantly influenced in the absence of bromadiolone selection. As the population size under investigation was small, random genetic drift may have played a role in this. In the presence of bromadiolone selection, however, the tolerance was significantly increased, suggesting that continuous selection will increase the proportion of highly resistant rats in the population. It was found that, for both males and females, surprisingly few individuals contributed to the next generation with numerous offspring, and most breeders contributed with none or a single offspring. The expected higher reproductive success and consequent increase in proportional numbers of sensitive rats in the absence of anticoagulant selection could not be observed. Among the resistant rats, moderately resistant females were found to be better breeders than highly resistant breeders, but for resistant males the reverse was true. This could be explained by the fact that the increased vitamin K requirement results in sex differential selection; in highly resistant males the selection presumably takes place at the immature stage, whereas in females the vitamin K requirement becomes crucial at the reproductive stage, as vitamin K is not only essential for the blood clotting process but also for bone formation.  相似文献

ABSTRACT We studied changes in pathogenicity, virulence, and aggressiveness of Globodera pallida populations over time. As a measure for pathogenicity, the reproduction factor on a partially resistant host was used; for aggressiveness, the reproduction factor on a susceptible reference host was used; and, for virulence, the ratio pathogenicity/aggressiveness was used. The G. pallida populations were reared in a glasshouse for four generations on potato cultivars with different levels of resistance. The cultivar Elkana did not increase pathogenicity significantly, but the more resistant cultivars Karakter and Darwina did. This increase in pathogenicity was caused by an increase in virulence, whereas aggressiveness generally was not altered significantly. The increase in virulence appeared to be caused by an enhanced ability of eggs to develop into cysts, and not by an increase in egg production per new cyst. The observed changes in virulence could be predicted reasonably well by a simple numerical model. The rate of selection depended strongly on the nematode population. Rearing a mix of two different populations on a susceptible host decreased the virulence strongly, as predicted by the Hardy-Weinberg equilibrium, but increased the aggressiveness because of heterosis.  相似文献

The rice root-knot nematode (RKN), Meloidogyne graminicola, is an important pathogen affecting rice production in South and Southeast Asia. Efficacy of resistance and tolerance in selected M. graminicola-resistant African rice genotypes TOG5674, TOG5675 and CG14 and -susceptible Asian rice genotypes IR64 and UPLRi-5 were examined under a range of initial population densities (Pi) and water regimes. Resistance to M. graminicola in resistant rice genotypes was not broken with increasing pathogen pressure (Pi = 15,000 to 60,000 J2/plant). Resistant rice genotypes were even tolerant to the damage and yield loss caused by high pathogen pressure. On the other hand, increasing Pi levels caused more damage on susceptible rice genotypes. Final nematode population densities in the root systems of resistant and susceptible rice genotypes were significantly lower under flooded conditions than under upland and drought conditions. TOG5674, TOG5675 and CG14 were more tolerant to M. graminicola infection even when grown under upland and drought conditions while IR64 and UPLRi-5 were highly sensitive.  相似文献

Experiments were conducted to determine the effectiveness and profitability of the Mi-resistance gene in tomato in suppressing populations of Meloidogyne javanica in a plastic-house with a natural infestation of the nematode. Experiments were also conducted to test for virulence and durability of the resistance. Monika (Mi-gene resistant) and Durinta (susceptible) tomato cultivars were cropped for three consecutive seasons in non-fumigated or in soil fumigated with methyl bromide at 75 g m–2 and at a cost of 2.44 euros m–2. Nematode densities were determined at the beginning and end of each crop. Yield was assessed in eight plants per plot weekly for 6 weeks. The Pf/Pi values were 0.28 and 21.6 after three crops of resistant or susceptible cultivars, respectively. Growth of resistant as opposed to susceptible tomato cultivars in non-fumigated soil increased profits by 30,000 euros ha–1. The resistant Monika in non-fumigated soil yielded similarly (P > 0.05) to the susceptible Durinta in methyl bromide fumigated soil but the resistant tomato provided a benefit of 8800 euros ha–1 over the susceptible one because of the cost of fumigation. Selection for virulence did not occur, although the nematode population subjected to the resistant cultivar for three consecutive seasons produced four times more eggs than the population on the susceptible one. Such a difference was also shown when the resistant cultivar was subjected to high continuous inoculum pressure for 14 weeks. The Mi-resistance gene can be an effective and economic alternative to methyl bromide in plastic-houses infested with root-knot nematodes, but should be used in an integrated management context to preserve its durability and prevent the selection of virulent populations due to variability in isolate reproduction and environmental conditions.  相似文献

A series of selection experiments on potato cyst nematode Globodera pallida, pathotype Pa1, tested the virulence response of the nematode to a range of resistant potato Solanum genotypes. Alleles conferring virulence against all four Solanum sources used in the study (i.e. Solanum vernei, S. multidissectum, S. sanctae-rosae and S. tuberosum ssp. andigena) were detected. Selection for multiple virulence against a combination of resistant sources resulted in the originally-selected virulence genes being retained or lost. The mechanism, or basis, of potato cyst nematode resistance differs between the Solanum species. The appropriate use of resistance cultivars produced from a range of Solanum-resistant species offers a management tool for controlling potato cyst nematode levels in infested land.  相似文献

 山东省荷泽地区薯蓣上根结线虫病发生严重。分离鉴定结果表明,该病的病原线虫是花生根结线虫Meloidogyne arenaria。该病在每年4月末开始发生,线虫在一年内发生完整的4代,完成一代平均约需要40 d,有明显的世代重叠现象。线虫主要在地表以下20~40 cm的土层内分布和为害。当地种植的4个主要品种中,肿脖温发病最重,明豆子发病最轻。  相似文献

Since the discovery of pathotypes of the potato cyst nematode (Heterodera rostochiensis Wollenweber, 1923), surveys on the distribution of these pathotypes have been carried out in different European countries. Test varieties of potato were selected according to findings resulting in the present situation of varying distinctive host-plants and of diverging programs for breeding potatoes resistant to the potato cyst nematode. With respect to rational selection and use of resistant potato varieties, there is a great need for standardization in pathotype identification (nomenclature), maintenance and supply of appropriate genotypes of Solanum species and methods for testing resistance in new potato varieties. Anticipating a cross-experiment with pathotypes and test varieties from Great Britain, the Federal Republic of Germany and the Netherlands in 1974, a preliminary test with all recognized or presumed pathotypes from these countries, but with only part of the potential differential host-plants was carried out by the Plant Protection Service at Wageningen. 为探明酸黄瓜南方根结线虫病抗性的内在机理,以抗病材料酸黄瓜与感病材料北京截头为试材,对南方根结线虫侵入与幼虫发育、根的解剖学及线虫侵入诱导形成的取食位点的细胞学结构进行了比较研究。结果表明,抗病材料酸黄瓜根系中线虫侵入率极显著低于感病材料北京截头(P0.01),酸黄瓜能够有效抵抗南方根结线虫的侵入;抗、感材料根系中雌雄虫个体比例分别为1∶12和1∶5(P0.01),抗性反应能够抑制线虫取食和幼虫发育;酸黄瓜根的解剖结构与普通栽培黄瓜相似,酸黄瓜的根不具有结构抗性;抗病反应中细胞发生过敏性坏死,感病反应中无此类现象发生。抗侵入、抑制取食和坏死反应是抗病材料酸黄瓜抗性反应的主要特征。  相似文献

 温室测定结果:拟粗壮螺旋线虫(Helicotylenchus pseudorobusius)接种量在每100cm3土壤100~1000条的条件下,接种30天后棉苗生长量比无线虫对照苗显著降低(P<0.05)。抗枯萎品种86-1在单接枯萎菌无线虫处理中不发生枯萎病,在枯萎菌(Fusarium oxysporum f. sp. vasinfectum)接种量为每克土7.5×105孢子和线虫(每100cm3±500~1000条)组合处理中发病,棉苗枯萎病情指数与线虫接种量之间呈正相关(γ=0.97)。田间试验结果表明:防治线虫和枯萎病复合症用薰蒸剂棉隆每m28~14g处理土壤比用非薰蒸性杀线虫剂克百威(呋哺丹),丙线磷(益收宝)和克线磷的效果更好。  相似文献

The response of four Mi-resistance gene tomato rootstocks to seven populations of Meloidogyne was determined in pot tests conducted in a glasshouse. Rootstocks PG76 (Solanum lycopersicum × Solanum sp.) and Brigeor (S. lycopersicum × S. habrochaites) and resistant cv. Monika (S. lycopersicum) were assessed against one population of M. arenaria, three of M. incognita, and three of M. javanica. Rootstocks Beaufort and Maxifort were assessed against one population of M. arenaria, two of M. incognita and two of M. javanica. Rootstock PG76 was highly resistant (reproduction index <10%) to all the populations, whereas rootstock Brigeor and cv. Monika were highly to moderate resistant. Rootstocks Beaufort and Maxifort showed reduced resistance or inability to suppress nematode reproduction, and their responses varied according to the population tested. Beaufort and Maxifort were susceptible to the two populations of M. javanica as Maxifort was to one of M. incognita. The reproduction index of the nematode was higher (P < 0.05) on Maxifort than Beaufort for all root-knot nematode populations.  相似文献
