In carrot (Daucus carota L.), the taproot colors orange, yellow and white are determined mostly by the Y, Y2, and Or loci. One of the most severe issues in carrot seed production is contamination by wild white carrot. To evaluate the contamination ratio, easily detectable DNA markers for white carrot are desired. To develop PCR-based DNA markers for the Y2 locus, we have re-sequenced two orange-colored carrot cultivars at our company (Fujii Seed, Japan), as well as six white- and one light-orange-colored carrots that contaminated our seed products. Within the candidate region previously reported for the Y2 locus, only one DNA marker, Y2_7, clearly distinguished white carrots from orange ones in the re-sequenced samples. The Y2_7 marker was further examined in 12 of the most popular hybrid orange cultivars in Japan, as well as ‘Nantes’ and ‘Chantenay Red Cored 2’. The Y2_7 marker showed that all of the orange cultivars examined had the orange allele except for ‘Beta-441’. False white was detected in the orange-colored ‘Beta-441’. The Y2_7 marker detected white root carrot contamination in an old open-pollinated Japanese cultivar, ‘Nakamura Senkou Futo’. This marker would be a useful tool in a carrot seed quality control for some cultivars.  相似文献

Edible coatings based on high molecular weight chitosan, pure or combined with methylcellulose or oleic acid, were applied to fresh-cut carrots cv. Nantesa by simple immersion and by applying a vacuum pulse (5 kPa for 4 min). Water vapour resistance, colour, mechanical properties and respiration rates of non-coated and coated samples were determined. Coatings improved sample appearance, since they diminished the occurrence of the white blush during storage. When applied by simple immersion, they neither conferred significant barrier properties nor the preservation of the mechanical properties of fresh-cut carrot samples. In contrast, coating application with a vacuum pulse enhanced all the positive effects, since the resistance of water vapour transmission of the samples was significantly improved, and better preservation of the sample colour and mechanical response during cold storage was obtained. Differences in film composition did not significantly affect the coating behaviour, probably due to the variability induced by different factors when coatings were applied to the carrot surface.  相似文献

以鲜牛乳和胡萝卜、菠萝为主要原料,以保加利亚乳杆菌、嗜热链球菌和双歧杆菌作为发酵剂,采用感官品质评价和正交试验方法,研究胡萝卜菠萝酸奶的加工工艺。结果表明,胡萝卜菠萝酸奶加工制作的最佳配方及工艺条件为:在鲜牛乳中添加10%的胡萝卜和菠萝汁(胡萝卜汁∶菠萝汁=1∶2),8%的白砂糖,菌种添加量为4%(保加利亚乳杆菌∶嗜热链球菌∶双歧杆菌=1∶2∶1),在42℃下发酵4 h,经冷却后熟后,制得的胡萝卜菠萝酸奶具有均匀的橙红色和独特的胡萝卜菠萝风味,其凝块均匀一致,表面细腻光滑,硬度、黏度适中,酸甜可口,清爽润喉,无异味,无或有少量的乳清析出,符合酸奶标准。  相似文献

In this study it was shown that wounding in combination with hormone stresses synergistically enhanced the accumulation of bioactive phenolic compounds in carrot tissue. Whole carrots and carrots cut in slices, pie-cuts and shreds were exposed to 250 μL/L methyl jasmonate and 1000 μL/L ethylene treatments. Results indicated that hormone stress-induced phenolic compounds and phenylalanine ammonia lyase enzyme activity were dependent upon wounding intensity. The synthesized phenolics correlated to an increase in antioxidant capacity (AOX). In addition, both stresses affected the phenolic profiles. The different proportions of chlorogenic acid, dicaffeoylquinic acid and ferulic acid, for each stress and combinations, influenced the specific AOX of the phenolic profiles (the effectiveness of the phenolic compounds to neutralize free radicals). The bitter compound isocoumarin was mostly affected by the ethylene treatment compared to methyl jasmonate or wounding alone. Exposure to combined abiotic stresses could provide more AOX to regular diets and be a less expensive alternative to genetic modifications and breeding programs.  相似文献

Heritabilities and Minimum Gene Number Estimates of Carrot Carotenoids   总被引:1,自引:0,他引:1  
Summary Broad sense heritabilities and gene numbers were estimated for the production of total carotenoids and the major component carotenoids of carrot storage roots: phytoene, ζ-carotene, β-carotene, α-carotene, and lycopene. Two crosses with different backgrounds were evaluated: orange B493 × white QAL and orange Brasilia × dark orange HCM. The HCM (high carotene mass selection), Brasilia and B493 parents had both α-carotene and β-carotene, but HCM had proportionally more α-carotene. Carotene content in F 2 populations ranged from 522 ppm to 1714 ppm in Brasilia × HCM and from 0 to 695 ppm in B493 × QAL progeny. F 2 plants segregating for absence of α-carotene were identified in B493 × QAL. Broad-sense heritabilities ranged from 28% to 48% for all carotenes except lycopene and phytoene where estimates were 44% to 89% in the Brasilia × HCM cross, All heritability values exceeded 88% for the B493 × QAL cross, except one estimate for lycopene. The estimated number of genes was 4 conditioning α-carotene, 2 to 3 each for β-carotene and total carotenes and one each for ζ-carotene, lycopene and phytoene in the orange × dark orange cross. In the orange × white cross, the estimates were 4 genes for α-carotene, 1 to 2 each for lycopene and total carotenes and 1 for each of the other carotenes. These results are in general agreement with QTL studies and they provided evidence for continuous inheritance of α-carotene, β-carotene and total carotenoids in the orange × dark orange cross and discrete inheritance for β-carotene and total carotenoids in the orange × white cross.Part of thesis “Biometrical studies and quantitative trait loci associated with major products of the carotenoids pathway of carrot (Daucus carota L.)” presented by the first author as partial fulfillment of the requirement of the PhD degree in Plant Breeding and Plant Genetics, UW-Madison, USA, 2001.  相似文献

Summary An investigation was made into the causes why bolters occur in carrot cultivation. In the first year of cultivation blooming plants are the result of a gene flow from wild to cultivated carrots. Hybridization may occur in areas where cultivated carrots are grown for seed production. Apart from bolters producing inedible roots such hybridization also causes a decline in quality of cultivated carrots in general. Small changes in morphological and phenological characteristics of the wild carrot are due to introgression from the cultivated carrot.  相似文献

Summary Investigations on the inheritance of root color in carrot (Daucus carota L.) were carried out by crossing uniformly colored roots to various tinge type roots, i.e. roots of which the xylem differs in color from the phloem.A single major gene (Y) was found to be responsible for the observed differences in progenies of orange x tinge orange-white (orange referring to phloem color, white to xylem color) crosses. Plants carrying the dominant Y-allele had either white or tinge orange-white roots, whereas plants with orange roots were of the genotype yy. Similarly one major gene (Y 2) determined the segregation found in progenies of orange x yellow crosses. In the latter crosses, plants having the dominant Y 2-allele had either yellow or tinge orange-yellow roots while the recessive would be orange. Variation in phloem color, i.e. differences between white and tinge orange-white or between yellow and tinge orange-yellow, was apparently caused by minor genes, modifiers, gene interactions, or by genes that are not involved in carotenogenesis in a direct way.When both the Y- and Y 2-genes were present, the roots were always white. Usually white roots gave a digenic segregation pattern in the F2 when crossed to orange, but there was some evidence that a third gene (Y 1) was segregating in some crosses. Tinge orange-white x yellow crosses gave approximately the same results as orange x white crosses, confirming that the same Y- and Y 2-genes were segregating.In crosses between orange lines and a light yellow line (RY) certain F1 's appeared to have a light orange xylem and a fairly dark orange phloem, which seems to be some evidence for the existence of recessive yellow. Although almost nothing is known yet about the genetics of RY it is assumed that it still carries a dominant inhibitor gene which may be leaky in heterozygous condition. The value of such a line as an aid in the selection of superior orange lines is discussed.Alpha- and beta-carotene were found to be the major pigments in orange carrot tissue; phytofluene, zetacarotene, gamma-carotene and xanthophylls were shown to be present in smaller amounts. Besides xanthophylls and a small amount of beta-carotene dark yellow carrot tissue appeared to contain an appreciable amount of an unidentified pigment (pigment I). It is well known that high hydrostatic pressure (HHP) treatment will increase the shelf life of different fruit. However, texture changes associated with these responses are not well understood in vegetables. The objective of this work was to study the effects of HHP treatment on quality and lignification of minimally processed water bamboo shoots cultivated in China. Water bamboo shoots were exposed to HHP treatment for 10 min at room temperature (25 °C) and then stored for 7 days at 4 °C. Firmness, lignin and cellulose contents, and enzyme activities of phenylalanine ammonia lyase (PAL) and peroxidase (POD), were investigated during storage. An increase in firmness of water bamboo shoots was observed, and this increase after harvest was positively correlated with higher lignin and cellulose contents, r = 0.96, P < 0.05; r = 0.98, P < 0.01 respectively. Accumulation of lignin in flesh tissue was also positively correlated with activities of PAL and POD, r = 0.97, P < 0.01; r = 0.90, P < 0.05 respectively. PAL and POD activities showed a persistent rise over the whole 7 days. HHP treatment resulted in lower firmness, delayed activities of PAL and POD, and retardation of lignin and cellulose accumulation. These studies showed that HHP might be an efficient nonthermal alternative for extended shelf life of water bamboo shoots.  相似文献

Carrot sticks are increasingly in demand as ready-to-eat products, with a major quality problem in the development of white discoloration. Modified atmosphere packaging (MAP) and edible coating have been proposed as postharvest treatments to maintain quality and prolong shelf-life. The combined application of an edible coating containing 5 mL L?1 of chitosan under two different MAP conditions (10 kPa O2 + 10 kPa CO2 in Pack A and 2 kPa O2 + 15–25 kPa CO2 in Pack B) over 12 d at 4 °C was studied. Respiration rate, microbial and sensory qualities as well as the contents of vitamin C, carotenoids and phenolics of coated and uncoated carrot sticks were evaluated. The use of the edible coating containing chitosan preserved the overall visual quality and reduced surface whiteness during storage. Microbial populations were very low and not influenced by coating or MAP. Edible coating increased respiration rates of carrot sticks, although this was only noticeable in the package with the less permeable film (Pack B). Vitamin C and carotenoids decreased during storage particularly in coated carrot sticks. In contrast, the content of total phenolics markedly increased in coated carrot sticks stored under moderate O2 and CO2 levels, while it was controlled under low O2 and high CO2 levels. The combined application of edible coating containing chitosan and moderate O2 and CO2 levels maintained quality and enhanced phenolic content in carrot sticks.  相似文献

摘要:【目的】揭示钾素水平对小麦后期蚜虫抗性的影响效果及其机理, 为指导钾肥合理施用,降低小麦蚜虫危害提供依据。【方法】利用设置K0(0 )、K1(0.1 )、K2(0.15 )三个钾 (K2Omg/kg土)处理的盆栽试验并结合室内分析,研究了供钾水平对小麦后期酚类物质、木质素含量及其代谢相关酶活性的影响,比较了处理间接种蚜虫种群变化动态的差异。【结果】接虫后14-28d,单株小麦蚜虫数量不同施钾处理间出现明显的差异,其中以K0蚜虫群体最高,其次是K2,K1最低。群体高峰期出现时间K2、K0较K1提前2-3天。三个处理相比,K1植株总酚、酚酸、木质素含量及其代谢相关酶(PAL、TAL、POD、PPO)活性最高,其次是K2,K0最低。相关分析表明,植株总酚、酚酸、木质素含量及其代谢相关酶活性与植株钾素含量相关性不显著,但与植株干物质积累量及籽粒产量相关性显著。【结论】适量供钾可增加了植株对钾素的吸收,促进了植株生长和总酚、酚酸、木质素的合成及代谢相关酶的活性,从而可增加小麦对蚜虫的抗性;而供钾不足或过量,或降低了植株对钾素的吸收,或使植株对钾素产生奢移吸收,均不利于植株干物质积累和酚类物质、木质素的合成,从而降低了小麦对蚜虫的抗性。  相似文献

以新鲜胡萝卜为原料,制作低糖果味胡萝卜果脯。在传统生产工艺中增加了缓冻预处理工序,果脯加工工艺的优化结果为:在-18℃下冻结6h,不仅能较好地促进渗糖,而且口感较佳;渗糖液添加组分为:25%的蔗糖,25%的淀粉糖浆,0.4%的柠檬酸,1.2%的鲜橘皮和0.4%的明胶;采用60~65℃恒温鼓风干燥6~7h,以1.0%的海藻酸钠胶体浸泡1min后继续干燥1~2h,制得成品的色、香、味、形俱佳。  相似文献

Salt stress was evaluated on root growth, enzyme activities (phenylalanine ammonia‐lyase or PAL and soluble plus cell wall‐bound peroxidase or POD), hydrogen peroxide (H2O2) production, total phenolic content and lignin content and composition in soybean (Glycine max L. Merrill) roots. Three‐day‐old seedlings were cultivated in half‐strength Hoagland’s solution (pH 6.0), with or without addition of 50–200 mm of NaCl, into a growth chamber (25 °C, 12/12 h light/dark photoperiod, irradiance of 280 μmol m?2 s?1) for 24 h. In general, root length and fresh and dry weights decreased after NaCl treatment. PAL activity decreased, soluble and cell wall‐bound POD activities increased, and H2O2 content significantly decreased after NaCl exposure. Consequently, total phenolic and lignin contents and p‐hydroxyphenyl (H) and syringyl (S) monomers of lignin increased in NaCl‐treated roots. Altogether, these results suggest that the effects caused by NaCl may be owing to the enhanced lignin production that solidifies the cell wall and restricts root growth.  相似文献

为研究植物生长促生细菌CH1(Brevibacillus brevis)诱导黄瓜对猝倒病的抗性的作用机制,采用生理生化的方法,测定了黄瓜根部酚类物质代谢中苯丙氨酸解氨酶(PAL)、多酚氧化酶(PPO)活性动态及阿魏酸、绿原酸、木质素、总酚、类黄酮含量变化。结果表明,病原菌(Pythium aphanidermatum)感染时,经CH1处理的植株根部PAL和PPO活性都明显高于未经CH1处理的,同时,经CH1处理的植株根部阿魏酸、绿原酸、木质素、总酚和类黄酮含量都高于未经CH1处理的。说明CH1是通过提高黄瓜植株体内酚类物质代谢的酶活性来激发酚类物质的积累,从而诱导黄瓜对猝倒病产生抗性。  相似文献

The influence of different growing periods on quality and decay of fresh-cut baby spinach leaves during chilling storage was assessed. Three different spinach crops were sown on October, December and January, respectively. On January–March the vegetable leaves were harvested, analysed for nitrate, oxalate, vitamin C and chlorophyll content and, after the minimal processing operations, the leaves were packaged and stored at 5 °C. During storage, respiration rate, microbiological quality, dry matter, chlorophyll content and colour parameters were monitored. The results show that for all harvests, the initial quality of leaves was acceptable, although slight changes may be due to variability in climatic conditions.  相似文献

研究了不同切片厚度与预冷终温对鲜切胡萝卜真空预冷效果的影响,分析了经过真空预冷和未经真空预冷的鲜切胡萝卜在贮藏期内的呼吸强度、失重率、色差和硬度的变化。结果表明,鲜切胡萝卜可比完整胡萝卜节省2/3的预冷时间;在切片厚度6 mm和预冷终温6℃时,鲜切胡萝卜的真空预冷效果最佳,失重率为1.3%、芯部温度1.5℃、预冷时间7.3 min。采用真空预冷处理方法可有效抑制鲜切胡萝卜贮藏期内的呼吸强度,降低失重率,保持其较好的感官品质。  相似文献

The effects of 40, 70 or 100 mg L?1 free chlorine neutral and acidic electrolyzed water (NEW and AEW) during the washing and disinfection step, on quality attribute changes during shelf life of fresh-cut mizuna baby leaves, were studied. Physiological, nutritional, enzymatic, sensory, and microbial changes throughout 11 days at 5 °C were monitored. Results were compared to those reached with a conventional industrial treatment of 100 mg L?1 NaClO at pH 6.5 and with a control washing with deionised water. Both NEW and AEW showed an inhibitory effect on natural microflora growth and retained the main quality attributes. Total chlorophyll content was preserved after shelf life. Initial total phenolic contents ranged between 1868 and 2518 mg CAE kg?1 fw for AEW 40 and AEW 100 treatments respectively and slightly increased throughout shelf life. In contrast, after shelf life the total antioxidant activity recorded on the processing day decreased around 35%. Throughout shelf life EW induced an increase in catalase activity while superoxide dismutase activity decreased. Scanning electron microscopy of the leaves showed that neither NEW nor AEW affected their surface structure. To the best of our knowledge, the effects of NEW and AEW on bioactive quality parameters, as well as on antioxidant enzyme activities for fresh-cut baby leaves are first reported here. EW provides an alternative sanitizing technique to NaClO for maintaining the quality of fresh-cut mizuna baby leaves up to 11 days at 5 °C.  相似文献

The aim of this work was to investigate the loss of freshness of fresh-cut pineapple samples stored at different temperatures using non-destructive spectroscopic methods. Three lots of fresh cut pineapples (Ananas comosus L. cv. Golden Ripe, from Costa Rica), packaged in PVC trays (250 g) were analyzed during storage at three different temperatures (5.3, 8.6 and 15.8 °C). Loss of quality of these fruit was evaluated by chemical and microbiological parameters and using NIR and MIR spectroscopy. The FT-NIR spectra were acquired in reflectance mode directly on the slice of fresh-cut pineapple, over the range 12,500–3900 cm−1, while FT-IR spectra were collected over the range 4000–700 cm−1 using an horizontal ATR cell. Some chemical and microbiological parameters were also measured. Principal component analysis (PCA) was applied to the second derivative of the spectra to uncover molecular modifications occurring over the storage time. A clear discrimination between “fresh” and “old” samples was obtained and a stability time corresponding to the time of the initial loss of freshness was defined at each temperature. The stability times revealed by NIR spectroscopy were in good accordance with those evaluated by MIR. At each temperature the stability times (i.e. the initial loss of freshness times) defined by spectroscopic techniques (4–5 d at 5.3 °C, 3–4 d at 8.6 °C and 1 d at 15.8 °C) were associated with a mesophilic bacteria count ranging between 105 and 106 CFU g−1 and lower than the maximum limit for mesophilic bacteria (<5 × 107 CFU g−1) given by French hygienic regulations at consumption.These results show that NIR and MIR spectroscopy could support conventional techniques (chemical and microbiological analysis) in studying shelf-life of fresh-cut fruit. In particular these techniques define the initial loss of freshness time, indicating a product which rapidly will be no longer acceptable if stored beyond that time. The main advantage of using IR spectroscopic techniques is to rapidly draw a profile of the product related to its change in quality.  相似文献

Peroxyacetic acid (PAA) is a strong oxidizer and exerts antimicrobial properties. The effect of a decontamination step with 80 and 250 mg L−1 PAA on shelf-life of grated carrots stored under equilibrium modified atmospheric packaging at 7 °C was determined and compared with the shelf-life of unwashed and water-washed carrots. Microbial parameters, including total aerobic plate count, numbers of lactic acid bacteria, Lactobacillae and yeasts, and sensory quality were evaluated. Next to these parameters, atmospheric gas composition, pH and nutrient content were also monitored. The suggested packaging configuration prevented CO2 accumulation, but at the end of the study anoxic conditions were reached for unwashed carrots and carrots washed with 80 mg L−1 PAA. The microbial shelf-life of water-washed carrots was 4 d based on the yeast count, whereas the flavour was not acceptable after 5 d. The total aerobic plate count and the yeast count determined the shelf-life of carrots treated with 80 mg L−1 PAA on 5 d, whereas the flavour was unacceptable after 7 d. None of the microbial parameters determined the shelf-life of carrots washed with 250 mg L−1 PAA. However, this treatment had already a pronounced adverse effect on the initial sensory quality. Water washing already decreased the content of all individually studied nutrients (−16 to −28%), except for lutein content and the antioxidant capacity. Additional losses after adding PAA on day 0 were found for α-tocopherol and phenols. Regardless of the applied treatment, α- and β-carotene remained stable during storage, whereas ζ-carotene, lutein and α-tocopherol were unstable. The phenol content and the antioxidant capacity of unwashed, water-washed and 80 mg L−1 PAA-treated carrots increased significantly at the end of the storage period, whereas no changes were found in carrots treated with 250 mg L−1 PAA.On the condition that carrots were packed under an adequate EMA, the 80 mg L−1 PAA treatment showed possibilities for extending shelf-life without pronounced effects on nutrient content.  相似文献

为明确氮肥施用模式对小麦茎秆木质素合成与积累及抗倒伏能力的影响,2011-2012和2012-2013生长季,选用济麦22 (抗倒伏)和山农16 (不抗倒伏)品种,分析不同生育期追施氮肥处理的茎秆木质素含量和木质素合成相关酶活性,及其与抗倒伏指数和产量的关系。各处理总施氮量为240 kg hm−2,其中80 kg hm−2播前基施,其余氮肥按4种模式追施,分别是起身期和孕穗期各一半(N1)、拔节期全施(N2)、拔节期和开花期各一半(N3)和孕穗期全施(N4)。与N2和N3处理相比,N1和N4处理的茎秆木质素含量高,茎秆抗折力大,茎秆抗倒能力强。各处理对木质素合成关键酶活性有显著影响,其中N1的苯丙氨酸解氨酶(PAL)、酪氨酸解氨酶(TAL)和过氧化物酶(POD)活性较高;N2处理第2节间形成后0~7 d时PAL、TAL和POD活性显著高于其他处理,但第2节间形成后21 d时迅速下降;N3处理第2节间形成后0~21 d时PAL、TAL和POD活性低,但开花期(21 d)追氮延缓了3种酶活性的降低;N4处理显著提高了第2节间形成14 d后的酶活性。在乳熟期和蜡熟期,两品种的基部茎秆抗折力和抗倒伏指数均以N1和N4处理显著优于其他处理,并且N1和N4处理的籽粒产量也显著高于其他处理。试验结果表明,孕穗期追肥的N1和N4处理有助于提高茎秆中PAL、TAL、POD活性及木质素含量,提高小麦茎秆的机械强度及抗倒伏能力,降低小麦倒伏面积和倒伏程度。  相似文献

Edible composite coatings based on hydroxypropyl methylcellulose (HPMC), hydrophobic components (beeswax and shellac), and food preservatives with antifungal properties were evaluated on ‘Valencia’ oranges during long-term cold storage. Selected food preservatives included potassium sorbate (PS), sodium benzoate (SB), sodium propionate (SP), and their mixtures. Intact oranges or oranges artificially inoculated with Penicillium digitatum or Penicillium italicum were coated and stored up to 60 d at 5 °C followed by 7 d of shelf-life at 20 °C. Some antifungal HPMC–lipid coatings significantly reduced incidence and severity of both green (GM) and blue (BM) molds on inoculated and cold-stored oranges, and PS + SP-based coating was the most effective. In general, the coatings controlled GM better than BM. After 30 and 60 d at 5 °C plus 7 d at 20 °C, fruit weight loss, rind firmness, internal gas concentrations, ethanol and acetaldehyde contents of the juice, sensory flavor, off-flavor, and fruit appearance were not adversely affected by application of the antifungal coatings, which showed promise as potential substitutes for citrus commercial waxes. However, further studies should follow to improve some coating physical characteristics in order to provide better water loss control and higher gloss on coated oranges.  相似文献
